Different forearm postures can modulate corticospinal excitability. However, there is no consensus on whether handedness plays a role in such a mechanism. This study investigated the effects of 3 forearm postures (pronation, neutral, and supination) on the corticospinal excitability of muscles from the dominant and nondominant upper limbs. Surface electromyography was recorded from the abductor digiti minimi, flexor pollicis brevis, and flexor carpi radialis from both sides of 12 right-handed volunteers. Transcranial magnetic stimulation pulses were applied to each muscle's hotspot in both cerebral hemispheres. Motor-evoked potential peak-to-peak amplitude and latency and resting motor threshold were measured. The data were evaluated by analysis of variance. The level of significance was set at 5%. The resting motor threshold was similar for the 3 muscles and both sides. Motor-evoked potential peak-to-peak amplitude from flexor pollicis brevis was lower during supination, and the dominant upper limb latency was longer. The flexor carpi radialis presented lower motor-evoked potential peak-to-peak amplitudes for neutral and shorter latencies during supination. Abductor digiti minimi seemed not to be affected by posture or side. Musculoskeletal modeling, typically implemented using marker-based systems in laboratory environments, is commonly used for noninvasive estimations of loads. Inertial measurement units (IMUs) have become an alternative for the evaluation of kinematics. However, estimates of spine joint contact forces using IMUs have yet to be thoroughly evaluated. Dynamics tasks and static postures from activities of daily living were captured on 11 healthy subjects using both systems simultaneously. Spine kinematics obtained from IMU- and marker-based systems and L4–L5 joint contact forces were compared. Lateral bending resulted in a weak agreement with significant differences between the 2 systems (P =.02, average root mean-squared error = 4.81), whereas flexion–extension and axial rotation exhibited the highest agreement with no significant differences (P <.05, average root mean-squared error = 5.51 and P <.31, average root mean-squared error = 5.08, respectively). All tasks showed excellent correlations (R2 =.76–.99) in estimated loads between systems. Differences in predicted loads at the L4–L5 were only observed during flexion–extension (1041 N vs 947 N, P =.0004) and walking with weights (814 N vs 727 N, P =.004). Different joint reaction force outcomes were obtained in 2 of the 8 tasks between systems, suggesting that IMUs can be robust tools allowing for convenient and less expensive evaluations and for longitudinal assessments inside and outside the laboratory setting. Background: Retirement transition has been shown to associate with changes in physical activity (PA) and self-rated health (SRH), but their interrelationship is less studied. The aim was to investigate changes in accelerometer-measured total PA, moderate to vigorous PA (MVPA), and sedentary time across retirement transition as a predictor of SRH. Methods: Data from the Swedish Retirement Study and the Finnish Retirement and Aging study were harmonized and pooled. Data from 3 waves (about 12 mo apart) were included: 1 preretirement (wave 1) and 2 postretirement follow-ups (wave 2–3). A totally of 245 participants (27% men) were included. Thigh-worn accelerometers were used to collect data for PA variables (wave 1–2), and SRH was obtained from the questionnaire (wave 1–3). Results: Between wave 1 and 2, total PA decreased with 11 (CI, −22 to −1) minutes per day, MVPA was stable (0 [CI, −3 to 3] min), and sedentary time decreased nonsignificantly with 9 (CI, −20 to 1) minutes. SRH changed between all 3 waves (all P <.001). At preretirement, 10 more minutes of MVPA was associated with greater odds of better SRH when adjusting for accelerometer wear-time, cohort, sex, age, and occupational status (odds ratio: 1.11 [95% CI, 1.02–1.22]). This association was no longer statistically significant when additionally adjusting for marital status, body mass index, and smoking. No significant associations were observed between changes in the PA variables during retirement transition and SRH at postretirement follow-ups. Conclusions: This study showed a cross-sectional association between MVPA and greater odds of reporting better SRH before retirement. Interoperability in computational chemistry is elusive, impeded by the independent development of software packages and idiosyncratic nature of their output files. The cclib library was introduced in 2006 as an attempt to improve this situation by providing a consistent interface to the results of various quantum chemistry programs. The shared API across programs enabled by cclib has allowed users to focus on results as opposed to output and to combine data from multiple programs or develop generic downstream tools. Initial development, however, did not anticipate the rapid progress of computational capabilities, novel methods, and new programs; nor did it foresee the growing need for customizability. Here, we recount this history and present cclib 2, focused on extensibility and modularity. We also introduce recent design pivots—the formalization of cclib's intermediate data representation as a tree-based structure, a new combinator-based parser organization, and parsed chemical properties as extensible objects. Purpose: To evaluate the clinical survival and impact on oral health--related quality of life (OHRQoL) of immediate loading of implant-supported full-arch CAD/CAM--milled PMMA interim restorations. Materials and Methods: This was a practice-based prospective cohort study performed in a Portuguese dental clinic. Eleven patients received a total of 17 CAD/CAM fully implant-supported, screw-retained, full-arch restorations with milled PMMA and were followed up for 1 year. The primary outcome was prosthesis survival, determined with a modification of the California Dental Association score and a Kaplan-Meier survival function analysis. The secondary outcome was the patient quality of life, as defined by application of the OHIP-14-PT questionnaire and standardized effect size variation between two visits. Significance was set at 5%. Results: Survival probability at 12 months was 76%, complete fracture of the prosthesis occurred in 17.6% of the cases, and small fractures without lab involvement occurred in 5.9% of cases. There was a significant improvement in OHRQoL between visit 1 and visit 4. The mean difference and effect size for total OHIP-14-PT score were -32.91 ± 3.68 and 3.66 (95% CI -1.83 to -5.80) respectively (P < .001* Wilcoxon matched paired rank test). Purpose: The monitoring of training loads and quantification of physical performance are common practices in youth soccer academies to support coaches in prescribing and programming training for individuals. The interaction between training load and physical performance is unknown during a preseason period in youth soccer players. The current study assessed changes in training load and physical assessments across a 4-week preseason period. The relationship between physical performance and match playing time in youth male soccer players was also investigated. Methods: The training loads of 25 professional youth academy male soccer players were monitored throughout a 4-week preseason period. Assessments of power, agility, speed, and aerobic capacity were undertaken in the first training session. Session ratings of perceived exertion (sRPE) and well-being questionnaires were collected during all training sessions and preseason matches. Playing time during subsequent competitive matches was recorded. Results: T test and 30-m-sprint assessments, conducted on the first day of preseason, were predictors of sRPE throughout preseason (t test χ2/df = 2.895, poor adjustment; 30-m sprint χ2/df = 1.608, good adjustment). YoYo Test performance was related with changes in perceived fatigue (χ2/df = 0.534, very good adjustment). Faster players reported higher values of sRPE, and players with higher aerobic capacity reported higher levels of fatigue across preseason. Well-being, perceived fatigue and soreness, and sRPE decreased across preseason. Greater match durations were related to higher levels of fatigue during preseason (P <.05). Conclusion: The current study highlights the relationship between training load, physical assessments, and playing time. Coaches and practitioners can use physical test data at the start of preseason as an indication of players that report higher sRPE, perceived fatigue, and reduced well-being across preseason, supporting decisions around individualized training prescriptions. Simple Summary: Tumor necrosis can be frequently found in malignant tumors and its prognostic value has been confirmed in multiple solid tumors in human and veterinary medicine. We evaluated the level of necrosis in 154 cases of feline mammary tumors (FMTs) using semi-quantitative and quantitative methods and found that tumor necrosis is particularly common in malignant tumors with clinicopathological characteristics associated with aggressive biological behavior. In addition, there was a positive correlation between the semi-quantitative and the quantitative methods, and the former had a high interobserver agreement. These results provide evidence of the importance of necrosis in FMTs and support the implementation of a semi-quantitative evaluation of necrosis in the FMT diagnostic routine. Necrosis is a common finding in human and animal neoplasms. The percentage of tumor necrosis is included in tumor grading schemes in veterinary oncology; however, evaluation methods are often overlooked. Different studies have assessed the prognostic value of tumor necrosis in feline mammary tumors with contradictory results, which could be related to methodologic variability. In this study, a comprehensive evaluation of tumor necrosis in feline mammary tumors (FMTs) was conducted, by applying a semi-quantitative and a quantitative methodology for assessing necrosis. The interobserver agreement, the relationship with clinicopathological characteristics and the prognostic value of necrosis were analyzed in 154 FMT cases. Although subjectivity in the assessment of necrosis existed, an almost perfect agreement (weighted quadratic k = 0.851) between two observers was obtained. Furthermore, there was a significant positive correlation between the semi-quantitative and quantitative methods. Necrosis was more common and more extensive in malignant tumors than in their benign counterparts. Despite the non-significant results in the survival analysis, extensive necrosis was significantly associated with aggressive clinicopathological features, such as higher histological grade, high mitotic count and lymphovascular invasion. Objective: Chronic inflammation's role in the pathogenesis, development, and rupture of vascular malformations is undebated. Advanced magnetic resonance imaging techniques with vessel wall studies, specifically Black Blood (bbMRI), may offer insights into vascular wall instability and predict rupture. This case series aims to assess bbMRI as a predictive diagnostic tool for brain arteriovenous malformations (bAVMs) rupture, suggesting early treatment. Material and methods: A prospective study included demographic, clinical, and neuroimaging data from a consecutive series of patients with ruptured or unruptured bAVMs, regardless of age or gender, between October 2018 and March 2024. All patients underwent MRI brain resonance with Black Blood study. Those with impaired renal function were excluded. Statistical analyses tested sample homogeneity. Univariate and multivariate logistical regressions assessed bbMRI as a rupture predictor for bAVMs, with a p-value set at < 0.05. Results: Ninety patients were retrieved: 64 with unruptured and 26 with ruptured bAVMs. The mean age was 31.9 years, and 55 were female. Admission symptoms were headache, neurological deficits, and seizure in 49, 21, and 19 cases, respectively. bbMRI showed wall enhancement in 56 cases, with 19 in the ruptured and 37 unruptured groups. Univariate and multivariate analyses revealed a significant correlation between bbMRI wall enhancement and bAVM rupture (p:0.033; p:0.047). Conclusion: bbMRI may be a useful and feasible diagnostic implement to determine vessel inflammation and the bAVMs prone to rupture. Post-translational cycles of α-tubulin detyrosination and tyrosination generate microtubule diversity, the cellular functions of which remain largely unknown. Here we show that α-tubulin detyrosination regulates kinetochore-microtubule attachments to ensure normal chromosome oscillations and timely anaphase onset during mitosis. Remarkably, detyrosinated α-tubulin levels near kinetochore microtubule plus-ends depend on the direction of chromosome motion during metaphase. Proteomic analyses unveil that the KNL-1/MIS12/NDC80 (KMN) network that forms the core microtubule-binding site at kinetochores and the microtubule-rescue protein CLASP2 are enriched on tyrosinated and detyrosinated microtubules during mitosis, respectively. α-tubulin detyrosination enhances CLASP2 binding and NDC80 complex diffusion along the microtubule lattice in vitro. Rescue experiments overexpressing NDC80, including variants with slower microtubule diffusion, suggest a functional interplay with α-tubulin detyrosination for the establishment of a labile kinetochore-microtubule interface. These results offer a mechanistic explanation for how different detyrosinated α-tubulin levels near kinetochore microtubule plus-ends fine-tune load-bearing attachments to both growing and shrinking microtubules. Background: Head injuries are the leading cause of trauma in Emergency Departments (EDs). Recent studies have shown epidemiological changes in patients consulting ED for head injuries. The main objective of this study was to describe the profile of head injury patients consulting in the EDs in France and assess i) head injury severity across age groups; ii) the delay between the occurrence of head injury and ED arrival; iii) factors associated with traumatic intracranial hemorrhage (ICH). Methods: This cross-sectional study collected patient data over a three-day period in March 2023. All adult patients (≥ 18 years old) admitted to the ED with a head injury (defined as a trauma to the head) were included. TBI severity was classified according to patients’ initial Glasgow Coma Scale score in the ED: severe (3–8); moderate (9 -12); mild (13–15); and simple head trauma in the absence of transient or persistent neurological symptoms. Results: Among the 71 participating EDs, 26,008 patients visited EDs and a total of 1070 patients (4.1%, IC 95 3.9—4.4) presented a head injury were included in the study, with a median age of 68.5 [37–85] years old. Most of the patients (66.7%) were referred to ED after a call to the Emergency Medical Dispatcher (EMD). The median time from head injury to ED visit was 2 h [1.0 – 5.5]. Ground-level falls were the leading cause of head injury (60.3%). Most of patient presented a simple head trauma (n = 715, 66.8%) followed by mild TBI (n = 337, 31.5%). CT head scans were performed for 636 patients (59.6%), of which 58 were positive. Traumatic ICH prevalence was 5.4% (95% CI: 4.1–6.9) and three patients (0.3%) required an urgent neurosurgical intervention. Neither preinjury anticoagulant (p = 0.97) nor antiplatelet (p = 0.93) use was associated with an increased risk of traumatic ICH. Conclusions: One head injury patient out of two presenting in the ED is aged over 65 years. Patients referred by EMD were more likely to visit ED promptly. Background: Post-stroke cognitive impairment (PSCI) occurs in up to 50% of stroke survivors. Presence of pre-existing vascular brain injury, in particular the extent of white matter hyperintensities (WMH), is associated with worse cognitive outcome after stroke, but the role of WMH location in this association is unclear. Aims: We determined if WMH in strategic white matter tracts explain cognitive performance after stroke. Methods: Individual patient data from nine ischemic stroke cohorts with magnetic resonance imaging (MRI) were harmonized through the Meta VCI Map consortium. The association between WMH volumes in strategic tracts and domain-specific cognitive functioning (attention and executive functioning, information processing speed, language and verbal memory) was assessed using linear mixed models and lasso regression. We used a hypothesis-driven design, primarily addressing four white matter tracts known to be strategic in memory clinic patients: the left and right anterior thalamic radiation, forceps major, and left inferior fronto-occipital fasciculus. Results: The total study sample consisted of 1568 patients (39.9% female, mean age = 67.3 years). Total WMH volume was strongly related to cognitive performance on all four cognitive domains. WMH volume in the left anterior thalamic radiation was significantly associated with cognitive performance on attention and executive functioning and information processing speed and WMH volume in the forceps major with information processing speed. The multivariable lasso regression showed that these associations were independent of age, sex, education, and total infarct volume and had larger coefficients than total WMH volume. Conclusion: These results show tract-specific relations between WMH volume and cognitive performance after ischemic stroke, independent of total WMH volume. This implies that the concept of strategic lesions in PSCI extends beyond acute infarcts and also involves pre-existing WMH. This research aims to enhance the design of an injection mold in order to minimize ejection time and increase temperature uniformity by repositioning cooling channels. For 3D transient thermal simulations, the ANSYS finite element method (FEM) program was employed, while the MATLAB program was used for optimization. It was discovered that the method could be used successfully to optimize the design of Conformal Cooling Channels (CCCs) in injection molds. The objective function value of the optimized model represents a near 40% improvement in relation to the initial model. The combined parameterization, simulation, and optimization method has proven beneficial for enhancing the arrangement of channels in an injection machine mold. The methodology is generalizable. Spirulina (Arthospira platensis) is known for its rich content of natural compounds like phycocyanin, chlorophylls, carotenoids, and high protein levels, making it a nutrient-dense food. Over the past decade, research has aimed to optimize the extraction, separation, and purification of these valuable metabolites, focusing on technologies such as high-pressure processing, ultrasound-assisted extraction, and microwave-assisted extraction as well as enzymatic treatments, chromatographic precipitation, and membrane separation. In this study, various extraction methods (conventional vs. ultrasound-assisted), solvents (water vs. phosphate buffer), solvent-to-biomass ratios (1:5 vs. 1:10), and ultrafiltration (PES membrane of MWCO 3 kDa, 2 bar) were evaluated. The quantities of total protein, phycocyanin (PC), chlorophyll a (Cla), and total carotenoids (TCC) were measured. The results showed that ultrasound-assisted extraction (UAE) with phosphate buffer at a 1:10 ratio yielded a metabolite-rich retentate (MRR) with 37.0 ± 1.9 mg/g of PC, 617 ± 15 mg/g of protein, 0.4 ± 0.2 mg/g of Cla, and 0.15 ± 0.14 mg/g of TCC. Water extraction in the concentration process achieved the highest concentrations in MRR, with approximately 76% PC, 92% total protein, 62% Cla, and 41% TCC. These findings highlight the effective extraction and concentration processes to obtain a metabolite-rich retentate from Spirulina biomass, reducing the volume tenfold and showing potential as a functional ingredient for the food, cosmetic, and pharmaceutical industries. Clinical Scenario: Injuries that affect the groin region are among the most common in football players. To prevent this condition, studies have focused on strengthening the adductors, hip flexors, or abdominal muscles. Recent investigations have used an eccentric-biased exercise (Copenhagen Adduction Exercise [CAE]) that promotes functional and architectural adaptations in the muscle tissue, though its effect on injury risk reduction is unknown. Clinical Question: Can the Copenhagen Adduction Exercise prevent groin injuries in soccer players? Summary of Key Findings: The literature was searched for studies investigating the potential groin injury risk reduction effect of the CAE. (1) Three studies met the inclusion criteria and were used for this appraisal; (2) one study observed a significantly lower injury rate ratio favoring the group that used the CAE program; and (3) 2 studies found similar or higher injury rates in the intervention groups, not supporting the inclusion of the CAE as a preventative tool. Clinical Bottom Line: There is conflicting evidence that usage of the CAE is superior to not performing adductor strengthening exercises in mitigating the risk of sustaining groin injuries. Given the evidence supporting these findings, it is advisable to exercise caution when contemplating the incorporation of the CAE into training regimens aimed at preventing groin injuries. This study determined whether natural preservatives, when used as substitutes for sulfur dioxide (SO2) in Boerewors, influenced water activity (aw), pH, and moisture content; microbial and sensory quality; and the lipid oxidative and color stability in Boerewors models. The inclusion of sulfur dioxide at 0.035% (positive control), formulation without preservatives (negative control); protective cultures (PrC1 and PrC2), containing lactic acid bacteria; and plant extract blends (KD1 and KD2), containing rosemary and acerola extracts and buffered vinegar, were evaluated. The use of KD2 increased pH on Days 0 and 6, but the increased pH did not affect the microbial preservative effect. The aw of the treatments was stable over 6 days and therefore also did not have an influence on the microbial preservative effect of the natural preservatives. Both plant extract preservatives (KD1 and KD2) increased the oxidative stability of lipids more than any other treatment, including the positive control. The microbial stability results were inconclusive because of the protective cultures, which increased the total bacteria count. The counts of coliform and Enterobacteriaceae, Escherichia coli, and Staphylococcus aureus were analyzed, but significant differences between the treatments were not observed. KD1, which contained rosemary plant extract, maintained the lowest TBC at the same level as the positive control but had a slightly lower redness score. Treatments had no effect on sensory evaluation. The use of KD1 and KD2 plant preservatives proofed to be worthy as replacers of SO2 in Boerewors. Inflammation is a frequent finding in feline mammary neoplasms. Recent research suggests that the presence and location of tumour‐associated immune cells might play a significant role in the clinical outcome of feline mammary carcinomas. The present study aimed to characterise the overall inflammatory infiltrates in healthy, hyperplastic/dysplastic, benign and malignant lesions of the feline mammary gland, and to evaluate its association with clinicopathological features. Perilesional and intralesional inflammatory foci were evaluated in 307 lesions from 185 queens, and categorised according to its distribution and intensity. The presence, location and density of tertiary lymphoid structures were also assessed. A control group included 24 queens without mammary changes. The presence of intralesional and perilesional inflammatory infiltrate was observed in a majority of the lesions (80.8% and 90.2%, respectively), but differed according to the type of mammary lesion, being more remarkable in malignant neoplasms. Only scarce individual cells were observed in 28.1% of the normal mammary glands. Data analysis revealed statistically significant associations (p < 0.05) between the presence of a more prominent intralesional and perilesional inflammatory reaction and several clinicopathological features associated with worse prognosis, including clinical stage, tumour size, mitotic count, lymphovascular invasion and lymph node metastasis. Furthermore, tertiary lymphoid structures were significantly more frequent in tumours with an infiltrative growth and lymph node metastasis. The present study was conducted with the aim of evaluating the effects, over 6 days, of different intensities of continuous light (20, 40, and 60 µmol photons m-2 s-1) on the growth of Arthrospira platensis and its impact on the production of phycocyanin, carotenoids, and intracellular/extracellular auxins, using a medium supplemented with 0.5 g L-1 of L-tryptophan. Additionally, the study aimed to assess the effect of treatments on the biostimulant activity of the supernatant (T0: untreated, T1: treated) compared to that of biomass extracts (T2). T0, T1, and T2 were adjusted to a concentration of 0.1 mg L-1 of indole-3-acetic acid (IAA) and a pH of 7.4. The results revealed that light intensities of 40 and 60 µmol photons m-2 s-1 produced the highest levels of extracellular IAA on day 2 (20.1 and 33.01 mg L-1, respectively), while an intensity of 20 µmol photons m-2 s-1 reached peak IAA production on day 3 (25.74 mg L-1), followed by a subsequent decrease. Phycocyanin concentrations markedly decreased after maximal IAA production under all light conditions. Tukey's analysis indicated that treatments T1 and T2 significantly increased the number and length of secondary roots in mung beans by 157% and 350%, respectively, with no significant differences between them, while the untreated supernatant (T0) exhibited a minor effect. The study concludes that under all assessed light conditions, concentrations of phycocyanin and carotenoids drastically decreased after peak IAA production, gradually recovering in the following days. Additionally, the treated supernatant and biomass extract significantly enhanced root growth in mung beans, highlighting the importance of the treatment method for biostimulation and suggesting potential for improved storage and transportation. Hard-to-reach communities represent Peru's main challenge for malaria elimination, but information about transmission in these areas is scarce. Here, we assessed Plasmodium vivax (Pv) and P. falciparum (Pf) transmission dynamics, resistance markers, and Pf hrp2/3 deletions in Nueva Jerusalén (NJ), a remote, indigenous community in the Peruvian Amazon with high population mobility. We collected samples from November 2019 to May 2020 by active (ACD) and passive case detection (PCD) in NJ. Parasites were identified with microscopy and PCR. Then, we analyzed a representative set of positive-PCR samples (Pv = 68, Pf = 58) using highly-multiplexed deep sequencing assays (AmpliSeq) and compared NJ parasites with ones from other remote Peruvian areas using population genetics indexes. The ACD intervention did not reduce malaria cases in the short term, and persistent malaria transmission was observed (at least one Pv infection was detected in 96% of the study days). In Nueva Jerusalen, the Pv population had modest genetic diversity (He = 0.27). Pf population had lower diversity (He = 0.08) and presented temporal clustering, one of these clusters linked to an outbreak in February 2020. Moreover, Pv and Pf parasites from NJ exhibited variable levels of differentiation (Pv Fst = 0.07–0.52 and Pf Fst = 0.11–0.58) with parasites from other remote areas. No artemisin resistance mutations but chloroquine (57%) and sulfadoxine-pyrimethamine (35–67%) were detected in NJ's Pf parasites. Moreover, pfhrp2/3 gene deletions were common (32–50% of parasites with one or both genes deleted). The persistent Pv transmission and the detection of a Pf outbreak with parasites genetically distinct from the local ones highlight the need for tailored interventions focusing on mobility patterns and imported infections in remote areas to eliminate malaria in the Peruvian Amazon. The green synthesis of silver nanoparticles (AgNPs) can be developed using safe and environmentally friendly routes, can replace potentially toxic chemical methods, and can increase the scale of production. This study aimed to synthesize AgNPs from aqueous extracts of guarana (Paullinia cupana) leaves and flowers, collected in different seasons of the year, as a source of active biomolecules capable of reducing silver ions (Ag+) and promoting the stabilization of colloidal silver (Ag0). The plant aqueous extracts were characterized regarding their metabolic composition by liquid chromatography coupled to high-resolution mass spectrometry (UHPLC-HRMS/MS), phenolic compound content, and antioxidant potential against free radicals. The synthesized AgNPs were characterized by UV/Vis spectrophotometry, dynamic light scattering (DLS), nanoparticle tracking analysis (NTA), transmission electron microscopy (TEM), and scanning electron microscopy coupled to energy-dispersive X-ray spectrometry (EDX). The results demonstrated that the chemical characterization indicated the presence of secondary metabolites of many classes of compounds in the studied aqueous extracts studied, but alkaloids and flavonoids were predominant, which are widely recognized for their antioxidant capabilities. It was possible to notice subtle changes in the properties of the nanostructures depending on parameters such as seasonality and the part of the plant used, with the AgNPs showing surface plasmon resonance bands between 410 and 420 nm using the leaf extract and between 440 and 460 nm when prepared using the flower extract. Overall, the average hydrodynamic diameters of the AgNPs were similar among the samples (61.98 to 101.6 nm). Polydispersity index remained in the range of 0.2 to 0.4, indicating that colloidal stability did not change with storage time. Zeta potential was above −30 mV after one month of analysis, which is adequate for biological applications. TEM images showed AgNPs with diameters between 40.72 to 48.85 nm and particles of different morphologies. EDX indicated silver content by weight between 24.06 and 28.81%. The synthesized AgNPs exhibited antimicrobial efficacy against various pathogenic microorganisms of clinical and environmental interest, with MIC values between 2.12 and 21.25 µg/mL, which is close to those described for MBC values. In this article, maintenance operations were incorporated to the sequence-dependent setup blocking flow shop to minimise total completion time subject to total maintenance costs. A mixed integer linear programming and procedures to incorporate maintenance to job sequence were developed. A constructive heuristic and three metaheuristics, greedy randomised adaptative search procedure (GRASP), discrete artificial bee colony (DABC), variable block insertion heuristic (VBIH) and iterated greedy algorithm (IG), designed to the blocking flow shop with total completion time minimisation were adapted to minimise total maintenance costs and the hierarchical function, respectively.All the methods were applied to solve small and medium and large size instance sets, with respective 1920 and 2200 problems. Experimental results shows that for small size instances set, DABC with α = 20 (DABC(20)) obtained the best trade-off between effectiveness and efficiency. For medium and large size instances set, DABC(20) VBIH with α = 20 (VBIH(20)) generated the best trade-off between quality of solution and computational time. Background: The goal of these experiments was to determine which learning and memory system(s) were necessary for the retention of visual discriminations and subsequent acquisition of a second problem. The dorsal striatum should be involved in the acquisition and expression of this task based on previous work implicating this region in instrumental learning and memory processes. The perirhinal cortex has been implicated in learning and memory processes associated with visual information like objects, and pictures and may also play a role in the acquisition and/or retention of visual discriminations. As there is no clear spatial/relational component to the task, the hippocampus should not be involved. Methods: Rats were trained on a two-choice visual discrimination task to criterion performance after which they received lesions to portions of the dorsal striatum (dorso-medial or dorso-lateral striatum) and medial temporal lobe (perirhinal cortex or hippocampus). After surgical recovery, the rats were tested for retention of the original discrimination, followed by training on a second problem on the same task. Results: The results showed that dorsal medial striatal lesions produced a retrograde deficit on picture discrimination, but dorsal lateral striatum lesions did not. Neither dorsal striatal lesion produced a deficit on acquisition of a second problem. Perirhinal cortex did not seem to make an essential contribution to the retention of the original discrimination or acquisition of the second problem. Surprisingly, subjects with hippocampal damage were severely impaired but eventually re-learned the discrimination. Damage to the hippocampus had no impact on acquisition of a second problem. Conclusions: Taken together, the results of the present experiments show that the dorsomedial striatum and the hippocampus may support performance on this instrumental task if intact during acquisition but is not required for acquisition of a new problem. With a growing emphasis on developing transverse competencies in students, many educational institutions are trying to revolutionize the traditional system of education. One among others is TEC de Monterrey in Mexico which refines its educational models so that the students can be nourished with the skills that prepare them not only for the present but for years to come. Among skill development transverse competencies are considered a key to the successful career development of a student that emphasizes on critical thinking aspect. So mainly the the educational models try to inculcate or enhance the critical thinking of students. Towards this goal, TEC introduced the TEC21 model that is oriented towards critical thinking skill development of the students. The current work in this regard utilizes real-time student data collected using the eOpen instrument and takes into consideration experts' opinions to analyze the TEC21 model. The determinate factors are taken from the TEC21 model and indeterminate factors are identified from the literature. As real-time data especially educational data contain much uncertain, indeterminate, and unknown information, therefore this research introduces the use of Neutrosophic Logic to address the uncertainty of the data. Through the use of neutrosophy, we have shown how indeterminate factors when considered for analysis give a better representation of information. We have also compared neutrosophic cognitive maps with used earlier fuzzy cognitive maps to show their effectiveness in this regard. Overall system developed using NCM gives a better analysis of educational models like TEC21. This is the only work now that has utilized neutrosophy to understand and analyze the TEC21 model. A significant degree of uncertainty surrounds security events in Mexico because of the interaction of multiple socioeconomic and political elements. Conventional probabilistic and logical approaches frequently fall short of properly handling this uncertainty. To effectively navigate and manage security incidents in Mexico, this study suggests applying neutrosophic logic, an extension of classical logic that allows for indeterminacy and partial knowledge. Neutrosophic logic provides a more adaptable framework for making decisions in uncertain contexts by simultaneously handling true, false, and indeterminate values. In this study, we have created a neutrosophic-based model that includes metrics like social unrest, economic instability, and crime rates to provide a thorough evaluation of the security environment. When the model is put to the test using actual data from different parts of Mexico, it outperforms conventional techniques in terms of prediction and interpretation of security occurrences. The findings demonstrate how neutrosophic logic helps security analysts and policymakers make more thoughtful and nuanced judgments by offering insightful information about the ambiguous nature of security occurrences. The ability to foresee and address possible risks is much improved when neutrosophic logic is incorporated into security event analysis. Background Young children and older adults are susceptible for invasive pneumococcal disease (IPD) caused by Streptococcus pneumoniae. Pneumococcal protein-specific antibodies play a protective role against IPD; however, not much is known about the pace of acquisition, maturation, and maintenance of these antibodies throughout life. Methods Immunoglobulin G (IgG) and IgA levels, avidity, and/or specificity to the pneumococcal proteome in serum and saliva from healthy young children, adults, and older adults, with known carriage status, were measured by enzyme-linked immunosorbent assay (ELISA) and 2-dimensional western blotting against ΔcpsTIGR4. Results Eleven-month-old children, the youngest age group tested, had the lowest pneumococcal proteome-specific IgG and IgA levels and avidity in serum and saliva, followed by 24-month-old children and were further elevated in adult groups. Among adult groups, the parents had the highest serum and saliva IgG and IgA antibody levels. In children, antibody levels and avidity correlated with daycare attendance and presence of siblings, posing as proxy for exposure and immunization. Immunodominance patterns slightly varied throughout life. Conclusions Humoral immunity against the pneumococcal proteome is acquired through multiple episodes of pneumococcal exposure. Low-level and low-avidity antiproteome antibody profiles in young children may contribute to their IPD susceptibility, while in overall antiproteome antibody-proficient older adults other factors likely play a role. Polyoxyalkylenes are widely studied as clay swelling inhibitors. Although many additives are available, the operations require more efficient formulations. This work synthesized and characterized two of molecules: poly(ethylene glycol)(PEG) modified with hydrocarbon chains of different lengths; and poly(amidoamine) dendrimers(PAMAM) and PEG-modified PAMAM. The clay swelling inhibition efficiency was evaluated by the Foster swelling test. For the hydrophobized PEG derivatives, there is an optimum hydrocarbon chain size (C6-C10) to achieve the best performance. The best result was obtained for PEG-modified PAMAM(1:1), tested for the first time, being its performance even better than a commercial additive widely used in the petroleum industry. Background: Trypanosoma cruzi is mainly transmitted to mammals by vectors, but other transmission routes exist. Opossums can harbor the infectious form of the parasite in their anal glands, underscoring their potential role in non-vectorial transmission. In Mexico, Didelphis virginiana and D. marsupialis are the main opossum species. While T. cruzi has been detected in the anal gland secretions of various opossum species, their infectivity has only been confirmed in D. marsupialis and D. albiventris. Vertical transmission has also been proposed in D. virginiana. However, if this occurs in opossums, it remains unclear whether it happens during pregnancy or lactation. Our objective was to investigate the possible contribution of urban opossums to non-vectorial transmission of T. cruzi in the metropolitan area of Merida, Yucatan, in southeastern Mexico. Methodology/principal findings: Blood, anal gland secretions, and milk were collected from opossums captured in Merida, Mexico, all identified as D. virginiana using taxonomic keys and Cytb sequencing. By PCR, T. cruzi was detected in 16/102 opossums (15.69%) in at least one type of sample. The prevalence was 14.71% (15/102) in blood and 0.98% (1/102) in anal gland secretions. 1/22 milk samples (4.55%) tested positive. Blood of 37 offspring from T. cruzi-positive mothers was collected and tested negative. qPCR revealed that females with offspring tended to have lower parasite load in blood compared to females without offspring and males. Genotyping of the parasite through multiplex PCR revealed only the DTU TcI. Conclusions/significance: This study agrees with previous works where D. virginiana was the most abundant opossum species in urban areas in southeastern Mexico and confirms that it is associated with TcI. Detection of T. cruzi in a sample of anal gland secretions underscores the potential risk represented by D. virginiana in non-vectorial transmission in urban areas of southeastern Mexico. Detection in the milk of a lactating female, along with the observed tendency towards a lower parasite load in females with offspring, highlight the importance of further investigating vertical transmission in D. virginiana. Author summary: Opossums are primary reservoirs of T. cruzi, the parasite responsible for Chagas disease, mainly transmitted to mammals by triatomine insects. Besides the vectorial route, opossums, which may have the unique ability among mammals to harbor the infective form of the parasite in their anal glands, may be involved in alternative transmission routes. Additionally, recent findings suggest that Virginia opossums (Didelphis virginiana) may vertically transmit T. cruzi, potentially increasing parasite prevalence within their populations and transmission risk via vectorial and non-vectorial routes. This makes opossums critical for studying T. cruzi transmission mechanisms, especially non-vector pathways. While T. cruzi has been detected in anal glands of various opossum species, transmission through gland secretions has only been confirmed in Didelphis marsupialis and D. albiventris. Moreover, if vertical transmission occurs in opossums, the mechanism remains unclear. We assessed the presence of T. cruzi in blood, anal gland secretions, and milk sampled in urban opossums from the metropolitan area of Merida, Yucatan, in southeastern Mexico. Detection of the parasite in anal gland secretions and milk suggests possible non-vectorial transmission routes. Absence of infection in offspring from infected opossum mothers, along with the observed tendency towards a lower parasite load in females with offspring, suggests that parasite load may constrain vertical transmission. Biothiols, such as cysteine (Cys), glutathione (GSH), and homocysteine (Hcy), play crucial roles in various physiological processes and serve as biomarkers for oxidative stress and redox homeostasis. Their structural similarities, however, pose significant challenges in selective detection and quantification, limiting the availability of suitable probes. Here, we report the design and synthesis of a novel ratiometric fluorescent sensor based on a seleno-BODIPY (Se-BODIPY) derivative, enabling rapid discrimination and quantification of Cys, Hcy, and GSH with low detection limits (Cys = 0.8 μM, Hcy = 20.4 μM, and GSH = 35.9 μM) via fluorescence. The probe exhibits high selectivity towards these biothiols over 11 amino acids, operating through dual-mode detection (absorption and emission spectra) with a visible color change from blue to orange (Cys/Hcy) or pink (GSH) in a turn-on fluorescence process. Notably, the distinct reaction mechanisms between Se-BODIPY and GSH versus Cys/Hcy lead to a more prominent blue shift for Cys/Hcy, facilitating their differentiation. Kinetic studies further differentiate Cys from Hcy, with the BODIPY reacting much faster with Cys than the latter. The effectiveness of the sensor was demonstrated in quantifying biothiols in urine samples, providing a non-invasive method with high recovery rates. Additionally, its incorporation into paper strips allows detection of biothiols in water samples via visible and UV light-induced color changes, indicating its potential for solid-state detection without organic solvents. Background: Anaplastic Large Cell Lymphoma (ALCL) is a rare and aggressive T-cell lymphoma, classified into ALK-positive and ALK-negative subtypes, based on the presence of chromosomal translocations involving the ALK gene. The current standard of treatment for ALCL is polychemotherapy, with a high overall survival rate. However, a subset of patients does not respond to or develops resistance to these therapies, posing a serious challenge for clinicians. Recent targeted treatments such as ALK kinase inhibitors and anti-CD30 antibody-drug conjugates have shown promise but, for a fraction of patients, the prognosis is still unsatisfactory. Methods: We investigated the genetic landscape of ALK + ALCL by whole-exome sequencing; recurring mutations were characterized in vitro and in vivo using transduced ALCL cellular models. Results: Recurrent mutations in FAT family genes and the transcription factor RUNX1T1 were found. These mutations induced changes in ALCL cells morphology, growth, and migration, shedding light on potential factors contributing to treatment resistance. In particular, FAT4 silencing in ALCL cells activated the β-catenin and YAP1 pathways, which play crucial roles in tumor growth, and conferred resistance to chemotherapy. Furthermore, STAT1 and STAT3 were hyper-activated in these cells. Gene expression profiling showed global changes in pathways related to cell adhesion, cytoskeletal organization, and oncogenic signaling. Notably, FAT mutations associated with poor outcome in patients. As a wide variety of intelligent technologies become part of everyday life, researchers have explored how people conceptualize agents that in some ways act and think like living things but are clearly machines. Much of this work draws upon the idea that people readily default to generalizing human-like properties to such agents, and only pare back on these generalizations with added thought. However, recent findings have also documented that people are sometimes initially hesitant to attribute minds to a machine but are more willing to do so with additional thought. In the current experiments, we hypothesized that these attribution-increasing reconsiderations could be spurred by situation-induced cognitive dissonance. In two experiments, participants completed a belief activation exercise designed to induce cognitive dissonance (writing arguments for or against prominent beliefs), viewed a video of an ambiguously intentional robot, and completed measures of cognitive dissonance. In both experiments, cognitive dissonance was associated with increased attributions of mind to the robot. Our findings provide evidence that people sometimes increase their attributions of minds when experiencing cognitive conflict, but also that activation of change-inducing concepts may impact attributions of a mind without producing conscious cognitive conflict in participants. Introduction: The main obstacle to achieving an HIV-1 cure is the proviral reservoir. To promote equity in HIV cure strategies, it is crucial to study the viral reservoir of the predominant HIV-1 subtype C in both women and men. Therefore, we investigated the dynamics of the (intact) viral reservoir in relation to plasma viral load (VL), CD4+ T cell count, and immune activation before and during 96 weeks of successful antiretroviral therapy (ART). Methods: Eighty-two participants (62% female) newly initiating ART in a rural clinic in South Africa were included in the study. Blood samples were collected at baseline, week 48, and week 96, and CD4 count was determined. Plasma was used for VL and immune marker analyses, while isolated peripheral blood mononuclear cells (PBMCs) were used for the quantification of cellular multiple spliced HIV-1 RNA (msRNA) and the intact proviral DNA assay. For the longitudinal analyses on ART, we selected only those participants who durably suppressed their VL to <200 copies/mL during 48 (n=65) and/or 96 (n=60) weeks of treatment. Results: At ART initiation, the median CD4 count was 234 cells/mm3 and VL was 68,897 copies/mL. Interestingly, at baseline the number of defective proviruses was significantly correlated with VL (p<0.0001), msRNA (p<0.0001), CD4 count (p=0.0008), CXCL10 (p=0.0003) and TNF-α (p=0.0394). During successful ART, a significant decrease of both the intact and defective proviral reservoir was observed (p<0.0001). The decrease of the intact proviral reservoir was more profound compared to the defective fraction after 96 weeks of therapy. In addition, a significant decrease in cellular msRNA and IL-6, IL-7, TNF-α, sCD14, sCD163, CCL2, CXCL10, and CRP was detected. Discussion: This study underscores the significant relationship observed prior to therapy initiation between the number of defective proviruses, viral transcription/production and their association with immune response indicators such as CD4 count, CXCL10, and TNF-α. Furthermore, the observation of a less pronounced decrease of the defective proviral DNA highlights the importance of addressing both intact and defective proviruses in therapeutic strategies to enhance clinical outcomes for people with HIV-1. Nature Positive is a concept and approach that focuses on restoring and enhancing nature to improve biodiversity, ecosystem health, and nature's benefits to humanity. While the Nature Positive movement is gaining significant momentum, with 90 countries currently signed on, achieving its ambitious vision will require engagement and contributions from all sectors of society. Notably, both governments and the private sector will need to align and embrace transformative change. A comprehensive understanding and implementation of the mitigation hierarchy as a foundation is a first step. This will require commitment, regulation, incentives and actions to both halt the drivers of biodiversity loss and support appropriate biodiversity restoration and protection along supply chains. While there is guidance on reporting, disclosure and target setting through frameworks such as Science-Based Targets Network (SBTN) and the Taskforce on Nature-Related Financial Disclosures (TNFD), there is a need for a clear pathway for companies, investors and policy makers for achieving Nature Positive. In this paper, we review and describe how the Nature Positive concept intersects with global and national-scale policy instruments and identify a set of key principles which can support companies on a Nature Positive pathway. Focusing initially on corporate action we explore how the mining sector can test the model for transforming company operations to achieve nature positive outcomes. We chose mining given its importance to the global economy, its impact on land, and its burgeoning role in the energy transition to a low carbon economy through the provision of critical minerals. The value of the contribution to this transition will be undermined if mining activities exacerbate the loss of biodiversity. We illustrate a conceptual approach that can guide the mining sector, including some key metrics that can be used to track and communicate progress toward nature positive goals. The mining sector has been testing and implementing a range of approaches such as No Net Loss, Net Positive Impact, biodiversity offsets, and lender standards over the last two decades. This provides an excellent foundation upon which to build nature positive ambition and outcomes. Ultimately, the guidance for Nature Positive can be adapted and replicated across other sectors and provide policy makers with the appropriate proof-points that can align on regulation so that robust business practices can drive Nature Positive outcomes with benefits for both people and nature. Author summary: The mining sector is poised to play a key role in the energy transition through the provision of critical minerals. This is leading to concerns over the impacts on nature that could result from increased global mining activity. It is important to consider such risks and create systems that can not only reduce the sector's impact on nature, but also foster its contribution to Nature Positive outcomes. Achieving success will depend on company commitments to mitigate and compensate their impacts on nature and then going beyond those commitments to invest in landscape-level conservation actions. This must also be accompanied by efforts to imbed nature in business decision making as well as in building more nature positive supply chains. The role of governments is essential in this process. Governments need to establish the enabling policies as well as the conservation targets to which companies can contribute and work closely with the sector to achieve the desired outcomes. A hitherto unknown modification of I2O5 was obtained from high‐pressure/high‐temperature syntheses in a Walker‐type multianvil device at 8 GPa and 250 °C. HP‐I2O5 crystallizes in the monoclinic crystal system with space group P21/c (no. 14) with the unit cell parameters a=12.0612(3) Å, b=4.8613(2) Å, c=6.9585(2) Å, β=100.10(1)° (at 173 K), and four formula units per cell. The single‐crystal structure data are accompanied by powder X‐ray diffraction data at ambient and elevated temperatures. Furthermore, DFT calculations were carried out to investigate the phase transition between the ambient‐pressure polymorph NP‐I2O5 to the newly synthesized high‐pressure phase. [ABSTRACT FROM AUTHOR]
