Abstract: We prepared the sirolimus liposome by rapid expansion of supercritical solution (RESS) technology of supercritical fluid, and studied the effects of temperature, pressure, and equilibrium time on the average particle size and envelop rate of liposome. The conditions of the minimal average particle size of liposome were 328K of temperature (35MPa), 35MPa of pressure (333K), and 50 minutes of equilibrium (343K) time, respectively. The conditions of the maximal envelop rate of liposome were 333K of temperature (30MPa), 35MPa of pressure (343K), and 50 minutes of equilibrium (323K) time, respectively. [ABSTRACT FROM AUTHOR]

To prepare timozolomide liposomes for administration through nasal mucous membrane, we studied the factors of the preparation of the liposomes. The timozolomide liposomes were prepared by the ammonium sulphate gradient method; electroscopy and laser particle analyzer were utilized to determine the conformation, size and distribution of timozolomide liposomes; high performance liquid chromatography (HPLC) was applied to determine the entrapping efficiency of timozolomide liposomes; then we studied the influences of the concentration of ammonium sulphate solution, temperature, and the drug-to-lipid ratio on the entrapping efficiency. The average size of timozolomide liposomes was 185 nm; the entrapping efficiency was 90.3%. The entrapping efficiency was enhanced with the increasing of the concentration of ammonium sulphate solution and the rising of temperature, and decreased with the increasing of the drug-to-lipid ratio. The timozolomide liposomes with high entrapping efficiency, small and even particle sizes could be prepared by the simple and convenient ammonium sulphate gradient method. The primary influencing factors on the entrapping efficiency of timozolomide liposomes were the concentration of ammonium sulphate solution, the temperature, and the drug-to-lipid ratio. We established a HPLC method for content determination of aniracetam in aniracetam inclusion complex. The chromato column was Agilent ODS (4.6mm×150mm,5µm), the mobile phase was methanol-0.01 mol/L Potassium dihydrogen phosphate buffer solution (25:75, pH 3.0), with the flow rate of 1.0 ml/min, column temperature of 30° and the detection wave at 280 nm, the sample size was 20µL. A good linear relationship was obtained between the peak areas and the concentrations of aniracetam in the range from 5~80µg/ml (r=0.9998), the mean recovery was 100.1% (n=15), RSD=0.19%. This method is convenient, rapid, accurate, and brings about good recovery; it can be used for content determination of aniracetam in aniracetam inclusion complex. [ABSTRACT FROM AUTHOR]
