The growing tendency toward non-compliance with international obligations to refugees requires UNHCR to make contingency plans for asylum denials. Making such plans calls for tools, such as game theory and confrontation analysis, that link non-compliance in refugee asylum with the configuration of interests for asylum and donor countries in a potential crisis. We illustrate such tools in a case study of Afghan refugees in Pakistan in 1997-2001, which generated asylum denials by Pakistan ranging from brief border closings to threats of mass refoulernent, linked to the withholding of assistance by the United States and other donors. Abstract: In collaboration with the Boeing Company, Wichita State University (WSU) Libraries and the Wichita State University Industrial Engineering Department employed Kaizen and Lean Management principles to successfully implement an accelerated improvement workshop (AIW) in the Libraries'' Technical Services Department. This case study explains the development of the Kaizen team, planning and training, Kaizen, and Lean Management theory and terminology, and the method used to reduce processing time. Materials processing time was reduced from 25 to 15 days. The team concluded that the AIW event had beneficial results and suggests AIW applications in other library units. [Copyright &y& Elsevier]

The cairn on Hardendale Nab was excavated in 1986 in advance of destruction by quarrying, and this allowed the reconstruction of a complex sequence of development. Construction of the cairn appears to have begun in the Early Bronze Age, although it might have been earlier. Phase 1 comprised a simple cist burial, and small earthen mound, with a number of secondary burials made in and around the original monument. Unusual rectangular features might have been used for pyres. Phase 2 saw the erection of a round enclosure around the mound, which was later (Phase 3) filled with loose limestone rubble. Again burials, both cremations and inhumations, were cut into the rubble, and bone scattered over and amidst it, with an apparent focus on the southern part of the structure. The entire complex was buried by a further layer of rubble in Phase 4, into which several further burials, this time inhumations, were cut. The excavation produced an unusually large amount of environmental evidence, much of it associated with the use of the cairn as a roost by owls in Phases 2 and 3. [ABSTRACT FROM AUTHOR]

Imaging with acoustic and optical televiewers results in continuous and oriented 360° views of the borehole wall from which the character, relation, and orientation of lithologic and structural planar features can be defined for studies of fractured-rock aquifers. Fractures are more clearly defined under a wider range of conditions on acoustic images than on optical images including dark-colored rocks, cloudy borehole water, and coated borehole walls. However, optical images allow for the direct viewing of the character of and relation between lithology, fractures, foliation, and bedding. The most powerful approach is the combined application of acoustic and optical imaging with integrated interpretation. Imaging of the borehole wall provides information useful for the collection and interpretation of flowmeter and other geophysical logs, core samples, and hydraulic and water-quality data from packer testing and monitoring. Evidence from barley [Hordeum distichum (L.) Lam. cv. Maris Mink], and from many other species, suggests that respiration is controlled by either supply of carbohydrate or demand for ATP. The relationship between root respiration rate (measured as O2 consumption or CO2 production) and ethanol-soluble carbohydrate content altered with time following selective pruning, and the change could not be accounted for by buffering of the cytoplasmic carbohydrate concentration by sugars in the vacuole. Exogenous sucrose supplied to the roots prevented any decline of the respiration rate in shoot-pruned plants, and if supplied for 24 h stimulated the respiration rate after any treatment. Root extension responded to sucrose in a similar manner. We suggest that respiration is under fine control by adenylates, but the capacity of the respiratory system is fixed by the supply of sucrose, possibly via coarse control of the respiratory machinery, or of the processes requiring metabolic energy. A state‐space model (SSM) of infiltration estimates daily groundwater recharge using time‐series of groundwater‐level altitude and meteorological inputs (liquid precipitation, snowmelt, and evapotranspiration). The model includes diffuse and preferential flow through the unsaturated zone, where preferential flow is a function of liquid precipitation and snowmelt rates and a threshold rate, above which there is direct recharge to the water table. Model parameters are estimated over seasonal periods and the SSM is coupled with the Kalman Filter (KF) to assimilate recent observations (hydraulic head) and meteorological inputs into recharge estimates. The approach can take advantage of real‐time hydrologic and meteorological data to deliver real‐time recharge estimates. The model is demonstrated on daily observations from two bedrock wells in carbonate aquifers of northwestern New York (USA) between 2013 and 2018. Meteorological inputs for liquid precipitation and snowmelt are compiled from SNODAS (2021). Results for recharge during winter and spring seasons show preferential flow events to the water table from liquid precipitation, snowmelt, or a combination of the two. Recharge estimates summed annually are consistent with previous estimates of recharge reported from groundwater flow and surface‐process models. Results from the SSM and KF point to errors in meteorological inputs, such as the snowmelt rate, that are not compatible with hydraulic head observations. Whereas liquid and solid precipitation are measured at discrete stations and extrapolated to 1‐km2 grid cells, snowmelt is a meteorological modeled outcome that may not represent conditions in the vicinity of monitoring well locations. Abstract: In situ chemical oxidation (ISCO) treatment with sodium permanganate, an electrically conductive oxidant, provides a strong electrical signal for tracking of injectate transport using time series geophysical surveys including direct current (DC) resistivity and electromagnetic (EM) methods. Effective remediation is dependent upon placing the oxidant in close contact with the contaminated aquifer. Therefore, monitoring tools that provide enhanced tracking capability of the injectate offer considerable benefit to guide subsequent ISCO injections. Time-series geophysical surveys were performed at a superfund site in New Hampshire, USA over a one-year period to identify temporal changes in the bulk electrical conductivity of a tetrachloroethylene (PCE; also called tetrachloroethene) contaminated, glacially deposited aquifer due to the injection of sodium permanganate. The ISCO treatment involved a series of pulse injections of sodium permanganate from multiple injection wells within a contained area of the aquifer. After the initial injection, the permanganate was allowed to disperse under ambient groundwater velocities. Time series geophysical surveys identified the downward sinking and pooling of the sodium permanganate atop of the underlying till or bedrock surface caused by density-driven flow, and the limited horizontal spread of the sodium permanganate in the shallow parts of the aquifer during this injection period. When coupled with conventional monitoring, the surveys allowed for an assessment of ISCO treatment effectiveness in targeting the PCE plume and helped target areas for subsequent treatment. Abstract: Field characterization of a trichloroethene (TCE) source area in fractured mudstones produced a detailed understanding of the geology, contaminant distribution in fractures and the rock matrix, and hydraulic and transport properties. Groundwater flow and chemical transport modeling that synthesized the field characterization information proved critical for designing bioremediation of the source area. The planned bioremediation involved injecting emulsified vegetable oil and bacteria to enhance the naturally occurring biodegradation of TCE. The flow and transport modeling showed that injection will spread amendments widely over a zone of lower‐permeability fractures, with long residence times expected because of small velocities after injection and sorption of emulsified vegetable oil onto solids. Amendments transported out of this zone will be diluted by groundwater flux from other areas, limiting bioremediation effectiveness downgradient. At nearby pumping wells, further dilution is expected to make bioremediation effects undetectable in the pumped water. The results emphasize that in fracture‐dominated flow regimes, the extent of injected amendments cannot be conceptualized using simple homogeneous models of groundwater flow commonly adopted to design injections in unconsolidated porous media (e.g., radial diverging or dipole flow regimes). Instead, it is important to synthesize site characterization information using a groundwater flow model that includes discrete features representing high‐ and low‐permeability fractures. This type of model accounts for the highly heterogeneous hydraulic conductivity and groundwater fluxes in fractured‐rock aquifers, and facilitates designing injection strategies that target specific volumes of the aquifer and maximize the distribution of amendments over these volumes. Konjac glucomannan is a hydrocolloid that has been used in food applications. The European ban on the use of Konjac glucomannan means that the detection and analysis has potential applications in the food industry, particularly detection of food adulteration. The aim of this work was to develop an assay capable of detecting Konjac glucomannan as an isolated sample and within food matrices. An indirect competitive ELISA was developed utilising a polyclonal antibody raised against Konjac glucomannan. The ELISA was found to be specific for Konjac glucomannan and sensitive, with a detection limit of 0.1 ng mL−1. Increasing salt concentration and freeze/thaw cycles did not affect the performance of the assay. The ELISA was able to detect Konjac glucomannan in admixtures with other gums and also in confectionery that had been spiked with Konjac glucomannan. The ELISA has potential as a kit for the differentiation of Konjac glucomannan from other hydrocolloids and detection in food. [ABSTRACT FROM AUTHOR]

Working relationships between vendors and academic libraries have extended from a traditional one of customer and bookseller to the testing of vendor-developed software. This extension has occurred since 1980, the last quarter century. What has changed since 2000 is the addition of tests of vendor/client interface informed by enterprise software that completely alters methodologies for accommodating library requirements for book supply, MARC record supply, and electronic invoices and purchase orders. This paper will examine the technical, administrative, and legal considerations now relevant to beta testing of vendor software by library staff and offer charts and checklists that may make these projects more easily comprehended. An increasing number of cell types, including peripheral blood mononuclear cells (PBMCs), have been demonstrated to release heat shock proteins (Hsps). In this paper we investigate further the hypothesis that Hsps are danger signals. PBMCs and Jurkat cells released Hsp70 (0·22 and 0·7 ng/106 cells, respectively) into medium over 24 h at 37°C. Release of Hsp70 was stimulated 10-fold by GroEL ( P < 0·001) and more than threefold by lipopolysaccharide (LPS) ( P < 0·001). Although Hsp60 could be detected in the medium of cells cultured at 37°C for 24 h, the low rates of release were due probably to cell damage. Significant release of Hsp60 was observed when Jurkat cells were exposed to GroEL (2·88 ng/106 cells) or LPS (1·40 ng/106 cells). The data are consistent with the hypothesis that Hsp70 and Hsp60 are part of a danger signalling cascade in response to bacterial infection. [ABSTRACT FROM AUTHOR]

A number of enzyme-linked immunosorbent assays (ELISAs) have been developed for the detection of milk adulteration in dairy products. Target antigens have been caseins, lactoglobulins, immunoglobulins and other whey proteins. Polyclonal and monoclonal antibodies have been used in a variety of formats including direct, indirect, competitive and sandwich ELISAs. ELISAs have been successfully applied to the detection of cows' milk adulteration of sheep, goat and buffalo milk. Goat milk adulteration of sheep milk has also been detected. A number of ELISAs have also been applied to cheese. It is recommended that ELISA should be used in combination with PCR to ensure compliance with current legislation. [ABSTRACT FROM AUTHOR]

This paper describes the development and use of a competitive enzyme-linked immunosorbent assay (ELISA) to detect prawns which have been irradiated. The ELISA utilizes a monoclonal antibody against a modified DNA base, dihydrothymidine. A comparison of extraction procedures demonstrated that DNA purification was not required and that crude prawn homogenate could be used in the ELISA. The ELISA was applied successfully to two prawn species, North Atlantic prawn (Pandalus borealis) and Tiger prawn (Penaeus monodon). The ELISA has a working range of 0.5–2 kGy with CVs typically below 10%. Storage of irradiated prawns for up to 12 months at -20 °C had no effect on ELISA performance. Heat shock proteins (Hsps) are known to be induced in response to short-term stress. The present study aimed to evaluate the potential of Hsp70 as a biomarker of stress produced by increased temperature, osmotic pressure, and exposure to cadmium and sodium chloride in marine macroalgae and fresh water plant species. An indirect competitive enzyme-linked immunosorbent assay (IC-ELISA) was developed with a working range of 0.025-10 μg ml -1 using a monoclonal antibody raised against purified Hsp70 of Phaseolus aureus (mung bean). Fucus serratus (toothed wrack), Chondrus crispus (Stackhouse or Carrageen moss), Ulva lactuca (sea lettuce) and Lemna minor (common duckweed) sample extracts were stressed for up to 24 h and then tested in the IC-ELISA. The presence of Hsp70 and cross-reactivity of the monoclonal antibody was confirmed by Western blot. The heat shock response was confirmed in each species using a 2-h 42°C treatment. Following heat shock, Hsp70 concentrations increased to a peak at 2 h ( F. serratus ) or 4 h ( L. minor ), after which concentrations decreased. Osmotic and cadmium stresses also resulted in elevated Hsp70 concentrations in samples of F. serratus and L. minor when compared with unstressed controls. In both, osmotic and metal stress, the production of Hsp70 increased to a maximum and subsequently decreased as the stressor levels increased. Results suggest that Hsp70 IC-ELISA could potentially be applied to the detection of stress in these aquatic species, although it would probably be most effective when used in conjunction with other measurements to provide a stressor-specific biomarker profile or fingerprint. Isoforms of the vitamin B12 carrier protein transcobalamin (TC) might influence its cellular availability and contribute to the association between disrupted single-carbon metabolism and Alzheimer's disease (AD). We therefore investigated the relationships between the TC 776C>G (Pro259Arg) genetic polymorphism, total serum cobalamin and holo-TC levels, and disease onset in 70 patients with clinically diagnosed AD and 74 healthy elderly controls. TC 776C>G polymorphism was also determined for 94 histopathologically confirmed AD patients and 107 controls. Serum holo-TC levels were significantly higher in TC 776C homozygotes (p = 0.04). Kaplan-Meier survival functions differed between homozygous genotypes (Cox's F-Test F(42, 46) = 2.1; p = 0.008) and between 776C homozygotes and heterozygotes (Cox's F test F(46, 108) = 1.7; p = 0.02). Proportionately fewer TC 776C homozygotes appear to develop AD at any given age, but this will require confirmation in a longitudinal study. [ABSTRACT FROM AUTHOR]

Background: Patients with implantable cardioverter–defibrillators often receive adjunctive antiarrhythmic therapy to prevent frequent shocks. We tested the efficacy and safety of sotalol, a beta-blocker with class III antiarrhythmic effects, for this purpose. Methods: In a multicenter trial, patients were stratified according to left ventricular ejection fraction (≤0.30 or >0.30), randomly assigned to double-blind treatment with 160 to 320 mg of sotalol per day (151 patients) or matching placebo (151 patients), and followed for 12 months. Kaplan–Meier analyses of the time to an event were performed. Three end points were used: the delivery of a first shock for any reason or death from any cause, the first appropriate shock for a ventricular arrhythmia or death from any cause, and the first inappropriate shock for a supraventricular arrhythmia or death from any cause. Results: Compliance with double-blind treatment was similar in the two groups. There were seven deaths in the placebo group and four in the sotalol group. As compared with placebo, treatment with sotalol was associated with a lower risk of death from any cause or the delivery of a first shock for any reason (reduction in risk, 48 percent; P<0.001 by the log-rank test), death from any cause or the delivery of a first appropriate shock (reduction in risk, 44 percent; P=0.007), or death from any cause or the delivery of a first inappropriate shock (reduction in risk, 64 percent; P=0.004). Sotalol also reduced the mean (±SD) frequency of shocks due to any cause (1.43± 3.53 shocks per year, as compared with 3.89±10.65 in the placebo group; P=0.008). In the sotalol group, the reduction in the risk of death from any cause or the delivery of a first shock for any reason did not differ significantly between patients with ejection fractions of more than 0.30 and those with ejection fractions of 0.30 or less. Conclusions: Oral sotalol was safe and efficacious in reducing the risk of death or the delivery of a first defibrillator shock whether or not ventricular function was depressed. The article presents a kinetic study on the reaction between superoxide and cobalamins II (Cbl (II)) and on the ability of cyanocobalamin, the vitamin B12 that forms in vitamin pills, in preventing intracellular superoxide levels in a cell. It notes that all of the kinetic experiments were made in the presence of catalase. It cites that the study has demonstrated that the intracellular Cbl form cob(II)alamin reacts with superoxide at rates nearing those of superoxide dismutase (SOD).
