Lead telluride and germanium telluride are well-known IV-VI semiconductors, which is now the focus of research due to the perspective of application as thermoelectrics for midrange temperatures. Solid solutions and heterostructures on this basis, obtained by molecular beam epitaxy, are a promising direction for the development of these materials. In this paper, we have focused on the Raman spectra excited by the 514.5 nm laser line (out of resonance) of PbTe, GeTe, (Pb, Ge)Te, and (Pb, Ge, Eu)Te layers grown on BaF2 (111) monocrystalline substrates. The obtained phonon properties are related to the properties of the corresponding bulk materials or can be explained by a model that takes into account the difference in the masses of the constituent elements only, as is the case with the local mode of Ge in PbTe (registered at about 181 cm−1). Multiphonon processes registered for this phonon are a consequence of the change in the electronic structure of PbTe and electron-phonon interaction. This paper presents an overview for the Special Issue of Clean Technology and Environmental Policy journal (CTEP), and it includes accepted papers from: 5th South East European Conferences on Sustainable Development of Energy, Water and Environment Systems (SDEWES) held from May 22–26, 2022, in Vlore, Albania. 3rd Latin American Conferences on Sustainable Development of Energy, Water and Environment Systems (SDEWES) held from July 24–28, 2022, in Sao Paulo, Brazil. 17th Conferences on Sustainable Development of Energy, Water and Environment Systems (SDEWES) held from November 6–10, 2022, in Paphos, Cyprus. Considering CTEP's policy of high-quality research papers, guest editors have invited 35 research articles, presented at the SDEWES 2022 conference. After a vigorous review process, 13 papers have been accepted for publication in this special issue. All 13 accepted papers are briefly presented in this overview together with a wider view that presents research efforts within the SDEWES community published through previous SDEWES special issues. The current applications in the energy sector are based largely on fossil fuels which release greenhouse gas emissions to the atmosphere. To face the issue of global warming, the energy sector has to transfer to and develop sustainable energy solutions that do not release carbon emissions. This is one of the primary motivators for the SDEWES conference as well as for this review, and previous ones, examining the most recent works based on sustainable and green energy production in such fields. The 17th Conference on the Sustainable Development of Energy, Water, and Environment Systems (SDEWES) was held on 6–10 November 2022 in Paphos, Cyprus. The SDEWES conference aims at solving complex and ongoing concerns that approach a long-term perspective and supporting innovative solutions and continuous monitoring and evaluation. This review paper aims at collecting the main presented papers focused on the following hot topics: low-carbon technologies based on renewable and clean-energy systems, including mainly biomass, solar, and wind energy applications; energy storage systems; hydrogen-based systems; energy-saving strategies in buildings; and the adoption of smart management strategies using renewable energy systems. These topics are investigated in order to propose solutions to address the issues of climate change, water scarcity, and energy saving. From the analyzed works, we note that some key issues for sustainable development remain to be further addressed: such as novel and advanced energy storage systems, green hydrogen production, novel low-temperature district heating and cooling networks, novel solar technologies for the simultaneous production of power and high temperature heat, solar desalination for hydrogen production systems, and agrivoltaic systems for the production of power and food. This paper presents the results of ceramic synthesis in the field of a powerful flux of high-energy electrons on powder mixtures. The synthesis is carried out via the direct exposure of the radiation flux to a mixture with high speed (up to 10 g/s) and efficiency without the use of any methods or means for stimulation. These synthesis qualities provide the opportunity to optimize compositions and conditions in a short time while maintaining the purity of the ceramics. The possibility of synthesizing ceramics from powders of metal oxides and fluorides (MgF2, BaF2, WO3, Ga2O3, Al2O3, Y2O3, ZrO2, MgO) and complex compounds from their stoichiometric mixtures (Y3Al3O12, Y3AlxGa(5−x) O12, MgAl2O4, ZnAl2O4, MgWO4, ZnWO4, BaxMg(2−x) F4), including activators, is demonstrated. The ceramics synthesized in the field of high-energy electron flux have a structure and luminescence properties similar to those obtained by other methods, such as thermal methods. The results of studying the processes of energy transfer of the electron beam mixture, quantitative assessments of the distribution of absorbed energy, and the dissipation of this energy are presented. The optimal conditions for beam treatment of the mixture during synthesis are determined. It is shown that the efficiency of radiation synthesis of ceramics depends on the particle dispersion of the initial powders. Powders with particle sizes of 1–10 µm, uniform for the synthesis of ceramics of complex compositions, are optimal. A hypothesis is put forward that ionization processes, resulting in the radiolysis of particles and the exchange of elements in the ion–electron plasma, dominate in the formation of new structural phases during radiation synthesis. Lung cancer remains the leading cause of cancer-related mortality worldwide, with non-small cell lung cancer (NSCLC) constituting 85% of cases. Among NSCLCs, squamous cell carcinoma (SqCC) is strongly associated with smoking. However, lung cancer in never smokers (LCINS) represents approximately 25% of lung cancer cases globally and shows increasing incidence, particularly in East Asia. LCINS-SqCC is less well-characterized, especially regarding its genomic alterations and their impact on clinical outcomes. We conducted a retrospective analysis over a 20-year period (July 2003–July 2023) at two major tertiary centers in the UK. The cohort included 59 patients with LCINS-SqCC who underwent radical surgical resection. Data collected included demographic information, comorbidities, histopathological details, and outcome metrics such as disease-free and overall survival. Molecular sequencing of tumor specimens was performed to identify genomic aberrations. The cohort had a median age of 71 years (IQR 62–77) and a median BMI of 25.4 (IQR 22.8–27.8), with a slight male predominance (53%). The majority of patients (93%) had a preoperative MRC of 1–2. Recurrent disease was observed in 23 patients (39%), and 32 patients (54%) had died at a median follow-up of 3 years. Median disease-free survival was 545 days (IQR 132–1496), and overall survival was 888 days (IQR 443–2071). Preoperative creatinine levels were higher in patients who experienced recurrence (p = 0.037). Molecular analysis identified biallelic SMARCB1 loss in two younger patients, associated with rapid disease progression despite R0 resection. These patients' tumors were PDL1-negative, TTF-1-negative, and positive for cytokeratin, CD56, and p40. SMARCB1-deficient SqCC in never smokers represents a highly aggressive variant with poor disease-free survival, highlighting the importance of integrating advanced molecular diagnostics in clinical practice. This study underscores the necessity for personalized treatment strategies, including targeted therapies such as EZH2 inhibitors and immune checkpoint blockade, to address the unique molecular pathways in SMARCB1-deficient cancers. In this work, we present the results of the analysis performed with near edge absorption fine structure spectroscopy of barium edges in a B+BaF2 optical filter. The radiation was generated by a laser-plasma soft X-ray source based on a double-stream gas puff target. The use of a flat-field spectrometer based on a diffraction grating allowed us to collect the absorption spectra of the radiation transmitted by the analyzed optical filter. Biological aerated filters (BAF) are attached growth aerobic systems used for the treatment of wastewater. This system can establish the physical filtration of solids along with the biological decomposition of organic matter. The performance of the BAF depends on the type of support media and its ability to meet objective of the treatment process. Natural zeolites, with properties such as ion exchange, larger surface area, capability to form biofilm, adsorption, regenerative properties etc., are a promising filter material in BAF systems. This paper investigates and discusses the effect of various factors like hydraulic retention time (HRT), carbon to nitrogen (C/N) ratio, aeration rate, depth of filter and particle size of natural zeolite which influence the performance of BAF. Based on the experimental results and optimization study using response surface methodology, carried out on the BAF system with varying process variables like HRT, C/N ratio, aeration rate and depth of filter, the natural zeolite media with particle size 1-3 mm range showed improved performance as compared to the coarser particles, that is, 3-5 mm and 5-10 mm. On conducting a final run using natural zeolite particle size of 1-3 mm under optimized value of HRT = 3.25 h, C/N = 6.2, aeration rate = 69 L/h, depth of filter = 83 cm, the BAF system observed chemical oxygen demand, biological oxygen demand, NH3-N, turbidity and total dissolved solids removal percentage of 97%, 96%, 99%, 98% and 58%, respectively. The mammalian SWI/SNF-like BAF complexes play critical roles during animal development and pathological conditions. Previous gene deletion studies and characterization of human gene mutations implicate that the complexes both repress and activate a large number of genes. However, the direct function of the complexes in cells remains largely unclear due to the relatively long-term nature of gene deletion or natural mutation. Here we generate a mouse line by knocking in the auxin-inducible degron tag (AID) to the Smarca4 gene, which encodes BRG1, the essential ATPase subunit of the BAF complexes. We show that the tagged BRG1 can be efficiently depleted by osTIR1 expression and auxin treatment for 6 to 10 h in CD4 + T cells, hepatocytes, and fibroblasts isolated from the knock-in mice. The acute depletion of BRG1 leads to decreases in nascent RNAs and RNA polymerase II binding at a large number of genes, which are positively correlated with the loss of BRG1. Further, these changes are correlated with diminished accessibility at DNase I Hypersensitive Sites (DHSs) and p300 binding. The acute BRG1 depletion results in three major patterns of nucleosome shifts leading to narrower nucleosome spacing surrounding transcription factor motifs and at enhancers and transcription start sites (TSSs), which are correlated with loss of BRG1, decreased chromatin accessibility and decreased nascent RNAs. Acute depletion of BRG1 severely compromises the Trichostatin A (TSA) -induced histone acetylation, suggesting a substantial interplay between the chromatin remodeling activity of BRG1 and histone acetylation. Our data suggest BRG1 mainly plays a direct positive role in chromatin accessibility, RNAPII binding, and nascent RNA production by regulating nucleosome positioning and facilitating transcription factor binding to their target sites. Brazil's Atlantic Forest (BAF) is a highly fragmented, strategic environmental and socio-economic region that represents the fourth biodiversity hotspot while also producing many commodities that are exported globally. Human disturbance plays a pivotal role as a driver of BAF's soil dynamics and behaviors. The soils under Late Primary and Secondary Semideciduous Seasonal Forests (LPSF and LSSF) were characterized by high to moderate resilience, with improved chemical properties as human disturbance decreased. The Transitional Forest to Cerrado (TFC) had the worst soil conditions. Disturbed Primary and Secondary Semideciduous Seasonal Forests (DPSF and DSSF) represent a transitional stage between LPSF/LSSF and TFC. Accordingly, SOCs stocks increased from TFC << DPSF, DSSF < LPSF, LSSF. In BAF soils, to avoid unreliable data, SOCs measurements should be (i) conducted to at least 1 m soil depth and (ii) quantified with a CHN analyzer. Human disturbance strongly affected the positive feedback between vegetation succession, SOCs, and soil nutrition. Soil development decreased as human disturbance increased, thus negatively affecting SOCs. Soils in the BAF require a long time to recover after the end of human disturbance, thus suggesting that preservation strategies should be prioritized in remnant BAF fragments. We present sub-50 fs soliton pulse generation from a diode-pumped Kerr-lens mode-locked laser based on an Yb3+-doped BaF2 crystal. Utilizing a spatially single-mode, fiber-coupled InGaAs laser diode at 976 nm as a pump source, the Yb:BaF2 laser generates pulses as short as 46 fs at 1060.1 nm with an average output power of 45 mW at a pulse repetition rate of ~65.6 MHz via soft-aperture Kerr-lens mode locking. To the best of our knowledge, this represents the first demonstration of Kerr-lens mode-locked operation of the Yb:BaF2 crystal, as well as the shortest pulse duration ever achieved from any diode-pumped mode-locked laser based on an Yb3+-doped alkaline-earth fluoride crystal. Cancer development is a complex process involving both genetic and epigenetic changes. The SWI/SNF (switch/sucrose non‐fermentable) chromatin remodelling complex, one of the most studied ATP‐dependent complexes, plays an important role in coordinating chromatin structural stability, gene expression and post‐translational modifications. The SWI/SNF complex can be classified into BAF, PBAF and GBAF according to their constituent subunits. Cancer genome sequencing studies have shown a high incidence of mutations in genes encoding subunits of the SWI/SNF chromatin remodelling complex, with abnormalities in one or more of these genes present in nearly 25% of all cancers, which indicating that stabilizing normal expression of genes encoding subunits in the SWI/SNF complex may prevent tumorigenesis. In this paper, we will review the relationship between the SWI/SNF complex and some clinical tumours and its mechanism of action. The aim is to provide a theoretical basis to guide the diagnosis and treatment of tumours caused by mutations or inactivation of one or more genes encoding subunits of the SWI/SNF complex in the clinical setting. The SWI/SNF chromatin remodeling complex consists of more than ten component proteins that form a large protein complex of >1 MDa. The catalytic proteins Smarca4 or Smarca2 work in concert with the component proteins to form a chromatin platform suitable for transcriptional regulation. However, the mechanism by which each component protein works synergistically with the catalytic proteins remains largely unknown. Here, we report on the function of Smarce1, a component of the SWI/SNF complex, through the phenotypic analysis of homozygous mutant embryonic stem cells (ESCs). Disruption of Smarce1 induced the dissociation of other complex components from the SWI/SNF complex. Histone binding to DNA was loosened in homozygous mutant ESCs, indicating that disruption of Smarce1 decreased nucleosome stability. Sucrose gradient sedimentation analysis suggested that there was an ectopic genomic distribution of the SWI/SNF complex upon disruption of Smarce1, accounting for the misregulation of chromatin conformations. Unstable nucleosomes remained during ESC differentiation, impairing the heterochromatin formation that is characteristic of the differentiation process. These results suggest that Smarce1 guides the SWI/SNF complex to the appropriate genomic regions to generate chromatin structures adequate for transcriptional regulation. [ABSTRACT FROM AUTHOR]

Prior to the next generation sequencing and characterization of the tumor genome landscape, mutations in the SWI/SNF chromatin remodeling complex and the KEAP1-NRF2 signaling pathway were underappreciated. While these two classes of mutations appeared to independently contribute to tumor development, recent reports have demonstrated a mechanistic link between these two regulatory mechanisms in specific cancer types and cell models. In this work, we expand upon these data by exploring the relationship between mutations in BAF and PBAF subunits of the SWI/SNF complex and activation of NRF2 signal transduction across many cancer types. ARID1A/B mutations were strongly associated with NRF2 transcriptional activity in head and neck squamous carcinomas (HNSC). Many additional tumor types showed significant association between NRF2 signaling and mutation of specific components of the SWI/SNF complex. Different effects of BAF and PBAF mutations on the polarity of NRF2 signaling were observed. Overall, our results support a context-dependent functional link between SWI/SNF and NRF2 mutations across human cancers and implicate ARID1A inactivation in HPV-negative HNSC in promoting tumor progression and survival through activation of the KEAP1-NRF2 signaling pathway. Purpose: Acute lung injury (ALI) is a severe pulmonary disease characterized by damage to the alveoli and pulmonary blood vessels, leading to severe impairment of lung function. Studies on the effect of capsaicin (8-methyl-N-geranyl-6-nonamide, CAP) on lipopolysaccharide (LPS)-induced ALI in bronchial epithelial cells transformed with Ad12-SV40 2B (BEAS-2B) are still limited. This study aimed to investigate the effect and specific mechanism by which CAP improves LPS-induced ALI.Methods: The present study investigated the effect of CAP and the potential underlying mechanisms in LPS-induced ALI in vitro and vivo via RNA sequencing, Western blotting (WB), quantitative real-time reverse transcription PCR (qRT‒PCR), enzyme-linked immunosorbent assay (ELISA), and transmission electron microscopy (TEM). The TRPV1 inhibitor AMG9810 and the AKT agonist SC79 were used to confirm the protective effect of the TRPV1/AKT axis against ALI. The autophagy agonist rapamycin (Rapa) and the autophagy inhibitors 3-methyladenine (3-MA) and bafilomycin A1 (Baf-A1) were used to clarify the characteristics of LPS-induced autophagy.Results: Our findings demonstrated that CAP effectively suppressed inflammation and autophagy in LPS-induced ALI, both in vivo and in vitro. This mechanism involves regulation by the TRPV1/AKT signaling pathway. By activating TRPV1, CAP reduces the expression of P-AKT, thereby exerting its anti-inflammatory and inhibitory effects on pro-death autophagy. Furthermore, prior administration of CAP provided substantial protection to mice against ALI induced by LPS, reduced the lung wet/dry ratio, decreased proinflammatory cytokine expression, and downregulated LC3 expression.Conclusion: Taken together, our results indicate that CAP protects against LPS-induced ALI by inhibiting inflammatory responses and autophagic death through the TRPV1/AKT signaling pathway, presenting a novel strategy for ALI therapy. Fast and heavy inorganic scintillators with suitable radiation tolerance are required to face the challenges presented at future hadron colliders of high energy and intensity. Up to 5 GGy and 5 × 1018 neq/cm2 of one-MeV-equivalent neutron fluence is expected by the forward calorimeter at the Future Hadron Circular Collider. This paper reports the results of an investigation of proton- and neutron-induced radiation damage in various fast and heavy inorganic scintillators, such as LYSO:Ce crystals, LuAG:Ce ceramics, and BaF2 crystals. The experiments were carried out at the Blue Room with 800 MeV proton fluence up to 3.0 × 1015 p/cm2 and at the East Port with one MeV equivalent neutron fluence up to 9.2 × 1015 neq/cm2, respectively, at the Los Alamos Neutron Science Center. Experiments were also carried out at the CERN PS-IRRAD proton facility with 24 GeV proton fluence up to 8.2 × 1015 p/cm2. Research and development will continue to develop LuAG:Ce ceramics and BaF2:Y crystals with improved optical quality, F/T ratio, and radiation hardness. [ABSTRACT FROM AUTHOR]

ATP-dependent chromatin remodeling SWI/SNF complexes exist in three subcomplexes: canonical BAF (cBAF), polybromo BAF (PBAF), and a newly described non-canonical BAF (ncBAF). While cBAF and PBAF regulate fates of multiple cell types, roles for ncBAF in hematopoietic stem cells (HSCs) have not been investigated. Motivated by recent discovery of disrupted expression of BRD9, an essential component of ncBAF, in multiple cancers, including clonal hematopoietic disorders, we evaluate here the role of BRD9 in normal and malignant HSCs. BRD9 loss enhances chromatin accessibility, promoting myeloid lineage skewing while impairing B cell development. BRD9 significantly colocalizes with CTCF, whose chromatin recruitment is augmented by BRD9 loss, leading to altered chromatin state and expression of myeloid-related genes within intact topologically associating domains. These data uncover ncBAF as critical for cell fate specification in HSCs via three-dimensional regulation of gene expression and illuminate roles for ncBAF in normal and malignant hematopoiesis. BRD9 is a core non-canonical BAF component. Here the authors show that BRD9 plays a pivotal role in regulating the disease-related cell fate of hematopoietic stem cells. Malignant rhabdoid tumor (MRT) is a highly malignant and often lethal childhood cancer. MRTs are genetically defined by bi-allelic inactivating mutations in SMARCB1, a member of the BRG1/BRM-associated factors (BAF) chromatin remodeling complex. Mutations in BAF complex members are common in human cancer, yet their contribution to tumorigenesis remains in many cases poorly understood. Here, we study derailed regulatory landscapes as a consequence of SMARCB1 loss in the context of MRT. Our multi-omics approach on patient-derived MRT organoids reveals a dramatic reshaping of the regulatory landscape upon SMARCB1 reconstitution. Chromosome conformation capture experiments subsequently reveal patient-specific looping of distal enhancer regions with the promoter of the MYC oncogene. This intertumoral heterogeneity in MYC enhancer utilization is also present in patient MRT tissues as shown by combined single-cell RNA-seq and ATAC-seq. We show that loss of SMARCB1 activates patient-specific epigenetic reprogramming underlying MRT tumorigenesis. The regulatory landscape of malignant rhabdoid tumor (MRT) due to SMARCB1 loss remains to be explored. TeO2-BaF2-Er2O3-Dy2O3 laser glasses were prepared using the melt-quenching method. The bound water that can capture the excited state energy was reduced by physical and chemical methods. We did not observe a significant Er3+ emission peak at 2.7 μm in fluorescence spectra, which may be due to the efficient energy transfer process (ET2). Meanwhile, we found a broadband gain span of approximately 400 nm in fluorescence spectra at the 2.85 μm band, attributed to the 'vector summation' of the energy level radiation transition and the change of the glass network. Subsequently, we explored the structural properties of the glass. The results indicated that the Gaussian peak located at 250 cm−1 drifts toward 370 cm−1, which may be caused by the fracture or recombination of Te-O-Te and a decrease in the bridge oxygen content with the increasing concentration of Er2O3. The topology cage structure around the luminescence center of rare earth ions is changed and the stability of the optically active center is enhanced, finally contributing to the enhancement of luminescence. Meanwhile, the maximum σemi and gain coefficient of Dy3+ reach up to 7.22 × 10−21 cm2 and 7.37 cm−1, respectively. The comprehensive results show that the fluorotellurite glass designed in this study is expected to be a gain medium for mid-infrared lasers in remote sensing monitoring, military, and other fields. Group 3 medulloblastoma is one of the most aggressive types of childhood brain tumors. Roughly 30% of cases carry genetic alterations in MYC, SMARCA4, or both genes combined. While overexpression of MYC has previously been shown to drive medulloblastoma formation in mice, the functional significance of SMARCA4 mutations and their suitability as a therapeutic target remain largely unclear. To address this issue, we combined overexpression of MYC with a loss of SMARCA4 in granule cell precursors. Both alterations did not increase proliferation of granule cell precursors in vitro. However, combined MYC overexpression and SMARCA4 loss successfully induced tumor formation in vivo after orthotopic transplantation in recipient mice. Resulting tumors displayed anaplastic histology and exclusively consisted of SMARCA4-negative cells although a mixture of recombined and non-recombined cells was injected. These observations provide first evidence for a tumor-promoting role of a SMARCA4 deficiency in the development of medulloblastoma. In comparing the transcriptome of tumors to the cells of origin and an established Sonic Hedgehog medulloblastoma model, we gathered first hints on deregulated gene expression that could be specifically involved in SMARCA4/MYC driven tumorigenesis. Finally, an integration of RNA sequencing and DNA methylation data of murine tumors with human samples revealed a high resemblance to human Group 3 medulloblastoma on the molecular level. Altogether, the development of SMARCA4-deficient medulloblastomas in mice paves the way to deciphering the role of frequently occurring SMARCA4 alterations in Group 3 medulloblastoma with the perspective to explore targeted therapeutic options. A series of CoCrFeNiMn high-entropy alloy matrix self-lubricating composites were prepared by spark plasma sintering. The composites are composed of an FCC phase, Cr7C3, Ag, and eutectic fluoride BaF2/CaF2 phases. The microstructure of the composites is uniform. The additional phases distribute along the boundary of equiaxed grains of the FCC phase. Tuberculosis is a widespread disease all over the world. Early diagnosis plays a crucial role in the treatment of patients and can reduce the mortality rate. A new biosensor based on a binary photonic crystal is designed for the detection of mycobacterium tuberculosis bacteria in the blood. The structure air/(A/B)N/(Defect)/(A/B)N/air is employed, where A, B and N are the first and second layers and the number of periods, respectively. Layers A and B are chosen as GaAs and BaF2, respectively. The defect layer is the blood sample. The transfer matrix method (TMM) is utilized to simulate the structure. Some parameters, such as central wavelength, defect layer thickness and incident angle are optimized to maximize the sensitivity. The optimum sensitivity is found 3197.18 nm/RIU, which is extremely high when compared to the most recent papers published in this field. Alternative techniques have been investigated for effectiveness in caries removal because conventional metallic dental burs can lead to an excessive loss of sound tissue. The aim of the present study is to realize a preliminary approach in obtaining effective polymer mixtures for polymeric bur development, capable of removing primary dental caries using combinations of polymers to ensure the requirements for such instruments, but also a greater compatibility with the teeth structure. This study assessed the main mechanical properties, water sorption, solubility and microscopic structure of four new polymer mixture recipes to provide essential features in obtaining experimental self-limited dental burs. Two mixtures have in their composition polymer mixtures of Bis-phenol A diglycidyl ether dimethacrylate/Triethylene glycol dimethacrylate/Urethane dimethacrylates (R1, R2), and two other mixtures have Bis-phenol A diglycidyl ether dimethacrylate/Polymethyl methacrylate/Methyl methacrylates (R3, R4). The incorporation of nanoparticles into the polymer matrix has become essential due to the need of polymer biocompatibility increasing along with teeth surface remineralization, so that the powder charge was added to four recipes, such as 5% glass with BaF2 and 0.5% graphene with silver particles. All data sets were analyzed using the One-Way ANOVA test. R3, R4 showed higher compressive strength and diametrical compression values; these values increased when glass and graphene were added. Moreover, the addition of glass particles lead to an increase in flexural strength. Regarding the sorption, sample R3 had the most significant differences between day 69 and the rest of the investigation days, while the solubility varied at different intervals. From the mechanical evaluation, we could conclude that the Bis-GMA/PMMA/MMA mixtures fit the mechanical characteristics supported by polymer burs, following future studies regarding their use on the affected dentin. Background: Disrupted intestinal epithelial barrier is one of the major causes of Crohn's disease (CD). Novel molecular targets for intestinal epithelial barrier are essential to treatment of CD. Transmembrane and immunoglobulin domain-containing protein 1 (TMIGD1) is an adhesion molecule that regulates cell adhesion, migration, and enterocyte differentiation. However, the function and mechanism of TMIGD1 in CD and intestinal epithelial barrier has rarely been studied. Furthermore, the association between TMIGD1 and the clinical features of CD remains unclear. Methods: Transcriptome analysis on colonic mucosa from CD patients and healthy individuals were performed to identify dysregulated genes. Multi-omics integration of the 1000IBD cohort including genomics, transcriptomics of intestinal biopsies, and serum proteomics identified the association between genes and characteristics of CD. Inflammation was assessed by cytokine production in cell lines, organoids and intestinal-specific Tmigd1 knockout (Tmigd1INT-KO) mice. Epithelial barrier integrity was evaluated by trans-epithelium electrical resistance (TEER), paracellular permeability, and apical junction complex (AJC) expression. Co-immunoprecipitation, GST pull-down assays, mass spectrometry, proteomics, and transcriptome analysis were used to explore downstream mechanisms. Results: Multi-omics integration suggested that TMIGD1 was negatively associated with inflammatory characteristics of CD. TMIGD1 was downregulated in inflamed intestinal mucosa of patients with CD and mice colitis models. Tmigd1INT-KO mice were more susceptible to chemically induced colitis. In epithelial cell lines and colonic organoids, TMIGD1 knockdown caused impaired intestinal barrier integrity evidenced by increased paracellular permeability and reduced TEER and AJC expression. TMIGD1 knockdown in intestinal epithelial cells also induced pro-inflammatory cytokine production. Mechanistically, TMIGD1 directly interacted with cytoplasmic BAF nuclear assembly factor 1 (BANF1) to inhibit NF-κB activation. Exogenous expression of TMIGD1 and BANF1 restored intestinal barrier function and inhibited inflammation in vitro and in vivo. TMIGD1 expression predicted response to anti-TNF treatment in patients with CD. Simple Summary: The generation of an artificial intestine could minimize the use of in vivo trials, which are presently necessary for testing aquafeeds. As well as being lengthy and expensive, they also require the use of many animals. Furthermore, although in vivo screening allows for a precise evaluation of the final effect of each feed, it does not improve our basic knowledge of the cellular and molecular mechanisms that determine the end results. This, in turn, severely limits our ability to understand and predict the biological value of ingredients, both individually and in differing combinations thereof. To circumvent these constraints, we previously derived and characterized two novel cell lines from the proximal (RTpi-MI) and distal intestine (RTdi-MI) of rainbow trout (Oncorhynchus mykiss), demonstrating that the two cell lines retain the main properties of the intestinal tracts from which they originate. In the present work, we used these cells to set up a functional intestinal barrier on a permeable membrane support to investigate the digestion and absorption of dietary amino acids in fish feed. The results demonstrate that the procedure we developed enables the quantification of individual amino acids that cross the intestinal wall. This new platform will now be used to screen and select new feed ingredients/formulations. The ever-increasing number and variation of raw materials utilized to provide alternative feed formulations continues to allow for a more sustainable and flexible approach. Testing all these options in vivo is still the most robust and reliable manner to pick the best raw material candidates, but it requires the use of large numbers of animals and is time-consuming and expensive. Therefore, we are developing an in vitro platform that can provide a reliable evaluation of new ingredients. The main aim of this work was to combine an in vitro digestion protocol of extruded, commercially relevant aquafeeds with the exposure of intestinal epithelial cells to the extracted bio-available fraction (BAF). The results show that 250,000 cells/cm2 represents the optimal seeding density and that up to 50% BAF concentration for up to 24 h had no negative effects on the epithelial barrier morphology and function. It is possible to determine amino acid digestibility and bioavailability in all the experimental conditions (with and without BSA, at 25% and 50% dilution) and at all time points (0, 6, and 24 h). Heavy metals (HMs) have the characteristics of high toxicity, long residual time, and difficult degradation, which pose a potential threat to aquatic ecosystems. The distribution and migration of HMs in different media can reflect their potential impact on aquatic ecosystem health. In this study, we analyzed the distribution characteristics of seven HMs (Zn, Cu, Cr, Ni, Hg, As, and Pb) in the water and sediment and five groups of organisms (fish, Crustacean, cephalopod, bivalvia, and gastropoda) in the sea ranching area of Haizhou Bay and explored the regularity of HM accumulation from water and sediments to organisms. The results showed that in the water, Zn and Hg had the smallest concentrations in the nearshore area, Cr had the lowest concentrations in the offshore area, and As, Cu, Pb, and Ni had the lowest concentrations in the central area. In the sediment, Hg had the smallest concentrations in the nearshore area and As, Pb, Cr, Cu, Ni, and Zn had the lowest concentrations in the offshore area; the average contents of seven heavy metals all meet the Class I standard of sediments in China. The concentrations of Cu and Zn in crustaceans were significantly higher than those in fishes (p < 0.05), and As showed a higher enrichment effect in cephalopods. Under the influence of feeding habits and habitat environment, the BAF value of benthic crustaceans and bivalvia accumulated HMs from sediments is high, and the BAF value of fish accumulated HMs from water and sediments is low. Overall, the concentration of HMs in water is high, and HMs in sediments are in good condition, crustaceans and bivalviaa have a strong ability to accumulate HMs from water and sediments, while fish are weak. We demonstrated the metal accumulation potential of Amaranthus retorflexus, a European weed species, both in moderately and strongly metal-contaminated sites. Metal accumulation in roots, stems, and leaves were studied. We also calculated the bioaccumulation factor (BAF), and translocation factor (TF) values to quantify the metal accumulation, and translocation between plant organs. Our findings indicated that metal accumulation correlated with metal concentration; that is plant organs accumulated higher concentration of metals in the contaminated area than in the control one. We found that the concentrations of Ba, Mn, Sr and Zn were the highest in leaves, and Al, Cr, Cu, Fe and Pb in roots. High BAF value was found for Sr in all studied areas, indicating this metal's high accumulation potential of Amaranthus retorflexus. High TF values were found for Al, Ba, Cu, Fe, Mn, Sr and Zn; these metals were successfully transported to aboveground plant organs. Growing concerns over environmental issues and sustainable living have resulted in increased interest in renewable energy and energy efficiency. The real estate market is no exception, with homeowners increasingly considering the market value of green and sustainable buildings, which can offer both energy efficiency and potential health benefits. This study investigates the level of interest among homeowners in investing in renewable energy sources and energy efficiency measures for their homes and how it relates to their perception of the market value of green or sustainable buildings in the real estate market. A survey was conducted in the Paphos urban complex in Cyprus, with 180 participants over the age of 18. The participants were selected through a random sampling method and were representative of the general population in terms of gender, age, and income. Data were collected on their attitudes towards renewable energy sources and energy efficiency, as well as their perceptions of the market value of green buildings. The data collected were analyzed using various statistical methods, including Cronbach's α coefficient, the non-parametric Friedman test, descriptive statistics, and factor analysis, with the Statistical Package for the Social Sciences (SPSS) being used for coding and analysis. Results indicate that 64% of the homeowners surveyed were interested in investing in renewable energy sources, and 72% were interested in energy efficiency measures. Additionally, findings suggest a moderate level of interest (58%) among homeowners in investing in renewable energy sources and that this is positively associated with their perception of the market value of green buildings. Furthermore, homeowners with higher income and education levels tend to be more interested in investing in renewable energy sources and energy efficiency measures and perceive green buildings as having higher market value. This study provides insights into the factors that drive homeowners' investment in renewable energy sources and energy efficiency measures, shedding light on the relationship between homeowners' perceptions of the market value of green buildings and their interest in such investments. Diffuse midline gliomas (DMGs) including diffuse intrinsic pontine gliomas (DIPGs) bearing lysine-to-methionine mutations in histone H3 at lysine 27 (H3K27M) are lethal childhood brain cancers. These tumors harbor a global reduction in the transcriptional repressive mark H3K27me3 accompanied by an increase in the transcriptional activation mark H3K27ac. We postulated that H3K27M mutations, in addition to altering H3K27 modifications, reprogram the master chromatin remodeling switch/sucrose nonfermentable (SWI/SNF) complex. The SWI/SNF complex can exist in two main forms termed BAF and PBAF that play central roles in neurodevelopment and cancer. Moreover, BAF antagonizes PRC2, the main enzyme catalyzing H3K27me3. We demonstrate that H3K27M gliomas show increased protein levels of the SWI/SNF complex ATPase subunits SMARCA4 and SMARCA2, and the PBAF component PBRM1. Additionally, knockdown of mutant H3K27M lowered SMARCA4 protein levels. The proteolysis targeting chimera (PROTAC) AU-15330 that simultaneously targets SMARCA4, SMARCA2, and PBRM1 for degradation exhibits cytotoxicity in H3.3K27M but not H3 wild-type cells. AU-15330 lowered chromatin accessibility measured by ATAC-Seq at nonpromoter regions and reduced global H3K27ac levels. Integrated analysis of gene expression, proteomics, and chromatin accessibility in AU-15330-treated cells demonstrated reduction in the levels of FOXO1, a key member of the forkhead family of transcription factors. The altered lysosomal function can induce drug redistribution which leads to drug resistance and poor prognosis for cancer patients. V-ATPase, an ATP-driven proton pump positioned at lysosomal surfaces, is responsible for maintaining the stability of lysosome. Herein, we reported that the potassium voltage-gated channel subfamily J member 15 (KCNJ15) protein, which may bind to V-ATPase, can regulate the function of lysosome. The deficiency of KCNJ15 protein in breast cancer cells led to drug aggregation as well as reduction of drug efficacy. The application of the V-ATPase inhibitor could inhibit the binding between KCNJ15 and V-ATPase, contributing to the amelioration of drug resistance. Clinical data analysis revealed that KCNJ15 deficiency was associated with higher histological grading, advanced stages, more metastases of lymph nodes, and shorter disease free survival of patients with breast cancer. KCNJ15 expression level is positively correlated with a high response rate after receiving neoadjuvant chemotherapy. Moreover, we revealed that the small molecule drug CMA/BAF can reverse drug resistance by disrupting the interaction between KCNJ15 and lysosomes. This work aimed to detect changes in trace element chemical speciation in sewage sludge (SS) after the ozonation process. The modified Community Bureau of Reference (BCR) sequential extraction procedure was performed to determine the chemical speciation of trace elements in SS after the ozonation process. To assess potential soil contamination with trace elements from sewage sludge after the ozonation process, the risk assessment code (RAC) coefficient was used. The bioaccumulation factor (BAF) and translocation factor (TF) values were also calculated to characterize the efficiency of trace element accumulation in the studied plant species from soil fertilized with sludges after the ozonation process. Generally, the mean concentration of total trace elements in the SS after the ozonation process was higher, but the differences were statistically significant only in the case of Mn, Cu, Pb, and Cd. The dominant fraction of Fe, Cr, Pb, and Cd was the residual fraction F4, while the extractable/exchangeable fraction F1 was present in the smallest amount. Therefore, in the case of Mn, Zn, and Ni, the ozonation process had a significant impact on the increase in the content of these elements in the F1 fraction. The application of the SS stabilized by ozonation process for maize and wheat fertilization did not significantly affect the bioaccumulation of most of the analyzed metals in aboveground biomass. Higher values of BAF coefficients after the application of ozonated SS were found only in the case of Cu and Ni. In turn, the determined TF coefficients were lower than 1 in most cases. The obtained results showed that the slight change in the concentration of Zn, Mn, and Ni in fraction F1 causes a specific risk of their mobility in the soil environment. It should be noted that due to the variable composition of sewage sludge, an analysis of the content of individual trace elements in chemical fractions should be carried out to assess its actual impact on the environment. Gammaherpesviruses, including Kaposi sarcoma-associated herpesvirus (KSHV) and Epstein-Barr virus (EBV), are DNA viruses that are globally associated with human cancers and establish lifelong latency in the human population. Detection of gammaherpesviral infection by the cGAS-STING innate immune DNA-sensing pathway is critical for suppressing viral reactivation from latency, a process that promotes viral pathogenesis and transmission. We report that barrier-to-autointegration factor 1 (BAF)-mediated suppression of the cGAS-STING signaling pathway is necessary for reactivation of KSHV and EBV. We demonstrate a role for BAF in destabilizing cGAS expression and show that inhibiting BAF expression in latently infected, reactivating, or uninfected cells leads to increased type I interferon-mediated antiviral responses and decreased viral replication. Furthermore, BAF overexpression resulted in decreased cGAS expression at the protein level. These results establish BAF as a key regulator of the lifecycle of gammaherpesviruses and a potential target for treating viral infections and malignancies. Although China has been the main manufacturer and consumer of phthalate esters (PAEs), human health ambient water quality criteria (AWQCs) have not been proposed for these chemicals. In this study, the distribution and bioaccumulation of six PAEs (dimethyl phthalate (DMP), diethyl phthalate (DEP), di-n-butyl phthalate (DBP), butyl benzyl phthalate (BBP), bis (2-ethylhexyl) phthalate (DEHP), and di-n-octyl phthalate (DnOP)) were investigated in 11 edible fish species collected from Poyang Lake, China. The results showed that the total concentrations of the six PAEs in the fish ranged between 118.63 and 819.84 μg/kg wet weight (mean of 327.50 ± 190.44 μg/kg). DMP, DEP, DBP, and DEHP were detected in all samples, of which DEHP and DBP were two of the most predominant phthalates, accounting for more than 90% of the total PAEs. The DEHP concentrations in fish with different habitat preferences were different, demersal species were significantly higher than pelagic species (p < 0.05). The mean natural logarithmic bioaccumulation factors (log BAFs) of PAEs increased with increasing lipophilicity of the substances, which yielded the following regression equation: log BAF (L/kg) = 0.103 log Kow + 2.158 (r2 = 0.940, p < 0.05, n = 4). Using this quantitative structure–activity relationship to calculate BAFs for the remaining undetected substances (BBP and DnOP) to derive AWQCs. According to the natural parameters, the human health AWQCs relating to PAE concentrations for water and fish consumption were derived as 9.4 × 103 (DMP), 5.0 × 102 (DEP), 4.2 × 101 (DBP), 1.1 (BBP), 8.6 × 10− 2 (DEHP), and 2.0 (DnOP) μg/L. Human health risk assessment indicated that the dietary intake of DEHP may exert a carcinogenic effect on residents of the Poyang Lake region. Mutations in DNAJC5/CSPα are associated with adult neuronal ceroid lipofuscinosis (ANCL), a dominant-inherited neurodegenerative disease featuring lysosome-derived autofluorescent storage materials (AFSMs) termed lipofuscin. Functionally, DNAJC5 has been implicated in chaperoning synaptic proteins and in misfolding-associated protein secretion (MAPS), but how DNAJC5 dysfunction causes lipofuscinosis and neurodegeneration is unclear. Here we report two functionally distinct but coupled chaperoning activities of DNAJC5, which jointly regulate lysosomal homeostasis: While endolysosome-associated DNAJC5 promotes ESCRT-dependent microautophagy, a fraction of perinuclear and non-lysosomal DNAJC5 mediates MAPS. Functional proteomics identifies a previously unknown DNAJC5 interactor SLC3A2/CD98hc that is essential for the perinuclear DNAJC5 localization and MAPS but dispensable for microautophagy. Importantly, uncoupling these two processes, as seen in cells lacking SLC3A2 or expressing ANCL-associated DNAJC5 mutants, generates DNAJC5-containing AFSMs resembling NCL patient-derived lipofuscin and induces neurodegeneration in a Drosophila ANCL model. These findings suggest that MAPS safeguards microautophagy to avoid DNAJC5-associated lipofuscinosis and neurodegeneration. BAF and PBAF are mammalian SWI/SNF family chromatin remodeling complexes that possess multiple histone/DNA-binding subunits and create nucleosome-depleted/free regions for transcription activation. Despite previous structural studies and recent advance of SWI/SNF family complexes, it remains incompletely understood how PBAF-nucleosome complex is organized. Here we determined structure of 13-subunit human PBAF in complex with acetylated nucleosome in ADP-BeF3-bound state. Four PBAF-specific subunits work together with nine BAF/PBAF-shared subunits to generate PBAF-specific modular organization, distinct from that of BAF at various regions. PBAF-nucleosome structure reveals six histone-binding domains and four DNA-binding domains/modules, the majority of which directly bind histone/DNA. This multivalent nucleosome-binding pattern, not observed in previous studies, suggests that PBAF may integrate comprehensive chromatin information to target genomic loci for function. Our study reveals molecular organization of subunits and histone/DNA-binding domains/modules in PBAF-nucleosome complex and provides structural insights into PBAF-mediated nucleosome association complimentary to the recently reported PBAF-nucleosome structure. Double-PHD fingers 3 (DPF3) is a BAF-associated human epigenetic regulator, which is increasingly recognised as a major contributor to various pathological contexts, such as cardiac defects, cancer, and neurodegenerative diseases. Recently, we unveiled that its two isoforms (DPF3b and DPF3a) are amyloidogenic intrinsically disordered proteins. DPF3 isoforms differ from their C-terminal region (C-TERb and C-TERa), containing zinc fingers and disordered domains. Herein, we investigated the disorder aggregation properties of C-TER isoforms. In agreement with the predictions, spectroscopy highlighted a lack of a highly ordered structure, especially for C-TERa. Over a few days, both C-TERs were shown to spontaneously assemble into similar antiparallel and parallel β-sheet-rich fibrils. Altered metal homeostasis being a neurodegeneration hallmark, we also assessed the influence of divalent metal cations, namely Cu2+, Mg2+, Ni2+, and Zn2+, on the C-TER aggregation pathway. Circular dichroism revealed that metal binding does not impair the formation of β-sheets, though metal-specific tertiary structure modifications were observed. Through intrinsic and extrinsic fluorescence, we found that metal cations differently affect C-TERb and C-TERa. Cu2+ and Ni2+ have a strong inhibitory effect on the aggregation of both isoforms, whereas Mg2+ impedes C-TERb fibrillation and, on the contrary, enhances that of C-TERa. Upon Zn2+ binding, C-TERb aggregation is also hindered, and the amyloid autofluorescence of C-TERa is remarkably red-shifted. Using electron microscopy, we confirmed that the metal-induced spectral changes are related to the morphological diversity of the aggregates. While metal-treated C-TERb formed breakable and fragmented filaments, C-TERa fibrils retained their flexibility and packing properties in the presence of Mg2+ and Zn2+ cations. Haemoglobinopathies, including thalassaemias and sickle-cell syndromes, are demanding, lifelong conditions that pose a significant burden to patients, families, and healthcare systems. Despite the therapeutic advances and the resulting improvements in prognosis accomplished in past decades, these patients still face important challenges, including suboptimal access to quality care in areas with developing economies, changing epidemiology due to massive migration flows, an evolving clinical spectrum due to ageing in well-treated patients, and limited access to novel high-cost therapies. We herein describe the organization of healthcare services for haemoglobinopathies in Cyprus--with particular focus on beta-thalassaemia, the most prevalent condition in this region--along with selected patient outcomes. This report aims at underscoring the fact that nationally funded and well-coordinated prevention and care programmes for chronic and complex conditions, such as haemoglobinopathies, with active involvement from patient organizations lead to effective disease control and excellent outcomes in survival, quality of life, social adaptation, and public health savings, and allow timely and effective responses to emerging crises, such as the COVID-19 pandemic. The Cyprus paradigm could therefore serve as a blueprint for the organization or adaptation of haemoglobinopathy programs in other countries since these disorders are still widely occurring. Four-wave mixing processes are ubiquitous in ultrafast optics and the determination of the coefficients of the χ(3) tensor is thus essential. We introduce a novel time-resolved ultrafast spectroscopic method to characterize the third-order nonlinearity on the femtosecond time-scale. This approach, coined as "nonlinear chirped interferometry," makes use of the variation of the optical group delay of a transmitted probe under the effect of an intense pump pulse in the nonlinear medium of interest. The observable is the spectral interference between the probe and a reference pulse sampled upstream and the metric is the transient swing of the probe group delay. We show that the detected signal is enhanced when the pulses are weakly chirped, and that, although interferometric, the method is intrinsically less sensitive to environmental phase fluctuations and drifts. By chirping adequately the reference pulse, the transient frequency shift of the probe pulses is also detected in the time domain and the detected nonlinear signal is enhanced. Nonlinear phase shifts as low as 10 mrad, corresponding to a frequency shift of 30 GHz, i.e., 0.01% of the carrier frequency, are detected without heterodyne detection or active phase-stabilization. The diagonal and/or non-diagonal terms of reference glasses (SiO2) and crystals (Al2O3, BaF2, CaF2) are characterized. With an incidence of ~1 in 800 births, Down syndrome (DS) is the most common chromosomal condition linked to intellectual disability worldwide. While the genetic basis of DS has been identified as a triplication of chromosome 21 (HSA21), the genes encoded from HSA21 that directly contribute to cognitive deficits remain incompletely understood. Here, we found that the HSA21-encoded chromatin effector, BRWD1, was upregulated in neurons derived from iPS cells from an individual with Down syndrome and brain of trisomic mice. We showed that selective copy number restoration of Brwd1 in trisomic animals rescued deficits in hippocampal LTP, cognition and gene expression. We demonstrated that Brwd1 tightly binds the BAF chromatin remodeling complex, and that increased Brwd1 expression promotes BAF genomic mistargeting. Importantly, Brwd1 renormalization rescued aberrant BAF localization, along with associated changes in chromatin accessibility and gene expression. These findings establish BRWD1 as a key epigenomic mediator of normal neurodevelopment and an important contributor to DS-related phenotypes. The current study examines the bioaccumulation potential of Taif rose shrubs by analyzing the shrubs' wastes. f. At Al-Shafa Highland, four farms with plants of different ages were chosen to collect soil samples and vegetative waste (leaves and stems) for morphological and chemical analysis. The tallest stem and largest crown diameter (184.2 and 243.5 cm, respectively) were found in the oldest (20-year-old) shrubs, which also produced the highest biomass of pruning wastes of stems and leaves (3.9 and 1.3 t/ha, respectively). The 10-year-old shrubs gathered the highest concentration of Co and Pb (1.74 and 7.34 mg kg−1) in the stem and the highest Fe, Mn, and Ni (18.55, 18.60, and 9.05 mg kg−1) in the leaves, while the youngest plants (4 years) accumulated the highest Cr and Zn (0.83 and 13.44 mg kg−1) in their leaves. The highest contents of Cd, Cr, Cu, Fe, Mn, Pb, and Zn were found in the oldest Taif rose stem (34.94, 1.16, 36.29, 49.32, 51.22, 24.76, and 32.51 g ha−1), while the highest contents of Co and Ni were found in the stems of plants that were 10 and 12 years old (3.21 and 9.54 g ha−1, respectively). The Taif rose's stem and leaves can absorb the majority of heavy metals that have been studied with BAF values greater than one. Significant relationships between various heavy metals in the soil and the same in the stems (Al, Co, and Pb) and leaves (Co, Fe, Mn, Ni, and Pb) of Taif roses have been observed. According to the current findings, the Taif rose is a promising viable and safe crop for heavy metals phytoremediation if it is grown in polluted soil because there is little to no risk of contamination in the use of its end products, high biomass of pruning wastes, and high efficiency of heavy metal removal. Observing phenological changes are important for evaluating the natural regeneration process of forests, especially in Mediterranean areas where the regeneration of coniferous forests depends on seeds and the changes in blossoming time are influenced by climate change. The high temporal resolution of Sentinel-1 data allows the time series analysis of synthetic aperture radar (SAR) data, but it is still unknown how these data could be utilised for better understanding forest phenology and climate-related alternations. This study investigates the phenological cycle of Paphos forest, Cyprus using SAR data from 1992 to 2021, acquired by ERS-1/2, Envisat and Sentinel-1. An average phenological diagram was created for each space mission and a more detailed analysis was performed from October 2014 to November 2021, using the higher temporal resolution of Sentinel-1 data. Meteorological data were used to better understand the drivers of blooming alternations. Using the interquartile range (IQR), outliers were detected and replaced using the Kalman filter imputation. Forecasting trend lines were used to estimate the amplitude of the summer peaks and the annual mean. The observation of the average phenology from each satellite mission showed that there were two main blooming peaks each year: the winter and the summer peak. We argue that the winter peak relates to increased foliage, water content and/or increased soil moisture. The winter peak was followed by a fall in February reaching the lower point around March, due to the act of pine processionary (Thaumetopoea pityocampa). The summer peak should relate to the annual regeneration of needles and the drop of the old ones. A delay in the summer peak—in August 2018—was associated with increased high temperatures in May 2018. Simultaneously, the appearance of one peak instead of two in the σ V H time series during the period November 2014–October 2015 may be linked to a reduced act of the pine processionary associated with low November temperatures. Furthermore, there was an outlier in February 2016 with very low backscattering coefficients and it was associated with a drought year. Finally, predicting the amplitude of July 2020 returned high relevant Root Mean Square Error (rRMSE). Seven years of time series data are limiting for predicting using trend lines and many parameters need to be taken into consideration, including the increased rainfall between November 2018 and March 2020. The effect of BaF2, MgF2, and AlF3 on the structural and luminescent properties of gallo-germanate glass (BGG) doped with erbium ions was investigated. A detailed analysis of infrared and Raman spectra shows that the local environment of erbium ions in the glass was influenced mainly by [GeO]4 and [GeO]6 units. Moreover, the highest number of non-bridging oxygens was found in the network of the BGG glass modified by MgF2. The 27Al MAS NMR spectrum of BGG glass with AlF3 suggests the presence of aluminum in tetra-, penta-, and octahedral coordination geometry. Therefore, the probability of the 4I13/2→4I15/2 transition of Er3+ ions increases in the BGG + MgF2 glass system. On the other hand, the luminescence spectra showed that the fluoride modifiers lead to an enhancement in the emission of each analyzed transition when different excitation sources are employed (808 nm and 980 nm). The analysis of energy transfer mechanisms shows that the fluoride compounds promote the emission intensity in different channels. Background: Nowadays, wound healing is one of the main problems of patients. Therefore, extensive research is underway to discover mechanisms associated with non-scarring of wounds. Using amniotic fluid and laser may potentially play a key role in wound healing and scar reduction due to its presence in tissue growth and repair agents. Aim: The present study evaluated the effect of bovine amniotic fluid (BAF)-derived cream and low-power laser (LPL) on accelerating skin wound healing and reducing scarring in an animal model. Materials and Methods: Therefore, 72 male Wistar rats were randomly divided into three groups (each group: 24). A wound 6 mm in diameter was then inflicted on the rats' backs. In the first group that was the control group, the wound was only used. Moreover, BAF was implemented for the second group, and in the third group, LPL radiation was utilized. On the 1st, 3rd, 5th, 14th, and 21st days, the healing condition of the wound and scar created were examined. Results: Hence, evaluation of wound healing status on days 5 and 14 showed that the wound healing scale in the BAF group and LPL group was significantly better than that of the control group. On the 21st day, the average Scar Scoring Scale in the BAF and LPL groups was significantly lower than that of the control group. Histological images showed a significant repair in the LPL and BAF groups. Conclusion: To conclude, considering the positive effect of LPL and BAF on wound healing and less scarring, it seems that LPL and BAF can heal wounds faster. YBa2Cu3O7−δ (YBCO) thin films with the addition of Cl were prepared on Hastelloy tape by the extremely low fluorine metal-organic deposition (MOD) technique. The composition and microstructure of the present samples were characterized by X-ray diffraction (XRD) and scanning electron microscopy (SEM). It was revealed that the Cl-added YBCO thin films achieved better texture and density compared with the pure YBCO films. The superconducting properties at magnetic fields were measured using the magnetic property measurement system (MPMS). The results showed that the sample onset transition temperature (Tc, on) did not change significantly after the addition of Cl. The critical current density (Jc) in the presence of YBCO with the addition of Cl increased significantly. Meanwhile, both the Jc and gravitational force (Fp) of the (Cl, Hf) co-added YBCO films were further enhanced at different magnetic fields. The thermodynamic analysis of the BaCO3 removal reaction revealed that the addition of Cl to YBCO formed chlorine compounds, which were effective in avoiding the formation of BaCO3. Further, the calculation results of the Gibbs free energy at different pressure quotients showed that the stability of the relevant chlorine compounds occurred in the order of BaCl2 > CuCl2 > YCl3 and the oxidation of BaCl2 to BaO required a higher temperature compared to BaF2, implying the potential advantages of inhibiting the a-axis growth. Transposable elements (TEs) through evolutionary exaptation have become an integral part of the human genome, offering ample regulatory sequences and shaping chromatin 3D architecture. While the functional impacts of TE-derived sequences on early embryogenesis have been recognized, their roles in malignancy are only starting to emerge. Here we show that many TEs, especially the pluripotency-related human endogenous retrovirus H (HERVH), are abnormally activated in colorectal cancer (CRC) samples. Transcriptional upregulation of HERVH is associated with mutations of several tumor suppressors, particularly ARID1A. Knockout of ARID1A in CRC cells leads to increased transcription at several HERVH loci, which involves compensatory contribution by ARID1B. Suppression of HERVH in CRC cells and patient-derived organoids impairs tumor growth. Mechanistically, HERVH transcripts colocalize with nuclear BRD4 foci, modulating their dynamics and co-regulating many target genes. Altogether, we uncover a critical role for ARID1A in restraining HERVH, whose abnormal activation can promote tumorigenesis by stimulating BRD4-dependent transcription. Here the authors show mutation of the BAF chromatin remodeler subunit ARID1A results in an ARID1B-dependent upregulation of HERVH, an ERV required for the pluripotency regulatory network. Background: The exit from pluripotency or pluripotent-somatic transition (PST) landmarks an event of early mammalian embryonic development, representing a model for cell fate transition. Results: In this study, using a robust JUN-induced PST within 8 h as a model, we investigate the chromatin accessibility dynamics (CAD) as well as the behaviors of corresponding chromatin remodeling complex SS18/BAFs, to probe the key events at the early stage of PST. Here, we report that, JUN triggers the open of 34661 chromatin sites within 4 h, accomplished with the activation of somatic genes, such as Anxa1, Fosl1. ChIP-seq data reveal a rapid relocation of SS18/BAFs from pluripotent loci to AP-1 associated ones. Consistently, the knockdown of Brg1, core component of BAF complexes, leads to failure in chromatin opening but not closing, resulting in delay for JUN induced PST. Notably, the direct interaction between SS18/BAFs and JUN-centric protein complexes is undetectable by IP-MS. Instead, we show that H3K27ac deposited by cJUN dependent process regulates SS18/BAFs complex to AP1-containing loci and facilitate chromatin opening and gene activation. Conclusions: These results reveal a rapid transfer of chromatin remodeling complexes BAF from pluripotent to somatic loci during PST, revealing a simple mechanistic aspect of cell fate control. Over millions of years, eukaryotes evolved from unicellular to multicellular organisms with increasingly complex genomes and sophisticated gene expression networks. Consequently, chromatin regulators evolved to support this increased complexity. The ATP-dependent chromatin remodelers of the SWI/SNF family are multiprotein complexes that modulate nucleosome positioning and appear under different configurations, which perform distinct functions. While the composition, architecture, and activity of these subclasses are well understood in a limited number of fungal and animal model organisms, the lack of comprehensive information in other eukaryotic organisms precludes the identification of a reliable evolutionary model of SWI/SNF complexes. Here, we performed a systematic analysis using 36 species from animal, fungal, and plant lineages to assess the conservation of known SWI/SNF subunits across eukaryotes. We identified evolutionary relationships that allowed us to propose the composition of a hypothetical ancestral SWI/SNF complex in the last eukaryotic common ancestor. This last common ancestor appears to have undergone several rounds of lineage-specific subunit gains and losses, shaping the current conformation of the known subclasses in animals and fungi. In addition, our results unravel a plant SWI/SNF complex, reminiscent of the animal BAF subclass, which incorporates a set of plant-specific subunits of still unknown function. The ancestral history of SWI/SNF subunits, responsible for nucleosome positioning, are explored in this work, revealing a plant SWI/SNF complex similar to the animal BAF subclass. A cross polarized wave (XPW) generation experiment with double BaF2 crystals in air was demonstrated with 120 µJ input pulse. A circular aperture was used to truncate the near field profile of the input beam and remove the outer part of the beam, and circularly symmetric beam profiles were generated at the BaF2 crystal positions. The measured focal spot size was close to the diffraction limit, and the energy stability of XPW was improved from ∼1.89% (without the aperture) to ∼0.67% (with the aperture). [ABSTRACT FROM AUTHOR]

In the present research work, we have theoretically analyzed the photonic band-gap properties of one-dimensional photonic structures composed of polycarbonate and non-glass materials. These photonic structures, PC1, PC2, PC3 and PC4, are composed of alternating layers of polycarbonate/Al2O3, polycarbonate/MgF2, polycarbonate/BaF2 and polycarbonate/TiO2 materials, respectively. The period of each photonic structure is made up of a thin non-glass material layer sandwiched between two identical polycarbonate layers. The transfer matrix method has been used to investigate the transmission properties of PC1 to PC4. The comparison between the transmission spectra of PC1 to PC4 shows that the polycarbonate and TiO2-based photonic structure (PC4) possess three PBGs of zero transmission located at far-ultraviolet, visible and near-infrared regions of the electromagnetic spectrum at normal and oblique incidence (θ0 = 55°), both corresponding to TE wave only. The index of refraction of all five materials used in this study was obtained by applying the Sellmeier-type dispersion relationship to ensure accuracy in the results. The purpose of selecting polycarbonate along with Al2O3, TiO2, MgF2 or BaF2 as constituent materials of these photonic structures is due to the heat resistance properties of polycarbonate and the unique optical properties of oxide and fluoride materials with wide transparency from the ultraviolet to the near-infrared regions of the electromagnetic spectrum. The proposed work can be used to design some influential wavelength-selective reflectors composed of 1D PCs behind the active region of the solar cells for improving the photovoltaic performance of solar panels. This study can further be utilized for the fabrication of advanced solar cell designs consisting of 1D photonic mirror-based luminescence and reflection concentrators.
