In this paper, three different Zn(II) complexes with (E)-2-(2-(1-(6-bromopyridin-2-yl)ethylidene)hydrazinyl)-N,N,N-trimethyl-2-oxoethan-1-aminium chloride (HLCl) have been synthesized and characterized by single crystal X-ray diffraction, elemental analysis, IR and NMR spectroscopy. All complexes are mononuclear, with the ligand (L) coordinated in a deprotonated formally neutral zwitterionic form via NNO donor set atoms. Complex 1 forms an octahedral geometry with the composition [ZnL2](BF4)2, while complexes 2 [ZnL(NCO)2] and 3 [ZnL(N3)2] form penta-coordinated geometry. Density functional theory (DFT) calculations were performed to enhance our understanding of the structures of the synthesized complexes and the cytotoxic activity of the complexes was tested against five human cancer cell lines (HeLa, A549, MDA-MB-231, K562, LS 174T) and normal human fibroblasts MRC-5. Additionally, antibacterial and antifungal activity of these complexes was tested against a panel of Gram-negative and Gram-positive bacteria, two fungal strains, and a yeast strain. It is noteworthy that all three complexes show selective antifungal activity comparable to that of amphotericin B. Molecular docking analysis predicted that geranylgeranyl pyrophosphate synthase, an enzyme essential for sterol biosynthesis, is the most likely target for inhibition by the tested complexes. Anaerobic fermentation converts organic waste into valuable products like volatile fatty acids (VFA). However, this process has weaknesses, such as delayed mass transfer, a slow microbe growth rate, poor hydrolysis, and surface heterogeneity. Such issues can be addressed by adding nanomaterials because they have unique physicochemical properties, such as a large surface area, many active sites, a high level of reactivity, a high level of specificity, the ability to self-assemble, and better mobility. This review focuses on the current efforts to investigate the effects of nanomaterials on improving VFA production. In addition, this work includes bibliometric information to aid in understanding current trends in this field of study. Furthermore, this work also reviewed the cytotoxicity and ecotoxicity of nanoparticles, followed by the applications and downstream processing of VFA. The liver is a crucial organ in the regulation of metabolism, signaling, and homeostasis. Using recent advanced sequencing technologies, several mutations of genes in major metabolic and signaling pathways have been discovered in the pathogenesis of hepatocellular carcinoma (HCC). These gene signatures alter expression and ultimately affect biochemical pathways by modifying enzyme/protein levels, resulting in numerous clinical outcomes related to HCC. It comes with varying forms of genetic and biochemical alterations, associated with carbohydrate, lipid, nucleic acid, and amino acid metabolism, as well as signaling pathways linked to tumorigenesis. Here, we aim to summarize the main components and mechanisms involved in the progression of HCC with a special focus on the metabolic regulation of key effectors of tumorigenesis, through the crosstalk between genetics and biochemistry. This paper provides an overview of hepatocellular carcinoma, underlying the fundamental effect of gene variations on metabolic and signaling pathways. Since there is still an unmet need for biomarkers and novel therapeutic targets, some of these signature genes or proteins can be used as novel biomarkers for diagnosis, prognosis, and novel potential therapeutic targets for the treatment of HCC. The solubility of pigments is of significant importance for the successful coloring of various products in the food industry. In this work, the solubility of anthocyanins and chlorophylls was analyzed by thin-layer chromatography. Plant pigments were applied to filter paper and placed in mixtures of hydrophilic, ethanol, methanol, hydrophobic solvents, heptane, chloroform, and petroleum ether. In addition, solutions of different pH were used to examine how pH changes the color of the plant pigments. By adding small amounts of a strong acid (hydrochloric acid), a strong base (sodium hydroxide), weak acids (citric acid and acetic acid), and salts (sodium hydrogen carbonate and sodium hypochlorite) to the pigment extracts, a palette of various colors in the pigments was observed. Based on the solubility data and the results of the color change in the pigments, anthocyanins were successfully used as a food color in a food product with acidic pH, lemonade, and a food product with basic pH, cookies. Recently, as doctors and researchers investigate into to the possible benefits of daily diets, ketogenic diets (KD) have started to gain popularity. As the growing up popularity of KD in social media, researchers had the chance to show their academic studies to the public more easily. Therefore, in this study, KD is discussed in terms of the recently popular “altmetric analysis” by scientists. The aim of study is to highlight the role of the ketogenic diet, figure out the mechanism of ketone bodies and the pivotal role of altmetric studies in health sciences. In order to investigate the novel aspects of the top 100 (T100) most cited scientific research paper in academia, 100 most discussed research paper was selected on social media platforms about KD from 1992 to 2019 and it was compared in terms of bibliometrics in web of science and in terms of social media impact in altmetric.com. While the total citations of the top 100 articles ranged from 143–109, altmetric values of T100 are 0–1269. According to the top social media platforms where the articles are mentioned, Twitter, Facebook, and Patent are the most popular social media platforms, respectively. It was not always coherent how altmetric data related to traditional bibliometric analysis. Thus, it might be suggested that bibliometric and altmetric assessments be viewed as complementary to one another. Biochemical sensors have important applications in biology, chemistry, and medicine. Nevertheless, many biochemical sensors are hampered by intricate techniques, cumbersome procedures, and the need for labeling. In the past two decades, it has been discovered that liquid crystals can be used to achieve the optical amplification of biological interactions. By modifying recognition molecules, a variety of label-free biochemical sensors can be created. Consequently, biochemical sensors based on the amplification of liquid crystals have become one of the most promising sensors. This paper describes in detail the optical sensing principle of liquid crystals, sensing devices, and optical detection technologies. Meanwhile, the latest research findings are elucidated. To analyze the research status and development trend of ionic liquids toxicology, VOSviewer and HistCite software were used to visually analyze the countries, institutions, authors, keywords, etc. of 1 178 ionic liquids toxicology related to literatures in the Web of Science core database from 2000 to 2021. The results show that: the number of literatures in the field of ionic liquids toxicology is increasing year by year, and China is far ahead of other countries in both the number of papers published and the total citation score. At present, the paper is mainly based on team cooperation within institutions, with strong independence. Although Chinese institutions and authors rank higher in the number of papers published, their total local citation score is not outstanding. The main focus of research in ionic liquids toxicology is imidazolium. The research focuses on the evaluation of lethality and composition changes in organisms, and the construction of structure-activity relationship models between ionic liquids and organisms. The ability to study chemical dynamics on ultrafast timescales has greatly advanced with the introduction of X-ray free electron lasers (XFELs) providing short pulses of intense X-rays tailored to probe atomic structure and electronic configuration. Fully exploiting the full potential of XFELs requires specialized experimental endstations along with the development of techniques and methods to successfully carry out experiments. The liquid jet endstation (LJE) at the Linac Coherent Light Source (LCLS) has been developed to study photochemistry and biochemistry in solution systems using a combination of X-ray solution scattering (XSS), X-ray absorption spectroscopy (XAS), and X-ray emission spectroscopy (XES). The pump–probe setup utilizes an optical laser to excite the sample, which is subsequently probed by a hard X-ray pulse to resolve structural and electronic dynamics at their intrinsic femtosecond timescales. The LJE ensures reliable sample delivery to the X-ray interaction point via various liquid jets, enabling rapid replenishment of thin samples with millimolar concentrations and low sample volumes at the 120 Hz repetition rate of the LCLS beam. This paper provides a detailed description of the LJE design and of the techniques it enables, with an emphasis on the diagnostics required for real-time monitoring of the liquid jet and on the spatiotemporal overlap methods used to optimize the signal. Enzymes are protein molecules that play a crucial role in various biological processes in living organisms. They function as catalysts in biological reactions such as digestion, metabolism, DNA replication and other physiological processes. Furthermore, enzymes are widely used in food production, pharmaceuticals and biofuel production. In these industries, they accelerate desired chemical reactions as biocatalysts. Therefore, applying computational methods and data-driven algorithms to predict enzyme properties is essential. Over the past decade, deep learning has made remarkable advancements in science and technology. Deep learning is a subset of machine learning algorithms that rely on artificial neural networks. These algorithms can be employed for supervised, semi-supervised and unsupervised learning. Here, to provide an update on the current literature, we provide an overview of various deep learning algorithms and recent advancements in their application to enzyme science. These applications can generally be categorized into diverse subjects: function prediction, enzyme kinetic parameters prediction, enzyme-substrate identification, condition optimization, thermophilic property prediction, enzyme catalytic site prediction and enzyme design. In conclusion, we discuss the convergence of enzyme science and deep learning, highlighting the potential opportunities and challenges. The COVID‐19 pandemic has forced a shift in thinking regarding the safe delivery of wet laboratory courses. While we were fortunate to have the capacity to continue delivering wet laboratory experiments with physical distancing and other measures in place, modifications to the mechanisms of delivery within courses were necessary to minimize risk to students and teaching staff. One such modification was introduced in BCH370H, an introductory biochemistry laboratory course, where a OneNote Class Notebook (ONCN) was used as an electronic laboratory notebook (ELN) in place of the traditional hardbound paper laboratory notebook (PLN) used prior to the pandemic. The initial reasoning for switching to an ELN was around safety—allowing course staff and students to maintain physical distancing whenever possible and eliminating the need for teaching assistants to handle student notebooks; however, the benefits of the ONCN proved to be significantly more. OneNote acted not only as a place for students to record notes but the Class Notebook's unique features allowed easy integration of other important aspects of the course, including delivery of laboratory manuals, posting of student results, notetaking feedback, sharing of instructional materials with teaching assistants, and more. Josep Rizo is a Professor of Biophysics, Biochemistry and Pharmacology at the University of Texas Southwestern Medical Center, where he is Virginia Lazenby O'Hara Chair in Biochemistry. We describe herein a method of recharging used commercial spin columns or assembling homemade spin columns using filter paper as binding material for cost-effective, low throughput nucleic acid purification. The efficiency of filter paper-based spin columns was evaluated for purification of nucleic acids from various sources. Following protocols of commercial kits, we found filter paper to be a useful binding material for purification of nucleic acids, including plant genomic DNA, plant total RNA, PCR products, and DNA from agarose gels. However, filter paper has a weak binding affinity to plasmid DNA in tested miniprep protocols. Protocols for the use of filter paper recharged spin columns or homemade spin columns for low throughput purification of plant genomic DNA and total RNA with unused commercial kit buffers or less expensive homemade buffers are presented. The coronavirus disease 2019 (COVID‐19) pandemic is a case that can be used as a biochemistry learning resource in relation to the topic of viruses. Learning to use cases will be effective if students are assigned to produce reports, articles, or other scientific papers. Self‐assessment can be used to assess the competencies that have been learnt in the writing process. This study aims to analyze the scientific papers and self‐assessment of students using the COVID‐19 pandemic case. It includes qualitative research with scientific paper and self‐assessment questionnaire analyses. The research sample was chemistry education study program students (N = 57) following biochemistry course, out of a population of 87 students. The results show that the students have a good understanding of explaining COVID‐19. Self‐assessment analysis shows that 80% of the students responded positively to eight statements; those who gave positive responses to each questionnaire statement were able to produce scientific papers. The emergence of ChatGPT as one of the most advanced chatbots and its ability to generate diverse data has given room for numerous discussions worldwide regarding its utility, particularly in advancing medical education and research. This study seeks to assess the performance of ChatGPT in medical biochemistry to evaluate its potential as an effective self‐learning tool for medical students. This evaluation was carried out using the university examination question papers of both parts 1 and 2 of medical biochemistry which comprised theory and multiple choice questions (MCQs) accounting for a total of 100 in each part. The questions were used to interact with ChatGPT, and three raters independently reviewed and scored the answers to prevent bias in scoring. We conducted the inter‐item correlation matrix and the interclass correlation between raters 1, 2, and 3. For MCQs, symmetric measures in the form of kappa value (a measure of agreement) were performed between raters 1, 2, and 3. ChatGPT generated relevant and appropriate answers to all questions along with explanations for MCQs. ChatGPT has "passed" the medical biochemistry university examination with an average score of 117 out of 200 (58%) in both papers. In Paper 1, ChatGPT has secured 60 ± 2.29 and 57 ± 4.36 in Paper 2. The kappa value for all the cross‐analysis of Rater 1, Rater 2, and Rater 3 scores in MCQ was 1.000. The evaluation of ChatGPT as a self‐learning tool in medical biochemistry has yielded important insights. While it is encouraging that ChatGPT has demonstrated proficiency in this area, the overall score of 58% indicates that there is work to be done. Legumes being a major source of protein have a wide range of economic importance. But the major constraints in growing legumes are poor germination, seedling emergence, non uniform stand establishment and crop mortality leading to lower pulse productivity. Thus, pre-sowing seed treatments are critical parameters which attribute to efficient plant growth and high yield. Uniform seed germination, seedling growth and uniform establishment are the critical stages during the growth of crops. Seed priming is a very promising presowing treatment employed to improve seed germination, better crop establishment, enhance the seed quality and induces tolerance against biotic and abiotic stresses in plants thereby providing a balanced healthy diet to the citizens. Priming is a cost effective and feasible strategy involving controlled hydration of seeds followed by drying to stimulate the pre-germinative metabolic activities to occur promoting rapid germination of seedlings, break dormancy and enhance crop yield. Enzymatic, metabolic and biochemical processes of the primed seeds attributes to rapid and uniform seedling emergence. Seed priming methods comprises of conventional methods including hydro priming, osmopriming, biopriming, chemical priming and the advanced methods including nano priming and priming with physical agents. Simple Summary: Penile cancer is a rare and aggressive disease. Current treatment options when the cancer is locally advanced are suboptimal and potentially mutilating. Insight into the immune dysregulation necessary for the emergence of penile cancer could suggest innovative ways to manipulate the immune system which have already demonstrated efficacy in other, more common malignancies. In this paper, we use multiplex immunofluorescence, a novel technology, to investigate the immune microenvironment of penile cancer for the first time. We describe a pattern of immune exhaustion as cancer becomes more advanced and identify tumor-associated macrophages as a potential key player in regulating this process. Penile squamous cell carcinoma (PSCC) is a rare and deadly malignancy. Therapeutic advances have been stifled by a poor understanding of disease biology. Specifically, the immune microenvironment is an underexplored component in PSCC and the activity of immune checkpoint inhibitors observed in a subset of patients suggests immune escape may play an important role in tumorigenesis. Herein, we explored for the first time the immune microenvironment of 57 men with PSCC and how it varies with the presence of human papillomavirus (HPV) infection and across tumor stages using multiplex immunofluorescence of key immune cell markers. We observed an increase in the density of immune effector cells in node-negative tumors and a progressive rise in inhibitory immune players such as type 2 macrophages and upregulation of the PD-L1 checkpoint in men with N1 and N2-3 disease. Fetal alcohol spectrum disorder (FASD) is commonly misdiagnosed because of the complexity of presentation and multiple diagnostic criteria. FASD includes four categorical entities (fetal alcohol syndrome, partial fetal alcohol syndrome, alcohol related neurodevelopmental disorder, and alcohol related birth defects). The four FASD diagnostic criteria are facial dysmorphology, growth deficiency, central nervous system dysfunction, and prenatal alcohol exposure. Sensory processing disorders (SPDs) are common in FASD and are observed as inappropriate behavioral responses to environmental stimuli. These can be either a sensory-based motor disorder, sensory discrimination disorder, or sensory modulation disorder. A child with SPD may experience challenges with their fine motor coordination, gross motor coordination, organizational challenges, or behavioral regulation impairments. FASD requires a multidimensional approach to intervention. Although FASD cannot be cured, symptoms can be managed with sleep-based therapies, sensory integration, and cognitive therapies. This paper reviews SPDs in FASD and the interventions that can be used by practitioners to help improve their therapeutic management, although it is unlikely that any single intervention will be the right choice for all patients. The combination of chemotherapy and photothermal therapy shows great potential in cancer treatment, and the carbon nanomaterials have been attracted great attention in biomedical technology. However, it is still a great challenge to design stimuli‐responsive nano‐carbon‐based drug release systems with integrated functions. In this paper, rod‐like carbon nanomaterials (RCNs) were prepared by the soft template hydrothermal method with glucose as raw materials, which were proved to have high photothermal efficiency (∼23.7%), as well as good biocompatibility and excellent drug‐loading capacity. After that, RCNs were used to load doxorubicin (DOX) as DOX@RCNs for integrated photothermal/chemotherapy towards cancer, which demonstrated 52.2% higher cancer cell killing efficiency than that of RCNs under near infrared (NIR) irradiation in vitro. The authors' approach provided a novel NIR‐responsive nano platform for combined photothermal/chemotherapy towards cancer, which was considered to be of great potential in anticancer applications. Purpose: To investigate cytokine levels in peri-implant crevicular fluid and thus evaluate the effects of concentrated growth factor (CGF) on osseointegration. Materials and Methods: A total of 40 mandibular implants were symmetrically placed in a group of 20 systemically healthy patients enrolled in the study. In each patient, one implant wetted with liquid infiltrated from fibrin matrix was placed in the test side (Group L), and the other implant was placed in the control side without the application of any material (Group C). Peri-implant crevicular fluid was collected at 2, 4, and 12 weeks later. Marginal bone loss was measured with panoramic radiographs taken immediately after implant placement and at 12 weeks. Resonance frequency analysis (RFA) of the implants was performed intraoperatively and at 4 and 12 weeks. Results: Stability values of the implants in the CGF liquid–treated sites were higher than those of the control group at week 12 (P = .005). There was no statistically significant difference between the two groups in terms of marginal bone loss (MBL). Group L showed increased levels of tumor necrosis factor alpha (TNF-α) and receptor activator of nuclear factor kappa-B ligand (RANKL) at 2 and 4 weeks. Also, levels of osteoprotegerin (OPG) were higher in Group L at week 4 compared to Group C (P = .033). Conclusions: The increased TNF-α, RANKL, and OPG levels in this study demonstrate that CGF liquid can be used to accelerate peri-implant bone remodeling in the early phase of osseointegration. Effluent found in the pulp and paper industry can cause considerable damage if it is discharged untreated, because of the high biochemical and chemical oxygen demands. Electrocoagulation is a physicochemical process widely used in industrial wastewater treatment. The removal of different pollutants depends on the sample type and operating conditions. The aim of this research was to evaluate the efficiency of an electrocoagulation system for COD removal from recycled paper production effluent via aluminum and iron electrodes. Different operational parameters, such as the electrolysis time (5 min to 15 min), current density (7 A/m² to 11 A/m²), and distance between each electrode (5 mm to 20 mm), were evaluated. The turbidity, total suspended solids, chlorides, sulfates, and COD had removal efficiencies of 92.7%, 91.3%, 70.4%, 66.6%, and 64%, respectively. A polynomial model was generated to estimate the optimum conditions for COD removal. The optimum times for the current densities 7 A/m², 8 A/m², 9 A/m², 10 A/m², and 11 A/m² were 39.5 min, 39.5 min, 35.7 min, 34.1 min, and 32.8 min, respectively, with a 15-mm electrode gap. The excellent characteristics of nanocellulose fiber-based papers have made them one of the most attractive and innovative materials for the development of electrochemical paper-based analytical devices (ePADs). Here, the authors describe a new ePAD based on cellulose nanofibers (CNs) for the determination of glucose concentration from whole blood samples. Cellulose acetate (CA) nanofibers were prepared by the electrospinning method. Then, the paper layer regenerated to cellulose by deacetylation in alkaline solution. To obtain a smooth and continuous CNs layer, it was treated with trimethyl chitosan (TMC). The formation of CA layer, its alkaline regeneration and TMC modification was confirmed by atomic force microscopy, scanning electron microscopy, Fourier-transform infrared spectroscopy, and energy dispersiveX-ray. Screen-printed three electrodes (SPEs) were fabricated by sputtering a thick layer of gold (Au) on the TMC/CNs substrate. Subsequently, reduced graphene oxide (rGO) was used to modify the surface of the working electrode. The fabrication steps of rGO-Au-SPE were characterized using cyclic voltammetry (CV), electrochemical impedance spectroscopy (EIS) and scanning electron microscop. The results showed that design, construction and modification process was well achieved. Next, glucose oxidase is successfully immobilized onto the regenerated CNs layer which has been previously located in the sensing area of the rGO-Au-SPEs. The ePAD glucose assay exhibited high sensitivity of $9.9\times 10^{-4}~\text{K}\Omega ^{-1} \cdot \text{mM} ^{-1}$ to glucose in the range 3.3 – 27.7 mM ($\text{R}^{2} =0.99$) with a detection limit of 0.1 mM, high reproducibility (RSD = 0.57-1.59%) and outstanding selectivity as well as stability. He was a pioneer in the application of electron microscopy to hair and wool ultrastructure and to that of the hair follicle. He discovered citrulline in keratin proteins, and the enzymes, roles, and histochemical localisation of transglutaminase and peptidylarginine deiminase (PAD). He was the first to demonstrate ribosomal-dependent keratin protein synthesis in 1965 followed by detailed studies of the biosynthesis of hair keratin proteins. His research initiated studies on the molecular events in the development of the feather follicle and later led to the cloning and characterisation of the clustered genes of feather and related avian keratins. He also cloned and characterised genes for the three classes of wool keratin proteins and studied the transcription of keratin genes. In 1977, he was awarded a DSc by the University of Adelaide in recognition for his work. In 2013, he was made an Officer in the Order of Australia. George's interest in science began at ten after his mother gave him a chemistry set and he became internationally renowned for the detailed structure and growth of hair and wool. Selective recognition of fructosyl amino acids in water by arylboronic acid-based receptors is a central field of modern supramolecular chemistry that impacts biological and medicinal chemistry. Fructosyl valine (FV) and fructosyl glycyl histidine (FGH) occur as N-terminal moieties of human glycated hemoglobin; therefore, the molecular design of biomimetic receptors is an attractive, but very challenging goal. Herein, we report three novel cationic Zn-terpyridine complexes bearing a fluorescent N-quinolinium nucleus covalently linked to three different isomers of strongly acidified phenylboronic acids (ortho-, 2Zn; meta-, 3Zn and para-, 4Zn) for the optical recognition of FV, FGH and comparative analytes (D -fructose, Gly, Val and His) in pure water at physiological pH. The complexes were designed to act as fluorescent receptors using a cooperative action of boric acid and a metal chelate. Complex 3Zn was found to display the most acidic –B(OH)2 group (pKa = 6.98) and exceptionally tight affinity for FV (K = 1.43 × 105 M−1) with a strong quenching analytical response in the micromolar concentration range. The addition of fructose and the other amino acids only induced moderate optical changes. On the basis of several spectroscopic tools (1H, 11B NMR, UV-Vis, and fluorescence titrations), ESI mass spectrometry, X-ray crystal structure, and DFT calculations, the interaction mode between 3Zn and FV is proposed in a 1 : 1 model through a cooperative two-point recognition involving a sp3 boronate–diol esterification with simultaneous coordination bonding of the carboxylate group of Val to the Zn atom. Fluorescence quenching is attributed to a static complexation photoinduced electron transfer mechanism as evidenced by lifetime experiments. The addition of FGH to 3Zn notably enhanced its emission intensity with micromolar affinity, but with a lower apparent binding constant than that observed for FV. FGH interacts with 3Zn through boronate–diol complexation and coordination of the imidazole ring of His. DFT-optimized structures of complexes 3Zn–FV and 3Zn–FGH show a picture of binding which shows that the Zn-complex has a suitable (B⋯Zn) distance to the two-point recognition with these analytes. Background Positive allosteric modulators (PAMs) of the GABAB receptor constitute a new class of GABAB-receptor ligands. GABAB PAMs reproduce several pharmacological effects of the orthosteric GABAB receptor agonist, baclofen, although displaying a better safety profile. Aims This paper reviews the reducing or, frequently, even suppressing effects of all GABAB PAMs tested to date on multiple alcohol-related behaviours in laboratory rodents exposed to validated experimental models of human alcohol use disorder. Results Acute or repeated treatment with CGP7930, GS39783, BHF177, rac -BHFF, ADX71441, CMPPE, COR659, ASP8062, KK-92A, and ORM-27669 reduced excessive alcohol drinking, relapse- and binge-like drinking, operant alcohol self-administration, reinstatement of alcohol seeking, and alcohol-induced conditioned place preference in rats and mice. Conclusions These effects closely mirrored those of baclofen; notably, they were associated to remarkably lower levels of tolerance and toxicity. The authors, a chemistry professor and a librarian, used a qualitative survey to assess student perceptions of a Wikipedia editing assignment that they included in a large upper-level biochemistry course. The assignment was initially intended as a public-facing alternative to a short research paper, emphasizing information literacy and scientific literacy. The goal of the survey was to use the results to enhance the assignment. The results of the survey and research for the literature review inspired a novel approach to the assignment using the perspective of metaliteracy. Background: The traditional pattern of theory assessment may not address the principles of assessment due to faulty paper-setting practices. This interventional study aimed to sensitize the faculty in designing a test blueprint template to set question papers (QPs). The set QPs were reviewed by experts to evaluate the effectiveness of blueprinting in the quality of the QPs. Methods: Based on the validated weightage of the biochemistry syllabus, each of the 10 faculty prepared a test blueprint, and set theory QPs without and with those test blueprints. The QPs were blinded and randomly allocated to 9 experts for evaluation. The test paper review score and feedback from both faculty and subject experts were statistically analyzed. Results: Reliability check of faculty feedback and review checklist of test papers validated its internal consistency. In all, 95% of participants expressed their agreement with various attributes of blueprinting and its future application in assessment. Statistically significant improvement (P < 0.005) was observed in the overall quality of the QPs with blueprinting. Conclusion: It was determined that blueprinting aligns objectives, content areas, and curriculum with assessment, thus improving reliability and content validity. This paper establishes a membraneless, co-laminar flow-based approach to develop a cost-effective microfluidic paper-based analytical device for enzymatic biofuel cell ($\mu $ PAD-EBFC). The developed $\mu $ PAD-EBFC supporting the self-capillary fluid transport action consists of Y-shaped paper microchannel with the fuel (glucose) and oxidant (O2) streaming in parallel over carbon nanotube-based bucky paper electrodes modified with biocompatible electrocatalytic enzymes, such as glucose oxidase and laccase without any additional redox cofactor. The electrochemical performance for the modified bioelectrodes, i.e., electrocatalytic oxidation and reduction reaction, was carried out using linear sweep voltammetry, cyclic voltammetry, and open circuit potential. The overall performance of $\mu $ PAD-EBFC was evaluated using the polarization studies. Subsequently, the catalytic activity of enzymes on the electrode surface was validated by the scanning electron microscope. This simple and portable $\mu $ PAD-EBFC can generate the maximum power density to the order of $100~\mu \text{W}$ /cm2($600~\mu \text{A}$ /cm2) at 0.505 V over prolonged durations of around 50 h. Metabolic pathway prediction and reconstruction play crucial roles in solving fundamental and applied biomedical problems. In the case of fundamental research, annotation of metabolic pathways allows one to study human health in normal, stressed, and diseased conditions. In applied research, it allows one to identify novel drugs and drug targets and to design mimetics (biomolecules with tailored properties), as well as contributes to the development of such disciplines as toxicology and nutrigenomics. It is important to understand the role of a metabolite as a substrate (the product or intermediate participant of an enzymatic reaction) in cellular signaling and phenotype implementation according to the pivotal paradigm of biology: "one gene–one protein–one function (one trait)". Due to the development of omics technologies, a vast body of data on the metabolome composition of living organisms has been accumulated over the past two decades. Systematization of the information on the roles played by metabolites in implementation of cellular signaling, as well as metabolic pathway reconstruction and refinement, have necessitated the development of bioinformatic tools for performing large-scale omics data mining. This paper reviews web-accessible databases relevant to metabolic pathways and considers the applications of the three types of bioinformatics methods for constructing metabolic networks (graphs for substrate–enzyme–product transformation; stoichiometric analysis of substrate–product transformation; and product retrosynthesis). It describes, step by step, a generalized algorithm for constructing biological pathway maps which explains to the researcher the workflow implemented in available bioinformatics tools and can be used to create new tools in projects requiring pathway reconstruction. This paper presents novel baseline health parameters on the Española lava lizard (Microlophus delanonis). Blood samples and morphological measurements were taken on 51 lizards (21 males, 30 females) captured from three locations on the island of Española. Morphological parameters measured included body weight, snout–vent length, and temperature. Blood samples were analyzed approximately 8 h after collection using an i-STAT portable blood analyzer that measured hemoglobin, total CO2, glucose, lactate, sodium, potassium, and ionized calcium. Hematologic characterization data were obtained using standard laboratory techniques. There were significant differences found in snout–vent length, weight, heart rate, respiratory rate, total CO2, and heterophil percentage between males and females. The data presented herein can be used to monitor the health of this population of Española lava lizards as well as help to understand the health of a related species, the San Cristóbal lava lizard (Microlophus bivittatus), which faces different evolutionary pressures including the presence of humans and feral cats. This study continues to widen our understanding of the genus Microlophus; however, further research should characterize the remaining species that inhabit the islands in the Galápagos archipelago. Background: Traditional problem-based learning (PBL) relying on tutored learning in small groups is very resource-intensive. Little is known about the benefits of PBL in a large classroom setting. This paper introduced a PBL case into the traditional didactic biochemistry course and investigated the acceptability of total online or partial online PBL in a large classroom setting introduced during the coronavirus pandemic. Methods: The students were allocated into either total online Group 1, partial online Group 2, or partial online and with poorer academic performance Group 3. A questionnaire comprising of 8 closed-ended questions and 2 open-ended questions and final exam performances were used to evaluate the acceptability of total online or partial online PBL in a large classroom setting. The 8 closed-ended questions were analysed by the Kruskal–Wallis test or chi-square tests. The word cloud analysis of the 2 open-ended questions were conducted by Wenjuanxing. Students' performances in the final examination were analysed by One-way Anova. Results: Both total online and partial online PBL were rated highly by the students. Overall, there were no significant differences in the effectiveness evaluation of PBL between Group 2 and Group 3. There were no significant differences in final exam performances between Group 1 and Group 2. However, Group 1 rated the effectiveness of PBL much higher than Group 2 and 3. Word cloud analysis of the 2 open-ended questions showed students' positive perspectives of PBL. In biochemistry teaching, from the perspective of the students, the expected optimal number of useful PBL cases might be 2. Background: Cigarette smoking remains the leading cause of preventable disease and death in the United States. Primary care offers an ideal setting to reach adults who smoke cigarettes and improve uptake of evidence-based cessation treatment. Although U.S. Preventive Services Task Force Guidelines recommend the 5As model (Ask, Advise, Assess, Assist, Arrange) in primary care, there are many barriers to its implementation. Automated, comprehensive, and proactive tools are needed to overcome barriers. Our team developed and preliminarily evaluated a proactive electronic visit (e-visit) delivered via the Electronic Health Record patient portal to facilitate evidence-based smoking cessation treatment uptake in primary care, with promising initial feasibility and efficacy. This paper describes the rationale, design, and protocol for an ongoing Hybrid Type I effectiveness-implementation trial that will simultaneously assess effectiveness of the e-visit intervention for smoking cessation as well as implementation potential across diverse primary care settings. Methods: The primary aim of this remote five-year study is to examine the effectiveness of the e-visit intervention vs. treatment as usual (TAU) for smoking cessation via a clinic-randomized clinical trial. Adults who smoke cigarettes are recruited across 18 primary care clinics. Clinics are stratified based on their number of primary care providers and randomized 2:1 to either e-visit or TAU. An initial baseline e-visit gathers information about patient smoking history and motivation to quit, and a clinical decision support algorithm determines the best evidence-based cessation treatment to prescribe. E-visit recommendations are evaluated by a patient's own provider, and a one-month follow-up e-visit assesses cessation progress. Main outcomes include: (1) cessation treatment utilization (medication, psychosocial cessation counseling), (2) reduction in cigarettes per day, and (3) biochemically verified 7-day point prevalence abstinence (PPA) at six-months. We hypothesize that patients randomized to the e-visit condition will have better cessation outcomes (vs. TAU). A secondary aim evaluates e-visit implementation potential at patient, provider, and organizational levels using a mixed-methods approach. Implementation outcomes include acceptability, adoption, fidelity, implementation cost, penetration, and sustainability. Background: Osteoarthritis (OA), characterized by inflammation and articular cartilage degradation, is a prevalent arthritis among geriatric population. This paper was to scrutinize the novel mechanism of long noncoding RNA (lncRNA) NEAT1 in OA etiology. Methods: A total of 10 OA patients and 10 normal individuals was included in this study. Cell model of OA was built in human normal chondrocytes induced by lipopolysaccharide (LPS). An OA Wistar rat model was established through intra-articular injection of L-cysteine and papain mixtures (proportion at 1:2) into the right knee. Quantitative reverse transcription-polymerase chain reaction was employed to ascertain the expression levels of NEAT1, microRNA (miR)-374b-5p and post-GPI attachment to protein 1 (PGAP1), while dual-luciferase reporter experiments were used for the validation of target relationship among them. Cell cycle and apoptosis were calculated by flow cytometry analysis. CCK-8 assay was done to evaluate the proliferative potentials of chondrocytes. The levels of cell cycle-related proteins (Cyclin A1, Cyclin B1 and Cyclin D2) and pro-apoptotic proteins (Caspase3 and Caspase9) were measured by western blotting. Tumor necrosis factor-alpha (TNF-α), interleukin-1beta (IL-1β) and IL-6 levels were determined via ELISA. Hematoxylin & eosin (HE) Staining was used for pathological examination in OA rats. Results: Pronounced downregulation of NEAT1 and PGAP1 and high amounts of miR-374b-5p were identified in OA patients, LPS-induced chondrocytes and OA rats. NEAT1 targeted miR-374b-5p to control PGAP1 expression. Loss of NEAT1 or upregulation of miR-374b-5p dramatically accelerated apoptosis, led to the G1/S arrest and promoted the secretion of inflammatory cytokines in LPS-induced chondrocytes, while ectopic expression of PGAP1 exhibited the opposite influences on chondrocytes. Additionally, we further indicated that upregulation of miR-374b-5p attenuated the effects of PGAP1 overexpression on LPS-induced chondrocytes. Conclusions: Reduced NEAT1 induces the development of OA via miR-374b-5p/PGAP1 pathway. Background: The role of dietary fat quality in promotion of cardiovascular diseases is studies before. However, the results are inconsistent. Recently, cholesterol to saturated fatty acid index (CSI) is suggested as a novel indicator of the atherogenicity and thrombogenicity potential of a diet. However, due to limited number of studies, in the current cross-sectional study, we aimed to evaluate the role of CSI in metabolic and inflammatory response among obese individuals. Methods: In the current cross-sectional study 488 obese individuals aged 18–50 years old were involved in volunteer based invitation from outpatient obesity clinics. Subjects underwent anthropometric assays including weight, height, waist circumference (WC) and body composition and their fasting blood sample were obtained for biochemical assessments including blood sugar, serum lipids, hs-CRP and IL-6 concentrations by commercial kits. Physical activity was also assessed by short form of international physical activity questionnaire (IPAQ). Results: According to our results, being at the top tetile of CSI was associated with higher anthropometric indices including weight, height, WC, FFM, and basal metabolic rate (BMR) compared with those at the lowest tertile (P < 0.05). Similarly, those at the highest category of CSI had significantly higher levels of serum glucose and hs-CRP both in crude and adjusted models in ANCOVA and in multinomial logistic regression models (P < 0.05). Conclusion: In the current study, for the first time, we identified the possible triggering role of dietary cholesterol to saturated fat index in increasing serum glucose and hs-CRP levels. due to cross-sectional design of the current study, causal inference is impossible. This review uses the total synthesis of vancomycin analogues by Boger and his colleagues as a case study of the drug discovery process. Vancomycin is a glycopeptide antibiotic used to treat staphylococcal infections. It acts by binding to the terminal chains attached to peptidoglycan in bacterial cell walls, which blocks these chains from crosslinking with each other. This inability to crosslink prevents the bacterium from forming a cell wall, ultimately resulting in bacterial cell death. However, over time, bacteria have evolved resistance to vancomycin. In response to this, Boger and his team studied single atom changes (O, S, NH) in the chemical structure of vancomycin to create new vancomycin analogues that these bacteria are not resistant to. The drug discovery process involves upstream and downstream flow of information about a potential drug candidate, and the interlude of Boger's work with vancomycin elegantly demonstrates this process. Boger found that replacing a single oxygen atom in vancomycin with NH significantly increases the ability of the analogue to bind with peptidoglycan terminal chains, increasing its effectiveness as an antibiotic. RNA's diversity of structures and functions impacts all life forms since primordia. We use calorimetric force spectroscopy to investigate RNA folding landscapes in previously unexplored low-temperature conditions. We find that Watson-Crick RNA hairpins, the most basic secondary structure elements, undergo a glass-like transition below TG ~ 20 °C where the heat capacity abruptly changes and the RNA folds into a diversity of misfolded structures. We hypothesize that an altered RNA biochemistry, determined by sequence-independent ribose-water interactions, outweighs sequencedependent base pairing. The ubiquitous ribose-water interactions lead to universal RNA phase transitions below TG, such as maximum stability at TS ~ 5 °C where water density is maximum, and cold denaturation at TC ~ 50 °C. Kynurenic acid (KYNA) is an antioxidant degradation product of tryptophan that has been shown to have a variety of cytoprotective, neuroprotective and neuronal signalling properties. However, mammalian transporters and receptors display micromolar binding constants; these are consistent with its typically micromolar tissue concentrations but far above its serum/plasma concentration (normally tens of nanomolar), suggesting large gaps in our knowledge of its transport and mechanisms of action, in that the main influx transporters characterized to date are equilibrative, not concentrative. In addition, it is a substrate of a known anion efflux pump (ABCC4), whose in vivo activity is largely unknown. Exogeneous addition of L-tryptophan or L-kynurenine leads to the production of KYNA but also to that of many other co-metabolites (including some such as 3-hydroxy-L-kynurenine and quinolinic acid that may be toxic). With the exception of chestnut honey, KYNA exists at relatively low levels in natural foodstuffs. However, its bioavailability is reasonable, and as the terminal element of an irreversible reaction of most tryptophan degradation pathways, it might be added exogenously without disturbing upstream metabolism significantly. Many examples, which we review, show that it has valuable bioactivity. Background: Obesity is a global health concern associated with increased risk of diseases like cardiovascular conditions including ischemic heart disease, a leading cause of mortality. The ketogenic diet (KD) has potential therapeutic applications in managing obesity and related disorders. However, the intricate effects of KD on diverse physiological conditions remain incompletely understood. The PI3K-Akt signaling pathway is critical for heart health, and its dysregulation implicates numerous cardiac diseases. Methods: We developed comprehensive mathematical models of the PI3K-Akt signaling pathway under high-fat diet (HFD) and KD conditions to elucidate their differential impacts and quantify apoptosis. Simulations and sensitivity analysis were performed. Results: Simulations demonstrate that KD can reduce the activation of key molecules like Erk and Trp53 to mitigate apoptosis compared to HFD. Findings align with experimental data, highlighting the potential cardiac benefits of KD. Sensitivity analysis identifies regulators like Trp53 and Bcl2l1 that critically influence apoptosis under HFD. Conclusions: Mathematical modeling provides quantitative insights into the contrasting effects of HFD and KD on cardiac PI3K-Akt signaling and apoptosis. Werner syndrome (WS) is an autosomal recessive disease caused by loss of function of WRN. WS is a segmental progeroid disease and shows early onset or increased frequency of many characteristics of normal aging. WRN possesses helicase, annealing, strand exchange, and exonuclease activities and acts on a variety of DNA substrates, even complex replication and recombination intermediates. Here, we review the genetics, biochemistry, and probably physiological functions of the WRN protein. Although its precise role is unclear, evidence suggests WRN plays a role in pathways that respond to replication stress and maintain genome stability particularly in telomeric regions. Social changes in higher education have led to a transformation in teaching innovation for developing a critical and creative mentality in students. To this end, video capsules have emerged as an optimal tool that allows combining the management of information and communication technologies and collaborative work. Video capsules were elaborated by groups of students for the virtualization of the laboratory practices of the subjects Environmental Microbiology and Biochemistry of the Degree in Environmental Sciences for two academic years (2022–2023 and 2023–2024), paying special attention to the principles of Universal Design of Learning. To assess the impact of this activity on the knowledge acquired by the students in the laboratory practices, the marks from these academic years were compared with the previous academic year (2021–2022) without the generation of video capsules. Moreover, an evaluation of the development and acquisition of cross-curricular competencies was carried out. The generation of video capsules by the students tended to increase the average grade obtained in the laboratory practices of both subjects. Furthermore, instrumental, interpersonal, and systemic competencies obtained by the students by performing the video capsules improved their transversal skills. The creation of video capsules during laboratory practices and the methodology used had a very positive direct influence on the academic outcomes of the subjects and facilitated the acquisition of transversal competencies for the students.
