Background: The GRAS gene family is a class of plant-specific transcription factors with important roles in many biological processes, such as signal transduction, disease resistance and stress tolerance, plant growth and development. So far, no information available describes the functions of the GRAS genes in Eucalyptus grandis. Results: A total of 82 GRAS genes were identified with amino acid lengths ranging from 267 to 817 aa, and most EgrGRAS genes had one exon. Members of the GRAS gene family of Eucalyptus grandis are divided into 9 subfamilies with different protein structures, while members of the same subfamily have similar gene structures and conserved motifs. Moreover, these EgrGRAS genes expanded primarily due to segmental duplication. In addition, cis-acting element analysis showed that this family of genes was involved involved in the signal transduction of various plant hormones, growth and development, and stress response. The qRT-PCR data indicated that 18 EgrGRAS genes significantly responded to hormonal and abiotic stresses. Among them, the expression of EgrGRAS13, EgrGRAS68 and EgrGRAS55 genes was significantly up-regulated during the treatment period, and it was hypothesised that members of the EgrGRAS family play an important role in stress tolerance. Conclusions: In this study, the phylogenetic relationship, conserved domains, cis-elements and expression patterns of GRAS gene family of Eucalyptus grandis were analyzed, which filled the gap in the identification of GRAS gene family of Eucalyptus grandis and laid the foundation for analyzing the function of EgrGRAS gene in hormone and stress response. As one of the largest megacity clusters in China, the Pearl River Delta (PRD) exhibited an obvious increase in the autumn mean maximum daily 8 hr average (MDA8) ozone (O3) concentration from 2014 to 2020, although O3 precursors were effectively controlled. To understand the cause of these elevated surface MDA8 O3 concentrations in the PRD, we explored potential meteorological causes. Based on the relationships between the MDA8 O3 concentrations and the meteorological conditions, the ozone weather index (OWI) was constructed from surface downward shortwave radiation flux (DSWRF), relative humidity (RH), and wind speed (WS) data to represent the variations in the autumn MDA8 O3 concentration in the PRD. Stronger DSWRF, lower RH, and lower WS were linked to larger OWI values and stronger O3 pollution. Discrepancies in DSWRF, RH, and WS were observed on days with OWI > 1 and OWI < −1. Atmospheric stagnation (AS) was also associated with O3 pollution. The average MDA8 O3 concentrations on AS days were 53% (51.0 μg m−3) higher than those on non‐AS days. The O3 exceedance rates on AS days were 36.5%, which is 4.8 times higher than those on non‐AS days (7.6%) in the PRD. In addition to lower WS and less precipitation, AS days were also accompanied by lower total cloud cover and stronger downdraft airflow, which accelerated O3 production. Since the occurrence of AS is projected to increase, efforts to mitigate AS would contribute to decreasing the risk of severe autumn O3 pollution events in the PRD. Purpose: To investigate the diagnostic value of the Prostate Imaging Reporting and Data System version 2.1 (PI-RADS v2.1) for clinically significant prostate cancer (CsPCa). We also aimed to combine PI-RADS v2.1 with prostate-specific antigen (PSA) derivatives to improve the predictive value of CsPCa. Methods: We retrospectively collected relevant data who underwent standard MRI examinations of the prostate and subjected to a prostate biopsy at Shenzhen People's hospital from November 2014 to November 2019. Included 125 cases of CsPCa and 383 cases of non-CsPCa. All cases were scored using the PI-RADS v2.1. The clinical data collected included age, PSA, free PSA/total PSA, prostate volume and PSA density (PSAD). A univariate analysis was performed to identify statistically significant indicators. Logistic regression was used to analyze the predictive value of the multi-parameter combination on CsPCa. Results: Except age, the difference in all of indicators between the CsPCa group and non-CsPCa group was statistically significant. The PI-RADS score and PSAD value had the highest diagnostic value. Logistic regression analysis revealed that the PI-RADS score and PSAD value were independent predictors of CsPCa, with a regression model AUC of 0.935. CsPCa detection rates were low when the PI-RADS score ≤ 2 or the PI-RADS score = 3 and the PSAD value ≤ 0.33 ng/ml/ml. Conclusion: Combining the PI-RADS score and PSAD value improved the predictive performance of CsPCa. Traffic congestion has become a major problem restricting the development of major cities. ITS (Intelligent Transportation System) can record the state of traffic and predict the future traffic state, then reasonably optimize the travel scheme, so as to achieve the purpose of alleviating traffic congestion. Meanwhile, traffic flow prediction can provide data support for ITS, so many researchers have done a lot of research on traffic flow prediction. Many researchers take the traffic network as an undirected graph, and use the GCN (Graph Convolution Network) model to study the traffic flow prediction, and have achieved good prediction results. However, the traffic network is directed, and the traffic network is regarded as an undirected graph, which loses the direction information of the road network. Therefore, this inspires us to propose a graph convolution operator DGCN (Directed GCN), which can make full use of the in degree and out degree information of each station in the traffic network. Background: Robotic colorectal cancer surgery is widely accepted and applied. However, there is still no objective and comprehensive assessment on the data of nationwide multicenter series. Method: A total of 28 medical centers in Mainland China participated in this nationwide retrospective observational study. From the first case performed in each center to the last until December 2017, patients with robotic resection for primary tumor and pathologically confirmed colorectal adenocarcinoma were consecutively enrolled. Clinical, pathological and follow-up data were collected and analyzed. Results: A total of 5389 eligible patients were finally enrolled in this study, composing 72.2% of the total robotic colorectal surgery volume of Mainland China in the same period. For resections of one bowel segment of primary tumor, the postoperative mortality rate was 0.08% (4/5063 cases), and the postoperative complication rate (Clavien–Dindo grade II or higher) was 8.6% (434/5063 cases). For multiple resections, the postoperative mortality rate was 0.6% (2/326 cases), and the postoperative complication rate was 16.3% (53/326 cases). Out of 2956 patients receiving sphincter-preserving surgery in only primary resection, 130 (4.4%) patients had anastomotic leakage. Traditional low anterior resection (tumor at middle rectum) (OR 2.384, P < 0.001), traditional low anterior resection (tumor at low rectum) (OR 1.968, P = 0.017) and intersphincteric resection (OR 5.468, P = 0.006) were significant independent risk factors for anastomotic leakage. Female gender (OR 0.557, P = 0.005), age ≥ 60 years (OR 0.684, P = 0.040), and preventive stoma (OR 0.496, P = 0.043) were significant independent protective factors. Body mass index, preoperative chemotherapy/radiotherapy, tumor size, and TNM stage did not independently affect the occurrence of anastomotic leakage. Background: Leptospirosis is one of the most important neglected tropical bacterial diseases worldwide. However, there is limited information on the genetic diversity and host selectivity of pathogenic Leptospira in wild small mammal populations. Methodology/Principal findings: Jiangxi Province, located in southern China, is a region highly endemic for leptospirosis. In this study, among a total of 3,531 trapped rodents dominated by Apodemus agrarius (59.7%), 330 Leptospira strains were successfully isolated from six different sites in Jiangxi between 2002 and 2015. Adding 71 local strains from humans, various kinds of livestock and wild animals in Jiangxi, a total of 401 epidemic strains were characterized using 16S rRNA gene senquencing, multilocus sequence typing (MLST) and the microscopic agglutination test (MAT). Among them, the most prevalent serogroup was Icterohaemorrhagiae (61.10%), followed by Javanica (19.20%) and Australis (9.73%); the remaining five serogroups, Canicola, Autumnalis, Grippotyphosa, Hebdomadis and Pomona, accounted for 9.97%. Species identification revealed that 325 were L. interrogans and 76 were L. borgpetersenii. Moreover, L. interrogans was the only pathogenic species in Fuliang and Shanggao and was predominant in Shangrao (95.0%); L. borgpetersenii was the most common in the remaining three sites. Twenty-one sequence types (STs) were identified. Similarly, ST1 and serogroup Icterohaemorrhagiae were most prevalent in Shangrao (86.0% and 86.4%) and Fuliang (90.4% and 90.4%), ST143 and serogroup Javanica in Shangyou (88.5% and 90.4%) and Longnan (73.1% and 73.1%), and ST105 and serogroup Australis in Shanggao (46.3% and 56.1%). Serogroup Icterohaemorhagiae primarily linked to A. agrarius (86.9%), serogroup Canicola to dogs (83.3%). There were significant differences in the distribution of leptospiral species/serogroups/STs prevalence across host species/collected locations among the 394 animal-associated strains (Fisher’s exact test, p<0.001). Conclusions/Significance: Our study demonstrated high genetic diversity of pathogenic Leptospira strains from wild small animals in Jiangxi from 2002 to 2015. A. agrarius was the most abundantly trapped animal reservoir, and serogroup Icterohaemorrhagiae and ST1 were the most dominant in Jiangxi. Significant geographic variation and host diversity in the distribution of dominant species, STs and serogroups were observed. Moreover, rat-to-human transmission might play a crucial role in the circulation of Leptospirosis in Jiangxi. Details of the serological and molecular characteristics circulating in this region will be essential in implementing prevention and intervention measures to reduce the risk of disease transmission in China. However, phylogenetic analysis of more Leptospira isolates should explore the impact of ecological change on leptospirosis transmission dynamics and investigate how such new knowledge might better impact environmental monitoring for disease control and prevention at a public health level. The liver is the most common anatomical site for hematogenous metastases of colorectal cancer, and colorectal liver metastases is one of the most difficult and challenging points in the treatment of colorectal cancer. To improve the diagnosis and comprehensive treatment in China, the Guidelines have been edited and revised several times since 2008, including the overall evaluation, personalized treatment goals and comprehensive treatments, to prevent the occurrence of liver metastases, improve the resection rate of liver metastases and survival. In the existing bandwidth-based methods, through traffic flows are considered as the coordination objects and offered progression bands accordingly. However, at certain times or nodes in the road network, when the left-turn traffic flows have a higher priority than the through traffic flows, it would be inappropriate to still provide the progression bands to the through traffic flows; the left-turn traffic flows should instead be considered as the coordination objects to potentially achieve better control. Considering this, a general maximum progression model to concurrently synchronize left-turn and through traffic flows is established by using a time-space diagram. The general model can deal with all the patterns of the left-turn phases by introducing two new binary variables into the constraints; that is, these variables allow all the patterns of the left-turn phases to deal with a single formulation. By using the measures of effectiveness (average delay time, average vehicle stops, and average travel time) acquired by a traffic simulation software, VISSIM, the validity of the general model is verified. The results show that, compared with the MULTIBAND, the proposed general model can effectively reduce the delay time, vehicle stops, and travel time and, thus, achieve better traffic control. Objective: The aim of this study was to evaluate the efficacy of early continuous veno-venous hemofiltration (CVVH) in decreasing the intra-abdominal pressure (IAP) and serum interleukin-8 (IL-8) level in severe acute pancreatitis (SAP) patients with abdominal compartment syndrome (ACS). Methods: Twenty-five ACS patients of SAP were enrolled in a prospective study conducted according to the standard management protocol. They were treated in the intensive care unit (ICU) of Affiliated Yidu Central Hospital of Weifang Medical College and underwent CVVH. Eleven patients were set up as the control group that received no hemofiltration and surgical treatment due to economic or other reasons but solely conventional treatment. Serum amylase, liver and kidney function, and C reactive protein were investigated before and after treatment. IAP and blood level of IL-8 were measured daily to investigate their time course of changes and the correlation between the 2 parameters. Results: Serum amylase levels, C-reactive protein and IAP were significantly lower and liver and kidney function was significantly better than those of the control group (p < 0.05). IAP on admission to the ICU was high, at 22.9 ± 2.1 mm Hg. The IAP was significantly lower to 17.2 ± 2.2 mm Hg (p < 0.01) 24 h after the initiation of CVVH, and thereafter decreased rapidly. The average blood level of IL-8 was high at 88.2 ± 25.1 ng/L on admission. However, it significantly decreased to 63.2 ± 18.7 ng/L (p < 0.01) 24 h after the initiation of CVVH, and subsequently decreased. There was a significant positive correlation between the blood level of IL-8 and IAP(r = 0.62, p < 0.01). Conclusions: CVVH is effective to decrease the IAP and the blood level of IL-8 in ACS patients of SAP. The blood level of IL-8 was significantly correlated with IAP, suggesting that IL-8 might play an important role in the pathogenesis of ACS. Early CVVH appeared to be effective in the treatment of ACS in patients with SAP through the removal of causative cytokines such as IL-8, and it thereby decreased interstitial edema to lower IAP and should be applied in the early stage of ACS. Connected-vehicles network provides opportunities and conditions for improving traffic signal control, and macroscopic fundamental diagrams (MFD) can control the road network at the macrolevel effectively. This paper integrated proposed real-time access to the number of mobile vehicles and the maximum road queuing length in the Connected-vehicles network. Moreover, when implementing a simple control strategy to limit the boundary flow of a road network based on MFD, we determined whether the maximum queuing length of each boundary section exceeds the road-safety queuing length in real-time calculations and timely adjusted the road-network influx rate to avoid the overflow phenomenon in the boundary section. We established a road-network microtraffic simulation model in VISSIM software taking a district as the experimental area, determined MFD of the region based on the number of mobile vehicles, and weighted traffic volume of the road network. When the road network was tending to saturate, we implemented a simple control strategy and our algorithm limits the boundary flow. Finally, we compared the traffic signal control indicators with three strategies: (1) no control strategy, (2) boundary control, and (3) boundary control with limiting queue strategy. • A novel analysis method based on the complex-factor model is proposed to analyze the vibration characteristics of the MSB due to the unbalance terms, and the precise vibration models are established. • The cross-feedback control model using the complex-exponential function method is proposed to effectively control the low-frequency precession and the high-frequency nutation of the MSB. • In comparison to the commonly used complex-factor cross-feedback model, the proposed complex-exponential function cross-feedback model could further reduce the precession amplitude by 38.88%, and mitigate the nutation by 60.42%. The research of vibration characteristics and active control methods of the magnetically suspended blower (MSB) are conducted for mitigation of the vibration amplitudes and the dynamic displacements of rotor shaft of the MSB at high speed. Firstly, the dynamic models with four degrees of freedom (DOFs) of the rotor shaft in the MSB are established. Secondly, the vibration characteristics of rotor shaft considering the rotor unbalance and misalignment are analyzed using the complex-factor model. Furthermore, the complex-factor and complex-exponential function cross-feedback models are respectively used to suppress the vibration amplitudes and the dynamic displacements of rotor shaft. Both numerical simulation and experimental tests of the cross-feedback model are conducted to verify the models. It is shown that a wide cutoff frequency of low-pass filter (LPF) could mitigate the precession, and a wide cutoff frequency of high-pass filter (HPF) could suppress the nutation. The proposed complex-exponential function cross-feedback model could be used to reduce the precession amplitude of the rotor shaft by 38.88%, and mitigate its nutation by 60.42%. Assisted reproductive technology is widely accepted as an effective treatment to improve female fertility, but the decline of aging oocyte quality remains an important factor in the decrease of female fecundity. However, the effective strategies for improving oocyte aging are still not well understood. In the study, we demonstrated that ROS content and abnormal spindle proportion were increased and mitochondrial membrane potential was decreased in aging oocytes. However, supplementation of α-ketoglutarate (α-KG), an immediate metabolite in the tricarboxylic acid cycle (TCA), for 4 months to aging mice, significantly increased the ovarian reserve showed by more follicle numbers observed. In addition, the oocyte quality was significantly improved, as demonstrated by reduced fragmentation rate and decreased reactive oxygen species (ROS) levels, in addition to a lower rate of abnormal spindle assembly, thereby improving the mitochondrial membrane potential. Consistent with the in vivo data, α-KG administration also improved the post-ovulated aging oocyte quality and early embryonic development by improving mitochondrial functions and reducing ROS accumulation and abnormal spindle assembly. With the rapid development of localization techniques and the prevalence of mobile devices, massive amounts of trajectory data have been generated, playing essential roles in areas of user analytics, smart transportation, and public safety. Measuring trajectory similarity is one of the fundamental tasks in trajectory analytics. Although considerable research has been conducted on trajectory similarity, the majority of existing approaches measure the similarity between two trajectories by calculating the distance between aligned locations, leading to challenges related to uncertain trajectories (e.g., low and heterogeneous data sampling rates, as well as location noise). To address these challenges, we propose Contra, a convolution-based similarity measure designed specifically for uncertain trajectories. The main focus of Contra is to identify the similarity of trajectory shapes while disregarding the time/order relevance of each record within the trajectory. To this end, it leverages a series of convolution and pooling operations to extract high-level geo-information from trajectories, and subsequently compares their similarities based on these extracted features. Moreover, we introduce efficient trajectory index strategies to enhance the computational efficiency of our proposed measure. We conduct comprehensive experiments on two trajectory datasets to evaluate the performance of our proposed approach. The experiments on both datasets show the effectiveness and efficiency of our approach. Abstract: The interparticle distance of CdSe(ZnS) quantum dots was accurately controlled by polymerization at the air–water interface which provided an increased homogeneity of the Langmuir–Blodgett film leading to a surface smoothness comparable to mica. The choice of a silane derivative is based on the fact that silicon is semiconductor, and the compound CdSe being the core of the quantum dot is also semiconductor. The combination of the two semiconductors could bring some unusual conduction properties as a polymeric silanized network. But first, it is most important to characterize the smoothness of the surface, which might be correlated to the formation of “trap” states, i.e. the photo-excited electron can fall, or the photo-excited hole can “float”. Quantum dots (QDs) have not been used to label cytoskeletonstructure of live cells owing to limitations in delivery strategies,and QDs conjugation methods and issues with nonspecific binding. We conjugated tubulin to QDs and applied the emerging method of photothermal nanoblade to deliver QD-tubulin conjugates into live Hela cells. We developed a new peptide, natural phytochelatin (PC),which tightlybinds to CdSe/ZnS quantum dots’ (QDs) surfaces and rendersthem water-soluble. Coating QDs with this flexible and all-hydrophilicpeptide offers high colloidal stability, adds only 0.8–0.9nm to the radius of the particles (as compared to their original inorganicradius), preserves very high quantum yield (QY) in water, and affordsfacile bioconjugation with various functional groups. There has been a lack of quick, simple and reliable methods for determination of nanoparticle size. An investigation of the size of hydrophobic (CdSe) and hydrophilic (CdSe/ZnS) quantum dots was performed by using the maximum position of the corresponding fluorescence spectrum. It has been found that fluorescence spectroscopy is a simple and reliable methodology to estimate the size of both quantum dot types. For a given solution, the homogeneity of the size of quantum dots is correlated to the relationship between the fluorescence maximum position (FMP) and the quantum dot size. This methodology can be extended to the other fluorescent nanoparticles. The employment of evolving factor analysis and multivariate curve resolution-alternating least squares for decomposition of the series of quantum dots fluorescence spectra recorded by a specific measuring procedure reveals the number of quantum dot fractions having different diameters. The size of the quantum dots in a particular group is defined by the FMP of the corresponding component in the decomposed spectrum. These results show that a combination of the fluorescence and appropriate statistical method for decomposition of the emission spectra of nanoparticles may be a quick and trusted method for the screening of the inhomogeneity of their solution. Abstract: Cyclosporine A (CsA) is an immunosuppressive drug commonly used for maintaining chronic immune suppression in organ transplant recipients. It is known that patients receiving CsA manifest increased growth of aggressive non-melanoma skin cancers. However, the underlying mechanism by which CsA augments tumor growth is not fully understood. Here, we show that CsA augments the growth of A431 epidermoid carcinoma xenograft tumors by activating tumor growth factor β-activated kinase1 (TAK1). The activation of TAK1 by CsA occurs at multiple levels by kinases ZMP, AMPK and IRAK. TAK1 forms heterodimeric complexes with TAK binding protein 1 and 2 (TAB1/TAB2) which in term activate nuclear factor κB (NFκB) and p38 MAP kinase. Transcriptional activation of NFκB is evidenced by IKKβ-mediated phosphorylation-dependent degradation of IκB and consequent nuclear translocation of p65. This also leads to enhancement in the expression of its transcriptional target genes cyclin D1, Bcl2 and COX-2. Similarly, activation of p38 leads to enhanced inflammation-related signaling shown by increased phosphorylation of MAPKAPK2 and which in turn phosphorylates its substrate HSP27. Abstract: The frequency shift of the ultra-narrow frequency shift keying (UNFSK) is far less than the ordinary binary frequency shift keying (FSK) method. Because the two frequencies of UNFSK are extremely close, the traditional demodulation method cannot separate them successfully. This study aims to successfully demodulate the UNFSK signals by applying cycle extension and notch filter. The signal processing procedure is as follows: First, the sampling signals have been made cycle extension to enlarge their spectrum difference between signal 0 and 1; secondly, the cycle extension signals have been processed by the notch filter algorithm and finally, we could get the information by sentencing the filtering result. Finally, the method is applied in transportation. The simulation results proved that the algorithm greatly enhanced UNFSK signal demodulation performance, compared to the traditional demodulation method. Abstract: TRB3, a human homolog of Drosophila Tribbles, has been recently shown as a critical negative regulator of Akt and S6 kinase activation in a number of cellular processes. Here we found that TRB3 interacted with an important cell cycle regulator CtIP (CtBP-interacting protein) and the interaction involved the C-terminus of both proteins. Interestingly, TRB3 and CtIP co-localized to the nucleus in HeLa cells and exhibited a unique dot-like pattern. Finally, we demonstrated that TRB3 was overexpressed in multiple tumor tissues. Since CtIP plays important roles in cell cycle checkpoint control and it has been implicated in tumorigenesis, our data suggest that TRB3 may be involved in these biological processes through interacting with CtIP. Abstract: Trioctylphosphine oxide (TOPO) capped CdSe and (CdSe)ZnS quantum dots (QDs) were prepared. The surface chemistry behavior of both QDs at the air–water interface was carefully examined by various physical measurements. Stable Langmuir films were formed for both QDs. The average limiting nanoparticle area derived from the π–A isotherms of the QDs could be used to estimate the average size of the QDs if the thickness of TOPO shell was counted. The epifluorescence image of the QDs Langmuir films revealed an aggregation in 2D during the early stage of the compression process. On the other hand, a diacetylene-peptide (PDA-Cys-Cys-Gly-OH) derivative, namely PDA-CCG, was used to modify the surface of QDs. Ozonization, a novel surface modification method, has been developed to improve hemocompatibility of polymeric biomaterials. The silicone film was exposed to ozone and then followed by grafted polymerization with 2-methacryloyloxyethyl phosphorylcholine (MPC). The surface graft polymerization was confirmed by X-ray photon spectroscopy and ATR-FTIR. Surface hydrophilicity was investigated by measuring the contact angles and MPC polymer grafted silicone film show a significant increase in hydrophilicity. The blood compatibility of the grafted film was evaluated by a platelet-rich plasma (PRP) adhesion study and scanning electron microscopy, using silicone film as the reference. No platelet adhesion was observed on the grafted film incubated with PRP at 37 °C for 60 and 180 min. The ozonization is an effective method to graft MPC onto the silicone surface with many advantages and therefore very useful for practical application of blood contacting medical devices. The average annual daily traffic (AADT) volumes can be estimated by using a short period count of less than twenty-four hour duration. In this paper, the neural network method is adopted for the estimation of AADT from short period counts and for the determination of the most appropriate length of counts. A case study is carried out by analysing data at thirteen locations on trunk roads and primary roads in urban area of Hong Kong. The estimation accuracy is also compared with the one obtained by regression analysis approach. Studies were conducted to characterize soil humin by acid hydrolysis. Two humin samples collected from two different types of soil, namely chernozem and laterite, which are widespread over a vast area from the north to south of China, were hydrolyzed under reflux with 0.5M H[sub 2]SO[sub 4] or 3M H[sub 2]SO[sub 4] for 4 h. The results showed that 25%-29% of organic carbon and 46%-54% of organic nitrogen could be hydrolyzed by 0.5M H[sub 2]SO[sub 4]; 36%-40% of organic carbon and 93% - 97% of organic nitrogen hydro1yzed by 3M H[sub 2]SO[sub 4]. The C/N ratio in hydrolyzed organic matter is lower than that in soil humin and that in organic matter hydrolyzed by 3M H[sub 2]SO[sub 4] is lower than in organic matter hydrolyzed by 0.5M H[sub 2]SO[sub 4]. The proportion of nitrogen hydrolyzed from humin is markedly larger than that from the original soil and also markedly larger than that from humic acid fraction. Only 3%-7% of nitrogen in humin exists in a relatively stable form, which is not easy to hydrolyze. There is little nitrogen that occurs in the form of heterocyclic rings in humin. Background: To investigate the clinical efficacy and outcomes of skin stretch suturing with self-locking sliding Nice knots in the treatment of small- or medium-sized wounds. Methods: From June 2015 to May 2018, 26 patients with small- or medium-sized wounds were included in the present study. Skin stretch suturing with self-locking slide Nice knots was performed to gradually close the soft-tissue defects in these patients. The time of wound closure and healing was recorded. The color and blood supply of the skin, cutaneous sensation, the stretch of skin, and the hair growth situation of the skin wound were observed and recorded. Results: There were 17 males and 9 females with an average age of 30.65 years (range, 15–48 years). The areas of the soft-tissue defects were between 3.2 × 7.1 cm and 8.0 × 15.2 cm. All patients underwent stretch suturing with self-locking slide Nice knots to close the soft-tissue defects. All wounds were successfully closed and healed. The mean time of wound closure was 10.69 days (range, 5–20 days), and the mean time of wound healing was 16.85 days (range, 10–24 days). The cutaneous sensation of skin wound recovered normally, and the color of the skin wounds was the same as that of normal skin at the last follow-up. The hair growth situation of the skin wounds also returned to normal. Conclusions: This study revealed that Nice knots yielded an accepted clinical result as a new method to close small- or medium-sized wounds that was simple and less minimally invasive, resulted in progressive tension, did not return to previous results, and partially replace flaps or free skin grafts. Eucalyptus grandis is an important planted hardwood tree worldwide with fast growth and good wood performance. The nitrate transporter (NRT) gene family is a major core involved in nitrogen (N) absorption and utilization in plants, but the comprehensive characterization of NRT genes in E. grandis remains mostly elusive. In this study, a total of 75 EgNRT genes were identified from the genome of E. grandis that were distributed unevenly across ten chromosomes, except Chr9. A phylogenetic analysis showed that the EgNRT proteins could be divided into three classes, namely NRT1, NRT2 and NRT3, which contained 69, 4 and 2 members, respectively. The cis-regulatory elements in the promoter regions of EgNRT genes were mainly involved in phytohormone and stress response. The transcriptome analysis indicated that the differentially expressed genes of leaf and root in E. grandis under different N supply conditions were mainly involved in the metabolic process and plant hormone signal transduction. In addition, the transcriptome-based and RT-qPCR analysis revealed that the expression of 13 EgNRT genes, especially EgNRT1.3, EgNRT1.38, EgNRT1.39 and EgNRT1.52, was significantly upregulated in the root under low-N-supply treatment, suggesting that those genes might play a critical role in root response to nitrate deficiency. Taken together, these results would provide valuable information for characterizing the roles of EgNRTs and facilitate the clarification of the molecular mechanism underlying EgNRT-mediated N absorption and distribution in E. grandis. Surface ozone (O 3) pollution in the Pearl River Delta (PRD), especially persistent O 3 pollution episodes lasting three days or longer (OPE3), showed an anomalous peak during the autumn of 2019 (bulge-2019). Our research reveals a robust connection between the occurrence of OPE3 events and days with low cloud cover. Specifically, low-cloud days comprised 55% of the total number of OPE3 events in the PRD during the autumn season spanning from 2015 to 2021. Notably, as much as 74% of the bulge-2019 is related to low-cloud days. However, establishing a direct causal link remains challenging. In autumn 2019, an anomalous cyclonic center developed over the Philippine Sea, with anomalous northerly winds on its northwestern flank, leading to anomalous downdraft over South China. This pattern was characterized by stagnant, low relative humidity, intensified solar radiation, and low cloud cover conditions that were conducive to the photochemical formation and accumulation of O 3. Further analysis of the relationship between low-cloud days and El Niño–Southern Oscillation shows that eastern Pacific El Niño events are associated with fewer low-cloud days in the PRD in autumn, whereas central Pacific (CP) El Niño events result in more low-cloud days. In addition, the anomalous cyclonic center over the Philippine Sea serves as the crucial atmospheric circulation system connecting CP El Niño and high O 3 level over southern China. Therefore, we propose that the increased downdraft of PRD brought by northward tropical cyclones (TCs) during more low-cloud days in 2019 is highly likely the primary cause of the occurrence of the bulge-2019. • Low-cloud days comprised 55% of PRD OPE3 events in 2015–2021 autumn, specially reaching 74% in 2019. • The anomalous cyclonic center over the Philippine Sea notably controlled PRD's autumn 2019 O 3 levels. • Increased downdrafts by northward tropical cyclones in low-cloud days might have caused the 2019 bulge in O 3 levels in PRD. Light is an important factor for plant development and has serious effects on the growth, production and quality of potatoes. However, the physical and molecular mechanisms by which potato plantlets cope with different light qualities are not understood. In this study, the potato “Zhuanxinwu”, which is a germplasm potato resource with a high anthocyanin content, was used for physiological and transcriptome profiling analyses to uncover the different mechanisms that occur in response to blue, red and white light conditions, with the white light condition serving as the control. Multiple growth indexes, protective enzyme activity and metabolite accumulation were measured. The results indicated that white light promoted a shift in biomass allocation away from tubers to leaves to enhance dry leaf matter and reduce tuber fresh/dry weight relative to the effects of blue or red light. The leaf area and anthocyanin content values were greater for plants grown in blue light than those grown in white or red light, suggesting that combinations of different spectra were more conducive to regulating potato growth. A total of 2220 differentially expressed genes (DEGs) were found among the three samples, and the DEGs in the three comparison sets were analyzed. A total of 1180 and 984 DEGs were identified in the red light (Red) and blue light (Blue) conditions compared to the control condition, respectively, and 359 DEGs overlapped between the two comparison sets (Blue_vs_White and Red_vs_White). Interestingly, the 24 most common overlapped DEGs were involved in photosynthesis, respiration, and reactive oxygen species (ROS) scavenging. Of these DEGs, four genes involved in photosynthesis and two genes involved in pigment synthesis were highly expressed, implying that some genes could be implemented to cope with different light spectra by regulating the expression of DEGs involved in the corresponding metabolic pathways. In conclusion, our study characterizes physiological responses of potato to different light qualities and identifies potential pathways and candidate genes involved in these responses, thus providing a basis for further research on artificial light regulation of potato plant growth. At present, enzyme debridement preparation has shown a good curative effect on eschar removal of burn wounds. Keratinase has shown great potential in enzymatic debridement because of its good fibrin-degrading ability. In this study, the debridement of keratinase was examined by using a third degree burn wound model in rats. We observed the wound, and keratinase shortened the time of eschar dissolution after debridement. Histopathology and immunofluorescence staining showed that the eschar in the keratinase group became thinner, inflammatory cell infiltration in the wound increased, the fluorescence intensity of the macrophage surface marker CD68 increased, and the CD163/CD86 ratio increased. In bone marrow-derived macrophages (BMDMs), there was no significant difference in the activity of CCK-8 in cells in the keratinase group compared with the control group. The fluorescence intensity of the keratinase group was higher than that of the control group. At 12 h, the cell scratches were obviously closed. The number of migrated Transwell cells increased. Flow cytometry and immunofluorescence analysis showed increased expression of CD206 and Arg-1 and decreased expression of CD86 and iNOS. The gene expression of the Arg-1, iNOS and IL-10 was increased, as shown by qPCR. The secretion of IL-10 was increased and TNF-α was decreased, as shown by ELISA. We concluded that keratinase dissolution of eschar not only has a hydrolytic effect on eschar but may also affect immune regulation to enhance the migration and phagocytosis of macrophages, promote the polarization of macrophages, and further enhance the effect of eschar dissolution. Background: Malaria was once widespread in Guangzhou, China. However, a series of control measures have succeeded in eliminating local malaria infections. Based on the analysis of the characteristics of malaria epidemics in Guangzhou, China, from 1950 to 2022, the changes and effectiveness of malaria control strategies and surveillance management in Guangzhou from 1950 to 2022 are described. Methods: Data on malaria prevention and treatment in Guangzhou from 1950 to 2022 were collected, and descriptive epidemiological methods were used to analyse the prevalence of malaria, preventive and control measures taken, and the effectiveness of prevention and treatment in different periods. Data on malaria cases were obtained from the Guangzhou Centre for Disease Control and Prevention (CDC) and the China Communicable Disease Reporting System. Results: The development of the malaria control system in Guangzhou has gone through four periods: 1. High malaria prevalence (1950–1979), 2. Intensive prevention and control stage (1980–2000), 3. Consolidating gains in malaria control (2001–2008), and 4. Preventing reestablishment of transmission (2009–2022). During Period 1, only medical institutions at all levels and the local CDCs, the Guangzhou CDC participated in the malaria prevention and control system, establishing a three-tier health system on malaria prevention and control. During Period 2, other types of organizations, including the agricultural sector, schools and village committees, the construction department and street committee, are involved in the malaria control system. During Period 3, more and more organizations are joining forces to prevent and control malaria. A well-established multisectoral malaria control mechanism and an improved post-elimination surveillance management system are in place. Between 1950 and 2022, a total of 420,670 cases of malaria were reported. During Period 1, there was an epidemic of malaria in the early 1950s, with an annual incidence rate of more than 10,000/100,000, including a high rate of 2887.98/100,000 in 1954. In Period 2 malaria was gradually brought under control, with the average annual malaria incidence rate dropping to 3.14/100,000. During Period 3, the incidence rate was kept below 1/100,000, and by 2009 local malaria infections were eliminated. Conclusion: For decades, Guangzhou has adopted different malaria control strategies and measures at different epidemic stages. Increased collaboration among civil organizations in Guangzhou in malaria control has led to a significant decline in the number of malaria cases and the elimination of indigenous malaria infections by 2009.The experience of Guangzhou can guide the development of malaria control strategies in other cities experiencing similar malaria epidemics. Recent advances highlight the pivotal role of nicotinamide adenine dinucleotide (NAD+) in ovarian aging. However, the roles of de novo NAD+ biosynthesis on ovarian aging are still unknown. Here, we found that genetic ablation of Ido1 (indoleamine‐2,3‐dioxygenase 1) or Qprt (Quinolinate phosphoribosyl transferase), two critical genes in de novo NAD+ biosynthesis, resulted in decreased ovarian NAD+ levels in middle‐aged mice, leading to subfertility, irregular estrous cycles, reduced ovarian reserve, and accelerated aging. Moreover, we observed impaired oocyte quality, characterized by increased reactive oxygen species and spindle anomalies, which ultimately led to reduced fertilization ability and impaired early embryonic development. A transcriptomic analysis of ovaries in both mutant and wild‐type mice revealed alterations in gene expression related to mitochondrial metabolism. Our findings were further supported by the observation of impaired mitochondrial distribution and decreased mitochondrial membrane potential in the oocytes of knockout mice. Supplementation with nicotinamide riboside (NR), an NAD+ booster, in mutant mice increased ovarian reserve and improved oocyte quality. To investigate the relationship between managerial compensation and research and development (R&D) investment under asymmetric information, this paper establishes a two-period principal-agency model, in which the firm owner hires a risk-averse manager to operate the firm and to make the R&D investment decision. The manager has private information regarding the profitability of the R&D investment. Firstly, we present an R&D investment problem under symmetric information as the benchmark. Then, we discuss the scenario with asymmetric information about the profitability, and the firm owner offers new optimal incentive contact. We compare the differences of the company’s R&D investment decisions and choices of the compensation contract under symmetric and asymmetric information cases by numerical experiments. We find that the manager receives an upward distortion in the first-period bonus share rate, and a downward distortion in the second-period bonus share rate. Meanwhile, the R&D investment is less than the symmetric information case. At last, the variable outside option has no effect on the optimal manager’s compensation contract when the marginal cost of R&D investment is relatively small or under symmetric information case. The prevention and treatment efficiency of ethanol-induced gastric epithelial injury are not satisfied. We have previously shown that AMP-activated protein kinase (AMPK) activation exerts a pro-survival function in human gastric epithelial cells (GECs). miroRNA-451 (“miR-451”)’s inhibitor, antagomiR-451, can activate AMPK signaling. In the present study, we show that forced-expression of antagomiR-451 via a lentiviral vector depleted miR-451, leading to AMPK activation in established GES-1 cells and primary human GECs. AntagomiR-451 efficiently protected GES-1 cells and primary human GECs from ethanol-induced viability reduction and apoptosis. AMPK activation is required for antagomiR-451-induced GEC protection. AMPKα1 knockdown (by targeted-shRNAs) or knockout (by CRISPR-Cas-9 KO plasmid) blocked antagomiR-451-induced AMPK activation, and GEC protection against ethanol. Further experimental results show that antagomiR-451 significantly attenuated ethanol-induced reactive oxygen species (ROS) production, lipid peroxidation and DNA damage. Background: The prevalence of colorectal cancer (CRC) worldwide is a huge challenge to human health. Primary tumor locations found to impact prognosis and response to therapy. The important role of gut microbiota in the progression and treatment of CRC has led to many attempts of alleviating chemotherapy-induced adverse effects using microecologics. However, the underlying mechanism of the difference in the prognosis of different primary tumor locations and the synergistic effect of prebiotics on chemotherapy need to be further elucidated. This study aims to explore the differences in tumor microbiota and examine the effectiveness of xylooligosaccharides (XOS) on gut microbiota, adverse effects, and bioavailability of chemotherapy drugs in CRC patients at different primary tumor locations. Methods: This is a double-blinded, randomized, parallel controlled clinical trial. Participants with left-sided CRC (LSCRC, n = 50) and right-sided CC (RSCC, n = 50) will randomly allocated to prebiotic group (n = 25) or control group (n = 25) and will receive either a daily XOS (3 g/day) or placebo, respectively, for 12 weeks. The primary outcomes will be the differences in the mucosa microbiota composition at different tumor locations and differences in gut microbiota composition, adverse effects, and blood concentration of capecitabine posttreatment. The secondary outcomes will include other blood indicators, short-chain fatty acids (SCFAs) concentration, quality of life, and mental health. Discussion: This study will reveal the potential benefits of prebiotic for improving the gut microbiota composition, alleviating the adverse effects, and improving the efficacy of chemotherapy in patients with CRC. In addition, this study will provide data on the different distribution of tumor microbiota and the different changes of gut microbiota during treatment in LSCRC and RSCC, which may provide novel insights into personalized cancer treatment strategies based on primary tumor locations and gut microbiota in the future. Background: The number of human rabies cases caused by pet dogs in Guangzhou has been decreasing after years of comprehensive interventions. Consequently, attacks by stray dogs become a major issue in rabies control. Objectives: To share our experience of successfully dealing with rabies to provide some inspiration for prevention and control in countries and regions affected by it. Methods: A multidisciplinary One Health response was initiated to control this outbreak. Rabies virus was detected by PCR in the brain tissue of the associated stray dog. The sequences were aligned with reference sequences downloaded from GenBank using ClustalX. The maximum likelihood method implemented in MEGA 5.0 software package was used in a phylogenetic analysis of the aligned sequences. Results: Twelve patients with exposure to the stray dog were identified in the field investigation. Rabies vaccines and immunoglobulin were administered to all patients within 48 h. After 1 year of follow‐up, no exposed patients showed symptoms. Maximum likelihood analysis of the nucleotide sequences obtained from the PCR products indicated that the rabies virus in the dog was closely related to isolates from neighbouring provinces of Guangdong as well as those from surrounding countries of China. Conclusions: Multidisciplinary One Health intervention is effective not only in the control of rabies but also in rapid emergency responses to attacks by rabid stray dogs. [ABSTRACT FROM AUTHOR]

Objectives: Patients with colorectal liver metastases (CRLM) who underwent hepatic resection after conversion therapy had a high recurrence rate of nearly 90%. Preoperative DEB-TACE has the potential to prevent postoperative recurrence which has not been elucidated. The objective of this study was to evaluate the safety and efficacy of preoperative DEB-TACE. Materials and methods: Patients with CRLM who underwent liver resection from June 1, 2016, to June 30, 2021, were collected and those who received conversional hepatectomy were included in this study. Patients with preoperative DEB-TACE were propensity-score matched in a 1:1 ratio to patients without preoperative DEB-TACE. Short-term outcomes and recurrence-free survival (RFS) were compared between the two groups. Results: After PSM, 44 patients were included in each group. The toxicities of DEB-TACE were mild and could be managed by conservative treatment. Overall response rate (ORR) of conversion therapy (75.0% vs. 81.2%, p = 0.437) and postoperative complication of hepatic resection (27.3% vs. 20.5%, p = 0.453) were similar between the two groups. The median RFS of the DEB-TACE group (10.7 months, 95%CI: 6.6–14.8 months) was significantly longer than that of the control group (8.1 months, 95%CI: 3.4–12.8 months) (HR: 0.60, 95%CI: 0.37–0.95, p = 0.027). Conclusions: In patients who became resectable after conversion therapy, preoperative DEB-TACE might be a safe option to achieve longer RFS. Sirtuin-3 (SIRT3), the main deacetylase in the mitochondria, maintains cellular energy metabolism and redox balance by deacetylating mitochondrial proteins in a NAD+-dependent manner. Growing evidence indicates that decreased Sirt3 expression is involved in various age-related maladies. However, the role of Sirt3 in ovarian and testicular senescence remains unclear. In this study, we observed that sirt3 expression showed age-dependent decreases in the ovary but not the testis. We generated Sirt3 null mice via CRISPR/Cas9-mediated genome editing. We observed that Sirt3 deletion accelerated ovarian aging, as shown by a decrease in offspring sizes, the follicle reserve and oocytes markers (Bmp15 and Gdf9) as well as increased expression of aging and inflammation-related genes (p16, p21 , Il-1α, and Il-1β). Sirt3 deficiency led to an accumulation of superoxide and disruption of spindle assembly accompanied by mitochondrial dysfunction (uneven mitochondria distribution, decreased mitochondrial potential as well as reduced mitochondrial DNA content) in aging oocytes. Meanwhile, in ovaries of Sirt3 null mice, the impaired mitochondrial functions were shown by decreases in mitochondrial respiratory complexes, along with lower levels of mitochondrial fusion (OPA1, MFN2) and fission (DRP1, FIS1) proteins. er levels of mitochondrial fusion (OPA1, MFN2) and fission (DRP1, FIS1) proteins. Interestingly, Sirt3 −/− male mice exhibited no changes on the testicular histology, serum testosterone levels, germ-cell proliferation, and differentiation of spermatogonia. Meiotic prophase I spermatocytes were also normal. Levels of superoxide, mitochondrial potential as well as expression of mitochondrially-encoded genes were unaltered in Sirt3 −/− testes. Collectively, the results indicated that SIRT3 plays a critical role in maintaining the ovarian follicle reserve and oocyte quality in aging mice, suggesting its important role in controlling ovarian senescence. To obtain the change tendency of output angular velocity and tangential contact force of a gear when the pinion under the step input during meshing of a new type of spiral bevel gear, which is a logarithmic spiral bevel gear, the tooth flank equation of logarithmic spiral bevel gear is deduced based on the formation mechanism of the tooth flank formation. A three-dimensional model of a pair of logarithmic spiral bevel gears whose number of teeth was 37:9, with modules being 4.5 mm, normal pressure angle being 20 degrees and spiral angle being 35 degrees were built and assembled. Based on Hertz elastic contact theory, the calculation formulas and parameters sets of contact force for conventional spiral bevel gear meshing simulation and logarithmic spiral bevel gear meshing simulation were done. Consider the dynamic simulation about meshing angular velocity and tangential contact force for conventional spiral bevel gear meshing and logarithmic spiral bevel gear meshing, respectively. Finally, by analyzing and comparing the simulation data, the results show that under the same input conditions, the fluctuation of the gear angular velocity and tangential contact force of logarithmic spiral bevel gear meshing are smaller than the conventional spiral bevel gear. Simple Summary: In this study, we first reported the infiltration and prognostic value of CXCR6+TAMs in all stages of colon cancer patients and assessed predictive ability as a biomarker for different duration adjuvant chemotherapy regimens in the primary cohort and validation cohort Patients with high CXCR6+ TAM infiltration tended to have worse overall survival. A 6-month chemotherapy regimen for high-risk stage II and stage III patients after curative operation benefited those with high CXCR6+ TAM density the most. Furthermore, we found there was a negative relationship between CXCR6+TAMs and activated CD8+ T cells. Cytokines from peripheral blood reflected the immunosuppressive state in patients with high accumulations of CXCR6+TAMs This indicates that CXCR6+TAMs may be involved in the formation of an immunosuppressive environment. We explored the infiltration and prognostic value of CXCR6+TAMs in all stages of colon cancer (CC) patients and assessed predictive ability as a biomarker for different ACT regimens among high-risk stage II and stage III patients in both primary and validation cohorts. Two independent cohorts of 360 and 126 consecutive colon cancer patients were enrolled from two medical centers of Zhongshan Hospital. Immunofluorescence and immunohistochemistry were performed to detect the density of CXCR6+TAMs and activated CD8+ T cells. The infiltration of CXCR6+TAMs was higher in tumor tissues and increased with advanced tumor stage. A high density of CXCR6+TAMs predicted worse overall survival (OS) in all CC patients (HR = 2.49, 95% CI = (1.68, 3.70), p < 0.001), and was an independent risk factor verified by Cox regression analysis (HR = 1.68, 95% CI = (1.09, 2.59), p = 0.019). For high-risk stage II and stage III patients with a high density of CXCR6+TAMs, better disease-free survival (DFS) (HR = 0.32, 95% CI = (0.11, 0.89), p = 0.003), and OS (HR = 0.28, 95% CI = (0.07, 1.11), p = 0.014) were observed in the 6-month treatment group. There was a negative relationship between the density of CXCR6+TAMs and CD8+ T cells (R = −0.51, p < 0.001) as well as activated CD8+ T cells (R = −0.54, p < 0.001). Higher levels of IL-6 and lower levels of IL-2R and TNF-α were expressed in high-CXCR6+ TAM-density patients, which indicates that CXCR6+TAMs contribute to an immunosuppressive microenvironment. CXCR6+TAMs predicted prognosis and response to different durations of ACT in CC patients. Simple Summary: This retrospective study evaluated the clinical implications of CUL9 expression on the prognosis and the predictive value for postoperative adjuvant chemotherapy in consecutive patients with colon cancer. Among all 1078 patients, high expression of CUL9 was an independent prognostic factor on disease-free survival and overall survival. Prognostic biomarkers are keys to the risk stratification of patients and the decision to recommend adjuvant chemotherapy. Among 564 patients with high-risk stage II and stage III disease who were recommended to receive the standard 6 months of chemotherapy, those with high CUL9 expression from the full dose group had better disease-free survival than those from the reduced dose group. A test for the interaction between CUL9 expression and the treatment reached significance and was not confounded by T stage, N stage and histopathological grade. This indicated that CUL9 expression may further filter those patients to identify truly high-risk cases that benefit from the full dose of the standard 6 months of chemotherapy. We evaluated the clinical implications of CUL9 expression on the prognosis and the predictive value for adjuvant chemotherapy in colon cancer. A total of 1078 consecutive patients treated with radical resection from 2008 to 2012 were included. Formalin-fixed, paraffin-embedded specimens were used as immunohistochemistry (IHC) for CUL9. For all patients, high expression of CUL9 was identified as an independent prognostic factor for overall survival (HR = 1.613, 95% CI 1.305–1.993, p < 0.001) and disease-free survival (HR = 1.570, 95% CI 1.159–2.128, p = 0.004). The prognostic value of high CUL9 expression was confirmed in an independent validation cohort from the GEO database. The efficacy of adjuvant chemotherapy was analyzed among patients with high-risk stage II and stage III disease. Those with high CUL9 expression from the full dose group had better disease-free survival (HR = 0.477, 95% CI 0.276–0.825, p = 0.006) than those from the reduced dose group. The interaction test between CUL9 expression and the treatment reached significance and was not confounded by T stage, N stage and histopathological grade. Background: N6-methyladenosine (m6A) modification plays crucial roles in cancers. However, its alteration in colorectal cancer (CRC) is still poorly described. The purpose of this study is to explore the change of m6A modification and the function of m6A binding protein YTHDC2 in CRC. Methods: The global level of m6A modification was detected by mass spectrometry and dot blotting assay. The expression of YTHDC2 was investigated using The Cancer Genome Atlas and using real-time polymerase chain reaction (RT-qPCR), western blotting, and immunohistochemistry based on CRC tissues. Kaplan–Meier analysis and Cox proportional hazards regression were performed to analyze the prognostic value of YTHDC2. RNA immunoprecipitation (RIP)-seq and m6A immunoprecipitation (MeRIP)-seq were used to explore the direct targets of YTHDC2. Gene oncology (GO) and Gene Set Enrichment Analysis (GSEA) were used to explore the pathways that could be influenced by YTHDC2. Results: No significant difference was observed in the global level of m6A modification on total RNA or mRNA between CRC and adjacent nontumor tissues. We further found a significant decreasing of YTHDC2 in CRC tissues. Kaplan–Meier analysis indicated that lower expression of YTHDC2 was related to the worse disease-free survival and overall survival. In addition, lower expression of YTHDC2 was an independent worse prognostic factor in univariate and multivariate Cox regression analysis. Using YTHDC2-RIP-seq and MeRIP-seq, we identified that YTHDC2 could participate in several important biological signal pathways. Conclusions: In summary, this study suggested that the global level of m6A did not change in CRC and identified that lower YTHDC2 as a prognostic marker for worse survival of CRC. Diazepam binding inhibitor (DBI)‐translocator protein (18kDa) (TSPO) signaling in the retina was reported to possess coordinated macroglia‐microglia interactions. We investigated DBI‐TSPO signaling and its correlation with vascular endothelial growth factor (VEGF), neurotrophic or inflammatory cytokines in neovascular retinopathy, and under hypoxic conditions. The vitreous expression of DBI, VEGF, nerve growth factor (NGF), and interleukin‐1beta (IL‐1β) were examined in proliferative diabetic retinopathy (PDR) patients with or without anti‐VEGF therapy and nondiabetic controls. Retinal DBI‐TSPO signaling and the effect of the anti‐VEGF agent were evaluated in a mouse model of oxygen‐induced retinopathy (OIR). Interactions between Müller cell‐derived VEGF and DBI, as well as cocultured microglial cells under hypoxic conditions, were studied, using Western blot, real‐time RT‐PCR, enzyme‐linked immunosorbent assay (ELISA), flow cytometry, and immunofluorescent labeling. Results showed that vitreous levels of DBI, VEGF, NGF, and IL‐1β were significantly higher in PDR patients compared with controls, which further changed after anti‐VEGF therapy. A statistical association was found between vitreous DBI and VEGF, NGF, IL‐1β, and age. The application of the anti‐VEGF agent in the OIR model induced retinal expression of DBI and NGF, and attenuated inflammation and microglial cell activation. Inhibition of Müller cell‐derived VEGF could increase its DBI expression under hypoxic conditions, while the DBI‐TSPO signaling pathway is essential for anti‐VEGF agents exerting anti‐inflammatory and neuroprotective effects, as well as limiting inflammatory magnitude, promoting its neurotrophin production and anti‐inflammatory (M2) polarization in microglial cells. This paper analyzes the evolution of the South Asian High (SAH) during and after the development of tropical cyclone Neoguri over the South China Sea (SCS) in mid-April 2008, the formation of tropical storm Nargis over the Bay of Bengal (BOB) in late April, and the Asian summer monsoon onset, as well as their interrelationships. Numerical sensitivity experiments are conducted to explore the underlying mechanism responsible for these seasonal transitions in 2008. It is demonstrated that strong latent heating related with tropical cyclone activities over the SCS can enhance the development of the SAH aloft and generate zonal asymmetric potential vorticity (PV) forcing, with positive vorticity advection to its east and negative advection to its west. Following the decay of the tropical cyclone, this asymmetric forcing leads to instability development of the SAH, presenting as a slowly westward-propagating Rossby wave accompanied by a westward shift of the high PV advection. A strong upper tropospheric divergence on the southwest of the SAH also shifts westward, while positive PV eddies are shed from the high PV advection and eventually arrives in the southern BOB. Such synoptic patterns provide favorable pumping conditions for local cyclonic vorticity to develop. The latent heating release from the cyclogenesis further intensifies the upper-layer divergence, and the lower and upper circulations become phase locked, leading to the explosive development of the tropical cyclone over the southern BOB. Purpose: To investigate the role of HER2 positivity in prognosis and unresponsiveness to anti-EGFR therapy for colorectal cancer. Methods: Patients who underwent primary CRC tumor resection were included. HER2 status of CRC was confirmed by immunohistochemistry and fluorescence in situ hybridization tests. Comparison of survival analysis between HER2 positivity and negativity was evaluated by a stratified log-rank test and summarized with the use of Kaplan–Meier and Cox proportional hazards methods. The treatment effects of cetuximab were further compared in full subgroup analyses. Result: 1240 patients were enrolled, including 763 with stage I-III CRC and 477 with stage IV CRC. 57 (4.6%) CRC patients presented HER2 positivity in the entire cohort. The survival analysis showed that patients with HER2 positivity had significantly worse disease-free survival and overall survival in stage III and IV CRC. The multivariable analysis also confirmed that HER2 positivity was a significantly independent risk factor in stage III and IV CRC. Univariate and multivariable survival analysis showed no prognostic significance of HER2 positivity in stage I–II CRC patients. Prespecified subgroup analysis showed no favorable trends in progression-free survival and overall survival for cetuximab in the patients with HER2 positivity, KRAS/NRAS/BRAF wild-type metastatic CRC (interaction P values = 0.005 and 0.014). Conclusion: For stage III and IV CRC patients, HER2 positivity was confirmed as an independent prognostic risk factor. Background: No.253 lymph node is the gateway to systemic metastasis for left-sided colorectal cancer. However, the value of D3 resection is still controversial. This study aimed to identify the incidence rate and prognostic value of 253LN metastasis in patients with left-sided colorectal cancer liver metastasis (CRLM) mainly through blood vessels and thus to provide theoretical basis for 253LN resection. Methods: From February 2012 to February 2019, a total of 281 patients who underwent curative resection for both primary and metastatic tumors were collected retrospectively. The clinicopathological and genetic characteristics were compared between 58 patients with positive 253LN and 223 patients with negative. Relapse-free survival (RFS) and overall survival (OS) were compared with Kaplan–Meier method. Cox regression analysis and a forest plot were conducted for RFS. Results: The incidence of 253LN metastasis in left-sided CRLM was 20.64% (58/281). Those with 253LN positive were T4 stage, N2 stage, and D1/D2 lymph nodes metastatic. About 10.3% (8/78) 253LN positive patients were D1/D2 negative. The 253LN metastasis was an independent risk factor for relapse after curative surgery, but not for OS. Patients with 253LN metastasis had worse RFS, especially in female, adenocarcinoma, poorly differentiated, pT3, preoperative serum CA199 < 37 U/mL, bilobar liver metastasis, without preoperative chemotherapy, KRAS, NRAS, or BRAF wild type. Conclusion: The incidence of 253LN metastasis in left-sided CRLM is 20.64%, and skip metastasis rate is 10.3%. The 253LN status is an independent prognostic risk factor for RFS but not for OS after curative surgery. Routine resection of 253LN should be applied in curative surgery of left-sided CRLM.
