Bovine serum albumin (BSA) hydrogels are non-immunogenic, low-cost, biocompatible, and biodegradable. In order to avoid toxic cross-linking agents, gellan was oxidized with NaIO4 to obtain new functional groups like dialdehydes for protein-based hydrogel cross-linking. The formed dialdehyde groups were highlighted with FT-IR and NMR spectroscopy. This paper aims to investigate hydrogel films for biomedical applications obtained by cross-linking BSA with oxidized gellan (OxG) containing immobilized β-cyclodextrin–curcumin inclusion complex (β-CD–Curc) The β-CD–Curc improved the bioavailability and solubility of Curc and was prepared at a molar ratio of 2:1. The film's structure and morphology were evaluated using FT-IR spectroscopy and SEM. The swelling degree (Q%) values of hydrogel films depend on hydrophilicity and pH, with higher values at pH = 7.4. Additionally, the conversion index of -NH2 groups into Schiff bases increases with an increase in OxG amount. The polymeric matrix provides protection for Curc, is non-cytotoxic, and enhances antioxidant activity. At pH = 5.5, the skin permeability and release efficiency of encapsulated curcumin were higher than at pH = 7.4 because of the interaction of free aldehyde and carboxylic groups from hydrogels with amine groups from proteins present in the skin membrane, resulting in a better film adhesion and more efficient curcumin release. Glioblastoma multiforme (GBM) is a highly aggressive malignant tumor, and the most prevalent primary malignant tumor affecting the brain and central nervous system. Recent research indicates that the genetic profile of GBM makes it resistant to drugs and radiation. However, the main obstacle in treating GBM is transporting drugs through the blood–brain barrier (BBB). Albumin is a versatile biomaterial for the synthesis of nanoparticles. The efficiency of albumin-based delivery systems is determined by their ability to improve tumor targeting and accumulation. In this review, we will discuss the prevalence of human glioblastoma and the currently adopted treatment, as well as the structure and some essential functions of the BBB, to transport drugs through this barrier. We will also mention some aspects related to the blood–tumor brain barrier (BTBB) that lead to poor treatment efficacy. The properties and structure of serum albumin were highlighted, such as its role in targeting brain tumors, as well as the progress made until now regarding the techniques for obtaining albumin nanoparticles and their functionalization, in order to overcome the BBB and treat cancer, especially human glioblastoma. Amylase is an enzyme used to hydrolyze starch in order to obtain different products that are mainly used in the food industry. The results reported in this article refer to the immobilization of α-amylase in gellan hydrogel particles ionically cross-linked with Mg2+ ions. The obtained hydrogel particles were characterized physicochemically and morphologically. Their enzymatic activity was tested using starch as a substrate in several hydrolytic cycles. The results showed that the properties of the particles are influenced by the degree of cross-linking and the amount of immobilized α-amylase enzyme. The temperature and pH at which the immobilized enzyme activity is maximum were T = 60 °C and pH = 5.6. The enzymatic activity and affinity of the enzyme to the substrate depend on the particle type, and this decreases for particles with a higher cross-linking degree owing to the slow diffusion of the enzyme molecules inside the polymer's network. By immobilization, α-amylase is protected from environmental factors, and the obtained particles can be quickly recovered from the hydrolysis medium, thus being able to be reused in repeated hydrolytic cycles (at least 11 cycles) without a substantial decrease in enzymatic activity. Curcumin has essential therapeutic benefits, but it is insoluble in water and thus has low bioavailability. This study aimed to immobilize curcumin into new polysaccharide-based microparticles (gellan, i-carrageenan, and chitosan) to increase its stability and bioavailability. Curcumin-loaded complex microparticles were obtained from three polysaccharides, of different ionic character, by ionic cross-linking and polyelectrolyte complexation. The immobilization efficiency was between 85.75% and 97.25%. The microparticles were characterized morphologically by SEM, and it was observed that the microparticles containing the i-carrageenan had a more pronounced porosity of the matrix. The swelling degree values at pH = 7.4 were superior to those obtained at pH = 6.8 or pH = 2 and depend on both the cross-linking degree and particles morphology. The polysaccharides microparticles, curcumin, and constituent polysaccharides were characterized by FT-IR spectroscopy. The curcumin release kinetics was studied in three different pH media, and the release efficiency ranged between 65.1% and 97.9% at pH = 7.4, between 60.2% and 82.2% at pH = 6.8 and between 56.1% and 64.0% at pH = 2. The purpose of this study is to obtain ionically cross-linked gellan particles containing immobilized yeast cells. The zinc acetate was chosen as a cross-linking agent due to its beneficial role in the growth and nutrition of yeast cells and also due to the advantages of its use in fermentation processes. The physicochemical and morphological properties as well as the fermentative activity of the obtained particles were studied. The zinc acetate concentration has an influence on particles stability in water, on the mechanical stability and also on the swelling degree. The morphology of the obtained particles was analyzed by SEM and proves that the yeast cells are immobilized in large numbers in the polymeric matrix. The cell viability is maintained at high values after several fermentation cycles. These obtained micro-bioreactors were tested in terms of fermentative activity and the fermentation process was optimized by studying the influence of several factors. The gellan gum particles can be easily recovered by filtration and they can be reused in repeated fermentation cycles. The paper reports the preparation of particles, having a hydrogel behavior, with immobilized yeast cells from a mixture of polysaccharide based on gellan, i-carrageenan, and carboxymethyl cellulose sodium salt. These particles were ionically cross-linked, using the extrusion method, with magnesium acetate, calcium acetate and zinc acetate and were characterized, by FTIR and SEM, in terms of some physicochemical properties, morphology, stability in water, and w ater retention capacity. These properties are influenced by the composition of polysaccharide mixture, which determines the number of cross-linkable functional groups, and also by the type and the concentration of the cross-linking agent. Analysis, by scanning electron microscopy, reveals that the yeast cells are well immobilized and in high amounts in the polysaccharides matrix. This matrix maintains the cell viability at high levels in aqueous media. High fermentation yields were obtained and the specific productivity in ethanol was equal to 0.79 g ethanol/h x g yeast cells. Highest ethanol yield was obtained for the sample cross-linked with Mg2*, and the lowest for the sample cross-linked with Zn2+. Polysaccharides matrix provides structural stability to yeast cells and maintains the cells viability, at values higher than 82% even after 10 fermentation cycles, as well as their ability to proliferate. Moreover, the obtained particles are stable, can be readily recovered by filtration from the fermentation medium and can be reused for at least 10 fermentation cycles. In this paper we report on the production of microbioreactors using ionically cross-linked gellan containing immobilized yeast cells with potential application in glucose fermentation. Cross-linking was achieved through a novel extrusion process in capillary by ionotropic gelation under the action of magnesium acetate. Compared to commonly used methods, this provides a host of practical advantages. The particles were physico-chemically and morphologically characterized as their mechanical stability, behavior in aqueous media, and bio-catalytic activity are influenced by the amount of cross-linker used. This demonstrated their ability to be reused in a large number of fermentation cycles without losing their bio-catalytic activity. Our results are wholly comparable with the behavior of free yeast. We show that fermentation cycles can succeed either immediately or at variable intervals, ensuring high yields of glucose transformation, comparable-if not superior-to results currently obtained using free yeast. Smart polymeric micelles (PMs) are of practical interest as nanocarriers for the encapsulation and controlled release of hydrophobic drugs. Two hydrophobic drugs, naturally-based curcumin (Cur) and synthetic 5-fluorouracil (5-FU), were loaded into the PMs formed by a well-defined pH-sensitive poly(2-vinyl pyridine)-b-poly(ethylene oxide) (P2VP90-b-PEO398) block copolymer. The influence of the drug loading on the micellar sizes was investigated by dynamic light scattering (DLS) and it appears that the size of the PMs increases from around 60 to 100 nm when Cur is loaded. On the contrary, the loading of the 5-FU has a smaller effect on the micellar sizes. This difference can be attributed to higher molar mass of Cur with respect to 5-FU but also to higher loading efficiency of Cur, 6.4%, compared to that of 5-FU, 5.8%. In vitro drug release was studied at pH 2, 6.8, and 7.4, and it was observed that the pH controls the release of both drugs. At pH 2, where the P2VP sequences from the "frozen-in" micellar core are protonated, the drug release efficiencies exceed 90%. Moreover, it was demonstrated, by in vitro assays, that these PMs are hemocompatible and biocompatible. Furthermore, the PMs protect the Cur against the photo-degradation, whereas the non-ionic PEO corona limits the adsorption of bovine serum albumin (BSA) protein on the surface.
