The adaxial leaf surface of butterworts (Pinguicula L.) presents specialized structures for carnivory, such as trichomes and sessile glands. The micromorphology of abaxial leaf surfaces has rarely been investigated; therefore, this study aimed to compare the micromorphology of adaxial and abaxial surfaces through electron scanning microscopy (SEM) and light microscopy (LM). We selected four species occurring in the Eastern Alps. All the species showed the same pattern: trichomes and sessile glands occurring only on the adaxial surface; stomata occurring on both surfaces with significant differences in density and size; and four-cells glands occurring only on the abaxial surface. The Euganean Thermal District, situated in North-East Italy, is one of Europe's largest and oldest thermal centres. The topical application of its therapeutic thermal muds is recognised by the Italian Health System as a beneficial treatment for patients suffering from arthro-rheumatic diseases. Polysaccharides produced by the mud microbiota have been recently identified as anti-inflammatory bioactive molecules. In this paper we analysed the efficacy of Microbial-Polysaccharides (M-PS) derived from mature muds obtained at different maturation temperatures, both within and outside the codified traditional mud maturation range. M-PSs were extracted from six mature muds produced by five spas of the Euganean Thermal District and investigated for their chemical properties, monosaccharide composition and in vivo anti-inflammatory potential, using the zebrafish model organism. Additionally, mature muds were characterized for their microbiota composition using Next-Generation Sequencing. The results showed that all M-PSs exhibit similar anti-inflammatory potential, referable to their comparable chemical composition. This consistency was observed despite changes in cyanobacteria populations, suggesting a possible role of the entire microbial community in shaping the properties of these biomolecules. These findings highlight the importance of scientific research in untangling the origins of the therapeutic efficacy of Euganean Thermal muds in the treatment of chronic inflammatory conditions. Introduction: Cyanobacteria appeared in the anoxic Archean Earth, evolving for the first time oxygenic photosynthesis and deeply changing the atmosphere by introducing oxygen. Starting possibly from UV-protected environments, characterized by low visible and far-red enriched light spectra, cyanobacteria spread everywhere on Earth thanks to their adaptation capabilities in light harvesting. In the last decade, few cyanobacteria species which can acclimate to far-red light through Far-Red Light Photoacclimation (FaRLiP) have been isolated. FaRLiP cyanobacteria were thus proposed as model organisms to study the origin of oxygenic photosynthesis as well as its possible functionality around stars with high far-red emission, the M-dwarfs. These stars are astrobiological targets, as their longevity could sustain life evolution and they demonstrated to host rocky terrestrial-like exoplanets within their Habitable Zone. Methods: We studied the acclimation responses of the FaRLiP strain Chlorogloeopsis fritschii sp. PCC6912 and the non-FaRLiP strain Synechocystis sp. PCC6803 to the combination of three simulated light spectra (M-dwarf, solar and far-red) and two atmospheric compositions (oxic, anoxic). We first checked their growth, O2 production and pigment composition, then we studied their transcriptional responses by RNA sequencing under each combination of light spectrum and atmosphere conditions. Results and discussion: PCC6803 did not show relevant differences in gene expression when comparing the responses to M-dwarf and solar-simulated lights, while far-red caused a variation in the transcriptional level of many genes. PCC6912 showed, on the contrary, different transcriptional responses to each light condition and activated the FaRLiP response under the M-dwarf simulated light. Surprisingly, the anoxic atmosphere did not impact the transcriptional profile of the 2 strains significantly. Results show that both cyanobacteria seem inherently prepared for anoxia and to harvest the photons emitted by a simulated M-dwarf star, whether they are only visible (PCC6803) or also far-red photons (PCC6912). They also show that visible photons in the simulated M-dwarf are sufficient to keep a similar metabolism with respect to solar-simulated light. Oxygenic photosynthetic organisms (OPOs) are primary producers on Earth and generate surface and atmospheric biosignatures, making them ideal targets to search for life from remote on Earth-like exoplanets orbiting stars different from the Sun, such as M-dwarfs. These stars emit very low light in the visible and most light in the far-red, an issue for OPOs, which mostly utilize visible light to photosynthesize and grow. After successfully testing procaryotic OPOs (cyanobacteria) under a simulated M-dwarf star spectrum (M7, 365–850 nm) generated through a custom-made lamp, we tested several eukaryotic OPOs: microalgae (Dixoniella giordanoi, Microchloropsis gaditana, Chromera velia, Chlorella vulgaris), a non-vascular plant (Physcomitrium patens), and a vascular plant (Arabidopsis thaliana). We assessed their growth and photosynthetic efficiency under three light conditions: M7, solar (SOL) simulated spectra, and far-red light (FR). Microalgae grew similarly in SOL and M7, while the moss P. patens showed slower growth in M7 with respect to SOL. A. thaliana grew similarly in SOL and M7, showing traits typical of shade-avoidance syndrome. Overall, the synergistic effect of visible and far-red light, also known as the Emerson enhancing effect, could explain the growth in M7 for all organisms. These results lead to reconsidering the possibility and capability of the growth of OPOs and are promising for finding biosignatures on exoplanets orbiting the habitable zone of distant stars. Key message: Stem photosynthesis seems to play an adaptive role for woody plants that prosper in hot and dry ecosystems. Stem photosynthesis is thought to be involved in tree resistance/resilience to water shortage. Recent studies have focused on the coordination between stem photosynthesis and hydraulics, but the generality of association of stem photosynthetic efficiency with species-specific adaptation to drought is still unclear. We quantified bark and wood chlorophyll a fluorescence (in terms of Fv/Fm) in current-year, 1-year and 2-year-old stems of several woody species harvested in diverse habitats. We ranked species in terms of relative drought tolerance on the basis of their vulnerability to xylem embolism (P50), and compared stem photosynthetic efficiency of drought-tolerant vs drought-sensitive species. Fv/Fm values decreased with increasing stem age, and were generally higher for Angiosperms than Gymnosperms. Fv/Fm both at the bark and wood level was higher for drought-tolerant Angiosperms compared to drought-sensitive ones. Our results highlight the potential adaptive role of stem photosynthesis in drought-tolerant species, thriving under arid conditions likely leading to prolonged stomatal closure and halt of leaf-level carbon gain. Phormidium sp. ETS-05 is one of the target cyanobacteria species conferring anti-inflammatory properties to the therapeutic muds applied by spas of the Euganean Thermal District (Italy) to treat arthro-rheumatic pathologies. Beneficial mud is prepared by spas following a traditional method, called maturation, leading to the growth of a specific microbiota on natural raw clay irrigated by flowing thermal water at 37–47 °C for about two months. The effectiveness of the mud is related to heat, electrolytes and bioactive molecules synthesized by the microbiota. A clear role in the anti-inflammatory activity of muds has been demonstrated for the exopolysaccharides, EPS, produced by the entire microbiota and Phormidium sp. ETS-05. Considering the interest in this species, we assessed its optimal growth conditions to obtain the higher EPS production in relation to temperature, light spectra, and intensity and nitrogen availability. The production of biomass and pigments was also taken into account, as other high-value compounds can be obtained in parallel with EPS. We found that exposure to a temperature of 45 °C under white light at 100 µmol photons m−2 s−1 is optimal to reach the highest biomass (1.13 g L−1) and an average production of 75 mg gDW−1 phycocyanin and of 150 mg gDW−1 EPS for Phormidium sp. ETS-05 cultured in lab-scale photobioreactors for 9 days. Putative genes linked with EPS assembly and export have also been identified in its genome, some of which have been investigated for their expression levels, opening up the possibility of biotechnologically boost EPS production. Simple Summary: Spirulina (Arthrospira platensis) is regarded as a functional food due to its valuable nutritional profile and components with potential antioxidant, anti-inflammatory, and immune-modulatory properties. However, the use of Spirulina in companion animals is still largely unexplored. Additionally, the palatability of nutraceuticals can be a primary factor in determining compliance with the protocol when they are used as complementary feed. The aim of this study was thus to assess the palatability and owners' perception of the effects of increasing amounts of Spirulina tablets in client-owned dogs and cats for a 6-week-long trial. Overall, the animals well accepted the supplementation of Spirulina, with no significant side effect detected. Owners appeared willing to adopt dietary Spirulina supplementation in the future for their pets, especially owners of senior dogs. The nutraceutical supplementation of Spirulina (Arthrospira platensis) in dogs and cats has not yet been investigated. The aim of this study was to evaluate if the dietary supplementation of increasing amounts of Spirulina for 6 weeks is palatable to pets and to assess the owner's perception of such supplementation. The owners of the 60 dogs and 30 cats that participated in this study were instructed to daily provide Spirulina tablets starting with a daily amount of 0.4 g, 0.8 g, and 1.2 g for cats as well as small dogs, medium dogs, and large dogs, respectively, and allowing a dose escalation of 2× and 3× every 2 weeks. The daily amount (g/kg BW) of Spirulina ranged from 0.08 to 0.25 for cats, from 0.06 to 0.19 for small-sized dogs, from 0.05 to 0.15 for medium-sized dogs, and from 0.04 to 0.12 for large-sized dogs. Each owner completed a questionnaire at the time of recruitment and the end of each 2-week period. No significant effect on the fecal score, defecation frequency, vomiting, scratching, lacrimation, general health status, and behavioral attitudes was detected by the owners' reported evaluations. Most animals accepted Spirulina tablets either administrated alone or mixed with food in the bowl. Daily supplementation of Spirulina for 6 weeks in the amounts provided in this study is therefore palatable and well tolerated by dogs and cats. [ABSTRACT FROM AUTHOR]

Introduction: The search for life on distant exoplanets is expected to rely on atmospheric biosignatures detection, such as oxygen of biological origin. However, it is not demonstrated how much oxygenic photosynthesis, which on Earth depends on visible light, could work under spectral conditions simulating exoplanets orbiting the Habitable Zone of M-dwarf stars, which have low light emission in the visible and high light emission in the far-red/near-infrared. By utilizing cyanobacteria, the first organisms to evolve oxygenic photosynthesis on our planet, and a starlight simulator capable of accurately reproducing the emission spectrum of an M-dwarf in the range 350–900 nm, we could answer this question. Methods: We performed experiments with the cyanobacterium Chlorogloeopsis fritschii PCC6912, capable of Far-Red Light Photoacclimation (FaRLiP), which allows the strain to harvest far-red in addition to visible light for photosynthesis, and Synechocystis sp. PCC6803, a species unable to perform this photoacclimation, comparing their responses when exposed to three simulated light spectra: M-dwarf, solar and far-red. We analysed growth and photosynthetic acclimation features in terms of pigment composition and photosystems organization. Finally, we determined the oxygen production of the strains directly exposed to the different spectra. Results: Both cyanobacteria were shown to grow and photosynthesize similarly under M-dwarf and solar light conditions: Synechocystis sp. by utilizing the few photons in the visible, C. fritschii by harvesting both visible and far-red light, activating the FaRLiP response. Discussion: Our results experimentally show that an M-dwarf light spectrum could support a biological oxygen production similar to that in solar light at the tested light intensities, suggesting the possibility to discover such atmospheric biosignatures on those exoplanets if other boundary conditions are met. Eukaryotic algae are photosynthetic organisms capable of exploiting sunlight to fix carbon dioxide into biomass with highly variable genetic and metabolic features. Information on algae metabolism from different species is inhomogeneous and, while green algae are, in general, more characterized, information on red algae is relatively scarce despite their relevant position in eukaryotic algae diversity. Within red algae, the best‐known species are extremophiles or multicellular, while information on mesophilic unicellular organisms is still lacunose. Here, we investigate the photosynthetic properties of a recently isolated seawater unicellular mesophilic red alga, Dixoniella giordanoi. Upon exposure to different illuminations, D. giordanoi shows the ability to acclimate, modulate chlorophyll content, and re‐organize thylakoid membranes. Phycobilisome content is also largely regulated, leading to almost complete disassembly of this antenna system in cells grown under intense illumination. Despite the absence of a light‐induced xanthophyll cycle, cells accumulate zeaxanthin upon prolonged exposure to strong light, likely contributing to photoprotection. D. giordanoi cells show the ability to perform cyclic electron transport that is enhanced under strong illumination, likely contributing to the protection of Photosystem I from over‐reduction and enabling cells to survive PSII photoinhibition without negative impact on growth. The therapeutic muds of the Euganean Thermal District, the oldest and largest thermal site in Europe, are recognized by the Italian Health System as an excellent treatment for osteoarticular diseases of inflammatory origin. The beneficial effects that have been demonstrated for these muds are due not only to heat and electrolytes, but also to bioactive compounds released from the microbial community, mainly composed of cyanobacteria, living in the thermal mud surface. We isolated and characterized a new coil-shaped filamentous cyanobacterium through a polyphasic approach. Molecular, morphological and biochemical data supported the description of the microorganism as a new species, Thermospirulina andreolii, of the new genus Thermospirulina, which includes other Spirulina-like cyanobacteria living in different thermal springs around the world. Biochemical analyses highlighted the capability of the strain to synthesize high amounts of phycocyanin and exopolysaccharides, with possible antioxidant and anti-inflammatory activities, suggesting its potential for contributing to the healing properties of the Euganean muds. WHIRLY2 is a single‐stranded DNA binding protein associated with mitochondrial nucleoids. In the why 2‐1 mutant of Arabidopsis thaliana, a major proportion of leaf mitochondria has an aberrant structure characterized by disorganized nucleoids, reduced abundance of cristae, and a low matrix density despite the fact that the macroscopic phenotype during vegetative growth is not different from wild type. These features coincide with an impairment of the functionality and dynamics of mitochondria that have been characterized in detail in wild‐type and why 2‐1 mutant cell cultures. In contrast to the development of the vegetative parts, seed germination is compromised in the why 2‐1 mutant. In line with that, the expression level of why 2 in seeds of wild‐type plants is higher than that of why 3, whereas in adult plant no difference is found. Intriguingly, in early stages of shoots development of the why 2‐1 mutant, although not in seeds, the expression level of why 3 is enhanced. These results suggest that WHIRLY3 is a potential candidate to compensate for the lack of WHIRLY2 in the why 2‐1 mutant. Such compensation is possible only if the two proteins are localized in the same organelle. Indeed, in organello protein transport experiments using intact mitochondria and chloroplasts revealed that WHIRLY3 can be dually targeted into both, chloroplasts and mitochondria. Together, these data indicate that the alterations of mitochondria nucleoids are tightly linked to alterations of mitochondria morphology and functionality. This is even more evident in those phases of plant life when mitochondrial activity is particularly high, such as seed germination. Moreover, our results indicate that the differential expression of why 2 and why 3 predetermines the functional replacement of WHIRLY2 by WHIRLY3, which is restricted though to the vegetative parts of the plant. Oxygenic photosynthetic microorganisms are a focal point of research in the context of human space exploration. As part of the bioregenerative life-support systems, they could have a key role in the production of breathable O2, edible biomasses and in the regeneration of CO2 rich-atmospheres and wastewaters produced by astronauts. The test of the organism's response to simulated physico-chemical parameters of planetary bodies could also provide important information about their habitability potential. It is believed that the success of future planetary and space missions will require innovative technologies, developed on the base of preliminary experiments in custom-made laboratory facilities. In this context, simulation chambers will play a pivotal role by allowing the growth of the microorganisms under controlled conditions and the evaluation in real-time of their biomass productivity and impact on atmosphere composition. We here present a system capable of addressing these requirements with high replicability and low costs. The setup is composed by three main parts: 1) a Star Light Simulator, able to generate different light intensities and spectra, including those of non-solar stars; 2) an Atmosphere Simulator Chamber where cultures of photosynthetic microorganisms can be exposed to different gas compositions; 3) a reflectivity detection system to measure from remote the Normalized Difference Vegetation Indexes (NDVI). Such a setup allows us to monitor photosynthetic microorganism's growth and gas exchange performances under selected conditions of light quality and intensity, temperature, pressure, and atmospheres simulating non-terrestrial environments. All parameters are detected by remote sensing techniques, thus without interfering with the experiments and altering the environmental conditions set. We validated the setup by growing cyanobacteria liquid cultures under different light intensities of solar illumination, collecting data on their growth rate, photosynthetic activity, and gas exchange capacity. We utilized the reflectivity detection system to measure the reflection spectra of the growing cultures, obtaining their relative NDVI that was shown to correlate with optical density, chlorophyll content, and dry weight, demonstrating the potential application of this index as a proxy of growth. Photosynthetic cell suspension cultures are a useful experimental system to analyze a variety of physiological processes, bypassing the structural complexity of the plant organism in toto. Nevertheless, cell cultures containing functional chloroplasts are quite difficult to obtain, and this process is usually laborious and time-consuming. In this work a novel and rapid method to set up photosynthetic cell suspension cultures from the model plant Arabidopsis thaliana was developed. The direct germination of Arabidopsis seeds on a sucrose-containing agarized culture medium supplemented with 0.25 µg/ml 6-benzylaminopurine and 0.5 µg/ml 2,4-dichlorophenoxyacetic acid caused the straightforward formation of green calli at the level of seedling hypocotyls. The subsequent transfer of these calli in liquid culture medium containing the same concentrations of phytohormones and gradually decreasing sucrose levels allowed for the establishment of chloroplast-containing cell suspension cultures, containing functional chloroplasts, in a much faster way than previously described procedures. Pulse amplitude modulation analyses, measurements of oxygen evolution and electron transport rate, together with confocal and electron microscopy observations, confirmed the photosynthetic efficiency of these cell suspension cultures. Cyanobacterium aponinum is a thermotolerant cyanobacterium species isolated from different thermal springs and showing potential therapeutic application thanks to the synthesis and release of exopolysaccharides with anti-inflammatory and immunomodulating properties. This work investigates the effects of temperature and light on growth rate, biomass yield, and released polysaccharides (RPS) synthesis in C. aponinum cultures. The highest biomass productivity was obtained at 40 °C (about 92 mg L d), while the highest RPS synthesis rate (about 10 mg L d) was detected at 35 °C. At this temperature, the influence of light intensities, between 15 and 650 μmol photons m s, was also evaluated. C. aponinum showed the highest productivity in biomass (about 280 mg L d) and RPS (about 20 mg L d) at 500 μmol photons m s. Interestingly, this strain also had the ability to accumulate other high-value compounds such as phycocyanin, zeaxanthin, and β-carotene suggesting that C. aponinum can be used for the industrial production of multiple molecules of biotechnological interest. Nannochloropsis is an eukaryotic alga of the phylum Heterokonta, originating from a secondary endosymbiotic event. In this work, we investigated how the photosynthetic apparatus responds to growth in different light regimes in Nannochloropsis gaditana. We found that intense illumination induces the decrease of both photosystem I and II contents and their respective antenna sizes. Cells grown in high light showed a larger capacity for electron transport, with enhanced cyclic electron transport around photosystem I, contributing to photoprotection from excess illumination. Even when exposed to excess light intensities for several days, N. gaditana cells did not activate constitutive responses such as nonphotochemical quenching and the xanthophyll cycle. These photoprotection mechanisms in N. gaditana thus play a role in acclimation to fast changes in illumination within a time range of minutes, while regulation of the electron flow capacity represents a long-term response to prolonged exposure to excess light. The family Scenedesmaceae is a taxonomically complicated group due to its simple morphology, high phenotypic plasticity, and the presence of cryptic taxa. Over the years several taxonomic revisions, based on molecular data, affected the family. Here, we describe a new scenedesmacean species from Antarctica, Chodatodesmus australis, based on phylogenetic analyses of data from nuclear (ITS2 spacer, 18S rDNA), and plastid (rbcL, tufA) markers. Morphological (LM and SEM) and ultrastructural (TEM) observations, carried out both on the holotype of C. australis and on the generitype of Chodatodesmus, allow us to emend the original generic description of this genus. Our molecular and phylogenetic data also reveal the existence of a new monotypic genus, Flechtneria, inside the family Scenedesmaceae and lead to the taxonomic reassignment of some microalgal strains available in International Culture Collections to new taxa. Of the considered genomic regions, the tufA gene was the easiest to amplify and sequence and it showed the highest phylogenetic signal, even if the number of sequences already available for this marker in the public databases was considerably lower than for the other chosen loci. The rbcL gene also provided good phylogenetic signal, but its amplification and sequencing were generally more problematic. The nuclear markers gave lower phylogenetic signals, but the 18S rDNA allowed distinction at the genus level and the ITS2 spacer had the advantage that secondary structures could be considered in the analyses. The use of more than one molecular locus is suggested to obtain reliable results in the characterization of scenedesmacean strains. [ABSTRACT FROM AUTHOR]

Antarctic algae play a fundamental role in polar ecosystem thanks to their ability to grow in an extreme environment characterized by low temperatures and variable illumination. Here, for prolonged periods, irradiation is extremely low and algae must be able to harvest light as efficiently as possible. On the other side, at low temperatures even dim irradiances can saturate photosynthesis and drive to the formation of reactive oxygen species. Colonization of this extreme environment necessarily required the optimization of photosynthesis regulation mechanisms by algal organisms. In order to investigate these adaptations we analyzed the time course of physiological and morphological responses to different irradiances in Koliella antarctica, a green microalga isolated from Ross Sea (Antarctica). Koliella antarctica not only modulates cell morphology and composition of its photosynthetic apparatus on a long‐term acclimation, but also shows the ability of a very fast response to light fluctuations. Koliella antarctica controls the activity of two xanthophyll cycles. The first, involving lutein epoxide and lutein, may be important for the growth under very low irradiances. The second, involving conversion of violaxanthin to antheraxanthin and zeaxanthin, is relevant to induce a fast and particularly strong non‐photochemical quenching, when the alga is exposed to higher light intensities. Leaf senescence is the last stage of development of an organ and is aimed to its ordered disassembly and nutrient reallocation. Whereas chlorophyll gradually degrades during senescence in leaves, mitochondria need to maintain active to sustain the energy demands of senescing cells. Here we analysed the motility and morphology of mitochondria in different stages of senescence in leaves of grapevine (Vitis vinifera), by stably expressing a GFP (green fluorescent protein) reporter targeted to these organelles. Results show that mitochondria were less dynamic and markedly changed morphology during senescence, passing from the elongated, branched structures found in mature leaves to enlarged and sparse organelles in senescent leaves. Progression of senescence in leaves was not synchronous, since changes in mitochondria from stomata were delayed. Mitochondrial morphology was also analysed in grapevine cell cultures. Mitochondria from cells at the end of their growth curve resembled those from senescing leaves, suggesting that cell cultures might represent a useful model system for senescence. Additionally, senescence-associated mitochondrial changes were observed in plants treated with high concentrations of cytokinins. Cyanobacteria (phylum Cyanophyta/Cyanobacteria, class Cyanophyceae) are among the most widespread organisms and are able to adapt themselves to different extreme environments. These micro-organisms have an important ecological role, given their ability to perform oxygenic photosynthesis, and are employed in different fields based on their ability to produce several bioactive compounds. Their prokaryotic nature, the presence of many cryptic species, and the coexistence of different nomenclature systems make the taxonomic identification of cyanobacteria particularly difficult. Moreover, for several species, the original reference strains (holotypes) are lacking. Increasingly, authors are using a polyphasic approach to characterize cyanobacteria, while typification is a recent trend that is being used to solve the problem of missing holotypes in other micro-organisms. Here we focus on a filamentous cyanobacterium, isolated from the Euganean Thermal District (Padova, Italy) and temporarily named strain ETS-02, using a polyphasic approach that includes morphological, ultrastructural, biochemical (pigment and fatty acid content), physiological (nitrogen fixation), and genetic (16S rRNA, 16S-23S ITS, cpcB-IGS- cpcA, rpoC1, gyrB, rbcL, nifD loci) analyses. A potassium channel (SynK) of the cyanobacterium Synechocystis sp. PCC 6803, a photoheterotrophic model organism for the study of photosynthesis, has been recently identified and demonstrated to function as a potassium selective channel when expressed in a heterologous system and to be located predominantly to the thylakoid membrane in cyanobacteria. To study its physiological role, a SynK-less knockout mutant was generated and characterized. Fluorimetric experiments indicated that SynK-less cyanobacteria cannot build up a proton gradient as efficiently as WT organisms, suggesting that SynK might be involved in the regulation of the electric component of the proton motive force. Accordingly, measurements of flash-induced cytochrome fa6f turnover and respiration pointed to a reduced generation of ApH and to an altered linear electron transport in mutant cells. The lack of the channel did not cause an altered membrane organization, but decreased growth and modified the photosystem ll/photosystem I ratio at high light intensities because of enhanced photosensitivity. These data shed light on the function of a prokaryotic potassium channel and reports evidence, by means of a genetic approach, on the requirement of a thylakoid ion channel for optimal photosynthesis. In maize vivipary, the precocious germination of the seed while it is still attached to the ear is a reliable phenotype for the identification of mutants impaired in the biosynthesis or response to abscisic acid (ABA). Here we present the characterization of a new allele of vp10, a gene encoding for a cofactor (MoCo) required for the last step of ABA biosynthesis. The lesion in this gene leads to a reduction in the endogenous ABA level. Embryonic messenger RNAs of the ABA inducible genes glb1, lea3, and rab17 are barely detectable, although their level increases when stimulated by exogenous ABA administration. These findings confirm that the mutant can be ascribed to a defect in ABA biosynthesis. In the absence of water stress, mutant plants grow like wild-type siblings; however when mutant tissues are exposed to air they differ from non-mutant ones by showing a higher rate of water loss, of transpiration and of stomatal conductance. These events are restored to almost normal values by adding exogenous ABA. All these defects are ascribable to an impairment in the regulation of stomatal opening since, in contrast to wild-type, some of the mutant stomata exhibit partially or totally open rims. The defect in ABA biosynthesis is also associated with loss of regulation of the expression of rab17 and rab28, two genes expressed in vegetative tissues under abiotic stress. These genes are constitutively expressed in the mutant plant tissues independently of the water regime applied. In this paper we report a morphological, ultrastructural, biochemical and molecular (16S rRNA, 16S-23S ITS, rbcL and rpoC1 gene sequencing) survey on a very thin, non-heterocystous, filamentous cyanobacterium, isolated from mats covering several mud maturation tanks of the Euganean Thermal District, at temperatures ranging from 26 to 59°C. Denaturing gradient gel electrophoresis results, obtained using cyanobacterial primers targeting the 16S rRNA gene, confirmed that this cyanobacterium is one of the commonest taxa growing in the mud tanks. Comparison with Geitlerinema sp. PCC 8501 (=Phormidium laminosum Gomont ex Gomont strain OH-1-p Cl 1), a thin thermobiotic species isolated from hot springs of Oregon and morphologically similar to our isolate, led us to hypothesize that the Euganean and PCC 8501 strains are either very similar sister species or ecotypes of the same species in a yet to be defined clade, clearly distinct within the paraphyletic Leptolyngbya group. Elucidation of the structure-function relationship of a small number of prokaryotic ion channels characterized so far greatly contributed to our knowledge on basic mechanisms of ion conduction. We identified a new potassium channel (SynK) in the genome of the cyanobacterium Synechocystis sp. PCC6803, a photosynthetic model organism. SynK, when expressed in a K+-uptake-system deficient E.coli strain, was able to recover growth of these organisms. The protein functions as a potassium selective ion channel when expressed in Chinese Hamster Ovary cells. The location of SynK in cyanobacteria in both thylakoid and plasmamembranes was revealed by immunogold electron microscopy and Western blotting of isolated membrane fractions. SynK seems to be conserved during evolution, giving rise to a TPK (two-pore K+ channel) family member which is shown here to be located in the thylakoid membrane of Arabidopsis. Our work characterizes a novel cyanobacterial potassium channel and indicates the molecular nature of the first higher plant thylakoid cation channel, opening the way to functional studies. Free porphyrins and their magnesium complexes, including chlorophylls, are potent photo-sensitizers. Plants usually accumulate these compounds bound to proteins together with protective compounds like carotenoids. Besides their protective role, carotenoids can play a structural role in these complexes. To analyze the effect of impaired carotenogenesis on plastid membranes we applied to barley seedlings the bleaching herbicide 2-(4-chlorophenylthio)triethylamine (CPTA) as a specific inhibitor for the cyclization of lycopene. To avoid interference with photo-oxidation, the essential experiments were performed on seedlings grown in darkness. While the amount of total carotenoids decreased, we found accumulation of more δ-carotene than lycopene in darkness clearly showing that CPTA inhibits the lycopene β-cyclase more effectively than the lycopene ε-cyclase. The CPTA treatment resulted in accumulation of non-photoactive protochlorophyllide a; the amount of photoactive protochlorophyllide and NADPH:protochlorophyllide oxidoreductase remained constant. Further, the level of Mg protophorphyrin and its monomethyl ester increased to an extent similar to that obtained by application of 5-aminolevulinic acid (ALA). The perturbation of the ultrastructure of etioplast inner membranes, observed after CPTA-treatment, was not found after ALA-treatment; this excluded the accumulated tetrapyrroles as responsible for the perturbation. By contrast, the down-regulation of Lhcb and RbcS genes found after CPTA-treatment was compatible with the presumed role of Mg protophorphyrin as “plastid signal” for regulation of nuclear gene expression. To understand the function of ascorbic acid (ASC) in root development, the distribution of ASC, ASC oxidase, and glutathione (GSH) were investigated in cells and tissues of the root apex of Cucubita maxima. ASC was regularly distributed in the cytosol of almost all root cells, with the exception of quiescent centre (QC) cells. ASC also occurred at the surface of the nuclear membrane and correspondingly in the nucleoli. No ASC could be observed in vacuoles. ASC oxidase was detected by immunolocalization mainly in cell walls and vacuoles. This enzyme was particularly abundant in the QC and in differentiating vascular tissues and was absent in lateral root primordia. Administration of the ASC precursor L-galactono-γ-lactone markedly increased ASC content in all root cells, including the QC. Root treatment with the ASC oxidized product, dehydroascorbic acid (DHA), also increased ASC content, but caused ASC accumulation only in peripheral tissues, where DHA was apparently reduced at the expense of GSH. The effects of impaired carotenogenesis on plastid membrane organization, functionality and stability were studied in etiolated barley plants grown at 20 and 30°C. The plants were treated with norflurazon or amitrole, two herbicides affecting phytoene desaturation and lycopene cyclization, respectively. At 20°C, the amitrole-treated etioplasts, which accumulated lycopene in their inner membranes, exhibited disorganized prolamellar bodies, containing a prevalent form of non-phototransformable protochlorophyllide (Pchlide). They also showed a certain difficulty in reducing the phototransformable pigment to chlorophyllide when exposed to light, and were unable to reform the active ternary complex [protochlorophyllide-oxidoreductase (POR)-Pchlide-NADPH] when placed back in darkness. No ultrastructural alterations were found in norflurazon-treated etioplasts, with carotenogenesis inhibited at the phytoene desaturation step. In these latter organelles, Pchlide, whose forms were comparable with those of the control etioplasts, was photoreduced quickly after illumination and the ternary complex was reformed during a subsequent dark period. Thus, the impaired carotenogenesis leading to the accumulation of lycopene showed greater interference with the etioplast membrane arrangement and functionality than did the earlier interruption of the biosynthetic pathway at the phytoene level. This might be due to the different interactions of the distinct carotenoid precursors with other membrane components. However, in etioplasts of norflurazon-treated plants, a rise in growth temperature caused a partial demolition of prolamellar bodies, showing a lowered thermostability of the carotenoid-deficient membranes. This latter effect strengthens the concept that a correct and complete carotenogenesis pathway, leading to the synthesis of polar carotenoids (i.e. xanthophylls), is required for the maintenance of stable plastid membranes. The effects of norflurazon (NF) and amitrole (AM), two bleaching herbicides which inhibit carotenogenesis, were compared in leaves of 7-day-old barley (Hordeum vulgare L. cv Express) plants grown in damaging light. The herbicide effects were analysed with respect to chloroplast organization, photosynthetic functionality and nuclear photodependent expression of the Lhcb1 gene, which codes for the Lhcb1 light-harvesting chlorophyll a/b binding protein of photosystem II. Both herbicides caused dramatic photooxidation of organelles, which were photosynthetically unfunctional. Plastids of NF-treated plants lacked thylakoids and pigments. Plastids of AM-treated plants had some strikingly altered membranes and contained only very small quantities of chlorophylls. Despite the presence of severely photodamaged plastids, cells of AM-treated leaves contained high levels of Lhcb1 transcript. This transcript, on the contrary, was completely absent in the cells of NF-treated plants. These findings suggest that in order to block expression of nuclear genes coding for plastid-resident proteins, photodamage leading to the complete dismantling of thylakoids and to the total absence of any form of photosynthetic pigment is required. In a few years, space telescopes will investigate our Galaxy to detect evidence of life, mainly by observing rocky planets. In the last decade, the observation of exoplanet atmospheres and the theoretical works on biosignature gasses have experienced a considerable acceleration. The most attractive feature of the realm of exoplanets is that 40% of M dwarfs host super-Earths with a minimum mass between 1 and 30 Earth masses, orbital periods shorter than 50 days, and radii between those of the Earth and Neptune (1–3.8 R ⊕ ). Moreover, the recent finding of cyanobacteria able to use far-red (FR) light for oxygenic photosynthesis due to the synthesis of chlorophylls d and f, extending in vivo light absorption up to 750 nm, suggests the possibility of exotic photosynthesis in planets around M dwarfs. Using innovative laboratory instrumentation, we exposed different cyanobacteria to an M dwarf star simulated irradiation, comparing their responses to those under solar and FR simulated lights. As expected, in FR light, only the cyanobacteria able to synthesize chlorophyll d and f could grow. Surprisingly, all strains, both able or unable to use FR light, grew and photosynthesized under the M dwarf generated spectrum in a similar way to the solar light and much more efficiently than under the FR one. Our findings highlight the importance of simulating both the visible and FR light components of an M dwarf spectrum to correctly evaluate the photosynthetic performances of oxygenic organisms exposed under such an exotic light condition. The Euganean Thermal District has been known since Roman times for the therapeutic properties of peloids, obtained from natural clays that have undergone a traditional maturation process. This leads to the growth of a green microbial biofilm with Cyanobacteria and the target species Phormidium sp. ETS-05 as fundamental components for their ability to synthetize anti-inflammatory molecules. Currently, in-depth studies on the microbiota colonizing Euganean peloids, as in general on peloids utilized worldwide, are missing. This is the first characterization of the microbial community of Euganean thermal muds, also investigating the effects of environmental factors on its composition. We analysed 53 muds from 29 sites (Spas) using a polyphasic approach, finding a stable microbiota peculiar to the area. Differences among mud samples mainly depended on two parameters: water temperature and shading of mud maturation plants. In the range 37–47 °C and in the case of irradiance attenuation due to the presence of protective roofs, a statistically significant higher mud Chl a content was detected. Moreover, in these conditions, a characteristic microbial and Cyanobacteria population composition dominated by Phormidium sp. ETS-05 was observed. The Euganean Thermal District (Italy) represents the oldest and largest thermal center in Europe, and its therapeutic mud is considered a unique product whose beneficial effects have been documented since Ancient Roman times. Mud properties depend on the heat and electrolytes of the thermal water, as well as on the bioactive molecules produced by its biotic component, mainly represented by cyanobacteria. The investigation of the healing effects of compounds produced by the Euganean cyanobacteria represents an important goal for scientific validation of Euganean mud therapies and for the discovering of new health beneficial biomolecules. In this work, we evaluated the therapeutic potential of exopolysaccharides (EPS) produced by Phormidium sp. ETS05, the most abundant cyanobacterium of the Euganean mud. Specifically, Phormidium EPS resulted in exerting anti-inflammatory and pro-resolution activities in chemical and injury-induced zebrafish inflammation models as demonstrated using specific transgenic zebrafish lines and morphometric and expression analyses. Moreover, in vivo and in vitro tests showed no toxicity at all for the EPS concentrations tested. The results suggest that these EPS, with their combined anti-inflammatory and pro-resolution activities, could be one of the most important therapeutic molecules present in the Euganean mud and confirm the potential of these treatments for chronic inflammatory disease recovery.
