Distinct patterns of circulating microRNAs (miRNAs) were found to be involved in misguided thrombus resolution. Thus, we aimed to investigate dysregulated miRNA signatures during the acute phase of pulmonary embolism (PE) and test their diagnostic and predictive value for future diagnosis of chronic thromboembolic pulmonary hypertension (CTEPH). Microarray screening and subsequent validation in a large patient cohort (n = 177) identified three dysregulated miRNAs as potential biomarkers: circulating miR-29a and miR-720 were significantly upregulated and miR-let7a was significantly downregulated in plasma of patients with PE. In a second validation study equal expression patterns for miR-29a and miR-let7a regarding an acute event of recurrent venous thromboembolism (VTE) or deaths were found. MiR-let7a concentrations significantly correlated with echocardiographic and laboratory parameters indicating right ventricular (RV) dysfunction. Additionally, circulating miR-let7a levels were associated with diagnosis of CTEPH during follow-up. Regarding CTEPH diagnosis, ROC analysis illustrated an AUC of 0.767 (95% CI 0.54–0.99) for miR-let7a. Using logistic regression analysis, a calculated patient-cohort optimized miR-let7a cut-off value derived from ROC analysis of ≥ 11.92 was associated with a 12.8-fold increased risk for CTEPH. Blast disease causes serious economic yield losses in pearl millet. Identification and introgression of genomic regions associated with blast resistance can help to develop resistant cultivars to minimize yield losses incurred from blast outbreaks. In this study, 384 advanced pearl millet genotypes were screened against six blast pathotype‐isolates (major pearl millet growing agro‐ecologies of India), namely, Pg 45, Pg 118, Pg 138, Pg 186, Pg 204 and Pg 232. Analysis of variance showed significant (P <.001) variation among genotypes for blast reaction (susceptible to resistance). ICMR 08111 and ICMR 10888 genotypes showed resistance to all six blast pathotypes. A genome‐wide association study performed with 264,241 single nucleotide polymorphic markers could successfully identify 15 SNPs (P = 1.26 × 10−7 to 9.22 × 10−12) underlying the genomic regions governing blast‐resistance across five different chromosomes. The SNPs reported had a significant association in at least two of the three models tested (GLM, MLM and Farm CPU). Pearl millet is an important cereal crop of semi-arid regions since it is highly nutritious and climate resilient. However, pearl millet is underutilized commercially due to the rapid onset of hydrolytic rancidity of seed lipids post-milling. We investigated the underlying biochemical and molecular mechanisms of rancidity development in the flour from contrasting inbred lines under accelerated aging conditions. The breakdown of storage lipids (triacylglycerols; TAG) was accompanied by free fatty acid accumulation over the time course for all lines. The high rancidity lines had the highest amount of FFA by day 21, suggesting that TAG lipases may be the cause of rancidity. Additionally, the high rancidity lines manifested substantial amounts of volatile aldehyde compounds, which are characteristic products of lipid oxidation. Lipases with expression in seed post-milling were sequenced from low and high rancidity lines. Polymorphisms were identified in two TAG lipase genes (PgTAGLip1 and PgTAGLip2) from the low rancidity line. Expression in a yeast model system confirmed these mutants were non-functional. We provide a direct mechanism to alleviate rancidity in pearl millet flour by identifying mutations in key TAG lipase genes that are associated with low rancidity. Surface radiative fluxes have been derived with the objective of supplementing top-of-atmosphere (TOA) radiative fluxes being measured under NASA's Clouds and the Earth's Radiant Energy System (CERES) project. This has been accomplished by using combinations of CERES TOA measurements, parameterized radiative transfer algorithms, and high-quality meteorological datasets available from reanalysis projects. Current CERES footprint-level products include surface fluxes derived from two shortwave (SW) and three longwave (LW) algorithms designated as SW models A and B and LW models A, B, and C. The SW and LW models A work for clear conditions only; the other models work for both clear and cloudy conditions. The current CERES Edition-4A computed surface fluxes from all models are validated against ground-based flux measurements from high-quality surface networks like the Baseline Surface Radiation Network and NOAA's Surface Radiation Budget Network (SURFRAD). Validation results as systematic and random errors are provided for all models, separately for five different surface types and combined for all surface types as tables and scatterplots. Validation of surface fluxes is now a part of CERES processing and is used to continually improve the above algorithms. Since both models B work for clear and cloudy conditions alike and meet the accuracy requirement, their results are considered to be the most reliable and most likely to be retained for future work. Genomic selection (GS) is an emerging methodology that helps select superior lines among experimental cultivars in plant breeding programs. It offers the opportunity to increase the productivity of cultivars by delivering increased genetic gains and reducing the breeding cycles. This methodology requires inexpensive and sufficiently dense marker information to be successful, and with whole genome sequencing, it has become an important tool in many crops. The recent assembly of the pearl millet genome has made it possible to employ GS models to improve the selection procedure in pearl millet breeding programs. Here, three GS models were implemented and compared using grain yield and dense molecular marker information of pearl millet obtained from two different genotyping platforms (C [conventional GBS RAD-seq] and T [tunable GBS tGBS]). The models were evaluated using three different cross-validation (CV) schemes mimicking real situations that breeders face in breeding programs: CV2 resembles an incomplete field trial, CV1 predicts the performance of untested hybrids, and CV0 predicts the performance of hybrids in unobserved environments. We found that (i) adding phenotypic information of parental inbreds to the calibration sets improved predictive ability, (ii) accounting for genotype-by-environment interaction also increased the performance of the models, and (iii) superior strategies should consider the use of the molecular markers derived from the T platform (tGBS). Background and Aim: Patients with primary sclerosing cholangitis (PSC) are at high risk for the development of cholangiocarcinoma (CC). Analysis of micro ribonucleic acid (MiRNA) patterns is an evolving research field in biliary pathophysiology with potential value in diagnosis and therapy. Our aim was to evaluate miRNA patterns in serum and bile of patients with PSC and/or CC. Methods: Serum and bile from consecutive patients with PSC (n = 40 (serum), n = 52 (bile)), CC (n = 31 (serum), n = 19 (bile)) and patients with CC complicating PSC (PSC/CC) (n = 12 (bile)) were analyzed in a cross-sectional study between 2009 and 2012. As additional control serum samples from healthy individuals were analyzed (n = 12). The miRNA levels in serum and bile were determined with global miRNA profiling and subsequent miRNA-specific polymerase chain reaction-mediated validation. Results: Serum analysis revealed significant differences for miR-1281 (p = 0.001), miR-126 (p = 0.001), miR-26a (p = 0.001), miR-30b (p = 0.001) and miR-122 (p = 0.034) between patients with PSC and patients with CC. All validated miRNAs were significantly lower in healthy individuals. MiR-412 (p = 0.001), miR-640 (p = 0.001), miR-1537 (p = 0.003) and miR-3189 (p = 0.001) were significantly different between patients with PSC and PSC/CC in bile. Conclusions: Patients with PSC and/or CC have distinct miRNA profiles in serum and bile. Aims Osteopontin (OPN) is a multifunctional cytokine critically involved in cardiac fibrosis. However, the underlying mechanisms are unresolved. Non-coding RNAs are powerful regulators of gene expression and thus might mediate this process. Methods and results OPN and miR-21 were significantly increased in cardiac biopsies of patients with myocardial fibrosis. Ang II infusion via osmotic minipumps led to specific miRNA regulations with miR-21 being strongly induced in wild-type (WT) but not OPN knockout (KO) mice. This was associated with enhanced cardiac collagen content, myofibroblast activation, ERK-MAPkinase aswell as AKT signalling pathway activation and a reduced expression of Phosphatase and Tensin Homologue (PTEN) as well as SMAD7 in WT but not OPNKO mice. In contrast, cardiotropic AAV9-mediated overexpression of OPN in vivo further enhanced cardiac fibrosis. In vitro, Ang II induced expression of miR-21 in WT cardiac fibroblasts, while miR-21 levels were unchanged in OPN KO fibroblasts. As pri-miR-21 was also increased by Ang II, we studied potential involved upstream regulators; Electrophoretic Mobility Shift and Chromatin Immunoprecipitation analyses confirmed activation of the miR-21 upstream-transcription factor AP-1 by Ang II. Recombinant OPN directly activated miR-21, enhanced fibrosis, and activated the phosphoinositide 3-kinase pathway. Locked nucleic acid-mediated miR-21 silencing ameliorated cardiac fibrosis development in vivo. Conclusion In cardiac fibrosis related to Ang II, miR-21 is transcriptionally activated and targets PTEN/SMAD7 resulting in increased fibroblast survival. Aims Differentiation of heart failure with reduced ( HFrEF) or preserved ( HFpEF) ejection fraction independent of echocardiography is challenging in the community. Diagnostic strategies based on monitoring circulating microRNA ( miRNA) levels may prove to be of clinical value in the near future. The aim of this study was to identify a novel miRNA signature that could be a useful HF diagnostic tool and provide valuable clinical information on whether a patient has HFrEF or HFpEF. Methods and results MiRNA biomarker discovery was carried out on three patient cohorts, no heart failure (no- HF), HFrEF, and HFpEF, using Taqman miRNA arrays. The top five miRNA candidates were selected based on differential expression in HFpEF and HFrEF ( miR-30c, −146a, −221, −328, and −375), and their expression levels were also different between HF and no- HF. These selected miRNAs were further verified and validated in an independent cohort consisting of 225 patients. The discriminative value of BNP as a HF diagnostic could be improved by use in combination with any of the miRNA candidates alone or in a panel. Combinations of two or more miRNA candidates with BNP had the ability to improve significantly predictive models to distinguish HFpEF from HFrEF compared with using BNP alone (area under the receiver operating characteristic curve >0.82). Conclusion This study has shown for the first time that various miRNA combinations are useful biomarkers for HF, and also in the differentiation of HFpEF from HFrEF. The utility of these biomarker combinations can be altered by inclusion of natriuretic peptide. MiRNA biomarkers may support diagnostic strategies in subpopulations of patients with HF. The Clouds and the Earth's Radiant Energy Systems (CERES) project utilizes radiometric measurements taken aboard the Terra and Aqua spacecrafts to derive the world-class data products needed for climate research. Achieving the exceptional fidelity of the CERES data products, however, requires a considerable amount of processing to assure quality and to verify accuracy and precision, which results in the CERES data being released more than 6 months after the satellite observations. For most climate studies such delays are of little consequence; however, there are a significant number of near-real time uses for CERES data products. The Fast Longwave and Shortwave Radiative Flux (FLASHFlux) data product was therefore developed to provide a rapid release version of the CERES results, which could be made available to the research and applications communities within 1 week of the satellite observations by exchanging some accuracy for speed. FLASHFlux has both achieved this 1-week processing objective and demonstrated the ability to provide remarkably good agreement when compared with the CERES data products for both the instantaneous single-scanner footprint (SSF) fluxes and the time- and space-averaged (TISA) fluxes. This paper describes the methods used to expedite the production of the FLASHFlux SSF fluxes by utilizing data from the CERES and Moderate Resolution Imaging Spectroradiometer instruments, as well as other meteorological sources. This paper also reports on the validation of the FLASHFlux SSF results against ground-truth measurements and the intercomparison of FLASHFlux and CERES SSF results. Crosses were performed to introgress genes for productivity and other desirable traits from ricebean ( Vigna umbellata) into black gram ( Vigna mungo). Crossability was very poor in black gram × ricebean crosses, and only two to nine true hybrid plants were obtained. Plant fertility was very poor in initial generations, but was improved gradually from F2 onwards. Twenty-four uniform progenies, bulked in F7, were evaluated for yield potential. The percentage increase/decrease in yield ranged from −35.48 to 50.31 over the check cultivar (' Mash338', female parent). All the progenies were found resistant to Mungbean yellow mosaic virus, Cercospora leaf spot and Bacterial leaf spot diseases. Overall, it was found that desirable traits such as high pod number, seed weight, productivity and resistance to diseases have been introgressed successfully into black gram from ricebean. A derivative line, KUG114, recorded yield superiority of 39.45% over the check cultivar ' Mash338' on the average of 14 multilocation research trials. An improvement was developed and tested for surface longwave flux algorithms used in the Clouds and the Earth’s Radiant Energy System processing based on lessons learned during the validation of global results of those algorithms. The algorithms involved showed significant overestimation of downward longwave flux for certain regions, especially dry–arid regions during hot times of the day. The primary cause of this overestimation was identified and the algorithms were modified to (i) detect meteorological conditions that would produce an overestimation, and (ii) apply a correction when the overestimation occurred. The application of this correction largely eliminated the positive bias that was observed in earlier validation studies. The Clouds and the Earth’s Radiant Energy System (CERES) project uses two shortwave (SW) and two longwave (LW) algorithms to derive surface radiative fluxes on an instantaneous footprint basis from a combination of top-of-atmosphere fluxes, ancillary meteorological data, and retrieved cloud properties. Since the CERES project examines the radiative forcings and feedbacks for Earth’s entire climate system, validation of these models for a wide variety of surface conditions is paramount. The present validation effort focuses upon the ability of these surface-only flux algorithms to produce accurate CERES Edition 2B single scanner footprint data from the Terra and Aqua spacecraft measurements. To facilitate the validation process, high-quality radiometric surface observations have been acquired that were coincident with the CERES-derived surface fluxes. For both SW models, systematic errors range from -20 to -12 W m-2 (from -2.8% to -1.6%) for global clear-sky cases, while for the all-sky SW model, the systematic errors range from 14 to 21 W m-2 (3.2%–4.8%) for global cloudy-sky cases. Larger systematic errors were seen for the individual surface types, and significant random errors where observed, especially for cloudy-sky cases. While the SW models nearly achieved the 20 W m-2 accuracy requirements established for climate research, further improvements are warranted. For the clear-sky LW model, systematic errors were observed to fall within ±5.4 W m-2 (±1.9%) except for the polar case in which systematic errors on the order from -15 to -11 W m-2 (from -13% to -7.2%) occurred. For the all-sky LW model, systematic errors were less than ±9.2 W m-2 (±7.6%) for both the clear-sky and cloudy-sky cases. The random errors were less than 17 W m-2 (6.2%) for clear-sky cases and 28 W m-2 (13%) for cloudy-sky cases, except for the desert cases in which very high surface skin temperatures caused an overestimation in the model-calculated surface fluxes. The annual cycles of surface shortwave flux are investigated using the 8-yr dataset of the surface radiation budget (SRB) components for the period July 1983–June 1991. These components include the downward, upward, and net shortwave radiant fluxes at the earth's surface. The seasonal cycles are quantified in terms of principal components that describe the temporal variations and empirical orthogonal functions (EOFs) that describe the spatial patterns. The major part of the variation is simply due to the variation of the insolation at the top of the atmosphere, especially for the first term, which describes 92.4% of the variance for the downward shortwave flux. However, for the second term, which describes 4.1% of the variance, the effect of clouds is quite important and the effect of clouds dominates the third term, which describes 2.4% of the variance. To a large degree the second and third terms are due to the response of clouds to the annual cycle of solar forcing. For net shortwave flux at the surface, similar variances are described by each term. The regional values of the EOFs are related to climate classes, thereby defining the range of annual cycles of shortwave radiation for each climate class. [ABSTRACT FROM AUTHOR]

Parameterized shortwave and longwave algorithms developed at the Langley Research Center have been used to derive surface radiative fluxes in the processing of the Clouds and the Earth's Radiant Energy System (CERES) data obtained from flight aboard the Tropical Rainfall Measuring Mission (TRMM) satellite. Retrieved fluxes were validated on an instantaneous-footprint basis using coincident surface measurements obtained from the Atmospheric Radiation Measurement (ARM) program's Southern Great Plains (SGP) central facility, the ARM/SGP network of extended facilities, and a number of surface sites of the Baseline Surface Radiation Network (BSRN) and the Climate Monitoring and Diagnostics Laboratory (CMDL). Validation was carried out separately for clear-sky and all-sky conditions. For the shortwave, systematic errors varied from -12 to 10 W m-2 for clear skies and from -5 to 35 W m-2 for all-sky conditions. Random errors varied from 20 to 40 W m-2 for clear skies but were much larger (45–85 W m-2) for all-sky conditions. For the longwave, systematic errors were comparatively small for both clear-sky and all-sky conditions (0 to -10 W m-2) and random errors were within about 20 W m-2. In general, comparisons with surface data from the ARM/SGP site (especially the central facility) showed the best agreement. Large systematic errors in shortwave comparisons for some sites were related to flaws in the surface measurements. Larger errors in longwave fluxes for some footprints were found to be related to the errors in cloud mask retrievals, mostly during the nighttime. Smaller longwave errors related to potential errors in the operational analysis products used in satellite retrievals were also found. Clouds and the Earth's Radiant Energy System (CERES) investigates the critical role that clouds and aerosols play in modulating the radiative energy flow within the Earth-atmosphere system. CERES builds upon the foundation laid by previous missions, such as the Earth Radiation Budget Experiment, to provide highly accurate top-of-atmosphere (TOA) radiative fluxes together with coincident cloud and aerosol properties inferred from high-resolution imager measurements. This paper describes the method used to construct empirical angular distribution models (ADMs) for estimating shortwave, longwave, and window TOA radiative fluxes from CERES radiance measurements on board the Tropical Rainfall Measuring Mission satellite. To construct the ADMs, multiangle CERES measurements are combined with coincident high-resolution Visible Infrared Scanner measurements and meteorological parameters from the European Centre for Medium-Range Weather Forecasts data assimilation product. The ADMs are stratified by scene types defined by parameters that have a strong influence on the angular dependence of Earth's radiation field at the TOA. The surface radiation budget of a region is strongly tied to its climate. An 8-yr climatology of surface radiation budget components for 2.5° regions over the earth is examined in order to learn how the regional climate and surface radiation are related. The yearly cycles of a few individual regions were studied by plotting monthly mean net longwave flux as a function of net shortwave flux at the surface. These plots show trajectories that are characteristic of the climate class. The behavior of the trajectories of surface radiation and their relation to the regional climate can be understood with simple conceptual models for many cases. From an examination of these trajectories, a set of parameters is developed, such as mean net longwave flux and range of net shortwave flux, which distinguish various climate classes on the basis of the surface radiation. These criteria are applied to produce a map of regional climate classes based on surface radiation, similar to those of Koeppen or Trewartha and Horn, which were based on vegetation, temperature, and precipitation. Climatological averages of surface radiation budget parameters, namely, the shortwave and longwave surface radiative fluxes, have been derived for each month of the year on a global scale. These climatological averages were derived from an 8-yr (96 month) time series of monthly average fluxes. The monthly averages were computed using fast radiation parameterizations and satellite data from the International Satellite Cloud Climatology Project and the Earth Radiation Budget Experiment. Results are presented as time series of hemispheric and global averages and as geographical distributions and time--latitude cross sections of climatological averages. The spatial/temporal variabilities of the results were found to be clearly related to the corresponding variabilities of meteorological and other inputs to the parameterizations. Numerous comparisons of the present results were made with available surface measurements for the purpose of validation. In most cases, the differences were found to be within the uncertainties of the measurements. In some cases, where they were large, the differences were attributable to identifiable deficiencies in the meteorological inputs and/or the surface measurements. However, large differences remained unexplained in a few cases. Anomalies of shortwave and longwave surface fluxes during the 1986/87 El Niño--Southern Oscillation episode show a strong relationship with corresponding top-of-atmosphere anomalies derived from an independent data source. Comparisons with results from several general circulation models showed large differences, but, in most cases, these were attributable to well-recognized deficiencies in model simulations. Global annual average downward and net shortwave fluxes were found to be about 185 and 161 W m 22 , respectively. An extensive study has been carried out to validate a satellite technique for estimating downward longwave radiation at the surface. The technique, mostly developed earlier, uses operational sun-synchronous satellite data and a radiative transfer model to provide the surface flux estimates. The satellite-derived fluxes were compared directly with corresponding ground-measured fluxes at four different sites in the United States for a common one-year data period. This provided a study of seasonal variations as well as a diversity of meteorological conditions. Dome heating errors in the ground-measured fluxes were also investigated and were corrected prior to the comparisons. Comparison of the monthly averaged fluxes from the satellite and ground sources for all four sites for the entire year showed a correlation coefficient of 0.98 and a standard error of estimate of 10 W m−2. With 1 table Abstract Foliar blast, caused by Pyricularia grisea (Cooke) Sacc, has recently emerged as a serious disease of pearl millet in India. To study the inheritance of resistance to this disease, two resistant restorer lines (ICMR 06222 and ICMR 07555) and two susceptible maintainer lines (ICMB 95444 and ICMB 89111) were selected on the basis of foliar blast reaction in tests conducted under field and greenhouse conditions. Each of the two resistant parents was crossed with two susceptible parents to generate four sets of F1s, F2s and their backcrosses with both resistant and susceptible parental lines. These were evaluated for disease reaction with artificial inoculation under both field and greenhouse conditions. The disease reaction of the F1s, and the segregation patterns of resistance in the F2s and backcross generations, showed that resistance to foliar blast in pearl millet is controlled by a single dominant gene. With continued population growth, increasing staple crop production is necessary. However, in dryland areas, this is negatively affected by various abiotic stresses, such as drought and salinity. The field screening of 10 improved genetic lines of pear millet originating from African dryland areas was conducted based on a set of agrobiological traits (i.e., germination rate, plant density, plant maturity rate, forage, and grain yields) in order to understand plant growth and its yield potential responses under saline environments. Our findings demonstrated that genotype had a significant impact on the accumulation of green biomass (64.4% based on two-way ANOVA), while salinity caused reduction in grain yield value. HHVBC Tall and IP 19586 were selected as the best-performing and high-yielding genotypes. HHVBC Tall is a dual purpose (i.e., forage and grain) line which produced high grain yields on marginal lands, with soil salinization up to electrical conductivity (EC) 6–8 dS m−1 (approximately 60–80 mM NaCl). Meanwhile, IP 19586, grown under similar conditions, showed a rapid accumulation of green biomass with a significant decrease in grain yield. Both lines were tolerant to drought and sensitive to high salinity (above 200 mM NaCl). The threshold salinity of HHVBC Tall calculated at the seedling stage was lower than that of IP 19586. Seedling viability of these lines was affected by oxidative stress and membrane peroxidation, and they had decreased chlorophyll and carotenoid biosynthesis. This study demonstrated that ionic stress is more detrimental for the accumulation of green and dry biomass, in combination with increasing the proline and malonic dialdehyde (MDA) contents of both best-performing pearl millet lines, as compared with osmotic stress.
