Ocean currents play a role in both facilitating and impeding connectivity of marine organisms' populations, serving as key drivers and potential barriers. The emergence of Taiwan around five million years ago (Mya) impacted the Kuroshio Current which later shaped the distribution of marine organisms. In this study, we examined how the Kuroshio Current influenced the distribution and population divergence of the marine red alga Dichotomaria elegans (Galaxauraceae, Nemaliales). Through phylogenetic analysis, species delimitation, and morphological comparison of Taiwanese species, we determined its phylogenetic position relative to other species within the D. marginata complex of the genus Dichotomaria. We uncovered the hidden cryptic diversity within the D. marginata complex in Taiwan and proposed the new combinations D. elegans and D. latifolia based on Galaxaura elegans and G. latifolia. Genetic analysis based on three loci (mitochondrial cox2-3 and cox1, and plastid rbcL) from 32 locations across the Philippines, Taiwan, and Japan revealed three allopatric lineages within D. elegans. The first lineage is restricted to the northern Philippines and southern Taiwan, while the second has a broader latitudinal range along the Kuroshio Current. The third lineage is found in southeast Luzon, Philippines, close to the bifurcation of the North Equatorial Current, near the start of the Kuroshio Current. The divergence of the first two lineages during the mid-Pliocene (~3.7 Mya) coincided with the emergence of Taiwan, with the current dynamics accentuating their separation. The Kuroshio Current exhibited a dual impact on the biogeography of D. elegans: promoting the dispersal of the second lineage towards higher latitudes and preventing the first lineage from moving northward. Principal component analysis showed that habitat conditions and morphologies differed between the two D. elegans lineages. This study provides insights into the cryptic diversity of Dichotomaria in Taiwan, as well as the genetic divergence, biogeographical patterns, and ecological associations of D. elegans, highlighting the interplay of island formation and currents shaping the diversification of seaweeds in the Northwest Pacific Ocean. Caulerpa J.V. Lamouroux is a genus of green macroalgae belonging to the family Caulerpaceae in the order Bryopsidales. The genus comprises 104 currently accepted species, of which 51 have been recorded from Pacific Islands. Among these islands, French Polynesia is found in the middle of the South Pacific Ocean and includes five archipelagos (i.e., the Austral, Gambier, Marquesas, Society, and Tuamotu Islands) where seaweed inventories have reported a total of 16 Caulerpa species so far based on morphology. Here, based on a sampling covering the five archipelagos of French Polynesia, we attempt to (i) verify the taxonomy of Caulerpa species present in these regions based on phylogeny, (ii) describe in more detail the specific diversity between the five archipelagos, and (iii) provide a morphological identification tool for these species. We successfully obtained 134 new tufA sequences for phylogenetic analyses, which corresponded to 13 species. We propose to resurrect C. pickeringii Harvey & Bailey for representatives of C. webbiana var. pickeringii and classify it in the Caulerpa subgenus Araucarioideae. We also transfer C. seuratii to C. pickeringii based on genetic results. The Andaman Sea-Malacca Strait is an area with high maritime complexity and rich seaweed biodiversity. The brown seaweed Sargassum plagiophyllum C.Agardh occurs abundantly in the intertidal in the area. However, its population genetics has not been studied. Here, we used the nuclear Internal Transcribed Spacer 2 of the ribosomal cistron ITS2 and the mitochondrial cytochrome c oxidase subunit 1 gene cox1 as genetic markers to determine the population genetic structure along the entire west coast of Thailand. A total of 351 individuals from the ten populations were genotyped. Nine different cox1 haplotypes and 17 different ITS2 ribotypes were found. Generally, S. plagiophyllum populations were found to have low genetic diversity. Two lineages were found based on ITS2 and two cryptic diversities were inferred from cox1 data. Most individuals belonged to the one single ribotype and haplotype. Ocean currents are believed to influence the S. plagiophyllum distributions and connectivity between populations. Higher phosphate concentrations during the northeast monsoon season could promote its growth and propagation. Our study suggested that the temporal and spatial transformation of the marine environment in the Andaman Sea influences the distribution, biomass, and population genetic structure of S. plagiophyllum, our results are beneficial for the management of seaweed resources. Background: Sargassum polycystum C. Agardh and Sargassum plagiophyllum C. Agardh are inhabitants of tropical coastal areas, their populations are negatively influenced by global warming and marine environment changes. The mitochondrial and chloroplast genomes of these species have not been sequenced. Results: The mitochondrial genomes of S. polycystum and S. plagiophyllum were 34,825 bp and 34,862 bp, respectively, and their corresponding chloroplast genomes were 124,493 bp and 124,536 bp, respectively. The mitochondrial and chloroplast genomes of these species share conserved synteny, sequence regions and gene number when compared with the organellar genomes of other Sargassum species. Based on sequence analysis of 35 protein-coding genes, we deduced that S. polycystum and S. plagiophyllum were closely related with S. ilicifolium; these species diverged approximately 0.3 million years ago (Ma; 0.1–0.53 Ma) during the Pleistocene period (0.01–2.59 Ma). Rates of synonymous and non-synonymous substitutions in the mitochondrial genome of the Sargassum genus were 3 times higher than those in the chloroplast genome. In the mitochondrial genome, rpl5, rpl31 and rps11 had the highest synonymous substitution rates. In the chloroplast genome, psaE, rpl14 and rpl27 had the highest synonymous substitution rates. Conclusions: Phylogenetic analysis confirms the close relationship between the two sequenced species and S. ilicifolium. Both synonymous and non-synonymous substitution rates show significant divergence between the group of mitochondrial genomes versus the group of chloroplast genomes. The deciphering of complete mitochondrial and chloroplast genomes is significant as it advances our understanding of the evolutionary and phylogenetic relationships between species of brown seaweeds. Seagrasses play a vital role in structuring coastal marine ecosystems, but their distributional range and genetic diversity have declined rapidly in recent decades. To improve conservation of seagrass species, it is important to predict how climate change may impact their ranges. Such predictions are typically made with correlative species distribution models (SDMs), which can estimate a species' potential distribution under present and future climatic scenarios given species' presence data and climatic predictor variables. However, these models are typically constructed with species‐level data, and thus ignore intraspecific genetic variability, which can give rise to populations with adaptations to heterogeneous climatic conditions. Here, we explore the link between intraspecific adaptation and niche differentiation in Thalassia hemprichii, a seagrass broadly distributed in the tropical Indo‐Pacific Ocean and a crucial provider of habitat for numerous marine species. By retrieving and re‐analysing microsatellite data from previous studies, we delimited two distinct phylogeographical lineages within the nominal species and found an intermediate level of differentiation in their multidimensional environmental niches, suggesting the possibility for local adaptation. We then compared projections of the species' habitat suitability under climate change scenarios using species‐level and lineage‐level SDMs. In the Central Tropical Indo‐Pacific region, models for both levels predicted considerable range contraction in the future, but the lineage‐level models predicted more severe habitat loss. Importantly, the two modelling approaches predicted opposite patterns of habitat change in the Western Tropical Indo‐Pacific region. Our results highlight the necessity of conserving distinct populations and genetic pools to avoid regional extinction due to climate change and have important implications for guiding future management of seagrasses. The genus Caulerpa is highly diverse, especially in the Caribbean Sea, the Indo-Malay archipelago and the temperate waters of southern Australia. Currently, more than 100 species are recognized worldwide. Despite recent advances in taxonomy and phylogeny, there is still confusion about the delimitation and identification of some species due to tremendous phenotypic plasticity. Caulerpa is known to occupy a wide range of environmental niches and to have great invasive potential. More studies are necessary to understand better the ecology (invasiveness, the functional role of its microbiome) and the biology (reproduction, life cycle, and metabolism) of Caulerpa species, especially for economical species. Very few species, mainly C. lentillifera and C. racemosa known as green caviar, have been cultivated. Caulerpa is consumed mostly in Asia and Oceania, but its popularity tends to be on the increase in western countries with new nutritional practices (vegetarians, vegans, health foods). Being naturally enriched in essential nutrients and various health-promoting compounds, Caulerpa species are promising candidates for the design of functional foods and in the health and well-being sectors. The diversity of Caulerpa offers a remarkable potential for valorization with innovation opportunities. The marine brown algal genus Colpomenia has a worldwide distribution, with five species reported in Korea and Japan. However, no studies to date attempted to identify the number of species and geographical distribution of Colpomenia along Chinese coast. To fill the biodiversity knowledge gap, we analyzed 63 mitochondrial cox3 and 62 mitochondrial atp6 sequences of Colpomenia specimens collected from 30 localities along the Chinese coast. Maximum likelihood and Bayesian inference trees suggest the presence of at least three Colpomenia species (i.e., C. peregrina, C. claytoniae, and C. sinuosa) in China. C. peregrina and C. claytoniae are documented for the first time. C. sinuosa was only found in the South China Sea and its distribution didn't overlap with that of C. peregrina which was found in the Yellow-Bohai Sea and the East China Sea. C. claytoniae appears to be confined to three isolated islands in the East and the South China Sea, where it occurs in sympatry with, respectively, C. peregrina and C. sinuosa. Future study can focus on comparing ecophysiological differences of Colpomenia species in response to environmental variables and exploring possible genetic hybridization / introgression at inter-specific contact zones. A phylogenetic analysis of DNA sequence data revealed that the morphological Caulerpa entities, previously reported as C. corynephora Montagne (or C. racemosa var. corynephora (Montagne) Weber-van Bosse) and C. ashmeadii Harvey from mangrove channels on the Thai Andaman Sea coast, are both representatives of C. macrodisca Decaisne. The analyzed Andaman Sea specimens formed one of four subclades in a monophyletic C. macrodisca clade that was sister to C. megadisca Belton & Gurgel. Ramuli arrangement of C. macrodisca from the Andaman Sea was mostly in opposite pairs and sometimes distichous. The shape of the ramuli varied from almost cylindrical to clavate or turbinate. No pyrenoid was observed. Caulerpa macrodisca with characteristic large peltate ramuli has only been reported in Thailand from the Gulf of Thailand. Its identity was confirmed with DNA sequence data from a herbarium specimen. It is recommended to refer to the three morphological C. macrodisca entities as ecads; i.e., C. macrodisca ecad macrodisca, ecad corynephora, and ecad ashmeadii. [ABSTRACT FROM AUTHOR]

Abstract: Aim: Biogeographical processes underlying Indo‐Pacific biodiversity patterns have been relatively well studied in marine shallow water invertebrates and fishes, but have been explored much less extensively in seaweeds, despite these organisms often displaying markedly different patterns. Using the marine red alga Portieria as a model, we aim to gain understanding of the evolutionary processes generating seaweed biogeographical patterns. Our results will be evaluated and compared with known patterns and processes in animals. Location: Indo‐Pacific marine region. Methods: Species diversity estimates were inferred using DNA‐based species delimitation methods. Historical biogeographical patterns were inferred based on a six‐gene time‐calibrated phylogeny, distribution data of 802 specimens, and probabilistic modelling of geographical range evolution. The importance of geographical isolation for speciation was further evaluated by population genetic analyses at the intraspecific level. Results: We delimited 92 candidate species, most with restricted distributions, suggesting low dispersal capacity. Highest species diversity was found in the Indo‐Malay Archipelago (IMA). Our phylogeny indicates that Portieria originated during the late Cretaceous in the area that is now the Central Indo‐Pacific. The biogeographical history of Portieria includes repeated dispersal events to peripheral regions, followed by long‐term persistence and diversification of lineages within those regions, and limited dispersal back to the IMA. Main conclusions: Our results suggest that the long geological history of the IMA played an important role in shaping Portieria diversity. High species richness in the IMA resulted from a combination of speciation at small spatial scales, possibly as a result of increased regional habitat diversity from the Eocene onwards, and species accumulation via dispersal and/or island integration through tectonic movement. Our results are consistent with the biodiversity feedback model, in which biodiversity hotspots act as both “centres of origin” and “centres of accumulation,” and corroborate previous findings for invertebrates and fish that there is no single unifying model explaining the biological diversity within the IMA. 1. In the planning of marine protected areas for the conservation of coral reef systems, it is important to be able to distinguish between certain bioregions, i.e. regions with distinct species assemblages. This was done off western Halmahera (Moluccas, Indonesia), where three such bioregions were distinguished based on species inventories of 41 coral reef sites. 2. The relative value of species belonging to different trophic groups was examined with regard to their possible role as indicators for these three regions. The study focused on ascidians (Ascidiacea), macroalgae, mushroom corals (Fungiidae), and a selection of coral-associated gastropods (Epitoniidae and Coralliophilidae). 3. The best results for the detection of bioregions were obtained when datasets of all four trophic groups were pooled. When comparing the taxa and their indicator values, ascidians were the most suitable, followed by macroalgae, corals and gastropods with 98, 83, 71 and 66 % certainties, respectively. The occurrence of 17 species correlated strongly with the bioregions, which therefore were identified as potential indicator species consisting of 13 ascidians, three macroalgae, and one mushroom coral. These data suggest that ascidians have a significant value as indicators to evaluate bioregion boundaries. 4. Water quality measurements indicated that salinity and turbidity could be responsible for at least part of the differences between the species assemblages in the three bioregions. The molecular phylogeny of brown algae was examined using concatenated DNA sequences of seven chloroplast and mitochondrial genes ( atpB, psaA, psaB, psbA, psbC, rbcL, and cox1). The study was carried out mostly from unialgal cultures; we included Phaeostrophion irregulare and Platysiphon glacialis because their ordinal taxonomic positions were unclear. Overall, the molecular phylogeny agreed with previously published studies, however, Platysiphon clustered with Halosiphon and Stschapovia and was paraphyletic with the Tilopteridales. Platysiphon resembled Stschapovia in showing remarkable morphological changes between young and mature thalli. Platysiphon, Halosiphon and Stschapovia also shared parenchymatous, terete, erect thalli with assimilatory filaments in whorls or on the distal end. Based on these results, we proposed a new order Stschapoviales and a new family Platysiphonaceae. We proposed to include Phaeostrophion in the Sphacelariales, and we emended the order to include this foliose member. Finally, using basal taxa not included in earlier studies, the origin and divergence times for brown algae were re-investigated. Results showed that the Phaeophyceae branched from Schizocladiophyceae ~260 Ma during the Permian Period. The early diverging brown algae had isomorphic life histories, whereas the derived taxa with heteromorphic life histories evolved 155-110 Ma when they branched from the basal taxa. Based on these results, we propose that the development of heteromorphic life histories and their success in the temperate and cold-water regions was induced by the development of the remarkable seasonality caused by the breakup of Pangaea. Most brown algal orders had diverged by roughly 60 Ma, around the last mass extinction event during the Cretaceous Period, and therefore a drastic climate change might have triggered the divergence of brown algae. Although recent molecular studies have indicated the presence of a number of distinct species within the Caulerpa racemosa-peltata complex, due to the difficulties presented by high levels of phenotypic plasticity and the large number of synonyms, infra-specific taxa, and names of uncertain affinity, taxonomic proposals are yet to be made. In this study, we aimed to resolve the taxonomy of the complex and provide an example of how historical nomenclature can best be integrated into molecular based taxonomies. We accomplished this by first determining the number of genetic species within our globally sampled data set through a combination of phylogenetic and species-delimitation approaches of partial elongation factor TU and RUBISCO large subunit gene sequences. Guided by these results, comparative morphological examinations were then undertaken to gauge the extent of phenotypic plasticity within each species, as well as any morphological overlap between them. Our results revealed the presence of 11 distinct species within the complex, five of which showed high levels of phenotypic plasticity and partial overlap with other species. On the basis of observations of a large number of specimens, including type specimens/descriptions, and geographic inferences, we were able to confidently designate names for the lineages. Caulerpa peltata, C. imbricata and C. racemosa vars. laetevirens, occidentalis and turbinata were found to represent environmentally induced forms of a single species, for which the earlier-described C. chemnitzia, previously regarded as a synonym of C. racemosa var. turbinata, is reinstated. C. cylindracea, C. lamourouxii, C. macrodisca, C. nummularia and C. oligophylla are also reinstated and two new species, C. macra stat. nov. and C. megadisca sp. nov., are proposed. Molecular characterization (plastid-encoded tufA gene) of New Caledonian members of the Caulerpa racemosa-Caulerpa peltata complex identified five lineages, each possibly harboring multiple species-level entities. On a global scale, the complex encompassed six lineages, two of which were pantropical and the remainder seemingly were restricted to the Indo-Pacific basin. Caulerpa racemosa var. cylindracea, a fast-spreading introduced taxon in the Mediterranean Sea and the Canary Islands, was previously thought to be restricted to southwestern Australia; it was newly reported here for New Caledonia and several northern Australian locations, including the Great Barrier Reef. Two new species of Padina - 1. Padina sulcata sp. nov. and 2. P. calcarea sp. nov. - from Malaysia, Indonesia and Palau were described based on morphological and molecular phylogenetic observations. Padina sulcata was a three-layered species characterised by a covering of thick fibrous hairs from the base to the middle of the inferior surface (away from the in-rolled margin) of the thallus; conspicuous equally spaced hairlines that alternated between both frond surfaces; and broad, indusiate oogonial and tetrasporangial sori that occupied nearly the entire fertile zones, and the fertile zones were separated by sterile zones of equal width. Padina calcarea was a two-layered species characterised by a bright yellow inferior surface and a thick calcification on the superior surface (facing to the in-rolled margin), which imparted a strikingly whitish color; inconspicuous hairlines were confined to the inferior surface; and indusiate tetrasporangial sori were just above the hairlines and were found only on the inferior surface. Molecular phylogenetic analyses used chloroplast rbcL and mitochondrial cox3 gene sequences and revealed that the two new species each form strongly supported clades that were genetically distant. Padina calcarea formed an isolated clade that made an early divergence; whereas, P. sulcata showed a sister relationship to P. ryukyuana, indicating a more recent divergence. Padina calcarea was very similar to the Hawaiian P. melemele in gross appearance, particularly in the bright orange to yellow color of the inferior thallus surface and the heavy calcification on the superior surface. However, they were distinguished mainly by the position and arrangement of reproductive sori that were found on the inferior surface and located just above the hairlines in P. calcarea but that were found on the superior surface between the hairlines of the opposite surface in P. melemele. A taxonomic study of the genus Padina from Japan, Southeast Asia, and Hawaii based on morphology and gene sequence data ( rbcL and cox3) resulted in the recognition of four new species, that is, Padina macrophylla and Padina ishigakiensis from Ryukyu Islands, Japan; Padina maroensis from Hawaii; and Padina usoehtunii from Myanmar and Thailand. All species are bistratose and morphologically different from one another as well as from any known taxa by a combination of characters relating to degree of calcification; the structure, position, and arrangement of hairlines (HLs) and reproductive sori; and the presence or absence of rhizoid-like groups of hairs and an indusium. Molecular phylogenetic analyses demonstrated a close relationship between P. ishigakiensis, P. macrophylla, P. maroensis, and Padina australis Hauck. The position of P. usoehtunii, however, was not fully resolved, being either sister to a clade comprising the other three new species and P. australis in the rbcL tree or more closely related to a clade comprising several other recently described species in the cox3 tree. The finding of the four new species demonstrates high species diversity particularly in southern Japan. The following characters were first recognized here to be useful for species delimitation: the presence or absence of small rhizoid-like groups of hairs on the thallus surface, structure and arrangement of HLs on both surfaces either alternate or irregular, and arrangement of the alternating HLs between both surfaces in equal or unequal distance. Comparative rbcL sequence analysis indicates that the species going under the name Schizymenia dubyi in the Azores should be referred to as S. apoda. Sequences of Schizymenia specimens from China and Namibia were also identified as S. apoda, of which the type locality is the Cape Province in South Africa. Schizymenia dubyi, described from Atlantic France, is clearly a distinct species that we here report for Japan and Sicily in the Mediterranean Sea. Both Schizymenia species, along with an unreported species from Japan, are distinct from S. pacifica described from Washington, in the Pacific Coast of North America. Secondary pit connections were observed in gametophytes of S. apoda from the Azores, a previously unknown character for the Nemastomatales. Examination of type material of Nemastoma confusum indicates that this species, currently placed in the Nemastomataceae, should be transferred to the genus Platoma in the Schizymeniaceae. A morphological comparison between Platoma confusum (Kraft & John) comb. nov. with descriptions of P. cyclocolpum and P. chrysymenioides suggests that the three species are closely related. Phylogenetic relationships in the Sargassaceae were explored using three DNA markers, and the monophyly of its genera was challenged. Nineteen out of 24 currently recognized genera were sampled, representing 63 species. The variable mt23S-tRNA Val intergenic spacer could only be aligned within genera and could not be used to infer intergeneric relationships. The partial mt23S was also useful to delineate genera and was alignable at the family level but provided few informative characters. Analysis of mt23S DNA sequences together with chloroplast-encoded psbA sequences resulted in a better resolved phylogeny. Hormophysa was the first genus to branch off within the Sargassaceae, followed by Myriodesma; then the three genera Caulocystis, Carpoglossum, and Scaberia in unresolved order; and then Acrocarpia. The other taxa studied here were divided over three major clades, but there was no branch support for the monophyly of two of these. The genera Bifurcaria, Cystoseira, Halidrys, and Sargassum appeared polyphyletic. The following taxonomic changes are proposed: a new genus Brassicophycus for Bifurcaria brassicaeformis (Kützing) E. S. Barton; reinstatement of the genus Sargassopsis for Sargassum decurrens (R. Brown ex Turner) C. Agardh; reinstatement of the genus Sirophysalis for Indo-Pacific Cystoseira trinodis (Forsskål) C. Agardh; reinstatement of the genus Polycladia for the western Indian Ocean species Cystoseira indica (Thivy et Doshi) Mairh, Cystoseira myrica (S. G. Gmelin) C. Agardh, and Acystis heinii Schiffner; and reinstatement of the genus Stephanocystis for the North Pacific Cystoseira species and Halidrys dioica N. L. Gardner. The European Cystoseira species should be split into three genera, but no name changes are proposed yet, because diagnostic characters were found only for the clade including the type species. Molecular phylogenetic analyses of Padina species collected from the western Pacific Ocean using rbcL and mitochondrial cox3 genes revealed the occurrence of four genetically distinctive clades of unknown Padina species: clade A (=Padina okinawaensis sp. nov.) from Ryukyu Islands (Japan), Hawaii, Indonesia and Thailand, clade B (=Padina undulata sp. nov.), clade C (=Padina terricolor sp. nov.) and clade D (=Padina fasciata sp. nov.) from Ryukyu Islands (Japan). Morphologically, these new taxa are all bistratose species, and different from any known species in the following morphological features: P. okinawaensis sp. nov., reniform or circular thallus of entire margin with inconspicuous hair lines on the superior surface of the thallus, and reproductive organs (oogonia and tetrasporangia) in small groups or discontinuous sori under thin flakes of calcium on the inferior surface; P. undulata sp. nov., circular or semicircular thallus with undulate margin, conspicuous hair lines on both surfaces of the thallus, oogonial and tetrasporangial sori in continuous or discontinuous lines covered with a persistent indusium, and cylindrical oogonia; P. terricolor sp. nov., grayish brown on the inferior surface of the thallus, and oogonial and tetrasporangial sori in continuous lines covered with a persistent indusium; P. fasciata sp. nov., broad white stripes on both surfaces of the thallus due to unique calcification, and oogonial and tetrasporangial sori in continuous lines, which are entirely embedded in a gelatinous layer, on the inferior surface of the thallus. Macaranga and Mallotus (Euphorbiaceae s.s.) are two closely related, large paleo(sub)tropical genera. To investigate the phylogenetic relationships between and within them and to determine the position of related genera belonging to the subtribe Rottlerinae, we sequenced one plastid (trnL-F) and three nuclear (ITS, ncpGS, phyC) markers for species representative of these genera. The analyses demonstrated the monophyly of Macaranga and the paraphyly of Mallotus and revealed three highly supported main clades. The genera Cordemoya and Deuteromallotus and the Mallotus sections Hancea and Oliganthae form a basal Cordemoya s.l. clade. The two other clades, the Macaranga clade and the Mallotus s.s. clade (the latter with Coccoceras, Neotrewia, Octospermum, and Trewia), are sister groups. In the Macaranga clade, two basal lineages (comprising mostly sect. Pseudorottlera) and a crown group with three geographically homogenous main clades were identified. The phylogeny of the Mallotus s.s. clade is less clear because of internal conflict in all four data sets. Many of the sections and informal infrageneric groups of Macaranga and Mallotus do not appear to be monophyletic. In both the Macaranga and Mallotus s.s. clades, the African and/or Madagascan taxa are nested in Asian clades, suggesting migrations or dispersals from Asia to Africa and Madagascar. A molecular phylogenetic analysis of the little-studied filamentous brown alga Discosporangium mesarthrocarpum (Meneghini) Hauck using rbcL and partial 18S rDNA sequences revealed that the species forms a monophyletic clade with Choristocarpus tenellus (Kütz.) Zanardini that is sister to all other brown algae. Although D. mesarthrocarpum has unique disk-shaped plurilocular reproductive organs, D. mesarthrocarpum and C. tenellus share the following basic morphological features, which are considered to be plesiomorphic characters in the brown algae: (1) apical (and diffuse) growth; (2) uniseriate, subdichotomously branched filaments; (3) multiple chloroplasts per cell without pyrenoids; and (4) lack of heterotrichy and of phaeophycean hairs. The rbcL DNA sequence of an Australian D. mesarthrocarpum specimen showed considerable deviation from Mediterranean and Macaronesian specimens. Therefore, the presence of a second species in the genus is suggested; however, the taxonomic treatment of this putative species is not pursued in the present report. Regarding the higher-ranking systematic position of D. mesarthrocarpum, reinstatement of Discosporangiaceae and Discosporangiales is proposed, and the inclusion of Choristocarpaceae in the order is also suggested. Under short-day and long-day culture conditions at 15°C–25°C, Mediterranean D. mesarthrocarpum exhibited a direct type of life history, with a succession of uniseriate filamentous thalli bearing characteristic disk-shaped plurilocular zoidangia, but thalli did not survive at 10°C and below. To better assess the current state of phaeophycean phylogeny, we compiled all currently available rbcL, 18S, and 26S rDNA sequences from the EMBL/GenBank database and added 21 new rbcL sequences of our own. We then developed three new alignments designed to maximize taxon sampling while minimizing information loss due to partial sequences. Phylogenetic analyses were performed on separate and combined data sets (with and without taxa from the sister classes Tribophyceae and Phaeothamniophyceae as outgroups) using a variety of assumption sets, tree-drawing algorithms (parsimony, neighbor joining, and likelihood), and resampling methods (bootstrap, decay, jackknife). Partition homogeneity testing (PHT) by codon position within rbcL showed that all positions could be used despite mild third position saturation. PHT by gene and domain within rDNA showed that the 26S D1 and D2 regions do not enhance phylogenetic signal even when combined with the 18S. The rbcL and rDNA (excluding the 26S D1 and D2) could be combined under PHT. The topology of the combined tree was the same as that of the rbcL tree alone, but bootstrap support was consistently higher in the combined analysis, applied to more branches, and enabled the establishment of sister group relationships among six orders. Although the taxon sampling for the combination tree was lower (n = 22) than for individual gene analyses (n = 58 for rbcL and n = 59 for rDNA), results show that the Laminariales (previously reported) and Sphacelariales (new) are both paraphyletic. Choristocarpus tenellus (Kützing) Zanardini is the most basal phaeophyte and the Dictyotales the most basal order. In contrast, the Laminariales sensu stricto (s.s.) and Ectocarpales sensu lato (s.l.) are the most derived. For phylogenetic studies in the Phaeophyceae, rbcL has more resolving power than rDNA, though the reason for this is unclear based on the fact that both genes are highly conserved. The article discusses a proposal to preserve the name Sphacelaria only for the Sphacelaria Propagulifera species. It states that when initially published, the genus Sphacelaria Lyngb. included only the species of Sphacelaria reticulata Lyngb. and thus future study of this species could lead to taxonomic instability. It notes that it is desirable to connect the name Sphacelaria to the popular members of the Sphacelaria Propagulifera species in order to avoid taxonomic instability in the future.
