Background: Fruit storage methods such as dynamic controlled atmosphere (DCA) technology enable adjusting the level of oxygen in the storage room, according to the physiological state of the product to slow down the ripening process. However, the successful application of DCA requires precise and reliable sensors of the oxidative stress of the fruit. In this study, respiration rate and chlorophyll fluorescence (CF) signals were evaluated after introducing a novel predictors of apples' hypoxic stress based on laser speckle imaging technique (LSI). Results: Both chlorophyll fluorescence and LSI signals were equally good for stress detection in principle. However, in an application with automatic detection based on machine learning models, the LSI signal proved to be superior, due to its stability and measurement repeatability. Moreover, the shortcomings of the CF signal appear to be its inability to indicate oxygen stress in tissues with low chlorophyll content but this does not apply to LSI. A comparison of different LSI signal processing methods showed that method based on the dynamics of changes in image content was better indicators of stress than methods based on measurements of changes in pixel brightness (inertia moment or laser speckle contrast analysis). Data obtained using the near-infrared laser provided better prediction capabilities, compared to the laser with red light. Conclusions: The study showed that the signal from the scattered laser light phenomenon is a good predictor for the oxidative stress of apples. Results showed that effective prediction using LSI was possible and did not require additional signals. The proposed method has great potential as an alternative indicator of fruit oxidative stress, which can be applied in modern storage systems with a dynamically controlled atmosphere. Significance: In the realm of cerebrovascular monitoring, primary metrics typically include blood pressure, which influences cerebral blood flow (CBF) and is contingent upon vessel radius. Measuring CBF noninvasively poses a persistent challenge, primarily attributed to the difficulty of accessing and obtaining signal from the brain. Aim: Our study aims to introduce a compact speckle contrast optical spectroscopy device for noninvasive CBF measurements at long source-to-detector distances, offering cost-effectiveness, and scalability while tracking blood flow (BF) with remarkable sensitivity and temporal resolution. Approach: The wearable sensor module consists solely of a laser diode and a board camera. It can be easily placed on a subject's head to measure BF at a sampling rate of 80 Hz. Results: Compared to the single-fiber-based version, the proposed device achieved a signal gain of about 70 times, showed superior stability, reproducibility, and signal-to-noise ratio for measuring BF at long source-to-detector distances. The device can be distributed in multiple configurations around the head. Conclusions: Given its cost-effectiveness, scalability, and simplicity, this laser-centric tool offers significant potential in advancing noninvasive cerebral monitoring technologies. Prior research has indicated the feasibility of assessing growth--associated activity in bacterial colonies through the application of laser speckle imaging techniques. A subpixel correlation method was employed to identify variations in sequential laser speckle images, thereby facilitating the visualization of specific zones indicative of microbial growth within the colony. Such differentiation between active (growing) and inactive (non-growing) bacterial colonies holds considerable implications for medical applications, like bacterial response to certain drugs or antibiotics. The present study substantiates the capability of laser speckle imaging to categorize bacterial colonies as growing or non-growing, a parameter which nonvisible in colonies when observed under white light illumination. The recent pandemic showed that the current global research strategies on vaccine development in an emergency period necessitates more optimized supplementary techniques to observe instant progressive vaccines' subtle effects on human metabolisms to make better and speedy evolutionary health assessments. To fill this gap, we have followed a multi-disciplinary approach exploiting AI, laser-optics, and specific imaging methods. The proposed technique can make progressive observations on Covid-19 Astra Zeneca vaccination effects on skin cellular network by use of the well-established technique—Intelligent Laser Speckle Classification (ILSC), as Covid-19 is a skin-affecting systemic disease. The method also managed to distinguish between three different subject groups via their laser speckle skin image samplings, grouped as early-vaccinated, late-vaccinated and non-vaccinated participants. The results have proven that the ILSC technique, in association with the parametrically optimised Bayesian network, can classify hidden skin changes of vaccinated and non-vaccinated individuals up to 90% accuracy and is also capable of detecting instant progressive developments pertaining to skin cellular properties. The proposed method has also proven that the continuous Covid-19 vaccine effect on the sub-skin layers can be observable by high frequency and speedy non-invasive data collection in real-time with high reliability. Introduction and hypothesis: Preclinical studies have shown that neuromodulation can increase vaginal blood perfusion, but the effect on vulvar blood perfusion is unknown. We hypothesized that pudendal and tibial nerve stimulation could evoke an increase in vulvar blood perfusion. Methods: We used female Sprague–Dawley rats for non-survival procedures under urethane anesthesia. We measured perineal blood perfusion in response to 20-minute periods of pudendal and tibial nerve stimulation using laser speckle contrast imaging (LSCI). After a thoracic-level spinalization and a rest period, we repeated each stimulation trial. We calculated average blood perfusion before, during, and after stimulation for three perineal regions (vulva, anus, and inner thigh), for each nerve target and spinal cord condition. Results: We observed a significant increase in vulvar, anal, and inner thigh blood perfusion during pudendal nerve stimulation in spinally intact and spinalized rats. Tibial nerve stimulation had no effect on perineal blood perfusion for both spinally intact and spinalized rats. Conclusions: This is the first study to examine vulvar hemodynamics with LSCI in response to nerve stimulation. This study demonstrates that pudendal nerve stimulation modulates vulvar blood perfusion, indicating the potential of pudendal neuromodulation to improve genital blood flow as a treatment for women with sexual dysfunction. This study provides further support for neuromodulation as a treatment for women with sexual arousal disorders. Studies in unanesthetized animal models of genital arousal disorders are needed to obtain further insights into the mechanisms of neural control over genital hemodynamics. [ABSTRACT FROM AUTHOR]

Background: To measure the degree of relative ischemia caused by skin closure, we explored the potential utility of intraoperative surface blood flow measurement with laser speckle imaging (LSI). Methods: Prospective observational study of eight subjects that underwent intraoperative LSI during elective cranial neurosurgical procedures at the time of skin closure. Results: Seven 1st time incisions, with closure techniques including sutures (n = 3), staples (n = 3), and one after galeal sutures. When compared to the control region, there was a mean 63.7% reduction in flow across all seven subjects (range 18.7-95.32%). Comparing by closure type, a higher flow reduction in the three subjects with suture closure (80.7% reduction) compared to staples (61.9% reduction, P = 0.0379). One subject had a complex wound where tightening and loosening of sutures were performed to ensure adequate perfusion. Suturing resulted in significantly more local decreased flow compared to staples (P < 0.0001). Conclusion: These findings demonstrate the relative feasibility of using LSI for preoperative vascular flow assessment in planning complex incision closure. These data also provide preliminary support for the hypothesis that skin closure itself causes relative ischemia compared to deep approximation or cautery of the skin edge and that the relative ischemia from staples closure is generally less than from suture closure. Objective: A handheld device was developed and qualified for in vivo human skin evaluation using laser speckle imaging technology. Methods: Each laser speckle device prototype allows the choice of up to three different laser wavelengths in the range of 400 nm to 800 nm in total. Speckle pattern analysis gives various speckle parameters, for example, speckle contrast, speckle size, speckle modulation or fractal dimension. The developed laser speckle device prototypes were evaluated investigating three skin issues. Results: We receive reproducible results from the speckle imaging device. For skin ageing, we found significant changes within three age groups. The effect of a methyl nicotinate treatment was clearly visible and quantifiable using a moorFLPI device as well as our speckle imaging device. In terms of basal cell carcinoma diagnosis, we found significant differences between normal and diseased skin, even though the number of samples was limited. Recurrent waves of spreading depolarization (SD) occur in brain injury and are thought to affect outcomes. What triggers SD in intracerebral hemorrhage is poorly understood. We employed intrinsic optical signaling, laser speckle flowmetry, and electrocorticography to elucidate the mechanisms triggering SD in a collagenase model of intracortical hemorrhage in mice. Hematoma growth, SD occurrence, and cortical blood flow changes were tracked. During early hemorrhage (0–4 h), 17 out of 38 mice developed SDs, which always originated from the hematoma. No SD was detected at late time points (8–52 h). Neither hematoma size, nor peri-hematoma perfusion were associated with SD occurrence. Further, arguing against ischemia as a trigger factor, normobaric hyperoxia did not inhibit SD occurrence. Instead, SDs always occurred during periods of rapid hematoma growth, which was two-fold faster immediately preceding an SD compared with the peak growth rates in animals that did not develop any SDs. Induced hypertension accelerated hematoma growth and resulted in a four-fold increase in SD occurrence compared with normotensive animals. Altogether, our data suggest that spontaneous SDs in this intracortical hemorrhage model are triggered by the mechanical distortion of tissue by rapidly growing hematomas. [ABSTRACT FROM AUTHOR]

Purpose: Laser speckle flowgraphy (LSFG) is a noninvasive method for quantitative evaluation of blood flow using the mean blur rate (MBR) as the blood flow index. We investigated whether LSFG can intraoperatively detect the demarcation line after vessel dissection and reduce the incidence of anastomotic leakage (AL). Methods: This study included 36 patients who underwent left-sided colorectal surgery. First, we compared the demarcation line (determined by LSFG) with the transection line (TL) at which the marginal vessels were divided. We then measured the MBR on both sides of the TL to determine where the MBR changed significantly. We investigated the presence or absence of significant differences between the MBR on the proximal side and that on the distal side of the TL. Finally, we retrospectively compared the patient characteristics and AL rates in the LSFG group (n = 36) and control group (n = 87). Results: In total, 58.3% (21/36) of the demarcation lines determined by LSFG matched the TL. The median distance between the demarcation line determined by LSFG and the TL was 0.0 mm (0.0–12.1 mm). The MBR sharply decreased at the TL in 80.6% (29/36) of cases. The median MBR was significantly lower on the distal than proximal side. The AL rate was not significantly lower in the LSFG group than in the control group. Conclusion: LSFG accurately detected the demarcation line during surgery. This paper reports a straightforward technique for three-dimensional (3D) visualization of a flow profile by a hybrid algorithm combining Fourier transform orthogonal fringe projection and laser speckle imaging techniques. The use of orthogonal projection aims to suppress the zero order allowing surface reconstruction with high spatial resolution and accuracy while analyzing the intensity fluctuations of diffuse backscattered laser light providing 2D flow information. Once both are achieved, 3D flow visualization can be displayed. The method is experimentally validated first with a plastic tube filled with scattering liquid (milk) running at various controlled flow rates and then with the tube embedded under scattering layers (chicken breast) of varying thickness. The system includes a single, common camera, a commercial projector (profilometry channel), a laser light source (flow channel), and a computer station. In addition, orthogonal projection processing was combined with Hilbert transform, increasing the visualization and resolution of the measured flow profile. [ABSTRACT FROM AUTHOR]

Neurovascular coupling is a fundamental response that links activity to perfusion. Traditional paradigms of neurovascular coupling utilize somatosensory stimulation to activate the primary sensory cortex through subcortical relays. Therefore, examination of neurovascular coupling in disease models can be confounded if the disease process affects these multisynaptic pathways. Optogenetic stimulation is an alternative to directly activate neurons, bypassing the subcortical relays. We employed minimally invasive optogenetic cortical activation through intact skull in Thy1-channelrhodopsin-2 transgenic mice, examined the blood flow changes using laser speckle imaging, and related these to evoked electrophysiological activity. Our data show that optogenetic activation of barrel cortex triggers intensity- and frequency-dependent hyperemia both locally within the barrel cortex (>50% CBF increase), and remotely within the ipsilateral motor cortex (>30% CBF increase). Intriguingly, activation of the barrel cortex causes a small (∼10%) but reproducible hypoperfusion within the contralateral barrel cortex, electrophysiologically linked to transhemispheric inhibition. Cortical spreading depression, known to cause neurovascular uncoupling, diminishes optogenetic hyperemia by more than 50% for up to an hour despite rapid recovery of evoked electrophysiological activity, recapitulating a unique feature of physiological neurovascular coupling. Background  Success of free tissue transfer depends on standardized intraoperative flap design, microsurgical technique, and postoperative monitoring. We sought to investigate whether laser speckle imaging (LSI) is suitable for optimization of intraoperative flap design and postoperative monitoring of free flaps with skin paddles. Methods  Skin perfusion was assessed with LSI in 27 free flaps after dissection at the donor site, after anastomosis at the recipient site, after inset and on postoperative days (POD) 1, 5, and 10. Skin perfusion of the whole flap (ROI [region of interest]-1) and the area over the pedicle (ROI-2) were compared between patients with and without postoperative complications (POC+ and POC − , respectively). A receiver operating characteristic (ROC) analysis was performed to determine the optimal cutoff for perfusion during LSI-guided microsurgery. Results  In flaps without or only minor POC, intraoperatively measured perfusion over ROI-2 was significantly higher compared with ROI-1, whereas no significant differences were found for flaps with major POC. Perfusion of ROI-1 and 2 intraoperatively and on POD 1 was significantly lower in the POC+ compared with the POC− group (p  < 0.05). ROC analysis yielded a threshold of 107 perfusion units (PU) at ROI-2 with an area under the curve (AUC) of > 0.8 for identification of flaps with major POC. Conclusion  LSI is an easy to use, noninvasive technique for identification of malperfused areas in free flaps, thus allowing for intraoperative decision-making on flap dimensions and postoperative monitoring. The mechanical anisotropy of aluminium alloy AA2024-T3 with strong cold-rolled crystallographic texture is investigated in this paper. A non-contact and non-destructive laser speckle imaging sensor is developed for surface strain measurement so that extensive mechanical tests can be readily performed without surface preparation on the specimens. Both Hill's 48 and Yld2004-18p yield functions are accurately calibrated using the experimental results from uniaxial tensile, pure shear and plane-strain tensile tests. The constitutive models based on these yield functions were successfully implemented in a commercial finite element (FE) package Abaqus via user material subroutine UMAT. Simulations of single element tensile tests and pure shear tests were carried out to validate the accuracy of UMAT subroutine. The performance of these constitutive models was further evaluated with deep drawing experiments in which the predicted earing profiles were compared with those obtained from FE simulations. The results showed that the constitutive model based on Yld2004-18p yield function accurate representing the anisotropic behaviour of AA2024-T3. Effective defects detection method for silicone polymer insulators is desperately needed to eliminate defective insulators in factory examination and make early warnings for accidents of in-service insulators. This paper investigates a laser shearography method that reveals surface deformation anomalies induced by internal defects after applying thermal excitation. Samples with artificial defects of different sizes and depths in the silicone rubber sheath were tested. Both fringe patterns and phase distribution maps were generated after the experiments for defects identification by indicating the displacement derivative with respect to the shearing direction. The relationship between the defect parameters and the relative phase difference was discussed, and a parameter named detection sensitivity was defined to evaluate the system detection ability on different defects. Finally, Canny edge detection algorithm and proposed image gradient method were employed to quantitatively estimate the defect size, and the ability of identifying defects was also improved. With its noncontact testing, full-field measurement, real-time imaging, and visual results, laser speckle imaging would have quite promising applications in nondestructive testing of silicone polymer insulators and it can be extended for other composite materials insulating equipment. Infrared thermal imaging of brain temperature changes is useful for evaluating cortical activity and disease states, such as stroke. However, the changes depend on a balance between changes in heat generation from metabolism and in heat convection related to blood flow. To discriminate between these effects and gain a clearer understanding of neurovascular metabolic coupling, brain temperature imaging must be improved to measure temperature and blood flow simultaneously. We develop an imaging technique that shows a twodimensional (2-D) distribution of absolute brain temperature and relative cerebral blood flow changes in anesthetized rats by combining infrared thermal imaging with laser speckle imaging. The changes in brain metabolism and cerebral blood flow are achieved using two different anesthetics (isoflurane and a-chloralose) to evaluate our system. Isoflurane increased cerebral blood flow but decreased metabolism, whereas a-chloralose decreased both parameters. This technique enables simultaneous visualization of brain surface changes in temperature and cerebral blood flow in the same regions. To realize visualization of the skin microvascular dysfunction of type 1 diabetic mice, we combined laser speckle contrast imaging and hyperspectral imaging to simultaneously monitor the noradrenaline (NE)- induced responses of vascular blood flow and blood oxygen with the development of diabetes through optical clearing skin window. The main results showed that venous and arterious blood flow decreased without recovery after injection of NE; furthermore, the decrease of arterious blood oxygen induced by NE greatly weakened, especially for 2- and 4-week diabetic mice. This change in vasoconstricting effect of NE was related to the expression of α1-adrenergic receptor. This study demonstrated that skin microvascular function was a potential research biomarker for early warning in the occurrence and development of diabetes. The in vivo skin optical clearing method provides a feasible solution to realize visualization of cutaneous microvessels for monitoring microvascular reactivity under pathological conditions. Objective: Monitoring hemodynamic and vascular changes in the acute stages of mouse stroke models is invaluable in studying ischemic stroke pathophysiology. However, there lacks a tool to simultaneously and dynamically investigate these changes. Methods: We integrated laser speckle imaging (LSI) and visible-light optical coherence tomography (Vis-OCT) to reveal dynamic vascular responses in acute stages in the distal middle cerebral artery occlusion (dMCAO) model in rodents. LSI provides full-field, real-time imaging to guide Vis-OCT imaging and monitor the dynamic cerebral blood flow (CBF). Vis-OCT offers depth-resolved angiography and oxygen saturation (sO2) measurements. Results: Our results showed detailed CBF and vasculature changes before, during, and after dMCAO. After dMCAO, we observed insignificant sO2 variation in arteries and arterioles and location–dependent sO2 drop in veins and venules. We observed that higher branch-order veins had larger drops in sO2 at the reperfusion stage after dMCAO. Conclusion: This work suggests that integrated LSI and Vis-OCT is a promising tool for investigating ischemic stroke in mouse models. In this study, we made use of dual‐wavelength laser speckle imaging (DW‐LSI) to assess cerebral blood flow (CBF) in the BTBR‐genetic mouse model of autism spectrum disorder, as well as control (C57Bl/6J) mice. Since the deficits in social behavior demonstrated by BTBR mice are attributed to changes in neural tissue structure and function, we postulated that these changes can be detected optically using DW‐LSI. BTBR mice demonstrated reductions in both CBF and cerebral oxygen metabolism (CMRO2), as suggested by studies using conventional neuroimaging technologies to reflect impaired neuronal activation and cognitive function. To validate the monitoring of CBF by DW‐LSI, measurements with laser Doppler flowmetry (LDF) were also performed which confirmed the lowered CBF in the autistic‐like group. Furthermore, we found in vivo cortical CBF measurements to predict the rate of hippocampal neurogenesis, measured ex vivo by the number of neurons expressing doublecortin or the cellular proliferation marker Ki‐67 in the dentate gyrus, with a strong positive correlation between CBF and neurogenesis markers (Pearson, r = 0.78; 0.9, respectively). These novel findings identifying cortical CBF as a predictive parameter of hippocampal neurogenesis highlight the power and flexibility of the DW‐LSI and LDF setups for studying neurogenesis trends under normal and pathological conditions. Laser speckle imaging (LSI) is a wide-field optical technique that enables superficial blood flow quantification. LSI is normally performed in a mounted configuration to decrease the likelihood of motion artifact. However, mounted LSI systems are cumbersome and difficult to transport quickly in a clinical setting for which portability is essential in providing bedside patient care. To address this issue, we created a handheld LSI device using scientific grade components. To account for motion artifact of the LSI device used in a handheld setup, we incorporated a fiducial marker (FM) into our imaging protocol and determined the difference between highest and lowest speckle contrast values for the FM within each data set (Kbest and Kworst). The difference between Kbest and Kworst in mounted and handheld setups was 8% and 52%, respectively, thereby reinforcing the need for motion artifact quantification. When using a threshold FM speckle contrast value (KFM) to identify a subset of images with an acceptable level of motion artifact, mounted and handheld LSI measurements of speckle contrast of a flow region (KFLOW) in in vitro flow phantom experiments differed by 8%. Without the use of the FM, mounted and handheld KFLOW values differed by 20%. To further validate our handheld LSI device, we compared mounted and handheld data from an in vivo porcine burn model of superficial and full thickness burns. The speckle contrast within the burn region (KBURN) of the mounted and handheld LSI data differed by <4% when accounting for motion artifact using the FM, which is less than the speckle contrast difference between superficial and full thickness burns. Collectively, our results suggest the potential of handheld LSI with an FM as a suitable alternative to mounted LSI, especially in challenging clinical settings with space limitations such as the intensive care unit. The present study was aimed to in vivo assess the blood flow oscillatory patterns in rat pial microvessels during 30 min bilateral common carotid artery occlusion (BCCAO) and 60 min reperfusion by laser speckle imaging (LSI). Pial microcirculation was visualized by fluorescence microscopy. The blood flow oscillations of single microvessels were recorded by LSI; spectral analysis was performed by Wavelet transform. Under baseline conditions, arterioles and venules were characterized by blood flow oscillations in the frequency ranges 0.005-0.0095 Hz, 0.0095-0.021 Hz, 0.021-0.052 Hz, 0.052-0.150 Hz and 0.150-0.500 Hz. Arterioles showed oscillations with the highest spectral density when compared with venules. Moreover, the frequency components in the ranges 0.052-0.150 Hz and 0.150-0.500 were predominant in the arteriolar total power spectrum; while, the frequency component in the range 0.150-0.500 Hz showed the highest spectral density in venules. After 30 min BCCAO, the arteriolar spectral density decreased compared to baseline; moreover, the arteriolar frequency component in the range 0.052-0.150 Hz significantly decreased in percent spectral density, while the frequency component in the range 0.150-0.500 Hz significantly increased in percent spectral density. However, an increase in arteriolar spectral density was detected at 60 min reperfusion compared to BCCAO values; consequently, an increase in percent spectral density of the frequency component in the range 0.052-0.150 Hz was observed, while the percent spectral density of the frequency component in the range 0.150-0.500 Hz significantly decreased. The remaining frequency components did not significantly change during hypoperfusion and reperfusion. The changes in blood flow during hypoperfusion/reperfusion caused tissue damage in the cortex and striatum of all animals. A new approach is proposed for statistically analysis of laser speckle signals emerged from a living biological tissue based on eigen-decomposition to separate the dynamic speckle signals due to moving blood cells from the static speckle signals due to static tissue components, upon which to achieve angiography of the interrogated tissue in vivo. The proposed approach is tested by imaging mouse ear pinna in vivo, demonstrating its capability of providing detailed microvascular networks with high contrast, and high temporal and spatial resolutions. Background: Previous neuroimaging studies have shown the hemodynamic effect of either preconditioning or postconditioning anesthesia in ischemic stroke model. However, the anesthetic effect in hemodynamics during and immediately after the stroke modeling surgery remains unknown due to the lack of appropriate anesthesia-free stroke model and intraoperative imaging technology. In the present study, we utilized our recently developed photothrombotic model of focal cerebral ischemia in conscious and freely moving rats, and investigated transient hemodynamic changes with or without isoflurane administration. Laser speckle imaging was applied to acquire real-time two-dimensional full-field cerebral blood flow (CBF) information throughout the surgical operations and early after.Results: Significantly larger CBF reduction area was observed in conscious rats from 8 min immediately after the onset of stroke modeling, compared with anesthetized rats. Stroke rats without isoflurane administration also showed larger lesion volume identified by magnetic resonance imaging 3 h post occlusion (58.9%), higher neurological severity score 24 h post occlusion (28.3%), and larger infarct volume from 2,3,5-triphenyltetrazolium chloride staining 24 h post occlusion (46.9%).Conclusions: Our results demonstrated that the hemodynamic features were affected by anesthetics at as early as during the stroke induction. Also, our findings about the neuroprotection of intraoperative anesthetics administration bring additional insights into understanding the translational difficulty in stroke research. Laser Doppler flowmetry (LDF) and laser speckle contrast analysis (LASCA) both utilize the spatiotemporal properties of laser speckle patterns to assess microcirculatory blood flow in tissue. Although the techniques analyze the speckle pattern differently, there is a close relationship between them. We present a theoretical overview describing how the LDF power spectrum and the LASCA contrast can be calculated from each other, and how both these can be calculated from an optical Doppler spectrum containing various degrees of Doppler shifted light. The theoretical relationships are further demonstrated using time-resolved speckle simulations. A wide range of Monte Carlo simulated tissue models is then used to show how perfusion estimates for LDF and LASCA are affected by changes in blood concentration and speed distribution, as well as by geometrical and optical properties. We conclude that perfusion estimates from conventional single exposure time LASCA are in general more sensitive to changes in optical and geometrical properties and are less accurate in the prediction of real perfusion changes, especially speed changes. Since there is a theoretical one-to-one relationship between Doppler power spectrum and contrast, one can conclude that those drawbacks with the LASCA technique can be overcome using a multiple exposure time setup. Background: Recent research has identified an increased rate of mortality associated with fluid bolus therapy for severe sepsis and septic shock, but the mechanisms are still not well understood. Fluid resuscitation therapy administered for sepsis and septic shock targets restoration of the macro-circulation, but the pathogenesis of sepsis is complex and includes microcirculatory dysfunction. Objective: The objective of the study is to systematically review data comparing the effects of different types of fluid resuscitation on the microcirculation in clinically relevant animal models of lipopolysaccharide-induced sepsis. Methods: A structured search of PubMed/MEDLINE and EMBASE for relevant publications from 1 January 1990 to 31 December 2015 was performed, in accordance with PRISMA guidelines. Results: The number of published papers on sepsis and the microcirculation has increased steadily over the last 25 years. We identified 11 experimental animal studies comparing the effects of different fluid resuscitation regimens on the microcirculation. Heterogeneity precluded any meta-analysis. Conclusions: Few animal model studies have been published comparing the microcirculatory effects of different types of fluid resuscitation for sepsis and septic shock. Biologically relevant animal model studies remain necessary to enhance understanding regarding the mechanisms by which fluid resuscitation affects the microcirculation and to facilitate the transfer of basic science discoveries to clinical applications. Laser speckle imaging (LSI) is an interferometric technique that provides information about the relative speed of moving scatterers in a sample. Photothermal LSI overcomes limitations in depth resolution faced by conventional LSI by incorporating an excitation pulse to target absorption by hemoglobin within the vascular network. Here we present results from experiments designed to determine the mechanism by which photothermal LSI decreases speckle contrast. We measured the impact of mechanical properties on speckle contrast, as well as the spatiotemporal temperature dynamics and bulk convective motion occurring during photothermal LSI. Our collective data strongly support the hypothesis that photothermal LSI achieves a transient reduction in speckle contrast due to bulk motion associated with thermally driven convection. Bell's palsy is a kind of peripheral neural disease that cause abrupt onset of unilateral facial weakness. In the pathologic study, it was evidenced that ischemia of facial nerve at the affected side of face existed in Bell's palsy patients. Since the direction of facial nerve blood flow is primarily proximal to distal, facial skin microcirculation would also be affected after the onset of Bell's palsy. Therefore, monitoring the full area of facial skin microcirculation would help to identify the condition of Bell's palsy patients. In this study, a non-invasive, real time and full field imaging technology - laser speckle imaging (LSI) technology was applied for measuring facial skin blood perfusion distribution of Bell's palsy patients. 85 participants with different stage of Bell's palsy were included. Results showed that Bell's palsy patients' facial skin perfusion of affected side was lower than that of the normal side at the region of eyelid, and that the asymmetric distribution of the facial skin perfusion between two sides of eyelid is positively related to the stage of the disease (P< 0.001). During the recovery, the perfusion of affected side of eyelid was increasing to nearly the same with the normal side. This study was a novel application of LSI in evaluating the facial skin perfusion of Bell's palsy patients, and we discovered that the facial skin blood perfusion could reflect the stage of Bell's palsy, which suggested that microcirculation should be investigated in patients with this neurological deficit. We introduce a microfluidic approach to simulate tumor hypoxia and vascular anomaly. Polydimethylsiloxane (PDMS) phantoms with embedded microchannel networks were fabricated by a soft lithography process. A dialysis membrane was sandwiched between two PDMS slabs to simulate the controlled mass transport and oxygen metabolism. A tortuous microchannel network was fabricated to simulate tumor microvasculature. A dual-modal multispectral and laser speckle imaging system was used for oxygen and blood flow imaging in the tumor-simulating phantom. The imaging results were compared with those of the normal vasculature. Our experiments demonstrated the technical feasibility of simulating tumor hypoxia and vascular anomalies using the proposed PDMS phantom. Such a phantom fabrication technique may be potentially used to calibrate optical imaging devices, to study the mechanisms for tumor hypoxia and angiogenesis, and to optimize the drug delivery strategies. The measurement of dynamic changes in brain hemodynamic and metabolism events following head trauma could be valuable for injury prognosis and for planning of optimal medical treatment. Specifically, variations in blood flow and oxygenation levels serve as important biomarkers of numerous pathophysiological processes. We employed the dual-wavelength laser speckle imaging (DW-LSI) technique for simultaneous monitoring of changes in brain hemodynamics and cerebral blood flow (CBF) at early stages of head trauma in a mouse model of intact head injury (n = 10). For induction of head injury, we used a weight-drop device involving a metal mass (~50 g) striking the mouse's head in a regulated manner from a height of ~90 cm. In comparison to baseline measurements, noticeable dynamic variations were revealed immediately and up to 1 h postinjury, which indicate the severity of brain damage and highlight the ability of the DW-LSI arrangement to track brain pathophysiology induced by injury. To validate the monitoring of CBF by DW-LSI, measurements with laser Doppler flowmetry (LDF) were also performed (n = 5), which confirmed reduction in CBF following injury. A secondary focus of the study was to investigate the effectiveness of hypertonic saline as a neuroprotective agent, inhibiting the development of complications after brain injury in a subgroup of injured mice (n = 5), further demonstrating the ability of DW-LSI to monitor the effects upon brain dynamics of drug treatment. Overall, our findings further support the use of DW-LSI as a noninvasive, cost-effective tool to assess changes in hemodynamics under a variety of pathological conditions, suggesting its potential contribution to the biomedical field. Subarachnoid hemorrhage (SAH) is often accompanied by cerebral vasospasm (CVS), which is the phenomenon of narrowing of large cerebral arteries, and then can produce delayed ischemic neurological deficit (DIND) such as lateralized sensory dysfunction. CVS was regarded as a major contributor to DIND in patients with SAH. However, therapy for preventing vasospasm after SAH to improve the outcomes may not work all the time. It is important to find answers to the relationship between CVS and DIND after SAH. How local cerebral blood flow (CBF) is regulated during functional activation after SAH still remains poorly understood, whereas, the regulation of CBF may play an important role in weakening the impact of CVS on cortex function. Therefore, it is worthwhile to evaluate the functional response of CBF in the activated cortex in an SAH animal model. Most evaluation of the effect of SAH is presently carried out by neurological behavioral scales. The functional imaging of cortical activation during sensory stimulationmay help to reflect the function of the somatosensory cortexmore locally than the behavioral scales do. We investigated the functional response of CBF in the somatosensory cortex induced by an electrical stimulation to contralateral forepaw via laser speckle imaging in a rat SAH model. Nineteen Sprague-Dawley rats from two groups (control group, n = 10 and SAH group, n = 9) were studied. SAH was induced in rats by double injection of autologous blood into the cisterna magna after CSF aspiration. The same surgical procedure was applied in the control group without CSF aspiration or blood injection. Significant CVS was found in the SAH group. Meanwhile, we observed a delayed peak of CBF response in rats with SAH compared with those in the control group, whereas no significant difference was found inmagnitude, duration, and areas under curve of relative CBF changes between the two groups. The results suggest that the regulation function of local CBF during functional activation induced by somatosensory stimulation might not be seriously impaired in the somatosensory cortex of rats with SAH. Quantitative assessment of wound tissue ischemia, perfusion, and inflammation provides critical information for appropriate detection, staging, and treatment of chronic wounds. However, few methods are available for simultaneous assessment of these tissue parameters in a noninvasive and quantitative fashion. We integrated hyperspectral, laser speckle, and thermographic imaging modalities in a single-experimental setup for multimodal assessment of tissue oxygenation, perfusion, and inflammation characteristics. Algorithms were developed for appropriate coregistration between wound images acquired by different imaging modalities at different times. The multimodal wound imaging system was validated in an occlusion experiment, where oxygenation and perfusion maps of a healthy subject's upper extremity were continuously monitored during a postocclusive reactive hyperemia procedure and compared with standard measurements. The system was also tested in a clinical trial where a wound of three millimeters in diameter was introduced on a healthy subject's lower extremity and the healing process was continuously monitored. Background/purpose Optical and parametric skin imaging methods which can efficiently identify invisible sub-skin features or subtle changes in skin layers are very important for accurate optical skin modelling. In this study, a hybrid method is introduced that helps develop a parametric optical skin model by utilizing interdisciplinary techniques including light back-scatter analysis, laser speckle imaging, image-texture analysis and Bayesian inference methods. The model aims to detect subtle skin changes and hence very early signs of skin abnormalities/diseases. Methods Light back-scatter and laser speckle image textural analysis are applied onto the normal and abnormal skin regions ( lesions) to generate set of attributes/parameters. These are then optimized by Bayesian inference method in order to build an optimized parametric model. Results The attributes selected by Bayesian inference method in the optimization stage were used to build an optimized model and then successfully verified. It was clearly proven that Bayesian inference based optimization process yields good results to build an optimized skin model. Conclusion The outcome of this study clearly shows the applicability of this hybrid method in the analysis of skin features and is therefore expected to lead development of non-invasive and low-cost instrument for early detection of skin changes. Previously published data demonstrate that the temporal processing algorithm for laser speckle contrast imaging (LSCI) can improve the visibility of deep blood vessels and is less susceptible to static speckle artifacts when compared with the spatial algorithm. To the best of our knowledge, the extent to which the temporal algorithm can accurately predict the speckle contrast associated with flow in deep blood vessels has not been quantified. Here, we employed two phantom systems and imaging setups (epi-illumination and transillumination) to study the contrast predicted by the spatial and temporal algorithms in subsurface capillary tubes as a function of the camera exposure time and the actual flow speed. Our data with both imaging setups suggest that the contrast predicted by the temporal algorithm, and therefore the relative flow speed, is nearly independent of the degree of static optical scattering that contributes to the overall measured speckle pattern. Collectively, these results strongly suggest the potential of temporal LSCI at a single-exposure time to assess accurately the changes in blood flow even in the presence of substantial static optical scattering. Renal perfusion signals contain dynamics arising from the renal autoregulation feedback mechanisms as the contraction and dilation of vessels alter flow patterns. We can capture the time-varying dynamics at points across the renal surface using laser speckle imaging. We segment an imaged area of the renal cortex into clusters with phase synchronized dynamics. Our approach first uses phase coherence with a surrogate data derived threshold to identify synchronized pixel pairs. Non-negative matrix factorization is then applied to segment phase coherence estimates into phase synchronized regions. The method is applied to laser speckle imaging of the renal cortex of anaesthetized rats to identify regions on the renal surface with phase synchronized myogenic activity. In three out of six animals imaged after bolus infusion of N^\omega-nitro-l-arginine methyl ester (NAM), the renal surfaces are segmented into clusters with high phase coherence. No more than two clusters were identified during control period for any animal. In the remaining three animals, a strong myogenic signal could not be detected in surface perfusion during control or NAM. Separation of arteries and veins in the cerebral cortex is of significant importance in the studies of cortical hemodynamics, such as the changes of cerebral blood flow, perfusion or oxygen concentration in arteries and veins under different pathological and physiological conditions. Yet the cerebral vessel segmentation and vessel-type separation are challenging due to the complexity of cortical vessel characteristics and low spatial signal-to-noise ratio. In this work, we presented an effective full-field method to differentiate arteries and veins in cerebral cortex using dual-modal optical imaging technology including laser speckle imaging (LSI) and optical intrinsic signals (OIS) imaging. The raw contrast images were acquired by LSI and processed with enhanced laser speckle contrast analysis (eLASCA) algorithm. The vascular pattern was extracted and segmented using region growing algorithm from the eLASCA-based LSI. Meanwhile, OIS images were acquired alternatively with 630 and 870 nm to obtain an oxyhemoglobin concentration map over cerebral cortex. Then the separation of arteries and veins was accomplished by Otsu threshold segmentation algorithm based on the OIS information and segmentation of LSI. Finally, the segmentation and separation performances were assessed using area overlap measure (AOM). The segmentation and separation of cerebral vessels in cortical optical imaging have great potential applications in full-field cerebral hemodynamics monitoring and pathological study of cerebral vascular diseases, as well as in clinical intraoperative monitoring. While the laser speckle imaging (LSI) is a powerful tool for multiple biomedical applications, such as monitoring of the blood flow, in many cases it can provide additional information when combined with spatio-temporal rhythm analysis. We demonstrate the application of Graphics Processing Units (GPU)-based rhythm analysis for the post processing of LSI data, discuss the relevant structure of GPU-based computations, test the proposed technique on surrogate 3D data, and apply this approach to kidney blood flow autoregulation. Experiments with surrogate data demonstrate the ability of the method to extract information about oscillation patterns from noisy data, as well as to detect the moving source of the rhythm. The analysis of kidney data allow us to detect and to localize the dynamics arising from autoregulation processes at the level of individual nephrons (tubuloglomerular feedback (TGF) rhythm), as well as to distinguish between the TGF-active and the TGF-silent zones. The occlusion effect of vascular targeted photodynamic therapy (V-PDT) for malformed vessels in port wine stains (PWS) often last for some time after the treatment. A relatively longer period after V-PDT is needed to accurately assess the final response of PWS microcirculation to the treatment. In this study, we intended to use laser speckle imaging (LSI) to assess the tissue perfusion changes of PWS at follow-up after V-PDT and preliminarily analyze the relationship between perfusion change and color bleaching. Seventeen patients with 40 PWS lesions were scanned by LSI before and 3-6 months after they received V-PDT. The speckle flow indices of PWS lesions and normal skin before and at follow-up after V-PDT were recorded. We also performed analyses on the correlation between perfusion changes and color bleaching. Before V-PDT, the 40 PWS lesions showed higher perfusion than the normal skin (1,421 ± 463 and 1,115 ± 386 perfusion unit (PU), respectively, P < 0.01). The PWS lesions scanned at follow-up showed decreased perfusion level compared to the preoperative values (1,282 ± 460 and 1,421 ± 463 PU, respectively, P < 0.01). After V-PDT, the perfusion change rates coincide well with the color bleaching rates (correlation coefficient, 0.73). In conclusion, the LSI system is capable of imaging PWS perfusion precisely, and it has shown promising results in assessing the changes of tissue perfusion of V-PDT for PWS, with objective and quantitative data, real-time images, and a shorter detection time. Objective The aims of this clinical study were to determine differences in plaque accumulation and to compare the effects of reinforced composite resin and titanium on peri-implant soft-tissue and residual-ridge inflammation. Material and methods A total of 19 subjects were enrolled in this clinical trial; 10 jaws had implant-supported fixed prostheses with composite resin mucosal surfaces, 11 jaws had titanium prostheses fabricated by computer-aided design/computer-aided manufacture ( CAD/ CAM), and 6 jaws had acrylic resin prostheses. Plaque area indexes ( PAIs) were calculated on the mucosal surfaces of prostheses, and blood flow in the mucosa was captured with two-dimensional laser speckle imaging to evaluate residual-ridge inflammation. Subjects were educated about oral hygiene and reevaluated after 3 months. Results The PAI was significantly lower on titanium mucosal surfaces than on reinforced composite resin surfaces at the initial and second measurements (initial, P = 0.0052; second, P = 0.0044). Self-curing acrylic resin surfaces did not show any significant difference when compared with reinforced resin or titanium. Blood flow was significantly lower in mucosa contacting titanium surfaces than in mucosa contacting reinforced composite resin surfaces at the initial measurement ( P = 0.0330). Although subjects were instructed about plaque control after the initial measurement, PAIs indicated that the difference between the two materials could not be overcome. Conclusion In terms of oral hygiene and mucosal inflammation, titanium was superior to reinforced composite resin in implant-supported fixed prostheses for edentulous subjects, and the short-term use of acrylic resin was superior to the use of reinforced composite resin. The mouse ischemic hindlimb model is used widely for studying collateral artery growth (i.e., arteriogenesis) in response to increased shear stress. Nonetheless, precise measurements of regional shear stress changes along individual collateral arteries are lacking. Our goal is to develop and verify trans-illumination laser speckle flowmetry (LSF) for this purpose. Studies of defibrinated bovine blood flow through tubes embedded in tissue-mimicking phantoms indicate that trans-illumination LSF better maintains sensitivity with an increasing tissue depth when compared to epi-illumination, with an ~50% reduction in the exponential decay of the speckle velocity signal. Applying trans-illuminated LSF to the gracilis muscle collateral artery network in vivo yields both improved sensitivity and reduced noise when compared to epi-illumination. Trans-illuminated LSF images reveal regional differences in collateral artery blood velocity after femoral artery ligation and are used to measure an ~2-fold increase in the shear stress at the entrance regions to the muscle. We believe these represent the first direct measurements of regional shear stress changes in individual mouse collateral arteries. The ability to capture deeper vascular signals using a trans-illumination configuration for LSF may expand the current applications for LSF, which could have bearing on determining how shear stress magnitude and direction regulate arteriogenesis. Stroke is a worldwidemedical emergency and an important issue in stroke research is looking for the early pathophysiological markers which can predict the severity of brain injury. Decreased cerebral blood flow (CBF) has been serving as the most important indicator of ischemic stroke. Particular attention is paid to study the spatio-temporal CBF changes immediately after the onset of stroke in a rat intraluminal filament middle cerebral artery occlusion (MCAO) model and investigation of its correlation with brain infarct volume after 24 h.We implement an on-line laser speckle imaging (LSI) system, which could provide real time high spatio-temporal resolution CBF information before, during, and immediately after the rat MCAO surgery. We found a significant correlation between the affected area with 50% CBF reduction (CBF50) at the first minute after occlusion with the infarct volume. To the best of our knowledge, this is the earliest CBF marker for infarct volume prediction. Based on such a CBF-infarct volume correlation, LSI may be used as a real time guidance for improving the consistency of intra-luminal filament MCAO model since the depth of filament insertion could be adjusted promptly and those unsuccessful models could be excluded in the earliest stage. Laser speckle perfusion imaging (LSPI) has become an increasingly popular technique for monitoring vascular perfusion over a tissue surface. However, few studies have utilized the full range of spatial and temporal information generated by LSPI to monitor spatial properties of physiologically relevant dynamics. In this study, we extend the use of LSPI to analyze renal perfusion dynamics over a spatial surface of ~5 x 7 mm of renal cortex. We identify frequencies related to five physiological systems that induce temporal changes in renal vascular perfusion (cardiac flow pulse, respiratory-induced oscillations, baroreflex components, the myogenic response, and tubuloglomerular feedback) across the imaged surface and compare the results with those obtained from renal blood flow measurements. We find that dynamics supplied from global sources (cardiac, respiration, and baroreflex) present with the same frequency at all locations across the imaged surface, but the local renal autoregulation dynamics can be heterogeneous in their distribution across the surface. Moreover, transfer function analysis with forced blood pressure as the input yields the same information with laser speckle imaging or renal blood flow as the output during control, intrarenal infusion of Nω-nitro-Larginine methyl ester to enhance renal autoregulation, and intrarenal infusion of the rho-kinase inhibitor Y-27632 to inhibit vasomotion. We conclude that LSPI measurements can be used to analyze local as well as global renal perfusion dynamics and to study the properties of physiological systems across the renal cortex. Objective: The aim of this study was to investigate the feasibility to measure microcirculatory blood flow changes in patients with venous ulcers, by using the laser speckle imaging. Methods: Nine patients with a leg ulcer were measured with the laser speckle imager in a sitting position before and after mimicking venous hypertension, with the legs raised, by applying a blood pressure cuff and inflating it to 60 mmHg. Results: The results were inconclusive, due to the fact that a lot of practical problems interfered with the measurements, e.g. movement artefacts and inadequate wound visualisation. Conclusion: The additional value of laser speckle imaging in daily practice for wound care still remains an important question for further research. A reliable, reproducible microcirculation measurement in venous ulcers might predict venous ulcer healing and recurrence and therefore would be a valuable diagnostic tool in daily practice. Laser speckle contrast imaging (LSCI) is a noninvasive full-field optical imaging technique that gives a 2-D microcirculatory blood flow map of tissue. Due to novelty of commercial laser speckle contrast imagers, image processing of LSCI data is new. By opposition, the numerous signal processing works of laser Doppler flowmetry (LDF) data—that give a 1-D view of microvascular blood flow—have led to interesting physiological information. Recently, analysis of multiscale entropy (MSE) of LDF signals has been proposed. A nonmonotonic evolution of MSE with two distinctive scales—probably dominated by the cardiac activity—has been reported. We herein analyze MSE of LSCI data. We compare LSCI results with the ones of LDF signals obtained during the same experiment. We show that when time evolution of LSCI single pixels is studied, MSE presents a monotonic decreasing pattern, similar to the one of Gaussian white noises. By opposition, when the mean of LSCI pixel values is computed in a region of interest (ROI) and followed with time, MSE pattern becomes close to the one of LDF data, for ROI large enough. Laser speckle contrast imaging (LSCI) is a high-resolution and high contrast optical imaging technique often used to characterize hemodynamic changes in short-term physiological experiments. In this study, we demonstrate the utility of LSCI for characterizing microvascular remodeling and hemodynamic changes during wound healing angiogenesis in vivo. A 2 mm diameter hole was made in the mouse ear and the periphery of the wound imaged in vivo using LSCI over 12 days. We were able to visualize and quantify the vascular and perfusion changes that accompanied wound healing in the microenvironment proximal to the wound, and validated these changes with histology. We found that consistent with the stages of wound healing, microvessel density increased during the initial inflammatory phase (i.e., day 0-3), stayed elevated through the tissue formation phase (i.e., until day 7) and returned to baseline during the tissue remodeling phase (i.e., by day 12). Concomitant 'wide area mapping' of blood flow revealed that tissue perfusion in the wound periphery initially decreased, gradually increased from day 3-7, and subsided as healing completed. Interestingly, some regions exhibited a reestablishment of tissue perfusion approximately 6 days earlier than the ~18 days usually reported for the long term remodeling phase. The results from this study demonstrate that LSCI is an ideal platform for elucidating in vivo changes in microvascular hemodynamics and angiogenesis, and has the potential to offer invaluable insights in a range of disease models involving abnormal hemodynamics, such as diabetes and tumors. During the last three decades, several laser systems, ancillary technologies, and treatment modalities have been developed for the treatment of port wine stains (PWSs). However, approximately half of the PWS patient population responds suboptimally to laser treatment. Consequently, novel treatment modalities and therapeutic techniques/strategies are required to improve PWS treatment efficacy. This overview therefore focuses on three distinct experimental approaches for the optimization of PWS laser treatment. The approaches are addressed from the perspective of mechanical engineering (the use of local hypobaric pressure to induce vasodilation in the laser-irradiated dermal microcirculation), optical engineering (laser-speckle imaging of post-treatment flow in laser-treated PWS skin), and biochemical engineering (light- and heat-activatable liposomal drug delivery systems to enhance the extent of post-irradiation vascular occlusion).
