Nasal spray treatments that inhibit the Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) entry into nose and nasopharynx at early stages can be an appropriate approach to stop or delay the progression of the disease. We performed a prospective, randomized, double-blind, placebo-controlled, parallel-group, multicentric, phase II clinical trial comparing the rate of hospitalization due to COVID-19 infection between azelastine 0.1% nasal spray and placebo nasal spray treatment groups. The study furthermore assessed the reduction in virus load in SARS-CoV-2-infected subjects estimated via quantitative reverse transcriptase polymerase chain reaction (RT-PCR) using nasopharyngeal swabs in both groups during the treatment period. A total of 294 subjects with mild COVID-19 infection were screened and randomized in a 1:1 ratio. There was no incidence of COVID-19-related hospitalization in either treatment group. Mean virus load was significantly reduced in both groups during the 11 treatment days as compared with baseline viral load values. The reduction in virus load in the azelastine 0.1% nasal spray group was significantly higher than the reduction in the placebo group at day 11 (log10 5.93 vs. log10 5.85 copies/mL, respectively, p = 0.0041). A total of 39 (32.0%) subjects in the azelastine 0.1% treatment group and 40 (31.0%) subjects in the placebo group reported 48 and 51 adverse events, respectively. The application of distributed fiber optic strain and temperature measurement can be utilized to address a multitude of measurement tasks across a diverse range of fields, particularly in the context of structural health monitoring in the domains of building construction, civil engineering, and special foundation engineering. However, a comprehensive understanding of the influences on the measurement method and the sensors is essential to prevent misinterpretations or measurement deviations. In this context, this study investigated the effects of moisture exposure, including various salt solutions and a high pH value, on a distributed strain measurement using Rayleigh backscattering. Three fiber optic sensors with different coating materials and one uncoated fiber were exposed to five different solutions for 24 h. The study revealed significant discrepancies (∼38%) in deformation between the three coating types depending on the surrounding solution. Furthermore, in contrast to the prevailing literature, which predominantly describes swelling effects, a negative deformation (∼−47 μ ε ) was observed in a magnesium chloride solution. The findings of this study indicate that corresponding effects can impact the precision of measurement, potentially leading to misinterpretations. With the changing epidemiology of COVID-19 and its impact on our daily lives, there is still an unmet need of COVID-19 therapies treating early infections to prevent progression. The current study was a randomized, parallel, double-blind, placebo-controlled trial. Ninety SARS-CoV-2 positive patients were randomized into 3 groups receiving placebo, 0.02% or 0.1% azelastine nasal spray for 11 days, during which viral loads were assessed by quantitative PCR. Investigators assessed patients' status throughout the trial including safety follow-ups (days 16 and 60). Symptoms were documented in patient diaries. Initial viral loads were log10 6.85 ± 1.31 (mean ± SD) copies/mL (ORF 1a/b gene). After treatment, virus load was reduced in all groups (p < 0.0001) but was greater in the 0.1% group compared to placebo (p = 0.007). In a subset of patients (initial Ct < 25) viral load was strongly reduced on day 4 in the 0.1% group compared to placebo (p = 0.005). Negative PCR results appeared earlier and more frequently in the azelastine treated groups: being 18.52% and 21.43% in the 0.1% and 0.02% groups, respectively, compared to 0% for placebo on day 8. Comparable numbers of adverse events occurred in all treatment groups with no safety concerns. The shown effects of azelastine nasal spray may thus be suggestive of azelastine's potential as an antiviral treatment. The histamine-1 receptor antagonist azelastine was recently found to impact SARS-CoV-2 viral kinetics in a Phase 2 clinical trial (CARVIN). Thus, we investigated the relationship between intranasal azelastine administrations and viral load, as well as symptom severity in COVID-19 patients and analyzed the impact of covariates using non-linear mixed-effects modeling. For this, we developed a pharmacokinetic (PK) model for the oral and intranasal administration of azelastine. A one-compartment model with parallel absorption after intranasal administration described the PK best, covering both the intranasal and the gastro-intestinal absorption pathways. For virus kinetic and symptoms modeling, viral load and symptom records were gathered from the CARVIN study that included data of 82 COVID-19 patients receiving placebo or intranasal azelastine. The effect of azelastine on viral load was described by a dose–effect model targeting the virus elimination rate. An extension of the model revealed a relationship between COVID-19 symptoms severity and the number of infected cells. The analysis revealed that the intranasal administration of azelastine led to a faster decline in viral load and symptoms severity compared to placebo. Infectious diseases of the conjunctiva and cornea usually leave behind both broad local and systemic immunity. Case reports of SARS-CoV-2-positive conjunctivitis with subsequent systemic immunity suggest a new route of immunization preventing the primary infection of the airways. Material and Methods: A total of 24 Syrian field hamsters were treated. In systematic animal experiments, we infected the eyes of n = 8 animals (group 1) and the airways of another n = 8 animals (group 2) with SARS-CoV-2 (Wuhan type); n = 8 hamsters served as controls (group 3). The weight development of the animals was recorded. After two weeks of observation of disease symptoms, all animals were re-exposed to SARS-CoV-2 in the respiratory tract (challenge) to determine whether immunity to the virus had been achieved. Results: The epi-ocularly infected animals (group 1) showed no clinically visible disease during the ocular infection phase. At most, there was a slightly reduced weight gain compared to the control group (group 3), while the respiratory infected animals (group 2) all lost weight, became lethargic, and slowly recovered after two weeks. After the challenge, none of the animals in groups 1 and 2 became ill again. The animals in the negative control (group 3) all became ill. Cytotoxic antibodies were detectable in the blood of the infected groups before and after challenge, with higher titers in the epi-ocularly infected animals. Conclusion: By epi-ocular infection with SARS-CoV-2, the development of systemic immunity with formation of cytotoxic antibodies without severe general disease could be observed in the experimental animals, which did not induce any more disease upon a second infection in the respiratory tract. The non‐thermal plasma (NTP) technique was shown to be a method to improve indoor air quality. In particular in kitchens, odorous emissions can be removed by NTP. A combined concept of adsorption of volatile organic compounds (VOCs) and plasma regeneration of the adsorber was tested in adsorption‐regeneration‐adsorption cycles. As reference VOCs, 2‐methylthiophene, 2‐methylpyrazine, 2‐acetylthiazole, nonanal, and trans‐2‐nonenal were selected in humid air streams. These odorous compounds are emitted during cooking and frying processes. The adsorption‐regeneration concept was also tested during a simulated frying process with garlic in rape oil. Ambient ionization mass spectrometry (AI-MS), the ionization of samples under ambient conditions, enables fast and simple analysis of samples without or with little sample preparation. Due to their simple construction and low resource consumption, plasma-based ionization methods in particular are considered ideal for use in mobile analytical devices. However, systematic investigations that have attempted to identify the optimal configuration of a plasma source to achieve the sensitive detection of target molecules are still rare. We therefore used a low-temperature plasma ionization (LTPI) source based on dielectric barrier discharge with helium employed as the process gas to identify the factors that most strongly influence the signal intensity in the mass spectrometry of species formed by plasma ionization. In this study, we investigated several construction-related parameters of the plasma source and found that a low wall thickness of the dielectric, a small outlet spacing, and a short distance between the plasma source and the MS inlet are needed to achieve optimal signal intensity with a process-gas flow rate of as little as 10 mL/min. In conclusion, this type of ion source is especially well suited for downscaling, which is usually required in mobile devices. Thermal pest control with hot air is widely accepted as an alternative to chemical methods. However, it requires relatively long treatment times owing to the low thermal conductivity of wood. Direct dielectric heating that applies radio waves or microwaves has the advantage of more homogeneous heating. However, sound experimental data on this technique are currently rare. Therefore, the thermal treatment of wood-destroying insects with radio waves and microwaves was studied with two model pests, Anobium punctatum and Hylotrupes bajulus, and with Tenebrio molitor as a reference. The secure elimination of pests was achieved, and the corresponding treatment time was in the range of a few minutes. Temperature profiles were more homogeneous when applying radio waves. Dielectric heating with radio waves (RW) and microwaves is an alternative treatment to hazardous chemicals or hot air for pest control. RW can be used to heat large bulky materials homogeneously and smoothly to eliminate embedded pest organisms. Dielectric heating enables selective overheating of certain materials in a heterogeneous mixture. This feature of electromagnetic heating methods, in contrast to techniques based on thermal conduction, is caused by significant differences in the dielectric properties of pest organisms and matrix materials. This study proves unambiguously that selective heating of cylindrical pest organisms in wood matrices occurs under defined conditions. The influence of electrical heating on microbiological processes in soil has been studied to evaluate the potential for enhancing biodegradation of pollutants by controlling the temperature. A frequency of 50 Hz (power line frequency) was applied for resistive heating. Dielectric heating was realized using a frequency of 13.56 MHz (radio frequency). Both techniques were compared with conventional heating in a water bath. For experiments in laboratory and full scale, a model soil and a contaminated original soil were used. It was shown that under conditions capable for heating soil to 35 °C or even 60 °C, soil respiration as a measure for microbial activity was not hindered by electrical heating when temperature and moisture content were comparable with conventional heating. The variations of soil respiration were reversible upon temperature changes. Under certain conditions, periodical fluctuations of microbiological activity were observed. Several possible explanations including chronobiology are discussed without being able to provide an unambiguous interpretation for this effect. [ABSTRACT FROM AUTHOR]

Applying a new experimental design with a capillary glass reactor and plate electrodes outside of the reactor allowed the initiation of discharges in aqueous electrolytes under the influence of a radio-frequency (RF) electromagnetic field. This study focused on the mechanism leading to the initiation of such discharges in the restriction of a glass tube. The light emission correlated with discharges was analysed with optical emission spectroscopy. Electrons with energies between 20 and 45 eV were responsible for the dissociation of water molecules into (excited) OH, H and O radicals. Current-voltage characteristics were measured before and under discharge conditions. Modelling of the experimental setup and simulation of electrical field strength distribution support the hypothesis of the origin of discharges in general and experimental findings such as ring-shaped discharges and a minimum solution conductivity of about 1 S m-1 required for discharge initiation with RF voltages of 2 kV. A new type of water dissociation at ambient pressure initiated by the irradiation of aqueous electrolytes using an electromagnetic field with a frequency of 13.56MHz is described in this study. A special reactor design allows the use of ex situ electrodes to form in situ electrical discharges in water vapour bubbles. The observed formation of molecular hydrogen (H2) and oxygen (O2) combined with the emission of light ('burning water' phenomenon) originates from a non-thermal plasma in water vapour bubbles. Formation rates of hydrogen peroxide (H2 O2), hydrogen (H2), and oxygen (O2) in gliding arc discharge with water spray and a variety of carrier gases, including Ar, He, O2, N2, and CO2, were determined. Significantly larger rates and concentrations of hydrogen peroxide were found in the present reactor with a modified liquid injection nozzle than in previous work. As long as nitrogen oxide species are minimized or not formed, the formation rates of hydrogen peroxide were less sensitive to the carrier gas (O2, CO2, Ar), although He gave about one-half the rate as these other gases. The stoichiometric ratios of H2 : O2 H2O2 exhibited a range of behaviors for the different carrier gases with the highest H2 formation rate in Ar and N2 and the largest O2 formation in CO2. Formation rates of hydrogen peroxide and electrode erosion rates for a range of different electrode materials were determined in a gas–liquid pulsed electrical discharge reactor with a high voltage electrode needle in the liquid and a ground electrode suspended in the gas over the liquid. It was found that the H2O2 formation rates and efficiencies did not depend on the electrode material. Electrode erosion from lowest to highest followed the series: nickel chromium, thoriated tungsten, diamond-coated tungsten, stainless steel, tungsten carbide and tungsten copper alloy. Smooth crater-like morphology was found for nickel chromium, titanium and tungsten carbide and a much finer surface structure with small protrusions for the tungsten, tungsten copper and the copper. Doubling the electrode diameter lead to an increase in the H2O2 formation per eroded length by a factor of three but it also decreased the energy efficiency yield of H2O2 by more than 20%.
