The influence of wave force linearisation methods on fatigue damage predictions for jack-up rigs is investigated, using Sesam software as the basis for comparison. The possible risk of significantly underpredicting fatigue damage is highlighted. Dynamic spectral fatigue analysis methodology is applied to a representative jack-up rig in 30 and 70 m water depth in the North Sea. Transfer functions are computed using two approaches: regular waves of constant steepness with capping of wave height at low frequencies; stochastic linearisation based on a single sea state per wave scatter diagram. It is concluded that stochastic linearisation based on a single sea state per scatter diagram is insufficient to provide an adequate characterisation of nonlinear drag forces for the dynamic spectral fatigue analysis of jack-up rigs. Superharmonic resonance effects, due to nonlinearities associated with variable submergence and drag forces, are found to be significant when computing transfer functions of structural dynamic response. The immune system of ectotherms, particularly non-avian reptiles, remains poorly characterized regarding the genes involved in immune function, and their function in wild populations. We used RNA-Seq to explore the systemic response of Mojave desert tortoise (Gopherus agassizii) gene expression to three levels of Mycoplasma infection to better understand the host response to this bacterial pathogen. We found over an order of magnitude more genes differentially expressed between male and female tortoises (1,037 genes) than differentially expressed among immune groups (40 genes). There were 8 genes differentially expressed among both variables that can be considered sex-biased immune genes in this tortoise. Among experimental immune groups we find enriched GO biological processes for cysteine catabolism, regulation of type 1 interferon production, and regulation of cytokine production involved in immune response. Sex-biased transcription involves iron ion transport, iron ion homeostasis, and regulation of interferon-beta production to be enriched. More detailed work is needed to assess the seasonal response of the candidate genes found here. How seasonal fluctuation of testosterone and corticosterone modulate the immunosuppression of males and their susceptibility to Mycoplasma infection also warrants further investigation, as well as the importance of iron in the immune function and sex-biased differences of this species. Finally, future transcriptional studies should avoid drawing blood from tortoises via subcarapacial venipuncture as the variable aspiration of lymphatic fluid will confound the differential expression of genes. Immune function plays an important role in an animal's defense against infectious disease. In reptiles, immune responses may be complex and counterintuitive, and diagnostic tools used to identify infection, such as induced antibody responses are limited. Recent studies using gene transcription profiling in tortoises have proven useful in identifying immune responses to various intrinsic and extrinsic stressors. As part of a larger experiment with Mojave desert tortoises (Gopherus agassizii), we facilitated the transmission of the pathogenic bacteria, Mycoplasma agassizii (Myag), to naïve adults and measured innate and induced immune reactions over time. Specifically, we evaluated clinical condition, presence of Myag in the nasal/oral cavity, induced antibody responses specific to Myag, and measured molecular reactions (gene transcript profiles) in 15 captive tortoises classified as naïve, exposed, or infected and 14 wild tortoises for comparison. Myag was confirmed inside the nasal/oral cavity in exposed tortoises within 30–60 days of introduction to infected animals, yet we did not detect Myag specific induced antibody responses in these individuals until 420–595 days post exposure. Surprisingly, we found no overall differences in the gene transcript profiles between our experimental treatment groups throughout this study. This work highlights the complexities in assessing immune function and diagnosing pathogen related infections in tortoises and other reptiles. Scholars have recognized T.F. Torrance as a ressourcement thinker in his later work while overlooking his earlier historical studies. In this article, I argue that in Calvin's Doctrine of Man (1949) Torrance presents a rereading of Calvin in order to advance contemporary theological understanding and facilitate the reception of Karl Barth in postwar Scotland. In that work, Torrance attempts to resolve the debate between Brunner and Barth over natural theology by reinterpreting Calvin's anthropology. Ceramic pot filters (CPFs) are an effective means of household water treatment, but the characterization of CPF lifetimes is ongoing. This paper describes a lifetime field study in Guatemala which was made possible by a collaboration between researchers, CPF-using households, and local non-governmental organizations (NGOs). Disinfection data were collected periodically for two years using field coliform enumeration kits as were flow rate data with the assistance of NGO staff. Consumer acceptance was characterized by surveying householders in the four subject villages at the beginning and end of the study. Flow rate data showed that average CPF flow rates decreased below the recommended minimum of 1 L h-1 after 10 months of use; however, the survey results indicated that the consumers were tolerant of the lower flow rates, and it is reasonable to assume that the daily volume of treated water can be readily increased by refilling the CPFs more frequently. Of greater concern was the finding that disinfection efficacy decreased below the recommended bacterial reduction after 14 months of use because it would not be obvious to users that effectiveness had declined. Historical mining activity (1850-1970) in the now inactive Tri-State Mining District provided an ongoing source of lead and zinc to the environment including the US Environmental Protection Agency Superfund site located in Cherokee County, southeast Kansas, USA. The resultant contamination adversely affected biota and caused human health problems and risks. Remediation in the Superfund site requires an understanding of the magnitude and extent of contamination. To provide some of the required information, a series of sediment and soil investigations were conducted in and near the Superfund site to characterize lead and zinc contamination in the aquatic and floodplain environments along the main-stem Spring River and its major tributaries. In the Superfund site, the most pronounced lead and zinc contamination, with concentrations that far exceed sediment quality guidelines associated with potential adverse biological effects, was measured for streambed sediments and floodplain soils located within or downstream from the most intensive mining-affected areas. Tributary streambeds and floodplains in affected areas are heavily contaminated with some sites having lead and zinc concentrations that are an order of magnitude (or more) greater than the sediment quality guidelines. For the main-stem Spring River, the streambed is contaminated but the floodplain is mostly uncontaminated. Measured lead and zinc concentrations in streambed sediments, lakebed sediments, and floodplain soils documented a persistence of the post-mining contamination on a decadal timescale. These results provide a basis for the prioritization, development, and implementation of plans to remediate contamination in the affected aquatic and floodplain environments within the Superfund site. Habitat disturbance from development, resource extraction, off-road vehicle use, and energy development ranks highly among threats to desert systems worldwide. In the Mojave Desert, United States, these disturbances have promoted the establishment of nonnative plants, so that native grasses and forbs are now intermixed with, or have been replaced by invasive, nonnative Mediterranean grasses. This shift in plant composition has altered food availability for Mojave Desert tortoises (Gopherus agassizii), a federally listed species. We hypothesized that this change in forage would negatively influence the physiological ecology, immune competence, and health of neonatal and yearling tortoises. To test this, we monitored the effects of diet on growth, body condition, immunological responses (measured by gene transcription), and survival for 100 captive Mojave tortoises. Tortoises were assigned to one of five diets: native forbs, native grass, invasive grass, and native forbs combined with either the native or invasive grass. Tortoises eating native forbs had better body condition and immune functions, grew more, and had higher survival rates (>95%) than tortoises consuming any other diet. At the end of the experiment, 32% of individuals fed only native grass and 37% fed only invasive grass were found dead or removed from the experiment due to poor body conditions. In contrast, all tortoises fed either the native forb or combined native forb and native grass diets survived and were in good condition. Health and body condition quickly declined for tortoises fed only the native grass (Festuca octoflora) or invasive grass (Bromus rubens) with notable loss of fat and muscle mass and increased muscular atrophy. Bromus rubens seeds were found embedded in the oral mucosa and tongue in most individuals eating that diet, which led to mucosal inflammation. Genes indicative of physiological, immune, and metabolic functions were transcribed at lower levels for individuals fed B. rubens, indicating potential greater susceptibility to disease or other health-related problems. This study highlights the negative indirect effects of invasive grasses, such as red brome, in desert ecosystems, and provides definitive evidence of a larger negative consequence to health, survival, and ultimately population recruitment for Mojave Desert tortoises than previously understood. Tortoises are susceptible to a wide variety of environmental stressors, and the influence of human disturbances on health and survival of tortoises is difficult to detect. As an addition to current diagnostic methods for desert tortoises, we have developed the first leukocyte gene transcription biomarker panel for the desert tortoise ( Gopherus agassizii), enhancing the ability to identify specific environmental conditions potentially linked to declining animal health. Blood leukocyte transcript profiles have the potential to identify physiologically stressed animals in lieu of clinical signs. For desert tortoises, the gene transcript profile included a combination of immune or detoxification response genes with the potential to be modified by biological or physical injury and consequently provide information on the type and magnitude of stressors present in the animal's habitat. Blood from 64 wild adult tortoises at three sites in Clark County, NV, and San Bernardino, CA, and from 19 captive tortoises in Clark County, NV, was collected and evaluated for genes indicative of physiological status. Statistical analysis using a priori groupings indicated significant differences among groups for several genes, while multidimensional scaling and cluster analyses of transcription C values indicated strong differentiation of a large cluster and multiple outlying individual tortoises or small clusters in multidimensional space. These analyses highlight the effectiveness of the gene panel at detecting environmental perturbations as well as providing guidance in determining the health of the desert tortoise. ABSTRACT Wildfires burned 24,254 ha of critical habitat designated for the recovery of the threatened Mojave desert tortoise ( Gopherus agassizii) in southern Nevada during 2005. The proliferation of non-native annual grasses has increased wildfire frequency and extent in recent decades and continues to accelerate the conversion of tortoise habitat across the Mojave Desert. Immediate changes to vegetation are expected to reduce quality of critical habitat, yet whether tortoises will use burned and recovering habitat differently from intact unburned habitat is unknown. We compared movement patterns, home-range size, behavior, microhabitat use, reproduction, and survival for adult desert tortoises located in, and adjacent to, burned habitat to understand how tortoises respond to recovering burned habitat. Approximately 45% of home ranges in the post-fire environment contained burned habitat, and numerous observations ( n = 12,223) corroborated tortoise use of both habitat types (52% unburned, 48% burned). Tortoises moved progressively deeper into burned habitat during the first 5 years following the fire, frequently foraging in burned habitats that had abundant annual plants, and returning to adjacent unburned habitat for cover provided by intact perennial vegetation. However, by years 6 and 7, the live cover of the short-lived herbaceous perennial desert globemallow ( Sphaeralcea ambigua) that typically re-colonizes burned areas declined, resulting in a contraction of tortoise movements from the burned areas. Health and egg production were similar between burned and unburned areas indicating that tortoises were able to acquire necessary resources using both areas. This study documents that adult Mojave desert tortoises continue to use habitat burned once by wildfire. Thus, continued management of this burned habitat may contribute toward the recovery of the species in the face of many sources of habitat loss. Published 2015. We present an application of mechanistic modeling and nonlinear longitudinal regression in the context of biomedical response-to-challenge experiments, a field where these methods are underutilized. In this type of experiment, a system is studied by imposing an experimental challenge, and then observing its response. The combination of mechanistic modeling and nonlinear longitudinal regression has brought new insight, and revealed an unexpected opportunity for optimal design. Specifically, the mechanistic aspect of our approach enables the optimal design of experimental challenge characteristics (e.g., intensity, duration). This article lays some groundwork for this approach. We consider a series of experiments wherein an isolated rabbit heart is challenged with intermittent anoxia. The heart responds to the challenge onset, and recovers when the challenge ends. The mean response is modeled by a system of differential equations that describe a candidate mechanism for cardiac response to anoxia challenge. The cardiac system behaves more variably when challenged than when at rest. Hence, observations arising from this experiment exhibit complex heteroscedasticity and sharp changes in central tendency. We present evidence that an asymptotic statistical inference strategy may fail to adequately account for statistical uncertainty. Two alternative methods are critiqued qualitatively (i.e., for utility in the current context), and quantitatively using an innovative Monte-Carlo method. Wildlife translocation is increasingly used to mitigate disturbances to animals or habitat due to human activities, yet little is known about the extent to which translocating animals causes stress. To understand the relationship between physiological stress and translocation, we conducted a multiyear study (2007-2009) using a population of desert tortoises ( Gopherus agassizii) near Fort Irwin, California. Blood samples were collected from adult tortoises in three treatment groups (resident, translocated and control) for 1 year prior to and 2 years after translocation. Samples were analyzed by radioimmunoassay for plasma total corticosterone ( CORT), a glucocorticoid hormone commonly associated with stress responses in reptiles. CORT values were analyzed in relation to potential covariates (animal sex, date, behavior, treatment, handling time, air temperature, home-range size, precipitation and annual plant production) among seasons and years. CORT values in males were higher than in females, and values for both varied monthly throughout the activity season and among years. Year and sex were strong predictors of CORT, and translocation explained little in terms of CORT. We examined loggerhead nest temperatures and hatchling sex ratios in an effort to more accurately predict hatchling sex ratios produced from 2 barrier islands in the northern management unit (Blackbeard Island National Wildlife Refuge and Wassaw National Wildlife Refuge, Georgia, United States) from 2000 to 2004. Temperature data loggers were placed into 169 nests to monitor incubation temperatures. Average critical period temperatures ranged from 26.3°C to 33.2°C (mean ± SE, 29.2° ± 0.1°C) and indicated seasonal variation in sex ratios. The sex of 669 hatchlings found dead in nests was histologically evaluated (n = 212 nests; 14-90 nests! yr). The sex ratios varied from 0% to 100% female per nest (n = 1-53 hatchlings/nest) and average sex ratio for all nests ranged from 55.5% female in 2003 to 85.4% female in 2002.. In addition to monitoring nest temperature, 10 hatchlings per nest were euthanized to verify sex during 2003 on Blackbeard Island National Wildlife Refuge (n = 10 nests) and 2004 on Wassaw National Wildlife Refuge (n = 9 nests). Sex ratios were analyzed by using an advanced statistical program for evaluating temperature-dependent sex determination and indicated a 1:1 temperature (temperature that produces a 1:1 sex ratio) of 28.9°C. We offer an equation for predicting northern management unit hatchling loggerhead sex ratios by using critical period temperature and tested its validity. Sixteen of 18 nests (n = 10 hatchlings/nest) showed no significant difference between the predicted sex ratios based on the equation vs. sex ratios obtained through histology. Our data indicated that rookery beaches north of Florida are important areas for the production and recruitments of male loggerhead hatchlings into the overall western North Atlantic Ocean and nests deposited earliest within a nesting season are primary contributors of male turtles. The Dust Trajectory Sensor (DTS) instrument is developed for the measurement of the velocity vector of cosmic dust particles. The trajectory information is imperative in determining the particles' origin and distinguishing dust particles from different sources. The velocity vector also reveals information on the history of interaction between the charged dust particle and the magnetospheric or interplanetary space environment. The DTS operational principle is based on measuring the induced charge from the dust on an array of wire electrodes. In recent work, the DTS geometry has been optimized [S. Auer, E. Grün, S. Kempf, R. Srama, A. Srowig, Z. Sternovsky, and V Tschernjawski, Rev. Sci. Instrum. 79, 084501 (2008)] and a method of triggering was developed [S. Auer, G. Lawrence, E. Grün, H. Henkel, S. Kempf, R. Srama, and Z. Sternovsky, Nucl. Instrum. Methods Phys. Res. A 622, 74 (2010)]. This article presents the method of analyzing the DTS data and results from a parametric study on the accuracy of the measurements. A laboratory version of the DTS has been constructed and tested with particles in the velocity range of 2-5 km/s using the Heidelberg dust accelerator facility. The methodology of auxiliary signal design for robust fault detection based on a multi-model (MM) formulation of normal and faulty systems is used to study the problem of incipient fault detection. The fault is modelled as a drift in a system parameter, and an auxiliary signal is to be designed to enhance the detection of variations in this parameter. It is shown that it is possible to consider the model of the system with a drifted parameter as a second model and use the MM framework for designing the auxiliary signal by considering the limiting case as the parameter variation goes to zero. The result can be applied very effectively to many early detection problems where small parameter variations should be detected. The potential impact of exposure to heavy metals and health problems was evaluated at the Tar Creek Superfund site, Ottawa County, Oklahoma, USA. Observed versus expected mortality was calculated for selected conditions in the County and exposed cities. Excess mortality was found for stroke and heart disease when comparing the exposed County to the state but not when comparing the exposed cities to the nonexposed rest of the County. However, sample sizes in the exposed area were small, population emigration has been ongoing, and geographic coding of mortality data was incomplete. In an exposed community, 62.5% of children under the age of 6 years had blood lead levels exceeding 10 μg/dl. The relationships between heavy-metal exposure and children’s health and chronic disease in adults are suggestive that a more thorough investigation might be warranted. A number of possible environmental and health studies are suggested, including those focusing on possible central nervous system impacts. Unfortunately, the exposed population is dispersing. One lesson learned at this site is that health studies need to be conducted as soon as possible after an environmental problem is identified to both study the impact of the most acute exposures and to maximize study sample size—including those exposed to higher doses—and minimize the loss of individuals to follow-up. The method of in situ time-resolved high-energy X-ray diffraction, using the intrinsically highly collimated X-ray beam generated by the European Synchrotron Radiation Facility, is demonstrated. A specially designed cell, which allows the addition of liquid components, has been used to study the reaction mechanisms of a foamed bioactive calcia–silica sol–gel glass immersed in simulated body fluid. Analysis of the X-ray diffraction data from this experiment provides atomic distances, via the pair correlation functions, at different stages of the dissolution of the glass and of the associated calcium phosphate, and ultimately hydroxyapatite, i.e. bone mineral, formation. Hence, changes in the atomic scale structure can be analysed as a function of reaction time, giving an insight into the evolution of the structure of both the glass matrix and the hydroxyapatite surface growth. Performance metrics and evaluation criteria are used to develop a novel taxonomy that classifies barrier mechanisms into categories ranging from simple Static-Binding Idle-Tasking methods to robust Dynamic-Binding Any-Tasking methods. Such capabilities include support for multiple barriers, reconfigurable and reusable synchronization resources, and provisions for dynamic process binding. Based on these features, the existing software-based approaches, dedicated-hardware mechanisms, and hybrid hardware/software techniques for synchronization are assessed in order of increasing complexity. 2-(Thiocyanomethylthio)benzothiazole (TCMTB) is a biocide used in the leather, pulp and paper, and water-treatment industries. TCMTB may enter aquatic ecosystems during its manufacture and use. TCMTB is environmentally unstable; therefore, it is important to evaluate the toxicity of the more persistent degradation products. This study compared the toxicity of TCMTB with its degradation products 2-mercaptobenzothiazole (2-MBT), 2-(methylthio) benzothiazole (MTBT), benzothiazole (BT), and 2-hydroxybenzothiazole (HOBT). Toxicity was determined using Ceriodaphnia dubia 48-hour acute and 7-day chronic test protocols. TCMTB was the most toxic compound evaluated in both the acute and chronic tests with EC50s of 15.3 and 9.64 μg/L, respectively. 2-MBT, the first degradation product, was the second most toxic compound with acute and chronic EC50s of 4.19 and 1.25 mg/L, respectively. The toxicity of MTBT and HOBT were similar with acute EC50s of 12.7 and 15.1 mg/L and chronic EC50s of 6.36 and 8.31 mg/L, respectively. The least toxic compound was BT with acute and chronic EC50s of 24.6 and 54,9 mg/L, respectively. TCMTB was orders of magnitude more toxic than its degradation products. Toxicity data on these benzothiazole degradation products is important because of concerns regarding their release, degradation, persistence, and non-target organism effects in aquatic ecosystems. In areas of the country where the U.S. Department of Energy has classified the available wind resources as Class 3 or greater, the use of wind turbines to provide power to relatively small remediation systems such as groundwater circulation wells may be technically and economically feasible. Groundwater circulation wells are a good candidate technology to couple with renewable energy, because the remediation system removes contamination from the subject aquifer with no net loss of the groundwater resource, while the wind turbine does not create potentially harmful air emissions. Wind data collected in the vicinity of the former Nebraska Ordnance Plant Superfund site were used to select a wind turbine system to provide a portion of the energy necessary to power a groundwater circulation well located in an area of high trichloroethylene groundwater contamination. Because utility power was already installed at the remediation system, a 10 kW grid inter-tie wind turbine system supplements the utility system without requiring batteries for energy storage. The historical data from the site indicate that the quantity of energy purchased correlates poorly with the quantity of groundwater treated. Preliminary data from the wind turbine system indicate that the wind turbine provides more energy than the remediation system treatment components and the well submersible pump require on a monthly average. The preliminary results indicate that the coupling of wind turbines and groundwater circulation wells may be an attractive alternative in terms of the system operation time, cost savings, and contaminant mass removal. © 2004 Wiley Periodicals, Inc. Wild type rat lens main intrinsic protein (MIP) and MIP mutated (F73I, F75L) to resemble the glycerol facilitator of Escherichia coli in the region of the NPA1 box were used to investigate the topology of MIP in the membrane of Spodoptera frugiperda (Sf21) cells using the baculovirus expression system and expression in mouse erythroid leukaemia cells (MEL C88). Differential fixation for staining was used, with paraformaldehyde for externally exposed antigenic sites, and acetone for both externally and internally exposed protein antigenic sites. Immunofluorescence using antibodies to synthetic MIP peptides showed that wild type MIP had a six transmembrane topography. The N- and C-termini were intracellular in both expression systems, and both NPA boxes were found to be extracellular. These results show that residues around the NPA1 box can influence the folding of the MIP in the membrane, and provide structural evidence for the poor water transport properties of MIP, as the NPA boxes lie outside the plane of the membrane. Wild-type rat lens main intrinsic protein (MIP) was heterologously expressed in the membrane of Spodoptera frugiperda (Sf21) cells using the baculovirus expression system and in mouse erythroid leukaemia cells (MEL C88). Both MEL and Sf21 cell lines expressing wild-type MIP were investigated for the conductance of ions using a whole cell patch clamp technique. An increase in conductance was seen in both expression systems, particularly on lowering the pH to 6.3. In Sf21 cells, addition of antibodies to the NPA1 box resulted in a reduction of current flow. These results suggest that MIP has pH-dependent ion channel activity, which involves the NPA1 box domain. [Copyright &y& Elsevier]

The wave surface profiles associated with extreme relative motion between a slender stationary vessel and the adjacent wave surface are investigated for long-crested head seas. The methodology is based upon an established statistical relationship for a Gaussian random process which indi-cates that in the vicinity of an extreme event the most probable shape of the time history approaches that of the autocorrelation function. Attention is given to the relative motion at a longitudinal position which is representative of the No. 1 hatch cover location on a bulk carrier. Second-order corrections to the wave surface profiles are used to provide estimates of nonlinear relative motions. It is shown that extreme relative motion at the hatch cover location is associated with the vessel encountering a steep-fronted wave with pronounced asymmetry in the horizontal and vertical directions. In order to evaluate the exceedence probabilities, it is hypothesized that the peaks of the nonlinear relative motions are closely correlated with the peaks of an underlying linear process. The overall methodology is applied to an investigation of relative motion exceedence probabilities for vessel lengths of 250 m and 300 m over a range of survival seastates. [ABSTRACT FROM AUTHOR]
