Postexercise hydration is fundamental to replace fluid loss from sweat. This study evaluated rehydration and gastrointestinal (GI) symptoms for each of three beverages: water (W), sports drink (SD), and skimmed, lactose-free milk (SLM) after moderate-intensity cycling in the heat. Sixteen college students completed three exercise sessions each to lose ≈2% of their body mass. They drank 150% of body mass loss of the drink assigned in randomized order; net fluid balance, diuresis, and GI symptoms were measured and followed up for 3 hr after completion of fluid intake. SLM showed higher fluid retention (∼69%) versus W (∼40%; p <.001); SD (∼56%) was not different from SLM or W (p >.05). Net fluid balance was higher for SLM (−0.26 kg) and SD (−0.42 kg) than W (−0.67 kg) after 3 hr (p <.001), resulting from a significantly lower diuresis with SLM. Reported GI disturbances were mild and showed no difference among drinks (p >.05) despite ingestion of W (1,992 ± 425 ml), SD (1,999 ± 429 ml), and SLM (1,993 ± 426 ml) in 90 min. In conclusion, SLM was more effective than W for postexercise rehydration, showing greater fluid retention for the 3-hr follow-up and presenting with low-intensity GI symptoms similar to those with W and SD. A successful hexagonal Cu 2 S p-type semiconductor thin film using DC magnetron sputtering is reported. Films with thickness gradients were deposited by taking advantage of deposition geometry and target dimensions. X-ray diffraction (XRD) analysis confirmed the exclusive formation of the hexagonal Cu 2 S phase. Elemental composition and thickness dependence with the sample position were determined using energy-dispersive x-ray spectroscopy. Optical properties, including the optical bandgap, refractive index, and extinction coefficient, were assessed by modeling transmittance spectra. The Tauc–Lorentz oscillator and Drude models were employed for this purpose. XRD data analysis successfully determined the film thickness (t X R D ) as a function of the sample position, aligning well with thickness values (t T) derived from transmittance spectra analyses. These results were further supported by film thickness values (t S E M ) obtained from cross-sectional SEM images. Charge carrier density and mobility, extracted from the optical models, were found to be consistent with DC electrical measurements. AC impedance curves were effectively modeled with RL–RC parallel circuits. Pre-service teachers' mental models of the nature of soil were investigated in a sample of 181 students from four different Spanish universities, using three different methodological approaches: a phenomenographic analysis of definitions, a categorisation of labelled-drawings and the analysis of answers to a questionnaire consisting of both open- and closed-ended questions. Based on the phenomenographic analysis, four explanatory categories were defined: paedological (soil as a highly complex system); anthropocentric (soil from a utilitarian point of view); structural (soil as a layer of Earth) and naive view (soil as a surface of unknown composition and function). The most represented category in the studied sample was the structural one. Based on the questionnaire and the drawing analysis, students have some notions about soil composition, but their understating of its origin and degradation processes is scarce. No significant correlation was found between the analyses conducted using the three different instruments, thus indicating the need to use different approaches to better understand students' conceptions and their 'intermediate' epistemic models of soil. Introduction: Early diagnosis of skin cancer is crucial for improving prognosis. Teledermatology (TD) usage can optimize referrals and reduce waiting times. This study aims to evaluate waiting times at the critical referral nodes in teleinterconsultations that raised suspicion of skin malignancy in the Chilean TD platform of the public health care system. Materials and Methods: A cross-sectional observational study that analyzed asynchronous teleinterconsultations and raised suspicion for skin malignancy following the teledermatologist evaluation was uploaded on the Chilean Ministry of Health's TD platform from January 1 to June 30, 2022. Results: Out of 20,522 teleinterconsultations, 1,853 raised suspicion of skin cancer. Among them, 1,119 patients were assessed by in-person examination, while 669 were still on the waiting list. Response times averaged 3.98 days for TD diagnostic suggestions. Overall referral times averaged 75.98 days from initial teleinterconsultation to the final specialist in-person evaluation. Waiting times showed significant differences among health care services and geographic regions. Discussion: In resource-limited settings, TD serves as a valuable tool to optimize referrals and manage the demand for oncologic dermatological consultation. The long waiting times emphasize the need for targeted interventions, especially in regions with longer delays. Conclusion: While TD has shown to be an effective tool in optimizing referrals, waiting times still exceed international recommendations, even in urban centers. The considerable heterogeneity in referral times within health care services and geographic regions highlights the necessity of establishing standardized referral protocols and explicit deadlines to fulfill teleinterconsultations that raise suspicion of skin malignancy in the Chilean public system. Mastitis, an inflammatory condition affecting more than 25% of breastfeeding women, is usually associated with reduced milk secretion, pain, and discomfort, which often leads to early cessation of breastfeeding. Although the etiology of mastitis is multifactorial, a pro-inflammatory state of the mammary gland might be a risk factor. However, changes in milk composition, and specifically in the milk immune profile, prior to and during mastitis have not been well described. To help close this research gap, we documented the immune profiles of milk produced by both breasts of 10 women experiencing clinical (CM) and 8 women experiencing subclinical (SCM) mastitis during the week of sign/symptom development as well as the week prior and compared them with milk produced by 14 healthy controls. CM was defined as having signs/symptoms of mastitis, whereas SCM was presumed if the participant did not have signs/symptoms of CM, but her milk had a somatic cell count >400,000 cell/mL and/or sodium-to-potassium (Na/K) ratio >1.0. Concentration of 36 immune factors (including immunoglobulins, cytokines, chemokines, and growth factors) was quantified via immunoassays. Milk produced by women who developed CM had distinct immune profiles the week prior to diagnosis, particularly elevated concentrations of pro-inflammatory cytokine IL-1β and regulatory cytokines IL-2, IL-4 and IL-10. In contrast, immune profiles in milk produced by women with SCM did not differ from that produced by healthy women or those with CM the week prior to mastitis onset. Once mastitis appeared, marked changes in milk's immune profile were observed in both CM and SCM groups. CM was characterized by elevated concentrations of 27 compounds, including pro-inflammatory cytokines (IL-1β, IL-1ra, and TNFα) and chemokines (including IL-8, eotaxin, IP-10, MCP-1, MIP1α, and MIP1β), compared to healthy controls. Milk's immune profile during SCM was intermediate, showing higher levels of IL-6, IFNγ, and MCP-1 compared to healthy controls, suggesting a milder, more controlled immune response compared to CM. Only milk produced by the mastitis-affected breast had altered immune profiles. Nonlinear dynamic simulations of mechanical resonators have been facilitated by the advent of computational techniques that generate nonlinear reduced order models (ROMs) using the finite element (FE) method. However, designing devices with specific nonlinear characteristics remains inefficient since it requires manual adjustment of the design parameters and can result in suboptimal designs. Here, we integrate an FE-based nonlinear ROM technique with a derivative-free optimization algorithm to enable the design of nonlinear mechanical resonators. The resulting methodology is used to optimize the support design of high-stress nanomechanical Si3N4 string resonators, in the presence of conflicting objectives such as simultaneous enhancement of Q-factor and nonlinear Duffing constant. To that end, we generate Pareto frontiers that highlight the trade-offs between optimization objectives and validate the results both numerically and experimentally. To further demonstrate the capability of multi-objective optimization for practical design challenges, we simultaneously optimize the design of nanoresonators for three key figure-of-merits in resonant sensing: power consumption, sensitivity and response time. Background: Porcine reproductive and respiratory syndrome virus (PRRSV) is a major threat to swine industry worldwide, especially virulent variants arising during the last years, such as Spanish PRRSV-1 Rosalia strain. The role of the nasal microbiota in respiratory viral infections is still to be unveiled but may be promisingly related with the health status of the animals and thus, their susceptibility. The goal of this project was to study the nasal microbiota composition of piglets during a highly virulent PRRSV-1 outbreak comparing animals that died due to the infection with animals that survived it. The microbiota composition was inferred by V3–V4 regions of the 16S rRNA gene sequencing and bioinformatics analysis. To deepen the analysis, we added samples taken from piglets before the outbreak as well as from the sows giving birth to piglets under study. Results: Piglets that survived the PRRSV-1 outbreak reported a more diverse and different nasal microbiota at three weeks of age compared to piglets dying, which was highly related with the litter of origin and the sow of the piglets. In addition, a high abundance of classical swine nasal colonizers belonging to genera such as Bergeyella, Glaesserella, Neisseria and Moraxella (among others), was related with good outcome. On the other hand, a dysbiotic community dominated by Escherichia and a different clade of Moraxella was found in piglets with bad outcome. Moreover, samples taken before the outbreak showed similar dynamics prior to virulent PRRSV-1 arrival, suggesting that microbiota-related susceptibility was already occurring in the animals and that the increase in mortality seen was related to the new highly virulent strain. Conclusion: Our study suggests that the susceptibility to an infection such as PRRSV could be related to the nasal microbiota composition at the moment of infection and may serve as starting point to explore animal resilience. Since the dysbiosis detected as an initial response to infection may be not specific for this virus, further investigations should explore this phenomenon in the context of other viral infections. Diabetes mellitus is a metabolic syndrome that has grown globally to become a significant public health challenge. Hypothesizing that the plasma membrane protein, transient receptor potential ankyrin-1, is a pivotal target in insulin resistance, we investigated the mechanism of action of cinnamaldehyde (CIN), an electrophilic TRPA1 agonist, in skeletal muscle, a primary insulin target. Specifically, we evaluated the effect of CIN on insulin resistance, hepatic glycogen accumulation and muscle and adipose tissue glucose uptake. Furthermore, the in vitro role of CIN in glucose uptake and intracellular signaling was determined in insulin-resistant rats whose calcium influx was analyzed. Moreover, the serum lipid profile was assessed following short-term CIN treatment in rats, and lipid tolerance was analyzed. The effects of CIN on insulin resistance were mediated by TRPA1, with downstream signaling involving the activation of PI3-K, MAPK, PKC, as well as extracellular calcium and calcium release from intracellular stores. Additionally, cytoskeleton integrity was required for the complete action of CIN on glucose uptake in muscle. Skin cancer is one of the most common types of cancer worldwide, with exposure to UVB radiation being a significant risk factor for its development. To prevent skin cancer, continuous research efforts have focused on finding suitable photoprotective ingredients from natural sources that are also environmentally friendly. This study aimed to develop oil-in-water photoprotective nanoemulsions containing marine macroalgae extract. A Box–Behnken experimental design was used to identify the most promising formulation composition, resulting in optimal physical properties. These properties, including droplet size, polydispersity index (PDI), and zeta potential, were evaluated using dynamic light scattering (DLS). To assess the photoprotection capacity of the formulations, electrical impedance spectroscopy (EIS) was employed to evaluate alterations in the electrical characteristics of excised pig skin membranes placed in Franz cells equipped with a 4-electrode set-up. The final composition of the nanoemulsion was caprylic/capric triglycerides 4%, Macrogolglycerol ricinoleate 30%, and algae extract 1%. The nanoemulsions had an average droplet size of 128.5 ± 8.6 nm, a PDI of 0.25 ± 0.06, and a zeta potential of 45.14 ± 0.02 mV. Compared to the control group, the photoprotective capacity of the oil-in-water nanoemulsions was statistically significant. Specifically, only a 15% reduction in the skin membrane electrical resistance following UVB exposure was observed when the formulation containing algae extract was used, whereas a 50% reduction was observed for the vehicle. In conclusion, this work demonstrates that the developed nanoemulsions based on natural ingredients show promising protective capacity against UVB exposure of the skin. [ABSTRACT FROM AUTHOR]

Objective: In this cross‐sectional study, we aim to investigate the interactions between obesity, siesta behavior, and the genetic propensity for siesta in a Mediterranean population, in whom siesta is deeply rooted. Methods: We applied a previously generated Siesta‐Polygenic Score (PGS) in the ONTIME study (n = 1278). Siesta and other Mediterranean lifestyle behaviors were characterized using questionnaires. We further determined obesity grade. Secondarily, we measured weight loss during treatment as well as long‐term weight‐loss maintenance. Logistic regression analyses were performed to address our aim. Results: A total of 42.4% of the population usually took siesta. A significant genetic influence on siesta propensity was found, with a higher genetic predisposition linked to taking siesta more frequently (odds ratio [OR] = 1.17, 95% CI: 1.03–1.32; p = 0.015). Participants with a higher genetic propensity for siesta showed poorer dietary habits (p < 0.05). Among individuals with a high genetic propensity for siesta, we found that those who usually take siesta have lower odds of having obesity (p = 0.038) compared with those who do not. Similarly, in exploratory analysis, among individuals with a high genetic propensity for siesta, we found that those who usually take siesta have higher odds of weight‐loss success (p = 0.007) compared with those who do not. Conclusions: Considering the ongoing debate regarding whether siesta is beneficial or detrimental, our findings suggest that individual genetic predisposition to siesta might influence the association between siesta and health. The objectives of this study were to determine the rates of natural mortality (M), fishing mortality (F), total mortality (Z), the exploitation rates (E), as well as the biological reference points (BRPs) and the annual removal rates (R) of the crocodile shark, Pseudocarcharias kamoharai, in the Ecuadorian Pacific. Thirty similar and different models were applied to determine all these rates. These equations were obtained from studies on teleost and chondrichthyan fish. The biological parameters, including age, growth, longevity, and reproduction, were obtained from the specialized literature based on the biology of P. kamoharai in Ecuadorian waters. These biological parameters were used in all the models considered here. The M estimations were 0.14 to 0.28 based on six models for chondrichthyans and osteichthyes. These values were similar to the six algorithms designed for cartilaginous fish, ranging from 0.16 to 0.35; for this reason, these mortality rates were considered low. The Z values ranged from 0.08 to 0.51; however, they were not considered given that the three estimations were less than M, and only the Z = 0.51 was considered. Given that Z = 0.51 and M = 0.24, an F = 0.27 was obtained by subtraction, indicating a low mortality by fishing. E had values between 0.21 and 0.53, which indicated overexploitation that exceeded the Eopt = 0.50 value. The obtained BRPs were Fopt = 0.10 and 0.12 and Flim = 0.16, which showed that the optimal fishing levels (best possible capture) to achieve long-term sustainable exploitation of the stock encompass 10 to 16% of the fishing effort applied for this species. However, the F surpassed this prudential range. The annual removal percentage (R = 21%) demonstrated that 21% of the population was being removed. Based on the biology and ecology of this species, all models applied in this study showed that P. kamoharai had low natural and fishing mortality rates and moderate total mortality; its exploitation rate exceeded the fishing limits. These values and their life history traits indicated that this shark species cannot tolerate any fishing level without threatening its populations. [ABSTRACT FROM AUTHOR]

Introduction: Many cancer patients experience high levels of anxiety during chemotherapy, which can negatively impact their mental health and their physiological, emotional, and spiritual well-being. Different complementary therapies aim to attenuate these effects, including music therapy. Although there is preliminary evidence on the positive effects of music therapy and music-based interventions in chemotherapy wards, few studies report live group interventions delivered by accredited music therapists. Objective: To determine the effect of a single live group music therapy intervention on state anxiety and well-being levels of adult cancer patients during chemotherapy. Methodology: This study protocol follows the SPIRT guidelines and reports a two-arm multicenter randomized clinical trial (RCT). The intervention group will receive standard care + a live group music therapy session and the control group will receive standard care only. The primary outcome is state anxiety, measured with the six-item State-Trait Anxiety Inventory (STAI-6). The secondary outcome is well-being, measured with the Well-being Numerical Rating Scales (WB-NRSs). The scales will be applied before and after each intervention. Sample size calculation resulted in a total of 102 participants. Conclusions: This study seeks to contribute to the improvement of psycho-emotional health and well-being of cancer patients during chemotherapy. It is the first multi-center RCT on music therapy with cancer patients in [country, de-identified for peer review] and aims to gather knowledge about music's role to improve patients' mental health during acute treatment. This study aims to evaluated the impact of poly(ε-caprolactone) (PCL) microspheres on the pharmacokinetics and pharmacodynamics (PopPK/PD) of 6-methylcoumarin (6MC). For this, PCL microspheres loaded with 6MC were prepared using the emulsification-evaporation method. Particle size, zeta potential, drug loading, and entrapment efficiency were characterised by dynamic light scattering and UV spectrophotometry. In vitro release and pharmacokinetics in Wistar rats were assessed for free and encapsulated 6MC. Anti-inflammatory activity was evaluated using the carrageenan-induced paw edoema model, with PopPK and PopPK/PD models developed. Microspheres showed diameters between 2.9 and 7.1 µm, zeta potentials of −10 to −15 mV, and drug loading of 0.24 mg/mg. Encapsulation efficiency was 45.5% to 75.9%. PopPK models showed enhanced absorption and distribution, with increased anti-inflammatory potency of encapsulated 6MC. PCL microspheres significantly improved the pharmacokinetic and pharmacodynamic profiles of 6MC, enhancing its therapeutic potential for lipophilic drugs. [ABSTRACT FROM AUTHOR]

Dystrophin Dp71 is the major product of the Duchenne muscular dystrophy (DMD) gene in the brain, and its loss in DMD patients and mouse models leads to cognitive impairments. Dp71 is expressed as a range of proteins generated by alternative splicing of exons 71 to 74 and 78, classified in the main Dp71d and Dp71f groups that contain specific C-terminal ends. However, it is unknown whether each isoform has a specific role in distinct cell types, brain regions, and/or stages of brain development. In the present study, we characterized the expression of Dp71 isoforms during fetal (E10.5, E15.5) and postnatal (P1, P7, P14, P21 and P60) mouse and rat brain development. We finely quantified the expression of several Dp71 transcripts by RT-PCR and cloning assays in samples from whole-brain and distinct brain structures. The following Dp71 transcripts were detected: Dp71d, Dp71d∆71, Dp71d∆74, Dp71d∆71,74, Dp71d∆71−74, Dp71f, Dp71f∆71, Dp71f∆74, Dp71f∆71,74, and Dp71fΔ71−74. We found that the Dp71f isoform is the main transcript expressed at E10.5 (> 80%), while its expression is then progressively reduced and replaced by the expression of isoforms of the Dp71d group from E15.5 to postnatal and adult ages. This major finding was confirmed by third-generation nanopore sequencing. In addition, we found that the level of expression of specific Dp71 isoforms varies as a function of postnatal stages and brain structure. Our results suggest that Dp71 isoforms have different and complementary roles during embryonic and postnatal brain development, likely taking part in a variety of maturation processes in distinct cell types. Deforestation linked to agricultural activities is a major sustainability concern. Planning towards sustainable agricultural landscapes in the (sub-)tropics requires indicators that capture the many aspects of social-ecological cost and benefit of agriculture. Agricultural production strategies are developed using the best available data and knowledge such as high-yield locations, distance to storage facilities, or certification bonuses. However, there is often a divide between sustainable production data generated by the scientific community and current data that are of interest to actors, such as those in the agribusiness sector. Here, we describe how the harmonization of crop production, conservation, and social data used by scientists and agribusiness is possible using a participatory exercise based on knowledge co-production (i.e., generation of knowledge in a collaborative way) of socio-economic and environmental indicators (such as agricultural production, logistics, or the location of indigenous communities). This was made available through an online decision support platform that facilitated the generation of sustainable entrepreneurial strategies. We tested this exercise for the social-environmentally vulnerable Argentine Chaco dry forest, subject to some of the highest rates of deforestation globally, mainly due to soybean production. The cooperation between participants of this exercise built a knowledge exchange network that was key for informing decision-makers and highlighted information gaps including agricultural productivity, accessibility of regions, and the vulnerability of rural communities. Our exercise may be applicable to other agricultural commodity frontiers and showcases the value of including actors’ priorities in the design of indicators to ensure their policy impact and to achieve food systems’ sustainability. In Ecuador, more than 50% of the milk produced was informally marketed, consequently, the mean of this study was to assess the quality of this milk specifically within the province of Pichincha, the largest milk producer. A total of 650 samples were gathered from the eight cantons within the province. The parameters that exhibited the highest levels of non-compliance, of Ecuadorian legislation, were titratable acidity, accounting for 72.77% of the samples, and protein stability, accounting for 49.34%; these findings indicate a significant presence of bacterial contamination. Concerning antibiotic detection, a frightening 28.46% of the samples tested positive for residues exceeding the maximum limits established by the Codex Alimentarius. The findings indicate that a significant proportion of milk sold through informal channels in Pichincha province-Ecuador poses a substantial risk to public health. [ABSTRACT FROM AUTHOR]

Amphibian populations are undergoing a major recession worldwide, likely triggered by global change components such as the global warming and pollutants, among which agrochemicals, in general, and fertilizers, in particular, play a central role given their relevance in agriculture. Potential synergies among these stressors could maximize their individual effects. In this work, we investigated the consequences of a controlled chronic exposure to increased temperature and a sublethal dose of ammonium during the larval stage of Pelophylax perezi frogs on the growth, development, and locomotor performance of tadpoles and the metamorphs they gave rise to. To that end, tadpoles were reared either in heated or nonheated tanks, with or without ammonium added. The parents of these tadpoles came from either a pine grove or an agrosystem. Survival was reduced in agrosystem tadpoles reared with ammonium. Increased temperature potentiated tadpole growth while giving way to smaller metamorphs. Faster growth could be a consequence of increased metabolism, whereas the smaller size could follow an accelerated development and metamorphosis, which reduced the growth period. Also, swimming speed was greater in tadpoles reared in heated tanks, while jumping distance was greater in metamorphs reared in nonheated tanks. The effects of temperature were more marked in agrosystem than in pine grove individuals, which could mirror reduced adaptability. Intimate partner violence (IPV) is prevalent worldwide, including in Latinx populations. Reported rates of IPV in Latinx populations vary widely, indicating that measurement errors may be impeding researchers' and clinicians' understanding of IPV in these populations. We conducted a systematic review across a range of social science databases to evaluate psychometric properties and translation methodologies of Spanish-language IPV measures. Records were included if they included Spanish measures assessing IPV victimization. We identified 91 records with a total of 70 measures and evaluated the measures' extant psychometric evidence using the COnsensus-based Standards for the selection of health Measurement Instruments. For the measures translated from English to Spanish, we evaluated the translation methodology based on best-practice recommendations for achieving translations that are psychometrically equivalent to their original versions. We found that validation information about measures was sparse and that few translations adhered to best-practice recommendations. Based on our a priori criteria we recommend the Plazaola-Castaño translation of the Index of Spouse Abuse. In closing, we discuss the validity evidence of translated measures independent of the original language version and best-practice recommendations in translating psychological measures. T cell-based immunotherapies for solid tumors have not achieved the clinical success observed in hematological malignancies, partially due to the immunosuppressive effect promoted by the tumor microenvironment, where PD-L1 and TGF-β play a pivotal role. However, durable responses to immune checkpoint inhibitors remain limited to a minority of patients, while TGF-β inhibitors have not reached the market yet. Here, we describe a bispecific antibody for dual blockade of PD-L1 and TFG-β, termed AxF (scFv)2, under the premise that combination with T cell redirecting strategies would improve clinical benefit. The AxF (scFv)2 antibody was well expressed in mammalian and yeast cells, bound both targets and inhibited dose-dependently the corresponding signaling pathways in luminescence-based cellular reporter systems. Moreover, combined treatment with trispecific T-cell engagers (TriTE) or CAR-T cells significantly boosted T cell activation status and cytotoxic response in breast, lung and colorectal (CRC) cancer models. Importantly, the combination of an EpCAMxCD3×EGFR TriTE with the AxF (scFv)2 delayed CRC tumor growth in vivo and significantly enhanced survival compared to monotherapy with the trispecific antibody. In summary, we demonstrated the feasibility of concomitant blockade of PD-L1 and TGF-β by a single molecule, as well as its therapeutic potential in combination with different T cell redirecting agents to overcome tumor microenvironment-mediated immunosuppression. The objective of this randomized controlled trial was to examine if exercise training can counteract energy restriction‐induced impairment of mitochondrial capacity in skeletal muscle of 55–70‐years people with prediabetes and metabolic syndrome. The potential impact of sex was explored. Fifty sedentary men and women with prediabetes and metabolic syndrome (age: 61 ± 6 (±SD) years, BMI: 29.6 ± 4.7 kg·m−2, body fat content: 37.5% ± 8.2% and VO2max: 22.3 ± 5.7 mL·min−1·kg−1) were randomized to either exercise training and dietary advice (EX‐DI) or dietary advice only (DI). Dietary advice aimed to induce weight loss and improve glycemic control. Exercise consisted of 32 ± 2 mixed 30–60 min training sessions with recreational small‐sided soccer distributed across 16 weeks. Maximal activity and protein abundance of key regulatory mitochondrial enzymes were determined in m. vastus lateralis pre‐ and post‐intervention. Muscle glycogen content was also determined. Dietary advice only (DI), impaired (p < 0.001) citrate synthase (CS), and 3‐hydroxyacyl‐CoA dehydrogenase (HAD) maximal activity by 18% ± 43% and 23% ± 19%, respectively. When combined with exercise training, no dietary impairment of CS or HAD maximal activity was detectable. Superoxide dismutase 2 (SOD2) and CS protein expression also declined (p < 0.05) in DI and remained unchanged in EX‐DI. In terms of sex differences, a decrease in maximal CS activity in both EX‐DI and DI was observed exclusively in men (all p < 0.05). In conclusion, 16 weeks of exercise training counteracts energy restriction‐induced impairment in skeletal mitochondrial function in 55‐ to 70‐year‐old women and men with prediabetes and metabolic syndrome. This study was designed with the purpose of testing the psychometric properties of the Spanish version of the Attitudes toward Prostitution and Prostitutes Scale through three studies with different samples. The first one explores the test's dimensional structure or constructs validity through confirmatory factor analysis, as well as internal consistency and test-retest reliability. The second one focuses on discriminant and criteria validity. Finally, the third one examines the scale's convergent validity and its sensitivity to detecting changes. The results support two subscales with an optimal index of internal consistency, structural stability over time, and discriminative power between groups of participants. This study focuses on the physiological response of lettuce grown on Technosols designed for the remediation of soils polluted by potentially harmful elements (PHEs: As, Cd, Cu, Fe, Pb, and Zn). Lettuce plants were grown in five treatments: recovered (RS) and polluted soil (PS) as controls, and three Technosols (TO, TS, and TV) consisting of 60% PS mixed with 2% iron sludge, 20% marble sludge, and 18% organic wastes (TO: composted olive waste, TS: composted sewage sludge, and TV: vermicompost of garden waste). The main soil properties and PHE solubility were measured, together with physiological parameters related to phytotoxicity in lettuce such as growth, photosynthetic capacity, oxidative stress, and antioxidant defence. All Technosols improved unfavourable conditions of PS (i.e., neutralised acidity and enhanced OC content), leading to a significant decrease in Cd, Cu, and Zn mobility. Nevertheless, TV was the most effective as the reduction in PHEs mobility was higher. Furthermore, lettuce grown on TV and TO showed higher growth (+90% and +41%) than PS, while no increase in TS. However, lower oxidative stress and impact on photosynthetic rate occurred in all Technosols compared to PS (+344% TV, +157% TO, and +194% TS). This physiological response of lettuce proves that PHE phytotoxicity is reduced by Technosols. Thus, this ecotechnology constitutes a potential solution for soil remediation, with effectiveness of Technosols depending largely on its components. [ABSTRACT FROM AUTHOR]

Background and Purpose: Gastrointestinal tumours overexpress voltage‐gated calcium (CaV3) channels (CaV3.1, 3.2 and 3.3). CaV3 channels regulate cell growth and apoptosis colorectal cancer. Gossypol, a polyphenolic aldehyde found in the cotton plant, has anti‐tumour properties and inhibits CaV3 currents. A systematic study was performed on gossypol blocking mechanism on CaV3 channels and its potential anticancer effects in colon cancer cells, which express CaV3 isoforms. Experimental Approach: Transcripts for CaV3 proteins were analysed in gastrointestinal cancers using public repositories and in human colorectal cancer cell lines HCT116, SW480 and SW620. The gossypol blocking mechanism on CaV3 channels was investigated by combining heterologous expression systems and patch‐clamp experiments. The anti‐tumoural properties of gossypol were estimated by cell proliferation, viability and cell cycle assays. Ca2+ dynamics were evaluated with cytosolic and endoplasmic reticulum (ER) Ca2+ indicators. Key Results: High levels of CaV3 transcripts correlate with poor prognosis in gastrointestinal cancers. Gossypol blockade of CaV3 isoforms is concentration‐ and use‐dependent interacting with the closed, activated and inactivated conformations of CaV3 channels. Gossypol and CaV3 channels down‐regulation inhibit colorectal cancer cell proliferation by arresting cell cycles at the G0/G1 and G2/M phases, respectively. CaV3 channels underlie the vectorial Ca2+ uptake by endoplasmic reticulum in colorectal cancer cells. Conclusion and Implications: Gossypol differentially blocked CaV3 channel and its anticancer activity was correlated with high levels of CaV3.1 and CaV3.2 in colorectal cancer cells. Heat pumping through thermoelectric devices has many advantages over traditional cooling. However, their current efficiency is a limiting factor in their implementation. In this paper, we approach the non-convex topology optimization of thermoelectrical elements for cooling applications through the method of moving asymptotes (MMA) to improve their cooling capabilities per watt usage. The optimization problem is defined for a given power budget, aiming for the minimum temperature with a known heat pumping need. The introduction of power as a constraint justifies the introduction of the voltage gradient across the thermocouple as a design variable to maintain the thermoelectrical device in its optimum power-to-heat extraction ratio. To better understand the convergence of this non-convex problem, we present a two-variable analytical thermoelectric optimization model. This example provides information on how to select the penalty parameters used to scale the three material coefficients involved in the problem to obtain lower objective values and better convergence using MMA. The analytical model shows the non-convexity of the problem and provides the recommendation to use penalization coefficients of the form p k = p σ > p α = 1 for the thermal conductivity, electrical conductivity, and Seebeck coefficients. We tested these penalization coefficients through optimizations of a model based on the 1MC10-031 commercial thermoelectric-cooler (TEC) using the finite element method (FEM). These penalization coefficients provided local minima without the need for volume constraints. With this procedure, we found designs that provided temperatures close to 10 degrees lower using 60% less semiconductor material volume compared to the initial design. [ABSTRACT FROM AUTHOR]

In this paper, a self-created analogy is presented based on comparing the process of osmosis with a situation recreated in a train carriage. It was implemented with upper-secondary education students at a secondary school in Chiclana de la Frontera (Cádiz, Spain). To assess the progression of the students' models on osmosis, their activities during the implementation were used. Their perceptions of the analogy and the strategy used were analysed from their answers to a questionnaire using Likert-type and open questions. A qualitative and quantitative analysis was carried out. The results show that the analogy helps the initial models of osmosis advance towards more complex models. The students value the analogy positively. They explain it helped them develop scientific skills and attitudes like teamwork. It is concluded that the analogy designed is a valuable tool to bring students closer to the concept of osmosis, although it needs to be integrated into teaching-learning sequences. Galaxy properties are known to be affected by their environment. This is well established for the extremes of the density scales, between the high-density cluster environment and the low-density field. It is, however, not fully understood how the intermediate-density regime of cosmic web filaments affects galaxy evolution. We investigate this environmental effect using a mass complete sample of 23 441 galaxies in the Sloan Digital Sky Survey DR8 Main Galaxy Sample (⁠|${M}_{\text{Stellar}} \gt 10^{9.91} \text{M}_{\odot }$|⁠). We define six environments, probing different density regimes and representing unique stages in the structure formation process, comparing the differences in star formation activity and morphology between them. We find that galaxies in filaments tend to be less star-forming and favour more early-type morphologies than those in the field. These differences persist when considering stellar mass-matched samples, suggesting that this is a consequence of the environment. We further investigate whether these trends are a result of the large-scale or local environment through constructing samples matched both in stellar mass and local galaxy density. We find that when also matching in local galaxy density, the differences observed between the filament and field population vanishes, concluding that the environmental effect of filaments can be entirely parametrized by a local galaxy density index. We find that differences can still be seen in comparisons with the interiors of clusters, suggesting these are unique environments which can impart additional physical processes not characterized by local galaxy density. Purpose: Uveitis can lead to secondary glaucoma, a condition with challenging management that can carry irreversible visual loss. Filtering surgery has demonstrated a higher failure rate, increased incidence of postoperative complications and reinterventions in uveitic patients. There is no consensus on the optimal surgical approach for uveitic glaucoma (UG) due to limited data comparing various intraocular pressure (IOP)-lowering surgeries. This retrospective cohort aims to assess the clinical outcomes of trabeculectomy (TBT), non-penetrating deep sclerectomy (NPDS), Ex-PRESS shunt and Ahmed glaucoma valve, providing additional insights into the long-term IOP control and safety of filtering surgeries in UG. Methods: The filtering surgery was performed on 32 eyes of 27 UG patients. Complete success was defined as IOP ≤ 18 mmHg or a 30% reduction. Qualified success allowed topical hypotensive treatment. Results: Complete success was 40.63% (13/32) at 12 months and 36.67% (11/30) at 36 months. Qualified success was 84.38% (27/32) at 12 months and 63.33% (19/30) at 36 months. In the survival analysis, both NPDS and Ex-PRESS demonstrated decreased failure rates compared to TBT (NPDS vs TBT: HR = 0.20, p = 0.049; Ex-PRESS vs TBT: HR = 0.28, p = 0.13). One or more reinterventions were required in 34.38% (11/32) of the eyes. NPDS had the lowest incidence of hypotony. Secondary cataract was a common complication in all groups. Conclusion: Various filtering surgeries are safe and effective procedures for lowering IOP and reducing the requirement of topical antihypertensives in UG at 36 months. In eukaryotic cells, aerobic energy is produced by mitochondria through oxygen uptake. However, little is known about the early mitochondrial responses to moderate hypobaric hypoxia (MHH) in highly metabolic active tissues. Here, we describe the mitochondrial responses to acute MHH in the heart and skeletal muscle. Rats were randomly allocated into a normoxia control group (n = 10) and a hypoxia group (n = 30), divided into three groups (0, 6, and 24 h post‐MHH). The normoxia situation was recapitulated at the University of Granada, at 662 m above sea level. The MHH situation was performed at the High‐Performance Altitude Training Centre of Sierra Nevada located in Granada at 2320 m above sea level. We found a significant increase in mitochondrial supercomplex assembly in the heart as soon as the animals reached 2320 m above sea level and their levels are maintained 24 h post‐exposure, but not in skeletal muscle. Furthermore, in skeletal muscle, at 0 and 6 h, there was increased dynamin‐related protein 1 (Drp1) expression and a significant reduction in Mitofusin 2. In conclusion, mitochondria from the muscle and heart respond differently to MHH: mitochondrial supercomplexes increase in the heart, whereas, in skeletal muscle, the mitochondrial pro‐fission response is trigged. Considering that skeletal muscle was not actively involved in the ascent when the heart was beating faster to compensate for the hypobaric, hypoxic conditions, we speculate that the different responses to MHH are a result of the different energetic requirements of the tissues upon MHH. Background: Manual therapy (MT) and occlusal splint therapy (OST) are the most conservative therapies applied on patients with temporomandibular disorders (TMDs). The aim was to compare the efficacy of MT vs. OST in improving pain, maximal mouth opening (MMO), disability, and health related-quality of life (hr-QoL) in these patients. Methods: According to PRISMA guidelines, a meta-analysis (CRD42022343915) was conducted including randomized controlled trials comparing the effectiveness of MT vs. OST in TMD patients, after searching in PubMed, PEDro, SCOPUS, and WOS up to March 2024. Methodological quality and risk of bias were assessed using the PEDro Scale. Cohen's standardized mean difference (SMD) and its 95% confidence interval (95% CI) were the pooled effect measures calculated. Results: Nine studies, providing data from 426 patients, were included. Meta-analyses revealed that MT is more effective than OST in reducing disability (SMD = −0.81; 95% CI −1.1 to −0.54) and increasing MMO (SMD = 0.52; 95% CI 0.27 to 0.76) without differences for improving pain intensity and hr-QoL. Subgroup analyses revealed the major efficacy of OST in reducing pain in myogenic patients (SMD = 0.65; 95% CI 0.02 to 1.28). Utilizing industrial and urban solid wastes is crucial for developing low-carbon cements. This study proposes a novel method to integrate recycled glass and blast furnace slag (BFS) into alkali-activated cements. It also investigates the influence of pulverized limestone (LSP), as a readily available cost-efficient partial replacement for BFS. The activators were alternative solid sodium silicates (SSWG), prepared by thermochemical treatment of soda lime silica waste glass in NaOH solutions. SSWG had moduli (Ms) SiO2/Na2O = 1 and 1.5 and were added at 6 and 8%wt. Na2O relative to the mass of BFS + LSP. After 3 years, pastes of 100% BFS at 20 °C yielded the highest compressive strength of 78 MPa, while incorporating 50–75% LSP resulted in 61–42 MPa, depending on the Ms and %Na2O. Microstructural analysis via XRD, thermal analysis, SEM, and 27Al and 29Si NMR, indicated the formation of cementitious products like C–S–H, C–(A)–S–H, hydrotalcite, natron, gaylussite and pyrssonite. LSP increased the chain length in C–(A)–S–H-type products and affected the Al uptake by a seeding effect. The eco-efficiency analysis showed that the studied cements emit 45–74% less kgCO2.eq per ton than Portland cement. [ABSTRACT FROM AUTHOR]

Context: Agriculture relies on irrigation in many parts of the world, and the need for irrigation is increasing due to rising demands for agricultural products and climate change-induced alterations in rainfall patterns. However, irrigated agriculture has been found to damage ecosystems and threaten landscape sustainability. Objectives: Against this background, there has been a recent development towards large-scale irrigation in Spain. The aim of this study is to understand this development at the landscape level and its impact in the context of landscape sustainability. Methods: We focused on two study sites in Castilla y León using a mixed-methods approach. We studied driving forces, landscape changes, and sustainability outcomes through document analysis, interviews, and aerial photograph analysis. Results: The development of a landscape-level underground pipe network took place at one study site and is planned for the second study site. Interviewees perceived institutional and social driving forces as particularly influential and technological driving forces as less influential. Political and economic driving forces were often interlinked. The irrigation development tied to land consolidation led to landscape changes, such as the removal of trees and increases in field size. Thus, in terms of environmental sustainability, trade-offs were found, while social sustainability outcomes were mainly positive. The impact on farmers’ economic security varied. Conclusions: For further landscape-level irrigation developments, we recommend integrating preserving seminatural habitats and the structural diversity of the agricultural landscape in planning processes. We also recommend a shift towards more water efficient crops, evapotranspiration management, and a new funding scheme for farmers to offset rising electricity costs. [ABSTRACT FROM AUTHOR]

Adolescence is a time of elation and sublime emotions which are also reflected in partner relationships, within a context characterised by materialism and social structures in which gender relations are affected by the domination/subordination model. This paper analyses the affective-sexual relationships of adolescents as a place of violence, power relations, domination and submission. It also studies the importance of social networks in their way of interacting and learning. The methodology used was, on the one hand, the review of bibliographic material regarding romance, partner relationships, gender violence and using of networks, and, on the other hand, the analysis of the results obtained from two studies carried out by the University of Granada, with quantitative and qualitative information on adolescents in Andalusia. The results of this research indicate that relationships are permeated by the idealisation of romantic love and by stereotypes leading to practices of control and possession. [ABSTRACT FROM AUTHOR]

Dichloromethane, as a readily available and inexpensive C1 synthon is proposed as a powerful building block for cyclopropanation of alkenes under mild conditions. Herein, we report a highly efficient and versatile dual photoredox system, involving a nickel aminopyridine coordination complex and a photocatalyst, for the cyclopropanation of aromatic olefins using dichloromethane, under visible‐light irradiation. The cyclopropanation protocol has been successfully applied at gram scale. Mechanistic studies suggest a Ni(II) pyridyl radical complex as the key intermediate for the homolytic cleavage of the Csp3−Cl bond, generating a chloromethyl radical that is captured by the olefin coupling partner. Our findings also highlight the versatility of this methodology. By directing the radical/polar crossover process, we were able to selectively drive the reaction towards either the formation of cyclopropyl derivatives or the corresponding non‐cyclic alkyl chloride products. The methodology also successfully apply to geminal dichloroalkanes, including the formation of spiro[2,2] compounds. Moreover, our methodology extends to the synthesis of deuterium‐labelled cyclopropanes, demonstrating its utility in isotopic labelling and broadening its applicability in chemical synthesis and drug development. Biopolymers derived from biomass can provide the advantages of both biodegradability and functional qualities from a circular economy point of view, where waste is transformed into raw material. In particular, avocado seeds can be considered an interesting residue for biobased packaging applications due to their high starch content. In this work, avocado seed starch (ASS)-based films containing different glycerol concentrations were prepared by solvent casting. Films were also reinforced with starch nanocrystals (SNCs) obtained through the acid hydrolysis of ASS. The characterization of the extracted starch and starch nanocrystals by scanning electron microscopy, X-ray diffraction, and thermogravimetric analysis has been reported. Adding 1% of SNCs increased elastic modulus by 112% and decreased water vapor permeability by 30% with respect to neat matrix. Interestingly, the bioactive compounds from the avocado seed provided the films with high antioxidant capacity. Moreover, considering the long time required for traditional plastic packaging to degrade, all of the ASS-based films disintegrated within 48 h under lab-scale composting conditions. The results of this work support the valorization of food waste byproducts and the development of reinforced biodegradable materials for potential use as active food packaging. Myeloproliferative neoplasms are stem cell–driven cancers associated with a large burden of morbidity and mortality. Most patients present with early-stage disease, but a substantial proportion progress to myelofibrosis or secondary leukemia, advanced cancers with a poor prognosis and high symptom burden. Currently, it remains difficult to predict progression, and therapies that reliably prevent or reverse fibrosis are lacking. A major bottleneck to the discovery of disease-modifying therapies has been an incomplete understanding of the interplay between perturbed cellular and molecular states. Several cell types have individually been implicated, but a comprehensive analysis of myelofibrotic bone marrow is lacking. We therefore mapped the cross-talk between bone marrow cell types in myelofibrotic bone marrow. We found that inflammation and fibrosis are orchestrated by a "quartet" of immune and stromal cell lineages, with basophils and mast cells creating a TNF signaling hub, communicating with megakaryocytes, mesenchymal stromal cells, and proinflammatory fibroblasts. We identified the β-galactoside–binding protein galectin-1 as a biomarker of progression to myelofibrosis and poor survival in multiple patient cohorts and as a promising therapeutic target, with reduced myeloproliferation and fibrosis in vitro and in vivo and improved survival after galectin-1 inhibition. In human bone marrow organoids, TNF increased galectin-1 expression, suggesting a feedback loop wherein the proinflammatory myeloproliferative neoplasm clone creates a self-reinforcing niche, fueling progression to advanced disease. This study provides a resource for studying hematopoietic cell–niche interactions, with relevance for cancer-associated inflammation and disorders of tissue fibrosis. Li, Colombo, and Wang et al. present a mouse cancer-stroma interactome for myelofibrosis and show that galectin-1 is a potential inflammation-related target for treating disease. Mesenchymal stem cells, eosinophils, basophils, and mast cells were each implicated in an inflammatory niche that drove myelofibrosis progression. Inhibiting galectin-1 with a monoclonal antibody in a mouse model of myeloproliferative neoplasm decreased fibrosis and cell proliferation and had a beneficial effect on survival. Galectin-1 was associated with myelofibrosis and poor survival in multiple human cohorts. Background/Objectives: An extract of calyces from Physalis peruviana with hypoglycemic activity is being considered as a potential herbal medicine. Preclinical pharmacokinetics (PK) studies of the extract in rats, focusing on plasma concentrations of its main compound, rutin, and its metabolites, revealed PK interactions in the extract matrix that improved the absorption of rutin metabolites compared to the pure compound, among other PK effects. This research aimed to study the PK of rutin alone and in the extract and assess potential PK interactions in the extract matrix on the flavonoid and its metabolites in rabbits, a nonrodent species; Methods: Animals received pure rutin or extract orally and intravenously. The PK analysis used noncompartmental and population pharmacokinetics (popPK) methods, and simple allometry was applied to predict human PK parameters; Results: The rutin concentration–time profile fit a two-compartment model with first-order elimination, while its metabolites fit a double first-order absorption model. The extract matrix led to increased absorption, distribution, and elimination of rutin as well as increased bioavailability of its metabolites in rabbits; Conclusions: The popPK model defined the equations for PK parameters describing these findings, and the increased volume of distribution and clearance of rutin was maintained in human predictions.
