It is unknown whether Ivor Lewis (IL) or McKeown (McK) esophagectomy is preferred in patients with potentially curable esophageal or gastro-esophageal junction (GEJ) cancer. Patients with mid- and distal esophageal and GEJ cancer without distant metastases who underwent IL or McK esophagectomy in the Netherlands between 2015 and 2017, were selected from the Netherlands Cancer Registry. Patients were propensity score matched for sex, age, American Society of Anesthesiologist classification, comorbidity, tumor type, tumor location, clinical stage, neoadjuvant treatment and year of diagnosis. The primary outcome was a 3-year relative survival (RS). Secondary outcome parameters were number of lymph nodes examined, number of positive lymph nodes, radical resection rate, tumor regression grade, post-operative complications and mortality. A total of 1627 patients who underwent IL (n  = 1094) or McK (n  = 533) esophagectomy were included. Patient and tumor characteristics were balanced after propensity score matching, leaving 658 patients to be compared. The 3-year RS was 54% after IL and 50% after McK esophagectomy, P  = 0.140. The median number of lymph nodes examined, median number of positive lymph nodes, radical resection rate and tumor regression grade were comparable between both groups. Recurrent laryngeal nerve palsy (2 vs. 5%, P  = 0.006) occurred less frequently after IL esophagectomy. No differences were observed in post-operative anastomotic leakage rate, pulmonary complication rate and mortality rates. There was no statistically significant difference in the 3-year RS between IL and McK esophagectomy. Background and aim Anastomotic leakage (AL) is a common and severe complication after esophagectomy, but it is largely unknown how to assess leakage severity at the time of diagnosis. We aimed to develop the Severity of Esophageal Anastomotic Leak (SEAL) score, a tool to predict leakage severity at the time of diagnosis. Methods The TENTACLE—Esophagus study is an international retrospective cohort study, in which 1509 patients with AL after esophagectomy between 2011 and 2019 were included in 71 centers from 20 countries. Potential predictors of mortality were patient parameters at diagnosis (e.g. diet at diagnosis, organ failure) and leakage parameters (e.g. fluid collections, gastric tube condition, leakage circumference). Primary outcome was 90-day mortality. Logistic regression was used to develop the SEAL score, comprising four grades of leakage severity (i.e. mild, moderate, severe and critical). Severity grade cut-offs were predefined and based on the individually predicted risk of 90-day mortality: 0–5%, 5–15%, 15–25% and 25–100%, respectively. Internal validation was performed by bootstrapping. The study protocol is accessible at www.tentaclestudy.com. Results The 90-day mortality rate in the included 1509 patients was 11.7%. The model had good discrimination (c-index 0.77; 95%CI 0.73–0.82) and calibration (calibration intercept 0.00; 95%CI -0.17– 0.17; calibration slope 1.14; 95%CI 0.95–1.33) after internal validation. The model classified 25.9% leakage patients as mild, 50.6% as moderate, 13.1% as severe and 10.4% as critical. The in-hospital, 30-day, 90-day and 180-day mortality rates, length of (ICU) stay, time to leak healing, Comprehensive Complication Index and Esophageal Complication Consensus Group classification worsened statistically significantly with higher classes of AL severity. Conclusion Anastomotic leakage is a severe complication with a 90-day mortality rate of 11.7%. The Severity of Esophageal Anastomotic Leak (SEAL) score is a tool which combines 12 patient and leak parameters at time of leak diagnosis and comprises four classes of leakage severity based on individual predictions of mortality. We aimed to investigate the effectiveness of different primary treatments of AL after esophagectomy. Methods The TENTACLE—Esophagus study is an international retrospective cohort study, in which 1509 patients with AL after esophagectomy between 2011 and 2019 were included. Detailed data were collected regarding case mix, resection (e.g. minimally invasive resection), leakage characteristics (e.g. organ failure, leak circumference, intrathoracic contamination) and leakage treatment (e.g. hours from diagnosis to treatment, primary and secondary treatment modalities such as endoscopic, radiologic and surgical treatment). An elaborate data quality validation and data accuracy assessment were performed. Primary outcome was 90-day mortality and secondary outcomes included length of stay and leak healing time. Regression analysis will be used to analyze the effectiveness of primary treatments after correction for patient-, surgery-, and leakage characteristics, and leakage severity. Different clinically relevant leakage groups have been defined. Efficacy of different treatment strategies adjusted for leakage severity will be analyzed in these clinical groups. In this study the ability of malignant and normal progenitors in peripheral blood (PB) and bone marrow (BM) of CML patients in chronic phase to proliferate and produce mature progeny after transplantation into hereditary immunodeficient (SCID and NOD/SCID) mice was examined. Engraftment in NOD/SCID mice preconditioned by total body irradiation (TBI) alone was 10-fold higher than in SCID mice preconditioned by macrophage depletion and TBI, demonstrating that NOD/SCID mice are more suitable for engraftment of chronic phase CML cells. Low-density cells at cell doses of 10-30 x 10(6) and purified CD34+ cells at doses of approximately 0.2 x 10(6) engrafted NOD/SCID mice, with levels of 2 to 20% CD45+ cells with production of monocytes, granulocytes, erythroid cells, B-lymphocytes, CD34+ cells and variable frequencies of erythroid and myeloid colony-forming cells. As demonstrated by fluorescent in situ hybridization (FISH) analysis, purified human myeloid, B-lymphoid, erythroid and CD34+ cells from chimeric mouse BM contained Philadelphia-chromosome (Ph)-positive cells and Ph- cells in similar frequencies as primary cells from the CML patients. These results demonstrate that production of mature normal as well as malignant cells of multiple lineages were supported with similar efficiency. In contrast, all human erythroid and myeloid clonogenic cells detected in the mice were Ph-, which can be attributed to less efficient maintenance or more rapid differentiation of immature Ph+ cells in the mouse microenvironment. CML blast crisis cells also grew well in NOD/SCID mice, with 80-90% of human cells produced containing the Ph- chromosome. The availability of an in vivo assay that supports outgrowth of normal and malignant stem cells from chronic phase and blast crisis CML patients will facilitate examination of differential effects of growth factors, inhibitory cytokines and cytotoxic drugs on survival of normal and malignant stem cells in vivo and on progression of chronic phase CML towards blast crisis. A study was conducted to determine the effect of two levels of linoleic acid (LA) intake at either high or low α-linolenic acid (ALA) intake on their conversion and subsequent deposition into long-chain (20-22 C-atoms) polyunsaturated fatty acids (LC PUFA) in muscle and backfat in growing pigs. In a 2 × 2 factorial arrangement, 32 gilts from 8 litters were assigned to one of four dietary treatments, varying in LA and ALA intakes. Low ALA and LA intakes were 0.15 and 1.31 g/(kg BW0.75/day), respectively, and high ALA and LA intakes were 1.48 and 2.65 g/(kg BW0.75/day) respectively. There was a close positive relation between intake of ALA and the concentration of ALA in backfat and in intramuscular fat. Dietary ALA did not affect the concentration of eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA), but increased docosapentaenoic acid (DPA) in backfat. High ALA intake did not significantly affect DHA but significantly increased EPA, 20:3 n-3 and DPA concentrations in intramuscular fat. The n-3 LC PUFA proportion in backfat was increased from approximately 1-3%, which may be useful to enrich meat with these fatty acids. The effect of ALA intake on n-3 LC PUFA was suppressed by LA intake. Dietary ALA suppressed the concentration of n-6 LC PUFA in blood plasma by more than 50%. Two experiments were conducted to determine the differences in starch digestion rate (KDS) among wheats from different cultivars and origins and to verify if chickens would benefit from a certain digestion rate of starch. In the first experiment, 192 chickens (21 d) were assigned to 4 diets containing 55% of each wheat sample (3 cultivars, one of them from 2 origins). Starch and protein digestion were calculated from the remaining starch and protein in 4 segments of the small intestine and in excreta, using chromic oxide as a marker. Mean retention time was measured in each segment, which enabled calculations of digestion rates. In the second experiment, 2,600 chickens were assigned to 5 isoenergetic and isonitrogenous diets (with KDS from 1.80 to 2.56 h-1) and growth performance was determined (1 to 34 d). In 3 treatments, dietary starch was provided each by the wheat cultivars (same origin), whereas in the other 2 treatments, 25 and 50% of the wheat starch with the highest KDS was substituted by pea starch. Clostridiurn perfringens and Lacto bacillus in the cecal chyme and glucose in the blood (glycemic index) were measured in broilers at d 19 and 25, respectively. Starch was gradually digested along the small intestine, mainly in the jejunum (48.5 and 80.4% at proximal and distal jejunurn) where the largest differences among wheat samples were found. Starch digestion rate varied with origin (from 1.96 to 2.56 h-1) and cultivar (from 2.17 to 2.56 h-1). Crude protein digestion rate (average 2.21 h') was not affected by either cultivar or origin. Broiler growth and feed conversion ratio improved in a quadratic way with KDS. The maximum broiler performance was observed with KDS around 2.2 h-1. Blood glucose response (glycemic index) was not affected by KDS; therefore, it cannot be used to predict broiler performance. Background and aim Transthoracic minimally invasive oesophagectomy (MIO) is increasingly performed as part of curative multimodality treatment. There is no robust evidence on the preferred location of the anastomosis after transthoracic MIO. The aim of this study was to compare an intrathoracic with a cervical anastomosis in a randomised controlled trial. Methods An open multicentre randomised controlled superiority trial was performed. Patients with mid to distal oesophageal or gastro-oesophageal junction cancer planned for curative resection were included. Patients were randomly assigned (1:1) to transthoracic MIO with intrathoracic or cervical anastomosis. The primary endpoint was anastomotic leakage requiring endoscopic, radiologic or surgical intervention. Secondary outcomes were overall anastomotic leak rate, other postoperative complications, length of stay, mortality and quality of life. The ICAN trial is registered in the Dutch trial register under number NTR4333. Results Two hundred and sixty-two patients were randomised and 245 were eligible for analysis. Anastomotic leakage necessitating re-intervention occurred in 15 of 122 (12.3%) patients with intrathoracic anastomosis and in 39 of 123 (31.7%) patients with cervical anastomosis (risk difference − 19.4%, 95% CI -29.5%—-9.3%). Overall anastomotic leak rate was 12.3% in the intrathoracic anastomosis group and 34.1% in the cervical anastomosis group (risk difference − 21.9%, 95% CI -32.1%—-11.6%). ICU length of stay, mortality rates and overall quality of life were comparable between groups, but intrathoracic anastomosis was associated with less severe complications, lower incidence of recurrent laryngeal nerve palsy and better quality of life in three subdomains. Conclusion Intrathoracic anastomosis is the preferred technique for patients treated with transthoracic MIO for mid to distal oesophageal or gastro-oesophageal junction cancer. A total of 5,000 one-day-old male broiler chickens were assigned to 8 different treatments in a 4 × 2 factorial design. Four wheat cultivars (Amiro, Guadalupe, Isengrain, and Horzal) and 2 levels (0 or 1 kg/t of feed) of an enzyme cocktail (Avizyme 1300, xylanase, 2,500 U/kg and protease, 800 U/kg) were used. Nutritionally complete mash diets contained 65 and 70% of the test wheat for the starter and grower period, respectively. Test wheats were used in diets for broilers, and growth performance and AME contents were measured. Broiler performance was measured in 4,800 broilers allocated to floor pens with 75 birds each and fed from 1 to 42 d of age. Digestibilities and AME contents of diets were measured in 200 broilers from 6 to 27 d of age individually allocated to battery cages. Chromic oxide (Cr2O3) at an inclusion rate of 0.5% in the diet was used as an indigestible marker. Apparent metabolizable energy was corrected by zero N balance to obtain AMEn. Wheat cultivar strongly influenced animal performance during the starter period (1 to 21 d of age). During the grower period (21 to 42 d of age), only BW and daily feed intake were influenced by wheat cultivar. Differences in daily feed intake were associated with differences in AMEn intake during the starter period, but not during the grower period. Nutrient digestibility was higher with the use of enzyme. Animal performance was not affected (i.e., wheat cultivar differences were not eliminated by using enzymes). During the grower period, significant interactions were detected with regard to nutrient digestibility and AMEn. Differences in AMEn content of wheat could not be explained by digestible starch. Cerebellar haemorrhage is an unusual complication of supratentorial neurosurgery. Several causative pre-operative factors and medical risk factors may predispose patients to cerebellar haemorrhage, however its etiology remains still unclear. Only two case reports have previously described the occurrence of cerebellar haemorrhage after subdural haematoma evacuation by burr-hole trepanation. We present two patients with this rare postoperative complication of minor supratentorial neurosurgery and possible underlying pathophysiological mechanisms are discussed. Our two cases support the post- rather than per-operative pathogenetic hypothesis. The objective of this study was to determine the effects of feeding frequency (FF) and feeding level (FL) on protein and energy metabolism in adapted, heavy preruminant calves. It was hypothesized that an increased FF would increase protein utilization by an improved synchrony between the supply of and requirements for protein during the day when a quickly hydrolyzable protein source was used. Eighteen Holstein Friesian calves of 136 ± 3 kg of body weight were assigned to FF (1, 2, or 4 meals daily) at 2 FL (1.5 or 2.5 times the metabolizable energy requirements for maintenance), except for calves fed once daily (only at a low FL). Calves were individually housed in respiration chambers during 2 experimental periods of 10 d. Whey protein was the only protein source in the diet. Neither FL nor FF affected apparent fecal nutrient digestibility. Increasing FF increased the efficiency with which digestible protein was utilized in calves. The increase was greater at a high FL (+11% from 2 to 4 meals/d) than at a low FL (+5% from 2 to 4 meals/d), but no significant interaction occurred between FL and FF. An increased FF and a higher FL enhanced fat deposition. Heat production was not affected by FF, but its circadian rhythm differed considerably between FF. Activity-related heat production was not affected by FF or FL. Abstract.The intracisternal (= ergastoplasmic) granules in the adipokinetic hormone-secreting cells of the glandular lobes of the corpora cardiaca in Locusta migratoria migratorioides represent accumulations of adipokinetic prohormones within cisternae of the rough endoplasmic reticulum. Solitary locusts have more and larger intracisternal granules than gregarious locusts. This coincides with the general locomotor activity and thus the energy metabolism in solitary locusts being quite different from that of gregarious locusts, which apparently has consequences for the amounts of adipokinetic hormones synthesized and/or released and, consequently, for the storage of these hormones in the intracisternal granules. 1. This study determined the effects of three protein sources (PS), each at two digestibility crude protein (DCP) levels, on performance, gut morphology and fermentation characteristics in the hindgut of broilers. 2. It was hypothesised that broilers fed ingredients high in indigestible CP, i.e. rapeseed meal (RSM) or maize gluten (MG), could potentially cause reduced growth, impaired gut health, and more protein fermentation products in caecal digesta. Increasing the DCP level in each of the indigestible CP diets may compensate for these detrimental effects. 3. In total, 288 one-d-old male Ross 308 broilers were used in a completely randomised 3 × 2 factorial design, with six replicate pens per treatment. Three PS: soybean meal (SBM), rapeseed meal (RSM) or maize gluten (MG), and two DCP levels: 15.8 and 17.2% were used. 4. Broilers fed SBM had increased feed intake and BWG and improved FCR compared with those fed RSM and MG diets. Broilers fed high DCP had better performance compared with those on low DCP. No significant effects of PS or DCP level were found on gastrointestinal tract development, caecal ammonia or volatile fatty acid concentrations. 5. Broilers fed SBM had longer villi, smaller crypts and increased villus height to crypt depth ratio compared with those fed RSM and MG diets. Broilers fed RSM diet had a lower caecal pH, and had 16.5% and 14.9% more branched chain fatty acid contents in caecal digesta compared with those fed SBM and MG diets, respectively, indicating more proteolytic fermentation. 6. Replacing SBM by RSM and MG negatively affected growth performance and gut morphology. Hindgut protein fermentation was substantially increased in RSM fed birds. 7. In pig (Sus scrofa) production, within-batch variation in BW gain of piglets during the nursery period (up to 10 wk of age) can be high and is of high economic importance. Homogeneity of BW within batches of animals is important as it influences the efficiency of use of the grower and finisher facilities, and provides an extra value for the fattening farms. In the current study, factors for a light BW at the end of the nursery period of pigs were determined by analyzing datasets from 3 different swine research centers in the Netherlands and France. The entire dataset contained information on 77,868 individual piglets bom between 2005 and 2010. Body weight was determined at different time points over the pre- and post-weaning phase, and sex, season of birth, litter information (litter size at day of birth and after cross-fostering, number of piglets bom alive per litter, number of total bom littermates, sow parity number), cross-fostered animals (yes or no), and pen group size over the post-weaning period were recorded. A risk factor analysis approach was used to analyze the datasets to determine factors that predict piglet BW at the end of the nursery period. Body weight at the end of the nursery period corrected for age was mainly determined by season (P < 0.001), birth weight (BiW, P < 0.001), weaning weight (WW, P < 0.001), and BW at 6 wk of age (P < 0.001). These variables were consistent among datasets and explained approximately 70% of the overall variation in BW at the end of the nursery period. Litter information did not significantly (P > 0.05) contribute to explaining the BW at the end of the nursery period. To discard the possibility of intrauterine growth retarded piglets (IUGR) being the reason for the influence of BiW as an explanatory factor in the regression model, a further analysis was performed on the effect of this category of piglets on the results of the regression analysis. Overall, it was concluded that the BW of piglets at the end of the nursery phase is mainly determined by season, sex, birth, WW, and BW at 6 wk of age. The period after hatch in broilers and around the time of weaning in pigs is critical for development and for adaptation of the small intestine to the nutritional changes. In broilers, the small-intestinal weight relative to body weight and villous height increase rapidly during the first week after hatch. After the first week, the relative small-intestinal weight decreases gradually, but the villous height continues to increase. In pigs, at 4 d after weaning, villous height decreases to about 60% of the pre-weaning height when weaned between 1 and 4 wk of age. Two weeks after weaning, this recovers to similar values as in unweaned control animals independent of weaning age. Small-intestinal development after hatch and after weaning consistently deteriorates at low feed intake levels and with suboptimal protein nutrition. The effect of dietary supplementation with N, N-dimethylglycine sodium salt (Na-DMG) was evaluated in a feeding trial with 1500 1-day-old broiler chicks (Cobb 500). DMG was supplemented at 0, 0.1, 0.2, 0.5 or 1 g Na-DMG/kg feed to a ration with either animal fat (chicken fat) or vegetal fat (soy oil) as main fat source. In the vegetal fat diets, production value was significantly linearly improved by supplementation with DMG up to 11%. Irrespective of dietary fat source, abdominal fat percentage was significantly linearly reduced up to 24% and meat yield tended to increase linearly with DMG level up to 4%. In the vegetal fat groups, DMG significantly lowered abdominal fat pad by up to 38% and tended to increase meat yield up to 6% at the highest dose. Fasted non-esterified fatty acid level significantly decreased with increasing DMG level up to 36% and thiobarbituric acid reactive species (TBARS) decreased with a statistical trend up to 46% at the highest dose. In vegetal fat diets, addition of DMG resulted in significant lower TBARS level by 56% at the highest dose. Finally, a significant quadratic effect on ascites heart index was present in the vegetal fat diets, with a minimal value at 0.5 g Na-DMG/kg. In conclusion, dietary supplementation with DMG may improve technical and slaughter performance, and may reduce oxidative stress and pulmonary hypertension, but the degree of effects is modulated by fatty acid profile of the diet. The increased productivity of sows increases the risk of a more pronounced negative energy balance during lactation. One possibility to prevent this is to increase the lactation efficiency (LE) genetically and thereby increase milk output for a given feed intake and mobilization of body tissue. The benefits of selection for LE depend on its heritability and the relationships with other traits of interest. The objectives of this study were to estimate genetic parameters for LE, its underlying traits, and to predict the consequences of current selection strategies in dam lines. Data from 4 farms were available to estimate genetic parameters. Heritabilities were estimated by using a univariate repeatability model, and genetic correlations were estimated bivariately. Selection index theory was used to predict the genetic progress by 3 alternative breeding programs: 1) a breeding program that aimed at balanced progress in the total number of piglets born, piglet mortality, and percent prolonged interval from weaning to estrus; 2) extension of this breeding goal with LE; and 3) a breeding goal that included only one selection criterion, litter weight gain, to demonstrate the effect of indirect selection for milk production. The heritability for LE was low (0.12). Body fat mass (0.52) and BW (0.45) of sows at the beginning of lactation showed the greatest heritabilities. Protein mass at the beginning of lactation, protein loss, weight loss, and ad libitum feed intake during lactation showed moderate heritabilities (0.39, 0.21, 0.20, and 0.30, respectively). Low to moderate heritabilities were found for litter weight at birth, within-litter SD in the birth weight of piglets, litter weight gain, fat loss, and restricted feed intake during lactation (0.19, 0.09, 0.18, 0.05, and 0.14, respectively). Within-litter SD in the weaning weight of piglets showed no genetic variability. It was predicted that a breeding goal for dam lines with an emphasis on the total number of piglets born, piglet mortality, and percent prolonged interval from weaning to estrus would not dramatically change BW or body composition at the beginning of lactation, or mobilization of body tissue and feed intake during lactation. Inclusion of LE in the breeding goal will improve stayability, as defined by the first-litter survival of sows and LE itself, without negative consequences for other economically important traits. Woolly monkeys (Lagothrix lagotricha) are a threatened specie in the wild with limited successful management in captivity due to diagnosed hypertension and suspected diabetic conditions. Six woolly monkeys with known hypertension problems were tested to determine if diabetes mellitus and current daily diet are underlying links to health problems for the captive population of this species. Blood and urine were collected and serum was analysed for fructosamine, glucose, glycated haemoglobin, insulin, triacylglycerides, total cholesterol, high-density lipoprotein cholesterol (HDL-Chol) and low-density lipoprotein cholesterol (LDL-Chol) while urine was tested for glucose concentrations. Diet disappearance was determined for 3 days prior to blood collection and nutrient content was calculated using Zoo Diet Analysis computer program. Serum analyses were within normal ranges (fructosamine (139–242 μmol/l), glucose (2.22–4.78 mmol/l), glycated haemoglobin (3.52–4.73%), insulin (6.2–13.0 μU/ml), triacylglycerides (0.38–3.4 mmol/l), total cholesterol (2.5–5.1 mmol/l), HDL-Chol (0.4–1.6 mmol/l) and LDL-Chol (1.8–3.4 mmol/l)). Urine glucose concentrations were below the detection limit. Diets were not limiting in starch and total sugars and were similar in non-starch polysaccharides. Potential dietary deficiencies were noted for vitamin A, vitamin D, calcium, phosphorus and selenium. When compared with the available primate reference ranges, the results do not indicate problems with diabetes mellitus or with glucose metabolism and therefore they are not causes of the diagnosed hypertension. ABSTRACT The effect of low-density diets on bird performance, egg composition, and embryonic development was studied with 2,100 female and 210 male Cobb broiler breeders from 25 to 60 wk of age. The experiment included 5 treatments. These included a control group with a normal density diet (ND, 2,800 kcal of AME/kg). Treatments 2 (LD11) and 3 (LD21) had a 11 and 21% lower nutrient density. Treatment 4 (LD11OP) had a 11% less dense diet, which was obtained by inclusion of other feed ingredients. In these 4 treatments similar diets were given during the rearing and the laying period. Treatment 5 combined LD12 in the rearing period and ND diets during the laying period (LD12-ND). Egg composition and embryonic development were measured in eggs of ND and LD21 birds at 29 and 41 wk of age. During the laying period from wk 25 to 60, live weights did not differ among treatments, except that birds fed LD11OP had lower live weights. A significantly higher rate of lay was provided by LD11 compared with ND. Egg weights were significantly higher when low-density diets were fed, particularly in LD11OP. Percentage of fertile eggs did not differ among treatments. Compared with the other treatments, LD11OP provided a significantly lower hatchability. We found that LD21 resulted in a better development of the area vitellina externa and heart and embryo weight at 29 wk of age. It was concluded that this was related to a higher egg weight and egg white proportion. Low-density diets might help to reduce hunger feeling in restricted-fed broiler breeders. Effects of low-density diets on nutrient digestibility and bird development were studied in Cobb 500 broiler breeder hens from 4 wk of age until the onset of the lay (wk 26). The experiment included 4 treatments. The control treatment was a normal density diet (ND; 2,600 kcal/kg). Treatments 2 and 3 had a 12 and 23% lower nutrient density than ND (LD12 and LD23, respectively) through inclusion of palm kernel meal, wheat bran, wheat gluten feed, and sunflower seed meal. Treatment 4 also had a 12% lower nutrient density than ND but included oats and sugar beet pulp (LD12°"). Total daily intake of energy, digestible lysine, calcium, retainable phosphorus, sodium, and linoleic acid was kept constant in the low-density dietary treatments. Animal performance and development of the intestinal tract and reproductive tract were determined in addition to digestibility and feed passage rate. The LD12OP provided a lower AME and nutrient digestibility than calculated and was related to lower live weights. Birds given LD23 and LD12OP showed a faster ovarian and oviduct development between wk 24 and 26 compared with ND. At 22 wk of age, LD23 and LD12OP diets resulted in higher empty jejunum and ileum weights. Low-density diets did not affect intestinal tract contents and decreased mean retention time of the contents, It was concluded that low-density diets can affect live weight and development of digestive and reproductive tracts. The effects of relative humidity (RH) and high ambient temperature (T) on physiological responses and animal performance were studied using 12 groups (10 gilts per group) in pens inside respiration chambers. The microclimate in the chamber was programmed so that T remained constant within a day. Each day, the T was increased by 2°C from low (16°C) to high (32°C). Relative humidity was kept constant at 50, 65, or 80%. The pigs' average initial BW was 61.7 kg (58.0 to 65.5 kg), and their average ending BW was 70.2 kg (65.9 to 74.7 kg). Respiration rate (RR), evaporative water (EW), rectal temperature (RT), skin temperature (ST), voluntary feed intake (VFI), water-to-feed ratio (rW:F), heat production (HP), and ADG were analyzed. The animals had free access to feed and water. We determined the T above which certain animal variables started to change: the so-called inflection point temperature (IPt) or upper critical temperature. The first indicator of reaction, RR, was in the range from 21.3 to 23.4°C. Rectal temperature was a delayed mdi- cat or of heat stress tolerance, with IPt values ranging from 24.6 to 27.1°C. For both these indicators the IPt was least at 80% RH (P < 0.05). Heat production and VFI were decreased above IPt of 22.9 and 25.5°C, respectively (P < 0.001). For each degree Celsius above IPt, the VFI was decreased by 81, 99, and 106 g/(pig·d) in treatments 50, 65, and 80% RH, respectively. The ADG was greatest at 50% RH (P < 0.05). Ambient temperature strongly affects the pigs' physiological changes and performance, whereas RH has a relatively minor effect on heat stress in growing pigs; however, the combination of high T and high RH lowered the ADG in pigs. The upper critical temperature can be considered to be the IPt above which VFI decreased and RT then increased. Temperatures of the magnitude of both these IPt are regularly measured in commercial pig houses. We conclude that the upper critical temperatures for 60-kg, group-housed pigs fed ad libitum are between 21.3 and 22.4°C for RR, between 22.9 and 25.5°C for HP and VFI, and between 24.6 and 27.1°C for RT. Earlier studies have shown that adult mice from a line selected for high litter size (S-line), in particular females, had higher residual food intake (RFI) than mice from a non-selected control line (C-line). It was suggested that this increase in RFI, in particular the mature selected females, may anticipate the metabolically stressful periods of pregnancy and lactation. The present study investigated whether body composition at maturity has been changed as a correlated response to selection, in order to support the offspring during pregnancy and lactation. Furthermore, part of the observed differences between individuals in RFI may be attributable to differing proportions of body protein and lipid. For these reasons, differences in body composition at maturity between males and females of the S-line and the C-line were investigated. Lipid percentage was similar for C-line animals and S-line females; S-line males had a significantly lower lipid percentage. Males had a higher protein percentage than females, in particular S-line males. The results show that body composition in adult non-reproductive females has not been affected as a correlated effect of selection for high litter size. Furthermore, the results suggest that the high lean content in S-line males may explain part of the high RFI compared with C-line animals. Body composition in S-line females probably does not explain the high RFI compared with S-line males and C-line animals. Transplantation of normal and malignant human hematopoietic cells into severe combined immunodeficient (SCID) mice allows for evaluation of long-term growth abilities of these cells and provides a preclinical model for therapeutic interventions. However, large numbers of cells are required for successful engraftment in preirradiated mice due to residual graft resistance, that may be mediated by cells from the mononuclear phagocytic system. Intravenous (i.v.) injection of liposomes containing dichloromethylene diphosphonate (Cl2MDP) may eliminate mouse macrophages in spleen and liver. In this study outgrowth of acute myeloid leukemia (AML) cells and umbilical cord blood (UCB) cells in SCID mice conditioned with a single i.v. injection of Cl2MDP liposomes in addition to sublethal total body irradiation (TBI) was compared to outgrowth of these cells in SCID mice that had received TBI alone. A two- to 10-fold increase in outgrowth of AML cells was observed in four cases of AML. Administration of 10(7) UCB cells reproducibly engrafted SCID mice that had been conditioned with Cl2MDP liposomes and TBI, whereas human cells were not detected in mice conditioned with TBI alone. As few as 2 x 10(4) purified CD34+ UCB cells engrafted in all mice treated with Cl2MDP liposomes. In SCID mice treated with macrophage depletion unexpected graft failures were not observed. Histological examination of the spleen showed that TBI and Cl2MDP liposomes i.v. resulted in a transient elimination of all macrophage subsets in the spleen, whereas TBI had a minor effect. Cl2MDP liposomes were easy to use and their application was not associated with appreciable side-effects. Sainfoin (Onobrychis viciifolia) is a tanniniferous legume forage that has potential nutritional and health benefits preventing bloating, reducing nematode larval establishment, improving N utilization, and reducing greenhouse gas emissions. However, the use of sainfoin as a fodder crop in dairy cow rations in northwestern Europe is still relatively unknown. The objective of this study was to evaluate the effect of sainfoin silage on nutrient digestibility, animal performance, energy and N utilization, and CH4 production. Six rumencannulated, lactating dairy cows with a metabolic body weight (BW0.75) of 132.5 ± 3.6 kg were randomly assigned to either a control (CON) or a sainfoin (SAIN)- based diet over 2 experimental periods of 25 d each in a crossover design. The CON diet was a mixture of grass silage, corn silage, concentrate, and linseed. In the SAIN diet, 50% of grass silage dry matter (DM) of the CON diet was exchanged for sainfoin silage. The cows were adapted to 95% of ad libitum feed intake for a 21-d period before being housed in climate-controlled respiration chambers for 4 d, during which time feed intake, apparent total-tract digestibility, N and energy balance, and CH4 production was determined. Data were analyzed using a mixed model procedure. Total daily DM, organic matter, and neutral detergent fiber intake did not differ between the 2 diets. The apparent digestibility of DM, organic matter, neutral detergent fiber, and acid detergent fiber were, respectively, 5.7, 4.0, 15.7, and 14.8% lower for the SAIN diet. Methane production per kilogram of DM intake was lowest for the SAIN diet, CH4 production as a percentage of gross energy intake tended to be lower, and milk yield was greater for the SAIN diet. Nitrogen intake, N retention, and energy retained in body protein were greater for the SAIN than for the CON diet. Nitrogen retention as CH4 a percentage of N intake tended to be greater for the SAIN diet. These results suggest that inclusion of sainfoin silage in dairy cow rations reduces CH4 per kilogram of DM intake and nutrient digestibility. An experiment with 288 male (Ross 308) 1-d-old broilers was conducted to test the hypothesis that a coarse diet supplemented with butyric acid (BA) and fermentable carbohydrates (FC) improves performance of broilers with a poorly digestible protein source. The interaction effects of diet structure (fine or coarse), FC supplementation (with or without), and BA supplementation (with or without) in a poorly digestible diet based on rapeseed meal (RSM) were tested in a factorial arrangement of 8 (2 × 2 × 2) dietary treatments. The coarseness of the diet affected feed intake (FI) (P < 0.001), BW gain (P = 0.001), and the feed conversion ratio (FCR) (P = 0.001) positively. Broilers fed the coarse diets had, on average, 14% heavier gizzards and 11, 7, 5, and 6% lower relative empty weights of the crop, duodenum, jejunum, and ileum, respectively, compared with those fed the fine diets. Dietary coarseness resulted in, on average, 6% greater ileal protein digestibility, 20% lower gizzard pH, 19% greater villus height, 18% lower crypt depth, and 23% reduced cecal branched chain fatty acids (BCFA) compared with chickens fed the fine diets. Broilers fed BA-supplemented diets had an improved FCR (P = 0.004) and decreased crypt depth (P < 0.001) compared with those fed diets without BA. Fermentable carbohydrate supplementation did not influence growth performance, gut development, or contents of total BCFA and total biogenic amines in the cecal digesta (P > 0.05). Supplementation with FC, however, decreased the cecal concentration of spermine by approximately 31% compared with broilers fed diets without FC (P = 0.002). In conclusion, feeding a coarse diet supplemented with BA improved performance of broilers fed a diet containing a poorly digestible protein source. An experiment with 210 male (Ross 308) 1-d-old broilers was conducted to test the hypothesis that a coarse diet improves performance of broilers fed a poorly digestible protein source. A highly digestible diet based on soybean meal was gradually replaced by a low digestible diet based on rapeseed meal (RSM) in 5 steps (RSM-0%, RSM-25%, RSM-50%, RSM-75%, and RSM-100%). Two diet structures (fine and coarse) were used as an additional factor. These 2 factors and their interactions were tested at different ages in a factorial arrangement with 10 dietary treatments. An increase in indigestible dietary protein negatively affected feed intake (P = 0.003), BW gain (P = 0.008), and feed conversion ratio (P = 0.034). This increase in dietary indigestible protein contents resulted in a decrease (P = 0.001) in total cecal volatile fatty acid concentration from 209.1 to 125.9 mmol/kg of DM digesta in broilers with increasing RSM in diets. Increase in the indigestible protein level, from RSM-0% to RSM-100%, resulted in a decrease (P = 0.042) in villus heights (1,782 vs. 1,574 pm), whereas crypt depths increased (P = 0.021; 237 vs. 274 µm). A coarse diet improved feed intake (P = 0.006), BW gain (P = 0.014), and feed conversion ratio (P = 0.009). Broilers fed coarse diets had approximately 11, 24, and 10% lower relative empty weights of the crop, proventriculus, and jejunum, respectively, whereas a 15% heavier gizzard was found compared with those fed the fine diets. Dietary coarseness resulted in approximately 16% lower gizzard pH, 21% greater villus heights, 27% lower crypt depths, 24% reduced branched-chain fatty acids, and 12% lower biogenic amines in the cecal digesta compared with broilers fed fine diets. In conclusion, feeding coarse particles improved broiler performance irrespective of digestibility of the diet. Hindgut protein fermentation can be reduced by coarse grinding of the diet. Milk fever is one of the most important metabolic diseases in dairy cattle. Reducing the dietary cation/anion balance ( DCAD) with anionic salts is a common prevention strategy. However, many small European farms cannot use total mixed rations ( TMR) in the close-up period. Including anionic salts in compound feeds can result in feed refusals and moderate inclusions to preserve feed palatability results in insufficient DCAD reduction. Rumen-protected rice bran induces the adaptation of Ca metabolism in dairy cows by a reduction of Ca intake and by a reduction of the availability of dietary Ca. In the presence of a negative control, rumen-protected rice bran (2.8 kg/day) was compared with a lowered DCAD diet (from 269 to 4 meq/kg DM) in their effect to prevent milk fever. In a randomized block design, 45 multiparous Holstein cows joined the trial sequentially from 21 days before the expected calving date and were observed until the 8th week of lactation. Feed and nutrient intakes were recorded, and Ca, P, Mg in serum and urine, urine pH, serum NEFA and milk production in early lactation were compared. Feeding rumen-protected rice bran before calving improved the recovery of calcaemia after calving and had a positive effect on DMI after calving. The moderately low DCAD diet did not positively influence serum Ca at calving. Calcaemia recovered even later than in control, and cows showed reduced DMI post-calving and higher NEFA levels in the first 36 h after calving. This moderate reduction of DCAD did not provide an intermediate prevention level indicating that DCAD needs to be reduced to the recommended levels to prevent milk fever. Rumen-protected rice bran may be a suitable feed to reduce hypocalcaemia post-partum and can be included in pre-calving compound feeds representing a palatable alternative to anionic salts. Litter moisture contents vary greatly between and within practical poultry barns. The current experiment was designed to measure the effects of 8 different dietary characteristics on litter and excreta moisture content. Additionally, free water content and water activity of the excreta and litter were evaluated as additional quality measures. The dietary treatments consisted of nonstarch polysaccharide content (NSP; corn vs. wheat), particle size of insoluble fiber (coarse vs. finely ground oat hulls), viscosity of a nonfermentable fiber (low- and high-viscosity carboxymethyl cellulose), inclusion of a clay mineral (sepiolite), and inclusion of a laxative electrolyte (MgS04). The 8 treatments were randomly assigned to cages within blocks, resulting in 12 replicates per treatment with 6 birds per replicate. Limited effects of the dietary treatments were noted on excreta and litter water activity, and indications were observed that this measurement is limited in highmoisture samples. Increasing dietary NSP content by feeding a corn-based diet (low NSP) compared with a wheat-based diet (high NSP) increased water intake, excreta moisture and free water, and litter moisture content. Adding insoluble fibers to the wheat-based diet reduced excreta and litter moisture content, as well as litter water activity. Fine grinding of the oat hulls diminished the effect on litter moisture and water activity. However, excreta moisture and free water content were similar when fed finely or coarsely ground oat hulls. The effects of changing viscosity and adding a clay mineral or laxative deviated from results observed in previous studies. Findings of the current experiment indicate a potential for excreta free water measurement as an additional parameter to assess excreta quality besides total moisture. Microbiota plays a role in the release and absorption of nutrients from feed components, thereby affecting digesta composition and moisture content of the excreta. The objective of the current study was to determine the effects of 5 different diets varying in ingredients (medium-chain fatty acids, nonstarch polysaccharides, and starch) on the microbiota composition of ileal digesta of broiler chickens and excreta DM content. Each treatment was repeated 6 times in cages each containing 18 Ross 308 broilers, with growth performance measured from 0 to 34 d of age and excreta DM and ileal microbiota composition analyzed at 34 d of age. Microbiota composition was evaluated using a novel ribosomal RNA microarray technology containing 370 different probes covering various genera, groups of microbial species, and individual species of the chicken gut microbiota, of which 321 had a signal above the background threshold. Replacing part of the animal fat and soybean oil in the wheat-based diet with medium-chain fatty acids (MCFA; 0.3% ClO and 2.7% C12) improved feed efficiency compared with the other dietary treatments. This coincided with a suppression of gram-positive bacteria belonging to the phylum of the Firmicutes, including Lacto bacillus species, and species belonging to the family of the Enterococcaceae and Micrococcaceae, whereas the gram-negative bacteria belonging to the family of the Enterobacteriaceae were promoted. None of the other diets used in the present study notably changed the ileal digesta bacteria composition. Excreta DM content was not affected by dietary treatment. The variation between individual birds per dietary treatment was more pronounced than variation caused by feed composition, with the exception of the digesta microbiota of the birds fed the MCFA diet. It is concluded that a diet with MCFA significantly changes the ileal microbiota composition, whereas the effect of the other diets on the composition of the microbiota and excreta DM content is small in broiler chickens. Reducing litter moisture is an effective measure to reduce the incidence of footpad dermatitis. Dietary mineral levels affect intestinal conditions with regard to osmolarity and water reabsorption. Magnesium is often used as a laxative, preventing reabsorption of water from the digesta, and as a consequence, more moisture in the excreta. The objective of the current experiment was to evaluate Mg in broiler diets as a model for reduced intestinal water reabsorption. Effects of magnesium source (magnesium sulfate, magnesium oxide, and magnesium chloride), each at 3 levels (0.255, 1.02, and 2.04 g-kg-1 diet), were studied. Measured effects were digesta moisture levels throughout the gastrointestinal tract and the moisture level of the excreta. The 10 treatments were randomly assigned to cages within 6 blocks, resulting in 6 replicates per treatments with 18 birds per replicate. Adding magnesium to the diet of broilers linearly increased the excreta moisture content, following the pattern MgCl > MgS04 = MgO. This rejects the hypothesis that MgO and MgCl are less laxative sources compared with MgSO4. The magnesium sources most likely changed the water reabsorption in the distal gastrointestinal tract, as confirmed by the increased digesta moisture percentage in the ceca and colon. Increasing dietary MgSCO4 linearly reduced BW gain and feed intake, though absolute differences were minor. Self-selection assumes that at high ambient temperature, birds are able to select a diet from different sources to minimize the heat load associated with the ingested nutrient metabolism. The objective was to test the hypothesis that young chickens are able to compose an adequate ration by adjusting dietary nutrient intake from 3 different diets that vary in energy and in protein contents from a cafeteria system at high temperature (HT; 31-32°C) and at normal temperature (NT; 31-21°C). Night temperature was set at 25°C at HT and at 18°C at NT and 12 h dark:12 h light. Control birds were fed a standard control diet (CP: 215 g/ kg; ME: 2,895 kcal/kg) for broiler chickens. The choicefed birds could choose between the control diet, a highprotein diet (CP: 299 g/kg; ME: 2,780 kcal/kg), and a high-energy diet (CP: 150.7 g/kg; ME: 3,241 kcal/kg). The diets had similar pellet size and color. Birds had access to each diet in a separate feeding trough from 1 to 42 d of age. Results showed that broilers spent 3.3% more time eating at NT than at HT and showed 42% more panting behavior at HT than at NT. High temperature decreased feed intake, protein intake, energy intake, and BW gain. Choice-fed birds had similar feed intake and BW gain, 14% lower protein intake, and 6.4% higher energy intake than control-fed birds. Body temperature and heterophil/lymphocyte ratio were higher at HT than at NT. Water intake was 8% higher in control-fed birds than in choice-fed birds but similar at both temperature regimens. It can be concluded that broilers can compose a diet by selecting less protein but higher energy density from different diets compared with the control. Choice-fed birds had similar feed efficiency as control-fed birds at HT, indicating similar body composition for both groups. Intestinal barrier function in pigs after weaning is almost exclusively determined in terminal experiments with Using chambers. Alternatively, the recovery in urine of orally administered lactulose can be used to assess intestinal permeability in living animals. This experiment was designed to study the barrier function of the small intestine of pigs over time after weaning. The aim was to relate paracellular barrier function (measured by lactulose recovery in the urine) with macromolecular transport [measured by horseradish peroxidase (HRP) using Ussing chambers] and bacterial translocation to assess whether lactulose recovery is related to possible causes of infection and disease. Forty gonadectomized male pigs (6.7 ± 0.6 kg) were weaned (d 0) at a mean age of 19 d, fitted with urine collection bags, and individually housed. Pigs were dosed by oral gavage with a marker solution containing lactulose (disaccharide) and the monosaccharides L-rhamnose, 3-0rnethylglucose, and D-xylose at 2 h and at 4, 8, and 12 d after weaning. The recovery of sugars in the urine was determined over 18 h after each oral gavage. The day after each permeability test, the intestines of 10 pigs were dissected to determine bacterial translocation to the mesenteric lymph nodes and jejunal permeability for HRP in Ussing chambers. Recovery of L-rharnnose in urine was affected by feed intake and by the time after weaning (P < 0.05). Recovery of lactulose from the urine was greater (P 0.05) at 4, 8, and 12 d after weaning compared with the first day after weaning and was negatively correlated with feed intake (r = -0.63, P < 0.001). The mean translocation of aerobic bacteria to the mesenteric lymph nodes was greater at 5 and 13 d after weaning compared with d 1 (P < 0.05). Lactulose recovery showed no correlation with permeability for HRP nor with bacterial translocation (P> 0.05). Although both lactulose recovery and bacterial translocation increased over time after weaning, lactulose recovery did not correlate with the permeability for HRP nor bacterial translocation within a pig (P> 0.05). Therefore, we conclude that lactulose recovery in the urine of pigs after weaning is not associated with risk factors for infections. However, it appears to be possible to measure paracellular barrier function with orally administered lactulose in pigs shortly after weaning. Milk fever in dairy cows can be prevented by activating Ca homeostasis before calving. Homeostatic adaptation can be achieved by reducing dietary Ca availability. Formaldehyde-treated rice bran was studied to supply rumen protected phytic acid to reduce Ca availability. Twelve multiparous dry cows were used in a 3 × 3 Latin square change-over design with 5-day periods to test three dietary treatments. Diets consisted of a forage mix (maize silage, grass silage and hay), being 77% of ration dry matter, supplemented with three concentrates: Control (no formaldehyde-treated rice bran), T1 (100% formaldehyde-treated rice bran) and T2 (99.5% formaldehyde-treated rice bran with 0.6% Ca carbonate, to equal Ca content of Control). Dietary treatments did not affect urine pH (8.14, 8.13 and 8.11 for Control, T1 and T2 respectively) or dry matter intake (13.9, 13.7 and 13.8 kg for Control, T1 and T2 respectively). Including formaldehyde-treated rice bran in the diet resulted in lower urinary Ca/creatinine ratio (0.970, 0.457 and 0.618 for Control, T1 and T2 respectively). A sudden increase of urinary Ca excretion took place after withdrawal of T1 and T2 at introduction of Control, peaking on the first day and coming back down progressively in the second and third days. Peak was greatest after T1 and was not observed in transitions between rice bran treatments. This is understood as indirect evidence of activation of intestinal Ca absorption during formaldehyde-treated rice bran feeding, because renal adaptations to changes in blood Ca clearance are immediate and intestinal adaptations delay 2 days. It was concluded that including formaldehyde-treated rice bran in rations before calving may represent a dietary strategy to prevent milk fever without reducing dry matter intake. Adapting Ca homeostasis of dairy cows before calving can prevent milk fever. Rice bran, treated with formaldehyde to prevent ruminal degradation of phytic acid, was fed to heifers to study its effect on Ca homeostasis. For 3 weeks 18 heifers were supplemented 3 kg of two feeds: placebo (PF) and rice bran (RBF), defining three treatments: control (CRT), low dose (LD) and high dose (HD). In weeks 1 and 3, all animals received 3 kg of PF and in week 2: CRT received 3 kg of PF, LD received 1.5 kg of PF and 1.5 kg of RBF and HD received 3 kg of RBF. Treatments did not affect dry matter intake (DMI). Feed intakes and growth rates indicated that all heifers had nutritional requirements that exceeded their Ca intakes. Serum Ca, urinary Ca, calcitriol or hydroxyproline remained unaffected. Urinary Ca was consistently low indicating high renal Ca reabsorption, which is indicative of insufficient Ca supply. Rice bran feed influenced P, Mg and Zn intakes and serum and urine presence of these minerals. Most heifers already presented an upregulated Ca metabolism, being inadequate to study adaptive changes in Ca homeostasis of multiparous dry cows. This metabolic difference can be explanatory to the very low susceptibility of heifers to milk fever, further supporting the induction of homeostatic adaptation before calving to prevent milk fever. Two experiments were conducted in growing pigs to determine the effects of dietary CP levels and specific crystalline amino acids supplementation to a diet on odor emission, odor intensity, hedonic tone, and ammonia emission from pig manure. In each experiment, pigs (n=18) were allocated in a randomized complete block arrangement having three treatments in six blocks. In the first experiment, treatment groups were 12%, 15% and 18% CP diets. In the second experiment, treatment groups were 1) diet with three times requirement of sulfurcontaining AA (Met + Cys), 14.2 g/kg diet, 2) diet with two times requirement of Trp, and Tyr + Phe, 2.9 and 20.4 g/kg diet as fed basis, respectively and 3) control diet with 15 % CP and with supplementation of these AA up to requirement. Pigs with initial body weight of around 39 kg were kept individually in partly slatted floor pens. Daily feed allowance was 2.8 x maintenance requirement for net energy (293 kJ W^0.75). Feed was mixed with water, 1/2.5 (w/w). Feces and urine of each pig were accumulated together in a separate manure pit during four weeks under the slatted floor. In the fifth week air samples for odor and ammonia analyses were collected directly from each manure pit. Reducing CP from 18% to 12% lowered odor emission and ammonia emission (P < 0.05) from pig manure by 80% and 53%, respectively, and increased odor hedonic tone (less unpleasant odor) (P < 0.05) but did not affect odor intensity (P > 0.05) of the odorous air. Supplementing S-containing AA in surplus of pigs' requirement increased odor emission by 723% and odor intensity and reduced odor hedonic tone (P < 0.05) of odorous air. Supplementing Trp, Tyr, and Phe in surplus of pigs' requirement did not affect odor emission, odor intensity and odor hedonic tone. No differences were observed in ammonia emission from manure of pigs fed different types of AA (P > 0.05). The hypothesis tested was that dietary vegetable fats rich in saturated fatty acids, when compared with a vegetable oil rich in linoleic acid, increase fat deposition in broiler chickens and affect synthesis or oxidation, or both, of individual fatty acids. Diets with native sunflower oil (SO), a 50:50 mix of hydrogenated and native SO, palm oil, and randomized palm oil were fed to broiler chickens. Intake of digestible fat and fatty acids, whole body fatty acid deposition, hepatic fatty acid profile, and hepatic enzyme activities involved in fatty acid oxidation and synthesis were measured. The fat deposition:digestible fat intake ratio was significantly lower for the SO group in comparison with the groups fed the vegetable fats rich in saturated fatty acids. The difference between digestible intake and deposition of C18:2, reflecting its maximum disappearance rate, was highest for the SO group and lowest for the palm oil- and randomized palm oil-fed birds. The calculated minimal rate of de novo synthesis of monounsaturated fatty acids (MUFA), calculated as deposition minus digestible intake, was more than 50% lower for the SO group than for the other 3 dietary groups. Based on the fatty acid profiles in the liver, it would appear that increasing contents of C18:2 decrease the desaturation of saturated fatty acids into MUFA. It is concluded that a diet rich in C18:2 in comparison with different kinds of vegetable saturated fatty acids decreases the deposition of fat, especially of MUFA. It appears to be caused by a higher 3-oxidation and a reduced de novo synthesis of MUFA, but this conclusion is not fully supported by the measured activities of enzymes involved in fatty acid synthesis and oxidation. This study investigated the effect of 2 different dietary amino acid treatments and feed restriction in early life versus a control treatment on development of the small intestine segments (weights), mortality, and broiler performance. Each treatment was applied to 6 cages with Ross 308 male broilers and to 6 cages with Cobb 500 male broilers with 24 birds per cage. A control treatment (100% ideal protein) was compared with a treatment with 30% extra ideal protein, a treatment with daily adjustment of the dietary amino acid level and profile, and a feed restriction treatment. The protein treatments were applied from 0 to 14 d of age. The feed restriction was applied from 4 to 21 d of age. Restriction was 15% from d 4 to 14 of age and diminished with equal daily steps thereafter to 5% at 21 d of age. Birds were weighed and dissected for evaluation of small intestine weights at 6, 9, 14, and 36 d of age. Feed intake restriction reduced leg problems in Ross and Cobb broilers. Extra dietary protein reduced leg problems in Ross broilers only. The present experiment does not show that small intestinal weight development is related to mortality. Thirty percent extra dietary ideal protein increased duodenum weight between 6 and 9 d of age. This was not further increased by the daily optimization of the dietary amino acid level and profile. The increased duodenum weights coincided with an improved BW gain. This indicates that duodenum weight may be important in facilitating BW gain in young broilers. At calving, many older cows fail to compensate the sudden demand of calcium by an adequate activation of intestinal absorption. This results in a variable degree of hypocalcaemia. Reducing intestinal availability of calcium during the close-up period can prevent milk fever. Fat-coated rice bran (FCRB) was investigated for its potential to reduce Ca availability in pre-calving cows. Fat-coated rice bran was incubated in situ to estimate ruminal degradation of dry matter and phytic acid. Also, seven dry multiparous dairy cows were used for a feeding trial in three periods of approximately 1 week each: P1: adaptation; P2: feeding of 2 kg of FCRB and P3: withdrawal of FCRB. Feed intake was recorded and daily urine samples were analysed for pH, Ca and creatinine. The bypass fraction of phytic acid (passage rate: 5%/h) was 30%. Fat-coated rice bran depressed dry matter intake in P2, resulting in a lower Ca intake. In P2 urine pH and calcium excretion were lower. Daily calcium excretion decreased after introduction of FCRB, peaked after withdrawal and dropped 2 days later. Postprandial oxidation of dietary free amino acids or egg white protein was studied using the [13CO2] breath test in rats, as well as in humans. Thirty-eight male rats were assigned to four dietary test groups. Two diets only differed in their protein fraction. Diet I contained 21% egg white protein. For the breath test egg white protein, intrinsically labelled with [1-13C]-leucine, was used as a substrate. Diet II contained the same amino acids as diet I, though not as egg white protein but in free form. Free [1-13C]-leucine was used to label this diet. In addition, two 1:1 mixtures of both diets were used. During the breath test either the free amino acid or the protein fraction was labelled as in diets I or II. The animals were breath-tested following short-term (day 5) and long-term adaptation (day 20) to their experimental diet. For all diets, including the mixed diets, postprandial oxidative losses on day 5 were significantly higher for the free leucine compared with the protein-derived leucine. Differences between free and protein-derived leucine oxidation had, however, largely disappeared on day 20. The human subjects were breath-tested without any adaptation period to the diets. The oxidative losses of free leucine were also higher than those of protein-derived leucine. None of the studies showed any indication for an interaction between the oxidation of protein-derived amino acids and free amino acids. An experiment was conducted with 768 non-cage-housed ISA Brown pullets, of which 576 hens were followed during the laying period, to investigate the separate effects of dietary energy dilution and non-starch polysaccharides (NSP) concentration (oat hulls as NSP source) on eating behavior and feather damage. Day old pullets were allotted to. 1 of 6 dietary treatments according to a 3 x 2 factorial arrangement (3 dilution and 2 NSP levels), with 8 replicates (pens) per treatment. At 17 wk of age, pens with hens were allotted to 1 of 8 dietary treatments according to a 4 x 2 factorial arrangement (4 dilution and 2 NSP levels), with 6 replicates per treatment. Compared with 0% dilution level, feed intake of laying hens of 10, 15, and 20% dilution levels increased by 8.4% (9.5 g/hen per d), 16.5% (18.1 g/hen per d), and 20.9% (23.6 g/hen per d), respectively. The ME,, intake was similar for all dilution levels. Hens fed standard-NSP laying diets had similar insoluble NSP intake for all dilution levels (9.3 g/hen per d). Insoluble NSP intake of hens fed high-NSP laying diets increased from 15.6 g/hen per day (0% dilution) to 18.9 g/hen per day (20% dilution). Providing high- vs. standard-NSP layer diet decreased relative proventriculus contents (1.1 vs. 0.3 g/kg of BW) and increased empty gizzard weight (14.3 vs. 24.4 g/kg of BW). Hens that were fed standard-NSP diets had more feather damage compared with hens fed high-NSP diets (0.58 vs. 0.30 arbitrary units). Increasing the insoluble NSP intake resulted in decreased proventricular weight and increased gizzard weight and its contents, which are indicators of improved functioning of the gut, there-by linearly reducing feather damage. Providing diluted rearing diets increased feed intake from the first weeks of life onwards. It was hypothesized that pullets were increasingly "imprinted" on feed as pecking substrate if dilution level increased.
