Mathematical models may aid researchers in describing biological processes, like growth, in animals. This study aimed to collect the body weight data of 18 Boer goat castrates and 20 Boer goat does, from birth until maturity, to model growth and determine growth trends. This is a novel investigation as sufficient information on an age-weight database for these two Boer goat sexes from birth to maturity, is lacking. Using age-weight data, four nonlinear models, namely the Brody, Gompertz, Logistic and Von Bertalanffy growth models, were plotted and evaluated. The model parameters of each growth model were compared for differences between the two sexes. The statistical effectiveness of fit was determined for each model using AIC and RMSE, with R2 also being considered. All models except the Brody model, predicted significantly heavier mature weights for castrates. The Brody model was deemed unfit to describe Boer goat growth as the function severely over-predict weights from birth until maturity for both sexes. The Von Bertalanffy (R2 = 91.3) and Gompertz functions (R2 = 91.3) showed the best fit for Boer goat castrates, while the Gompertz model (R2 = 95.1) showed the best fit for Boer goat does. The Gompertz function is the preferred model to depict Boer goat growth overall, as it accurately characterized growth of both sexes. According to the Gompertz model the age at which the inflection point of the growth curve was reached, did not differ significantly between castrates and does (141.80 days versus 136.31 days). Producers require an accurate predictive tool that can determine the optimal point of slaughter based on fat depth. The modelling of fat deposition with a simple mathematical model could supply in this need. Dohne Merino and Merino ewes were crossed with Dorper, Dormer and Ile de France rams or rams of their own breeds to create two purebred (Dohne Merino and Merino) and six crossbred groups (Dohne x Dorper, Dohne x Dormer, Dohne x Ile de France, Merino x Dorper, Merino x Dormer and Merino x Ile de France) of offspring. Fat deposition of four lambs of each sex per genotypic group was monitored from 80 to 360 days using ultrasound, and the data subsequently fitted to various equations and evaluated for goodness of fit. A linear fitting of fat depth to age (R2 > 0.77) and live weight (R2 > 0.56) were deemed to provide the best fit. The slope parameters of the equations indicated that ewes deposited fat faster than rams and that Dorper crosses had the highest fat deposition rate. An attempt was also made to model loin muscle growth, but the model fit was judged to be unsatisfactory. Nanomaterials are a central pillar in modern medicine. They are thought to optimize drug delivery, enhance therapeutic efficacy, and reduce side-effects. To foster this technology, analytical methods are needed to validate not only the localization and distribution of these nanomaterials, but also their compatibility with cells, drugs, and drug release. In the present work, we assessed nanoparticles based on porous silicon (pSiNPs) loaded with the clinically used tyrosine kinase inhibitor sunitinib for their effectiveness of drug delivery, release, and toxicity in colon cancer cells (HCT 116 cells) and cardiac myoblast cells (H9c2) using Raman micro-spectroscopy, highresolution fluorescence microscopy, along with biological methods for toxicological effects. We produced pSiNPs with a size of about 100 nm by grinding mesoporous silicon layers. pSiNPs allowed an effective loading of sunitinib due to their high porosity. Photoluminescence properties of the nanoparticles within the visible spectrum allowed the visualization of their uptake in cardiac cells. Raman micro-spectroscopy allowed not only the detection of the uptake and distribution of pSiNPs within the cells via a characteristic silicon Raman band at about 518-520 cm-1, but also the localization of the drug based on its characteristic molecular fingerprints. Cytotoxicity studies by Western blot analyses of apoptotic marker proteins such as caspase-3, and the detection of apoptosis by subG1-positive cell fractions in HCT 116 and MTT analyses in H9c2 cells, suggest a sustained release of sunitinib from pSiNPs and delayed cytotoxicity of sunitinib in HCT 116 cells. The analyses in cardiac cells revealed that pSiNPs are well tolerated and that they may even protect from toxic effects in these cells to some extent. Analyses of the integrity of mitochondrial networks as an early indicator for apoptotic cellular effects seem to validate these observations. Our study suggests pSiNPs-based nanocontainers for efficient and safe drug delivery and Raman micro-spectroscopy as a reliable method for their detection and monitoring. Thus, the herein presented nanocontainers and analytical methods have the potential to allow an efficient advancement of nanoparticles for targeted and sustained intracellular drug release that is of need, e.g., in chronic diseases and for the prevention of cardiac toxicity. Marine phytoplankton contribute about one half of global primary production and play a key role in global biogeochemical cycles. High cell densities in extensive phytoplankton blooms are expected to be modified by global changes in ocean circulation and stratification, acidification and carbonation, solar radiation, temperature, and eutrophication. Although photochemical gas production from chromophoric dissolved organic matter (CDOM) has been widely studied, ultraviolet (UV) effects on emissions from phytoplankton cells themselves have not been fully explored. We therefore investigated UV‐driven emissions of carbon monoxide (CO), carbon dioxide (CO2), methane (CH4), ethene (C2H4), ethane (C2H6), and nitrous oxide (N2O) from cell suspensions of 16 phytoplankton species and their filtrates under controlled experimental conditions. These gases make direct or indirect contributions to radiative forcing of the atmosphere or contribute to atmospheric chemistry including stratospheric ozone (O3) depletion. We observed production of CH4, CO, CO2, C2H4, and N2O from cell suspensions and CO, CO2, and N2O after 0.45 μm‐filtration to remove phytoplankton cells. CH4 production was only observed with cells present, whereas N2O was still produced after filtration. Production of CO from filtrates was 30%–90% of that from cell suspensions in all but two species with a CO2:CO mole ratio from filtrates always below one. Our results clearly demonstrate a need to quantify the production potentials of these climate‐relevant gases in situ under natural sunlight using key phytoplankton species, especially those forming blooms which are predicted to change in prevalence and distribution with future global change scenarios. Plain Language Summary: The oceans play an important role in the global exchange of gases with the atmosphere including the greenhouse gases carbon dioxide and methane that contribute to global warming. Several recent studies have shown that phytoplankton metabolism can produce methane during growth. Other studies also demonstrated that sunlight can degrade dissolved organic matter and plastic particles in seawater to produce methane and other gases. However, few studies have examined whether the ultraviolet radiation in sunlight can drive production of methane and other climate‐relevant gases from the cells of phytoplankton. In this study, we exposed cell suspensions of 16 phytoplankton species to controlled levels of ultraviolet radiation in the laboratory. We observed production of methane, carbon dioxide, carbon monoxide, ethene and also the important greenhouse gas nitrous oxide from cell suspensions. Nitrous oxide was still produced after phytoplankton cells were removed by filtration, whereas methane was only produced with cells present. These gases make direct or indirect contributions to global warming and atmospheric chemistry. Blooms of phytoplankton cells may cover extensive areas of the ocean. Consequently, it is important to evaluate the role of the ultraviolet component of sunlight to emissions of these gases under future global change scenarios. In the intensification of sheep production systems, feedlot finishing plays a fundamental role in preparing lambs for slaughter, as well as relieving the grazing pressure on pasture. The profit margins in feedlot operations are often narrow and require the economics of scale to generate a sufficient income. In order to minimise expenses, intensive management and precision rearing of lambs to an ideal slaughter weight is needed to obtain premium carcass prices. The South African sheep industry is made up of wool, dual-purpose as well as meat type breeds, which also vary in terms of maturity. In order to implement precision finishing of South African lamb, a complete understanding of the growth, intake and fat deposition trends of growing lambs of different breed types is needed. This review outlines feedlot lamb production within the Southern African context for the major commercial breeds, while also providing insight in the considerations necessary to develop a decision support system for lamb rearing. Integrating such a decision support system into a lamb feedlot operation can then be used for precision finishing of lambs by predicting the optimal length of the feeding period and ideal slaughter weights of lambs. Background Information is collected worldwide on the diagnosis and assessment of occupational diseases (ODs) by occupational physicians (OPs). However, information on perceived facilitators and barriers to assessment is scarce. Aims To evaluate the perceived barriers and facilitators in the assessment of ODs by OPs. Methods We conducted a qualitative study, using interviews and focus groups. We held 12 interviews and two focus groups with Dutch OPs, to identify barriers and facilitators in the assessment of ODs. Results Case definition, exposure assessment, attribution to work, guidelines and decision tools, external expertise, individual motivation and consequences were identified as themes. Barriers and facilitators were mainly reported regarding assessing work attribution, e.g. how to assess multifactorial causes or the need for training, and individual motivation such as the perceived lack of usefulness or the need for monitoring suspected OD cases. Within the theme of consequences, only barriers to the assessment of ODs were reported, including the liability of employers. Conclusions Perceived facilitators in the assessment of ODs were practical assessment tools, multifaceted education, ability to assess work exposures and professional independence. Perceived barriers were lack of usefulness, lack of urgency, complexity of assessment and concerns about liability issues. [ABSTRACT FROM AUTHOR]

Context. Studies of galaxy clusters provide stringent constraints on models of structure formation. Provided that selection effects are under control, large X-ray surveys are well suited to derive cosmological parameters, in particular those governing the dark energy equation of state. Aims. We forecast the capabilities of the all-sky eROSITA (extended ROentgen Survey with an Imaging Telescope Array) survey to be achieved by the early 2020s. We bring special attention to modelling the entire chain from photon emission to source detection and cataloguing. Methods. The selection function of galaxy clusters for the upcoming eROSITA mission is investigated by means of extensive and dedicated Monte-Carlo simulations. Employing a combination of accurate instrument characterisation and a state-of-the-art source detection technique, we determine a cluster detection efficiency based on the cluster fluxes and sizes. Results. Using this eROSITA cluster selection function, we find that eROSITA will detect a total of approximately 105 clusters in the extra-galactic sky. This number of clusters will allow eROSITA to put stringent constraints on cosmological models. We show that incomplete assumptions on selection effects, such as neglecting the distribution of cluster sizes, induce a bias in the derived value of cosmological parameters. Conclusions. Synthetic simulations of the eROSITA sky capture the essential characteristics impacting the next-generation galaxy cluster surveys and they highlight parameters requiring tight monitoring in order to avoid biases in cosmological analyses. Background The prevention of occupational diseases is limited by a lack of insight into occupational exposure to risk. We developed a six-step approach to improve the diagnosis and reporting of occupational diseases and the selection of subsequent preventive actions by occupational physicians (OPs). Aims To evaluate the effect of the six-step approach on the transparency and quality of assessing occupational diseases and the usability of the six-step approach according to OPs and their satisfaction with it. Methods A randomized controlled trial. OPs in the control group used the standard information available. OPs in the intervention group used the six-step approach and accompanying educational materials. The actions and decisions of OPs in both groups were analysed using 17 performance indicators. To address the second issue, OPs used the six-step approach over 6 weeks and rated its usability and their satisfaction in relation to several aspects. Results The average score of the OPs in the intervention group (n = 110) was statistically significantly higher (11/17 performance indicators, 62% of the maximum score) than that of the OPs in the control group (n = 120, 5/17 performance indicators, 30% of the maximum score, P < 0.001). The usability aspects of the six-step approach had mean scores of 7 and 8. Mean satisfaction with the six-step approach was 8. Conclusions The six-step approach resulted in better evidence-based and transparent decision-making about occupational diseases by OPs. The activity of sulfate reducing bacteria (SRB) in domestic wastewater treatment plants (WWTP) is often considered as a problem due to HS formation and potential related odour and corrosion of materials. However, when controlled well, these bacteria can be effectively used in a positive manner for the treatment of wastewater. The main advantages of using SRB in wastewater treatment are: (1) minimal sludge production, (2) reduction of potential pathogens presence, (3) removal of heavy metals and (4) as pre-treatment of anaerobic digestion. These advantages are accessory to efficient and stable COD removal by SRB. Though only a few studies have been conducted on SRB treatment of domestic wastewater, the many studies performed on industrial wastewater provide information on the potential of SRB in domestic wastewater treatment. A key-parameter analyses literature study comprising pH, organic substrates, sulfate, salt, temperature and oxygen revealed that the conditions are well suited for the application of SRB in domestic wastewater treatment. Since the application of SRB in WWTP has environmental benefits its application is worth considering for wastewater treatment, when sulfate is present in the influent. [ABSTRACT FROM AUTHOR]

Low-fill-factor laser bars composed of five narrow-stripe broad-area lasers (30 \mu \textm \,\, \times \,\, 6000~\mu \textm ) with monolithic spectral stabilization are presented. Each laser on the bar emits at a unique wavelength from 970 to 980 nm with a channel spacing of 2.5 nm. Narrow spectral line width <1 nm per emitter is obtained by implementing 40th Bragg order distributed feedback surface gratings with varied grating periods. The design and fabrication of these laser bars is reviewed, with optimal grating etch depth determined using simulations that combine 2D-simulation of the reflectivity and optical losses of a 40th-order surface grating with coupled mode theory. The selected etch depth delivers a grating coupling coefficient of \kappa \textL~\sim 0.2, which is shown to be sufficient to suppress lasing in Fabry-Perot modes for narrow line operation across all wavelength channels. However, additional optical scattering losses of the gratings are found to limit output power and efficiency. The final realized minibars deliver 25-W continuous wave output power with a conversion efficiency >40% and operate with a beam parameter product of $\sim 1.6$ mm mrad per single emitter, twofold improved in comparison with typical 90- \mu \text{m} wide broad area laser. In the sewage or wastewater treatment plant, biological sulphate reduction can occur spontaneously or be applied beneficially for its treatment. The results of this study can be applied to control SRB in the sewage and WWTP. Therefore, population diversity analyses of SRB for nine activated sludge wastewater treatment plants (WWTP) in the Netherlands and the effect of long-term (months) oxygen exposures on the SRB activity were carried out. T-RFLP and clone sequencing analyses of winter and summer samples revealed that (1) all WWTP have a similar SRB population, (2) there is no seasonal impact (10-20 °C) on the SRB population present in the WWTP and (3) Desulfobacter postgatei, Desulfovibrio desulfuricans and Desulfovibrio intestinalis were the most common and dominant SRB species observed in these samples, and origin from the sewage. Short term activity tests demonstrated that SRB were not active in the aerobic WWTP, but while flushed with N-gas SRB became slightly active after 3 h. In a laboratory reactor at a dissolved oxygen concentration of <2 %, sulphate reduction occurred and 89 % COD removal was achieved. SRB grew in granules, in order to protect themselves for oxygen exposures. SRB are naturally present in aerobic WWTP, which is due to the formation of granules. [ABSTRACT FROM AUTHOR]

Alzheimer's disease (AD) displays a high degree of heterogeneity in terms of its etiology, presentation, prognosis, and treatment response. This can partly be explained by high-penetrance mutations in the amyloid precursor protein, presenilin 1 and presenilin 2 genes causing amyloid beta aggregation, which is a major pathogenic mechanism in the development of early-onset AD in a small subgroup of patients. Late-onset AD is considered a polygenic disorder in which cumulative risk resulting from interaction with modifiable environmental risk factors may be responsible for the majority of cases. The ϵ-4 allele of the apolipoprotein E (APOE) gene has emerged as the most significant genetic risk factor for late-onset AD, influencing nearly every pathogenic domain affected in AD. It is a major risk factor for cerebral amyloid angiopathy, recognized as a common pathological finding in an AD subtype associated with white matter dysfunction. The APOE ϵ-4 allele is also a known risk factor for ischemic stroke, which can result in vascular dementia or contribute to subcortical vascular dysfunction. In this review, we evaluate the clinical relevance of APOE genotyping in relation to cholesterol metabolism and available evidence on risk reduction strategies applicable to AD. The effects of different dietary energy concentrations on ostrich production variables were examined in two separate trials. The first trial tracked changes in production variables from the pre-starter phase through the starter phase and grower phase. The second trial was based on the finisher phase per se. In both trials, the influence of dietary energy on feed intake, feed conversion ratio (FCR) and growth variables was investigated. Additionally, basic abattoir weights were recorded, and measurements of the feathers and skin were performed.In both trials, three diets with different concentrations of dietary energy were given during each phase where the low-, medium- and high-energy concentrations for each phase were as follows: 13.5, 14.5 and 15.5 MJ ME/kg feed pre-starter; 12.5, 13.5 and 14.5 MJ ME/kg feed starter; 10.5, 11.5 and 12.5 MJ ME/kg feed grower and 9.5, 10.5 and 11.5 MJ ME/kg feed finisher. Feed and water were available ad libitum in both trials.Overall, it was found that the best performance for growth, FCR, skin size and grade, live weight, carcass weight and thigh weight were obtained on the medium-energy diet during the pre-starter, starter and grower phases.During the finisher phase, improved growth rate and tanned skin size was found in birds given the diet with the highest energy concentration (11.5 MJ ME/kg feed). Seawater toilet flushing, seawater intrusion in the sewerage, and discharge of sulfate-rich industrial effluents elevates sulfate content in wastewater. The application of sulfate-reducing bacteria (SRB) in wastewater treatment is very beneficial; as for example, it improves the pathogen removal and reduces the volume of waste sludge, energy requirement and costs. This paper evaluates the potential to apply biological sulfate reduction using acetate and propionate to saline sewage treatment in moderate climates. Long-term biological sulfate reduction experiments at 10 and 20 °C were conducted in a sequencing batch reactor with synthetic saline domestic wastewater. Subsequently, acetate and propionate (soluble organic carbon) conversion rate were determined in both reactors, in the presence of either or both fatty acids. Both acetate and propionate consumption rates by SRB were 1.9 times lower at 10 °C than at 20 °C. At 10 °C, propionate was incompletely oxidized to acetate. At 10 °C, complete removal of soluble organic carbon requires a significantly increased hydraulic retention time as compared to 20 °C. The results of the study showed that biological sulfate reduction can be a feasible and promising process for saline wastewater treatment in moderate climate. [ABSTRACT FROM AUTHOR]

The epidermal growth factor receptor (EGFR) is a central regulator of tumor progression in human cancers. Cetuximab is an anti-EGFR antibody that has been approved for use in oncology. Previously we investigated mechanisms of resistance to cetuximab using a model derived from the non-small cell lung cancer line NCI-H226. We demonstrated that cetuximab-resistant clones (CtxR) had increased nuclear localization of the EGFR. This process was mediated by Src family kinases (SFKs), and nuclear EGFR had a role in resistance to cetuximab. To better understand SFK-mediated nuclear translocation of EGFR, we investigated which SFK member(s) controlled this process as well as the EGFR tyrosine residues that are involved. Analyses of mRNA and protein expression indicated upregulation of the SFK members Yes (v-Yes-1 yamaguchi sarcoma viral oncogene) and Lyn (v-yes-1 Yamaguchi sarcoma viral-related oncogene homolog) in all CtxR clones. Further, immunoprecipitation analysis revealed that EGFR interacts with Yes and Lyn in CtxR clones, but not in cetuximab-sensitive (CtxS) parental cells. Using RNAi interference, we found that knockdown of either Yes or Lyn led to loss of EGFR translocation to the nucleus. Conversely, overexpression of Yes or Lyn in low nuclear EGFR-expressing CtxS parental cells led to increased nuclear EGFR. Chromatin immunoprecipitation (ChIP) assays confirmed nuclear EGFR complexes associated with the promoter of the known EGFR target genes B-Myb and iNOS. Further, all CtxR clones exhibited upregulation of B-Myb and iNOS at the mRNA and protein levels. siRNAs directed at Yes or Lyn led to decreased binding of EGFR complexes to the B-Myb and iNOS promoters based on ChIP analyses. SFKs have been shown to phosphorylate EGFR on tyrosines 845 and 1101 (Y845 and Y1101), and mutation of Y1101, but not Y845, impaired nuclear entry of the EGFR. Taken together, our findings demonstrate that Yes and Lyn phosphorylate EGFR at Y1101, which influences EGFR nuclear translocation in this model of cetuximab resistance. [ABSTRACT FROM AUTHOR]

The purpose of this study was to compare the effect of different transport distances on ostrich meat quality parameters, weight loss, and dressing percentage. Twenty-four ostriches were transported for 0 (T0), 60 (T60) and 600 (T600) km before being slaughtered. There was a significant difference (P = 0.038) in the percentage liveweight loss during transport and lairage between T60 km (2.4 ± 2.185%) and T600 km (8.13 ± 1.156%), respectively. There was also a significant difference (P = 0.003) in dressing percentage (farm weight to cold carcass weight) between the two abovementioned groups (T60 = 49.77%; T600 = 46.86%). Treatments also had a significant effect (P < 0.05) on ultimate pH (pH[sub u]), with the ostriches that did not travel exhibiting the lowest average pH[sub u] (5.77 ± 0.053) and the birds that travelled 600 km having the highest average pH[sub u] (6.11 ± 0.053). No differences (P > 0.05) were found in shear force and percentage cooking loss between the treatments, although all three treatments differed significantly (P < 0.05) from each other in drip loss (T0 km = 0.40%; T60 km = 1.36%; T600 km = 0.97%). Significant differences (P < 0.05) were found for L*, b* and hue-angle colour ordinates between the three treatments, while there were no differences (P > 0.05) in a* and chroma values. A significant correlation was found between hue angle (r = 0.528; P = 0.008) and pH[sub u]. The results of this study indicate that ostrich transport distance influences liveweight loss and meat quality. Little is known of the effect of transport distance on the meat quality of ostriches. Ostriches travelled 60 or 600 km to quantify the effect of transport distance on their meat quality. Objectives Work-related risk factors, such as long work hours, and physically demanding work have been suggested to adversely influence pregnancy outcome. The authors aimed to examine associations between various aspects of physically demanding work with fetal growth in different trimesters during pregnancy and the risks of adverse birth outcomes. Methods Associations between physically demanding work and fetal growth were studied in 4680 pregnant women participating in a population-based prospective cohort study from early pregnancy onwards in the Netherlands (2002-2006). Mothers who filled out a questionnaire during mid-pregnancy (response 77% of enrolment) were included if they conducted paid employment and had a spontaneously conceived singleton live born pregnancy. Questions on physical workload were obtained from the Dutch Musculoskeletal Questionnaire and concerned questions on lifting, long periods of standing or walking, night shifts and working hours. Fetal growth characteristics were repeatedly measured by ultrasound and were used in combination with measurements at birth. Results There were no consistent significant associations between physically demanding work nor working hours in relation to small for gestational age, low birth weight or preterm delivery. Women exposed to long periods of standing had lower growth rates for fetal head circumference (HC), resulting in a reduction of approximately 1 cm (3%) of the average HC at birth. Compared with women working <25 h/week, women working 25-39 h/week and >40 h/week had lower growth rates for both fetal weight and HC, resulting in a difference of approximately 1 cm in HC at birth and a difference of 148-198 g in birth weight. Objective To investigate the influence of maternal working conditions on fertility and pregnancy outcomes. Methods 8880 women were enrolled in a large prospective birth cohort during early (76%), mid (21%) or late pregnancy (3%) (61% participation). Complete questionnaire information was available for 6302 women (71% response). Outcomes were prolonged time to pregnancy (TTP) (>6 months), preterm birth (<37 weeks) and decreased birth weight (<3000 g). Self-reported exposure to chemical agents was based on a limited list of chemicals. Physical load questions concerned manual materials handling, prolonged sitting and long periods of standing. A job-exposure matrix (JEM) linked reported job title to workplace chemical exposure within jobs according to expert judgement. Associations between maternal occupational exposure and fertility and pregnancy outcomes, adjusted for age, education, minority, parity, smoking and alcohol use, were studied using logistic regression analysis. Results Women in jobs with regular handling of loads ≥5 kg had better fertility and pregnancy outcomes. No self-reported exposure to chemicals was associated with any outcomes and self-assessments had very low reliability compared with JEM-based assessments. JEM-based maternal occupational exposure to phthalates was associated with prolonged TTP (OR 2.16, 95% CI 1.02 to 4.57) and exposure to pesticides was associated with decreased birth weight (OR 2.42, 95% CI 1.10 to 5.34). The population attributable fractions were small at 0.7% for phthalates and 0.7% for pesticides. Conclusion This birth cohort study presents evidence of health-based selection into the workforce and adverse effects of maternal occupational exposure to phthalates and pesticides on fertility and pregnancy outcomes. The purpose of this study was to determine if the efficiency of energy retention in pregnant cows was dependent on the time during the pregnancy that feed was offered. Our hypothesis was that restricting feed intake during the second trimester of gestation and providing the saved feed during the third trimester was less energetically efficient than providing the feed during the second trimester. Twenty cows (4 breed composite: 1/4 Hereford, 1/4 Angus, 1/4 Red Poll, and 1/4 Pinzgauer) that had produced 1 calf before the study were fed a diet that consisted of(DM basis) 67.3% chopped corn silage, 27.0% alfalfa hay, 5.5% corn, and 0.2% NaCl. When the cows were 87 ± 0.6 d pregnant, the first nutrient balance measurement was conducted. Six subsequent nutrient balance measurements were taken on d 122 ± 0.6, 143 ± 0.6, 171 ± 0.6, 206 ± 0.6, 241 ± 0.6, and 262 ± 0.6 of gestation. Each nutrient balance measurement consisted of a 96-h total collection of feces and urine and a 24-h indirect calorimetry measurement. Ten cows were fed for moderate BW gain during the entire pregnancy, and 10 cows were feed-restricted in the second trimester and realimented during the third trimester (low-high, L-H). The BW of the cows at parturition (559 ± 14 kg) did not differ between treatments (P = 0.20). There was a general trend for the proportion of ME intake retained to decrease in moderate cows as pregnancy progressed. The proportion of ME intake retained in L-H cows decreased during the first 49 d of feed restriction, but the proportion of ME retained after 77 d of restriction was greater than that retained at 49 d of restriction. During realimentation, there were no time effects for efficiency of ME conversion to retained energy, but efficiency was greater for L-H cows than moderate cows (P < 0.001). The ability of the cow to adapt its energy metabolism during periods of moderate feed restriction and realimentation allows development of management strategies that alter the time interval of the production cycle during which supplemental feed is offered. Left-right asymmetry of internal organs is widely distributed in the animal kingdom. The chick and mouse embryos have served as important model organisms to analyze the mechanisms underlying the establishment of the left-right axis. In the chick embryo many genes have been found to be asymmetrically expressed in and around the node, while the same genes in the mouse show symmetric expression patterns. In the mouse there is strong evidence for an establishment of left-right asymmetry through nodal cilia. In contrast, in the chick and in many other organisms left-right asymmetry is probably generated by an early-acting event involving membrane depolarization. In both birds and mammals a conserved Nodal-Lefty-Pitx2 module exists that controls many aspects of asymmetric morphogenesis. This review also gives examples of divergent mechanisms of establishing asymmetric organ formation. Slow filtration is a widespread and environmentally sound method for the disinfestation of nutrient solutions in closed hydroponic systems. The efficacy of slow filters can be explained in part by straining and adsorption of pathogens onto filter material surfaces. Both mechanisms are influenced by choice of filter material, flow rate of the nutrient solution, biomass and extent of extracellular polymers on the filter skin. In a 2-year study, the total polysaccharide content and enzyme activity of the filter skin, and filter efficacy were monitored. Closed hydroponic systems using the nutrient film technique connected to slow filters were run with or without weekly inoculation with a fungal cell wall preparation (10.5 g m-2 of filter surface). This organic matter amendment increased the amount of total polysaccharide and enzyme activity within the filter skin. The fungal cell wall preparation did not accumulate, but was degraded rapidly. The total polysaccharide content (tPS; in mg g-1) of the filter skin increased linearly with time at 34.50 mg g-1 + 0.26 mg g-1 d-1 × days (d) (R² = 0.556; P = 0.02) for untreated filters, and at 43.40 mg g-1 + 0.70 mg g-1 d-1 × days (d) (R² = 0.826; P = 0.000) for treated filters. The extracellular polysaccharide content in the filter skin, determined after 16 and 20 weeks of slow filter operating time, ranged from 87.9 - 109.7 mg g-1 mineral wool (rockwool) sampled. The amount of extracellular polysaccharide was not influenced by the total polysaccharide content of the filter skin, by enzyme activity, or by addition of the fungal cell wall preparation. There was a trend towards higher filter efficacies when fungal cell wall preparations were added to the filter skin (treated: 98.3% ± 0.96; untreated: 97.9% ± 1.8); however, on the basis of the present data, no correlation between the assessed parameters and filter efficacies were observed. For a beef cow to continue in an annual production cycle, she must rebreed within 3 mo after calving. Malnutrition during this period frequently results in failure of the cow to become pregnant. The energetic needs of the cow are increased by lactation, and additional energy is required for growth of the primiparous cow. Determining energy expenditures during the first 40 to 60 d postpartum is critical to developing feed programs that will allow cows to become pregnant with a second calf. Sixty-seven balance trials were conducted on 25 MARC III cows (4-breed composite: ¼ Hereford, ¼ Angus, ¼ Red Poll, and ¼ Pinzgauer) that were between 3 and 53 d in milk. Cows' BW were 481 ± 4 kg. Metabolizable energy intake ranged from 14.8 to 28.9 Mcal/d. Milk yields ranged from 4.7 to 13.3 kg/d. Recovered energy (RE) increased linearly with increased ME intake. Forty-seven observations were obtained with cows in negative tissue energy (TE) balance, and 20 observations were obtained with cows in positive TE balance. Estimated zero RE from regression analysis of RE on ME intake was 146 kcal of ME/kg of BW0.75. Efficiency of conversion of ME to lactation energy (LE) was 72%. The efficiency for conversion of ME to TE and the conversion of TE to LE was 78%. We hypothesized that adaptation of heat production in the realimented cow would occur over an extended period, and the length of time would be influenced by the level of feed. Our objectives were to quantify the changes in heat production of cows after feed restriction and to quantify the effect of level of realimentation on the dynamics of heat production in lightweight cows. Forty 4-yr-old nonpregnant, nonlacting cows (4-breed composite: 1/4 Hereford, 1/4 Angus, 1/4 Red Poll, and 1/4 Pinzgauer) were randomly assigned to receive 1 of 4 levels of a common alfalfa hay source. All cows were feed-restricted [50.0 g of DMI metabolic body size (MBS, kg of BW0.75); period 1], and individual fed heat production measurements were taken 0, 7, 13, 28, 56, and 91 d after feed restriction (period 1). In period 2, cows were fed their assigned feed level for their treatment after d 91 of restriction: 50.0 (T50.0), 58.5 (T58.5), 67.0 (T67.0), and 75.5 (T75.5) g of DM/MBS. Measures were taken at 7, 13, 28, 42, 56, 91, 119, and 175 d. In period 3, all cows were fed 75.5 g of DM/MBS after their 175-d measurement, and measures were taken at 7, 14, 28, 56, and 112 d later. In period 1, heat production decreased rapidly during the first 7 d of feed restriction, and heat production continued to decrease during the 91-d restriction. Heat production increased rapidly within the first 7 d, but chronic adaptation continued for T75.5 and T67.0 cows. In period 3, heat production increased rapidly during the first 7 d. Heat production scaled for metabolic body size tended to differ among treatments (P = 0.11). Daily heat production increased by 2.5 kcal/d. The potential of the microflora in nutrient solutions to produce cell wall degrading enzymes (CWDE) was investigated by adding glucose or substrates of CWDE, such as chitin, cellulose, curdlan and preparations of fungal mycelia (0, 0.01 and 0.1%, w/v). The results indicate the potential of the microflora in nutrient solutions to produce proteolytic, chitinolytic, cellulolytic as well as β-1,3-glucanolytic enzymes. All enzyme complexes were induced by addition of preparations of Fusarium oxysporum f. sp. cyclaminis (Focy) and Pythium ultimum, respectively. In contrast, addition of glucose to nutrient solution resulted in only slight increase of protease and chitinase. No correlation between increased activity of CWDE and survival of Focy was found. Slow filtration is an effective, reliable and economically reasonable technique for disinfestation of nutrient solution in closed irrigation systems. However, the problematic control of its efficacy is a major shortcoming of this technique. The efficacy of slow filters against pathogenic microorganisms is caused by a complex combination of mechanical, physical-chemical and biological factors. Due to the importance of biological factors, parameters describing biological processes are possible factors for predicting the efficacy of slow filters. The natural occurrence, induction and significance of cell wall-degrading enzymes (CWDE, such as protease, chitinase, cellulase and β-1,3-glucanase) in slow filters were investigated. On non-treated slow filters of rockwool, proteolytic and chitinolytic activity was enhanced mainly in the top filter layer compared with the nutrient solution. For cellulase and β-1,3-glucanase activity, no accumulation could be found. Induction of all CWDE was possible in situ with a preparation of Fusarium oxysporum f. sp. cyclaminis. Statistical analysis revealed that enzyme activity on rockwool was correlated with the efficacy of slow filters (r2 = 0.495). Chitinase had the strongest influence, followed by protease and cellulase, whereas β-l,3-glucanase had no significance. 2,2'-Azoisobutyronitril (AIBN) initiated radical polymerization of trimethylsilyloxyethyl methacrylate (TMS-HEMA) was carried out in N,N-dimethylformamide (DMF) with the triazolinyl radical (T•) as additive. The latter effected a controlled polymerization process at least up to 60 % monomer conversion. Background: A recent cross sectional community readiness assessment carried out in Islamabad, Pakistan indicated that the community is at the denial or resistant stage for implementing school-based sexuality education in Pakistan. The resistance was attributed to lack of awareness and prevalent misconceptions regarding sexuality education. This study aims at qualitatively exploring strategies for targeting misconceptions and increase awareness regarding sexuality education with stakeholder involvement. Methods: 35 in-depth interviews and a focus group discussion was conducted to discuss the results of the readiness assessment comprising of community members, researchers, NGO representatives, and social media influencers. Discussion guide included topics on engagement strategies, intervention modes of delivery, target audience, misconceptions, community support, and optimizing sexuality education in local context. Qualitative content analysis was conducted for analyzing the data. Results: Results indicate that current community support is usually passive in nature and digital or social media platforms are mostly used to talk about sexuality education. Participants believed social media can be used as a platform for disseminating information by involving social media influencers with careful narrative building around sexuality for rebranding the topic, as talking about sexuality is seen as a taboo in the society. Targeting misconceptions through counter narratives and scientific evidence was considered as a way forward for increasing community acceptability and readiness. Violence, staying safe, and age appropriateness were identified as some of the key features of sexuality education that need to be highlighted for garnering community support. Conclusions: For promoting sexuality education in Pakistan, it is essential to tackle resistance by sensitizing the community and digital social media platforms offer an opportunity to do this through influencer marketing. Disadvantaged populations, including unemployed adults, often exhibit low levels of health literacy. Exploring subjectively perceived health literacy needs in these populations can serve as an entry point for participatory intervention development. We aimed to assess health literacy needs of unemployed adults by triangulating qualitative interviews data and a scoping review. Using a parallel approach, we combined results of an interview study with 10 participants of a job reintegration program in Germany and a scoping review. The interviews, conducted in early 2021, focused on health topics of interest to the participants and their sources of health related information. Data were analysed using thematic analysis. For the scoping review, we searched MEDLINE, CINAHL, PsycInfo and SCOPUS up to January 2021 for studies containing ‘‘unemployed’’ AND ‘‘health literacy’’ in titles/abstracts. Study selection and data extraction were done independently by two researchers. Nutrition and physical activity emerged as core themes during the interviews, with some participants referring to the importance of both for mental health. Doctors and the institution running the job reintegration program were the sources of health-related information often mentioned. The Corona pandemic was reported to have limited physical activity and affected psychosocial well-being. Five out of 2696 studies were included in the review. Four focused on mental health literacy, the fifth assessed information seeking practices in unemployed adults. The qualitative analysis revealed that health literacy needs of unemployed adults go beyond mental health literacy. As nutrition and physical activity impact mental health, interventions targeting such topics might also improve mental health literacy in unemployed adults. Study findings will be discussed with unemployed persons and social workers in coproduction workshops that aim to identify and prioritize health literacy needs for intervention development.
