This paper introduces a mathematical model that incorporates the pertinent optical and physiological properties of skin reflections with the objective to increase our understanding of the algorithmic principles behind remote photoplethysmography (rPPG). The model is used to explain the different choices that were made in existing rPPG methods for pulse extraction. The understanding that comes from the model can be used to design robust or application-specific rPPG solutions. We illustrate this by designing an alternative rPPG method, where a projection plane orthogonal to the skin tone is used for pulse extraction. Recent years have witnessed extensive research activity in modeling biological phenomena as well as in developing intervention strategies for such phenomena. S-systems, which offer a good compromise between accuracy and mathematical flexibility, are a promising framework for modeling the dynamical behavior of biological phenomena. In this paper, two different intervention strategies, namely direct and indirect, are proposed for the S-system model. In the indirect approach, the prespecified desired values for the target variables are used to compute the reference values for the control inputs, and two control algorithms, namely simple sampled-data control and model predictive control (MPC), are developed for transferring the control variables from their initial values to the computed reference ones. In the direct approach, a MPC algorithm is developed that directly guides the target variables to their desired values. The proposed intervention strategies are applied to the glycolytic–glycogenolytic pathway and the simulation results presented demonstrate the effectiveness of the proposed schemes. Oscillometry is a popular technique for automatic estimation of blood pressure (BP). However, most of the oscillometric algorithms rely on empirical coefficients for systolic and diastolic pressure evaluation that may differ in various patient populations, rendering the technique unreliable. A promising complementary technique for automatic estimation of BP, based on the dependence of pulse transit time (PTT) on cuff pressure (CP) (PTT–CP mapping), has been proposed in the literature. However, a theoretical grounding for this technique and a nonparametric BP estimation approach are still missing. In this paper, we propose a novel coefficient-free BP estimation method based on PTT–CP dependence. PTT is mathematically modeled as a function of arterial lumen area under the cuff. It is then analytically shown that PTT–CP mappings computed from various points on the arterial pulses can be used to directly estimate systolic, diastolic, and mean arterial pressure without empirical coefficients. Analytical results are cross-validated with a pilot investigation on ten healthy subjects where 150 simultaneous electrocardiogram and oscillometric BP recordings are analyzed. The results are encouraging whereby the mean absolute errors of the proposed method in estimating systolic and diastolic pressures are 5.31 and 4.51 mmHg, respectively, relative to the Food and Drug Administration approved Omron monitor. Mock circulatory loops (MCLs) have been widely used to test left ventricular assist devices. The hydraulic properties of the mock systemic arterial system are usually described by two alternative four-element windkessel (W4) models. Compared with three-element windkessel model, their parameters, especially the inertial term, are much more difficult to estimate. In this paper, an estimator based on the iterated unscented Kalman filter (IUKF) algorithm is proposed to identify model parameters. Identifiability of these parameters for different measurements is described. Performance of the estimator for different model structures is first evaluated using numerical simulation data contaminated with artificial noise. An MCL is developed to test the proposed algorithm. Parameter estimates for different models are compared with the calculated values derived from the mechanical and hydraulic properties of the MCL to validate model structures. In conclusion, the W4 model with an inertance and an aortic characteristic resistance arranged in series is proposed to represent the mock systemic arterial system. Once model structure is appropriately selected, IUKF can provide reasonable estimation accuracy in a limited time and may be helpful for future clinical applications. [ABSTRACT FROM PUBLISHER]

A partially perturbed particle swarm optimization (PPSO) has been proposed for identifying the parameters of the Beeler–Reuter (BR) equation from action potential data. In the PPSO algorithm, the 63 BR equation parameters are divided into groups, and parameter patterns are made from the combination of the groups. PPSO enhances the capability of conventional particle swarm optimization (CPSO) by partially perturbing the coordinates of the globally best particle with the patterns when the searching process is locally confined. “Experimental data” were produced for cardiac myocytes simulated by the BR equation and the equation of Luo and Rudy (1991), and were used to test the algorithm of PPSO. The test results show that PPSO was able to identify the parameters of the BR equation effectively for different cardiac myocytes, while still retaining the conceptual simplicity and easy implementation of CPSO. We study the use of the FitzHugh–Nagumo (FHN) model for capturing neural spiking. The FHN model is a widely used approximation of the Hodgkin–Huxley model that has significant limitations. In particular, it cannot produce the key spiking behavior of bursting. We illustrate that by allowing time-varying parameters for the FHN model, these limitations can be overcome while retaining its low-order complexity. This extension has applications in modeling neural spiking behaviors in the thalamus and the respiratory center. We demonstrate the use of the FHN model from an estimation perspective by presenting a novel parameter estimation method that exploits its multiple time-scale properties, and compare the performance of this method with the extended Kalman filter in several illustrative examples. Quantitative methods such as model-based predictive control are known to facilitate the design of strategies to manipulate biological systems. This study develops a sparse-grid-based adaptive model predictive control (MPC) strategy to direct HL60 cellular differentiation. Sparse-grid sampling and interpolation support a computationally efficient adaptive MPC scheme in which multiple data-consistent regions of the model parameter space are identified and used to calculate a control compromise. The algorithm is evaluated in silico with structural model mismatch. Simulations demonstrate how the multiscenario control strategy more effectively manages the mismatch compared to a single scenario approach. Furthermore, the controller is evaluated in vitro to differentiate HL60 cells in both normal and perturbed environments. The controller-derived input sequence successfully achieves and sustains the specified target level of granulocytes when implemented in the laboratory. The results and analysis given here imply that adoption of this experiment planning technique to direct cell differentiation within more complex tissue engineered constructs will require the use of a reasonably accurate mathematical model and an extension of this algorithm to multiobjective controller design. Patient studies based on diffusion tensor images (DTI) require spatial correspondence between subjects. We propose to obtain the correspondence from white matter tracts, by introducing a new method for nonrigid matching of white matter fiber tracts in DTI. The method boils down to point set registration that involves simultaneously clustering and matching of the data points. The tracts are implicitly warped to a common frame of reference to avoid the potential bias toward one of the datasets. The algorithm gradually refines from global to local registration, which is implemented through deterministic annealing. Special care was taken to incorporate the spatial relation between fiber points and the uncertainty in principal diffusion orientation. As a result, the computed clusters are oriented along the fiber tracts and discriminate between adjacent but distinct fiber tracts. This is validated on synthetic and clinical data. The root-mean-squared distance with respect to expert-annotated landmarks is low (3 mm). In contrast to a state-of-the-art nonrigid registration technique, the proposed method is more robust to residual misalignments in terms of measured fractional anisotropy values. The efficient solution of the bidomain equations is a fundamental tool in the field of cardiac electrophysiology. When choosing a finite element discretization of the coupled system, one has to deal with the solution of a large, highly sparse system of linear equations. The conjugate gradient algorithm, along with suitable preconditioning, is the natural choice in this scenario. In this study, we identify the optimal preconditioners with respect to both stimulus protocol and mesh geometry. The results are supported by a comprehensive study of the mesh-dependence properties of several preconditioning techniques found in the literature. Our results show that when only intracellular stimulus is considered, incomplete LU factorization remains a valid choice for current cardiac geometries. However, when extracellular shocks are delivered to tissue, preconditioners that take into account the structure of the system minimize execution time and ensure mesh-independent convergence.
