Psittacanthus calyculatus parasitizes mesquite trees through a specialized structure called a haustorium, which, in the intrusive process, can cause cellular damage in the host tree and release DAMPs, such as ATP, sugars, RNA, and DNA. These are highly conserved molecules that primarily function as signals that trigger and activate the defense responses. In the present study, we generate extracellular DNA (exDNA) from mesquite (P. laevigata) tree leaves (self-exDNA) and P. calyculatus (non-self exDNA) mistletoe as DAMP sources to examine mesquite trees' capacity to identify specific self or non-self exDNA. We determined that mesquite trees perceive self- and non-self exDNA with the synthesis of O2•−, H2O2, flavonoids, ROS-enzymes system, MAPKs activation, spatial concentrations of JA, SA, ABA, and CKs, and auxins. Our data indicate that self and non-self exDNA application differs in oxidative burst, JA signaling, MAPK gene expression, and scavenger systems. Psittacanthus calyculatus is a hemiparasite mistletoe that represents an ecological problem due to the impacts caused to various tree species of ecological and commercial interest. Although the life cycle for the Psittacanthus genus is well established in the literature, the development stages and molecular mechanism implicated in P. calyculatus host infection are poorly understood. In this study, we used a manageable infestation of P. laevigata with P. calyculatus to clearly trace the infection, which allowed us to describe five phenological infective stages of mistletoe on host tree branches: mature seed (T1), holdfast formation (T2), haustorium activation (T3), haustorium penetration (T4), and haustorium connection (T5) with the host tree. Proteomic analyses revealed proteins with a different accumulation and cellular processes in infective stages. Activities of the cell wall-degrading enzymes cellulase and β-1,4-glucosidase were primarily active in haustorium development (T3), while xylanase, endo-glucanase, and peptidase were highly active in the haustorium penetration (T4) and xylem connection (T5). Patterns of auxins and cytokinin showed spatial concentrations in infective stages and moreover were involved in haustorium development. These results are the first evidence of proteins, cell wall-degrading enzymes, and phytohormones that are involved in early infection for the Psittacanthus genus, and thus represent a general infection mechanism for other mistletoe species. N-acyl-L-homoserine lactones (AHLs) are involved in cell-to-cell communication in Gram-negative bacteria through a process termed quorum-sensing (QS). In this report, we evaluated the response of Arabidopsis thaliana primary roots to abscisic acid (ABA) in wild-type (WT) and decanamide resistant root 1 (drr1) mutant, previously reported to be resistant to N-decanoyl-L-homoserine lactone (C10-HL). When compared to WT seedlings, drr1 mutants were hypersensitive to ABA and had primary roots shorter, which correlated with lower cell division in meristems, a higher concentration of endogenous ABA, and a greater expression of ABI5 gene; and this shortened primary root phenotype was reversed in drr1abi5 double mutants. An analysis of expression of ABSCISIC ACID INSENSITIVE 4 (ABI4) showed an ABA-inducible pattern in primary root tips, which was further increased in drr1 mutant seedlings. Comparison of seed germination in WT, drr1, abi5, and drr1abi5 lines showed higher germination percentages in the following order under control condition: abi5 > drr1abi5 > WT > drr1, while abi5 and drr1abi5 germinated faster and drr1 germinated slower with respect to WT under ABA condition. Taken together, our results suggest that DRR1 is a negative regulator of ABA signaling probably acting upstream of the transcription factors ABI4 and ABI5, which influence ABA responsiveness in primary roots and seed germination. ALTERED MERISTEM PROGRAM 1 (AMP1) encodes a putative glutamate-carboxypeptidase important for plant growth and development. In this study, by comparing the growth of Arabidopsis wild-type, amp1–10 and amp1–13 mutants, and AMP1-GFP/OX2- and AMP1-GFP/OX7-overexpressing seedlings in vitro and in soil, we uncover the role of AMP1 in biomass accumulation in Arabidopsis. AMP1-overexpressing plants had longer primary roots and increased lateral root number and density than the WT, which correlated with improved root, shoot, and total biomass accumulation. AMP1-overexpressing seedlings had an enhanced rate of growth of primary roots, and accordingly, sucrose supplementation restored primary root growth and promoted lateral root formation in amp1 mutants, while reproductive development, fruit size, and seed content were also modified according to disruption or overexpression of AMP1. We further found that AMP1 plays an important role for stomatal development, guard cell functioning, and carbon assimilation. These data help explain the pleiotropic functions of AMP1 in both root and shoot system development, possibly acting in a sugar-dependent manner for regulation of root architecture, biomass accumulation, and seed production. Metallothioneins (MTs) constitute a heterogeneous family of ubiquitous metal ion-binding proteins. In plants, MTs participate in the regulation of cell growth and proliferation, protection against heavy metal stress, oxidative stress responses, and responses to pathogen attack. Despite their wide variety of functions, the role of MTs in symbiotic associations, specifically nodule-fabacean symbiosis, is poorly understood. Here, we analyzed the role of the PvMT1A gene in Phaseolus vulgaris-Rhizobium tropici symbiosis using bioinformatics and reverse genetics approaches. Using in silico analysis, we identified six genes encoding MTs in P. vulgaris, which were clustered into three of the four classes described in plants. PvMT1A transcript levels were significantly higher in roots inoculated with R. tropici at 7 and 30 days post inoculation (dpi) than in non-inoculated roots. Functional analysis showed that downregulating PvMT1A by RNA interference (RNAi) reduced the number of infection events at 7 and 10 dpi and the number of nodules at 14 and 21 dpi. In addition, nodule development was negatively affected in PvMT1A:RNAi transgenic roots, and these nodules displayed a reduced nitrogen fixation rate at 21 dpi. These results strongly suggest that PvMT1A plays an important role in the infection process and nodule development in P. vulgaris during rhizobial symbiosis. [ABSTRACT FROM AUTHOR]
