Agronomy biofortification is an important crop management strategy to enhance concentrations of micronutrients in edible portions. The availability of micronutrients for plant uptake is reduced by the high salinity, pH, and lime of soils. Hence, this study aimed to determine the most appropriate time and the best ratio of zinc (Zn) and iron (Fe) consumption during the wheat growth stages in a saline and calcareous soil. The experiment consisted of two factors with four levels; 0, 30, 60, and 120 kg Zn ha−1 as Zn0, Zn1, Zn2, and Zn3 respectively, and 0, 2.5, 5, and 7.5 kg Fe ha−1 as Fe0, Fe1, Fe2, and Fe3 respectively, in a randomized complete block design with four replications. The results showed that the interaction effects of Fe and Zn fertilizers application were significant on the wheat yield indices and concentration of Fe and Zn in wheat shoots and grain. The simultaneous application of 120 kg Zn ha−1 and 5 kg Fe ha−1 caused significant improvement (p < 0.05) in thousand kernel weight (9.1%) and subsequently grain yield (23.4%) compared to the control treatment. The Zn concentration in the wheat shoot during tillering (37.0–58.2 mg Zn kg−1) was higher than the other two growth stages (elongation (29.2–40.8 mg Zn kg−1) and booting (20.0–31.0 mg Zn kg−1)). While the highest Fe concentration in wheat shoots was observed at the stem elongation stage (98.5–268.8 mg Fe kg−1), then the booting stage (91.0–150.8 mg Fe kg−1) and finally the least absorption was at the tillering (80.0–120.0 mg Fe kg−1). Because of the high calcium carbonate, salinity and Zn- deficient soil of the experimental site, the Zn concentration in wheat grain was obtained in a range of 3 to 19 mg kg−1. The results clarified that the fertilizer application of 30 kg Zn ha−1 and 2.5 kg Fe ha−1 can be proportionally and simultaneously increased the concentration of Fe and Zn in wheat grain by 100%. Therefore, the soil application of 30 kg Zn and 2.5 kg Fe per hectare proposed for improvement in quantitative (619 kg ha−1 increase in wheat grain yield) and qualities (100% increase in grain Zn and Fe concentration and 8% increase in thousand kernel weight) indices of wheat yield in calcareous and saline soils. Zinc (Zn) deficiency and salt stress are well-known soil problems and often happen parallelly in cultivated soils. In this study, Zn-amino acid complexes (Zn-AAc) were used as a source of Zn to determine their effects on salt-induced damage in wheat plants. The bread wheat (Triticum aestivum L. cvs. Kavir) was supplied with Zn-glycine (Zn-Gly), Zn-alanine (Zn-Ala), and ZnSO4 as Zn sources at three salinity levels (EC 2, 4 and 6 dS m-). Salinity caused a significant decrease in shoot dry matter and grain yield of wheat, but this negative effect was significantly improved by the application of Zn-AAc. Salt stress decreased shoot and grain Zn concentration, but this reduction was lower in plants supplied by Zn-AAc. Calcium (Ca) and potassium (K) concentrations were increased in a shoot by salinity stress while decreased in grain. Sodium (Na) concentration decreased in shoot and grain by using Zn-AAc. At all of the salinity levels, wheat supplied with Zn-AAc had lower lipid peroxidation compared to those grown under the ZnSO4 source. Application of Zn-AAc increased the activities of catalase (CAT) and superoxide dismutase (SOD) in the roots of wheat plants in saline conditions. Selenium (Se) is a micronutrient that has multiple biochemical effects ranging from a nutritional deficiency at low levels to toxicity at high levels. Its quantification in environmental systems is vital to protect the health of people and animals. This study developed and validated methods to determine the total selenium (Se) concentration in soil, maize plants, and maize flour. The performance of the methods was ascertained using matrix certified reference materials (CRMs), namely NMIJ CRM 7303-a (trace elements in lake sediments), NIST SRM 1646-a (estuarine sediments), NIST SRM 1570-a (spinach leaves), NIST SRM 1571 (orchard leaves), NCS ZC 73010 (maize flour) and NIST RM 8437 (hard red spring wheat flour). All methods fulfilled the performance requirements of the AOAC International. An acceptable accuracy was achieved with percentage recoveries between 57 and 102% and E n ‐ scores within ±1 limits. Repeatability ( RSD r , %) and intermediate precision (RSD R , %) ranged from 4 to 12%, and 9 to 15%, respectively. The limit of detection (LOD) and limit of quantification (LOQ) for soil were 0.024 ng. g−1 and 0.082 ng. g−1, respectively. The LODs and LOQs for maize plants and flour were 0.0036 and 0.0179 ng. g−1, respectively. The performance characteristics of the methods were comparable with previously reported methods, confirming reliability. The measurement uncertainty was evaluated following the guide to the expression of uncertainty in measurement (GUM). Background: Septoria tritici blotch (STB) disease causes yield losses of up to 50 per cent in susceptible wheat cultivars and can reduce wheat production. In this study, genomic architecture for adult-plant STB resistance in a Septoria Association Mapping Panel (SAMP) having 181 accessions and genomic regions governing STB resistance in a South Asian wheat panel were looked for. Results: Field experiments during the period from 2019 to 2021 revealed those certain accessions, namely BGD52 (CHIR7/ANB//CHIR1), BGD54 (CHIR7/ANB//CHIR1), IND92 (WH 1218), IND8 (DBW 168), and IND75 (PBW 800), exhibited a high level of resistance. Genetic analysis revealed the presence of 21 stable quantitative trait nucleotides (QTNs) associated with resistance to STB (Septoria tritici blotch) on all wheat chromosomes, except for 2D, 3A, 3D, 4A, 4D, 5D, 6B, 6D, and 7A. These QTNs were predominantly located in chromosome regions previously identified as associated with STB resistance. Three Quantitative Trait Loci (QTNs) were found to have significant phenotypic effects in field evaluations. These QTNs are Q.STB.5A.1, Q.STB.5B.1, and Q.STB.5B.3. Furthermore, it is possible that the QTNs located on chromosomes 1A (Q.STB.1A.1), 2A (Q.STB_DH.2A.1, Q.STB.2A.3), 2B (Q.STB.2B.4), 5A (Q.STB.5A.1, Q.STB.5A.2), and 7B (Q.STB.7B.2) could potentially be new genetic regions associated with resistance. Conclusion: Our findings demonstrate the importance of Asian bread wheat as a source of STB resistance alleles and novel stable QTNs for wheat breeding programs aiming to develop long-lasting and wide-ranging resistance to Zymoseptoria tritici in wheat cultivars. Background: Heat stress is one of the abiotic stresses that make wheat crops vulnerable, which significantly impacts crop production around the world. An increase in temperature during the reproductive phase (anthesis) beyond the optimal range of 15–20 °C leads to decreased crop production, poor quality of the grain, and altered physiological and biochemical processes. To study the association between high temperature and physio-biochemical traits under normal and late sown, a set of fifteen genotypes was utilized. Results: Relative water content under high temperatures had an overall decrease of 8.7%. However, grain protein and malondialdehyde content were higher in the stressed conditions than in the control, with increases of 20.2% and 38.9%, respectively. Marker Xgwm67, located on chromosome 5B, was found to be significantly associated with malondialdehyde content (R2 = 21%) and Xgwm570, located on 6A, was closely linked to relative water content as well as grain protein content (R2 = 16%) revealed by regression analysis. The correlation matrix displays a positive association between the control and stressed condition by R2 = 0.92, 0.82, and 0.53 in malondialdehyde, relative water content, and grain protein, respectively. However, there was a negative correlation between water content–malondialdehyde and malondialdehyde–grain protein, though there was only a 4% correlation between grain protein content (control) and relative water content (stressed). Based on the tolerance matrix, WH730 and RAJ4079 were heat tolerant, and DBW173 and HD3086 were sensitive. Conclusions: These findings indicate that to identify tolerant genotypes, physiological and biochemical traits can be utilized as an alternate criterion, and these closely associated markers can be applied for improved late-planted wheat production through MAS. The breeding scheme and genome editing by recognizing novel genes through physio-biochemical parameters, marker-assisted selection, and prospective screening of tolerant genotypes are proclaimed by the study. Wheat is an important staple crop not only in Pakistan but all over the globe. Although the area dedicated to wheat cultivation expands annually, the quantity of wheat harvested is declining due to various biotic and abiotic factors. Global wheat production and output have suffered as a result of the drought, which is largely driven by a lack of water and environmental factors. Organic fertilizers have been shown to reduce the severity of drought. The current research was conducted in semi-arid climates to mitigate the negative effects of drought on wheat during its critical tillering (DTS), flowering (DFS), and grain filling (DGFS) stages through the application of three different abscisic acid treatments: ABA0 (0 mgL−1) control, ABA1 (100 mgL−1) and ABA2 (200 mgL−1). Wheat growth and yield characteristics were severely harmed by drought stress across all critical development stages, with the DGFS stage being particularly vulnerable and leading to a considerable loss in yield. Plant height was increased by 24.25%, the number of fertile tillers by 25.66%, spike length by 17.24%, the number of spikelets per spike by 16.68%, grain count per spike by 11.98%, thousand-grain weight by 14.34%, grain yield by 26.93% and biological yield by 14.55% when abscisic acid (ABA) was applied instead of the control treatment. Moreover, ABA2 increased the more physiological indices (water use efficiency (36.12%), stomatal conductance (44.23%), chlorophyll a (24.5%), chlorophyll b (29.8%), transpiration rate (23.03%), photosynthetic rate (24.84%), electrolyte leakage (− 38.76%) hydrogen peroxide (− 18.09%) superoxide dismutase (15.3%), catalase (20.8%), peroxidase (− 18.09%), and malondialdehyde (− 13.7%)) of drought-stressed wheat as compared to other treatments. In the case of N, P, and K contents in grain were maximally improved with the application of ABA2. Through the use of principal component analysis, we were able to correlate our results across scales and provide an explanation for the observed effects of ABA on wheat growth and production under arid conditions. Background: Beneficial effects from practising a Paleolithic diet as compared to a diabetes diet on weight, waist circumference, satiety, leptin, HbA1c and glucose control in randomised controlled trial participants with type 2 diabetes could be due to lower leptin resistance. Support for this hypothesis comes from an in vitro experiment that showed that digested wheat gluten, which is excluded from a Paleolithic diet, inhibits leptin from binding to its receptor, thus indicating a possible dietary cause of leptin resistance. However, the clinical relevance of the latter finding is unclear since removal of enzyme activity from the gluten digest by heat treatment also abolished leptin binding inhibition. Assessment of leptin binding inhibition in vivo is possible by comparison of total leptin levels with those of 'biologically active' leptin bound to its receptor (bioLep). Objectives: To assess the effects of a Paleolithic diet compared to a diabetes diet on leptin binding inhibition and to replicate our in vitro study. Methods: BioLep and total leptin levels were measured in secondary analysis of fasting plasma samples from our open label random order three plus three-month long cross-over trial performed in 2005–2007, that compared a Paleolithic diet with a diabetes diet in participants with type 2 diabetes without insulin treatment (per protocol). BioLep was also measured in vitro for known recombinant leptin concentrations incubated with a series of concentrations of 10 kDa spin-filtered digested wheat gluten, with or without prior heat treatment, at 100ºC for 30 min and centrifugation. Results: There was no difference between diets when comparing differences between bioLep and total leptin levels and their ratio in the 13 participants, three women and 10 men, aged 52–74 years with a mean BMI of 30 kg/m2 and a mean diabetes duration of eight years. We found no carry-over or period effect for bioLep and total leptin. In vitro, wheat gluten digest inhibited leptin binding in a dose-dependent manner but not after heat treatment. Conclusions: We found no leptin binding inhibition after the Paleolithic or diabetes diet, possibly due to its abolishment from cooking-related heat treatment of wheat gluten. Introduction: The association between food protein-induced enterocolitis syndrome (FPIES) and wheat ingestion in children with celiac disease is unknown at this time. Methods: We present seven cases of children with celiac disease who presented with symptoms of wheat-triggered acute FPIES (a-FPIES). An oral food challenge (OFC) with wheat allergen followed by 4 h of observation was performed. Activation of innate system cells was measured at baseline (T0), during symptoms (Ts), and 4 h after symptom onset (Ts + 4). A panel of human inflammatory cytokines was also performed. Results: All patients reacted to the first allergen dose. Three patients experienced a decrease of 30 mm Hg in systolic blood pressure and tachycardia and required hemodynamic resuscitation. Neutrophilia and a decrease in eosinophil count were evident at 4 h after symptom onset. At 4 h after symptom onset, cytokines (IL-6 and IL-8, and to a lesser degree, IL-10) were elevated. Conclusion: In a small sample of celiac patients with wheat exposure in an OFC, symptoms and acute immunological changes in serum inflammatory cytokine profile were consistent with a-FPIES. Fusarium graminearum causes Fusarium head blight (FHB) disease in wheat worldwide. Although F. graminearum is reported to secrete several effectors, their role in virulence and pathogenicity is unknown. The study aimed at identifying candidate genes with a role in pathogenicity and virulence using two different host systems, Arabidopsis thaliana and wheat, challenged with F. graminearum TN01. Detached leaf assay and histological studies revealed the virulent nature of TN01. A genome-wide in silico search revealed several candidate genes, of which 23 genes were selected based on reproducibility. Gene expression studies by reverse transcriptase–polymerase chain reaction (RT-PCR) in leaf tissues of Arabidopsis and the two wheat genotypes, the susceptible (Sonalika) and the resistant (Nobeoka Bozu/Nobeoka), compared with mock-treated controls in a time-course study using fungal- and plant-specific genes as internal controls revealed that these genes were differentially regulated. Further, expression of these candidates in F. graminearum–inoculated Sonalika and Nobeoka spikes compared with mock-treated controls revealed their role in pathogenicity and virulence. Gene ontology studies revealed that some of these secretory proteins possessed a role in apoptosis and ceratoplatanin and KP4 killer toxin syntheses. A three-dimensional protein configuration was performed by homology modeling using trRosetta. Further, real-time quantitative PCR (RT-qPCR) studies in F. graminearum–inoculated Arabidopsis and wheat at early time points of inoculation revealed an increased expression of the majority of these genes in Sonalika, suggesting their possible role in pathogenicity, whereas low mRNA abundance was observed for 11 of these genes in the resistant genotype, Nobeoka, compared with Sonalika, indicating their role in virulence of F. graminearum. Synthetic genes for the two subunits of phenylalanyl-tRNA synthetase (PheRS) from wheat were expressed in Escherichia coli. When each gene was induced individually, the α subunit with a cleavable 6 × His tag at the amino terminus was largely soluble, while the β subunit was almost completely insoluble. When the two subunits were co-expressed, a soluble fraction containing the two subunits were obtained. This was purified by a standard method in which the tag was cleaved off with a specific protease after affinity purification. As the sample contained slightly more PheRSα than PheRSβ, we further resolved the sample by gel filtration to obtain the fraction that showed the size of the conventional α2β2 tetrameric complex and contains an almost equal amount of the two subunits. The final yield was 0.6 mg per 1 liter of the culture medium, and the specific activity was 28 nmol min−1 mg−1, which was higher than that of a fraction purified from wheat germ. This recombinant PheRS was used, along with purified samples of the elongation factors and the ribosomes from wheat germ, for a poly(U)-dependent poly(Phe) synthesis reaction. Summary: Drought, especially terminal drought, severely limits wheat growth and yield. Understanding the complex mechanisms behind the drought response in wheat is essential for developing drought‐resistant varieties. This study aimed to dissect the genetic architecture and high‐yielding wheat ideotypes under terminal drought.An automated high‐throughput phenotyping platform was used to examine 28 392 image‐based digital traits (i‐traits) under different drought conditions during the flowering stage of a natural wheat population. Of the i‐traits examined, 17 073 were identified as drought‐related. A genome‐wide association study (GWAS) identified 5320 drought‐related significant single‐nucleotide polymorphisms (SNPs) and 27 SNP clusters.A notable hotspot region controlling wheat drought tolerance was discovered, in which TaPP2C6 was shown to be an important negative regulator of the drought response. The tapp2c6 knockout lines exhibited enhanced drought resistance without a yield penalty. A haplotype analysis revealed a favored allele of TaPP2C6 that was significantly correlated with drought resistance, affirming its potential value in wheat breeding programs.We developed an advanced prediction model for wheat yield and drought resistance using 24 i‐traits analyzed by machine learning. Summary: Fusarium diseases pose a severe global threat to major cereal crops, particularly wheat. Existing biocontrol strains against Fusarium diseases are believed to primarily rely on antagonistic mechanisms, but not widely used under field conditions.Here, we report an endophytic fungus, Purpureocillium lilacinum YZ1, that shows promise in combating wheat Fusarium diseases. Under glasshouse conditions, YZ1 inoculation increased the survival rate of Fusarium graminearum (Fg)‐infected wheat seedlings from 0% to > 60% at the seedling stage, and reduced spikelet infections by 70.8% during anthesis. In field trials, the application of YZ1 resulted in an impressive 89.0% reduction in Fg‐susceptible spikelets.While a slight antagonistic effect of YZ1 against Fg was observed on plates, the induction of wheat systemic resistance by YZ1, which is distantly effective, non‐specific, and long‐lasting, appeared to be a key contributor to YZ1's biocontrol capabilities. Utilizing three imaging methods, we confirmed YZ1 as a potent endophyte capable of rapid colonization of wheat roots, and systematically spreading to the stem and leaves. Integrating dual RNA‐Seq, photosynthesis measurements and cell wall visualization supported the link between YZ1's growth‐promoting abilities and the activation of wheat systemic resistance.In conclusion, endophytes such as YZ1, which exhibits non‐antagonistic mechanisms, hold significant potential for industrial‐scale biocontrol applications. Soil contamination with heavy metals (HMs) has become a severe environmental problem for crop production. The efficacy of bacillus strain (N18) and Fe-modified biochar (FeB2%) on plant growth, lead (Pb) bioaccumulation and oxidative stress in wheat plants was studied under Pb contaminated soil. Lead stress caused significant reduction in plant growth, chlorophyll contents, antioxidant enzymes activity and Pb tolerance index. The Bacillus strain inoculated seeds stimulated the plant growth, chlorophyll content, antioxidative enzymatic activity with reduced oxidative stress and Pb bioavailability in soil, Pb biological accumulation, translocation and uptake factor in plants. The FeB2% treatment had shown prominently positive results relative to N18 inoculated treatment. Whereas, combined application of N18 and FeB2% depicted maximum significant improvement in root length (37, 36 and 30%), shoot length (45 %, 46 % and 47 %), total chlorophyll (43 %, 51 % and 48 %), carotenoid contents (36 %, 49 % and 48 %) and antioxidative enzymes activities such as; superoxide dismutase (19 %, 35 % and 40 %), peroxidase (29 %, 18 % and 20 %), catalase (19 %, 35 % and 40 %) and ascorbate peroxidase (23 %, 23 % and 13 %) in Pb-0, 400 and 600 mg kg-1 soil, respectively. Reduction in malondialdehyde content (MDA) (52 %, 61 % and 56 %), Pb biological accumulation (48 %, 41 % and 37 %), soil-root translocation factor (11 %, 15 % and 9 %), root-shoot translocation factor (42 %, 36 % and 35 %) and uptake factor (19 %, 22 % and 17 %) in wheat plants observed with combined N18+ FeB2% application as compared to control treatments in Pb-0, 400 and 600 mg kg-1 soil, respectively. The study suggested that combined application of N18+ FeB2% treatment prominently increased plant health with reduction in Pb bio-availability compared to sole application of N18 strain and FeB2% treatment under (Pb) contaminated soils. Drought threatens plant growth and related ecosystem services. The emergence of plant drought stress under edaphic drought is well studied, whilst the importance of atmospheric drought only recently gained momentum. Yet, little is known about the interaction and relative contribution of edaphic and atmospheric drought on the emergence of plant drought stress. We conducted a gradient experiment, fully crossing gravimetric water content (GWC: maximum water holding capacity—permanent wilting point) and vapour pressure deficit (VPD: 1−2.25 kPa) using five wheat varieties from three species (Triticum monococcum, T. durum & T. aestivum). We quantified the occurrence of plant drought stress on molecular (abscisic acid), cellular (stomatal conductance), organ (leaf water potential) and stand level (evapotranspiration). Plant drought stress increased with decreasing GWC across all organizational levels. This effect was magnified nonlinearly by VPD after passing a critical threshold of soil water availability. At around 20%GWC (soil matric potential 0.012 MPa), plants lost their ability to regulate leaf water potential via stomata regulation, followed by the emergence of hydraulic dysfunction. The emergence of plant drought stress is characterized by changing relative contributions of soil versus atmosphere and their non‐linear interaction. This highly non‐linear response is likely to abruptly alter plant‐related ecosystem services in a drying world. The response of mesophyll conductance (gm) to CO2 plays a key role in photosynthesis and ecosystem carbon cycles under climate change. Despite numerous studies, there is still debate about how gm responds to short‐term CO2 variations. Here we used multiple methods and looked at the relationship between stomatal conductance to CO2 (gsc) and gm to address this aspect. We measured chlorophyll fluorescence parameters and online carbon isotope discrimination (Δ) at different CO2 mole fractions in sunflower (Helianthus annuus L.), cowpea (Vigna unguiculata L.), and wheat (Triticum aestivum L.) leaves. The variable J and Δ based methods showed that gm decreased with an increase in CO2 mole fraction, and so did stomatal conductance. There were linear relationships between gm and gsc across CO2 mole fractions. gm obtained from A‐Ci curve fitting method was higher than that from the variable J method and was not representative of gm under the growth CO2 concentration. gm could be estimated by empirical models analogous to the Ball‐Berry model and the USO model for stomatal conductance. Our results suggest that gm and gsc respond in a coordinated manner to short‐term variations in CO2, providing new insight into the role of gm in photosynthesis modelling. Summary: To assess the effect of wheat milling or bran separation method, five wheat bran fractions, one from stone milling (SB), two from roller flour milling (coarse: RB1, fine: RB2), and two from grain pearling (0–3%: PB1, 3–6%: PB2) were analysed. Physiological properties provided good physical and flow characteristics of SB fraction; however, it was the darkest. Particle size was in the order of RB1 > SB > RB2 > PB1 > PB2. The fibre analysis showed the highest insoluble fibre components for RB1, PB1, and exceptionally higher soluble fibre for PB1. The protein, fat, ash, and starch content were highest for RB2 and PB2. Antioxidant properties followed the order PB2 > RB2 > PB1 > RB1 > SB. Pearled fractions (PB1 and PB2) showed better functional properties than others. SEM, XRD, and FT‐IR verified the changes in the properties of bran fractions. The characterisation data will find applications to help increase the usage of bran fractions in specific food products. A two-year field experiment was conducted at Lovely Professional University, Phagwara during the Rabi season of 2018–2019 and 2019-20 to study the effect of crop establishment methods and sowing schedule on the growth and yield of wheat under the rice-wheat cropping system. The experiment comprised different rice residue management-based wheat establishment methods, sowing dates, and varying nitrogen levels. The results confirmed that growth, yield and yield attributes were all influenced by different crop establishment methods, sowing dates and level of nitrogen application. The results showed that maximum plant height, number of tillers per plant, number of spikes per plant, spike length, number of grains per spike and grain yield were observed under 20th November sowing (timely sowing) and with 100% RDN (recommended dose of nitrogen). Moreover, the maximum number of spikes per plant, spike length and grain yield were significantly highest for the residue incorporation method of wheat establishment. The optimum time of sowing also improved the grain yield of wheat. The majority of farmers (85%) in Asia operate on small plots (< 2 ha) under low-input conditions to support their livelihoods. Wheat production in India faces challenges due to water scarcity and suboptimal nitrogen (N) management practices. To address this problem, a multi-location (9) field experiment comprising of 9 treatments by including nano and prilled urea alone or in combination was conducted under restricted irrigation conditions (only two irrigations i.e., first at crown root initiation stage and 2nd at flag leaf stage) during 2021–2022 and 2022–2023 in randomised block design with three replications in three agro-climatic regions (NWPZ- North Western Plains Zone, NEPZ- North Eastern Plains Zone and CZ- Central Zone) of India. Results indicated that combining recommended nitrogen (RDN) i.e., 90 kg/ha with two sprays of 5% urea increased grain yield by 4.1% and 41.9% in NWPZ and NEPZ, respectively, over RDN alone. Two sprays of nano urea at tillering and jointing stages significantly outperformed the control (Zero N), increasing grain yield by 14.5%, 34.0%, and 19.2% in NWPZ, NEPZ, and CZ, respectively. Moreover, RDN with two sprays of nano urea increased the grain yield and agronomic efficiency by 12.1% and 12.0% in CZ and 25.2% and 24.6% in NEPZ over RDN alone, respectively. From a net benefit perspective, RDN with two sprays of 5% urea was 6.7 and 70.4% higher in NWPZ and NEPZ over RDN under restricted irrigation. In CZ, applying RDN with two sprays of nano urea provided 15.5% higher net benefit over RDN. The recommendation for wheat cultivation in small-scale farming systems in India under restricted irrigation conditions suggests adopting a nitrogen management approach combining RDN with two sprays of 5% urea in NWPZ and NEPZ or with two sprays of nano urea in NEPZ and CZ improved the productivity, profitability and agronomic efficiency. The grain protein concentration (GPC) and yield are intimately and negatively correlated in wheat. However, how carbon (C) and nitrogen (N) supplies influence the wheat growth that underlies yield and quality formation has not been fully explored. Two wheat varieties with high GPCs (ZM578) and low GPCs (LY502) were grown under low and optimum N supplies in the field, and their growth, yields, and GPCs were closely investigated. The grain yield and total N uptake of the wheat variety ZM578 were comparable to those of LY502, but the GPCs increased by 12.4–16.1% on average. Compared with LY502, ZM578 had lower maximum stem numbers but similar spike numbers at maturity due to a lower rate of tiller mortality, which partly contributed to its grain yield. The grain N and C assimilation rates strongly and linearly correlated with each other, while steeper slopes were observed for the optimum- vs. low-N treatments, as well as for the high- vs. low-GPC treatments. Similarly, the C to N ratios in the developing grains of ZM578 were lower than those in the developing grains of LY502, regardless of N availability, which indicated that more N was transported to the grains per unit C in high-GPC ZM578. The random forest model revealed that the grain N uptake and leaf senescence (SPAD during grain filling) were the major controlling factors of yields and GPCs in cultivar ZM578. Ancient wheats are valuable genetic resources, though knowledge on their response to micronutrients in the presence of saline irrigation water is scanty. Two studies were conducted to unravel the behavior of ancient emmer and spelt wheats upon exposure to saline (75 and 150 mM NaCl) water and foliar-applied Zn (4 g L− 1) under pot and field conditions. Two weeks after implementing the salt treatment, Zn treatment was implemented twice with one week interval and the plants were exposed to the prolonged salt stress until physiological maturity. Then, an array of physiological processes underlying differential grain yield and quality responses of the ancient and standard wheats to these treatments were scrutinized. Salinity suppressed chlorophyll, relative water content, root volume, stubble yield, and grain yield of emmer and spelt and standard durum and bread wheats. Though, it increased the proline concentration, and Na+/K+ in all wheat genotypes. Emmer wheats indicated smaller salt-induced suppressions in stubble yield and grain yield, despite indicating a greater Na+/K+. Ancient emmer and spelt wheats indicated smaller grain yield components, but out-ranked the standard durum and bread wheats in terms of root volume, grain Zn, and protein concentrations. Zn's effect on the grain yield attributes and grain yield was moderate but it enhanced the grain Zn, particularly in emmer wheats. Novel findings of this study suggest that emmer wheats supplied with foliar-applied Zn are superior to standard durum and bread wheats in terms of grain protein and Zn, root volume, and tolerance to saline water. A comprehensive modelling methodology is proposed to describe wheat seeds using the discrete element method. By analysing the geometrical characteristics of wheat seeds, the multi-sphere approach is employed to establish 7-, 11-, 15-, 19-, and 23-sphere models based on ellipsoids. The physical and mechanical characteristics of wheat grain are measured and calibrated. Then, the proposed model is verified with several assessment criteria by contrasting the results of the experiment and simulation, including the wheat seed volume fraction, static angle of repose, hopper discharge, rotating drum and "self-flow screening". By balancing the accuracy of the multi-sphere model and computational efficiency, the 7-sphere or 11-sphere model is found to be the optimal model for determining the static stacking behaviour and hopper discharge of wheat seeds. For the rotating drum and the "self-flow screening", there is a considerable discrepancy between the simulation and experimental findings due to the surface roughness of the 7- and 11-sphere models. However, 15-, 19-, and 23-sphere models show a high accuracy, which can be applied for drying seeds of the rotating drum and accurately reproducing the sieve permeability of the "self-flow screening" experiment. After the 1960s, breeding activities carried out on many crops, including wheat, and the use of chemical fertilizers and pesticides, saw increased criticism of the negative consequences in terms of nutrition, social, and environmental impacts. There is a current re-emphasis upon changing the methods of flour and bread production technologies, and wheat landrace varieties have made a resurgence. Wheat landrace varieties have become widespread and create the demand for their consumption as an alternative to industrial wheat. Producers have come together to form collectives of different structures and seek ways to expand production of wheat landraces. As a result, alternative food consumption communities have become established in which consumers can access wheat landrace products. The factors related to the development of wheat, flour, bulgur, bread, etc. should be put forward to consumers and producers. The fieldwork of this research is based on five community supported food groups formed by consumers in the İzmir province of Turkey, and associations/groups that produce wheat varieties in Çanakkale, Balıkesir and Kars provinces. The aim of the research is to identify common problems that reduce the separation between wheat production and consumption and how to increase the use of wheat landraces and their products by using participatory action research. Tan spot disease of wheat (also known as yellow spot) is caused by the necrotrophic fungal pathogen Pyrenophora tritici‐repentis (Ptr). Initially described as a grass pathogen, Ptr has become a wheat pathogen of global importance. In Australia, tan spot was first recorded in 1952 as a minor wheat disease. However, by the 1970s tan spot had reached epidemic levels in Australia with yield losses of up to 49% reported in the north‐eastern region. A national survey carried out in 2007/2008 placed tan spot as the most economically important wheat disease with an estimated yield loss of 6%, corresponding to nearly a quarter of all disease losses. The incidence of tan spot in Australia has now markedly reduced with some wheat breeding programmes no longer considering tan spot disease resistance to be a priority trait. The disease can be effectively managed with integrated control strategies such as crop rotation, timely application of fungicides and removal of surface stubble. However, the key to the success of controlling tan spot disease in Australia has been achieved through wheat breeding programmes. The development of tan spot resistance resources suitable for the Australian climate was a concerted effort between national and international research organizations and Australian wheat breeders, supported by the Australian Grains Research Development Corporation (GRDC). While traditional breeding was essential to combat the disease, this article highlights the value of effector biology in assisting the development of resistant cultivars. [ABSTRACT FROM AUTHOR]

The resistance gene Lr34 conditions durable disease resistance to many biotrophic wheat pathogens and has been incorporated into many wheat cultivars throughout the world. Lr67 is a similar adult plant resistance gene that also conditions resistance to multiple wheat diseases. Both genes significantly reduce disease severity and work in an additive manner with other rust resistance genes. To determine the effect of Lr34 and Lr67 on Fusarium head blight (FHB), two doubled haploid populations, segregating for each of these genes, were developed in the Thatcher wheat background by crossing near‐isogenic lines. Progeny from these populations were tested in five Fusarium graminearum‐inoculated disease nurseries and assessed for FHB symptoms, using a visual rating index (VRI), and deoxynivalenol (DON) accumulation. Both Lr34 and Lr67 significantly reduced visual symptoms of FHB and DON overall, though those effects were not significant in all environments. The overall reduction in the group of progeny with the resistant allele compared to the group with the susceptible allele for VRI was 9.4% for Lr34 and 6.8% for Lr67, while for DON it was 16.4% for Lr34 and 14.9% for Lr67. The importance of monitoring and management of yellow rust, caused by Puccinia striiformis f. sp. tritici (Pst), has markedly increased in Russia over recent years because of intensifying negative effects of this destructive disease on wheat production. The Pst virulence survey was conducted in 2019–2021 in the European (North‐West, Volga, North Caucasus) and Asian (Siberia) parts of Russia, and the same 109 isolates have been genotyped here with 20 SSR markers. Fifty‐three multilocus genotypes (MLGs) were detected, seven of which occurred in two or more regions. The most frequent MLG was found in three European Pst collections (except Dagestan) and Siberia. SSR‐based variation within the regional collections was low because many MLGs were closely related. Nevertheless, each collection contained relatively different small genotype groups. No clear geographic structure of the Russian Pst population was observed because the five studied regional collections shared groups of similar genotypes. The most singular SSR genotype was identified in the North‐West collection, but genotypes of four invasive isolates belonging to the PstS2 temperature‐adapted aggressive strain did not considerably differ from others. The observed heterozygosity at all SSR loci exceeded the expected one in all regional Pst collections, which could imply asexual reproduction of the pathogen. Both the SSR and virulence analyses demonstrated similar patterns of relationships between the regional Pst collections, although the magnitude of differences between the collections was larger for virulence pathotypes. However, no significant association was established between the virulence pathotypes and SSR genotypes because multiple pathotypes had the same genotype and vice versa. [ABSTRACT FROM AUTHOR]

SUMMARY: Polyploidy is a prominent driver of plant diversification, accompanied with dramatic chromosomal rearrangement and epigenetic changes that affect gene expression. How chromatin interactions within and between subgenomes adapt to ploidy transition remains poorly understood. We generate open chromatin interaction maps for natural hexaploid wheat (AABBDD), extracted tetraploid wheat (AABB), diploid wheat progenitor Aegilops tauschii (DD) and resynthesized hexaploid wheat (RHW, AABBDD). Thousands of intra‐ and interchromosomal loops are de novo established or disappeared in AB subgenomes after separation of D subgenome, in which 37–95% of novel loops are lost again in RHW after merger of D genome. Interestingly, more than half of novel loops are formed by cascade reactions that are triggered by disruption of chromatin interaction between AB and D subgenomes. The interaction repressed genes in RHW relative to DD are expression suppressed, resulting in more balanced expression of the three homoeologs in RHW. The interaction levels of cascade anchors are decreased step‐by‐step. Leading single nucleotide polymorphisms of yield‐ and plant architecture‐related quantitative trait locus are significantly enriched in cascade anchors. The expression of 116 genes interacted with these anchors are significantly correlated with the corresponding traits. Our findings reveal trans‐regulation of intrachromosomal loops by interchromosomal interactions during genome merger and separation in polyploid species. Significance Statement: Understanding the chromatin interaction changes in response to ploidy transition can provide further insight into the role of chromatin loops in regulation of gene expression and phenotypes in polyploidy. We find that the ploidy transition can result in cascaded changes in chromatin interactions, which are associated with balancing homoeologous gene expression and altering the expression of coexpressed genes related to specific phenotypes in polyploids. SUMMARY: Global wheat production amounted to >780 MMT during 2022–2023 whose market size are valued at >$128 billion. Wheat is highly susceptible to high‐temperature stress (HTS) throughout the life cycle and its yield declines 5–7% with the rise in each degree of temperature. Previously, we reported an array of HTS‐response markers from a resilient wheat cv. Unnat Halna and described their putative role in heat acclimation. To complement our previous results and identify the key determinants of thermotolerance, here we examined the cytoplasmic proteome of a sensitive cv. PBW343. The HTS‐triggered metabolite reprograming highlighted how proteostasis defects influence the formation of an integrated stress‐adaptive response. The proteomic analysis identified several promising HTS‐responsive proteins, including a NACα18 protein, designated TaNACα18, whose role in thermotolerance remains unknown. Dual localization of TaNACα18 suggests its crucial functions in the cytoplasm and nucleus. The homodimerization of TaNACα18 anticipated its function as a transcriptional coactivator. The complementation of TaNACα18 in yeast and overexpression in wheat demonstrated its role in thermotolerance across the kingdom. Altogether, our results suggest that TaNACα18 imparts tolerance through tight regulation of gene expression, cell wall remodeling and activation of cell defense responses. [ABSTRACT FROM AUTHOR]

Spring wheat (Triticum aestivum L.) nitrogen-use efficiency (NUE) and recovery with nitrogen (N) fertilization rates in crop rotations need further exploration. The effect of cropping sequence and N fertilization rate were evaluated on spring wheat N concentration and uptake, N harvest index (NHI), N-use efficiency (NUE), and N recovery index (NRI) from 2012 to 2019 in the northern Great Plains, USA. Cropping sequences were conventional till spring wheat-fallow (CTWF), no-till spring wheat-fallow (NTWF), no-till spring wheat-pea (Pisum sativum L.) (NTWP), and no-till continuous spring wheat (NTCW), and N fertilization rates to spring wheat were 0, 50, 100, and 150 kg N ha−1. Straw N concentration and N uptake were greater for CTWF than NTWP and NTCW in 4 out of 8 years and increased with increased N fertilization rate. Grain N concentration and NHI varied with treatments and years. Grain N uptake was greater for CTWF and NTWF than NTWP and NTCW at most N fertilization rates and years. In contrast, NUE was greater for NTCW than CTWF and NTWF at all N fertilization rates in most years. The NRI was greater for NTWP or NTCW than CTWF or NTWF in most years. Increasing N fertilization rate increased grain N uptake for most cropping sequences in all years but had a variable effect on NUE and NRI. Although spring wheat N concentration and N uptake were superior for CTWF and NTWF, and NUE for NTCW, NTWP with 50 to 100 kg N ha−1 can be used to sustain grain yield, N uptake, NUE, and NRI. Consumer demand for healthier and more sustainable foods has led to a multitude of innovative food products. The aim of the work was to monitor the physico-chemical quality parameters (color, taste, smell, infestation, granularity, humidity, gluten index, falling number, alveographic properties, ash content, hectoliter mass of wheat) for the raw material: wheat, for the flours resulting from the milling, and for different types of flours and resulting mixtures. The mixtures were made with wheat flour and carob, oats, rice, buckwheat, and coconut flour added in proportions of 5%, 10%, and 20%. Heavy metals (Pb, Cd, Zn, and Cu) were also determined for different types of flour. The metal content was determined by the atomic absorption method using a GBC Avanta PM atomic absorption spectrometer. The detection and quantification limits were evaluated using calibration curves. The degree of coloring of the flours differs depending on the degree of extraction and, therefore, also depends on the amount of ash contained. As for infestation contamination, it was not identified in any of the analyzed flour. Rheological properties indicate the quality of flours. Flour types 480 and 550 recorded exceptional values of the rheological parameters, presenting a deformation energy (W) above 200 10-4J and a curve configuration ratio (P/E) of 1.08. [Display omitted] Flexible pressure sensors with high sensitivity and wide sensing range are highly desired in e-skins and wearable electronics. However, there is always a trade-off between high sensitivity and broad sensing range for most flexible pressure sensors. Herein, an artificial wheat awn-like hierarchical structure is designed onto the dielectric layer of the iontronic pressure sensor, realizing both high sensitivity and broad working range. The sensor is constructed by sandwiching a wheat awn-like polyvinyl alcohol/H 3 PO 4 dielectric layer between two transparent electrodes of silver nanowires/thermoplastic polyurethane/ionic liquid. The obtained sensor exhibits a high precision of 1 Pa, a high sensitivity of 47.65 kPa−1 (1–200 Pa), a wide measurement range from 1 Pa to 238 kPa, short response/recovery time of 13 ms/12 ms, outstanding stability over 6000 cycles, as well as good transparency. Long‐term genetic studies of wild populations are very scarce, but are essential for connecting ecological and population genetics models, and for understanding the dynamics of biodiversity. We present a study of a wild wheat population sampled over a 36‐year period at high spatial resolution. We genotyped 832 individuals from regular sampling along transects during the course of the experiment. Genotypes were clustered into ecological microhabitats over scales of tens of metres, and this clustering was remarkably stable over the 36 generations of the study. Simulations show that it is difficult to determine whether this spatial and temporal stability reflects extremely limited dispersal or fine‐scale local adaptation to ecological parameters. Using a common‐garden experiment, we showed that the genotypes found in distinct microhabitats differ phenotypically. Wheat plants infested by Russian wheat aphids (RWA) induce a cascade of defence responses, which include increased activity of β-1,3-glucanase and peroxidase (POD). There is a lack of information regarding β-1,3-glucanase and POD synergistic effects on the plant cell wall modification and characterisation during wheat-RWA infestation. This study aimed to characterise the physicochemical properties of the cell wall-bound POD and β-1,3-glucanase during RWA-wheat interaction. The susceptible Tugela, moderately resistant Tugela-Dn1, and resistant Tugela-Dn5 cultivars were planted in a glasshouse to a seedling stage before being infested with RWASA2 for 14 days. The findings showed a significant increase in β-1,3-glucanase and POD activities in the infested Tugela-Dn5 and Tugela-Dn1 cultivars over the 14 days. However, in the Tugela enzymes were repressed. In addition, it was shown for the first time that β-1,3 − 1,4-glucanase activity specific toward mixed-linked glucan was significant in the resistant cultivar over 14 days. β-1,3-glucanase, β-1,3 − 1,4-glucanase and POD displayed optimum at pH 5. β-1,3-glucanase and POD displayed temperature optimum at 40 and 50 °C, respectively. However, β-1,3 − 1,4-glucanase had temperature optimum at 25 °C. β-1,3-glucanase and POD had a thermo-stability at 37 °C followed by about 80% relative activity at 70 °C, but β-1,3 − 1,4-glucanase displayed thermostability at 25 °C and retained more than 75% at 70 °C, confirming that β-1,3 − 1,4-glucanase and β-1,3-glucanase induced in the resistant cultivars cell wall were two different enzymes. Mechanism of actions and oligosaccharide displayed that β-1,3-glucanase was highly active against β-1,3-glucan and required a triose and higher oligosaccharide to be active. Our findings demonstrated cell-wall bound POD and β-1,3-glucanase activities significantly increased in wheat after RWASA2 infestation, revealing they acted synergistically to reinforce the cell wall to deter RWASA2 feeding in resistant cultivars. The stress-protective effect of melatonin (N-acetyl-5-methoxytryptamine) on plant cells is mediated by key signaling mediators, in particular calcium ions and reactive oxygen species (ROS). However, the links between changes in calcium and redox homeostasis and the formation of adaptive responses of cultivated cereals (including wheat) to the action of high temperatures have not yet been studied. In the present study, we investigated the possible involvement of ROS and calcium ions as signaling mediators in developing heat resistance in wheat (Triticum aestivum L.) seedlings and activating their antioxidant system. Treatment of 3-day-old etiolated seedlings with melatonin solutions at concentrations 0.01–10 µM increased their survival after exposure to 45 °C for 10 min. The most significant stress-protective effect was exerted by melatonin treatment at 1 µM concentration. Under the influence of melatonin, a transient enhancement of superoxide anion radical (O2•–) generation and an increase in hydrogen peroxide content were observed in roots, with a maximum at 1 h. Four hours after treatment with melatonin, the activity of catalase and guaiacol peroxidase increased in roots, while the activity of superoxide dismutase did not change significantly. After exposure to 45 °C, the activity of catalase and guaiacol peroxidase was higher in the roots of melatonin-treated wheat seedlings, and the indices of ROS generation, content of the lipid peroxidation product malonic dialdehyde, and cell membrane damage were lower than in control seedlings. Melatonin-induced changes in root ROS generation and antioxidant enzyme activities were eliminated by pretreatment with the hydrogen peroxide scavenger dimethylthiourea (DMTU), NADPH oxidase inhibitor imidazole, and calcium antagonists (the extracellular calcium chelator EGTA and phospholipase C inhibitor neomycin). Treatment with DMTU, imidazole, EGTA, and neomycin also abolished the melatonin-induced increase in survival of wheat seedlings after heat stress. The role of calcium ions and ROS, generated with the participation of NADPH oxidase, as signaling mediators in the melatonin-induced antioxidant system and heat stress resistance of wheat seedlings have been demonstrated. Societal Impact Statement: Increasing crop productivity is often proposed as a key goal for meeting the food security demands of a growing global population. However, achieving high crop yields alone without meeting end‐use quality requirements is counter to this objective and can lead to negative environmental and sustainability issues. High yielding feed wheat crops in the United Kingdom are a typical example of this. The historical context of UK agricultural industrialisation, developments in plant breeding and wheat end‐use processing are examined. We then outline how employing innovations in plant breeding methods offer the potential to redress the balance between wheat quantity and quality. Summary: Bread wheat (Triticum aestivum L.) has historically been an important crop for many human civilisations. Today, variability in wheat supply and trade has a large influence on global economies and food security. The United Kingdom is an example of an industrialised country that achieves high wheat yields through intensive cropping systems and a favourable climate. However, only a minority of the wheat grain produced is of suitable end‐use quality for modern bread baking methods and most wheat produced is fed to livestock. A large agricultural land area and input use dedicated to producing grain for animal rather than human food has wide‐ranging negative impacts for environmental sustainability and domestic food production. Here we present an historical perspective of agricultural and economic changes that have resulted in UK production primarily focussing on wheat quantity over quality. Agricultural intensification, liberalisation of free trade in agricultural commodities, innovations in the milling and baking sector, developments in scientific understanding of genetics and plant breeding, and geopolitical changes have all played a role. We propose that wheat breeding plays a crucial role in influencing these issues and although wheat breeders in the United Kingdom have historically applied the most‐up‐to‐date scientific advances, recent advances in genomics tools and quantitative genetics present a unique opportunity for breeders to redress the balance between quantity and quality. Societal Impact Statement: The Green Revolution is commonly understood as the dramatic increase in food production in Mexico and India between the 1940s and 1970s due to the spread of new agricultural technologies, especially high‐yielding seed varieties. Beyond this conventional understanding, however, historians are now revealing the occurrence of—and connections between—various Green Revolutions across space and time. This article identifies a "fascist Green Revolution" through the history of agrarian development and scientific wheat breeding in fascist Italy (1922–1943). Through this case study, the article provides new insights into key themes of discussion around the Green Revolution such as the relationship between plant breeding, the environment, food security, and food self‐sufficiency. Summary: In 1925, the fascist regime launched the "Battle of Wheat," a plan of agrarian development aimed at achieving self‐sufficiency in wheat, Italy's main food crop. Key to this project was the replacement of traditional landraces with "elite" semi‐dwarf and early‐maturing wheat cultivars released by Italy's star plant breeder Nazareno Strampelli. Through the historical analysis of Strampelli's breeding program and of the implementation of the fascist "seed replacement" strategy, this article makes the case for viewing the Battle of Wheat as a "fascist Green Revolution."The article draws on new methods and perspectives developed by the historiography of the Green Revolution. It is grounded on the historical analysis of fascist‐era publications and archival documents produced by Italian crop scientists and agricultural research institutions.While "elite" cultivars spread rapidly in fertile and wealthy northern Italy, they struggled to take hold in the dry and poor Italian South. The article delineates the environmental, social, and economic reasons underpinning this divergence. Focusing in particular on agrarian scientists' approaches to crop improvement in the Italian South, the article shows how this "marginal" region became a key site for the development of scientific counter‐narratives and concrete alternatives to the official fascist strategy of "seed replacement."The article reveals the co‐existence of multiple approaches over space and time within fascist‐era agrarian science. Through this case study, it provides new elements to analyze key themes in science and agriculture historiography such as the tension between hyper‐modernization and agroecological concerns, centralized and decentralized research, imported technologies and local resources. Leaves play a crucial role in photosynthesis and respiration, ultimately affecting the final grain yield of crops, including wheat (Triticum aestivum L.); however, the molecular mechanisms underlying wheat leaf development remain largely unknown. Here, we isolated a narrow-leaf gene, TaWAK2-A, through a map-based cloning strategy. TaWAK2-A encodes a wall-associated kinase (WAK), for which a single Ala-to-Val amino acid substitution reduces the protein stability, leading to a narrow-leaf phenotype in wheat. Further investigation suggests that TaWAK2 directly interacts with and phosphorylates TaNAL1, a trypsin-like serine/cysteine protease. The phosphorylated TaNAL1 is then involved in the degradation of the zinc finger transcription factor TaDST, which acts as a repressor of leaf expansion by activating the expression of the cytokinin oxidase gene TaCKX9 and triggering in vivo cytokinin degradation. Therefore, our findings elucidate a signaling cascade involving TaWAK2-TaNAL1-TaDST that sheds light on the regulation of wheat leaf development. [ABSTRACT FROM AUTHOR]

BACKGROUND: The continuous cultivation of rice–wheat in the same field is a key element of double‐cropping systems in the Indo‐Gangetic plains. Yields of such cropping systems are increasingly challenged as climate change drives increases in temperature, terminal stress and uneven rainfall, delaying rice harvesting and subsequently delaying sowing of wheat. In this paper, we evaluate the optimum sowing dates to achieve high grain yield and quality of wheat cultivars in northwest India. Three cultivars of wheat, HD‐2967, HD‐3086 and PBW‐723, were sown on three different dates at the research farm of ICAR‐IARI, New Delhi, to generate different weather conditions at different phenological stages. Different biophysical attributes, photosynthetic rate, stomatal conductance and transpiration rate, were measured at different phenological stages. Yield and grain quality parameters such as protein, starch, amylopectin, amylose and gluten were measured in different cultivars sown on different dates. RESULTS: Biophysical parameters were found to be higher in timely sown crops followed by late‐sown and very late‐sown crops. Further, the different sowing dates had a significant (P < 0.05) impact on the grain quality parameters such as protein, starch, amylopectin, amylose and gluten content. Percentage increases in the value of starch and amylose content under timely sown were ~7% and 11.6%, ~5% and 8.4%, compared to the very late‐sown treatment. In contrast, protein and amylopectin contents were found to increase by ~9.7% and 7.5%, ~13.8% and 16.6% under very late‐sown treatment. CONCLUSION: High‐temperature stress during the grain‐filling periods significantly decreased the grain yield. Reduction in the grain yield was associated with a reduction in starch and amylose content in the grains. The protein content in the grains is less affected by terminal heat stress. Cultivar HD‐3086 had higher growth, yield as well as quality parameters, compared to HD‐2967 and PBW‐723 in all treatments, hence could be adopted by farmers in northwest India. © 2024 Society of Chemical Industry. Wheat is a crop of great food importance for food production in Brazil. However, the improvement of this crop is still concentrated in isolated areas, which hinders advances in new cultivation regions due to the extensive demand for existing information. In this investigation, four wheat genotypes analysed to study associations and interrelationships between wheat traits across nine environments during the agricultural years of 2016, 2017 and 2018. The experiment was conducted using a randomised block design with four repetitions, and the analysis was divided into two groups: morphological and productive traits of wheat, comprising three and eleven characteristics, respectively. Canonical correlations between the morphological and productive groups were found to be dependent in most environments. Taller plants with longer main spikes and longer tiller spikes can be indirectly selected to increase grain yield under different sowing conditions. The information generated in this study indicate that some care must be taken when using the interrelations between trait groups to select or recommend genotypes for broader regions, as these relationships are influenced by the specific conditions of each region and sub-region. Heat stress poses a significant environmental challenge that profoundly impacts wheat productivity. It disrupts vital physiological processes such as photosynthesis, by impeding the functionality of the photosynthetic apparatus and compromising plasma membrane stability, thereby detrimentally affecting grain development in wheat. The scarcity of identified marker trait associations pertinent to thermotolerance presents a formidable obstacle in the development of marker-assisted selection strategies against heat stress. To address this, wheat accessions were systematically exposed to both normal and heat stress conditions and phenotypic data were collected on physiological traits including proline content, canopy temperature depression, cell membrane injury, photosynthetic rate, transpiration rate (at vegetative and reproductive stage and 'stay-green'. Principal component analysis elucidated the most significant contributors being proline content, transpiration rate, and canopy temperature depression, which exhibited a synergistic relationship with grain yield. Remarkably, cluster analysis delineated the wheat accessions into four discrete groups based on physiological attributes. Moreover, to explore the relationship between physiological traits and DNA markers, 158 wheat accessions were genotyped with 186 SSRs. Allelic frequency and polymorphic information content value were found to be highest on genome A (4.94 and 0.688), chromosome 1A (5.00 and 0.712), and marker Xgwm44 (13.0 and 0.916). Population structure, principal coordinate analysis and cluster analysis also partitioned the wheat accessions into four subpopulations based on genotypic data, highlighting their genetic homogeneity. Population diversity and presence of linkage disequilibrium established the suitability of population for association mapping. Additionally, linkage disequilibrium decay was most pronounced within a 15–20 cM region on chromosome 1A. Association mapping revealed highly significant marker trait associations at Bonferroni correction P < 0.00027. Markers Xwmc418 (located on chromosome 3D) and Xgwm233 (chromosome 7A) demonstrated associations with transpiration rate, while marker Xgwm494 (chromosome 3A) exhibited an association with photosynthetic rates at both vegetative and reproductive stages under heat stress conditions. Additionally, markers Xwmc201 (chromosome 6A) and Xcfa2129 (chromosome 1A) displayed robust associations with canopy temperature depression, while markers Xbarc163 (chromosome 4B) and Xbarc49 (chromosome 5A) were strongly associated with cell membrane injury at both stages. Notably, marker Xbarc49 (chromosome 5A) exhibited a significant association with the 'stay-green' trait under heat stress conditions. These results offers the potential utility in marker-assisted selection, gene pyramiding and genomic selection models to predict performance of wheat accession under heat stress conditions. Background: CINNAMYL ALCOHOL DEHYDROGENASE (CAD) catalyzes the NADPH-dependent reduction of cinnamaldehydes into cinnamyl alcohols and is a key enzyme found at the final step of the monolignol pathway. Cinnamyl alcohols and their conjugates are subsequently polymerized in the secondary cell wall to form lignin. CAD genes are typically encoded by multi-gene families and thus traditionally organized into general classifications of functional relevance. Results: In silico analysis of the hexaploid Triticum aestivum genome revealed 47 high confidence TaCAD copies, of which three were determined to be the most significant isoforms (class I) considered bone fide CADs. Class I CADs were expressed throughout development both in RNAseq data sets as well as via qRT-PCR analysis. Of the 37 class II TaCADs identified, two groups were observed to be significantly co-expressed with class I TaCADs in developing tissue and under chitin elicitation in RNAseq data sets. These co-expressed class II TaCADs were also found to be phylogenetically unrelated to a separate clade of class II TaCADs previously reported to be an influential resistance factor to pathogenic fungal infection. Lastly, two groups were phylogenetically identified as class III TaCADs, which possess distinct conserved gene structures. However, the lack of data supporting their catalytic activity for cinnamaldehydes and their bereft transcriptional presence in lignifying tissues challenges their designation and function as CADs. Conclusions: Taken together, our comprehensive transcriptomic analyses suggest that TaCAD genes contribute to overlapping but nonredundant functions during T. aestivum growth and development across a wide variety of agroecosystems and provide tolerance to various stressors. The cultivation of wheat in North America represents a relatively recent endeavour, spanning a mere 150 years. This relatively brief period of its recent cultivation has allowed the opportunity to document the threats and diseases farmers had to face from the start. In this minireview, our primary objective is to provide a detailed exploration of the historical context on how we end up effectively managing what was once among the most destructive and hard to manage diseases affecting wheat, namely, common bunt (covered smut). This review delves into the early research efforts dedicated to understanding the biology of the causal pathogens and developing effective management approaches. These efforts encompass a wide spectrum of potential methods, ranging from seed treatments to cropping practices, and the development of genetic resistance. Throughout this exploration, we will also spotlight the remarkable scientific success story that has unfolded within the Canadian context. In essence, this review aims to provide a scientific examination of the history, challenges and innovative approaches associated with mitigating the impact of bunt pathogens on wheat cultivation in North America, and future challenges. Summary The integrity of wheat (Triticum aestivum) production is increasingly jeopardized by the fungal pathogen Blumeria graminis f. sp. tritici (Bgt), particularly amid the vicissitudes of climate change. Here, we delineated the role of a wheat transcription factor, TaNAC1, which precipitates cellular apoptosis and fortifies resistance against Bgt. Utilizing BiFC, co‐immunoprecipitation, protein quantification, luciferase report assays, we determined that cytoplasmic TaNAC1‐7A undergoes phosphorylation at the S184/S258 sites by TaCDPK20, facilitating its nuclear translocation. This migration appears to prime further phosphorylation by TaMPK1, thereby enhancing transcriptional regulatory activity. Notably, the apoptotic activity of phosphorylated TaNAC1‐7A is negatively modulated by the nuclear protein phosphatase PP2Ac. Furthermore, activation of TaNAC1 phosphorylation initiates transcription of downstream genes TaSec1a and TaCAMTA4, through binding to the C[T/G]T[N7]A[A/C]G nucleic acid motif. Suppression of TaNAC1, TaCDPK20, and TaMPK1 in wheat compromises its resistance to Bgt strain E09, whereas overexpression of TaNAC1 and silencing of PP2Ac markedly elevate resistance levels. Our results reveal the pivotal role of TaNAC1 in basal resistance which is mediated by its effects on homotypic fusion, vacuolar protein sorting, and the expression of defense‐related genes. Simple crop rotations like maize and soyabean grown in succession often coincide with simple weed management tactics, resulting in weed infestations that are highly adapted to the conditions maintained for the selected crops. Amaranthus tuberculatus is a prominent weed species which possesses biological characteristics that allow it to persist in maize–soyabean rotations. Spring wheat and oats emerge earlier than A. tuberculatus and are grown in narrower rows than maize and soyabean, which may increase their competitive capability. A field study which measured morphological and phenological characteristics of three A. tuberculatus cohorts that emerged under fallow, maize, soyabean, spring wheat and oats was conducted to compare their competitive ability. Early emerged A. tuberculatus grew similarly in fallow, maize and soyabeans and grew to heights of >150 cm but was significantly reduced in spring wheat and oats, never exceeding 40 cm. Heights of the last emerged cohorts were more influenced by crop canopies never reaching 15 cm in spring wheat and oats compared to >70 cm in fallow. Early emerged A. tuberculatus canopy diameter and branches plant−1 were reduced by >80% in spring wheat and oats compared to fallow. Flowering was delayed in spring wheat and oats compared to fallow and seed production and biomass were reduced by 99% across cohorts. Growth suppression of A. tuberculatus, based on the findings, can be achieved under dense, competitive crop canopies such as in spring wheat and oats and these crops should be considered as additional crops in maize–soyabean rotations. The water absorption capacity (WAC) of hard wheat (Triticum aestivum L.) flour affects end‐use quality characteristics, including loaf volume, bread yield, and shelf life. However, improving WAC through phenotypic selection is challenging. Phenotyping for WAC is time consuming and, as such, is often limited to evaluation in the latter stages of the breeding process, resulting in the retention of suboptimal lines longer than desired. This study investigates the potential of univariate and multivariate genomic predictions as an alternative to phenotypic selection for improving WAC. A total of 497 hard winter wheat genotypes were evaluated in multi‐environment advanced yield and elite trials over 8 years (2014–2021). Phenotyping for WAC was done via the solvent retention capacity (SRC) using water as a solvent (SRC‐W). Traits that exhibited a significant correlation (r ≥ 0.3) with SRC‐W and were evaluated earlier than SRC‐W were included in the multivariate genomic prediction models. Kernel hardness and diameter were obtained using the single kernel characterization system (SKCS), and break flour yield and total flour yield (T‐Flour) were included. Cross‐validation showed the mean univariate genomic prediction accuracy of SRC to be r = 0.69 ± 0.005, while bivariate and multivariate models showed an improved prediction accuracy of r = 0.82 ± 0.003. Forward validation showed a prediction accuracy up to r = 0.81 for a multivariate model that included SRC‐W + All traits (SRC‐W, Diameter, SKCS hardness and diameter, F‐Flour, and T‐Flour). The pervasive existence of nanoplastics (NPs) and microplastics (MPs) in soil has become a worldwide environmental concern. N/MPs exist in the environment in a variety of forms, sizes, and concentrations, while multi-omics studies on the comprehensive impact of N/MPs with different properties (e.g. type and size) on plants remain limited. Therefore, this study utilized multi-omics analysis methods to investigate the effects of three common polymers [polyethylene-NPs (PE-NPs, 50 nm), PE-MPs (PE-MPs, 10 μm), and polystyrene-MPs (PS-MPs, 10 μm)] on the growth and stress response of wheat, as well as the rhizosphere microbial community at two concentrations (0.05 and 0.5 g/kg). PS and PE exhibited different effects for the same particle size and concentration. PE-NPs had the most severe stress effects, resulting in reduced rhizosphere bacteria diversity, plant biomass, and antioxidant enzyme activity while increasing beneficial bacteria richness. N/MPs altered the expression of nitrogen-, phosphorus-, and sulfur-related functional genes in rhizosphere bacteria, thereby affecting photosynthesis, as well as metabolite and gene levels in wheat leaves. Partial least squares pathway models (PLSPMs) indicated that concentration, size, and type play important roles in the impact of N/MPs on the plant ecological environment, which could have essential implications for assessing the environmental risk of N/MPs. SUMMARY Hypersensitive response‐programmed cell death (HR‐PCD) regulated by Ca2+ signal is considered the major regulator of resistance against Puccinia triticina (Pt.) infection in wheat. In this study, the bread wheat variety Thatcher and its near‐isogenic line with the leaf rust resistance locus Lr26 were infected with the Pt. race 260 to obtain the compatible and incompatible combinations, respectively. The expression of translationally controlled tumor protein (TaTCTP) was upregulated upon infection with Pt., through a Ca2+‐dependent mechanism in the incompatible combination. The knockdown of TaTCTP markedly increased the area of dying cell and the number of Pt. haustorial mother cells (HMCs) at the infection sites, whereas plants overexpressing the gene exhibited enhanced resistance. The interaction between TaTCTP and calcineurin B‐like protein‐interacting protein kinase 23 (TaCIPK23) was also investigated, and the interaction was found occurred in the endoplasmic reticulum. TaCIPK23 phosphorylated TaTCTP in vitro. The expression of a phospho‐mimic TaTCTP mutant in Nicotiana benthamiana promoted HR‐like cell death. Silencing TaCIPK23 or TaCIPK23/TaTCTP co‐silencing resulted in the same results as silencing TaTCTP. Stripe rust, induced by Puccinia striiformis f. sp. tritici, is the most harmful and prevalent disease in temperate regions worldwide, affecting wheat production areas globally. An effective strategy for controlling the disease involves enhancing genetic resistance against stripe rust, achieved through Egyptian breeding efforts not previously conducted on wheat genotypes. The resistance level to stripe rust in thirty-eight wheat genotypes was assessed using marker-assisted selection methods. The investigation suggests that wheat breeding programs can utilize slow-rusting Yr genes, which are effective resistance genes, to develop novel genotypes with stripe rust resistance through marker-assisted breeding. Based on the four disease responses of the wheat genotypes under investigation, the results categorized the genotypes into three groups. The first group included resistant genotypes, the second group exhibited a slow-rusting character with the lowest disease symptom rates, and the last group displayed the highest disease characteristics rates throughout the three seasons, comprising fast-rusting genotypes. The rust-resistant genes identified were Yr5, Yr9, Yr10, Yr15, Yr17, Yr18, Yr26, Yr29, Yr30, and Yr36. Genes Yr26, Yr30, and Yr36 were present in all genotypes. Genotypes Misr3, Misr4, Giza168, Giza167, Giza170, Giza171, Gemmeiza9, and Gemmeiza10 carried the Yr9 gene. Only one genotype, Sids13, was found to have the Yr17 gene. Background: This study explores the use of Unmanned Aerial Vehicles (UAVs) for estimating wheat biomass, focusing on the impact of phenotyping and analytical protocols in the context of late-stage variety selection programs. It emphasizes the importance of variable selection, model specificity, and sampling location within the experimental plot in predicting biomass, aiming to refine UAV-based estimation techniques for enhanced selection accuracy and throughput in variety testing programs. Results: The research uncovered that integrating geometric and spectral traits led to an increase in prediction accuracy, whilst a recursive feature elimination (RFE) based variable selection workflowled to slight reductions in accuracy with the benefit of increased interpretability. Models, tailored to specific experiments were more accurate than those modelling all experiments together, while models trained for broad-growth stages did not significantly increase accuracy. The comparison between a permanent and a precise region of interest (ROI) within the plot showed negligible differences in biomass prediction accuracy, indicating the robustness of the approach across different sampling locations within the plot. Significant differences in the within-season repeatability (w2) of biomass predictions across different experiments highlighted the need for further investigation into the optimal timing of measurement for prediction. Conclusions: The study highlights the promising potential of UAV technology in biomass prediction for wheat at a small plot scale. It suggests that the accuracy of biomass predictions can be significantly improved through optimizing analytical and modelling protocols (i.e., variable selection, algorithm selection, stage-specific model development). Background: The need for rapid in-field measurement of key traits contributing to yield over many thousands of genotypes is a major roadblock in crop breeding. Recently, leaf hyperspectral reflectance data has been used to train machine learning models using partial least squares regression (PLSR) to rapidly predict genetic variation in photosynthetic and leaf traits across wheat populations, among other species. However, the application of published PLSR spectral models is limited by a fixed spectral wavelength range as input and the requirement of separate custom-built models for each trait and wavelength range. In addition, the use of reflectance spectra from the short-wave infrared region requires expensive multiple detector spectrometers. The ability to train a model that can accommodate input from different spectral ranges would potentially make such models extensible to more affordable sensors. Here we compare the accuracy of prediction of PLSR with various deep learning approaches and an ensemble model, each trained and tested using previously published data sets. Results: We demonstrate that the accuracy of PLSR to predict photosynthetic and related leaf traits in wheat can be improved with deep learning-based and ensemble models without overfitting. Additionally, these models can be flexibly applied across spectral ranges without significantly compromising accuracy. Conclusion: The method reported provides an improved prediction of wheat leaf and photosynthetic traits from leaf hyperspectral reflectance and do not require a full range, high cost leaf spectrometer. We provide a web service for deploying these algorithms to predict physiological traits in wheat from a variety of spectral data sets, with important implications for wheat yield prediction and crop breeding.
