In the present research, thermal investigation of amorphous solid dispersions (ASDs) was performed in order to compare their relaxation transitions. Two different active pharmaceutical ingredients (APIs), namely naproxen (NAP) and spironolactone (SPIR), were used as model drugs and combined with polyvinylpyrrolidone vinyl acetate (PVPVA64) copolymer to form stable ASDs. The reason for the API selection was their different interacting potentials based on the presence (in the case of NAP) and the lack of H-bond donor group (in the case of SPIR). Determination of both Tg and sub-Tg transitions suggested that ASDs containing NAP and PVPVA64 are 'interacting' systems with respect to the H-bonding. Besides, temperature-dependent Raman spectroscopic experiments confirmed that the naphthalene ring of the NAP has a significant role in the sub-Tg relaxations. In contrast, SPIR-PVPVA64 systems proved to be 'non-interacting' ASDs according to the MDSC, TSDC, and Raman measurements. This study highlights that the combination of the results of different thermoanalytical investigations with spectroscopic methods helps to understand the molecular background of the relaxations in ASDs and thus contributes to the conscious design of stable amorphous pharmaceuticals in the early stage of development. Metal organic frameworks (MOFs) and particularly copper benzene-1,3,5-tricarboxylate (HKUST-1) are excellent materials for gas storage (e.g., CH4, N2, H2 adsorption) and gas separation. In this work, reduced graphene oxide (RGO)–HKUST-1 mechanical mixtures were studied in order to reveal the effect of RGO content on the pressure tolerance of the texture and heat conductivity. HKUST-1 was obtained by two different synthesis routes. Air-dried MOF and RGO were thoroughly mixed prior to the compression. Powder XRD and Raman spectroscopy were used to characterize the response of the crystal structure, while low-temperature nitrogen adsorption was used the follow the adsorption properties of the pellets. Finally, the "flash" heat pulse method was used to assess the thermal properties. The gas adsorption isotherms revealed that the adsorption capacity decreases when RGO is added. Based on Raman and XRD results, we found that the synthesis route has an effect on multiple scales. We experimentally confirmed that evaluation of the thermal diffusivity requires a model more complex than the simple Fourier equation, due to the inherent heterogeneous structure of the material. A good approximation of the Fourier coefficient of thermal diffusivity was obtained using the parameters of the Guyer–Krumhansl equation. [Carbonatotetraamminecobalt(III)] permanganate monohydrate was synthesized first in the metathesis reaction of [Co(NH3)4CO3]NO3 and NaMnO4 in aqueous solution. Its thermal dehydration at 100 °C resulted in phase-pure [Co(NH3)4CO3]MnO4 (compound 1). Compounds 1 and 2 (i.e., the hydrated form) were studied with IR, far-IR, and low-temperature Raman spectroscopies, and their vibrational modes were assigned. The lattice parameters were determined by powder X-ray diffraction (PXRD) and single crystal X-ray diffraction (SXRD) methods for the triclinic and orthorhombic compounds 1 and 2, respectively. The detailed structure of compound 2 was determined, and the role of hydrogen bonds in the structural motifs was clarified. UV studies on compounds 1 and 2 showed the distortion of the octahedral geometry of the complex cation during dehydration because of the partial loss of the hydrogen bonds between the crystal water and the ligands of the complex cation. The thermal decomposition consists of a solid phase quasi-intramolecular redox reaction between the ammonia ligands and permanganate anions with the formation of ammonia oxidation products (H2O, NO, N2O, and CO2). The solid phase reaction product is amorphous cobalt manganese oxide containing ammonium, carbonate (and nitrate) anions. The temperature-controlled thermal decomposition of compound 2 in toluene at 110 °C showed that one of the decomposition intermediates is ammonium nitrate. The decomposition intermediates are transformed into Co1.5Mn1.5O4 spinel with MnCo2O4 structure upon further heating. Solid compound 2 gave the spinel at 500 °C both in an inert and air atmosphere, whereas the sample pre-treated in toluene at 110 °C without and with the removal of ammonium nitrate by aqueous washing, gave the spinel already at 300 and 400 °C, respectively. The molten NH4NO3 is a medium to start spinel crystallization, but its decomposition stops further crystal growth of the spinel phase. By this procedure, the particle size of the spinel product as low as ~4.0 nm could be achieved for the treatments at 300 and 400 °C, and it increased only to 5.7 nm at 500 °C. Major histocompatibility complex (MHC) genes are commonly used markers for monitoring adaptive genetic and evolutionary potential of species. In this study, we investigated genetic variation of the MHC class II DRB locus in the chamois genus Rupicapra by using next-generation sequencing. Sequencing of 102 samples led to the identification of 25 alleles, 11 of which are novel. The high ratio of the relative rates of nonsynonymous to synonymous mutations (dN/dS) suggests a signal of positive selection on this locus. We analyzed patterns of genetic variation within and among 2 subspecies of Northern Chamois and compared them to previously published studies using neutral markers to provide a basis for assessing the effects of demographic processes. Our analyses have shown that alleles are likely to be maintained by balancing selection in different populations with similar frequencies and that this mechanism also works in small, isolated populations that are strongly affected by genetic drift. Sustainable battery concepts are of great importance for the energy storage demands of the future. Organic batteries based on redox‐active polymers are one class of promising storage systems to meet these demands, in particular when combined with environmentally friendly and safe electrolytes. Deep Eutectic Solvents (DESs) represent a class of electrolytes that can be produced from sustainable sources and exhibit in most cases no or only a small environmental impact. Because of their non‐flammability, DESs are safe, while providing an electrochemical stability window almost comparable to established battery electrolytes and much broader than typical aqueous electrolytes. Here, we report the first all‐organic battery cell based on a DES electrolyte, which in this case is composed of sodium bis(trifluoromethanesulfonyl)imide (NaTFSI) and N‐methylacetamide (NMA) alongside the electrode active materials poly(2,2,6,6‐tetramethylpiperidin‐1‐yl‐oxyl methacrylate) (PTMA) and crosslinked poly(vinylbenzylviologen) (X‐PVBV2+). The resulting cell shows two voltage plateaus at 1.07 V and 1.58 V and achieves Coulombic efficiencies of 98 %. Surprisingly, the X‐PVBV/X‐PVBV+ redox couple turned out to be much more stable in NaTFSI : NMA 1 : 6 than the X‐PVBV+/X‐PVBV2+ couple, leading to asymmetric capacity fading during cycling tests. Background: Non-intubated thoracic surgery has not achieved widespread acceptance despite its potential to improve postoperative outcomes. To ensure airway safety, our institute has developed a technique combining spontaneous ventilation with double-lumen tube intubation (SVI). This study aimed to verify the feasibility and limitations of this SVI technique. Methods: For the SVI method, anesthesia induction involves fentanyl and propofol target-controlled infusion, with mivacurium administration. Bispectral index monitoring was used to ensure the optimal depth of anesthesia. Short-term muscle relaxation facilitated double-lumen tube intubation and early surgical steps. Chest opening preceded local infiltration, followed by a vagal nerve blockade to prevent the cough reflex and a paravertebral blockade for pain relief. Subsequently, the muscle relaxant was ceased. The patient underwent spontaneous breathing without coughing during surgical manipulation. Results: Between 10 March 2020 and 28 October 2022, 141 SVI surgeries were performed. Spontaneous respiration with positive end-expiratory pressure was sufficient in 65.96% (93/141) of cases, whereas 31.21% (44/141) required pressure support ventilation. Only 2.84% (4/141) of cases reversed to conventional anesthetic management, owing to technical or surgical difficulties. Results of the 141 cases: The mean maximal carbon dioxide pressure was 59.01 (34.4–92.9) mmHg, and the mean lowest oxygen saturation was 93.96% (81–100%). The mean one-lung, mechanical and spontaneous one-lung ventilation time was 74.88 (20–140), 17.55 (0–115) and 57.73 (0–130) min, respectively. Conclusions: Spontaneous ventilation with double-lumen tube intubation is safe and feasible for thoracic surgery. The mechanical one-lung ventilation time was reduced by 76.5%, and the rate of anesthetic conversion to relaxation was low (2.8%). [ABSTRACT FROM AUTHOR]

A trace amount of water is typically unavoidable as an impurity in ionic liquids, which is a huge challenge for their application in Mg‐ion batteries. Here, we employed molecular sieves of different pore diameters (3, 4, and 5 Å), to effectively remove the trace amounts of water from 1‐methyl‐1‐propylpiperidinium bis(trifluoromethylsulfonyl)imide (MPPip‐TFSI) and 1‐butyl‐1‐methylpyrrolidinium bis(trifluoromethylsulfonyl)imide (BMP‐TFSI). Notably, after sieving (water content <1 mg ⋅ L−1), new anodic peaks arise that are attributed to the formation of different anion‐cation structures induced by minimizing the influence of hydrogen bonds. Furthermore, electrochemical impedance spectroscopy (EIS) reveals that the electrolyte resistance decreases by ∼10 % for MPPip‐TFSI and by ∼28 % for BMP‐TFSI after sieving. The electrochemical Mg deposition/dissolution is investigated in MPPip‐TFSI/tetraglyme (1 : 1)+100 mM Mg(TFSI)2+10 mM Mg(BH4)2 using Ag/AgCl and Mg reference electrodes. The presence of a trace amount of water leads to a considerable shift of 0.9 V vs. Mg2+/Mg in the overpotential of Mg deposition. In contrast, drying of MPPip‐TFSI enhances the reversibility of Mg deposition/dissolution and suppresses the passivation of the Mg electrode. [ABSTRACT FROM AUTHOR]

Inflammasomes, primarily responsible for the activation of IL-1β, have emerged as critical regulators of the tumor microenvironment. By using in vivo and in vitro brain metastasis models, as well as human samples to study the role of the NLRP3 inflammasome in triple-negative breast cancer (TNBC) brain metastases, we found NLRP3 inflammasome components and IL-1β to be highly and specifically expressed in peritumoral astrocytes. Soluble factors from TNBC cells induced upregulation and activation of NLRP3 and IL-1β in astrocytes, while astrocyte-derived mediators augmented the proliferation of metastatic cells. In addition, inhibition of NLRP3 inflammasome activity using MCC950 or dampening the downstream effect of IL-1β prevented the proliferation increase in cancer cells. In vivo, MCC950 reduced IL-1β expression in peritumoral astrocytes, as well as the levels of inflammasome components and active IL-1β. Most importantly, significantly retarded growth of brain metastatic tumors was observed in mice treated with MCC950. Overall, astrocytes contribute to TNBC progression in the brain through activation of the NLRP3 inflammasome and consequent IL-1β release. We conclude that pharmacological targeting of inflammasomes may become a novel strategy in controlling brain metastatic diseases. [ABSTRACT FROM AUTHOR]

In this work, the performance of two fast chemical imaging techniques, Raman and near-infrared (NIR) imaging is compared by utilizing these methods to predict the rate of drug release from sustained-release tablets. Sustained release is provided by adding hydroxypropyl methylcellulose (HPMC), as its concentration and particle size determine the dissolution rate of the drug. The chemical images were processed using classical least squares; afterwards, a convolutional neural network was applied to extract information regarding the particle size of HPMC. The chemical images were reduced to an average HPMC concentration and a predicted particle size value; these were used as inputs in an artificial neural network with a single hidden layer to predict the dissolution profile of the tablets. Both NIR and Raman imaging yielded accurate predictions. As the instrumentation of NIR imaging allows faster measurements than Raman imaging, this technique is a better candidate for implementing a real-time technique. Purpose: In continuous manufacturing (CM), the material traceability and process dynamics can be investigated by residence time distribution (RTD). Many of the unit operations used in the pharma industry were characterized by dead time–dominated RTD. Even though feasible and proper feedback control is one of the many advantages of CM, its application is challenging in these cases. This study aims to develop a feedback control, implementing the RTD in a Smith predictor control structure in a continuous powder blender line. Methods: Continuous powder blending was investigated with near-infrared spectroscopy (NIR), and the blending was controlled through a volumetric feeder. A MATLAB GUI was developed to calculate and control the concentration of the API based on the chemometric evaluation of the spectra. The programmed GUI changed the feeding rate based on the proportional integral derivative (PID) and the Smith predictor, which implemented the RTD of the system. The control structures were compared even on a system with amplified dead time. Results: In this work, the control structure of the Smith control was devised by utilizing the RTD of the system. The Smith control was compared to a classic PI control structure on the normal system and on an increased dead time system. The Smith predictor was able to reduce the response time for various disturbances by up to 50%, and the dead time had a lower effect on the control. Conclusions: Implementing the RTD models in the control structure improved the process design and further expanded the wide range of applications of the RTD models. Both control structures were able to reduce the effect of disturbances on the system; however, the Smith predictor presented more reliable and faster control, with a wider space for control tuning. Thermal processing of Zr-loaded ion-exchangers is a facile route to synthetize (ZrO2, ZrC)@C composites. In the present paper, furnace and RF-thermal plasma processing of ZrOCl2 loaded thiourea-functionalized styrene-divinylbenzene copolymer was investigated and led to composites containing ZrO2 and ZrC. Different ZrO2@C composites were formed between 1000 and 1400 °C in 2 h, whereas the composite containing ZrC was created at 1400 °C in 8 h. The ratio of ZrO2/ZrC, the prevailing ZrO2 modifications, and the crystallite sizes strongly depend on the synthesis conditions. The ZrC-containing composites formed only at 1400 °C in 8 h and by the plasma treatment of the ZrO2@C sample prepared in the furnace, resulting in 8 and 16% ZrC content, with 44 and 41 nm ZrC crystallite sizes, respectively. The ZrO2-containing composites (tetragonal, monoclinic, and cubic modifications with 65–88 nm ZrO2 crystallite sizes and 15–43 m2/g BET surface areas) formed in a tube furnace between 1000 and 1400 °C in 2 h. All ZrO2@C composites had both amorphous carbon and graphite, and their ratio is temperature dependent. The carbonaceous compounds were characterized by Raman spectroscopy with analysis of the G and D band intensities. XPS studies showed the surface oxidation of ZrC. [ABSTRACT FROM AUTHOR]

Epithelial ion and fluid secretion determine the physiological functions of a broad range of organs, such as the lung, liver, or pancreas. The molecular mechanism of pancreatic ion secretion is challenging to investigate due to the limited access to functional human ductal epithelia. Patient-derived organoids may overcome these limitations, however direct accessibility of the apical membrane is not solved. In addition, due to the vectorial transport of ions and fluid the intraluminal pressure in the organoids is elevated, which may hinder the study of physiological processes. To overcome these, we developed an advanced culturing method for human pancreatic organoids based on the removal of the extracellular matrix that induced an apical-to-basal polarity switch also leading to reversed localization of proteins with polarized expression. The cells in the apical-out organoids had a cuboidal shape, whereas their resting intracellular Ca2+ concentration was more consistent compared to the cells in the apical-in organoids. Using this advanced model, we demonstrated the expression and function of two novel ion channels, the Ca2+ activated Cl− channel Anoctamin 1 (ANO1) and the epithelial Na+ channel (ENaC), which were not considered in ductal cells yet. Finally, we showed that the available functional assays, such as forskolin-induced swelling, or intracellular Cl− measurement have improved dynamic range when performed with apical-out organoids. Taken together our data suggest that polarity-switched human pancreatic ductal organoids are suitable models to expand our toolset in basic and translational research. [ABSTRACT FROM AUTHOR]

Three octahedral iron(III) nitrate complexes with dimethylated urea ligand isomers, [hexakis(N,N'-dimethylurea-O)iron(III)] nitrate (compound 1), trans-[diaquatetrakis(N,N-dimethylurea-O)iron(III)] nitrate (compound 2), and [hexakis(N,N-dimethylurea-O)iron(III)] nitrate trihydrate (compound 3) were prepared and characterized with single crystal X-ray diffraction, IR, Raman and UV–Vis methods. In compounds 1 and 3, six dimethylurea ligands coordinate to the central FeIII ion via the oxygen in octahedral geometry and the ligands are arranged in a propeller-like manner, dividing the complex cations into two sides. In compound 1, the dimethylurea propellers screw in the opposite direction on the two sides of the complex and in compound 3, they are arranged with the same handedness on the two sides. The complexes have helical chirality. The two sides of the complex cations differ not only in the rotation direction of the ligands but also in the hydrogen bond formation. On one side of the complex cation, the ligands form intermolecular hydrogen bonds only with the crystal waters, meanwhile on the other side of the complex, the ligands form hydrogen bonds only with the nitrate ions. In compound 2, [Fe(N,N-dimethylurea)4(H2O)2]3+ cations form layers that are separated by interconnected NO3− ions forming a hydrogen bonding system and connecting the complex cations A-s and B-s. The three crystallographically different nitrate ions each form four hydrogen bonds in a way that they have one bidentate O atom and two monodentate O atoms; however, the anions differ in their hydrogen bonding. Biohydrogen production from wastewater using eukaryotic green algae can be facilitated by appropriately selected bacterial partners and cultivation conditions. Two Chlorella algal species were chosen for these experiments, based on their robust growth ability in synthetic wastewater. The applied three Bacillus bacterial partners showed active respiration and efficient biomass production in the same synthetic wastewater. Bacillus amyloliquefaciens, Bacillus mycoides, and Bacillus cereus as bacterial partners were shown to specifically promote algal biomass yield. Various inter-kingdom co-culture combinations were investigated for algal–bacterial biomass generation, for co-culture-specific exopolysaccharide patterns, and, primarily, for algal biohydrogen evolution. Chlorella sp. MACC-38 mono- and co-cultures generated significantly higher biomass compared with that of Chlorella sp. MACC-360 mono- and co-cultures, while in terms of hydrogen production, Chlorella sp. MACC-360 co-cultures clearly surpassed their Chlorella sp. MACC-38 counterparts. Imaging studies revealed tight physical interactions between the algal and bacterial partners and revealed the formation of co-culture-specific exopolysaccharides. Efficient bacterial respiration was in clear correlation with algal hydrogen production. Stable and sustainable algal hydrogen production was observed in synthetic wastewater for Chlorella sp. MACC-360 green algae in co-cultures with either Bacillus amyloliquefaciens or Bacillus cereus. The highest algal hydrogen yields (30 mL H2 L−1 d−1) were obtained when Chlorella sp. MACC-360 was co-cultured with Bacillus amyloliquefaciens. Further co-culture-specific algal biomolecules such as co-cultivation-specific exopolysaccharides increase the valorization potential of algal–bacterial co-cultures and might contribute to the feasibility of algal biohydrogen production technologies. The aim of this study was to develop antisense oligonucleotide tablet formulations using high-speed electrospinning. Hydroxypropyl-beta-cyclodextrin (HPβCD) was used as a stabilizer and as an electrospinning matrix. In order to optimize the morphology of the fibers, electrospinning of various formulations was carried out using water, methanol/water (1:1), and methanol as solvents. The results showed that using methanol could be advantageous due to the lower viscosity threshold for fiber formation enabling higher potential drug loadings by using less excipient. To increase the productivity of electrospinning, high-speed electrospinning technology was utilized and HPβCD fibers containing 9.1% antisense oligonucleotide were prepared at a rate of ~330 g/h. Furthermore, to increase the drug content of the fibers, a formulation with a 50% drug loading was developed. The fibers had excellent grindability but poor flowability. The ground fibrous powder was mixed with excipients to improve its flowability, which enabled the automatic tableting of the mixture by direct compression. The fibrous HPβCD–antisense oligonucleotide formulations showed no sign of physical or chemical degradation over the 1-year stability study, which also shows the suitability of the HPβCD matrix for the formulation of biopharmaceuticals. [κ2-O,O′-Carbonatotetraamminecobalt(III)] iodide, or [Co(NH3)4CO3]I, named in this paper as compound 1, was prepared and characterized comprehensively with spectroscopic (IR, Raman and UV) and single-crystal X-ray diffraction methods. Compound 1 was orthorhombic, and isomorphous with the analogous bromide. The four ammonia ligands and the carbonate anion were coordinated to the central cobalt cation in a distorted octahedral geometry. The carbonate ion formed a four-membered symmetric planar chelate ring. The complex cations were bound to each other by N-H···O hydrogen bonds and formed zigzag sheets via an extended 2D hydrogen bond network. The complex cations and iodide ions were arranged into ion pairs and each cation bound its iodide pair through three hydrogen bonds. The thermal decomposition started with the oxidation of the iodide ion by CoIII in the solid phase resulting in [Co(NH3)4CO3] and I2. This intermediate CoII-complex in situ decomposed into Co3O4 and C-N bond containing intermediates. In inert atmosphere, CO or C-N bond containing compounds, and also, due to the in situ decomposition of CoCO3 intermediate, Co3O4 was formed. The quasi-intramolecular solid-phase redox reaction of [Co(NH3)4CO3] might have resulted in the formation of C-N bond containing compounds with substoichiometric release of ammonia and CO2 from compound 1. The C-N bond containing intermediates reduced Co3O4 into CoO and Co, whereas in oxygen-containing atmosphere, the end-product was Co3O4, even at 200 °C, and the endothermic ligand loss reaction coincided with the consecutive exothermic oxidation processes. The recognition that egg volume variation has widespread implications in avian biology led us to test the accuracy and precision of the most commonly used egg volume determination methods. As a benchmark for the tests, we used real egg volume values determined by water submersion. We identified some evident limitations of this approach and attempted to improve the method by using distilled water and accurate temperature correction. Starting from the assumption that our methodological proposals can effectively improve the accuracy of egg volume measurements, we compared the outcomes with two widely used volume estimation methods based on Hoyt's equation and Troscianko software estimate for five gallinaceous bird species (forty eggs from each species). We found that Hoyt's and Troscianko's egg volume estimation methods strongly correlate with our volume measurements. Despite the highly significant and relatively high values of coefficients of determination, further analyses reveal some important differences among the methods. [ABSTRACT FROM AUTHOR]

We synthesized and characterized (IR, Raman, UV, SXRD) hexaamminecobalt(III) dichloride permanganate, [Co(NH3)6]Cl2(MnO4) (compound 1) as the precursor of Co–Mn–spinel composites with atomic ratios of Co:Mn = 1:1 and 1:3. The 3D−hydrogen bond network includes N–H···O–Mn and N–H···Cl interactions responsible for solid-phase redox reactions between the permanganate anions and ammonia ligands. The temperature-limited thermal decomposition of compound 1 under the temperature of boiling toluene (110 °C) resulted in the formation of (NH4)4Co2Mn6O12. which contains a todorokite-like manganese oxide network (MnII4MnIII2O1210−). The heat treatment products of compounds 1 and [Co(NH3)5Cl](MnO4)2 (2) synthesized previously at 500 °C were a cubic and a tetragonal spinel with Co1.5Mn1.5O4 and CoMn2O4 composition, respectively. The heating of the decomposition product of compounds 1 and 2 that formed under refluxing toluene (a mixture with an atomic ratio of Co:Mn = 1:1 and 1:2) and after aqueous leaching ((NH4)4Co2Mn6O12, 1:3 Co:Mn atomic ratio in both cases) at 500 °C resulted in tetragonal Co0.75Mn2.25O4 spinels. The Co1.5Mn1.5O4 prepared from compound 1 at 500 °C during the solid-phase decomposition catalyzes the degradation of Congo red with UV light. The decomposition rate of the dye was found to be nine times faster than in the presence of the tetragonal CoMn2O4 spinel prepared in the solid-phase decomposition of compound 2. The todorokite-like intermediate prepared from compound 1 under N2 at 115 °C resulted in a 54 times faster degradation of Congo red, which is a great deal faster than the same todorokite-like phase that formed from compound 2 under N2. One of the main issues of metal organic framework (MOF)‐based solid electrolytes (SE) is their high guest solvent content reaching up to >50 wt% of the total mass of SE pellets. The presence of large solvent amounts reduces the SE hardness and the electrochemical stability in presence of a magnesium (Mg) anode. Moreover, this often leads to misleading ionic conductivity values. In the present work, a strategy to minimize the guest solvent in MOF‐based SE from 44–55 wt% to 20–30 wt% of the total SE's mass is presented. Moreover, mixed metal organic frameworks of different structures and crystallinity are demonstrated for the first time to enhance the ionic conductivity of Mg2+ ions inside the MOFs' structures. The presence of both highly crystalline and amorphous MOFs increases the degree of disorder in the mixture and consequently opens up extra pathways for Mg2+ ion diffusion. The ionic conductivity of mixed MOFs [amorphous Mgbp3dc and crystalline α‐Mg3(HCOO)6] showed an enhanced value of 3.8×10−5 S cm−1 at 30 °C compared to 1.1×10−6 S cm−1 for α‐Mg3(HCOO)6. Mixed MOF‐SEs with a transference number (t+) of 0.335 showed a good stability in the presence of Mg electrodes with an enhanced reversibility upon galvanostatic cycling. [ABSTRACT FROM AUTHOR]

A hemipyridine solvate of bis(pyridine)silver(I) perchlorate, [Agpy2ClO4]·0.5py (compound 1) was prepared and characterized by single crystal X-ray analysis and vibrational spectroscopy (R and low-temperature Raman). Compound 1 was prepared via the trituration of [Agpy2ClO4] and 4[Agpy2ClO4]·[Agpy4]ClO4 (as the source of the solvate pyridine) in a mixed solvent of acetone:benzene =1:1 (v = v) at room temperature. The monoclinic crystals of compound 1 were found to be isomorphic with the analogous permanganate complex (a = 19.1093(16) Å, b = 7.7016(8) Å, c = 20.6915(19) Å, β = 105.515(7)°; space group: C2/c). Two [Agpy2]+ cations formed a dimeric unit [Agpy2ClO4]2, and each silver ion was connected to two ClO4− anions via oxygen atoms. The Ag∙∙∙Ag distance was 3.3873(5) Å, the perchlorate ions were coordinated to silver ions, and the Ag∙∙∙O distances were 2.840(2) Å and 2.8749(16) Å in the centrosymmetric rectangle of Ag-O-Ag-O. The stoichiometric ratio of the monomer [Agpy2ClO4] and the solvent pyridine was 1:0.5. The guest pyridine occupied 527.2 Å3, which was 18.0% of the volume of the unit cell. There was no additional residual solvent-accessible void in the crystal lattice. The solvate pyridine was connected via its a-CH to one of the O atoms of the perchlorate anion. Correlation analysis, as well as IR and low-temperature Raman studies, were performed to assign all perchlorate and pyridine vibrational modes. The solvate and coordinated pyridine bands in the IR and Raman spectra were not distinguishable. A perchlorate contribution via Ag-O coordination to low-frequency Raman bands was also assigned. Creating supersaturating drug delivery systems to overcome the poor aqueous solubility of active ingredients became a frequent choice for formulation scientists. Supersaturation as a solution phenomenon is, however, still challenging to understand, and therefore many recent publications focus on this topic. This work aimed to investigate and better understand the pH dependence of supersaturation of telmisartan (TEL) at a molecular level and find a connection between the physicochemical properties of the active pharmaceutical ingredient (API) and the ability to form supersaturated solutions of the API. Therefore, the main focus of the work was the pH-dependent thermodynamic and kinetic solubility of the model API, TEL. Based on kinetic solubility results, TEL was observed to form a supersaturated solution only in the pH range 3–8. The experimental thermodynamic solubility-pH profile shows a slight deviation from the theoretical Henderson–Hasselbalch curve, which indicates the presence of zwitterionic aggregates in the solution. Based on pKa values and the refined solubility constants and distribution of macrospecies, the pH range where high supersaturation-capacity is observed is the same where the zwitterionic form of TEL is present. The release of the FDA's guidance on Process Analytical Technology has motivated and supported the pharmaceutical industry to deliver consistent quality medicine by acquiring a deeper understanding of the product performance and process interplay. The technical opportunities to reach this high-level control have considerably evolved since 2004 due to the development of advanced analytical sensors and chemometric tools. However, their transfer to the highly regulated pharmaceutical sector has been limited. To this respect, data fusion strategies have been extensively applied in different sectors, such as food or chemical, to provide a more robust performance of the analytical platforms. This survey evaluates the challenges and opportunities of implementing data fusion within the PAT concept by identifying transfer opportunities from other sectors. Special attention is given to the data types available from pharmaceutical manufacturing and their compatibility with data fusion strategies. Eukaryotic algae represent a highly heterogeneous group in terms of organization, lifestyle, and metabolic capabilities. Unicellular green microalgae are capable of biohydrogen production through direct and indirect photolysis as well as dark fermentation. Most algae hydrogen studies focus on axenic algal cultures, although these are difficult and expensive to maintain for continuous operation. Moreover, the complex interplays and metabolic fluxes between algae and bacteria in natural ecosystems provide a number of clear biological and technological benefits to large-scale functional algae-based systems. Two green algae species from the Chlamydomonas and Chlorella genera were used to engineer stable synthetic communities by incorporating a starch-degrading bacterium from the Bacillus genus into the inter-kingdom consortium. Continuous photoheterotrophic biohydrogen production was achieved by elaborating an appropriate algal–bacterial ratio and fine-tuning the culture conditions for the synthetic consortia. Medium with starch as only carbon source served as a simple model of cheap substrate for algal hydrogen generation. The engineered pairwise algal–bacterial associations showed increased biomass and biohydrogen yield compared to the axenic control conditions. Chlorella sp. MACC-360 produced a significantly higher amount of hydrogen when both the bacterium partner and starch were added to the media compared to the axenic algae. Continuous, elevated algal hydrogen production was achieved in media supplemented with 8 g L−1 starch as sole carbon source when carefully selected initial cell number values were used for the Chlorella sp. Industry 4.0 has started to transform the manufacturing industries by embracing digitalization, automation, and big data, aiming for interconnected systems, autonomous decisions, and smart factories. Machine learning techniques, such as artificial neural networks (ANN), have emerged as potent tools to address the related computational tasks. These advancements have also reached the pharmaceutical industry, where the Process Analytical Technology (PAT) initiative has already paved the way for the real-time analysis of the processes and the science- and risk-based flexible production. This paper aims to assess the potential of ANNs within the PAT concept to aid the modernization of pharmaceutical manufacturing. The current state of ANNs is systematically reviewed for the most common manufacturing steps of solid pharmaceutical products, and possible research gaps and future directions are identified. In this way, this review could aid the further development of machine learning techniques for pharmaceutical production and eventually contribute to the implementation of intelligent manufacturing lines with automated quality assurance. The failures of anti-β-amyloid immunotherapies suggested that the very low fraction of injected antibodies reaching the brain parenchyma due to the filtering effect of the BBB may be a reason for the lack of therapeutic effect. However, there is no treatment, as yet, for the amyotrophic lateral sclerosis (ALS) despite substantial evidence existing of the involvement of TDP-43 protein in the evolution of ALS. To circumvent this filtering effect, we have developed a novel approach to facilitate the penetration of antibody fragments (Fabs) into the brain parenchyma. Leveraging the homing properties of endothelial progenitor cells (EPCs), we transfected, ex vivo, such cells with vectors encoding anti-β-amyloid and anti-TDP43 Fabs turning them into an "antibody fragment factory". When injected these cells integrate into the BBB, where they secrete anti-TDP43 Fabs. The results showed the formation of tight junctions between the injected engineered EPCs and the unlabeled resident endothelial cells. When the EPCs were further modified to express the anti-TDP43 Fab, we could observe integration of these cells into the vasculature and the secretion of Fabs. Results confirm that production and secretion of Fabs at the BBB level leads to their migration to the brain parenchyma where they might exert a therapeutic effect. [ABSTRACT FROM AUTHOR]

We have developed an easy route to prepare (nano-ZrO2,nano-ZrCx)@C composites with varying ZrO2/ZrCx content. The process consists of preparing a zirconium-loaded, iminodiacetate-functionalized styrene-divinylbenzene (STY-DVB) copolymer, and its subsequent carbonization in a tube furnace and/or a thermal plasma reactor. Depending on the zirconium salt used (zirconyl chloride, zirconyl nitrate or zirconium (IV) sulfate) in the Zr loading, the Zr-loaded resins resulted in ZrO2@C pre-pyrolizates with C to Zr molar ratios of 5.8, 6.8 and 6.60. This carbon surplus is sufficient for the partial or even complete reduction of ZrO2 into ZrC0.58 at 1400 °C. The reaction products also contain 5 to 55 mass% residual free carbon. The plasma processing of the ZrO2@C composite formed at 1000 °C in a tube furnace led to ZrC0.94@C composites. The transformation of amorphous carbon content during the plasma treatment strongly depended on the atmosphere (He or H2) in the reactor and the anion type of the Zr salt. In the presence of He, amorphous carbon could be completely transformed into graphite. In the presence of H2, amorphous carbon and graphite were found at roughly the same ratio. No ZrO2 could be detected in the plasma-treated samples, whilst different ZrO2 polymorphs were found in the samples prepared in the tube furnace, depending on the synthesis conditions. Spreading depolarizations (SDs) indicate injury progression and predict worse clinical outcome in acute brain injury. We demonstrate in rodents that acute brain swelling upon cerebral ischemia impairs astroglial glutamate clearance and increases the tissue area invaded by SD. The cytotoxic extracellular glutamate accumulation (>15 µM) predisposes an extensive bulk of tissue (4–5 mm2) for a yet undescribed simultaneous depolarization (SiD). We confirm in rat brain slices exposed to osmotic stress that SiD is the pathological expansion of prior punctual SD foci (0.5–1 mm2), is associated with astrocyte swelling, and triggers oncotic neuron death. The blockade of astrocytic aquaporin-4 channels and Na+/K+/Cl− co-transporters, or volume-regulated anion channels mitigated slice edema, extracellular glutamate accumulation (<10 µM) and SiD occurrence. Reversal of slice swelling by hyperosmotic mannitol counteracted glutamate accumulation and prevented SiD. In contrast, inhibition of glial metabolism or inhibition of astrocyte glutamate transporters reproduced the SiD phenotype. Finally, we show in the rodent water intoxication model of cytotoxic edema that astrocyte swelling and altered astrocyte calcium waves are central in the evolution of SiD. This review article summarizes the history the Hungarian Scientific Cloud Infrastructure project. This research infrastructure was launched officially on 1 October 2016, funded by the Hungarian Academy of Sciences. With the support of ELKH, the infrastructure’s capacity has been substantially boosted; the features and workflows that it offers to scientists were significantly expanded to celebrate the arrival of the year 2022. The article reviews the types of work Hungarian researchers implemented on the infrastructure, thereby providing an overview of the state of cloud-computing enabled science in Hungary. Plant pathogenic fungi are responsible for enormous crop losses worldwide. Overcoming this problem is challenging as these fungi can be highly resistant to approved chemical fungicides. There is thus a need to develop and introduce fundamentally new plant and crop protection strategies for sustainable agricultural production. Highly stable extracellular antifungal proteins (AFPs) and their rationally designed peptide derivatives (PDs) constitute feasible options to meet this challenge. In the present study, their potential for topical application to protect plants and crops as combinatorial biofungicides is supported by the investigation of two Neosartorya (Aspergillus) fischeri AFPs (NFAP and NFAP2) and their γ-core PDs. Previously, the biofungicidal potential of NFAP, its rationally designed γ-core PD (γNFAP-opt), and NFAP2 was reported. Susceptibility tests in the present study extended the in vitro antifungal spectrum of NFAP2 and its γ-core PD (γNFAP2-opt) to Botrytis, Cladosporium, and Fusarium spp. Besides, in vitro additive or indifferent interactions, and synergism were observed when NFAP or NFAP2 was applied in combination with γNFAP-opt. Except for γNFAP2-opt, the investigated proteins and peptides did not show any toxicity to tomato plant leaves. The application of NFAP in combination with γNFAP-opt effectively inhibited conidial germination, biofilm formation, and hyphal extension of the necrotrophic mold Botrytis cinerea on tomato plant leaves. However, the same combination only partially impeded the B. cinerea-mediated decay of tomato fruits, but mitigated the symptoms. Today, graphene oxide (GO) has gained well-deserved recognition, with its applications continuing to increase. Much of the processing of GO-based devices occurs in a dispersed form, which explains the commercialization of GO suspensions. Aging of these suspensions can, however, affect the shelf life and thus their application potential. Aging of GO preparations is often acknowledged, but no longer-term systematic study has been reported on the alteration of GO suspensions. This paper investigates high-concentration (10 mg/mL) aqueous GO suspensions over a 2-year time scale. In addition to steady shear tests, the dynamic behavior of the suspensions was studied in more detail by transient shear and frequency sweep measurements. Both the viscosity and the dynamic moduli increased with age, particularly within the first year. The results of the complementary Raman spectroscopic studies indicate that the change in the rheological behavior with aging results from a slow oxidation process occurring in the highly acidic aqueous medium during the relatively long-term storage. The (over)oxidized layers peel off spontaneously or are removed by high shear stress, resulting in increased viscosity, as it was corroborated by XRD and XPS. [ABSTRACT FROM AUTHOR]

In the present study, the effect of long-term exercise training was investigated on myocardial morphological and functional remodeling and on proarrhythmic sensitivity in a rabbit athlete's heart model. New-Zealand white rabbits were trained during a 12-week long treadmill running protocol and compared with their sedentary controls. At the end of the training protocol, echocardiography, in vivo and in vitro ECG recordings, proarrhythmic sensitivity with dofetilide (nM) were performed in isolated hearts, and action potential duration (APD) measurements at different potassium concentrations (4.5 and 2 mM) were made in the isolated papillary muscles. Expression levels of the slow component of delayed rectifier potassium current and fibrosis synthesis and degradation biomarkers were quantified. Echocardiography showed a significantly dilated left ventricle in the running rabbits. ECG PQ and RR intervals were significantly longer in the exercised group (79 ± 2 vs. 69 ± 2 ms and 325 ± 11 vs. 265 ± 6 ms, p < 0.05, respectively). The in vivo heart rate variability (HRV) (SD of root mean square: 5.2 ± 1.4 ms vs. 1.4 ± 0.2 ms, p < 0.05) and Tpeak-Tend variability were higher in the running rabbits. Bradycardia disappeared in the exercised group in vitro. Dofetilide tended to increase the QTc interval in a greater extent, and significantly increased the number of arrhythmic beats in the trained animals in vitro. APD was longer in the exercised group at a low potassium level. Real-time quantitative PCR (RT-qPCR) showed significantly greater messenger RNA expression of fibrotic biomarkers in the exercised group. Increased repolarization variability and higher arrhythmia incidences, lengthened APD at a low potassium level, increased fibrotic biomarker gene expressions may indicate higher sensitivity of the rabbit "athlete's heart" to life-threatening arrhythmias. The work aimed to develop the Absorption Driven Drug Formulation (ADDF) concept, which is a new approach in formulation development to ensure that the drug product meets the expected absorption rate. The concept is built on the solubility-permeability interplay and the rate of supersaturation as the driving force of absorption. This paper presents the first case study using the ADDF concept where not only dissolution and solubility but also permeation of the drug is considered in every step of the formulation development. For that reason, parallel artificial membrane permeability assay (PAMPA) was used for excipient selection, small volume dissolution-permeation apparatus was used for testing amorphous solid dispersions (ASDs), and large volume dissolution-permeation tests were carried out to characterize the final dosage forms. The API-excipient interaction studies on PAMPA indicated differences when different fillers or surfactants were studied. These differences were then confirmed with small volume dissolution-permeation assays where the addition of Tween 80 to the ASDs decreased the flux dramatically. Also, the early indication of sorbitol's advantage over mannitol by PAMPA has been confirmed in the investigation of the final dosage forms by large-scale dissolution-permeation tests. This difference between the fillers was observed in vivo as well. The presented case study demonstrated that the ADDF concept opens a new perspective in generic formulation development using fast and cost-effective flux-based screening methods in order to meet the bioequivalence criteria. This study examined the concentrations of Al, Ca, Cd, Cr, Cu, Fe, K, Mg, Mn, Na, Ni, Pb, and Zn in the liver and kidney samples of golden jackals (n = 163) and red foxes (n = 64). Additionally, we studied how sex affected element concentration in both species, and in the case of golden jackals, how concentration levels were affected by age. Liver was the most important storage organ for Cu, Fe, Mn, and Zn, while kidney for Cd and Na in jackals and foxes. We found no sex-related differences in concentration of any of the selected elements in fox samples. In jackals, the mean concentrations of Al, Mn, and Zn were significantly higher in kidneys of males, while Zn and Cu had higher concentrations in female livers. Our results suggest bioaccumulation processes do not occur in mesocarnivores. Lead and Cd concentrations fell within ranges accepted as normal for canine species and were below toxic levels in the organs of the examined species. Concentrations of Cr, Cu, Fe, Mn, and Zn were far below the toxic levels in both organs of both species, with a slight sign of deficiency of Fe, Mn, and Zn in jackal and fox kidneys. In comparison with toxicity levels and results from other European study sites, the current study demonstrated that species living in the examined area of Somogy County, Hungary, are generally exposed to low levels of environmental contamination. The swine industry is comprised of highly productive and healthy sows capable of weaning large litters for multiple parities. This prolificacy has resulted in greater metabolic demands during lactation due to increased capacity and demand for milk production. Modern sows also have increased genetic merit for leanness which has altered the amount of energy reserves available for mobilization to support the growing demands of lactation. Together, these factors may render modern sows more vulnerable to metabolic disorders such as postpartum dysgalactia syndrome (PDS) or mastitis-metritis-agalactia (MMA). Described as a metabolic and hormonal disturbance, a hallmark of PDS is a reduction (dysgalactia) or cessation (agalactia) in milk production after farrowing. Insufficient milk production by the sow leads to increased piglet mortality, causing substantial economic losses, reducing sow welfare, and resulting in premature removal of sows from the herd. Our prior reports have shown differences in circulating analytes and inflammatory markers between healthy sows and sows affected with PDS following parturition. However, very few differences were observed before clinical signs were evident, rendering these analytes ineffective in early detection of PDS. Therefore, the objective of the current study was to utilize gas-chromatography mass-spectrometry (GC-MS) to further characterize the biological conditions and metabolic pathways contributing to PDS in sows. Through this, we aimed to identify small molecules in circulation with the potential for use as a biomarker to aid in early detection of PDS. To accomplish this, a study was conducted on a commercial sow farm with a history of poor lactation performance. Sows (n = 36) with clinical signs of PDS were matched by parity and day of lactation with healthy control (CON) sows (n = 36) that remained unaffected. Blood samples collected at farrowing (timepoint 1) and at onset of clinical PDS (timepoint 2) were subjected to GC-MS. As expected, piglets from PDS sows had decreased (P < 0.01) average daily gain and greater mortality than piglets from control sows. Following GC-MS analysis, 16 and 41 molecules were identified as significantly different between CON and PDS sows at timepoint 1 and timepoint 2, respectively (P < 0.10). A difference in 2-hydroxybutyric acid, an early marker for insulin resistance and impaired glucose regulation, was observed at timepoint 1. Additional molecules identified as significantly different at timepoint 1 include D-pinitol, D-glucose, inositol (3,6)-bisphosphate, and others. Detailed vibrational (IR, Raman, far-IR) and thermal (TGA, TG–MS, DSC) analysis has been performed on di[κ1O,κ2O-carbonatotetraamminecobalt(III)] sulfate trihydrate, ([Co(NH3)4CO3]2SO4·3H2O (1). Its isothermic heating at 100 °C leads to formation of [Co(NH3)4CO3]2SO4 (compound 2). UV and IR studies showed that the distorted octahedral arrangement around cis-O2CoN4 core in compound 1 does not change during dehydration, which explains the reversible water loss and ability of compound 2 to rehydrate into compound 1. Compound 2 decomposes at ~ 240 °C in inert atmosphere giving final decomposition products, which are two modifications of nanosized metallic cobalt (hcp-15 nm, fcc-250 nm) and CoO (55 nm). The redox reaction results in N2 as an ammonia oxidation product. The decomposition intermediate is a cobalt(II) compound, Co2O1,14+δ(SO4)0.86 (δ = the oxygen surplus due to the presence of 2.8% of Co(III) ion). The same reaction in air atmosphere resulted in Co2O1.25+δ(SO4)0.75 (δ = the oxygen surplus due to the presence of 5.3% of Co(III) ion (compound 3a). Compound 3a is oxidized in air at 793 °C into Co3O4. The compound 3a exhibits catalytic activity in photodegradation in Congo red. Ezrin–Radixin–Moesin (ERM) proteins play an essential role in the cytoplasm by cross‐linking actin filaments with plasma membrane proteins. Research has identified the nuclear localization of ERMs, as well as the involvement of a single Drosophila ERM protein, Moesin, in nuclear mRNA exports. However, the question of how important the nuclear activity of ERM proteins are for the life of an organism has so far not been explored. Here, we present the first attempt to reveal the in vivo relevance of nuclear localization of Moesin in Drosophila. With the help of a nuclear export signal, we decreased the amount of Moesin in the nuclei of the animals. Furthermore, we observed various developmental defects, demonstrating the importance of ERM function in the nucleus for the first time. Transcriptome analysis of the mutant flies revealed that the lack of nuclear Moesin function leads to expression changes in nearly 700 genes, among them heat‐shock genes. We prepared and characterized a high-t tetragonal polymorph of diamminesilver(I) sulfate (a = 8.6004(4) Å, c = 6.2123(6) Å) (compound 1-HT). The phase transformation of the low-temperature polymorph of diamminesilver(I) sulfate (compound 1-LT) into the high-temperature polymorph (compound 1-HT) partly coincides with the initial stage of the thermal decomposition of 1-LT. Selecting appropriate conditions, we separated the two processes and determined the standard heat of formation of the LT polymorph (ΔH298 = −1208.92 kJ/mol) and the enthalpy of polymorphic transformation (ΔHLT→HT = 13.18 kJ/mol). The high-temperature XRD results showed that the lattice constants of the LT polymorph (1-LT) extend in the direction of a, b, and compress in the direction of c in the 1-HT polymorph. According to high temperature SXRD data, the Ag–Ag distance (in the direction of c) shortens from 3.20 Å (1-LT) to 3.11 Å (1-HT), while the extension in the a,b direction weakens/disrupts the N-H...O-S hydrogen bonds and the coordinative Ag...O-S interactions. The IR and Raman spectroscopic data confirm the weakening/breaking of N-H...O-S hydrogen bonds. A vacuum-assisted thermal deammoniation experiment of 1-LT in the preparation of monoamminesilver(I) sulfate showed that the product is the mixture of silver(I) sulfate and the initial diamminesilver(I) sulfate complex. The 1 : 1 type double salt of AgNO3 and NH4NO3 (NH4NO3 ⋅ AgNO3, compound 1) was prepared and its Raman spectrum was evaluated. IR and far‐IR spectra of compound 1 and its deuterated analog (ND4NO3 ⋅ AgNO3, compound 1‐D) were also studied. We identified two types of nitrate ions, one had a relatively strong, whereas the other is only weakly hydrogen bonded to the surrounding ammonium ions. The thermal decomposition features of compound 1 and its formation mechanism from [Ag(NH3)2]MnO4 (compound 2) at 80 °C with consecutive aqueous leaching of the decomposition residue has been elucidated. The solid phase decomposition product of compound 2 is presumably [Ag(NH3)NO3] (compound 3), which disproportionates into AgNO3 and [Ag(NH3)2]NO3 (compound 4). Compound 4 thermally decomposes in the solid phase into Ag+MnOx, while in aqueous solution its hydrolysis results in NH4NO3, which crystallizes out with an equimolar amount of AgNO3 formed during the disproportionation as compound 1. We present facile methods to obtain purified sporopollenin exine capsules, and provide mass balances for classical and novel purification procedures. An ionic liquid, tetrabutyl phosphonium hydroxide turned out to be the most effective in removing the intine wall. The sporopollenin capsules were investigated by fluorescent microscopy, AFM, solid-state NMR and infrared Raman spectroscopy. The latter two methods showed that sunflower and rape exines have different proportions of O-aliphatic and aromatic constituents. Purified exine capsules were coated with functionalized fluorophores. The procedures presented in this paper could contribute to further spread of the applications of this hollow, and chemically highly resistant material. [ABSTRACT FROM AUTHOR]

Age-related blood-brain barrier (BBB) disruption and cerebromicrovascular rarefaction contribute importantly to the pathogenesis of both vascular cognitive impairment and dementia (VCID) and Alzheimer's disease (AD). Recent advances in geroscience research enable development of novel interventions to reverse age-related alterations of the cerebral microcirculation for prevention of VCID and AD. To facilitate this research, there is an urgent need for sensitive and easy-to-adapt imaging methods that enable longitudinal assessment of changes in BBB permeability and brain capillarization in aged mice and that could be used in vivo to evaluate treatment efficiency. To enable longitudinal assessment of changes in BBB permeability in aged mice equipped with a chronic cranial window, we adapted and optimized two different intravital two-photon imaging approaches. By assessing relative fluorescence changes over the baseline within a volume of brain tissue, after qualitative image subtraction of the brain microvasculature, we confirmed that, in 24-mo-old C57BL/6J mice, cumulative permeability of the microvessels to fluorescent tracers of different molecular masses (0.3 to 40 kDa) is significantly increased compared with that of 5-mo-old mice. Real-time recording of vessel cross-sections showed that apparent solute permeability of single microvessels is significantly increased in aged mice vs. young mice. Cortical capillary density, assessed both by intravital two-photon microscopy and optical coherence tomography was also decreased in aged mice vs. young mice. The presented methods have been optimized for longitudinal (over the period of 36 wk) in vivo assessment of cerebromicrovascular health in preclinical geroscience research. [ABSTRACT FROM AUTHOR]
