Calcium is an element widely used in the development of biomaterials for bone tissue engineering as it plays important roles in bone metabolism and blood coagulation. The Ca ions can condition the microenvironment at the tissue–material interface, affecting the protein deposition process and cell responses. The aim of this study was to analyze the changes in the patterns of protein adsorption on the silica hybrid biomaterials supplemented with different amounts of CaCl2, which can function as release vehicles. This characterization was carried out by incubating the Ca-biomaterials with human serum. LC–MS/MS analysis was used to characterize the adsorbed protein layers and compile a list of proteins whose affinity for the surfaces might depend on the CaCl2 content. The attachment of pro- and anti-clotting proteins, such as THRB, ANT3, and PROC, increased significantly on the Ca-materials. Similarly, VTNC and APOE, proteins directly involved on osteogenic processes, attached preferentially to these surfaces. To assess correlations with the proteomic data, these formulations were tested in vitro regarding their osteogenic and inflammatory potential, employing MC3T3-E1 and RAW 264.7 cell lines, respectively. Twitter is one of the most popular social media networks that, in recent years, has been increasingly used by researchers as a platform to share science and discuss ongoing work. Despite its popularity, Twitter is not commonly used as a medium to teach science. Here, we summarize the results of #EUROmicroMOOC: the first worldwide Microbiology Massive Open Online Course taught in English using Twitter. Content analytics indicated that more than 3 million users saw posts with the hashtag #EUROmicroMOOC, which resulted in over 42 million Twitter impressions worldwide. These analyses demonstrate that free Microbiology MOOCs shared on Twitter are valuable educational tools that reach broad audiences throughout the world. We also describe our experience teaching an entire Microbiology course using Twitter and provide recommendations when using social media to communicate science to a broad audience. Abstract: The prime objectives in the development of biomaterials for dental applications are to improve the quality of osseointegration and to short the time needed to achieve it. Design of implants nowadays involves changes in the surface characteristics to obtain a good cellular response. Incorporating osteoinductive elements is one way to achieve the best regeneration possible post-implantation. This study examined the osteointegrative potential of two distinct biomaterials: sandblasted acid-etched titanium and a silica sol-gel hybrid coating, 70% MTMOS-30% TEOS. In vitro, in vivo, and proteomic characterisations of the two materials were conducted. Enhanced expression levels of ALP and IL-6 in the MC3T3-E1 cells cultured with coated discs, suggest that growing cells on such surfaces may increase mineralisation levels. 70M30T-coated implants showed improved bone growth in vivo compared to uncoated titanium. Complete osseointegration was achieved on both. However, coated implants displayed osteoinductive properties, while uncoated implants demonstrated osteoconductive characteristics. Coagulation-related proteins attached predominantly to SAE-Ti surface. Surface properties of the material might drive the regenerative process of the affected tissue. Analysis of the proteins on the coated dental implant showed that few proteins specifically attached to its surface, possibly indicating that its osteoinductive properties depend on the silicon delivery from the implant.Graphical abstract: [ABSTRACT FROM AUTHOR]

Abstract: In most minimally‐invasive procedures used to address severe pain arising from compression fractures of the vertebral bodies, such as percutaneous vertebroplasty (PVP), a poly(methyl methacrylate) (PMMA) bone cement is used. Shortcomings of this type of cement, such as high exotherm temperature and lack of bioactivity, are well known. We prepared different formulations of a composite bone cement, whose solid constituents consisted of PMMA beads and particles of a bioactive glass (BG), where 0–20%(w/w) of the calcium component was substituted by strontium. The difference between the formulations was in the relative amounts of the solid phase constituents and in the Sr‐content of BG. We determined the influence of the mixture of solid phase constituents of the cement formulation on a collection of properties, such as maximum exotherm temperature (Tmax), setting time (tset), and injectability (I). The selection of the PMMA beads was crucial to obtain cement composite formulations capable to be efficiently injected. Results allowed to select nine solid phase mixtures to be further tested. Then, we determined the influence of the composition of these composite bone cements on Tmax, tset, I, and cell proliferation. The results showed that the performance of various of the selected composite cements was better than that of PMMA cement reference, with lower Tmax, lower tset, and higher I. We found that incorporation of Sr‐substituted BGs into these materials bestows bioactivity properties associated with the role of Sr in bone formation, leading to some composite cement formulations that may be suitable for use in PVP. © 2017 Wiley Periodicals, Inc. The interactions of implanted biomaterials with the host organism determine the success or failure of an implantation. Normally, their biocompatibility is assessed usingin vitrotests. Unfortunately,in vitroandin vivoresults are not always concordant; new, effective methods of biomaterial characterisation are urgently needed to predict thein vivooutcome. As the first layer of proteins adsorbed onto the biomaterial surfaces might condition the host response, mass spectrometry analysis was performed to characterise these proteins. Four distinct hybrid sol-gel biomaterials were tested. Thein vitroresults were similar for all the materials examined here. However,in vivo, the materials behaved differently. Six of the 171 adsorbed proteins were significantly more abundant on the materials with weak biocompatibility; these proteins are associated with the complement pathway. Surface engineering of biomaterials could promote the osseointegration of implants. In this work, two types of hybrid sol-gel materials were developed to stimulate cell attachment, proliferation and differentiation of osteogenic cells. One type was synthesised from vinyl triethoxysilane (VTES) and tetraethyl-orthosilicate (TEOS) at different molar ratios, while the other from VTES and hydroxyapatite particles (HAp). Hybrid materials were systematically investigated using nuclear magnetic resonance, Fourier transform infrared spectroscopy and contact angle metrology. The biocompatibility and osseoinduction of the coatings were evaluated by measuring mesenchymal stem cell proliferation using MTT assays and analysing the mineralised extracellular matrix production by quantifying calcium-rich deposits. The results highlighted the versatility of these coatings in obtaining different properties by changing the molar ratio of the VTES:TEOS precursors. Thus, mineralisation was stimulated by increasing TEOS content, while the addition of HAp improved cell proliferation but worsened mineralisation. [ABSTRACT FROM AUTHOR]

The controlled release of active agents from a matrix has become increasingly important for oral, transdermal or implantable therapeutic systems, due to the advantages of safety, efficacy and patient convenience. Controlled-release hybrid (organic-inorganic) sol-gel coating synthesis has been performed to create a sol with an active molecule included (procaine). Synthesis procedures included acid-catalysed hydrolysis, sol preparation, the addition of a procaine solution to the sol, and the subsequent gelation and drying. The alkoxide precursors used were triethoxyvinylsilane and tetraethyl-orthosilicate (TEOS) in molar ratios of 1:0, 9:1, 8:2 and 7:3. After the determination of the optimal synthesis parameters, the material was physicochemically characterised by silicon-29 nuclear magnetic resonance (Si-NMR) and Fourier transform infrared spectroscopy, contact angle analysis and electrochemical impedance spectroscopy tests. Finally, the materials were assayed in vitro for their ability to degrade by hydrolysis and to release procaine in a controlled manner. The sustained release of procaine over a 3-day period was demonstrated. A close correlation between release and degradation rates suggests that film degradation is the main mechanism underlying the control of release. Electrochemical analysis reveals the formation of pores and water uptake during the degradation. The aim of this work was to estimate the intestinal bioaccessibility of polyphenols and antioxidant capacity of cactus pear using an in vitro gastrointestinal model. The amount of polyphenols released from the food matrix by the action of digestive enzymes was about 69-83% in pulp and 0-14% in seeds. The rest of polyphenols was associated with indigestible compounds conferring antioxidant capacity. The results suggest that these compounds could reach the colon and induce healthy effects. Dietary polyphenols are substrates for colonic microbiota. They and their metabolites contribute to the maintenance of gastrointestinal health by interacting with epithelial cells, and largely by modulating the gut microbial composition. Polyphenols may act as promoting factors of growth, proliferation, or survival for beneficial gut bacteria-mainly Lactobacillus strains-and thus, exerting prebiotic actions and inhibiting the proliferation of some pathogenic bacteria such as Salmonella and Helicobacter pylori species. To date the interactions affecting metabolic pathways and numerous metabolites of dietary polyphenols have been widely documented. However, the effects of dietary polyphenols on the modulation of the intestinal ecology and on the growth of gut microbial species are still poorly understood. This paper summarizes data on the influence of dietary polyphenols on gut microbiota and the main interactions between dietary polyphenols and beneficial and pathogenic intestinal bacteria. The utilization of injectable acrylic bone cement is crucial to the outcome of vertebroplasty and kyphoplasty. However, only a few cements that are in clinical use today are formulated specifically for use in these procedures and even these formulations are not regarded as “ideal” injectable bone cements. The aim of this work is to prepare bioactive bone cements by adding strontium hydroxyapatite (SrHA) to a cement formulation based on polymethylmethacrylate. Thus, the cement combines the immediate mechanical support given by the setting of the acrylic matrix with optimum radiopacity and bioactivity due to the incorporation of the SrHA. Formulations of bioactive cement were prepared with 10 and 20 wt% of SrHA as synthesised and after a surface treatment with the monomer. Cements loaded with treated particles showed an enhancement of their handling properties, and hence, an improvement on their rheological behaviour, injectabilities and compressive parameters. Abstract: Thirty-seven Brucella reference and field strains representing all the species and their biovars were analysed by PCR–RFLP to determine the degree of variation in the genes encoding the new members of group 3 outer membrane protein (Omp) family. Analysis of the omp22 and omp25c/omp25d genes indicated that the restriction patterns were identical for all species and biovars with all restriction enzymes tested, except for Brucella ovis that showed a short 30bp deletion close to omp22 gene, and for B. abortus biovar 6 and B. ovis that lacked a DdeI site and a HinfI site, respectively, in the omp25c/omp25d genes. Analysis of PCR products of the omp31b gene digested with 20 restriction enzymes revealed that this gene has a greater level of DNA polymorphism than the other genes encoding the new members of group 3 Omp family. A deletion of 232bp was detected in fourteen B melitensis strains from different hosts and from different geographic origins, confirming that this feature is indeed a hallmark of B. melitensis. PCR–RFLP analysis of omp31b with DdeI allowed us to identify species-specific markers for B. abortus, B. melitensis, and B. ovis. Finally, by PCR analysis, Southern blot hybridization and DNA sequencing we showed that a large deletion of 15kb, comprising the entire omp25b gene and 21 more genes, is present in all B. ovis strains, thus confirming previous observations from other authors. OBJECTIVE:: To compare the intakes of dietary fibre (DF) and dietary indigestible fraction (DIF) in the Spanish diet and also to show the limitations of DF values for nutritional and epidemiological studies. DESIGN:: This includes the following: (i) estimation of plant foods consumption in Spanish diet from national food consumption data obtained from annual surveys (6000 households, 700 hotels and restaurants and 200 institutions); (ii) determination of DIF content in plant foods using analytical methods that mimic physiological conditions; (iii) estimation of daily intakes of DIF and DF in the Spanish diet. RESULTS:: DIF intake in the Spanish population was estimated at 41.5?g/person/day. DF intake (18.3?g/person/day) was considerably lower than the amount of carbohydrate necessary to maintain the daily bacterial cell turnover in large intestine (‘carbohydrate gap’). The differences between DIF and DF values are a consequence of conceptual and methodological aspects. DIF, as the part of foods reaching the colon, comprises not only DF but also other indigestible compounds such as a fraction of resistant starch, protein, polyphenols and other associated compounds. Analytical conditions are closer to actual physiological conditions in the determination of DIF than in DF analysis. CONCLUSIONS:: DF intake underestimates a major part of the dietary substrates that enter the colon. DIF intake more closely matches the amount of substrates needed to maintain a typical human colonic microflora. The objective of this study was to improve the wear behaviour of acrylic bone cements by substituting the conventional inorganic radiopaque agents (BaSO4 ZrO2) for different iodinated radiopaque monomers which can co-polymerize with the methyl methacrylate monomer, MMA. Hydrophilic matrices are an interesting option when developing drug delivery systems. With this aim, hydroxypropyl methacrylate was grafted onto hydroxypropyl starch and hydroxypropyl cellulose substrates by following the Ce(IV) redox initiation method. Different amounts of ethyleneglycol dimethacrylate, 7 and 34 mol%, as the crosslinking monomer, were also added. The drying of grafted products was carried out by lyophilization, obtaining white powders. Reaction yields (percent grafting, grafting efficiency, etc.) and some physical characteristics of the powders (particle size, moisture uptake, density, morphology, etc.) were determined. These parameters indicate how useful these products may be as potential matrices for direct compressed tablets. In this light, the powder flowability and the binding properties of each copolymer were determined. The graft copolymers can be considered of great interest as direct compression excipients. Due to their different chemical structure and composition, they showed differences in viscoelastic properties that revealed an interesting range of possibilities for use in drug delivery formulations. -amylase resistant corn starches were in vitro fermented (72 h) using cecal content of rat as inoculum. Resistant starches were prepared from native (NCS), retrograded (RSA ), and retrograded and boiled (RSB ) corn starch by treatment with α-amylase to remove digestible starch. Short-chain fatty acid (SCFA) production, molar ratios (acetate: propionate: butyrate), fermentability with respect to lactulose and dry matter disappearance were measured. SCFA production was significantly correlated with non-fermented residue (P <0.001). The three starches showed a similar fermentability, although the rate of SCFA production and disappearance of substrate were slower for retrograded starches. NCS was fermented early while RSA and RSB prolonged their degradation to 72 h. There were slight differences in the molar ratios of acetate, propionate and butyrate. Structural changes in the materials were observed by scanning electron microscopy (SEM). In summary, commonly utilised domestic cooking and technological treatments modified the in vitro fermentative characteristics of resistant corn starches probably due to structural changes. This paper describes the rheological behavior of starch and cellulose acrylic graft copolymers synthesized with the aim of obtaining controlled-release excipients. The rheological characteristics determine the final release properties of matrix tablets. The study of the storage and loss moduli (G′ and G″, respectively) and the viscosity allowed us to know if the polymer behavior was that of a gel and, hence, if it could act as a barrier to drug diffusion. Since dynamic measurements showed a storage modulus higher than the loss modulus, we assessed that all the polymers were gels. Thus, knowing that all the graft copolymers had acceptable properties for compression, the release of theophylline as a model drug at different pH was studied. The activities of a short therapeutic regimen with azithromycin and the classic treatment doxycycline with streptomycin were compared and evaluated in mice infected with Brucella melitensis. In a chronic model, starting therapy 31 days after challenge, azithromycin (10 days, 50 mg/kg/day) significantly reduced the infection (2.9 logs, day 48 post-infection). The effectiveness of doxycycline (21 days, 50 mg/kg/12 hourly) was greater than azithromycin (4.1 logs of reduction, day 48 post-infection), and when doxycycline was administered for a period of 45 days, all the animals were bacteriologically cured from day 78. The combination with streptomycin (14 days, 10 mg/kg/day) did not improve the effect of any of the regimens. In an acute model infection, treatments with doxycycline or doxycycline-streptomycin, for a period of 3 days, starting 1 day after lethal challenge, were able to protect all the mice. In contrast, only 50% of the mice treated with azithromycin survived the challenge. In conclusion, although a short oral treatment with azithromycin was able to reduce the infection significantly, it was not able to cure the animals as effectively as the classic regimen with doxycycline administered for a longer period of time. A total of 29 strains of Brucella abortus, B. melitensis, B. suis, B. ovis, and B. neotomae were examined for growth and catechol production in a semisynthetic low-iron medium. All strains showed reduced growth yields and, quantitatively, production of catechols varied widely among the different strains with no relationship to species, biotypes, or serotypes of Brucella. No clear correlation between catechol production and growth under iron-limiting conditions was observed. The major catechol was identified as 2,3-dihydroxybenzoic acid, and neither other iron-regulated catechols nor hydroxamate type compounds were detected when representative strains of B. abortus or B. melitensis were grown in tryptic soy broth in the presence of iron-sequestering agents.
