The eclipses seen in the radio emission of some pulsars can be invaluable to study the properties of the material from the companion stripped away by the pulsar. We present a study of six consecutive eclipses of PSR J0024–7204O in the globular cluster 47 Tucanae as seen by the MeerKAT radio telescope in the UHF (544–1088 MHz) band. A high scintillation state boosted the signal during one of the orbits and allowed a detailed study of the eclipse properties. We measure significant dispersion measure (DM) variations and detect strong scattering that seems to be the dominating mechanism of the eclipses at these frequencies. A complete drop in the linear polarization together with a small increase in the rotation measure suggests the presence of a magnetic field of |$\sim 2$|  mG. The study of multiple eclipses allowed us to measure difference in the lengths of the eclipses and DM differences of |$\sim 0.01$|  pc cm |$^{-3}$| in consecutive orbits. One orbit, in particular, shows a delay in recovery of the linear polarization and a visible delay in the arrival of the pulses caused by a stronger scattering event. Using MeerKAT , we have discovered three new millisecond pulsars (MSPs) in the bulge globular cluster M62: M62H, M62I, and M62J. All three are in binary systems, which means all ten known pulsars in the cluster are in binaries. M62H has a planetary-mass companion with a median mass M c, med ∼ 3 MJ and a mean density of ρ ∼ 11 g cm−3. M62I has an orbital period of 0.51 d and a M c, med ∼ 0.15 M⊙. Neither of these low-mass systems exhibit eclipses. M62J has only been detected in the two Ultra High Frequency band (816 MHz) observations with a flux density S 816 = 0.08 mJy. The non-detection in the L-band (1284 MHz) indicates it has a relatively steep spectrum (β < −3.1). We also present 23-yr-long timing solutions obtained using data from the Parkes 'Murriyang', Effelsberg , and MeerKAT telescopes for the six previously known pulsars. For all these pulsars, we measured the second spin-period derivatives and the rate of change of orbital period caused by the gravitational field of the cluster, and their proper motions. From these measurements, we conclude that the pulsars' maximum accelerations are consistent with the maximum cluster acceleration assuming a core-collapsed mass distribution. Studies of the eclipses of the redback M62B and the black widow M62E at four and two different frequency bands, respectively, reveal a frequency dependence with longer and asymmetric eclipses at lower frequencies. The presence of only binary MSPs in this cluster challenges models which suggest that the MSP population of core-collapsed clusters should be dominated by isolated MSPs. [ABSTRACT FROM AUTHOR]

We report the results of an observational campaign using the Effelsberg 100-m telescope of the pulsars J1746−2849, J1746−2850, J1746−2856, and J1745−2912 located in the central molecular zone (CMZ) close to the Galactic Centre in order to study rotation measure (RM) variations. We report for the first time the RM value of PSR J1746−2850 to be −12234 ± 181 rad m−2. This pulsar shows significant variations of RM of 300–400 rad m−2 over the course of months to years that suggest a strongly magnetized environment. The structure function analysis of the RM of PSR J1746−2850 revealed a steep power-law index of |$1.87_{-0.3}^{+0.4}$| comparable to the value expected for isotropic turbulence. This pulsar also showed large dispersion measure (DM) variation of ∼50 pc cm−3 in an event lasting a few months where the RM increased by ∼200 rad m−2. The large difference in RM between PSR J1746−2849 and PSR J1746−2850 despite the small angular separation reveals the presence of a magnetic field of at least 70 μG in the CMZ and can explain the lack of polarization in the radio images of the region. These results contribute to our understanding of the magnetic field in the CMZ and show similarities between the RM behaviours of these pulsars and some fast radio bursts. [ABSTRACT FROM AUTHOR]

The most massive globular cluster in our Galaxy, Omega Centauri, is an interesting target for pulsar searches, because of its multiple stellar populations and the intriguing possibility that it was once the nucleus of a galaxy that was absorbed into the Milky Way. The recent discoveries of pulsars in this globular cluster and their association with known X-ray sources was a hint that, given the large number of known X-ray sources, there is a much larger undiscovered pulsar population. We used the superior sensitivity of the MeerKAT radio telescope to search for pulsars in Omega Centauri. In this paper, we present some of the first results of this survey, including the discovery of 13 new pulsars; the total number of known pulsars in this cluster currently stands at 18. At least half of them are in binary systems and preliminary orbital constraints suggest that most of the binaries have light companions. Context. PSR J1910−5959A is a binary millisecond pulsar in a 0.837 day circular orbit around a helium white dwarf (HeWD) companion. The position of this pulsar is 6.3 arcmin (∼74 core radii) away from the optical centre of the globular cluster (GC) NGC 6752. Given the large offset, the association of the pulsar with the GC has been debated. Aims. We aim to obtain precise measurements of the masses of the stars in the system along with secular orbital parameters, which will help identify if the system belongs to the GC. Methods. We have made use of archival Parkes 64 m 'Murriyang' telescope data and carried out observations with the MeerKAT telescope with different backends and receivers over the last two decades. Pulse times of arrival were obtained from these using standard pulsar data reduction techniques and analysed using state-of-the-art Bayesian pulsar timing techniques. We also performed an analysis of the pulsar's total intensity and polarisation profile to understand the interstellar scattering along the line of sight, and we determined the pulsar's geometry by fitting the rotating vector model to the polarisation data. Results. We obtain precise measurements of several post-Keplerian parameters: the range, r = 0.202(6) T⊙, and shape, s = 0.999823(4), of the Shapiro delay, from which we infer: the orbital inclination to be 88.9−0.14+0.15 deg; the masses of the pulsar and the companion to be 1.55(7) M⊙ and 0.202(6) M⊙, respectively; a secular change in the orbital period Ṗb = −53−6.0+7.4 × 10−15 s s−1 that proves the GC association; and a secular change in the projected semi-major axis of the pulsar, ẋ = −40.7−8.2+7.3 × 10−16 s s−1, likely caused by the spin–orbit interaction from a misaligned HeWD spin, at odds with the likely isolated binary evolution of the system. We also discuss some theoretical models for the structure and evolution of white dwarfs in neutron star–white dwarf binaries, using PSR J1910−5959A's companion as a test bed. Conclusions. PSR J1910−5959A is a rare system for which several parameters of both the pulsar and the HeWD companion can be accurately measured. We present the polarization profiles of 22 pulsars in the globular cluster 47 Tucanae using observations from the MeerKAT radio telescope at UHF band (544–1088 MHz) and report precise values of dispersion measure (DM) and rotation measure (RM). We use these measurements to investigate the presence of turbulence in electron density and magnetic fields. The structure function of DM shows a break at ∼30 arcsec (∼0.6 pc at the distance of 47 Tucanae) that suggests the presence of turbulence in the gas in the cluster driven by the motion of wind-shedding stars. On the other hand, the structure function of RM does not show evidence of a break. This non-detection could be explained either by the limited number of pulsars or by the effects of the intervening gas in the Galaxy along the line of sight. Future pulsar discoveries in the cluster could help confirm the presence and localize the turbulence. [ABSTRACT FROM AUTHOR]

We report four new pulsars discovered in the core-collapsed globular cluster (GC) NGC 6624 by the TRAPUM Large Survey Project with the MeerKAT telescope. All of the new pulsars found are isolated. PSR J1823−3021I and PSR J1823−3021K are millisecond pulsars with period of respectively 4.319 and 2.768 ms. PSR J1823−3021J is mildly recycled with a period of 20.899 ms, and PSR J1823−3022 is a long period pulsar with a period of 2.497 s. The pulsars J1823−3021I, J1823−3021J, and J1823−3021K have position and dispersion measure (DM) compatible with being members of the GC and are therefore associated with NGC 6624. Pulsar J1823−3022 is the only pulsar bright enough to be re-detected in archival observations of the cluster. This allowed the determination of a timing solution that spans over two decades. It is not possible at the moment to claim the association of pulsar J1823−3022 with the GC given the long period and large offset in position (∼3 arcmin) and DM (with a fractional difference of 11 percent compared the average of the pulsars in NGC 6624). The discoveries made use of the beamforming capability of the TRAPUM backend to generate multiple beams in the same field of view which allows sensitive searches to be performed over a few half-light radii from the cluster centre and can simultaneously localize the discoveries. We describe the ongoing Relativistic Binary programme (RelBin), a part of the MeerTime large survey project with the MeerKAT radio telescope. RelBin is primarily focused on observations of relativistic effects in binary pulsars to enable measurements of neutron star masses and tests of theories of gravity. We selected 25 pulsars as an initial high priority list of targets based on their characteristics and observational history with other telescopes. In this paper, we provide an outline of the programme, and present polarization calibrated pulse profiles for all selected pulsars as a reference catalogue along with updated dispersion measures. We report Faraday rotation measures for 24 pulsars, twelve of which have been measured for the first time. More than a third of our selected pulsars show a flat position angle swing confirming earlier observations. We demonstrate the ability of the Rotating Vector Model, fitted here to seven binary pulsars, including the Double Pulsar (PSR J0737–3039A), to obtain information about the orbital inclination angle. We present a high time resolution light curve of the eclipse of PSR J0737–3039A by the companion's magnetosphere, a high-phase-resolution position angle swing for PSR J1141–6545, an improved detection of the Shapiro delay of PSR J1811–2405, and pulse scattering measurements for PSRs J1227–6208, J1757–1854, and J1811–1736. Finally, we demonstrate that timing observations with MeerKAT improve on existing data sets by a factor of, typically, 2–3, sometimes by an order of magnitude. [ABSTRACT FROM AUTHOR]

Using the state-of-the-art SKA precursor, the MeerKAT radio telescope, we explore the limits to precision pulsar timing of millisecond pulsars achievable due to pulse stochasticity (jitter). We report new jitter measurements in 15 of the 29 pulsars in our sample and find that the levels of jitter can vary dramatically between them. For some, like the 2.2 ms pulsar PSR J2241−5236, we measure an implied jitter of just ∼4 ns h−1, while others, like the 3.9 ms PSR J0636−3044, are limited to ∼100 ns h−1. While it is well known that jitter plays a central role to limiting the precision measurements of arrival times for high signal-to-noise ratio observations, its role in the measurement of dispersion measure (DM) has not been reported, particularly in broad-band observations. Using the exceptional sensitivity of MeerKAT, we explored this on the bright millisecond pulsar PSR J0437−4715 by exploring the DM of literally every pulse. We found that the derived single-pulse DMs vary by typically 0.0085 cm−3 pc from the mean, and that the best DM estimate is limited by the differential pulse jitter across the band. We postulate that all millisecond pulsars will have their own limit on DM precision which can only be overcome with longer integrations. Using high-time resolution filterbank data of 9 μs, we also present a statistical analysis of single-pulse phenomenology. Finally, we discuss optimization strategies for the MeerKAT pulsar timing program and its role in the context of the International Pulsar Timing Array. [ABSTRACT FROM AUTHOR]

In the last decade, zebrafish has been used as a model for the study of several human skin diseases. The epidermis of Danio rerio is composed of keratinocytes and two types of secretory cells: mucous cells and club cells. Club cells have multiple biological functions and among them may be important in the protection against ultraviolet damage through the proliferative response or through the increased production of protective substances. Calcium-binding proteins such as calbindin D28K and calretinin are used as markers of nervous and enteric nervous systems, but they are present in numerous other cells. These proteins are involved in a wide variety of cell activities, such as cytoskeletal organization, cell motility and differentiation, cell cycle regulation and neuroprotective function. In this study we demonstrated, for the first time, the presence of calretinin and calbindin D28K in skin club cells of Danio rerio exposed to different wavelengths by immunohistochemistry analysis. Exposure to white-blue light and blue light causes the expression and colocalization of calbindin-D28K and calretinin. These proteins were moderately expressed and no colocalization was observed in the club cells of the control fish. In zebrafish exposed to continuous darkness for 10 days, in the club cells the two antibodies did not detect any proteins specifically. These results demonstrate that calbindin and calretinin could be involved in the pathophysiology of skin injury due to exposure to short-wavelength visible light spectrums. [ABSTRACT FROM AUTHOR]

The millisecond pulsar J1823−3021A is a very active giant pulse emitter in the globular cluster NGC 6624. New observations with the MeerKAT radio telescope have revealed 14350 giant pulses over 5 h of integration time, with an average wait time of about 1 s between giant pulses. The giant pulses occur in phases compatible with the ordinary radio emission, follow a power-law distribution with an index of −2.63 ± 0.02, and contribute 4 per cent of the total integrated flux. The spectral index of the giant pulses follows a Gaussian distribution centred around −1.9 with a standard deviation of 0.6 and is on average flatter than the integrated emission, which has a spectral index of −2.81 ± 0.02. The waiting times between the GPs are accurately described by a Poissonian distribution, suggesting that the time of occurrence of a GP is independent from the times of occurrence of other GPs. 76 GPs show multiple peaks within the same rotation, a rate that is also compatible with the mutual independence of the GP times of occurrence. We studied the polarization properties of the giant pulses finding, on average, linear polarization only at the 1 per cent level and circular polarization at the 3 per cent level, similar to the polarization percentages of the total integrated emission. In four cases, it was possible to measure the RM of the GPs that are highly variable and, in two cases, is inconsistent with the mean RM of the total integrated pulsar signal. [ABSTRACT FROM AUTHOR]

We report here on initial results from the Thousand-Pulsar-Array (TPA) programme, part of the Large Survey Project 'MeerTime' on the MeerKAT telescope. The interferometer is used in the tied-array mode in the band from 856 to 1712 MHz, and the wide band coupled with the large collecting area and low receiver temperature make it an excellent telescope for the study of radio pulsars. The TPA is a 5 year project, which aims at to observing (a) more than 1000 pulsars to obtain high-fidelity pulse profiles, (b) some 500 of these pulsars over multiple epochs, and (c) long sequences of single-pulse trains from several hundred pulsars. The scientific outcomes from the programme will include the determination of pulsar geometries, the location of the radio emission within the pulsar magnetosphere, the connection between the magnetosphere and the crust and core of the star, tighter constraints on the nature of the radio emission itself, as well as interstellar medium studies. First, results presented here include updated dispersion measures, 26 pulsars with Faraday rotation measures derived for the first time, and a description of interesting emission phenomena observed thus far. Despite considerations of mass-loss from stellar evolution suggesting otherwise, the content of gas in globular clusters seems poor and hence its measurement very elusive. One way of constraining the presence of ionized gas in a globular cluster is through its dispersive effects on the radiation of the millisecond pulsars included in the cluster. This effect led Freire et al. to the first detection of any kind of gas in a globular cluster in the case of 47 Tucanae. By exploiting the results of 12 additional years of timing, as well as the observation of new millisecond pulsars in 47 Tucanae, we revisited this measurement: we first used the entire set of available timing parameters in order to measure the dynamical properties of the cluster and the three-dimensional position of the pulsars. Then we applied and tested various gas distribution models: assuming a constant gas density, we confirmed the detection of ionized gas with a number density of n  = 0.23 ± 0.05 cm−3, larger than the previous determination (at 2σ uncertainty). Models predicting a decreasing density or following the stellar distribution density are highly disfavoured. We are also able to investigate the presence of an intermediate mass black hole in the centre of the cluster, showing that is not required by the available data, with an upper limit for the mass at ∼4000 M⊙. Purpose: Overweight and obesity are important risk factors for diabetes, cardiovascular diseases, and premature death in modern society. Recently, numerous natural and synthetic compounds have been tested in diet-induced obese animal models, to counteract obesity. Melatonin is a circadian hormone, produced by pineal gland and extra-pineal sources, involved in processes which have in common a rhythmic expression. In teleost, it can control energy balance by activating or inhibiting appetite-related peptides. The study aims at testing effects of melatonin administration to control-fed and overfed zebrafish, in terms of expression levels of orexigenic (Ghrelin, orexin, NPY) and anorexigenic (leptin, POMC) genes expression and morphometry of visceral and subcutaneous fat depots.Methods: Adult male zebrafish (n = 56) were divided into four dietary groups: control, overfed, control + melatonin, overfed + melatonin. The treatment lasted 5 weeks and BMI levels of every fish were measured each week. After this period fishes were sacrificed; morphological and morphometric studies have been carried out on histological sections of adipose tissue and adipocytes. Moreover, whole zebrafish brain and intestine were used for qRT-PCR.Results: Our results demonstrate that melatonin supplementation may have an effect in mobilizing fat stores, in increasing basal metabolism and thus in preventing further excess fat accumulation. Melatonin stimulates the anorexigenic and inhibit the orexigenic signals.Conclusions: It seems that adequate melatonin treatment exerts anti-obesity protective effects, also in a diet-induced obesity zebrafish model, that might be the result of the restoration of many factors: the final endpoint reached is weight loss and stabilization of weight gain. Summary: The mechanosensory lateral line system of fish is responsible for several functions such as balance, hearing, and orientation in water flow and is formed by neuromast receptor organs distributed on head, trunk and tail. Superficial and canal neuromasts can be distinguished for localization and morphological differences. Several information is present regarding the superficial neuromasts of zebrafish and other teleosts especially during larval and juvenile stages, while not as numerous data are so far available about the ultrastructural characteristics of the canal neuromasts in adult zebrafish. Therefore, the aim of this study was to investigate by transmission electron microscopy the ultrastructural aspects of cells present in the canal neuromasts. Besides the typical cellular aspects of the neuromast, different cellular types of hair cells were observed that could be identified as developing hair cells during the physiological turnover. The knowledge of the observed cellular types of the canal neuromasts and their origin could give a contribution to studies carried out on adult zebrafish used as model in neurological and non‐neurological damages, such as deafness and vestibular disorders. The origin of the nuclear star cluster in the centre of our Galaxy is still unknown.One possibility is that it formed after the disruption of stellar clusters that spiralled into the Galactic Centre due to dynamical friction.We trace the formation of the nuclear star cluster around the central black hole, using state-of-the-art N-body simulations, and follow the dynamics of the neutron stars born in the clusters.We then estimate the number of millisecond pulsars (MSPs) that are released in the nuclear star cluster during its formation. The assembly and tidal dismemberment of globular clusters lead to a population ofMSPs distributed over a radius of about 20 pc, with a peak near 3 pc. No clustering is found on the subparsec scale. We simulate the detectability of this population with future radio telescopes like the MeerKAT radio telescope and SKA1, and find that about an order of 10 MSPs can be observed over this large volume, with a paucity of MSPs within the central parsec. This helps discriminating this scenario from the in situ formation model for the nuclear star cluster that would predict an overabundance of MSPs closer to the black hole. The linearly polarized component of a pulsar signal at different radio frequencies can help to constrain the parallel component of the magnetic field along the line of sight. In this work we measured the polarimetric properties of the pulsars in the globular cluster 47 Tucanae and we report the Rotation Measure (RM) for 13 of them. A gradient in the RM values of the pulsars across the cluster is detected suggesting the presence of significant variations in the magnetic field across the very small angular scales associated with the lines of sight to the pulsars in 47 Tucanae. Both magnetic fields located in the globular cluster or in the Galactic disk in the direction of the cluster are taken into consideration. However, more detailed modelling of the dynamics of the cluster and deeper observations with the MeerKAT and/or the SKA1 radio telescopes are necessary to discriminate among the models. The oral cavity of vertebrates has a very important role in many functions strictly related to the food processing. Two species, sea bass Dicentrarchus labrax and seabream Sparus aurata, are mainly involved in the aquaculture industry in the mediterranean area; moreover, the white seabream Diplodus sargus sargus was recently chosen as the best candidate for the diversification of species of commercial interest. This investigation was carried out, using the standard procedures for light and scanning electron microscopy, to analyse the morphology of the tongue dorsal surface in order to show whether relationships are present between the tongue morphology and the nutritional choices of these species. In all the three investigated species, three different areas were observed on the dorsal lingual surface: an apex, a body and a root. In the sea bass D. labrax, numerous caninelike teeth, organized in pads, were present along the dorsal surface. The presence of numerous taste buds, scattered on the tongue, was shown. In the seabream S. aurata, the apical part of the tongue is inserted in a pouch with a characteristic medial ridge on the body surface. Numerous taste buds were scattered on the whole surface. In the white seabream D. sargus sargus, a pouch partially covering the apex was clearly observed. In the pouch, filiform-like and foliate-like papillae can be distinguished. The results show the important role of the fish tongue in the food ingestion process, demonstrating the interaction of food processing, taste and morphological characteristics. Contents Orexin A and B, also known as hypocretin A and B, are hypothalamic neuropeptides arising from a precursor to the 130 amino acid, called pre-pro orexin. They are synthesized mainly in lateral and posterior hypothalamus and are involved in different functions such as regulation of food intake and energy balance. Orexins and orexin receptors were previously described also in different tissues and organs outside the brain. The aim of this study was to demonstrate by means of the immunofluorescence technique, the presence of orexin A in the ovary of cat and dog, to support the hypothesis of the role of this substance also at the level of the female genital system. The presence of orexin A in the ovary either in dog or in cat is in agreement with previous data on the presence and role of orexins in the female genital system of other species. Ionic channels play key roles in the sensory cells, such as transducing specific stimuli into electrical signals. The acid-sensing ion channel (ASIC) family is voltage-insensitive, amiloride-sensitive, proton-gated cation channels involved in several sensory functions. ASIC2, in particular, has a dual function as mechano- and chemo-sensor. In this study, we explored the possible role of zebrafish ASIC2 in olfaction. RT-PCR, Western blot, chromogenic in situ hybridization and immunohistochemistry, as well as ultrastructural analysis, were performed on the olfactory rosette of adult zebrafish. ASIC2 mRNA and protein were detected in homogenates of olfactory rosettes. Specific ASIC2 hybridization was observed in the luminal pole of the non-sensory epithelium, especially in the cilia basal bodies, and immunoreactivity for ASIC2 was restricted to the cilia of the non-sensory cells where it was co-localized with the cilia marker tubulin. ASIC2 expression was always absent in the olfactory cells. These findings demonstrate for the first time the expression of ASIC2 in the olfactory epithelium of adult zebrafish and suggest that it is not involved in olfaction. Since the cilium sense and transduce mechanical and chemical stimuli, ASIC2 expression in this location might be related to detection of aquatic environment pH variations or to detection of water movement through the nasal cavity. The gilthead seabream is a food fish, one of the most frequently used in aquaculture. In the species of commercial interest, feeding in captivity is very important and this process is strictly related to the morphological characteristics of the oral cavity. The aim of this study is, using the standard procedures for light and scanning electron microscopy, to analyze the morphology of the tongue dorsal surface to show if relationships are present between the tongue morphology and the nutritional habits and choices of this farmed species. The main characteristic of the gilthead seabream oral cavity floor is the presence of an apical pouch, with, probably, a protective role mainly for the apical, free part of the tongue. Three zones, like in other teleosts, an apex, a body and a root, can be clearly distinguished. In the pouch foliate-like papillae were observed, while the whole tongue is characterized by the presence of two types of papillae, respectively with a fungiform and cylindroid aspect, both randomly distributed throughout the whole dorsal surface of the tongue. Scattered and numerous taste buds, with the typical pear-onion shape, together with small and numerous taste pores are also present, distributed throughout the tongue surface. Our results demonstrate that in the gilthead seabream important mechanic and sensory roles are carried out by specific anatomical structures. Our anatomical data could give, together with further biochemical and physiological data, an important support with the aim of improving the nutrition of aquaculture species. Microsc. Res. With 4 figures The Italian lizard ( Podarcis sicula) is the most diffused reptile in Italy, but it is also present in other European countries. This lizard belongs to the Lacertidae family, lives near walls, slants and along the borders of the paths; its diet includes bugs and aracnids. No data are so far available in literature about the three-dimensional morphology of the tongue of Podarcis sicula, therefore the aim of the present paper was to study by scanning electron and light microscopy the three-dimensional characteristics of the dorsal lingual surface and moreover the presence of chemosensory receptors like the taste buds in the oral cavity. Our results demonstrate that the Podarcis sicula tongue is a triangular muscular membranous organ, dorsoventrally flattened and that three different areas can be observed: a bifid apex, a body and a root. No papillae were observed in the apex, characterized by a flattened mucosa and by two deep median pouches. In the body cylindrical papillae with a flat surface are present, aborally gradually substituted by imbricated papillae. Foliate-like papillae were observed in the lateral parts of the tongue body. No sensory structures were showed on the lingual dorsal surface, while they were numerous in the oral cavity, particularly on the gingival epithelium. The light microscopy shows, on the dorsal surface, a stratified pavimentous not keratinized epithelium, conversely keratinized along the ventral surface. Many caliciform cells on the lateral parts of the papillae, deputed to the secretion of mucus, were also observed. Therefore, the results obtained in this paper could give a contribution to the knowledge of the tongue anatomy in a species widely diffused in different European countries and could be of help for clinical purposes in reptiles. The zebrafish is a common model for developmental studies including those regarding tooth, palate and tongue. Nevertheless, little information is available about the morphology of the oral cavity in this teleost, especially in adult animals. In this study we used light, scanning and transmission electron microscopy to describe in detail the morphology of the oral cavity of adult zebrafish. The oral cavity could be divided into three different zones: the outer containing the lips, the intermediate corresponding to the internal valves and the internal that corresponds to the tongue. In the upper and lower intermediate zones, there were semilunar shaped valves, more prominent in the upper part. The internal lower zones correspond to the palate and the tongue, which is an individualized structure filled with numerous transversal ridges. Both the intermediate and internal zones were covered by a stratified epithelium containing numerous mucous and rodlet cells. Present data provide the first description of the morphology and structure of the oral cavity in the adult zebrafish and might serve as a baseline for developmental studies of the oral cavity using this teleost as a model. Neurotrophins play a critical role in the development of the mammalian ovary, oogenesis and folliculogenesis. In this study we investigated the cell localization of the two main receptors for nerve growth factor (NGF), TrkA and p75NTR, in the cow and pig ovary, using immunohistochemistry. Specific immunoreactivity for TrkA and p75NTR was detected in the ovary of both species, but the pattern and intensity of immunostaining were marginally different between them. The follicular cells regularly expressed immunoreactivity for both receptors. Immunoreactivity was also detected in the oocytes, independently of the maturational stage of the follicles, with the exception of primordial and primary follicles of the pig which did not display p75NTR. Taken together, these results suggest a possible direct role of NGF on oocytes expressing TrkA and p75NTR, in addition to the well-known roles in other ovary functions. We studied the effects of (post-tetanic) potentiation on myosin light chain (MLC-2) phosphorylation, work and energy cost in skeletal muscle. Experiments were performed using in situ medial gastrocnemius muscles of male Wistar rats, which were electrically stimulated through the severed sciatic nerve. One group of muscles was first potentiated with an isometric tetanus before a series of 10 concentric contractions (PRC). A second group performed the same series of contractions without previous potentiation (RC). Following the last contraction the muscles were rapidly frozen and excised after which the high-energy phosphate content, lactate concentration and the level of MLC-2 phosphorylation were measured. The results indicate that PRC muscles had a higher ( P < 0.05) total work output 144.5 ± 17.0 (SD) ( n = 6) vs. 121.6 ± 11.4 (SD) ( n = 6) mJ and level of MLC-2 phosphorylation (49.2 ± 7.3 vs. 40.8 ± 3.6%) than RC muscles. The energy cost of the series of concentric contractions in the PRC muscles (9.8 ± 1.9 μmol∼P/muscle) was significantly higher ( P < 0.05) than the energy cost in the RC muscles (6.2 ± 0.97 μmol∼P/muscle). It was shown that the relative increase in energy cost of PRC muscles was higher ( P < 0.05) than in total work output. It is proposed that the relative high increase in energy cost is the direct result of the increase in muscle performance rather than a property of potentiation. [ABSTRACT FROM AUTHOR]

Abstract Rationale: The prefrontal cortical (PFC) dopamine (DA) system has been implicated in anxiety-related behavioral changes, but direct, unequivocal support for this idea is sparse. Objectives: The present aim was to study the functional significance of prefrontal DA using the pentylenetetrazol (PTZ) discrimination model of anxiety. A comparison was made with its role in the cue of the anxiolytic drug chlordiazepoxide (CDP). Methods: Two groups of rats were trained to discriminate either PTZ (20 mg/kg, s.c.) or CDP (10 mg/kg, J.p.) from saline using an operant drug discrimination procedure. After prolonged training, half of each group was used to assess biochemical changes induced by both drugs in different sub areas of the PFC. For the remaining rats, discrimination training continued and generalization tests with PTZ and CDP were performed. Rats were then provided with bilateral guide cannulae aimed at the ventromedial (vm) PFC, and the effects of local infusions of DAergic drugs on discriminative performance were evaluated. Results: CDP did not affect PFC DA activity, but PTZ increased the DOPAC/DA ratio in the vmPFC selectively. Generalization tests showed that the cues of PTZ and CDP were dose dependent. In PTZ-trained rats, infusions of the DA receptor antagonist cis-flupenthixol into the vmPFC blocked the PTZ cue dose dependently, whereas the agonist apomorphine partially generalized to this cue. In CDP-trained rats, neither drug antagonized or generalized to the CDP cue, showing that PFC DA is not critically involved in the CDP cue and that local pharmacological manipulations of PFC DA do not affect discriminative abilities per se. In recent years, data have accumulated suggesting that the role of neurotrophins and Trk receptors may not be limited to the nervous system, and the presence of these substances has been detected in a variety of vertebrate and invertebrate non-nervous tissues. This study was designed to map the expression of immunoreactivity (IR) for Trk-like proteins in alevins of the teleost Dicentrarchus labrax, with particular emphasis on non-nervous structures. We used antibodies against specific epitopes of the intracellular domain of these proteins, a region that is highly conserved in phylogeny. Trk-like IR was seen in segregate cell populations of the nervous system, and non-nervous tissues. In the central nervous system TrkA-like and TrkC-like IR was abundant, whereas TrkB-like IR was restricted to a low number of brain areas. Expression of Trk-like protein IR was observed in the peripheral nervous system and sensory organs, with the exception of the lateral line organ. Outside the nervous system, TrkA-like IR was mainly found in different epithelia, TrkB-like IR in the endocrine and digestive system, and TrkC-like IR in the cardiovascular and immune systems. The gills showed IR for all three Trk-like proteins, whereas they were absent from the gonads. Furthermore, scattered cells positive for Trk-like proteins were found in most of the investigated tissues. The distribution of Trk-like IR in this teleost is compared with that of mammals and birds, which it often paralleled, and the possible role of neurotrophins and Trk-like receptor proteins in different non-neuronal tissues is discussed. Gram-negative bacterial infections of the central nervous system are generally associated with high morbidity and mortality rates. In patients with ventriculitis induced by gram-negative liquoral sepsis, a reduction in cerebrospinal fluid formation has been reported, suggesting that gram-negative ventriculitis is able per se to alter the normal functioning of the choroid plexus. The aim of the present study was to analyse, for the first time in the rat, the effects of tcute ventriculitis on the ultrastructure of the choroid plexus. A simple and inexpensive experimental model of acute ventriculitis was developed: we injected into the cisterna magna of rats 10 CFU of live Escherichia coli, inducing septic ventriculitis without major neurological deficits. Histological examinations of rodent choroid plexus 24 h after the injection revealed patches of altered epithelium, with swollen and vacuolated ependymal cells associated with leukocyte infiltration. Electron microscopy demonstrated a reduced number of microvilli and flattening of the epithelial surface. These results (a) indicate that gramnegative septic ventriculitis is able to induce visible ultrastructural alterations of the choroid plexus which (b) are consitent with a picture of marked reduction of the functioning epithelial choroid plexus surface, and (c) highlight the potential usefulness of our rodent acute ventriculitis model for developing treatment modalities.
