The paper investigates the occurrence of relaxation processes in enzymatic hydrolysis of whey proteins in phosphate buffer using high-resolution ultrasonic spectroscopy. The hydrolysis produces a frequency-dependent evolution of ultrasonic velocity and attenuation originated by the proton transfer between the phosphate ions and the terminal − N H 2 group of protein hydrolysates. Such frequency dependence is absent in other systems, e.g., Tris buffer. Real-time profiles of ultrasonic velocity and attenuation measured during the hydrolysis of β–lactoglobulin, α–lactalbumin and bovine serum albumin by α–chymotrypsin were employed in quantitative analysis of the relaxation mechanisms, relaxation times, determination of the proton transfer rate constants and the reaction volume. The proton transfer is characterised by the rate constant 9.4 × 107 L mol−1 s−1 and the adiabatic reaction volume 24 × 10−6 m3 mol−1. The obtained reaction volume agrees well with thermodynamic data for ionisation volumes and enthalpies of the terminal amino group and phosphate. The described methodology for estimating the relaxation contribution to ultrasonic characteristics allows for precision real-time ultrasonic measurements of the concentration of peptide bonds cleaved during protein hydrolysis in a broad range of environmental conditions. Abstract Load transmission is an important function of the meniscus. In articular cartilage, proteoglycans help maintain hydration via negatively charged moieties. We aimed to investigate the influence of electrostatic effects on stiffness of meniscal tissue. Circular discs from bovine menisci of 8 mm diameter and 5 mm thickness were placed within a confined compression chamber. The apparatus was bathed in distilled water, 0.14 M PBS or 3 M PBS before being subjected to 5% ramp compressive strain and held for 300s. FEBio software was used to fit resultant relaxation curves to a non-linear poroviscoelastic model with strain dependent Holmes-Mow permeability. Analysis was conducted using one-way ANOVA with Tukey post-hoc analysis. 10 samples were tested in each solution. Significant differences (p < 0.05) were observed between the values for Young's modulus, zero strain dependent permeability and the viscoelastic coefficient for samples tested in 3 M PBS as compared to deionised water/0.14 M PBS. No significant differences were observed in the strain dependent/stiffening coefficients or the relaxation time. Approximately 79% of the stiffness of the meniscus appears attributable to ionic effects. Ionic effects play a significant role in the mechanical stiffness of the meniscus. Coalescence in liquid–liquid systems is determined by multiple factors, including droplet size ratios and the concentration and type of any continuous phase ions present. When using the standardized EFCE (European Federation of Chemical Engineering) toluene/water standard test system, these factors were systematically investigated based on a high statistical relevant database. The influence on the film drainage time was captured with a high speed imaging system. Shorter coalescence times were found to correlate with higher coalescence probabilities and an observed increase in the coalescence time with increasing droplet diameter agrees well with simple drainage modeling. Coalescence inhibition was also observed with the addition of continuous phase electrolyte. An empirical correlation was developed to account for the influence of equivalent droplet diameter and ion concentration on the coalescence time. [ABSTRACT FROM AUTHOR]

Normal and less polarised water models are used as the solvent to investigate Hofmeister effects and alkali metal ionic effects on dodecamer d(CGCGAATTCGCG) B-DNA with atomic dynamics simulations. As normal water solvent is replaced by less polarised water, the Hofmeister series of alkali metal ions is changed from Li+> Na+≃ K+≃ Cs+≃ Rb+to Li+> Na+> K+> Rb+> Cs+. In less polarised water, DNA experiences the B→A conformational transition for the lighter alkali metal counterions (Li+, Na+and K+). However, it keeps B form for the heavier ions (Rb+and Cs+). We find that the underlying cause of the conformation transition for these alkali metal ions except K+is the competition between water molecules and counterions coupling to the free oxygen atoms of the phosphate groups. For K+ions, the ‘economics’ of phosphate hydration and ‘spine of hydration’ are both concerned with the DNA helixes changing. [ABSTRACT FROM PUBLISHER]

The effects of ionic stress on the physiology and gene expression of two rice genotypes (IR4630 and IR15324) that differ in salt tolerance, were investigated by evaluating changes in the biomass, Na+ and K+ concentrations and applying the cDNA-AFLP technique to highlight changes in gene expression. Over 8 days of salinisation, the effect of NaCl on the reduction of biomass (dry weight) was apparent from 24 h after salinisation (the first time point), indicating that the consequences of the build up of Na+ (and Cl-) in the leaves of both lines was rapid. Furthermore, root growth of IR15324 was much more sensitive to salt than that of IR4630 (the reduction in root dry weight compared to non-salinised plants was three times greater in IR15324 than IR4630). The two rice lines also differed in their Na+ accumulation in saline conditions, a difference that was more marked in the shoots, particularly at the final harvest, than in the roots. Under salt stress, the K+ content (µmol/shoot) increased over four successive harvests (24, 48, 96, 192 h) in both lines, but was always greater in IR4630 than in IR15324: differences in Na+/K+ ratio appear to be an important determinant of salt tolerance in rice. To separate osmotic from ionic effects of salt, mannitol was applied as a non-ionic osmoticum at an osmotic potential estimated to be equivalent to 50 mM NaCl. Messenger RNA was sampled at 0.5, 6, 24, 48 and 192 hours after salinisation. Several products (AFLP-bands) were detected, which were upregulated in the response to ionic effects of salt in the tolerant line (IR4630) and not expressed in the sensitive line (IR15324). Bioinformatic analysis indicated three of these AFLP-bands have a high-degree of sequence similarity with the genes encoding a proline rich protein, senescence associated protein and heat-shock protein. The data are novel in that they differentially highlight changes induced by the ionic rather than osmotic effects of salt and in a tolerant rather than a sensitive genotype. Background: Salinity as a most significant environmental challenges affects the growth and productivity of plants worldwide. In this study, the ionic and iso-osmotic effects of salt stress were investigated in Aeluropus littoralis L., a halophyte grass species from Poaceae family, by cDNA-amplified fragment length polymorphism (cDNA-AFLP) technique. To dissect the two different effects (ionic and osmotic) exerted by salt stress, various ionic agents including 200 and 400 mM sodium chloride (NaCl), 200 and 400 mM potassium chloride (KCl) as well as 280 and 406 gl− 1 (− 0.9 and − 1.4 MPa) polyethylene glycol 6000 (PEG) as their iso-osmotic concentrations were applied. Results: Application of KCl and PEG significantly reduced the fresh weight (FW) of A. littoralis seedlings compared to control while NaCl treatment markedly enhanced the FW. At the transcriptome level, different observations of changes in gene expression have been made in response of A. littoralis to ionic and osmotic stresses. Out of 69 transcript derived fragments (TDFs), 42 TDFs belong to 9 different groups of genes involved in metabolism (11.6%), transcription (10.2%), ribosomal protein (8.7%), protein binding (8.7%) transporter (5.8%), translation (5.8%), signal transduction (4.3%), nucleosome assembly protein (2.9%) and catabolism (2.9%). The 44 and 28 percent of transcripts were expressed under ionic stress (NaCl-specific and KCl-specific) and osmotic stress (common with NaCl, KCl and PEG), respectively which indicating a greater response of plants to ionic stress than osmotic stress. Expression pattern of eight candidate TDFs including; SYP81, CAND1, KATN, ISB1, SAMDC, GLY1, HAK18 and ZF30 was evaluated by RT-qPCR at high salinity levels and recovery condition. Conclusion: Differential regulation of these TDFs was observed in root and shoot which confirm their role in salt stress tolerance and provide initial insights into the transcriptome of A. littoralis.
