Background The European Renal Association (ERA) Registry collects data on kidney replacement therapy (KRT) in patients with end-stage kidney disease (ESKD). This paper is a summary of the ERA Registry Annual Report 2021, including a comparison across treatment modalities. Methods Data was collected from 54 national and regional registries from 36 countries, of which 35 registries from 18 countries contributed individual patient data and 19 registries from 19 countries contributed aggregated data. Using this data, incidence and prevalence of KRT, kidney transplantation rates, survival probabilities and expected remaining lifetimes were calculated. Result In 2021, 533.2 million people in the general population were covered by the ERA Registry. The incidence of KRT was 145 per million population (pmp). In incident patients, 55% were 65 years or older, 64% were male, and the most common primary renal disease (PRD) was diabetes (22%). The prevalence of KRT was 1040 pmp. In prevalent patients, 47% were 65 years or older, 62% were male, and the most common PRDs were diabetes and glomerulonephritis/sclerosis (both 16%). On 31 December 2021, 56% of patients received haemodialysis, 5% received peritoneal dialysis, and 39% were living with a functioning graft. The kidney transplantation rate in 2021 was 37 pmp, a majority coming from deceased donors (66%). For patients initiating KRT between 2012–2016, 5-year survival probability was 52%. Compared to the general population, life expectancy was 65% and 68% shorter for males and females receiving dialysis, and 40% and 43% shorter for males and females living with a functioning graft. 3. Hepatocyte Nuclear Factor-1α Increases Fibrinogen Gene Expression in Liver and Plasma Fibrinogen Concentration in Rats with Experimental Chronic Renal Failure.

Chronic kidney disease (CKD) is associated with elevated plasma fibrinogen concentration. However, the underlying molecular mechanism for elevated plasma fibrinogen concentration in CKD patients has not yet been clarified. We recently found that HNF1α was significantly upregulated in the liver of chronic renal failure (CRF) rats, an experimental model of CKD in patients. Given that the promoter region of the fibrinogen gene possesses potential binding sites for HNF1α, we hypothesized that the upregulation of HNF1α can increase fibrinogen gene expression and consequently plasma fibrinogen concentration in the experimental model of CKD. Here, we found the coordinated upregulation of Aα-chain fibrinogen and Hnfα gene expression in the liver and elevated plasma fibrinogen concentrations in CRF rats, compared with pair-fed and control animals. Liver Aα-chain fibrinogen and HNF1α mRNAs levels correlated positively with (a) liver and plasma fibrinogen levels and (b) liver HNF1α protein levels. The positive correlation between (a) liver Aα-chain fibrinogen mRNA level, (b) liver Aα-chain fibrinogen level, and (c) serum markers of renal function suggest that fibrinogen gene transcription is closely related to the progression of kidney disease. Knockdown of Hnfα in the HepG2 cell line by small interfering RNA (siRNA) led to a decrease in fibrinogen mRNA levels. Clofibrate, an anti-lipidemic drug that reduces plasma fibrinogen concentration in humans, decreased both HNF1α and Aα-chain fibrinogen mRNAs levels in (a) the liver of CRF rats and (b) HepG2 cells. The obtained results suggest that (a) an elevated level of liver HNF1α can play an important role in the upregulation of fibrinogen gene expression in the liver of CRF rats, leading to an elevated concentration of plasma fibrinogen, a protein related to the risk of cardiovascular disease in CKD patients, and (b) fibrates can decrease plasma fibrinogen concentration through inhibition of HNF1α gene expression. Biochar (BC) is a material that has many applications in agricultural and environmental activities. The aim of the study was to define the influence of BC produced in low-temperature pyrolysis from various organic waste materials, including one-month-old compost (OMOC), pine bark (PB), pine needle mulch (NM), pine cones (PC) and maple leaves (ML), on soil enzyme activity as well as its relation with organic matter properties. A 60-day incubation pot experiment was set up to investigate the influence of BC amendment on soil (S) characteristics. After incubation, we investigated the activity of soil enzymes, the content of available phosphorus (AP), potassium (AK) and magnesium (AMg), total organic carbon (TOC), total nitrogen (TN), dissolved organic matter (DOM) and its fractional composition (content and share of carbon and nitrogen of humic (CHAs, NHAs) and fulvic (CFAs, NFAs) acids and humin fractions). The effect of the amended biochars differed depending on the feedstock material. In general, the use of biochar enriched the soil with AP, AK and AMg increased the soil carbon stock, increased the intensity of nitrogen transformation and influenced the soil enzyme activity. OMOC and ML biochars significantly increased soil fertility, which was expressed by the high value of the CHA/CFA ratio. [ABSTRACT FROM AUTHOR]

One of the main tasks in the search for environmentally friendly crop-growing methods is to increase soil fertility by improving its physical, chemical and biological parameters. The aim of this study was to determine the effect that the long-term annual application of different types of soil fertility agents (exogenous organic matter: 1. manure, 2. straw in combination with nitrogen fertilization and liming and 3. the addition of biostimulants) had on organic matter properties, including humic acid (HAs) properties. The research was carried out on the basis of soil samples from a ten-year pot experiment which was set up as single-factor pot experiment with four replications. PVC pots with perforated bottoms were filled with soil samples taken from the tilled layer of an arable field where winter wheat was grown in monoculture. The pots were exposed directly to the weather and were left without vegetation. The soil samples were assayed for the content of total organic carbon (TOC), total nitrogen and fractional composition of humus. HAs were extracted with the Schnitzer method and analyzed for the elemental composition, spectrometric parameters in the FT-IR and UV-VIS range and hydrophilic and hydrophobic properties. In addition, EPR spectra were produced. The results showed that the content of organic matter compared to soil without additives increased with the use of manure and the use of straw in the CaO variant and in the form of a mulch. The content of dissolved organic carbon (DOC) ranged from 124.6 to 286.1 mg kg
−1 and had strong positive correlation with TOC content. The values of the ratio of carbon content in humic acids to carbon content in fulvic acids (CHAs/CFAs) ranged from 0.71 to 0.99. The use of a biostimulator—with or without the addition of straw—increased carbon sequestration in humic acid molecules, as well as their oxidation level and their share of hydrophobic fractions with the longest retention time. 6. Hepatocyte Nuclear Factor 1α Proinflammatory Effect Linked to the Overexpression of Liver Nuclear Factor–κB in Experimental Model of Chronic Kidney Disease.

Chronic kidney disease (CKD) is associated with low-grade inflammation that activates nuclear factor–κB (NF–κB), which upregulates the expression of numerous NF–κB responsive genes, including the genes encoding IL-6, ICAM-1, VCAM-1, and MCP-1. Herein, we found the coordinated overexpression of genes encoding RelA/p65 (a subunit of NF–κB) and HNF1α in the livers of chronic renal failure (CRF) rats—an experimental model of CKD. The coordinated overexpression of RelA/p65 and HNF1α was associated with a significant increase in IL-6, ICAM-1, VCAM-1, and MCP-1 gene expressions. A positive correlation between liver RelA/p65 mRNA levels and a serum concentration of creatinine and BUN suggest that RelA/p65 gene transcription is tightly related to the progression of renal failure. The knockdown of HNF1α in the HepG2 cell line by siRNA led to a decrease in Rel A/p65 mRNA levels. This was associated with a decrease in IL-6, ICAM-1, VCAM-1, and MCP-1 gene expressions. The simultaneous repression of HNF-1α and RelA/p65 by clofibrate is tightly associated with the downregulation of IL-6, ICAM-1, VCAM-1, and MCP-1 gene expression. In conclusion, our findings suggest that NF–κB could be a downstream component of the HNF1α-initiated signaling pathway in the livers of CRF rats. [ABSTRACT FROM AUTHOR]

7. Management Options for Fetal Bronchopulmonary Sequestration.

To evaluate the prenatal course and perinatal outcome of fetuses with bronchopulmonary sequestration (BPS) managed expectantly or using minimally invasive methods. This was a retrospective study of 29 fetuses with suspected BPS managed between 2010 and 2021 in three fetal medicine centers in Poland. Medline was searched to identify cases of BPS managed expectantly or through minimally-invasive methods. In 16 fetuses with BPS, there was no evidence of cardiac compromise. These fetuses were managed expectantly. Thirteen hydropic fetuses with BPS qualified for intrauterine intervention: a thoraco-amniotic shunt (TAS) was inserted in five fetuses, laser coagulation of the feeding vessel was performed in seven cases, and one fetus had combined treatment. In the combined data from the previous and the current study of various percutaneous interventions for BPS associated with hydrops, the survival rate was 91.2% (31/34) for TAS, 98.1% (53/54) for laser coagulation, and 75% (3/4) for intratumor injection of sclerosant. After taking into account cases with available data, the rate of preterm birth before 37 weeks in the group treated with laser coagulation was 14.3% (7/49) compared to 84.6% (22/26) in the group treated with TAS. The need for postnatal sequestrectomy was lower in the group of fetuses treated with laser coagulation 23.5% (12/51) in comparison to fetuses treated with TAS 84% (21/26). In fetuses with BPS without hydrops, progression of the lesion's volume, leading to cardiac compromise, is unlikely. In hydropic fetuses with BPS, intrauterine therapy using minimally invasive methods prevents fetal demise. Both, the rate of preterm birth and the need for postnatal surgery is significantly lower in the group treated with laser coagulation compared to the group treated with TAS. [ABSTRACT FROM AUTHOR]

9. Properties of humic acids depending on the land use in different parts of Slovakia.

Many studies report organic carbon stabilization by clay minerals, but the effects of land use and soil type on the properties of humic acids (HAs) are missing. The aim of the paper is to determine the effects of land use and soil types on the characteristics of HAs, which have a considerable influence on organic matter quality. It was hypothesised that the effect of the land use on HAs properties depends on the particular size distribution. The research was performed in three ecosystems: agricultural, forest, and meadow, located in Slovakia. From each of them, the samples of 4 soil types were taken: Chernozem, Luvisol, Planosol, and Cambisol. The soil samples were assayed for the content of total organic carbon (TOC) and the particle size distribution. HAs were extracted with the Schnitzer method and analysed for the elemental composition, spectrometric parameters in the UV-VIS range, and hydrophilic and hydrophobic properties, and the infrared spectra were produced. The research results have shown that the properties of HAs can be modified by the land use and the scope and that the direction of changes depends on the soil type. The HAs of Chernozem and Luvisol in the agri-ecosystem were identified with a higher "degree of maturity", as reflected by atomic ratios (H/C, O/C, O/H), absorbance coefficients, and the FT-IR spectra, as compared with the HAs of the meadow and forest ecosystem. However, as for the HAs of Cambisol, a higher "degree of maturity" was demonstrated for the meadow ecosystem, as compared with the HAs of the agri- and forest ecosystem. The present research has clearly identified that the content of clay is the factor determining the HAs properties. Introduction: We sought to identify patients with amyotrophic lateral sclerosis (ALS) who displayed suspected peripheral nervous system (PNS) inflammation to compare them to those with suspected PNS degeneration.Methods: We measured sonographic median and ulnar nerve cross-sectional area (CSA) and cerebrospinal fluid albumin/serum albumin ratio (Qalb ) in patients with ALS to classify them as having suspected PNS degeneration (small CSA/low Qalb ) or inflammation (larger CSA/high Qalb ).Results: Fifty-seven percent of patients had suspected PNS degeneration, 21% had suspected PNS inflammation, and 21% displayed suspected "normal PNS state." Suspected PNS degeneration was related to classic ALS, shorter disease duration, and a smaller hypoechoic nerve area. Suspected PNS inflammation was associated with men, longer disease duration, and a larger hypoechoic nerve area and was the dominant finding in superoxide dismutase 1 mutation carriers.Discussion: Our simple approach might aid in the in vivo differentiation of supposed ALS subtypes, those with suspected PNS degeneration vs. inflammation, for stratification in clinical trials. The article presents the approach to the design process of acoustic panels based on the scientific research. This approach is based on combining the technical and the design competences to develop the innovative product value for the concept of acoustic panels. The article presents the concepts of two new acoustic panels -- an absorbing and scattering panel and a panel reflecting sound waves. The first part of the article presents the starting point for the presented project -- the acoustic research and the inspiration for both types of presented solutions. Next, the materials possible to use were discussed, which could reproduce the natural acoustic properties of the lava and glacier caves. The next part presents consecutive stages of the product development in a modern form, ensuring the expected acoustic properties. 12. Prestress state evolution during thermal activation of memory effect in concrete beams strengthened with external SMA wires.

- Abstract
The memory effect of shape-memory alloys (SMAs) has opened interesting perspectives to create prestress states in concrete elements. However, the procedure has not been yet fully resolved due to the complex thermomechanical behavior of these alloys, in addition to the practical difficulties of mechanical coupling between SMA and concrete elements. The present study deals with tests on the development of prestressing forces in concrete beams during the thermal cycle required in the procedure. Pre-stretched nickel–titanium wires were externally placed on concrete prismatic beams equipped with strain gauges. As concrete rupture may occur during the heating by the Joule effect, a compromise must be found between the SMA pre-stretch level and the maximum temperature to be applied before returning to ambient temperature. A macroscopic model was developed to analyze this compromise. The complex thermomechanical response of SMAs implies a particular attention in the definition of the ambient temperature and heating conditions for the creation of prestress states in concrete components. These guidelines follow the mission of the World Association of Perinatal Medicine, which brings together groups and individuals throughout the world with the goal of improving outcomes of maternal, fetal and neonatal (perinatal) patients. Guidelines for auditing, evaluation, and clinical care in perinatal medicine enable physicians diagnose, treat and follow-up of COVID-19-exposed pregnant women. These guidelines are based on quality evidence in the peer review literature as well as the experience of perinatal expert throughout the world. Cardiac ischaemia‐reperfusion (I/R) injury has been attributed to stress signals arising from an impaired mitochondrial electron transport chain (ETC), which include redox imbalance, metabolic stalling and excessive production of reactive oxygen species (ROS). The alternative oxidase (AOX) is a respiratory enzyme, absent in mammals, that accepts electrons from a reduced quinone pool to reduce oxygen to water, thereby restoring electron flux when impaired and, in the process, blunting ROS production. Hence, AOX represents a natural rescue mechanism from respiratory stress. This study aimed to determine how respiratory restoration through xenotopically expressed AOX affects the re‐perfused post‐ischaemic mouse heart. As expected, AOX supports ETC function and attenuates the ROS load in post‐anoxic heart mitochondria. However, post‐ischaemic cardiac remodelling over 3 and 9 weeks was not improved. AOX blunted transcript levels of factors known to be up‐regulated upon I/R such as the atrial natriuretic peptide (Anp) whilst expression of pro‐fibrotic and pro‐apoptotic transcripts were increased. Ex vivo analysis revealed contractile failure at nine but not 3 weeks after ischaemia whilst label‐free quantitative proteomics identified an increase in proteins promoting adverse extracellular matrix remodelling. Together, this indicates an essential role for ETC‐derived signals during cardiac adaptive remodelling and identified ROS as a possible effector. [ABSTRACT FROM AUTHOR]

Introduction: We aimed to investigate whether sonographic peripheral cross-sectional nerve area (CSA) and progranulin (PGRN), a neuritic growth factor, are related to each other and whether they interact to predict clinical and paraclinical measures in amyotrophic lateral sclerosis (ALS).Methods: We included 55 ALS patients who had forearm median and ulnar nerve CSA, cerebrospinal fluid (CSF) PGRN, and serum PGRN measures available. CSF PGRN was normalized against the CSF / serum albumin ratio (Qalb ). Using age, sex, height, and weight adjusted general linear models, we examined CSA × CSF PGRN interaction effects on various measures.Results: There was a medium-effect size inverse relationship between CSA and CSF PGRN, but not between CSA and serum PGRN. Lower CSA values together with higher CSF PGRN levels were linked to smaller motor amplitudes.Discussion: In ALS, the constellation of peripheral nerve atrophy together with higher CSF PGRN levels indicates pronounced axonal damage. Classic prognostic factors, such as clinical advancement of the disease and histological grade of the tumor, continue to have a decisive role in the selection of therapeutic strategy in patients with carcinoma of the oral cavity floor and oral surface of the tongue (OCC). YAP1/Yes-associated protein 1 (YAP) and transcriptional co-activator with PDZ-binding motif, WWTR1 (TAZ) proteins, appear to be promising markers that may be used to develop personalized therapies. The aim of the present study was to analyze the associations between the levels of YAP, TAZ and tyrosine-protein phosphatase non-receptor type 14 (PTPN14) and to determine whether the increased expression of YAP and TAZ had an effect on tumor cell proliferation, as determined by minichromosome maintenance 7, DNA replication licensing factor 7 expression. Their prognostic value was also assessed. In total, 127 patients who underwent radical surgery and were subjected to adjuvant radiation therapy due to squamous cell OCC were enrolled in the present study. The results demonstrated an evident effect as YAP expression increased in cancer-associated fibroblasts, which induced unfavorable prognosis in patients. In addition, a positive association between proliferation in cancer cells and YAP expression in stromal cells was observed. A lack of YAP expression in the cytoplasm of tumor cells was a factor for poor prognosis with regard to disease-free survival and disease specific survival. No statistically significant correlations between YAP and TAZ expression and PTPN14 expression were identified, nor was a correlation between cell proliferation and the presence of YAP and TAZ in tumor cells observed. Cerebrospinal fluid (CSF) neurofilament light chain (NfL) has emerged as putative diagnostic biomarker in amyotrophic lateral sclerosis (ALS), but it remains a matter of debate, whether CSF total tau (ttau), tau phosphorylated at threonine 181 (ptau) and the ptau/ttau ratio could serve as diagnostic biomarker in ALS as well. Moreover, the relationship between CSF NfL and tau measures to further axonal and (neuro)degeneration markers still needs to be elucidated. Our analysis included 89 ALS patients [median (range) age 63 (33-83) years, 61% male, disease duration 10 (0.2-190) months] and 33 age- and sex-matched disease controls [60 (32-76), 49%]. NfL was higher and the ptau/ttau ratio was lower in ALS compared to controls [8343 (1795-35,945) pg/ml vs. 1193 (612-2616), H(1) = 70.8, p < 0.001; mean (SD) 0.17 (0.04) vs. 0.2 (0.03), F(1) = 14.3, p < 0.001], as well as in upper motor neuron dominant (UMND, n = 10) compared to classic (n = 46) or lower motor neuron dominant ALS [n = 31; for NfL: 16,076 (7447-35,945) vs. 8205 (2651-35,138) vs. 8057 (1795-34,951)], Z ≥ 2.5, p ≤ 0.01; for the ptau/ttau ratio: [0.13 (0.04) vs. 0.17 (0.04) vs. 0.18 (0.03), p ≤ 0.02]. In ALS, NfL and the ptau/ttau ratio were related to corticospinal tract (CST) fractional anisotropy (FA) and radial diffusivity (ROI-based approach and whole-brain voxelwise analysis). Factor analysis of mixed data revealed a co-variance pattern between NfL (factor load − 0.6), the ptau/ttau ratio (0.7), CST FA (0.8) and UMND ALS phenotype (− 2.8). NfL did not relate to any further neuroaxonal injury marker (brain volumes, precentral gyrus thickness, peripheral motor amplitudes, sonographic cross-sectional nerve area), but a lower ptau/ttau ratio was associated with whole-brain gray matter atrophy and widespread white matter integrity loss. Higher NfL baseline levels were associated with greater UMN disease burden, more rapid disease progression, a twofold to threefold greater hazard of death and shorter survival times. The findings that higher CSF NfL levels and a reduced ptau/ttau ratio are more associated with clinical UMN involvement and with reduced CST FA offer strong converging evidence that both are markers of central motor degeneration. Furthermore, NfL is a marker of poor prognosis, while a low ptau/ttau ratio indicates extramotor pathology in ALS. [ABSTRACT FROM AUTHOR]

Inflammation related to chronic kidney disease (CKD) is an important clinical problem. We recently determined that hepatocyte nuclear factor 1α (HNF1α) was upregulated in the livers of chronic renal failure (CRF) rats—experimental model of CKD. Considering that the promoter region of gene encoding C-reactive protein (CRP) contains binding sites for HNF1α and that the loss-of-function mutation in the Hnfs1α leads to significant reduction in circulating CRP levels, we hypothesized that HNF1α can activate the Crp in CRF rats. Here, we found coordinated upregulation of genes encoding CRP, interleukin-6 (IL-6), HNF1α, and HNF4α in the livers and white adipose tissue (WAT) of CRF rats, as compared to the pair-fed and control animals. This was accompanied by elevated serum levels of CRP and IL-6. CRP and HNFs’ mRNA levels correlated positively with CRP and HNFs’ protein levels in the liver and WAT. Similar upregulation of the Crp, Il-6, and Hnfs in the liver and WAT and increased serum CRP and IL-6 concentrations were found in lipopolysaccharide (LPS)-induced systemic inflammation in rats. Moreover, silencing HNF1α in HepG2 cells by small interfering RNA led to decrease in CRP mRNA levels. Our results suggests that (a) HNFs act in concert with IL-6 in the upregulation of CRP production by the liver and WAT, leading to an increase in circulating CRP concentration in CRF rats and (b) CRF-related inflammation plays an important role in the upregulation of genes that encode HNFs and CRP in the liver and WAT of CRF rats. Purpose: The aim of this paper has been to determine the seasonal changes in the content of dissolved organic matter (DOM) in the soils under agricultural use based on assaying changes in dissolved organic carbon (DOC) and dissolved nitrogen (DNt) as well as determining the factors which can define the DOM in soils.Materials and methods: The research has involved the soils under agricultural use sampled in the Kujawsko-Pomorskie province (Poland). Phaeozems and Luvisols were sampled from the depth of 0-30, 30-60, and 60-100 cm, November 2011 through September 2013, in November, March, May, July, and September. The soil samples were assayed for the grain size composition, pH, dry weight content, content of total organic carbon, and total nitrogen. Dissolved organic matter was extracted with 0.004 mol dm
3 CaCl2 ; in the DOM extracts, the content of dissolved organic carbon (DOC) and dissolved nitrogen (DNt) were assayed. The research results were statistically verified.Results and discussion: It has been demonstrated that in the first year of research, the content of dissolved organic carbon in the soils was changing throughout the year. The highest differences in the content of that carbon fraction occurred across the soil sampled in autumn and the soil sampled in spring. In the second year of research, an inverse dependence was noted. DOC was migrating to deeper layers of the soil profile; yet, the migration got more intensive in summer. The content of dissolved nitrogen was not changing significantly throughout the year. Higher DNt content in the surface layer, in general, resulted in a higher content of dissolved nitrogen in deeper profile layer, which could have been due to leaching of the nutrient deep down the soil profile.Conclusions: The content of dissolved organic carbon was significantly related to the content of total organic carbon and total nitrogen. Significant changes in the content of dissolved forms of nitrogen were reported in the profile of Phaeozems due to mineral fertilization and irrigation. The soils where irrigation and higher nitrogen rates had been applied demonstrated a higher content and share of soluble forms of nitrogen, as compared with the soils non-irrigated and the soils where lower nitrogen rates had been supplied. The study deals with the influence of thermal boundary effects on the process of creating recovery stresses in a SMA wire activated by Joule heating, during a thermal cycle (up to the return to ambient temperature). First, a thermal characterization is performed using infrared thermography for temperature profile measurements along the wire in a steady-state regime. Second, recovery stress tests are performed using a uniaxial testing machine. Finally, tests are analyzed using a thermomechanical model, taking the inhomogeneous temperature distribution along the wire into account. The influence of the initial distribution of martensite (before thermal activation of the memory effect) is discussed, as well as the influence of the wire length. It is shown that the thermal boundary effects at the contact with the grips of the testing machine significantly influence the response of the wire. For instance, during the heating of the wire, an austenite-to-martensite transformation may occur in the zones near the wire ends (where the temperature remains close to ambient) due to the increased stress. A length of influence of the thermal boundary effects on the overall wire response is defined, and a condition to neglect this influence is proposed. 22. Impact of gender and dialysis adequacy on anaemia in peritoneal dialysis.

- Abstract
Purpose: In the general population, haemoglobin (Hb) concentration is higher in men than in women. However, target Hb levels in dialysis patients are set constant regardless of the patient's sex. The aim of this study was to evaluate Hb concentration and the use of erythropoiesis-stimulating agents (ESA) in peritoneal dialysis (PD) patients taking gender and dialysis adequacy into account. Methods: The study comprised two parts. The first was a cross-sectional analysis of Hb and ESA in 2180 prevalent PD patients. The second included 88 incident PD patients, followed for 36 months. During this time, the major parameters recorded at 12-month intervals included: Hb concentration, weekly ESA, total, renal, and peritoneal Kt/V. Erythropoietin resistance index (ERI) was calculated as the ratio between ESA dose and achieved Hb. Results: In prevalent PD patients, Hb concentration was significantly lower in women, (11.2 ± 1.4 vs. 11.5 ± 1.6 g/dl; p < 0.001), despite higher doses of ESA (2691 ± 1821 vs. 2344 ± 1422; p = 0.001). Hb concentrations were related to dialysis adequacy in both cohorts. However, despite significantly higher Kt/V, women were characterized by a lower Hb level. In incident patients, this association was present throughout the observation period, while the ESA dose in women was significantly higher at every time point. In multiple regression analysis, gender was an independent determinant of ERI ( b = 0.34; p < 0.05). Conclusions: Despite higher dialysis adequacy, Hb concentration in women treated with PD is significantly lower, and the ability to correct it impaired, as compared to men. [ABSTRACT FROM AUTHOR]

Background. This is the first report on the epidemiology of biopsy-proven kidney diseases in Poland. Methods. The Polish Registry of Renal Biopsies has collected information on all (n = 9394) native renal biopsies performed in Poland from 2009 to 2014. Patients' clinical data collected at the time of biopsy, and histopathological diagnoses were used for epidemiological and clinicopathologic analysis. Results. There was a gradual increase in the number of native renal biopsies performed per million people (PMP) per year in Poland in 2009-14, starting from 36 PMP in 2009 to 44 PMP in 2014. A considerable variability between provinces in the mean number of biopsies performed in the period covered was found, ranging from 5 to 77 PMP/year. The most common renal biopsy diagnoses in adults were immunoglobulin A nephropathy (IgAN) (20%), focal segmental glomerulosclerosis (FSGS) (15%) and membranous glomerulonephritis (MGN) (11%), whereas in children, minimal change disease (22%), IgAN (20%) and FSGS (10%) were dominant. Due to insufficient data on the paediatric population, the clinicopathologic analysis was limited to patients ⩾18 years of age. At the time of renal biopsy, the majority of adult patients presented nephroticrange proteinuria (45.2%), followed by urinary abnormalities (38.3%), nephritic syndrome (13.8%) and isolated haematuria (1.7%). Among nephrotic patients, primary glomerulopathies dominated (67.6%in those 18-64 years of age and 62.4% in elderly patients) with leading diagnoses being MGN (17.1%), FSGS (16.2%) and IgAN (13.0%) in the younger cohort and MGN (23.5%), amyloidosis (18.8%) and FSGS (16.8%) in the elderly cohort. Among nephritic patients 18-64 years of age, the majority (55.9%) suffered from primary glomerulopathies, with a predominance of IgAN (31.3%), FSGS (12.7%) and crescentic GN (CGN) (11.1%). Among elderly nephritic patients, primary and secondary glomerulopathies were equally common (41.9% each) and pauci-immune GN (24.7%), CGN (20.4%) and IgAN (14.0%) were predominant. In both adult cohorts, urinary abnormalities were mostly related to primary glomerulopathies (66.8% in younger and 50% in elderly patients) and the leading diagnoses were IgAN (31.4%), FSGS (15.9%), lupus nephritis (10.7%) and FSGS (19.2%), MGN (15.1%) and pauci-immune GN (12.3%), respectively. There were significant differences in clinical characteristics and renal biopsy findings between male and female adult patients. Conclusions. Endothelial dysfunction could be related to the limited availability of nitric oxide (NO). NO is synthesized with the participation of an NO synthase whose activity is inhibited by asymmetric dimethylarginine (ADMA). The synthesis of ADMA is exacerbated by oxidative stress, and several studies have shown the efficacy of drugs acting on the renin-angiotensin-aldosterone system (RAAS) (converting enzyme inhibitors and angiotensin II receptor antagonists) in reducing the level of ADMA. The probable mechanism of drug action is a reduction of oxidative stress through a decrease of angiotensin II formation. The aim of this study was to assess the influence of RAAS blockers on the plasma concentration of ADMA in patients with chronic glomerulonephritis (ChGN). The study included 37 patients, placed into group A and group B, depending on the treatment. Both groups were treated with RAAS blockers. In group B, immunosuppressive drugs were additionally administered. The control visits were at the 0, 6 and 12 months of observation. In both the studied groups (A+B), a significant reduction of ADMA (0.77 vs 0.4 μmol/l; p<0.05) was noticed. In patients suffering from ChGN, the use of RAAS blockers resulted in a significant decrease of plasma ADMA concentration, independently of immunosupressive treatment. [ABSTRACT FROM AUTHOR]

Most of our perceptions of and engagements with the world are shaped by our immersion in social interactions, cultural traditions, tools and linguistic categories. In this study we experimentally investigate the impact of two types of language-based coordination on the recognition and description of complex sensory stimuli: that of red wine. Participants were asked to taste, remember and successively recognize samples of wines within a larger set in a two-by-two experimental design: (1) either individually or in pairs, and (2) with or without the support of a sommelier card--a cultural linguistic tool designed for wine description. Both effectiveness of recognition and the kinds of errors in the four conditions were analyzed. While our experimental manipulations did not impact recognition accuracy, bias-variance decomposition of error revealed non-trivial differences in how participants solved the task. Pairs generally displayed reduced bias and increased variance compared to individuals, however the variance dropped significantly when they used the sommelier card. The effect of sommelier card reducing the variance was observed only in pairs, individuals did not seem to benefit from the cultural linguistic tool. Analysis of descriptions generated with the aid of sommelier cards shows that pairs were more coherent and discriminative than individuals. The findings are discussed in terms of global properties and dynamics of collective systems when constrained by different types of cultural practices. [ABSTRACT FROM AUTHOR]

Introduction: In this study we investigated whether peripheral nerve sonography could be used as a biomarker to monitor disease progression in amyotrophic lateral sclerosis (ALS).Methods: In 37 patients, ulnar and median nerve cross-sectional area (CSA) was determined in at least 2 ultrasound sessions; mean follow-up was 14.5 months. Linear mixed-effects models were conducted to analyze time effects on CSA.Results: Ulnar nerve CSA declined significantly at a monthly rate of -0.04 mm(2) (forearm) and -0.05 mm(2) (wrist); the decrease was more pronounced when baseline CSA was greater. To detect a 50% treatment effect on ulnar nerve CSA, 332 patients would need to be entered in a hypothetical randomized, controlled clinical trial. Time had no significant impact on median nerve CSA.Conclusions: Distal ulnar nerve ultrasound may be a useful biomarker to monitor disease progression in ALS, especially as hypothetical treatment effects on CSA seem to be detectable in manageable cohort sizes. Introduction In various countries, standard doses of anti-D IgG used for postpartum immunoprophylaxis of hemolytic disease of fetus and newborn ( HDFN) vary from 100 μg to 300 μg. Results In all artificial mixtures with known concentrations, FCTs and KBT with counting 10 000 RBCs had similar satisfying sensitivity and specificity. KBT with counting 2000 RBCs had to be disqualified because of significant discrepancies between expected and measured values of FMH. The test procedure with anti-D was easier and shorter than the remaining tests, but it can be only used for FMH assessment in RhD-negative mothers with RhD-positive newborns. In one clinical sample, it was impossible to distinguish fetal RBCs from maternal F cells in KBT and FC with anti-HbF but other tests were useful. Conclusion In the four tests, correlation between expected and obtained results was appropriate ( CCC ̴1). Each test had some advantage and limitation in any clinical situation. ABSTRACT Introduction: In this study we sought to determine the cross-sectional area (CSA) of peripheral nerves in patients with distinct subtypes of amyotrophic lateral sclerosis (ALS). Methods: Ulnar and median nerve ultrasound was performed in 78 ALS patients [classic, n = 21; upper motor neuron dominant (UMND), n = 14; lower motor neuron dominant (LMND), n = 20; bulbar, n = 15; primary lateral sclerosis (PLS), n = 8] and 18 matched healthy controls. Results: Compared with controls, ALS patients had significant, distally pronounced reductions of ulnar CSA (forearm/wrist level) across all disease groups, except for PLS. Median nerve CSA (forearm/wrist level) did not differ between controls and ALS. Conclusion: Ulnar nerve ultrasound in ALS subgroups revealed significant differences in distal CSA values, which suggests it has value as a marker of LMN involvement. Its potential was particularly evident in the UMND and PLS groups, which can be hard to separate clinically, yet their accurate separation has major prognostic implications. The aim of the paper was to determine the stability and the decomposition intensity of selected polycyclic aromatic hydrocarbons (fluorene, anthracene, pyrene, and chrysene) in soils that are under agricultural use. Soil was sampled from the arable layer that is representative of the Kujawy and Pomorze Provinces, which are located in the northwestern part of Poland. The soil samples were polluted with selected PAHs at an amount corresponding to 10 mg PAHs/kg. PAH-polluted soil samples were incubated for 10, 30, 60, 120, 180, and 360 days at a temperature of 20-25 °C and a fixed moisture of 50 % field water capacity. High-performance liquid chromatography (HPLC) was used to determine the content of PAHs. It was found that the process of the degradation of PAHs was most intensive during the first 30 days of the experiment; however, three-ring PAHs (fluorene and anthracene) definitely decomposed faster than the four-ring ones (pyrene and chrysene). Mitochondrial defects have been shown to be associated with the pathogenesis of Parkinson's disease (PD). Yet, experience in PD research linking mitochondrial dysfunction, e.g., deregulation of oxidative phosphorylation, with neuronal degeneration and behavioral changes is rather limited. Using the 6-hydroxydopamine (6-OHDA) rat model of PD, we have investigated the potential role of mitochondria in dopaminergic neuronal cell death in the substantia nigra pars compacta by high-resolution respirometry. Mitochondrial function was correlated with the time course of disease-related motor behavior asymmetry and dopaminergic neuronal cell loss, respectively. Unilateral 6-OHDA injections (>2.5 μg/2 μl) into the median forebrain bundle induced an impairment of oxidative phosphorylation due to a decrease in complex I activity. This was indicated by increased flux control coefficient. During the period of days 2-21, a progressive decrease in respiratory control ratio of up to −58 % was observed in the lesioned compared to the non-lesioned substantia nigra of the same animals. This decrease was associated with a marked uncoupling of oxidative phosphorylation. Mitochondrial dysfunction, motor behavior asymmetry, and dopaminergic neuronal cell loss correlated with dosage (1.25-5 μg/2 μl). We conclude that high-resolution respirometry may allow the detection of distinct mitochondrial dysfunction as a suitable surrogate marker for the preclinical assessment of potential neuroprotective strategies in the 6-OHDA model of PD. The area of environmental protection concern minimises the impact that technical objects have on the environment. Usually the most effective way of protecting the environment is to influence the source of the problem. For this reason studies are conducted to modify the construction of machines, power machines in particular, so as to minimise their impact on the environment. In the case of environmental protection from noise it is most convenient to carry out measurements in an anechoic chamber. Unfortunately, this is possible only in very limited circumstances. In all other cases measurements are performed using an engineering method or the survey method, both of which are described in the standards and by taking into account the so-called environmental corrections. The obtained results are burdened with greater error than those of measurements in an anechoic chamber. Therefore, it would seem advantageous to develop a method of obtaining similar and reliable results as those in an anechoic chamber, but in a reverberant field. The authors decided to use numerical modelling for this purpose. The main objective of this work is a comprehensive analysis of the numerical model of a laboratory designed for acoustic tests of selected power machines. The geometry of a room comprising an area of analysis is easy to design. The main difficulty in modelling the phenomena occurring in the analysed area can be the lack of knowing the boundary conditions. 37. Effect of soil pollution with polycyclic aromatic hydrocarbons on the properties of humic acids.

Purpose: Interestingly, soil is the component of the natural environment in which most hydrophobic organic pollution including polycyclic aromatic hydrocarbons (PAHs) gets accumulated. The aim of the present paper was to determine the effect of soil pollution with PAHs on the elemental composition, spectral properties, and hydrophobic and hydrophilic properties of humic acids. The research was performed on different types of soil samples that were artificially polluted with selected PAHs (anthracene, pyrene, fluorene and chrysene). Materials and methods: The soil samples were polluted with selected PAHs in an amount corresponding to 10 mg PAHs kg. The PAHs-polluted soil samples were incubated for 180 and 360 days at a temperature of 20-25 °C and fixed moisture (50 % of field water capacity). Humic acids (HAs) were extracted from the soil samples prior to the incubation (additionally, soils not polluted with PAHs) and after 180 and 360 days of incubation. For isolated HAs, the following analyses were performed: elemental composition, UV-Vis and IR spectra, susceptibility to oxidation, and hydrophilic (HIL) and hydrophobic (HOB) properties were determined using high-performance liquid chromatography. Results and discussion: The research demonstrated that introducing anthracene, fluorene, pyrene and chrysene to soil samples resulted in a change in some of the quality parameters of humic acids. However, the intensity and the direction of those changes were determined by soil properties. The changes of the parameters, once PAHs were introduced, that did not depend on the soil properties were Δ A and Δ A (susceptibility to oxidation at wavelengths of 465 and 665 nm) as well as HIL/ΣHOB. The same tendency in changes in the structure of humic acids, once PAHs were introduced, was also observed based on the Fourier transform infrared spectra pattern. Conclusions: A single pollution of soils with PAHs that leads to changes in the quality parameters of humic acids shows that, as for the soils permanently exposed to pollution with those compounds, significant changes can occur in the properties of humic acids. Charcot-Marie-Tooth neuropathy type 2A (CMT2A) is associated with heterozygous mutations in the mitochondrial protein mitofusin 2 (Mfn2) that is intimately involved with the outer mitochondrial membrane fusion machinery. The precise consequences of these mutations on oxidative phosphorylation are still a matter of dispute. Here, we investigate the functional effects of MFN2 mutations in skeletal muscle and cultured fibroblasts of four CMT2A patients applying high-resolution respirometry. While maximal activities of respiration of saponin-permeabilized muscle fibers and digitonin-permeabilized fibroblasts were only slightly affected by the MFN2 mutations, the sensitivity of active state oxygen consumption to azide, a cytochrome c oxidase (COX) inhibitor, was increased. The observed dysfunction of the mitochondrial respiratory chain can be explained by a twofold decrease in mitochondrial DNA (mtDNA) copy numbers. The only patient without detectable alterations of respiratory chain in skeletal muscle also had a normal mtDNA copy number. We detected higher levels of mtDNA deletions in CMT2A patients, which were more pronounced in the patient without mtDNA depletion. Detailed analysis of mtDNA deletion breakpoints showed that many deleted molecules were lacking essential parts of mtDNA required for replication. This is in line with the lack of clonal expansion for the majority of observed mtDNA deletions. In contrast to the copy number reduction, deletions are unlikely to contribute to the detected respiratory impairment because of their minor overall amounts in the patients. Taken together, our findings corroborate the hypothesis that MFN2 mutations alter mitochondrial oxidative phosphorylation by affecting mtDNA replication. [ABSTRACT FROM AUTHOR]

40. Method validation for determination of 13 elements in honey samples by ICP-MS.

- Abstract
A method for simultaneous ICP-MS determination of 13 elements in three types of honey from Poland is described. The method was validated, and the uncertainty budget was set up. The results obtained for the relative expanded uncertainties U ( k = 2) were 15.1% for Al, 18.6% for B, 18.8% for Ba, 7.9% for Ca, 24.4% for Cd, 7.24% for Cu, 7.9% for K, 4.8% for Mg, 8.3% for Mn, 12.7% for Na, 14.9% for Ni, 12.5% for Pb and 13.4% for Zn. Traceability of the measurement results was established based on the use of the corn flour CRM INCT-CF-3 and the apple leaves CRM SRM 1515 and by analyzing spiked samples. Recovery rates between 94% (Zn) and 107% (Na) were found. The detection limits of all elements studied showed the suitability of the procedure for routine analyses. Summarizing it can be concluded that the described analytical procedures to measure the mass fractions of 13 elements in honey samples with established traceability and evaluated uncertainty allow to obtain reliable and internationally comparable results. The paper presents an original method for sound reinforcement in open areas. The method enabled both a regular sound reinforcement and the required spatial impressions of sound to be achieved in the area used for the study. The inverse image source method was used for the disposition of sound sources in order to find the inverse problem solution for determining the configuration of additional sound sources. Simulations demonstrated the improvement of sound impressions in the area in question and the simulations results were verified experimentally. The intended result of the proposed method was the increase of the lateral energy fraction and lateral energy fraction coefficient parameter values by 6 and 8 points, respectively, for the simulation by 5 and 7 points for the experiment. It should be stated that, in both the simulation and the experiment, eligible values for the acoustic parameters were obtained after using the sound system with additional sound sources, the speech intelligibility value parameters remain at an excellent level. In conclusion, it may be claimed that the proposed sound reinforcement system makes the creation of the intended spatial sound impressions in an open area possible. [ABSTRACT FROM AUTHOR]

Serotonin (5-hydroxytryptamine, 5-HT) is an important neuromodulator in learning and memory processes. A functional genetic polymorphism of the 5-HT 2a receptor (5-HTR2a His452Tyr), which leads to blunted intracellular signaling, has previously been associated with explicit memory performance in several independent cohorts, but the underlying neural mechanisms are thus far unclear. The human hippocampus plays a critical role in memory, particularly in the detection and encoding of novel information. Here we investigated the relationship of 5-HTR2a His452Tyr and hippocampal novelty processing in 41 young, healthy subjects using functional magnetic resonance imaging (fMRI). Participants performed a novelty/familiarity task with complex scene stimuli, which was followed by a delayed recognition memory test 24 hours later. Compared to His homozygotes, Tyr carriers exhibited a diminished hippocampal response to novel stimuli and a higher tendency to judge novel stimuli as familiar during delayed recognition. Across the cohort, the false alarm rate during delayed recognition correlated negatively with the hippocampal novelty response. Our results suggest that previously reported effects of 5-HTR2a on explicit memory performance may, at least in part, be mediated by alterations of hippocampal novelty processing. The aim of this study was to determine the effect of the size of the soil particle containing organic matter, the type of organic fertiliser (cattle manure, vermicompost, straw) and the decomposition time of these fertilisers in soil on the hydrophilic-hydrophobic properties and polydispersity of humic acid molecules. In the model incubation experiment, organic fertilisers (cattle manure, vermicompost and wheat straw) were mixed with soil material (sandy texture). The soil was sampled for testing after one and three years of incubation. Each sample was separated into two fractions according to graining, grain diameter>0.1 mm (sand fraction) and<0.1 mm (silt and clay fraction), and humic acids (HAs) were isolated. For the HAs isolated, hydrophilic-hydrophobic properties were defined using RP-HPLC, and the division was made into low- and high-molecular-mass fractions (HPSEC). It was demonstrated that introducing organic fertilisers into soil resulted in an increase in the proportion of hydrophobic and high-molecular mass fractions and a decrease in the proportion of hydrophilic and low-molecular mass fractions in the HA molecules. Highest values for the ratio HIL/ΣHOB and lowest values for S1/S2 were noted for HAs of soil mixed with straw. HAs isolated from the fine-grained soil fraction demonstrated a higher degree of 'maturity' than HAs from the coarse-grained fraction, which was seen as higher values for HIL/ΣHOB and lower values for S1/S2. (ATP) , mtBK, and mtKv1.3. While the latter two share similarities with their plasma membrane counterparts, mtK(ATP) exhibits considerable differences with the plasma membrane K(ATP) -channel. One important function seems to be suppression of release of proapototic substances from mitochondria through the PTP. Open potassium channels tend to keep the PTP closed thus acting as antiapoptotic. Nevertheless, in their mode of action there are considerable differences among them. Objectives To describe the prevalence, management and outcome of spontaneous twin anemia polycythemia sequence (TAPS) diagnosed in the prenatal period. Method Retrospective analysis of 142 consecutive monochorionic twin pregnancies not diagnosed with twin to twin transfusion syndrome. TAPS cases were identified based on the presence of discordant middle cerebral artery peak systolic velocity (MCA-PSV) measurements and signs suggestive of a chronic intertwin transfusion imbalance: either an elevated reticulocyte count in the anemic twin or the presence of few small unidirectional anastomoses during fetoscopy or at postnatal placental examination. Results Three cases were identified, giving an estimated prevalence of 2%. Prenatal interventions were tailored to the characteristics of each case and consisted of intrauterine transfusion and interruption of the shared circulation by cord coagulation or laser separation. Conclusion In monochorionic twin pregnancies, TAPS is an uncommon prenatal finding. Nonetheless, its incidence seems high enough to recommend screening for this disease by MCA-PSV measurements. Patients with chronic renal failure (CRF) receive recombinant human erythropoietin (rhEPO) for the correction of anemia. However, rhEPO also has an immunomodulatory effect. Detailed changes of phenotype and function of CD4
+ T lymphocytes in CRF patients receiving rhEPO have not been reported yet; their study may bring insight into understanding of this immunomodulatory action of rhEPO. Two groups of CRF patients were included into the study: those treated; and those not receiving rhEPO. The expression of activation markers on CD4+ lymphocytes was measured with flow cytometry, both ex vivo and in vitro. The kinetics of CD4+ T lymphocytes proliferation was calculated using a dividing cells tracing method and numerical approach. Significantly higher percentages of CD4+ CD95+ , CD4+ HLA-DR+ cells, and lower percentages of CD4+ CD69+ and CD4+ CD28+ cells were observed in both rhEPO-treated and untreated patients when compared with healthy controls. Changes in the proportions of CD4+ CD28+ and CD4+ HLA-DR+ subpopulations were dependent on the type of rhEPO, being more pronounced for rhEPOβ. CD4+ lymphocytes from untreated patients exhibited decreased expression of CD28 and CD69 after stimulation in vitro, whereas the expression of these antigens on lymphocytes of rhEPO-treated patients was similar to that observed in healthy controls. Fewer CD4+ CD28+ T lymphocytes of untreated patients proliferated in vitro; these cells had longer G0→G1 time, which negatively correlated with surface expression of CD28. Our study confirms that rhEPO treatment normalizes activation parameters of CD4+ T lymphocytes and their proliferative capacity, which could explain earlier described immunomodulatory effects of rhEPO in patients suffering from CRF. Quality of life (QOL) in the context of education is an understudied topic and research related to its’ relation to the sense of coherence (SOC), level of optimism (LOO) and self-efficacy (SE) among students seems to be missing. The rationale for this survey was to fill this gap in knowledge and therefore, the aim of the study is to examine the relationship between students’ QOL and psychological variables such as SOC, LOO and SE, and social ones such as type of faculty, students’ social background and sex. The quantitative cross sectional survey has been conducted at three higher education institutions in Poland in January 2006. The random sample of 396 undergraduate students (average age was 20.5 years) representing 5 different faculties namely physiotherapy, physical education, tourism and recreation, English and Polish philology has been selected and participated in the study. Four reliable and validated research tools have been used to collect the data. These included the SF-36, SOC-29, Seligman’s scale, and GSES. The results have shown to indicate that different variables had a significant effect on particular subscales of the QOL outcomes. For example, among social variables faculty had a significant effect on physical functioning, role limitation due to physical functioning, and changes in health. Sex had a significant effect on role limitation due to emotional problems, social functioning, and energy vitality. Social background had a significant effect on general health. Among psychological variables the sense of coherence was significantly related to all subscales of QOL. A significant effect was noted by self-efficacy on physical and social functioning. Level of optimism had a significant effect on energy-vitality and general health. This research survey has suggested that changes in academic curriculum might be implemented to improve students QOL. Biochar (BC) is a material that finds many applications in agriculture and environmental activities. The aim of the study was to define the influence of biochar produced from various organic materials: mellow compost (MC), stabilized municipal sewage sludge (MSS), pine sawdust (PS), sycamore sawdust (SS) and oak leaves (OL) on soil enzyme activity, as well as its relations with carbon and nitrogen content. After a 60-day incubation of soil and BC, the activity of dehydrogenases (DEH), catalase (CAT), alkaline (AlP) and acid (AcP) phosphatases was investigated. The basic parameters of soil were also determined: TOC, TN, DOM, pH in H
2 O, available phosphorus (AP). The highest AP content was obtained in the S + MSS, S + OL and S + MC variants. Enzyme activity was highest in soil with MSS BC, regardless of incubation time. After 60 days, the activity of soil enzymes was inhibited. The obtained results indicate that the response of enzymatic activity to biochar depends on the feedstock material and the incubation time. When using BC as an exogenous matter, it is necessary to determine the TOC/TN ratio. For the very wide range of this parameter, supplemental nitrogen fertilization or mixtures of different biochars should be applied. [ABSTRACT FROM AUTHOR]- Published

In patients with mesial temporal lobe epilepsy (MTLE) it remains an unresolved issue whether the interictal decrease in N-acetyl aspartate (NAA) detected by proton magnetic resonance spectroscopy (
1 H-MRS) reflects the epilepsy-associated loss of hippocampal pyramidal neurons or metabolic dysfunction. To address this problem, we applied high-resolution1 H-MRS at 14.1 Tesla to measure metabolite concentrations in ex vivo tissue slices from three hippocampal subfields (CA1, CA3, dentate gyrus) as well as from the parahippocampal region of 12 patients with MTLE. In contrast to four patients with lesion-caused MTLE, we found a large variance of NAA concentrations in the individual hippocampal regions of patients with Ammon's horn sclerosis (AHS). Specifically, in subfield CA3 of AHS patients despite of a moderate preservation of neuronal cell densities the concentration of NAA was significantly lowered, while the concentrations of lactate, glucose, and succinate were elevated. We suggest that these subfield-specific alterations of metabolite concentrations in AHS are very likely caused by impairment of mitochondrial function and not related to neuronal cell loss. A subfield-specific impairment of energy metabolism is the probable cause for lowered NAA concentrations in sclerotic hippocampi of MTLE patients. [ABSTRACT FROM AUTHOR]- Published

Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative disease characterized by a progressive loss of motor neurons. Visualizing corresponding metabolic changes in the brain of patients with ALS with proton magnetic resonance spectroscopy (
1 H-MRS) may provide surrogate markers for an early disease detection, for monitoring the progression and for evaluating a treatment response. The primary objective of our study was to evaluate whether modifications in MR metabolite levels occur before clinical disease onset, and whether these changes are directly linked to a distinct spatial progression pattern in the CNS. Therefore, age-dependent alterations in the cerebral and spinal metabolic profile in the mouse model of ALS overexpressing the mutated human G93A-superoxide dismutase 1 (G93A-SOD1) were determined by high-resolution MRS of tissue extracts at 14.1 Tesla. Both non-transgenic mice (control mice) and transgenic mice overexpressing the non-mutated human SOD1 (tg-SOD1) served as controls. In the spinal cord of G93A-SOD1 mice significantly decreased levels of N-acetyl aspartate were already detected 34 days postpartum, i.e. about 60 days before the average disease onset caused by motor neuron decline. In addition, glutamine and γ-aminobutyric acid concentrations were significantly diminished at Day 75, which is still in the presymptomatic phase of the disease. These metabolic changes were further progressive in the course of the disease and started to involve the brainstem at Day 75. Overall, high-resolution1 H-MRS allows a sensitive spatial and temporal metabolite profiling in the presymptomatic phase of ALS even before significant neuronal cell loss occurs.
