Simple Summary: Granulosa cells play an essential role in oocyte meiotic competence acquisition by supplying nutrients and metabolites to oocytes and secreting paracrine signals that regulate oocyte maturation. Identifying biomarkers in granulosa cells to measure an oocyte's ability to mature in vitro noninvasively predicts success or failure prior to maturation. This technique also offers the opportunity to develop treatment strategies to overcome potential maturation failures. This study aimed to evaluate potential biomarkers associated with follicle development, meiotic competence, cell death and atresia, and embryonic genome activation in granulosa cells from oocytes that did or did not mature after in vitro fertilization in the southern white rhinoceros. This study determined eight potential biomarkers associated with the success or failure of an oocyte to mature. Two genes were correlated with follicle development, three genes with meiotic competence, and three genes with cell death and atresia. This is the first study in which in vivo granulosa cells were used as a diagnostic predictor of oocyte maturation competence in the southern white rhinoceros. It represents a critical evaluation needed to improve assisted reproductive technologies in this species. In recent years, biomarkers in granulosa cells (GC) have been determined and associated in several species with oocyte maturation, in vitro fertilization success, and embryo development outcomes. The identification of biomarkers of oocyte competence can aid in improving assisted reproductive technologies (ARTs) in the southern white rhino (SWR). This study aimed to identify biomarkers present in SWR GC associated with oocytes that either did or did not mature in vitro. We evaluated follicle development (FD), meiotic competence (MC), cell death and atresia (CDA), and embryonic genome activation (EGA). Our objective was to design biomarkers to predict oocyte in vitro maturation results in the SWR. RNA was isolated from GC obtained during ovum pick up (OPU) for qPCR analysis. Overall, 22 genes were assessed, and nine were differentially expressed between GC from oocytes that did or did not mature in vitro (FD-GDF9 and mTOR; MC-GGPS1, JMY, and NPR2; CDA-COL4A1, MACIR, and TMPO; EGA-NFYA). From these data, we determined that GC can be used as a predictor for oocyte in vitro maturation outcome in the SWR. Background: Blastocyst morphology has been demonstrated to be associated with ploidy status. Existing artificial intelligence models use manual grading or 2D images as the input for euploidy prediction, which suffer from subjectivity from observers and information loss due to incomplete features from 2D images. Here we aim to predict euploidy in human blastocysts using quantitative morphological parameters obtained by 3D morphology measurement. Methods: Multi-view images of 226 blastocysts on Day 6 were captured by manually rotating blastocysts during the preparation stage of trophectoderm biopsy. Quantitative morphological parameters were obtained by 3D morphology measurement. Six machine learning models were trained using 3D morphological parameters as the input and PGT-A results as the ground truth outcome. Model performance, including sensitivity, specificity, precision, accuracy and AUC, was evaluated on an additional test dataset. Model interpretation was conducted on the best-performing model. Results: All the 3D morphological parameters were significantly different between euploid and non-euploid blastocysts. Multivariate analysis revealed that three of the five parameters including trophectoderm cell number, trophectoderm cell size variance and inner cell mass area maintained statistical significance (P < 0.001, aOR = 1.054, 95% CI 1.034–1.073; P = 0.003, aOR = 0.994, 95% CI 0.991–0.998; P = 0.010, aOR = 1.003, 95% CI 1.001–1.006). The accuracy of euploidy prediction by the six machine learning models ranged from 80 to 95.6%, and the AUCs ranged from 0.881 to 0.984. Particularly, the decision tree model achieved the highest accuracy of 95.6% (95% CI 84.9-99.5%) with the AUC of 0.978 (95% CI 0.882–0.999), and the extreme gradient boosting model achieved the highest AUC of 0.984 (95% CI 0.892-1.000) with the accuracy of 93.3% (95% CI 81.7-98.6%). No significant difference was found between different age groups using either decision tree or extreme gradient boosting to predict euploid blastocysts. The quantitative criteria extracted from the decision tree imply that euploid blastocysts have a higher number of trophectoderm cells, larger inner cell mass area, and smaller trophectoderm cell size variance compared to non-euploid blastocysts. Conclusions: Using quantitative morphological parameters obtained by 3D morphology measurement, the decision tree-based machine learning model achieved an accuracy of 95.6% and AUC of 0.978 for predicting euploidy in Day 6 human blastocysts. Until now, North Sumatra's food security is still unstable. This condition gives a clear picture that North Sumatra Province experiences problems in the field of food security. Because of the position of the North Sumatra region's very strategic position, this province should be prosperous and independent in terms of food sources. This research aims to develop a policy strategy for the development of food commodities towards national food security. In supporting this research, SWOT, Interpretative Structural Modeling (ISM), and Balanced Score Card (BSC) methods are used. The SWOT method was used to formulate a strategy based on the identification of internal factors and external factors that had a significant influence based on the Mckinsey 7S and PEST models. Furthermore, the ISM method is used to determine strategy priorities. The BSC method is used to determine the implementation plan and mapping of the strategies formulated. The development of food commodity development policy strategies towards food security in North Sumatra based on the results of the research consists of 11 (eleven) main strategies consisting of Strategy SO-1,2,3, Strategy ST-1,2,3, Strategy WO-1,2,3 and Strategy WT-1,2. It is hoped that the eleven strategies resulting from this research can increase the development of food commodities in North Sumatra towards stable and sustainable food security. Drought is a major factor affecting coffee production, and different genotypes exhibit varying degrees of resistance to drought stress. We examined root traits and biomasses of drought‐resistant (74110, Angafa, Bultum, Chala, and Gawe) and drought‐sensitive (75227, Koti, Melko CH2, Menasibu, and Mokah) Coffea arabica varieties at seedling stage under contrasting watering regimes (water stressed and well watered) for 30 days followed by 15 days of recovery to identify the association between drought resistance and root traits and dry matter partitioning, and the impact of drought stress on growth performance of arabica coffee varieties. We used a split‐plot design with three blocks, where watering regime was the whole‐plot factor and variety was the subplot factor. During water‐stress and recovery periods, the interaction effect between watering regime and variety significantly affected root traits and dry matter partitioning, while the watering‐regime main effect affected biomass. We observed a higher (1) tap root diameter (0.34 cm), lateral root number (80.7), and root volume (4.7 cm−3) for 74110; (2) lateral root number (79.3), specific root length (24.8 cm g−1), and root‐mass ratio (0.41 g g−1) for Bultum; and (3) root length density (3.3–5.2 cm cm−3), root angle (42.6°–47.8°), root‐mass ratio (0.40–0.42 g g−1), and root‐shoot ratio (0.67–0.72 g g−1) for Angafa, Chala, and Gawe under water‐stressed condition. During both study periods, biomasses were much lower under water‐stressed than under well‐watered condition. The findings show the association between drought resistance and root traits and dry matter partitioning, and the impact of drought stress on growth performance of young arabica coffee. Cell-free DNA (cfDNA), a burgeoning class of molecular biomarkers, has been extensively studied across a variety of biomedical fields. As a key component of liquid biopsy, cfDNA testing is gaining prominence in disease detection and management due to the convenience of sample collection and the abundant wealth of genetic information it provides. However, the broader clinical application of cfDNA is currently impeded by a lack of standardization in the preanalytical procedures for cfDNA analysis. A number of fundamental challenges, including the selection of appropriate preanalytical procedures, prevention of short cfDNA fragment loss, and the validation of various cfDNA measurement methods, remain unaddressed. These existing hurdles lead to difficulties in comparing results and ensuring repeatability, thereby undermining the reliability of cfDNA analysis in clinical settings. This review discusses the crucial preanalytical factors that influence cfDNA analysis outcomes, including sample collection, transportation, temporary storage, processing, extraction, quality control, and long-term storage. The review provides clarification on achievable consensus and offers an analysis of the current issues with the goal of standardizing preanalytical procedures for cfDNA analysis. It is widely postulated that the majority of pathologically elevated extracellular or cell-free DNA (cfDNA) in cancer originates from tumor cells; however, evidence has emerged regarding the significant contributions of other cells from the tumor microenvironment. Here, the effect of cfDNA originating from murine B16 melanoma cells and L929 fibroblasts on B16 cells was investigated. It was found that cfDNAL929 increased the viability and migration properties of B16 cells in vitro and their invasiveness in vivo. In contrast, cfDNAB16 exhibited a negative effect on B16 cells, reducing their viability and migration in vitro, which in vivo led to decreased tumor size and metastasis number. It was shown that cell treatment with both cfDNAs resulted in an increase in the expression of genes encoding DNases and the oncogenes Braf, Kras, and Myc. cfDNAL929-treated cells were shown to experience oxidative stress. Gene expression changes in the case of cfDNAB16 treatment are well correlated with the observed decrease in proliferation and migration of B16 cells. The obtained data may indicate the possible involvement of fibroblast DNA in the tumor microenvironment in tumor progression and, potentially, in the formation of new tumor foci due to the transformation of normal cells. Background: Elevated FSH often occurs in women of advanced maternal age (AMA, age ≥ 35) and in infertility patients undergoing controlled ovarian stimulation (COS). There is controversy on whether high endogenous FSH contributes to infertility and whether high exogenous FSH adversely impacts patient pregnancy rates. Methods: The senescence-accelerated mouse-prone-8 (SAMP8) model of female reproductive aging was employed to assess the separate impacts of age and high FSH activity on the percentages (%) of viable and mature ovulated oocytes recovered after gonadotropin treatment. Young and midlife mice were treated with the FSH analog equine chorionic gonadotropin (eCG) to model both endogenous FSH elevation and exogenous FSH elevation. Previously we showed the activin inhibitor ActRIIB:Fc increases oocyte quality by preventing chromosome and spindle misalignments. Therefore, ActRIIB:Fc treatment was performed in an effort to increase % oocyte viability and % oocyte maturation. Results: The high FSH activity of eCG is ootoxic to ovulatory oocytes, with greater decreases in % viable oocytes in midlife than young mice. High FSH activity of eCG potently inhibits oocyte maturation, decreasing the % of mature oocytes to similar degrees in young and midlife mice. ActRIIB:Fc treatment does not prevent eCG ootoxicity, but it restores most oocyte maturation impeded by eCG. Conclusions: FSH ootoxicity to ovulatory oocytes and FSH maturation inhibition pose a paradox given the well-known pro-growth and pro-maturation activities of FSH in the earlier stages of oocyte growth. We propose the FOOT Hypothesis ("FSH OoToxicity Hypothesis), that FSH ootoxicity to ovulatory oocytes comprises a new driver of infertility and low pregnancy success rates in DOR women attempting spontaneous pregnancy and in COS/IUI patients, especially AMA women. We speculate that endogenous FSH elevation also contributes to reduced fecundity in these DOR and COS/IUI patients. Restoration of oocyte maturation by ActRIB:Fc suggests that activin suppresses oocyte maturation in vivo. This contrasts with prior studies showing activin A promotes oocyte maturation in vitro. Improved oocyte maturation with agents that decrease endogenous activin activity with high specificity may have therapeutic benefit for COS/IVF patients, COS/IUI patients, and DOR patients attempting spontaneous pregnancies. Involving the global issues as listed in Sustainable Development Goals (SDGs) in education is necessarily done in the education process, especially in English as a Foreign Language (EFL) learning. Exposure to global issues is known to improve students' understanding, awareness, and ability to solve urgent issues faced by global society. This paper aims to find out the trend of research on the coverage of SDGs in students' learning process. This systematic literature analysis was done by applying Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) Method. A total of 25 studies were recognized through a systematic search by using Sustainability, SDGs, and EFL as keywords. The result shows that the trend of associating SDGs with EFL settings was done mostly in Indonesia. In the recent year 2022, it reached the highest number of studies in the particular matter with 7 total of research. It was also found that the study involving SDGs on EFL learning was mostly done in the tertiary setting, compared with K-12, junior high school, secondary, high school, and other educational institutions. It was also discovered that the specific area of study enhances EFL students' learning achievement, environmental awareness, global citizen values, as well as students' levels of selfnorms, beliefs, and self-value. The leading indicator for successful outcomes in in-vitro fertilization (IVF) is the quality of gametes in oocytes and sperm. Thus, advanced research aims to highlight the parameter in assessing these qualities - DNA fragmentation in sperm and oocyte development capacity (ODC) via evaluation of microenvironments involving its maturation process. Regarding oocytes, most evidence reveals the role of cumulus cells as non-invasive methods in assessing their development competency, mainly via gene expression evaluation. Our review aims to consolidate the evidence of GDF-9 derivatives, the HAS2, GREM1, and PTGS2 gene expression in cumulus cells used as ODC markers in relevant publications and tailored to current IVF outcomes. In addition to that, we also added the bioinformatic analysis in our review to strengthen the evidence aiming for a better understanding of the pathways and cluster of the genes of interest - HAS2, GREM1, and PTGS2 in cumulus cell level. Otherwise, the current non-invasive method can be used in exploring various causes of infertility that may affect these gene expressions at the cumulus cell level. Nevertheless, this method can also be used in assessing the ODC in various cohorts of women or as an improvement of markers following targeted tools or procedures by evaluating the advancement of these gene expressions following the targeted intervention. Age-related changes in the mitochondrial status of human cumulus cells (hCCs) impact oocyte quality; however, the relationship between hCC mitochondrial (dys)function and reproductive aging remains poorly understood. This study aimed to establish the interplay between hCC mitochondrial dysfunction and women's reproductive potential. In this investigation, 266 women were enrolled and categorized into two groups based on their age: a young group (<35 years old) and an advanced maternal age (AMA) group (≥35 years old). Comprehensive analysis of reproductive outcomes was conducted in our population. Various mitochondrial-related parameters were analyzed across distinct subsets. Specifically, mitochondrial membrane potential (∆Ψm) and mitochondrial mass were examined in 53 samples, mtDNA content in 25 samples, protein levels in 23 samples, bioenergetic profiles using an XF24 Extracellular Flux Analyzer in 6 samples, and levels of reactive oxygen species (ROS) and adenosine triphosphate (ATP) in 39 and 43 samples, respectively. In our study, the reproductive potential of AMA women sharply decreased, as expected. Additionally, an impairment in the mitochondrial function of hCCs in older women was observed; however, no differences were found in terms of mitochondrial content. Regarding oxidative phosphorylation, metabolic profiling of hCCs from AMA women indicated a decrease in respiratory capacity, which was correlated with an age-dependent decrease in the ATP synthase (ATP5A1) protein level. However, intracellular ROS and ATP levels did not differ between groups. In conclusion, our study indicates that age-related dysfunction in hCCs is associated with impaired mitochondrial function, and, although further studies are required, ATP synthase could be relevant in this impairment. Postovulatory aging is known to impair the oocyte quality and embryo development due to oxidative stress in many different animal models, which reduces the success rate or pregnancy rate in human assisted reproductive technology (ART) and livestock timed artificial insemination (TAI), respectively. Salidroside (SAL), a phenylpropanoid glycoside, has been shown to exert antioxidant and antitumor effects. This study aimed to investigate whether SAL supplementation could delay the postovulatory oocyte aging process by alleviating oxidative stress. Here, we show that SAL supplementation decreases the malformation rate and recovers mitochondrial dysfunction including mitochondrial distribution, mitochondrial membrane potential (ΔΨ) and ATP content in aged oocytes. In addition, SAL treatment alleviates postovulatory aging-caused oxidative stress such as higher reactive oxygen species (ROS) level, lower glutathione (GSH) content and a reduced expression of antioxidant-related genes. Moreover, the cytoplasmic calcium ([Ca2+]c) and mitochondrial calcium ([Ca2+]mt) of SAL-treated oocytes return to normal levels. Notably, SAL suppresses the aging-induced DNA damage, early apoptosis and improves spindle assembly in aged oocytes, ultimately elevating the embryo developmental rates and embryo quality. Finally, the RNA-seq and confirmatory experience showed that SAL promotes protective autophagy in aged oocytes by activating the MAPK pathway. Taken together, our research suggests that supplementing SAL is an effective and feasible method for preventing postovulatory aging and preserving the oocyte quality, which potentially contributes to improving the successful rate of ART or TAI. Purpose: Although significant improvements in assisted reproductive technology (ART) outcomes have been accomplished, a critical question remains: which embryo is most likely to result in a pregnancy? Embryo selection is currently based on morphological and genetic criteria; however, these criteria do not fully predict good-quality embryos and additional objective criteria are needed. The cumulus cells are critical for oocyte and embryo development. This systematic review assessed biomarkers in cumulus-oocyte complexes and their association with successful IVF outcomes. Methods: A comprehensive search was conducted using PubMed, Embase, Scopus, and Web of Science from inception until November 2022. Only English-language publications were included. Inclusion criteria consisted of papers that evaluated genetic biomarkers associated with the cumulus cells (CCs) in humans and the following three outcomes of interest: oocyte quality, embryo quality, and clinical outcomes, including fertilization, implantation, pregnancy, and live birth rates. Results: The search revealed 446 studies of which 42 met eligibility criteria. Nineteen studies correlated genetic and biochemical biomarkers in CCs with oocyte quality. A positive correlation was reported between oocyte quality and increased mRNA expression in CCs of genes encoding for calcium homeostasis (CAMK1D), glucose metabolism (PFKP), extracellular matrix (HAS2, VCAN), TGF-β family (GDF9, BMP15), and prostaglandin synthesis (PTGS2). Nineteen studies correlated genetic and biochemical biomarkers in CCs with embryo quality. A positive correlation was reported between embryo quality and increased mRNA expression in CCs of genes encoding for extracellular matrix (HAS2), prostaglandin synthesis (PTGS2), steroidogenesis (GREM1), and decreased expression of gene encoding for hormone receptor (AMHR2). Twenty-two studies assessed genetic and biochemical biomarkers in CCs with clinical outcomes. Increased expression of genes encoding for extracellular matrix (VCAN), and TGF-β family (GDF9, BMP15) were positively correlated with pregnancy rate. Conclusion: Genetic biomarkers from cumulus cells were associated with oocyte quality (CAMK1D, PFKP, HAS2, VCAN, GDF-9, BMP-15, PTGS2), embryo quality (GREM1, PTGS2, HAS2), and pregnancy rate (GDF9, BMP15, VCAN). Background: The exponential data growth emphasises the importance of efficient information flow in organisations, especially in the financial sector. Data quality significantly influences decision-making, necessitating reliable Data Quality Management (DQM) frameworks. Previous studies propose DQM to maintain data quality through regulation, technology, measurement, evaluation, and improvement. Researchers highlight high-quality data benefits in private organisations but note the lack of improvement in data utilisation in public organisations. In Indonesia, data accuracy and quality are crucial for financial policies, with frequent reports of data inaccuracies in the Directorate General of Customs and Excise (DJBC), demanding standardised DQM practices. However, However, prior studies have yet to provide comprehensive and practical solutions to improve DQM practices. This study therefore aims to measure the DQM maturity, provide recommendations based on best practices, and formulate a practical strategy for improvements along with indicators tailored to the organisation, a topic that previous research has not explored. Methods: This study falls under a mixed method approach (a quantitative study followed by a qualitative study) and employs a three-stage methodology. The authors conduct maturity assessment using Loshin model through an assisted enumeration from 5 key stakeholders followed by recommendations based on the Data Management Body of Knowledge (DMBOK) and strategy formulation from internal documents and interview. Results: The data analysis yielded a DQM maturity score of 3.10, indicating a "defined to managed" level of maturity. Among eight components, only one receives a Managed level, two components are in the Defined level and the rest belongs to a Repeatable level. This study also proposes three strategies to bolster DQM by targeting 49 weak points, which will be progressively and sequentially implemented over a three-year period, using twelve possible solutions. Conclusion: The study highlights the importance of efficient data flow, particularly in the financial sector, and suggests DQM for maintaining data quality. DJBC's import DQM level is assessed using Loshin's measurements, revealing areas for improvement through key DMBOK activities. Recommendations include data governance, strategic planning, and sequential DQM implementation. This review elucidates diverse microfluidic systems in mimicking the ovary, fallopian tube, uterus, placenta and cervix, and we delve into the culture of follicles and oocytes, gametes' manipulation, cryopreservation, and permeability especially. We investigate the role of microfluidics in endometriosis and hysteromyoma, and explore their applications in ovarian cancer, endometrial cancer and cervical cancer. At last, the current status of assisted reproductive technology and integrated microfluidic devices are introduced briefly. Through delineating the multifarious advantages and challenges of the microfluidic technology, we chart a definitive course for future research in the woman health field. As the microfluidic technology continues to evolve and advance, it holds great promise for revolutionizing the diagnosis and treatment of female reproductive health issues, thus propelling us into a future where we can ultimately optimize the overall wellbeing and health of women everywhere. Objective(s): Polycystic ovary syndrome (PCOS), the primary cause of anovulatory infertility in women, may change the gene expression profile of cumulus cells. In human ART (assisted reproductive technology), gene expression profiling in cumulus cells, a non-invasive method, may be used to identify the most competent oocytes. We aim to identify key genes according to the networkbased data and assess the suitability of these genes as markers to predict oocyte competence and PCOS diagnosis. Materials and Methods: The GSE34526 microarray dataset was obtained from the Gene Expression Omnibus (GEO) database. The function and pathway enrichment analysis for DEGs were analyzed. A protein-protein interaction (PPI) network analysis and candidate gene screening were conducted. A two-layer network consisting of mRNA and lncRNA was constructed. Expression levels of hub genes were verified using quantitative RT-PCR (qRT-PCR). Results: A total of 2721 DEGs were retained. The PPI network of selected genes associated with the biological process of "cell communication" was analyzed, and the first 10 key genes were determined by degree. Additionally, 2 hub genes and 2 hub lncRNAs, including STAT3, RHOA, GAS5, and LINC01116, were selected from the lncRNA-mRNA network. Finally, expression levels of STAT3, RHOA, GAS5, and LINC01116 were significantly increased in the cumulus cells of PCOS patients compared to the control group (P<0.05). However, there was no significant difference in expression between the pregnant and non-pregnant groups. Conclusion: STAT3, RHOA, GAS5, and LINC01116 may serve as possible diagnostic markers for PCOS. However, further studies on a larger population are needed to validate this finding. The search for simple morphological predictors of oocyte quality is an important task for assisted reproduction technologies (ARTs). One such predictor may be the morphology of the oocyte nucleus, called the germinal vesicle (GV), including the level of chromatin aggregation around the atypical nucleolus (ANu)—a peculiar nuclear organelle, formerly referred to as the nucleolus-like body. A prospective cohort study allowed distinguishing three classes of GV oocytes among 135 oocytes retrieved from 64 patients: with a non-surrounded ANu and rare chromatin blocks in the nucleoplasm (Class A), with a complete peri-ANu heterochromatic rim assembling all chromatin (Class C), and intermediate variants (Class B). Comparison of the chromatin state and the ability of oocytes to complete meiosis allowed us to conclude that Class B and C oocytes are more capable of resuming meiosis in vitro and completing the first meiotic division, while Class A oocytes can resume maturation but often stop their development either at metaphase I (MI arrest) or before the onset of GV breakdown (GVBD arrest). In addition, oocytes with a low chromatin condensation demonstrated a high level of aneuploidy during the resumption of meiosis. Considering that the degree of chromatin condensation/compaction can be determined in vivo under a light microscope, this characteristic of the GV can be considered a promising criterion for selecting the best-quality GV oocytes in IVM rescue programs. Shallot (Allium cepa var. aggregatum.) is a horticultural plant that is widely consumed in the world. However, the productivity of shallots in Indonesia is still relatively low, if compared to the actual optimum production potential of shallot. Shallot cultivation in Indonesia often experiences many problems. One of the problems is twisted diseases caused by Fusarium sp. This research aimed to study the effect of the application of organic material enriched with Bacillus in suppressing the development of twisted disease of shallot and increasing yield of shallot. This study was arranged in Randomized Complete Block Design (RCBD) with 5 treatments namely (A) compost + Bacillus velezensis isolate B-27, (B) compost + Bacillus cereus isolate RC76, (C) B. velezensis isolate B-27+B. cereus isolate RC76+compost, (D) compost + Trichoderma asperellum and (E) control (compost 1 ton/ha) with 5 replications on glasshouse treatment and 3 replications on field treatment. The results showed that the combination of B. velezensis in compost effectively reduced the incidence of twisted disease, the number of Fusarium spp. colonies, and the number of infected bulbs by Fusarium sp. Besides, the combination of compost with microbial agents showed better results than compost single treatment. Summary: Here we report a quantitative analysis of human metaphase II (MII) oocytes from a 22-year-old oocyte donor, retrieved after ovarian-controlled hyperstimulation. Five surplus donor oocytes were processed for transmission electron microscopy (TEM), and a stereological analysis was used to quantify the distribution of organelles, using the point-counting technique with an adequate stereological grid. Comparisons between means of the relative volumes (Vv) occupied by organelles in the three oocyte regions, cortex (C), subcortex (SC) and inner cytoplasm (IC), followed the Kruskal–Wallis test and Mann–Whitney U -test with Bonferroni correction. Life cell imaging and TEM analysis confirmed donor oocyte nuclear maturity. Results showed that the most abundant organelles were smooth endoplasmic reticulum (SER) elements (26.8%) and mitochondria (5.49%). Significant differences between oocyte regions were found for lysosomes (P = 0.003), cortical vesicles (P = 0.002) and large SER vesicles (P = 0.009). These results were quantitatively compared with previous results using prophase I (GV) and metaphase I (MI) immature oocytes. In donor MII oocytes there was a normal presence of cortical vesicles, SER tubules, SER small, medium and large vesicles, lysosomes and mitochondria. However, donor MII oocytes displayed signs of cytoplasmic immaturity, namely the presence of dictyosomes, present in GV oocytes and rare in MI oocytes, of SER very large vesicles, characteristic of GV oocytes, and the rarity of SER tubular aggregates. Results therefore indicate that the criterion of nuclear maturity used for donor oocyte selection does not always correspond to cytoplasmic maturity, which can partially explain implantation failures with the use of donor oocytes. This study is prompted by the election of female legislative candidates in East Java's 2019 General Elections. The notable presence of women in the East Java Regional Legislative Assembly serves as a cause for celebration, as it has observed a commendable quantitative surge from a previous count of seven individuals in the 2014 election to a noteworthy tally of 21 individuals in the 2019 election. The research aims to investigate how these four female legislative candidates utilized their social capital to secure their positions as elected members in the 2019 General Elections. The primary informants selected for the study met certain criteria, including being the incumbent legislative member of their electoral area, being the representative of their respective regions, and having a top or early sequence number in the election. This study is founded upon Robert Putnam's social capital theory and is bolstered by Claudia Derich's, Andrea Fleschenberg's, and Momoyo Hustebeck's moral capital theory. The methodology employed in this study is qualitative, employing a case study approach centered around in-depth interviews with four female legislative candidates who were successfully elected as legislative members. The primary findings of this study highlight several key factors that contributed to the election of these four female legislative candidates. These factors include significant support from their family members (spouses and parents) as well as endorsements from various colleagues representing religious organizations, social organizations, and alumni networks. Additionally, their success in the elections can be attributed to the moral capital they possess, which derived from their professional backgrounds as advocates, businesswomen, retired civil servants, and academicians. One of the most popular technologies is the internet of things (IoT). It refers to the number of users and penetration in the industry (I-IoT) and consumer (C-IoT) sectors. The previous stud-ies indicated that the usage rate of the C-IoT is outperforming the I-IoT worldwide. However, the contrary indication occurred in Indonesia. Among developing countries, the spending level of IoT in Indonesia is significant, but the use level of the technology is less developed. This survey study purposed to predict what factors influence the behavior intention to use C-IoT. The researchers extended the unified theory of acceptance and use of technology (UTAUT) model by adopting the network externality aspects. Around 400 valid data were collected from urban communities in the six most populous provinces in the country. The scholars used the partial least squares structural equation modeling (PLS-SEM) method using SmartPLS 3.3 in the data analysis stage. The findings expressed that the number of users and social influence factors are not influential factors influencing behavior intention to use IoT. Besides that, the UTAUT model extension may also be one of the theoretical references for future similar studies. Mass spectrometry (MS)-based lipidomic has become a powerful tool for studying lipids in biological systems. However, lipidome analysis at the single-cell level remains a challenge. Here, we report a highly sensitive lipidomic workflow based on nanoflow liquid chromatography and trapped ion mobility spectrometry (TIMS)-MS. This approach enables the high-coverage identification of lipidome landscape at the single-oocyte level. By using the proposed method, comprehensive lipid changes in porcine oocytes during their maturation were revealed. The results provide valuable insights into the structural changes of lipid molecules during porcine oocyte maturation, highlighting the significance of sphingolipids and glycerophospholipids. The damage and fracture of coal is accompanied by a complex energy conversion process, and these different stages of energy evolution are closely related to coal failure. In this paper, an evolution model describing the behavior of coal failure was proposed using the energy dissipation under cyclic loading/unloading. The energy growth pattern and energy consumption characteristics of the coal fracture were analyzed under cyclic loading/unloading. An evolution model of the energy behavior of coal fracture was established. The damage variables of energy dissipation were defined, and a theoretical model was established. The parameters included the relationship between the energy state, damage state, and strength state according to the uniaxial cyclic loading/unloading test. The results show that there are energy excitation and inhibition effects in the process of coal fracture; that is, the accumulation rate and level of energy are affected by the energy storage state, and the energy storage rate changes in the mode of "low promotion and high inhibition". The abrupt increase in dissipated energy can be regarded as the precursor of coal fracture. Based on the analysis of the characteristics of the damage and failure state and dissipated energy, the discriminant equation for the stability of the coal energy state was constructed; it is a meaningful discovery for predicting and evaluating coal failure. Brittle failure, also known as rockburst, is a violent phenomenon that occurs during the excavation of hard rock in areas with high geostress, particularly at great depths. The induced hazards have been a global concern since the first half of the 20th century, leading to extensive efforts to understand the mechanisms and develop effective approaches to predict and control such rock mass failures. To understand the current state of research and future prospects of rockburst, a comprehensive review of past developments has been conducted. Firstly, we briefly summarize different definitions and types of rockburst. Next, we delve into the various technologies and methods employed in rockburst research and analyze the current knowledge on the mechanisms, classifications, and prediction of rockbursts. Then, we examine the existing methods for preventing and controlling these events. Despite the advancements made, the relationship between the behavior of rock masses at different scales, both in the laboratory and real-world conditions, remains an area of challenge. Additionally, the absence of rock mass classifications under high in-situ stress and lack of accuracy in rockburst prediction methods remain persistent issues that need to be addressed. This study delves into cutting-edge rockburst research, illuminating potential ground support strategies and empowering the mining industry in order to unleash innovative solutions for controlling these explosive phenomena. The stability of the cemented paste backfill roof (CPB roof) is critical to safe production in mines using the underhand drift cut-and-fill stopping. To investigate the scientific and reasonable design method of key parameters (size and strength) of the CPB roof and the stress state of the CPB roof during the mining process, field measurements were carried out with Jinchuan Group's third mining area as the engineering background. Based on the measurement results, a mechanics model was constructed based on the thick plate theory. The field measurement results show that the overlying load on the CPB roof tends to increase first and then decrease with the gradual mining of the stope, and the maximum overlying load values of the two CPB roofs measured are 0.240 MPa and 0.244 MPa, respectively. With the gradual mining of the stope, the deformation of the CPB roof shows a trend of increasing first and then stabilizing. Based on the thick plate theory, the stress model of the CPB roof is constructed, and the error between the calculation results of the model and the field measurement results does not exceed 5%. Applying the research results to the three mines of Jinchuan Group, the span of the stope can be expanded from 5 m to 6 m under the existing strength standard of the filling body, which can increase its mining capacity by 20%. Combined mining is when two working faces separated by a certain distance simultaneously advance in the same direction, which commonly results in stress concentration and rockburst. This paper investigates the rockburst mechanism due to inclined coal seam combined mining under complex geological conditions in the Da'anshan Coal Mine, China. Based on the experimental analysis, there is an impact tendency of the No.10 coal seam. A theoretical model is established to evaluate rockburst risk due to combined mining. In addition, a 3D multifactor numerical model of inclined coal seam combined mining is established. The novel "PyFlac-Exc" calculation code and the "PyFlac-Eng" elastic strain energy calculation script are compiled. The evolution of elastic strain energy is obtained during combined mining. Theoretical and numerical results show that a rockburst occurs in the western fourth working face of the No. 10 coal seam, which is consistent with the on-site rockburst accident. In addition, the kernel density estimation method is proposed to reduce stress concentration by changing the coal pillar widths and misalignment distances. Numerical results show that the elastic strain energy levels are low for the same coal pillar width, and the misalignment distance is arranged from 50 to 80 m. With the same misalignment distance, a coal pillar width greater than 45 m can effectively reduce the rockburst risk. [ABSTRACT FROM AUTHOR]

Lontar is a palm-leaf or lontar-based manuscript containing a Balinese script passed down from generation to generation. Lontar is derived from cellulose- rich palm leaf, which can serve as a substrate for the growth of fungi producing cellulase enzymes. Nevertheless, the activity of these fungi could potentially damage the lontar itself. The objective of this study is to identify the fungi that contaminated the Balinese lontar. The fungus was isolated from several Lontar storage areas in Balinese society using swabs. In addition, the fungus was cultivated on PDA media prior to isolation and identification. Seven lontar-degrading fungus isolates, including Penicillium restricum, Aspergillus fumigatus, Mucor racemosus, Candida krusei, A. niger, Fusarium sp., and Rhodotorula mucilaginosa, have been identified as the primary causes of damage to the Balinese lontar. A gas outburst risk level prediction method, based on the Whale Optimization Algorithm (WOA) Improved Extreme Learning Machine (ELM), is proposed to predict the coal and gas outburst hazard level more accurately. Based on this method, recommendations are given according to the gas outburst risk level with the help of the Case-Based Reasoning (CBR) method. Firstly, we analyze the accident reports of gas outburst accidents, select the gas outburst risk prediction index, and construct the gas outburst risk prediction index system by combining the gas outburst prevention and control process. The WOA-ELM model was used to predict the gas outburst risk level by selecting data from 150 accident reports from 2008 to 2021. Again, based on the coal and gas outburst risk level, CBR is used to match the cases and give corresponding suggestions for different levels of gas outburst risk conditions to help reduce the gas outburst risk. The results show that the WOA-ELM algorithm has better performance and faster convergence than the ELM algorithm, when compared in terms of accuracy and the error of gas outburst hazard prediction. Purpose. A methodology development for predicting the geomechanical situation when mining an ore deposit with steepdipping layers, taking into account the uncertainty in determining the rock properties, which is a consequence of the rock mass heterogeneity. Methods. The assessment of the open-pit wall stability is based on a combination of numerical simulation of the rock stress-strain state (SSS) and probabilistic analysis. The finite element method is used to determine the changes in the SSS that occur at various stages of mining operations due to design changes in the overall open-pit slope angle. The elastic-plastic model of the medium and the Mohr-Coulomb failure criterion are implemented in the codes of the 3D finite element analysis program RS3 (Rocscience). Stochastic simulation is used to assess random risks associated with natural object state variations. Findings. The distribution of maximum shear strains, which localizes the real or potential sliding surfaces in the open-pit wall at various stages of ore mining, has been identified. Based on the Shear Strength Reduction procedure, the open-pit wall Strength Reduction Factor (SRF) has been determined. The probabilities of open-pit wall stability loss, as well as the decrease in the strength reduction factor below the standard level at all stages of the ore body mining, have been revealed. Originality. For the first time, for real mining-geological conditions of a deep ore open pit, the dependence of the strength reduction factor on the overall wall slope angle, which changes during mining of each steep layer, has been determined. For each stage of mining operations, for the first time, the probability of a decrease in the open-pit wall stability below the standard level has been determined based on stochastic simulation. Practical implications. The ratio between the open-pit contour characteristic (overall slope angle) and the probabilistic safety factor is the basis for practical solutions to ensure the efficiency and safety of mining at various stages of friable and hard overburden excavation, ore extraction, as well as for the subsequent optimization of the open-pit design contours. Polycystic ovary syndrome (PCOS) is a hormonal disorder that affects women of reproductive age. It is characterised by several clinical signs and symptoms, including hyperandrogenism, ovarian dysfunction, and chronic anovulation. Obesity and hormonal imbalances can be reduced in PCOS by modifying lifestyle habits. The effects of lifestyle changes on PCOS were investigated, and it was found that consuming less fat and engaging in physical activity reduced metabolic disturbances. The purpose of this study was to examine the effects of lifestyle modifications on metabolic parameters, anthropometric parameters, and the hirsutism score as they relate to the pathogenesis of PCOS. This study involved a total of 10 patients diagnosed with PCOS at the Sultan Agung Semarang Islamic Hospital. Blood glucose, triglycerides, HDL (high density lipoprotein), blood pressure, waist circumference, hirsutism score, and vital sign data related to PCOS were measured before and after a 4-month treatment. PCOS patients' fasting blood sugar levels, triglycerides (TG), high density lipoprotein (HDL), hirsutism scores, blood pressure (systolic and diastolic), and waist circumference averaged 92, 60; 48.10; 96.90; 10.5; 121.5/77; 86.80, respectively, at baseline (pretreatment). After four months of treatment, the average levels of fasting blood sugar, triglycerides, high-density lipoproteins, blood pressure (systolic and diastolic), hirsutism scores, and waist circumference were 108.7, 50, 99.8, 7, 122.3, 82.7, and 87.50, respectively. Changes in lifestyle led to significant variations in fasting blood sugar levels as a metabolic parameter and hirsutism scores, but not in triglycerides, high-density lipoprotein, waist circumference, or blood pressure. The study investigates the effect of the Balanced Scorecard, one of the performance measurement systems, on employee well-being (EWB) in organizations. As a result of the literature research, it is seen that employee welfare is examined in 3 different areas: Job Satisfaction, Organizational Commitment, and Job Engagement. One of the most important factors for the success of an organization is the happy employees who contribute tremendously to the improvement of the organization's performance. Both qualitative and quantitative measurement methods were used for this study. The surveys that have used for this study included 27334 employees in the state of Michigan and the surveys data were analyzed by Statistical Package for Social Sciences (SPSS) 28. The study revealed that the BSC strategies implemented in the State of Michigan improved organizational performance in the four perspectives of financial, customer, internal process, and learning & growth. The statistical results of employee satisfaction surveys show that BSC increases job satisfaction by 68.30% (R-squared: 0.683), work engagement by 71.00% (R-squared: 0.710), and organizational commitment by 21.30% (R-squared: 0.213). With the BSC strategy and learning-growth perspective that has been in effect for 20 years, employee satisfaction has been increased, organizational commitment and job engagement have been prioritized in the State of Michigan. Background: Embryo production in vitro requires three steps: in vitro maturation (IVM), in vitro fertilization (IVF) and in vitro culture (IVC). The first step in the in vitro maturation of oocytes is germinal vesicle breakdown (GVBD), followed by completion of the 1st meiotic division and formation of the 1st polar body. These parameters are critical during ovary and oocyte selection. This study aimed to evaluate bovine ovary and oocyte collection for IVM. Methods: Ovaries collected from cows in a 0.9% NaCl saline solution at the central slaughterhouse were divided into two normal and abnormal ovaries according to their morphological appearance. The morphometric dimensions of bovine ovaries, such as weight and deamination (length, width and volume), were recorded. The oocyte was extruded and its deamination occurred before and after culture in maturation medium. Result: The mean ovary weight, volume, length and width were not significantly different between the two ovary types. Additionally, the dominant and subordinate follicle diameters in both ovary types showed no significant differences. Furthermore, the oocyte number per ovary of the normal and abnormal oocytes showed no significant differences. The mean cumulus oocyte complex before maturation showed no significant difference (52.73±7.23 mm for the normal ovary vs. 43.015±5.41 mm for the abnormal ovary). However, after maturation, a highly significant difference was found (P<0.001) between the normal ovaries before and after maturation (178.10±15.36 µm) and abnormal ovaries (10.45±7.99 µm). Additionally, data analysis of oocytes with or without 1st polar bodies revealed a highly significant difference (P<0.001) between oocytes of the normal ovary with (32.15±4.19) and without (10.95±1.59) 1st polar bodies and oocytes of the abnormal ovary with (7.0±078) and without 1st polar bodies (3.3±0.32). Thus, the critical point at which the normal ovary produces better in vitro matured follicles with good oocyte quality and produces the 1st polar body determines developmental competence. Therefore, the best selection of normal ovaries will enhance in vitro maturation for subsequent experiments, such as in vitro fertilization or cloning and in vitro embryo development. Coal burst prediction is an important research hotspot in coal mine production safety. This paper presents FDNet, which is a knowledge and data fusion-driven deep neural network for coal burst prediction. The main idea of FDNet is to extract explicit features based on the existing mine seismic physical model and utilize deep learning to automatically extract the implicit features of mine microseismic data. The key innovations of FDNet include an expert knowledge indicator selection method based on a subset search strategy, a mine microseismic data extraction method based on a deep convolutional neural network, and a feature deep fusion method of mine microseismic data based on an attention mechanism. We conducted a set of engineering experiments in Gaojiapu Coal Mine to evaluate the performance of FDNet. The results show that compared with the state-of-the-art data-driven machines and knowledge-driven methods, the prediction accuracy of FDNet is improved by 5% and 16%, respectively. In mammalian species an optimal fertilization window during which successful fertilization occurs. In the majority of mammals estrus marks ovulation time and coincident with mating, thereby allowing the synchronized meeting in the fallopian tubes, between freshly ejaculated sperm and freshly ovulated oocytes. Conversely, women do not show natural visual signs of ovulation such that fertilization can occur hours later involving an aged oocyte and freshly ejaculated spermatozoa. During this time, the oocyte undergoes a rapid degradation known as "postovulatory aging" (POA). POA may become particularly important in the human-assisted reproductive technologies, as the fertilization of retrieved mature oocytes can be delayed due to increased laboratory workload or because of unforeseeable circumstances, like the delayed availability of semen samples. This paper is an updated review of the consequences of POA, either in vivo or in vitro, on oocyte quality with particular attention to modifications caused by POA on oocyte nuclear, cytoplasmic, genomic, and epigenetic maturation, and embryo development. Fertility preservation through gamete vitrification has become one of the critical strategies to secure a childbearing potential in patients who are diagnosed with cancer or risks of infertility. Preserving the gametes would prevent the deleterious effects of cancer drugs or radiotherapy exposure on the quality of the gametes. Furthermore, in vitro fertilisation of vitrified mature human oocytes has lately demonstrated promising results that are reflected in the increased survival rate of thawed oocytes and the resultant clinical pregnancy rate. However, limitations in the cryopreservation of mature oocytes of cancer patients persist. Ovarian stimulation protocols which comprise administering gonadotrophin-releasing hormones could aggravate cancer or delay essential cancer therapy. Considering such circumstances, vitrification of immature oocytes would become a rational option. While the vitrification procedure of mature oocytes has been established, the vitrification of immature oocytes remains controversial due to a low post-thaw in vitro maturation and fertilisation rate. Apparent cryoinjuries to the immature oocytes post thawing or warming have been observed in both human and animal model oocytes. An alternative strategy was therefore proposed to improve the effectiveness of utilising immature oocytes for fertility preservation by conducting the in vitro oocyte maturation process first before vitrification. This method has prevailed, especially in oncofertility patients. Although the success rate of the clinical outcomes remains low, this approach, in conjugation with proper counselling, might provide oncofertility patients with an opportunity to preserve their reproductive potential. Rockburst is a kind of complex and catastrophic dynamic geological disaster in the development and utilization of underground space, which seriously threatens the safety of personnel and environment. Due to the suddenness in time and randomness in space, the prediction of rockburst becomes a great challenge. Microseismic monitoring is capable to continuously capture rock microfracture signals in real time, which offers an effective means for rockburst prediction. With the explosive growth of monitoring data, the conventional manual forecasting methods are laborious and time-consuming. Therefore, artificial intelligence was introduced to improve prediction efficiency. A novel tree-based algorithm was proposed. Its basic idea was to automatically recognize precursory microseismic sequences for the real-time prediction of rockburst intensity. The database consisting of 1500 microseismic events was analyzed. To establish precursory microseismic sequences, dimensionality reduction of the database was first implemented by t-SNE algorithm. Then, k-means clustering algorithm was employed for labelling 1500 microseismic events. Before that, canopy algorithm was adopted to determine the number of clusters. Finally, 300 precursory microseismic sequences were formed by the grouping rule. They were further partitioned into two parts through stratified sampling: 70% for training and 30% for validation. The validation results indicated that the precursor tree with pruning achieved a higher prediction accuracy of 98.9% than one without pruning on the validation set. And the increase was separately 12.2%, 9.2% and 28.6% on the whole validation set and each classes (low/moderate rockburst). In comparison with low rockburst, moderate rockburst was minority class. The improved accuracy on moderate rockburst suggested that pruning can enhance the recognition ability of precursor tree for the minority class. Additionally, two extra rockburst cases were collected from a diversion tunnel in northwestern China, which provided a complete workflow about how to apply the built precursor tree model to achieve field rockburst warning in engineering practice. The tree-based algorithm served as a new and promising way for the real-time rockburst prediction, which successfully integrated field microseismic monitoring and artificial intelligence. During a survey of oomycetes in ornamental plants carried out at the University of Aberdeen in 2014–2015, Pythium kashmirense was isolated from a specimen of Viburnum plicatum ‘Lanarth’, the first report of this oomycete in the UK (and in Europe). Pathogenicity of a Py. kashmirense isolate was examined using a range of plant species. Inoculations were carried out under controlled conditions in the absence of other Pythium and Phytophthora species, on Glycine max (soya bean), Phaseolus vulgaris (common bean), Lupinus angustifolius (blue lupin), Cucumis sativa (cucumber) and Viburnum opulus. The majority of inoculations caused pre-emergence damping-off, as well as seed rot and root rot. In the in vitro assays, germination rates (%) of soya bean and blue lupin seeds were less than 50%; in the in vivo inoculations on plants, over 50% of soya bean, blue lupin and common bean plants died; in contrast, cucumber plants showed lower susceptibility in pathogenicity tests, with an approximately 80% germination rate in in vitro tests, and 25% dead plants in the in planta inoculations. Inoculations carried out on root systems of Viburnum opulus caused severe necrosis and root rot. Little research was previously conducted on pathogenicity of Py. kashmirense and its relationship with losses in crop yield and quality. The present study showed varying virulence on the different plant species tested after inoculation with Py. kashmirense. Despite the lack of clear host specialization, infection by Py. kashmirense decreased seedling survival and health of plants in a range of important agricultural and ornamental plant species.
