Various biophysical techniques have been extensively employed to study protein aggregation due to its significance. Traditionally, these methods detect aggregation at micrometer length scales and micromolar concentrations. However, unlike in vitro, protein aggregation typically occurs at nanomolar concentrations in vivo. Here, using fluorescence correlation spectroscopy (FCS), we captured bromelain aggregation at concentrations as low as ~20 nM, surpassing the detection limit of traditional methods like thioflavin T fluorescence, scattering, and fluorescence microscopy by more than one order of magnitude. Moreover, using thioflavin T fluorescence‐based FCS, we have detected larger aggregates at higher bromelain concentrations, which is undetectable in FCS otherwise. Importantly, our study reveals inherent heterogeneity in bromelain aggregation, inaccessible to ensemble‐averaged techniques. The presented report may provide a platform for the characterization of premature aggregates at very low protein concentrations, which are thought to be functionally significant species in protein aggregation‐induced diseases. This paper aims to understand the changing dynamics of internal labour market segmentation intertwined with the history of working-class struggle in an industrial belt in India's National Capital Region (NCR). Nuances of workers' class struggle in the automobile sector in Delhi-NCR over the last 15 years have shown that unity between permanent and contract workers was key to several successful struggles against worsening work conditions and management control within the global value chains of automobile production. Our fieldwork findings, along with direct and indirect political engagement in the region, reveal that increasing salaries and allotment of supervisory roles to small sections of permanent workers, coupled with the simultaneous fragmentation a lower strata of workers into multiple categories with very low salaries, benefits and heavy workloads, have been significant attempts by capital to break class solidarity among different sections of workers. Relatively pro-labour legislation and trade unions tend to concentrate in high-wage zones, while low-wage and precarious employment remains unprotected by such institutional mechanisms. Labour segmentation is inherent to capitalist exploitation, guarded by institutional mechanisms. Advanced mechanisation reduces distinction of skill requirements between highly paid permanent and lowly paid contract workers. Among benign neoplasms of the salivary gland, malignancies are less common. One of the rarest tumours found in the oral cavity is oncocytoma, comprising less than 2% of all oral neoplasms and carrying a 1% risk of malignant transformation. Oncocytoma is rare among benign neoplasms, typically affecting females and impacting the major salivary glands. Literature discussing the cytodiagnosis of oncocytoma through Fine Needle Aspiration Cytology (FNAC) is even scarcer. Cytomorphological features indicating a predominantly oncocytic population with benign nuclear characteristics suggest the diagnosis of oncocytoma. The presence of oncocytes alongside other cell types in cytology smears suggests oncocytosis or a tumour-like presentation of Warthin's tumour. Due to its unique features and clinical presentation, clinicians may misdiagnose this tumour as pleomorphic adenoma, haemangioma, or another form of oncocytosis due to its rarity and clinical features. Therefore, only histopathological examination can provide a definitive diagnosis. The present report discusses two unusual cytodiagnoses of oncocytoma identified through FNAC of the parotid gland in a 50-year-old female and a 70-year-old male, along with their clinicoradiological findings and subsequent tumour excision. In recent years, there has been phenomenal growth in internet usage worldwide, with a substantial proportion of children and adolescents actively engaging with online platforms. While the internet presents numerous opportunities for children and adolescents, the lack of digital literacy and adequate online safety measures exposes them to various cybercrimes, including cyberbullying, cyberstalking, identity theft, and sexual predation. Moreover, there is growing concern regarding internet addiction among this population. To investigate the determinants of internet addiction among adolescents, we conducted a cross-sectional study in peri-urban Delhi-NCR, India. We used a self-administered questionnaire to gather information on internet usage, and 630 adolescents aged 13–18 participated in the study, also completing an Internet Addiction Test. The findings indicate that 415 adolescents (65.9 %) exhibited no signs of internet addiction, suggesting a healthy relationship with the internet. However, 215 adolescents (33.1 %) displayed symptoms of internet addiction. Among those exhibiting internet addictions, 159 (74.0 %) were classified as mild internet addicts, indicating moderate levels of internet usage. Furthermore, 56 (26.0 %) adolescents were classified as moderate internet addicts, reflecting a higher level of internet addiction. Our study highlights the significant influence of various factors, including family dynamics, environmental factors, and personal experiences, on internet addiction among adolescents. Based on these findings, we propose implementing measures at different levels to foster responsible internet use among adolescents, thereby substantially reducing internet addiction. Pollution by end-of-life electronics is a rapid ever-increasing threat and is a universal concern with production of million metric tons of these wastes per annum. Electronic wastes (E-waste) are rejected electric or electronic equipment which have no other applications. The aggrandized unproper land filling of E-waste may generate hazardous effects on living organisms and ecosystem. At present, millions of tons of E-waste await the advancement of more efficient and worthwhile recycling techniques. Recovery of base and critical elements from electronic scraps will not only reduce the mining of these elements from natural resources but also reduces the contamination caused by the hazardous chemicals (mostly organic micropollutants) released from these wastes when unproperly disposed of. Bioleaching is reported to be the most eco-friendly process for metal recycling from spent electronic goods. A detailed investigation of microbial biodiversity and a molecular understanding of the metabolic pathways of bioleaching microorganisms will play a vital function in extraction of valuable minerals from the end-of-life scraps. Bioleaching technique as an economic and green technology costs around 7 USD per kg for effective reusing of E-waste as compared to other physical and chemical techniques. This review provides a summary of worldwide scenario of electronic pollutants; generation, composition and hazardous components of electronic waste; recycling of valuable elements through bioleaching; mechanism of bioleaching; microorganisms involved in base and critical element recovery from E-waste; commercial bioleaching operations; and upcoming aspects of this eco-friendly technique. There is a pressing need to identify recent directions in the field of aptamer-based sensing. DNA aptamers that are synthetically generated by in vitro selection mechanisms using the SELEX technique are single-stranded oligonucleotides which are selected to bind to a target with favorable sensitivity and selectivity. These aptamers have attracted significant attention due to their high binding affinity and ability to be easily engineered and provide various detection modes in what are known as aptasensors. Our aim is to focus on specialized detection strategies that have gained less attention but are of vital importance, such as optical detection in live cells, fluorescence polarization sensing, multi-analyte detection, colorimetric bioassays, wavelength shifting, and electrochemical-based detection. This will provide us with a perspective to facilitate developments in aptasensor technology for various targets, promising a bright future for biological receptors in the field of biosensing. Introduction: Breast cancer is the most common malignant tumour and the leading cause of cancer-related death in women is breast cancer. The Immunohistochemistry (IHC) method utilised antigens and antibodies to interact to identify cellular or tissue constituents (antigens). This research has been employed as a diagnostic tool for specific cancers. When Programmed Cell Death Ligand 1 (PD-L1) binds to Programmed Cell Death 1 (PD-1), it suppresses the cellular immune response by killing and depleting T-cells. Monoclonal antibodies that block the PD-1/PD-L1 pathway have shown promise as a treatment strategy currently being tested in human cancer trials. Need for the study: Breast cancer is a global issue, and PDL1 expression is emerging as a promising biomarker for breast cancer prognosis. It can provide valuable information treatment planning. Aim: The current study aims to examine the expression of PD-1 and PD-L1 in breast cancer using IHC in all subgroups of breast cancer patients. Both tests can serve as a biomarkers to guide immunotherapeutic interventions, improving prognosis, and correlating with other clinicopathological individual parameters such as age, tumour size, distant metastasis, lymph node involvement, Estrogen Receptor (ER) and Progesterone Receptor (PR) status, Her2neu expression, histological type, and TNM stage. Materials and Methods: This will be a two-year cohort study conducted in the Department of Pathology, Jawaharlal Nehru Medical College (JNMC), Maharashtra, India. The study will include 70 specimens from all cases with a histopathological diagnosis of breast cancer. The Nottingham variant of the Bloom-Richardson Grading System will be used to determine the histological grade of the tumour, and immunostaining for PD-1 and PD-L1 will be performed to evaluate their protein expression. Mission-critical applications refer to the real-time applications, which require fast and secure service provisioning, such as defense sector and disaster management. This paper proposes a delay-aware and secure service provisioning model for such types of applications. As a use-case, we have considered the defense sector, which is a vital sector for a country's all-round well-being including security, safety, society, and economy. In the conventional sensor-cloud model, the sensor data is stored and processed in the cloud. However, the sensor nodes have small coverage and the use of the long distant cloud servers increases the delay. Therefore, the conventional sensor-cloud model may not be efficient for defense application. Moreover, data hiding for security purposes is another important aspect of this field. To address these challenges, this paper proposes a mobility-aware sensor-fog paradigm for mission-critical applications based on network coding and steganography, referred to as Mobi-Sense. In Mobi-Sense, steganography is used for hiding the data during transmission. The theoretical results demonstrate that Mobi-Sense outperforms the existing frameworks with respect to delay and power consumption by ∼ (40 - 80) %. The simulation results present that Mobi-Sense reduces the delay by ∼ (18 - 40) % than the conventional sensor-cloud framework for mission-critical applications. An optimal path finding algorithm based on deep learning has been deployed in the context of disaster scenario. The experimental analysis shows that the proposed optimal path finding method achieves precision and accuracy above 90%. The chaperone Hsp104, a member of the Hsp100/Clp family of translocases, prevents fibril formation of a variety of amyloidogenic peptides in a paradoxically substoichiometric manner. To understand the mechanism whereby Hsp104 inhibits fibril formation, we probed the interaction of Hsp104 with the Alzheimer's amyloid-ß42 (Aβ42) peptide using a variety of biophysical techniques. Hsp104 is highly effective at suppressing the formation of Thioflavin T (ThT) reactive mature fibrils that are readily observed by atomic force (AFM) and electron (EM) microscopies. Quantitative kinetic analysis and global fitting was performed on serially recorded 1H-15N correlation spectra to monitor the disappearance of Aβ42 monomers during the course of aggregation over a wide range of Hsp104 concentrations. Under the conditions employed (50 µM Aβ42 at 20 ℃), Aβ42 aggregation occurs by a branching mechanism: an irreversible on-pathway leading to mature fibrils that entails primary and secondary nucleation and saturating elongation; and a reversible off-pathway to form nonfibrillar oligomers, unreactive to ThT and too large to be observed directly by NMR, but too small to be visualized by AFM or EM. Hsp104 binds reversibly with nanomolar affinity to sparsely populated Aβ42 nuclei present in nanomolar concentrations, generated by primary and secondary nucleation, thereby completely inhibiting on-pathway fibril formation at substoichiometric ratios of Hsp104 to Aβ42 monomers. Tight binding to sparsely populated nuclei likely constitutes a general mechanism for substoichiometric inhibition of fibrillization by a variety of chaperones. Background Electroconvulsive seizure therapy is often used in both treatment-resistant and geriatric depression. However, preclinical studies identifying targets of chronic electroconvulsive seizure (ECS) are predominantly focused on animal models in young adulthood. Given that putative transcriptional, neurogenic, and neuroplastic mechanisms implicated in the behavioral effects of chronic ECS themselves exhibit age-dependent modulation, it remains unknown whether the molecular and cellular targets of chronic ECS vary with age. Methods We subjected young adult (2–3 months) and middle-aged (12–13 months), male Sprague Dawley rats to sham or chronic ECS and assessed for despair-like behavior, hippocampal gene expression, hippocampal neurogenesis, and neuroplastic changes in the extracellular matrix, reelin, and perineuronal net numbers. Results Chronic ECS reduced despair-like behavior at both ages, accompanied by overlapping and unique changes in activity-dependent and trophic factor gene expression. Although chronic ECS had a similar impact on quiescent neural progenitor numbers at both ages, the eventual increase in hippocampal progenitor proliferation was substantially higher in young adulthood. We noted a decline in reelin⁺ cell numbers following chronic ECS only in young adulthood. In contrast, an age-invariant, robust dissolution of perineuronal net numbers that encapsulate parvalbumin⁺ neurons in the hippocampus were observed following chronic ECS. Conclusion Our findings indicate that age is a key variable in determining the nature of chronic ECS-evoked molecular and cellular changes in the hippocampus. This raises the intriguing possibility that chronic ECS may recruit distinct, as well as overlapping, mechanisms to drive antidepressant-like behavioral changes in an age-dependent manner. Several global health incidents and evidences show the increasing likelihood of pandemics (large‐scale outbreaks of infectious disease), which has adversely affected all aspects of human lives. It is essential to develop an analytics framework by extracting and incorporating the knowledge of heterogeneous data‐sources to deliver insights for enhancing preparedness to combat the pandemic. Specifically, human mobility, travel history, and other transport statistics have significantly impact on the spread of any infectious disease. This article proposes a spatio‐temporal knowledge mining framework, named STOPPAGE, to model the impact of human mobility and other contextual information over the large geographic areas in different temporal scales. The framework has two key modules: (i) spatio‐temporal data and computing infrastructure using fog/edge based architecture; and (ii) spatio‐temporal data analytics module to efficiently extract knowledge from heterogeneous data sources. We created a pandemic‐knowledge graph to discover correlations among mobility information and disease spread, a deep learning architecture to predict the next hotspot zones. Further, we provide necessary support in home‐health monitoring utilizing Femtolet and fog/edge based solutions. The experimental evaluations on real‐life datasets related to COVID‐19 in India illustrate the efficacy of the proposed methods. STOPPAGE outperforms the existing works and baseline methods in terms of accuracy by ≈$$ \approx $$(18–21)% in predicting hotspots and reduces the power consumption of the smartphone significantly. The scalability study yields that the STOPPAGE framework is flexible enough to analyze a huge amount of spatio‐temporal datasets and reduces the delay in predicting health status compared to the existing studies. With the enormous growth of wireless technology, and location acquisition techniques, a huge amount of spatio-temporal traces are being accumulated. This dataset facilitates varied location-aware services and helps to take real-life decisions. Efficiently handling and processing spatio-temporal queries are necessary to respond in real-time. Processing the vast spatio-temporal data requires scalable computing infrastructure. In this regard, an efficient query resolution system can be deployed if we predict the infrastructure requirement of the user query apriori along with the identification of the geospatial service chain. In this work, we propose a framework, namely LYRIC (deadLine and budget aware spatio-temporal querY pRocessing In Cloud), where the spatio-temporal queries are resolved efficiently considering user-defined deadline and budget constraint. Our framework shows high deadline completion accuracy in the range of 1.0 - 0.937, which is more accurate than SparkGIS, GeoSpark, GeoMesa and JUST. This also reduces the resource prediction error by 11 percent, considering the geospatial service chain than without it. The cost of the spatio-temporal query is reduced by $\approx$ ≈ 23% in LYRIC, further, the simulation study (using CloudSim) illustrates the efficacy and scalability of LYRIC in terms of optimal budget usage and execution time compared to four baseline approaches. Antibiotic resistance of Mycobacterium tuberculosis is exclusively a consequence of chromosomal mutations. Translesion synthesis (TLS) is a widely conserved mechanism of DNA damage tolerance and mutagenesis, executed by translesion polymerases such as DinBs. In mycobacteria, DnaE2 is the only known agent of TLS and the role of DinB polymerases is unknown. Here we demonstrate that, when overexpressed, DinB1 promotes missense mutations conferring resistance to rifampicin, with a mutational signature distinct from that of DnaE2, and abets insertion and deletion frameshift mutagenesis in homo-oligonucleotide runs. DinB1 is the primary mediator of spontaneous −1 frameshift mutations in homo-oligonucleotide runs whereas DnaE2 and DinBs are redundant in DNA damage-induced −1 frameshift mutagenesis. These results highlight DinB1 and DnaE2 as drivers of mycobacterial genome diversification with relevance to antimicrobial resistance and host adaptation. This manuscript elucidates new mechanisms of mutagenesis in mycobacteria by implicating two translesion DNA polymerases in genome diversification, including creating the mutations that underlie all antibiotic resistance in these global pathogens. Internet of Things (IoT) has a pivotal role in developing intelligent and computational solutions to facilitate varied real‐life applications. To execute high‐end computations and data analytics, IoT and cloud‐based solutions play the most significant role. However, frequent communication with long distant cloud servers is not a delay‐aware and energy‐efficient solution while providing time‐critical applications such as healthcare. This article explores the possibilities and opportunities of integrating cloud technology with fog and edge‐based computing to provide healthcare services to users in exigency. Here, we propose an end‐to‐end framework named RESCUE (enabling green healthcare services using integrated iot‐edge‐fog‐cloud computing environments), consisting efficient spatio‐temporal data analytics module for efficient information sharing, spatio‐temporal data analysis to predict the path for users to reach the destination (healthcare center or relief camps) with minimum delay in the time of exigency (say, natural disaster). This module analyzes the collected information through crowd‐sourcing and assists the user by extracting optimal path postdisaster when many regions are nonreachable. Our work is different from the existing literature in varied aspects: it analyses the context and semantics by augmenting real‐time volunteered geographical information (VGI) and refines it. Furthermore, the novel path prediction module incorporates such VGI instances and predicts routes in emergencies avoiding all possible risks. Also, the design of development of a latency‐aware, power‐aware data‐driven analytics system helps to resolve any spatio‐temporal query more efficiently compared to the existing works for any time‐critical application. This article discusses the prediction of cohesiveness of a group of people in images. The cohesiveness of a group is an essential indicator of the emotional state, structure, and success of the group. We study the factors that influence the perception of group-level cohesion and propose methods for estimating the human-perceived cohesion on the group cohesiveness scale. To identify the visual cues (attributes) for cohesion, we conducted a user survey. Image analysis is performed at a group-level via a multi-task convolutional neural network. A capsule network is explored for analyzing the contribution of facial expressions of the group members on predicting the Group Cohesion Score (GCS). We add GCS to the Group Affect database and propose the ‘GAF-Cohesion database’. The proposed model performs well on the database and achieves near human-level performance in predicting a group's cohesion score. It is interesting to note that group cohesion as an attribute, when jointly trained for group-level emotion prediction, helps in increasing the performance for the later task. This suggests that group-level emotion and cohesion are correlated. Probiotics are potential health-promoting microorganisms representing a significant health aid with diverse pharmaceutical applications. But its role in modulating the human gut microbiome necessities a profound analysis. Undoubtedly, the human gut is a reservoir of infinite species such as bacteria, viruses, fungi, etc. But host with chronic illness bids enrichment of the pathogenic strain which may alter the composition gut microbiome. Culture-based techniques are providing unclear taxonomic data where the possibility of the discrepancy is very high. Therefore the use of next-generation sequencing (NGS) analysis helps to explore depth understanding of the curd microbiome and can generate big genomic data. The present investigation aims to explore the microbial diversity of curd samples from different local dairy farms of Odisha, India using a metagenomic approach. The data read comprising 304,725 sequences totaling 148,546,295 base pairs were interpreted. Taxonomical distribution at different levels unfolded the absolute abundance of phylum, class, order, family, genus, and species within each microbial community. The most dominant species found to be Lactobacillus at order level is 49%, whereas phyla proteobacteria and firmicutes have shown taxonomic percentage abundance of 50.01% and 48.77%, respectively. However, nearly 26.41% of unclassified species from the Lactococcus genus were identified in the sample at different taxonomic levels. Automatic chronic pain assessment and pain intensity estimation has been attracting growing attention due to its widespread applications. One of the prevalent issues in automatic pain analysis is inadequate balanced expert-labelled data for pain estimation. This work proposes an anomaly detection based network addressing one of the existing limitations of automatic pain assessment. The evaluation of the network is performed on pain intensity estimation and protective behaviour estimation tasks from body movements in the EmoPain Challenge dataset. The EmoPain dataset consists of body part based sensor data for both the tasks. The proposed network, PLAAN (Pain Level Assessment with Anomaly-detection based Network), is a lightweight LSTM-DNN network which considers features based on sensor data as the input and predicts intensity level of pain and presence or absence of protective behaviour in chronic low back pain patients. Joint training considering body movement patterns, such as exercise type, corresponding to pain exhibition as a label improves the performance of the network. However, contrary to perception, protective behaviour rather exists sporadically alongside pain in the EmoPain dataset. This induces yet another complication in accurate estimation of protective behaviour. This problem is resolved by incorporating anomaly detection in the network. A detailed comparison of different networks with varied features is outlined in the paper, presenting a significant improvement with the final proposed anomaly detection based network. There has been alarming depletion of manganese (Mn) reserves owing to the ongoing extensive mining operations for catering the massive industrial demand of this element. Moreover, the mining operations have been leading to the generation of Mn-rich waste, thereby contaminating both terrestrial and aquatic bodies. The current scenario necessitates the development of alternative processes for bioremediation as well as economic recovery of Mn from mining wastes. The present investigation aims to report the bioleaching of Mn by Lysinibacillus sp. from mining waste residues in the context of mine waste remediation. Results confirmed that the native isolate had a high Mn biosolubilization potential with a solubilizing efficiency of 84% at the end of a 21-day study under optimized conditions of pulp density 2% (< 150-μm particle size), pH 6.5, and temperature 30 °C. Fourier transform infrared spectroscopy (FTIR) studies followed by liquid chromatography mass spectrometry (LC-MS) analysis were used to ascertain the change in microbial protein conformation, configuration, and protein identification. The results revealed the expression of heat shock proteins (HSP) from the family HSP which is predominantly expressed in bacteria during stress conditions. This study represents the application of native bacterial strain in Mn biosolubilization. We foresee the utility of proteomics-based studies to provide a methodological framework to the underlying mechanism of metal solubilization, thereby facilitating the two-tier benefit of recovery of Mn from alternative sources as well as bioremediation of waste having high manganese content. This paper proposes a mobile healthcare framework based on edge-fog-cloud collaborative network. It uses edge and fog devices for parameterized health monitoring, and cloud for further health data analysis in case of abnormal health status. The continuous location change of users is a critical issue, and the connection interruption and delay in delivering health related data may be fatal in case of emergency. In this direction, in the proposed framework, mobility information of the users is considered and the users' mobility pattern detection is performed inside the cloud for advising the user regarding nearby health centre. From the theoretical analysis, it is observed that the proposed framework reduces the delay and energy consumption of user device by ∼ 28 % and ∼ 27 % respectively than the cloud only health care model. The proposed healthcare framework has been implemented in the laboratory and health data of few student volunteers are analyzed to predict their health status. Pulsed electron paramagnetic resonance (EPR) based distance measurements using the recently developed Cu2+-DPA label present a promising strategy for measuring DNA backbone distance constraints. Herein we develop force field parameters for Cu2+-DPA in order to understand the features of this label at an atomic level. We perform molecular dynamics (MD) simulations using the force field parameters of Cu2+-DPA on four different DNA duplexes. The distance between the Cu2+ centers, extracted from the 2 μs MD trajectories, agrees well with the experimental distance for all the duplexes. Further analyses of the trajectory provide insight into the orientation of the Cu2+-DPA inside the duplex that leads to such agreement with experiments. The MD results also illustrate the ability of the Cu2+-DPA to report on the DNA backbone distance constraints. Furthermore, measurement of fluctuations of individual residues showed that the flexibility of Cu2+-DPA in a DNA depends on the position of the label in the duplex, and a 2 μs MD simulation is not sufficient to fully capture the experimental distribution in some cases. Finally, the MD trajectories were utilized to understand the key aspects of the double electron electron resonance (DEER) results. The lack of orientational selectivity effects of the Cu2+-DPA at Q-band frequency is rationalized in terms of fluctuations in the Cu2+ coordination environment and rotameric fluctuations of the label linker. Overall, a combination of EPR and MD simulations based on the Cu2+-DPA labelling strategy can contribute towards understanding changes in DNA backbone conformations during protein–DNA interactions. This paper presents a study of the response of FRET based DNA aptasensors in the intracellular environment. Herein, we extend previous studies of aptasensors functioning in the extracellular environment to detection of antigens in the intracellular environment. An essential step in this research is the use of a novel means of achieving the endocytosis of aptasensors. Plumbagin derived from the plant Plumbago indica, known as Chitrak in India, is an example of a medicinal compound used traditionally to cure a variety of ailments. Previous reports have indicated that it can inhibit the growth of Mycobacterium tuberculosis (Mtb), the causative agent of the deadly disease TB. In this investigation, we provide an insight into its mode of action. We show here that a significant mycobacterial target that is inhibited by plumbagin is the enzyme ThyX, a form of thymidylate synthase, that is responsible for the synthesis of dTMP from dUMP in various bacterial pathogens, including Mtb. Using a purified preparation of the recombinant version of Mtb ThyX, we demonstrate that plumbagin, a 2,4 napthoquinone, but not lawsone, a structurally related medicinal compound, inhibits its activity in vitro. We also show that the intracellular [dTTP]/[dATP] ratio in Mycobacterium smegmatis (Msm) cells decrease upon treatment with plumbagin, and this, in turn, leads to cell death. Such a conclusion is supported by the observation that over-expression of thyx in the plumbagin treated Msm cells leads to the restoration of viability. The interactions between proteins and their specific DNA sequences are the basis of many cellular processes. Hence, developing methods to build an atomic level picture of these interactions helps improve our understanding of key cellular mechanisms. CueR is an Escherichia coli copper‐sensing transcription regulator. The inhibition of copper‐sensing transcription regulators can kill pathogens, without harming the host. Several spectroscopic studies and crystallographic data have suggested that changes in the conformation of both the DNA and the protein control transcription. However, due to the inadequate resolution of these methods, the exact number of active conformations of CueR has not been determined. Resolving the structure of CueR in its active state is highly important for the development of specific inhibitors. Herein, the potential of double‐histidine (dHis)‐based CuII spin labeling for the identification of various conformational states of CueR during transcription is shown. Mineral resources have been counted as public assets with economic benefit since time immemorial. Due to the rising issue of decreasing mineral deposits, recovery of metals from several waste residues has become progressively more essential. Novel and efficient recycling processes have been on the rise globally. Manganese (Mn) as the fourth most industrially applicable metal generates an extensive quantity of metallic waste which not only leads to loss of precious metal but also results in environmental toxicity. Globally, around 7 million tons of high-grade ores are produced, whereas 8 million tons of Mn alloys are produced yearly. Therefore, it is of greater significance to recover and recycle Mn from various waste residues. Various physical and biological techniques have been developed for recycling Mn from waste residues. Traditional Mn extraction processes are costly and labor intensive in nature, on the contrary, bioleaching techniques using diverse microorganism's, form the basis of an efficient, eco-friendly, and economically sustainable process of metal recovery. The quick progress in current methodologies to counteract the fast consumption of innate mineral resources involves the proper utilization of unused waste residues containing industrially important metals like Mn. This review focuses to enumerate diverse features of Mn recovery, efficient methodologies, bioleaching of Mn, merits of Mn bioleaching, and applications of recycled Mn along with the futuristic applications. The global consumption of manganese is rising due to its growing industrial requirement while the natural reserves of manganese are diminishing at an alarming rate. Spin labels attached to two residues of a protein chain have less conformational flexibility than those attached to a single residue and thus lead to a narrower spatial distribution of the unpaired electron. The case of Cu2+ labels based on the double-histidine (dHis) motif is of particular interest, as it combines the advantage of precise localization of the unpaired electron with a labelling scheme orthogonal to the more common cysteine-based labelling. Here, we introduce an approach for in silico spin labelling of a protein by dHis motifs and Cu2+ complexes of iminodiacetic acid or nitrilotriacetic acid. We discuss a computerized scan for native histidine pairs that might be prone to bind such Cu2+ complexes and spin-labelling site pair scans that can identify suitable double mutants for labelling. Predicted distance distributions between two Cu2+ labels are compared to experimental distance distributions. We also test the hypothesis that elastic network modelling of conformational transitions with Cu2+-dHis labels can provide more accurate structural models than with nitroxide labels. S100A12 or Calgranulin C is a homodimeric antimicrobial protein of the S100 family of EF-hand calcium-modulated proteins. S100A12 is involved in many diseases such as inflammation, tumor invasion, cancer and neurological disorders such as Alzheimer’s disease. The binding of transition metal ions to the protein is important as the sequestering of the metal ion induces conformational changes in the protein, inhibiting the growth of various pathogenic microorganisms. In this work, we probe the Cu2+ binding properties of Calgranulin C. We demonstrate that the two Cu2+ binding sites in Calgranulin C show different coordination environments in solution. Continuous wave-electron spin resonance (CW-ESR) spectra of Cu2+-bound protein clearly show two distinct components at higher Cu2+:protein ratios, which is indicative of the two different binding environments for the Cu2+ ions. The g|| and A|| values are also different for the two components, indicating that the number of directly coordinated nitrogen in each site differs. Furthermore, we perform CW-ESR titrations to obtain the binding affinity of the Ca2+-loaded protein to Cu2+ ions. We observe a positive cooperativity in binding of the two Cu2+ ions. To further probe the Cu2+ coordination, we also perform electron spin echo envelope modulation (ESEEM) experiment. We perform ESEEM at two different fields where one Cu2+ binding site dominates the other. At both sites we see distinct signatures of Cu2+-histidine coordination. However, we clearly see that the ESEEM spectra corresponding to the two Cu2+ binding sites are significantly different. There is clear change in the intensity of the double quantum peak with respect to the nuclear quadrupole interaction peak at the two different fields. Furthermore, ESEEM along with hyperfine sublevel correlation show that only one of the two Cu2+ binding sites has backbone coordination, confirming our previous observation. Finally, we perform double electron-electron resonance spectroscopy to probe if the difference in binding environment is due to the Cu2+ binding to different sites in the protein. We obtain a distance distribution with a sharp peak at ~ 3 nm and a broad peak at ~ 4 nm. The shorter distance agrees with the Cu2+-Cu2+ distance expected for a dimer from the crystal structure. This paper reports an optical “TURN OFF” aptasensor, which is comprised of a deoxyribonucleic acid aptamer attached to a quantum dot on the $5^{\prime }$ terminus and gold nanoparticle on the $3^{\prime }$ terminus. The photoluminescence intensity is observed to decrease upon progressive addition of the target protein tumor necrosis factor-alpha (TNF- $\alpha $) to the sensor. For PBS-based TNF- $\alpha $ samples, the beacon exhibited 19%–20% quenching at around 22 nM concentration. The photoluminescence intensity and the quenching efficiency showed a linear decrease and a linear increase, respectively, between 0 to 22.3 nM TNF- $\alpha $. The detection limit of the sensor was found to be 97.2 pM. Specificity test results determined that the sensor has higher selectivity toward TNF- $\alpha $ than other control proteins such as C—reactive protein, albumin, and transferrin. The beacon successfully detected different concentrations of TNF- $\alpha $ in human serum-based samples exhibiting around 10% quenching efficiency at 12.5 nM of the protein. [ABSTRACT FROM AUTHOR]

Bone grafting is one of the commonest surgical procedures, yet all bone substitutes developed so far suffer from specific weaknesses and the search for a bone graft material with ideal physical and biological properties is still ongoing. Calcium phosphate pastes are the most frequently used synthetic bone grafts, yet they (a) often take an impractically long time to set, (b) release the drug content too fast, and (c) do not form pores large enough to accommodate host cells and foster osseointegration. To make up for these deficiencies, we introduced gelatin and silica to pastes composed of 5–15 nm sized hydroxyapatite nanoparticles and yielded a bioresorbable composite that is compact, yet flowing upon injection; that prevents setting at room temperature, but sets promptly, in minutes, at 37 °C; that displays an increase in surface porosity following immersion in physiological fluids; that allows for sustained release of antibiotics; and that sets in a tunable manner and in clinically relevant time windows: 1–3 minutes at its fastest. Timelapse, in situ X-ray diffraction analysis demonstrated that the setting process is accompanied by an increase in crystallinity of the initially amorphous hydroxyapatite, involving no polymorphic phase transitions in its course. Setting time can be tuned by controlling the weight content of gelatin or powder-to-liquid ratio. The release of vancomycin was slow, ∼8% after 2 weeks, and unaffected by the gelatin content. While vancomycin-loaded pastes were effective in reducing the concentration of all bacterial species analyzed, the bacteriostatic effects of the antibiotic-free pastes were pronounced against S. liquefaciens and E. coli. S. liquefaciens bacilli underwent beading and filamentation during the treatment, suggesting that the antimicrobial effects are attributable to cell wall disruption by hydroxyapatite nanoparticles. Vancomycin-loaded pastes augmented the activity of the antibiotic against P. aeruginosa and S. liquefaciens, while exhibiting no negative effects against human mesenchymal stem cells. They were also uptaken three times more abundantly than pure hydroxyapatite, indicating the theoretical favorability of their use for intracellular delivery of therapeutics. Background: Electroconvulsive seizure treatment is a fast-acting antidepressant therapy that evokes rapid transcriptional, neurogenic, and behavioral changes. Epigenetic mechanisms contribute to altered gene regulation, which underlies the neurogenic and behavioral effects of electroconvulsive seizure. We hypothesized that electroconvulsive seizure may modulate the expression of epigenetic machinery, thus establishing potential alterations in the epigenetic landscape. Methods: We examined the influence of acute and chronic electroconvulsive seizure on the gene expression of histone modifiers, namely histone acetyltransferases, histone deacetylases, histone methyltransferases, and histone (lysine) demethylases as well as DNA modifying enzymes, including DNA methyltransferases, DNA demethylases, and methyl-CpG-binding proteins in the hippocampi of adult male Wistar rats using quantitative real time-PCR analysis. Further, we examined the influence of acute and chronic electroconvulsive seizure on global and residue-specific histone acetylation and methylation levels within the hippocampus, a brain region implicated in the cellular and behavioral effects of electroconvulsive seizure. Results: Acute and chronic electroconvulsive seizure induced a primarily unique, and in certain cases bidirectional, regulation of histone and DNA modifiers, and methyl-CpG-binding proteins, with an overlapping pattern of gene regulation restricted to Sirt4, Mll3, Jmjd3, Gadd45b, Tet2, and Tet3. Global histone acetylation and methylation levels were predominantly unchanged, with the exception of a significant decline in H3K9 acetylation in the hippocampus following chronic electroconvulsive seizure. Conclusions: Electroconvulsive seizure treatment evokes the transcriptional regulation of several histone and DNA modifiers, and methyl-CpG-binding proteins within the hippocampus, with a predominantly distinct pattern of regulation induced by acute and chronic electroconvulsive seizure. Vitamin C is known to inhibit mycobacterial growth by acting as a hypoxia inducing agent. While investigating how mycobacteriophage growth is influenced by hypoxic conditions induced by vitamin C, using Mycobacterium smegmatismycobacteriophage D29 as a model system, it was observed that prior exposure of the host to such conditions resulted in increased burst size of the phage. Vitamin C pre-exposure was also found to induce synchronous growth of the host. A mutant defective in DevR, the response regulator that controls hypoxic responses in mycobacteria, neither supported higher phage bursts nor was it able to undergo synchronized growth following vitamin C pre-exposure, indicating thereby that the two phenomena are interrelated. Further evidence supporting such an interrelationship was obtained from the observation that phage burst sizes varied depending on the stage of synchronous growth that the host cells were in, at the time of infection--higher bursts were observed in the resting/synthetic phases and lower in the dividing ones. The effects were specific in nature as synchronization by an unrelated method, known as 'crowding', did not lead to the same consequence. The results indicate that growth synchronization induced by vitamin C treatment is a DevR-dependent phenomenon which is exploited by mycobacteriophage D29 to grow in larger numbers. Manganese (Mn) ranks twelfth among the most exuberant metal present in the earth's crust and finds its imperative application in the manufacturing steel, chemical, tannery, glass, and battery industries. Solubilisation of Mn can be performed by several bacterial strains which are useful in developing environmental friendly solutions for mining activities. The present investigation aims to isolate and characterize Mn solubilising bacteria from low grade ores from Sanindipur Manganese mine of Sundargh district in Odisha state of India. Four morphologically distinct bacterial strains showing visible growth on Mn supplemented plates were isolated. Mn solubilising ability of the bacterial strains was assessed by visualizing the lightening of the medium appearing around the growing colonies. Three isolates were gram negative and rod shaped while the remaining one was gram positive, coccobacilli. Molecular identification of the isolates was carried out by 16S rRNA sequencing and the bacterial isolates were taxonomically classified as Bacillus anthrasis MSB 2, Acinetobacter sp. MSB 5, Lysinibacillus sp. MSB 11, and Bacillus sp. MMR-1 using BLAST algorithm. The sequences were deposited in NCBI GenBank with the accession number KP635223, KP635224, KP635225 and JQ936966, respectively. Manganese solubilisation efficiency of 40, 96, 97.5 and 48.5% were achieved by MMR-1, MSB 2, MSB 5 and MSB 11 respectively. The efficiency of Mn solubilisation is suggested with the help of a pH variation study. If nations are 'imagined communities', 1 as many theorists like to define them, then they need an ideology to create a cohesive imagination. In modern times, the project of writing 'history' has been an important instrument in the service of this ideological purpose of justifying and reproducing the modern nation-state as the predestined and legitimate container of collective consciousness. School textbooks, at least in South Asia, have long been among the most exploited media for the presentation of the history of the national collective. This essay is a study of school textbooks in Bangladesh. It looks at narrative representations of selected episodes from the past, both pre- and postindependence, in order to reflect on how they construct 'history'. Through this work I endeavor to relate textual images to issues of community relations and identity by identifying and sharing the ways in which the audience for nationalist discourse is created, nurtured, and secured through symbolic means.
