Tissue engineering is an interdisciplinary field that combines materials, methods, and biological molecules to engineer newly formed tissues to replace or restore functional organs. Biomaterials-based scaffolds play a crucial role in developing new tissue by interacting with human cells. Tissue engineering scaffolds with ideal characteristics, namely, nontoxicity, biodegradability, and appropriate mechanical and surface properties, are vital for tissue regeneration applications. However, current biocomposite scaffolds face significant limitations, particularly in achieving structural durability, controlled degradation rates, and effective cellular integration. These qualities are essential for maintaining long-term functionality in vivo. Although commonly utilized biomaterials can provide physical and chemical properties needed for tissue regeneration, inadequate biomimetic properties, as well as insufficient interactions of cells-scaffolds interaction, still need to be improved for the application of tissue engineering in vivo. It is impossible to achieve some essential features using a single material, so combining two or more materials may accomplish the requirements. In order to achieve a proper scaffold design, a suitable fabrication technique and combination of biomaterials with controlled micro or nanostructures are needed to achieve the proper biological responses. This review emphasizes advancements in scaffold durability, biocompatibility, and cellular responsiveness. It focuses on natural and synthetic polymer combinations and innovative fabrication techniques. Developing stimulus-responsive 3D scaffolds is critical, as these scaffolds enhance cell adhesion and promote functional tissue formation while maintaining structural integrity over time. This review also highlights the natural polymers, smart materials, and recent advanced techniques currently used to create emerging scaffolds for tissue regeneration applications. Pectin has recently drawn much attention in biomedical applications due to its distinctive chemical and biological properties. Polymers like pectin with cell-instructive properties are attractive natural biomaterials for tissue repair and regeneration. In addition, bioactive pectin and pectin-based composites exhibit improved characteristics to deliver active molecules. Pectin and pectin-based composites serve as interactive matrices or scaffolds by stimulating cell adhesion and cell proliferation and enhancing tissue remodeling by forming an extracellular matrix in vivo. Several bioactive properties, such as immunoregulatory, antibacterial, anti-inflammatory, anti-tumor, and antioxidant activities, contribute to the pectin's and pectin-based composite's enhanced applications in tissue engineering and drug delivery systems. Tissue engineering scaffolds containing pectin and pectin-based conjugates or composites demonstrate essential features such as nontoxicity, tunable mechanical properties, biodegradability, and suitable surface properties. The design and fabrication of pectic composites are versatile for tissue engineering and drug delivery applications. This article reviews the promising characteristics of pectin or pectic polysaccharides and pectin-based composites and highlights their potential biomedical applications, focusing on drug delivery and tissue engineering. A refractive index (RI) sensor has been demonstrated and numerically analyzed by the finite element method (FEM) to gain supreme sensitivity on the basis of localized surface plasmon resonance (LSPR). Plasmonic material, such as silver, has been used as a coating layer outside of the dielectric core. Various structural parameters have been analyzed for the proposed sensors, which have a significant influence on the sensing field. To make the sensing strength much stronger, a thin MgF2 layer has been added to the silver layer. The wavelength and amplitude interrogation method achieves extremely high wavelength sensitivity (WS) responses of 948.67 μm/RIU and amplitude sensitivity (AS) responses of − 1602.82 RIU−1 in the analyte RI range of 1.35 to 1.38. The operating wavelength range of 5.5–7.5 μm shows efficient results in this structure. The proposed sensor is used for identifying analytes in the biological and biochemical fields and is also applicable in bio-photonics. Owing to its low confinement losses and simply designed structure, the proposed RI sensor will be extensively appropriate for bio-sensing because of its outstanding sensitivity response. This work has developed a theoretical model to investigate the mechanisms of nonlinear steady-state photocurrent versus dose rate characteristics in Cs 2 AgBiBr 6 -based X-ray detectors. The photocurrent analysis on Cs 2 AgBiBr 6 -based X-ray detector shows that there is a photocurrent gain, primarily due to the presence of dark current. Charge injection from metal contacts is the primary source of dark current in Cs 2 AgBiBr 6. The accumulation of X-ray induced ions near the electrodes elevates the metal/perovskite interface electric field, and hence increases the charge injection with the X-ray dose. The Schottky barrier height for charge injection is found to be dependent on the X-ray dose as well. Therefore, the variation of dark current under X-ray exposures are mainly responsible for the non-linear steady-state photocurrent. Considering the important factor of bioactive nanohydoxyapatite (nHA) to enhance osteoconductivity or bone-bonding capacity, nHA was incorporated into an electrospun polycaprolactone (PCL) membrane using electrospinning techniques. The viscosity of the PCL and nHA/PCL with different concentrations of nHA was measured and the morphology of the electrospun membranes was compared using a field emission scanning electron microscopy. The water contact angle of the nanofiber determined the wettability of the membranes of different concentrations. The surface roughness of the electrospun nanofibers fabricated from pure PCL and nHA/PCL was determined and compared using atomic force microscopy. Attenuated total reflectance Fourier transform infrared spectroscopy was used to study the chemical bonding of the composite electrospun nanofibers. Beadless nanofibers were achieved after the incorporation of nHA with a diameter of 200-700 nm. Results showed that the fiber diameter and the surface roughness of electrospun nanofibers were significantly increased after the incorporation of nHA. In contrast, the water contact angle (132° ± 3.5°) was reduced for PCL membrane after addition of 10% (w/w) nHA (112° ± 3.0°). Ultimate tensile strengths of PCL membrane and 10% (w/w) nHA/PCL membrane were 25.02 ± 2.3 and 18.5 ± 4.4 MPa. A model drug tetracycline hydrochloride was successfully loaded in the membrane and the membrane demonstrated good antibacterial effects against the growth of bacteria by showing inhibition zone for E. coli (2.53 ± 0.06 cm) and B. cereus (2.87 ± 0.06 cm). The development of nano-sized scaffolds with antibacterial properties that mimic the architecture of tissue is one of the challenges in tissue engineering. In this study, polycaprolactone (PCL) and PCL/gelatine (Ge) (70:30) nanofibrous scaffolds were fabricated using a less toxic and common solvent, formic acid and an electrospinning technique. Nanofibrous scaffolds were coated with silver (Ag) in different concentrations of silver nitrate (AgNO) aqueous solution (1.25, 2.5, 5, and 10 %) by using dipping method, drying and followed by ultraviolet (UV) photoreduction. The PCL/Ge (70:30) nanofibrous scaffold had an average fibre diameter of 155.60 ± 41.13 nm. Characterization showed that Ag was physically entrapped in both the PCL and PCL/Ge (70:30) nanofibrous scaffolds. Ag ions release study was performed and showed much lesser release amount than the maximum toxic concentration of Ag ions in human cells. Both scaffolds were non-toxic to cells and demonstrated antibacterial effects towards Gram-positive Bacillus cereus ( B. cereus) and Gram-negative Escherichia coli ( E. coli). Background: Despite high prevalence of intimate partner violence (IPV) and its adverse social and health consequences, the rate of help seeking for IPV is generally low. Although the level of IPV is much higher in urban slums of Bangladesh, the level and nature of help seeking of the victims are unknown. This paper aims to address this gap in the literature.Methods: Using a cross-sectional survey conducted between August 2011-February 2012, we explored disclosure of violence, help seeking behavior, and their correlates among randomly selected currently married women aged 15-29 in Dhaka slums (n = 2604).Results: About 60 % of the currently married women reported past year spousal physical violence, but only 21 % disclosed and 19 % sought any help. High acceptance of violence was the main reason for not seeking help. Help was most commonly sought from informal sources (89 %). Any education, frequent and severe physical abuse, and presence of children increased the likelihood of disclosure and help seeking. Most survivors from slum who disclosed also sought help.Conclusions: Despite widespread physical abuse, many survivors never sought help. Wide acceptance of violence hampering help seeking needs to be challenged. Increasing disclosure would also enhance help seeking. Awareness rising regarding rights of women to live a violence free life is essential. Although many services are available in the urban area, information about these services needs to be available to women. Cellulose-based materials are one of the most commonly used materials for biomedical applications, which normally applied as carriers for pharmaceuticals and drug-releasing scaffolds. In this study, cellulose acetate (CA) was used to fabricate the nanofibrous membrane using the electrospinning technique. CA solutions at different concentrations were prepared by dissolving the polymer in a mixture of acetic acid/acetone solvents with the ratio of 3 : 1. The field emission scanning electron microscope results showed that electrospinning of 10% (w/v) CA produced nanofibres with many beads. When the CA concentration was increased to 14% (w/v), bead-free nanofibres were produced. The contact angle measurement results confirmed the hydrophilic properties of nanofibres. In order to prevent common bacterial infections, a model drug, Tetracycline ⋅ HCL was incorporated into the CA nanofibres. The drug-loaded CA nanofibres showed antibacterial activity against Gram-positive and Gram-negative bacteria. CA nanofibres had high water uptake properties. The CA nanofibrous membrane was non-toxic to human skin fibroblast cells. Thus the CA nanofibres with 14% (w/v) concentration exerted suitable properties for wound healing application. Adrenomedullins (AM) is a multifaceted distinct subfamily of peptides that belongs to the calcitonin gene-related peptide (CGRP) superfamily. These peptides exert their functional activities via associations of calcitonin receptor-like receptors (CLRs) and receptor activity-modifying proteins (RAMPs) RAMP2 and RAMP3. Recent studies established that RAMPs and CLRs can modify biochemical properties such as trafficking and glycosylation of each other. However there is very little or no understanding regarding how RAMP or CLR influence ligand-induced events of AM-receptor complex. In this study, using pufferfish homologs of CLR (mfCLR1–3) and RAMP (mfRAMP2 and mfRAMP3), we revealed that all combinations of CLR and RAMP quickly underwent ligand-induced internalization; however, their recycling rates were different as follows: mfCLR1–mfRAMP3 > mfCLR2–mfRAMP3 > mfCLR3–mfRAMP3. Functional receptor assay confirmed that the recycled receptors were resensitized on the plasma membrane. In contrast, a negligible amount of mfCLR1–mfRAMP2 was recycled and reconstituted. Immunocytochemistry results indicated that the lower recovery rate of mfCLR3–mfRAMP3 and mfCLR1–mfRAMP2 was correlated with higher proportion of lysosomal localization of these receptor complexes compared to the other combinations. Collectively our results indicate, for the first time, that the ligand-induced internalization, recycling, and reconstitution properties of RAMP–CLR receptor complexes depend on the receptor-complex as a whole, and not on individual CLR or RAMP alone. Conventional control of sucking pests infesting various vegetables has several drawbacks. Biorational insecticides are increasingly used for pest management due to their limited side effects. A study involving several biorational compounds, viz. spinosad, buprofezin, Beauveria bassiana, Lecanicillium muscarium and a chemical insecticide, fenitrothion was performed by leaf-dip bioassay to measure their efficacy against the bean aphid, A. craccivora Koch. as a representative of sucking pests. Mortality of aphids against treatment with biorational insecticides was recorded in three replicates under laboratory conditions. The lethal concentration (LC50 and LC90) of the treatments were calculated by probit analysis. Efficacy was concentration and time dependent. Spinosad has been found to be more effective than buprofezin. The spinosad (LC50 = 2582.04 ppm) and buprofezin (LC50 = 543.03 ppm) were found to be approximately 88 and 52 times less toxic than fenitrothion. At 48 h post-treatment (HPT), the highest mortality for spinosad and buprofezin was 66.67 and 60.00%, respectively, and reached 93.33% at 72 HPT. Among the two fungal biopesticides, L. muscarium was more effective than B. bassiana. For both, mortality (80–100%) at the higher concentrations was statistically different than others at 144 and 168 HPT. Abstract: Adrenomedullins (AM) form a multifunctional subfamily of the calcitonin gene-related peptide (CGRP) superfamily, the members of which exert their physiological roles through a 1:1 combination of calcitonin receptor-like receptors (CLRs) and receptor activity-modifying proteins (RAMPs). It has been shown that RAMPs can modify the biochemical properties of CLRs; for example, RAMP escorts CLR to the plasma membrane, affects glycosylation state of CLR, and transforms the ligand selectivity of CLR, but on the other hand the effects of CLRs on the biochemical and functional properties of the partner RAMPs are not well established. In this study, using pufferfish (mefugu, mf) homolog, we revealed that mfCLR1 could affect the post-translational modification and trafficking pathway of mfRAMP1. In addition, mfCLRs boosted mfRAMP1, mfRAMP2b, and mfRAMP3 translocation to cell surface. We further revealed that mfRAMPs, except mfRAMP1 and mfRAMP3, could be expressed as multimers on the plasma membrane. However, only monomeric form of mfRAMP2a, mfRAMP4, and mfRAMP5 could heteromerize with mfCLR1 but not with mfCLR2 or mfCLR3, which was consistent with their abilities to induce cAMP response. Collectively our results indicate that the glycosylation, subcellular trafficking, and pharmacological properties of the components of RAMP–CLR receptor complexes are regulated in an interdependent manner. Heart development is a precisely coordinated process of cellular proliferation, migration, differentiation, and integrated morphogenetic interactions, and therefore it is highly susceptible to developmental anomalies such as the congenital heart disease (CHD). One of the major causes of CHD has been shown to be the mutations in key cardiac transcription factors, including nkx2.5. Here, we report the analysis of zebrafish mutant ftk that showed a progressive heart malformation in the later stages of heart morphogenesis. Our analyses revealed that the cardiac muscle maturation and heart morphogenesis in ftk mutants were impaired because of the disorganization of myofibrils. Notably, we found that the expression of nkx2.5 was down-regulated in the ftk heart despite the normal expression of gata4 and tbx5. suggesting a common mechanism for the occurrence of ftk phenotype and CHD. We identified ftk to be a loss-of-function mutation in a connexin gene, cx36.7/early cardiac connexin (ecx), expressed during early heart development. We further showed by a rescue experiment that Nkx2.5 is the downstream mediator of Ecx-mediated signaling. From these results, we propose that the cardiac connexin Ecx and its downstream signaling are crucial for establishing nkx2.5 expression, which in turn promotes unidirectional, parallel alignment of myofibrils and the subsequent proper heart morphogenesis. Abstract: Calcitonin receptor (CTR) is a member of class B G protein-coupled receptor (GPCR) superfamily. We have recently identified the CTR gene and its two transcriptional isoforms, mfCTR and mfCTRΔN, in mefugu (mf) (Takifugu obscurus). Here we characterized the mfCTRΔN that lacks hormone-binding extracellular N-terminal domain. Strong expression in the liver and weak but broad tissue distribution of its mRNA, revealed by Northern analysis, suggested physiological significance of this headless splice variant. Biochemical and immunocytochemical analyses revealed that it acts as a naturally occurring dominant negative isoform by forming a heterodimer with normal CTR. The headless mfCTRΔN characterized here is the first case of N-terminally truncated dominant negative form of GPCR, and immunocytochemistry used for detecting the heterodimer formation may be useful as a novel method for analyzing membrane protein interaction in a living cell. Abstract: Endothelin, a vasoconstrictor peptide, plays important roles not only in the mammalian circulatory system but also in non-mammalian systems, such as the gill lamellar vascular network with complex structural characteristics. Here, we show that (i) the contraction of pillar cells that delimit the lamellar vasculature is controlled by endothelin through the type A endothelin receptor (ETA) linked to the intracellular calcium signaling system and (ii) ETA receptor is also highly expressed on fugu erythrocytes, a hitherto unexpected finding. Database mining revealed the presence of five endothelin receptor (ETR) sequences in the fugu genome. By Northern blotting, cDNA cloning, and fura-2 monitoring, the branchial ETR subtype was shown to be ETA able to induce a Ca2+ transit. Immunohistochemistry revealed its pillar cell and erythrocyte localization. These results suggest an endothelin/ETA-mediated coordinated regulation of the pillar cell shape and erythrocyte membrane flexibility. [Copyright &y& Elsevier]

The Embodiment Lab, rooted in critical human geography, is grounded in embodiment, belonging, mentorship, care, and temporal dynamics to challenge norms in the neoliberal university. We argue that the Lab serves as a counter-practice within the academy by prioritizing our individual and collective well-being over productivity metrics. Weekly practices cultivate radical vulnerability, creating a foundation for a caring environment. Delving into multifaceted spatial dimensions our experiences suggest that the Lab becomes a living example of a feminist ethic of care. Belonging emerges as an antidote to the exclusions ingrained in academic spaces. The Lab empowers its scholars to challenge uneven power dynamics, fostering inclusion where diverse voices are heard. The Lab's emphasis on collective action and intentional processes of growth contrasts with a conventional fast, metric-driven tempo. In this paper, we offer a model to center care in lab spaces by reflecting on our own experiences in a space that values scholars as whole individuals rather than vessels of productivity. We illustrate the reflexive character of the Lab, acknowledging its adaptability and dynamism over time. Rejecting the neoliberal norms that too often dictate research spaces, the Lab exemplifies the messy and ongoing process of creating care-full academic spaces. [ABSTRACT FROM AUTHOR]

Non-thermal atmospheric pressure plasma jet (NTAPPJ) was shown as a promising tool for sterilization, treatment of chronic wounds & tumors. A portable and facile NTAPPJ system will broaden the application of plasma for many other biomedical applications. This paper demonstrates the feasibility of a customized dimmer controllable atmospheric pressure plasma jet (APPJ) in treating HeLa cells. A plasma torch was energized at 8 kVpp and 35 kHz from a neon transformer and discharged with argon gas-producing plasma plume of various gas species. The output intensity of the plasma discharge was adjusted by the voltage input from the dimmer controller circuit. Between 60 - 220 V, a different power output of the dimmer can be varied between 8.4 to 83.6 W, to generate different intensities of non-thermal APPJ (NTAPPJ). The NTAPPJ was then applied to HeLa cells for 20 sec & it was found that cells reduced their adhesion at a dimmer power < 20 W but stayed viable. This progressed to observations of damaged cell body when the power was increased beyond 20 W & the cell membranes shattered when the power was up to 83.6 W. This was due to the ultraviolet rays and reactive oxygen species (ROS), that induced detrimental effects to the cells. The controllable NTAPPJ system, based on dimmer control, induced different levels of effects on the biophysical structures of cells, ranging from reduced cell spread to apoptosis and eventually cell membrane disintegration. In this study, biodegradable and biocompatible polylactic acid (PLA) and poly(3-hyroxybutyrate-co-3-hydroxyvalerate) (PHBV) blend polymer solutions were electrospun using electrospinning technique. Polymer blend ratio and electrospinning voltage were varied and optimized. The fabricated electrospun PLA/PHBV membranes were dipped into poly(3,4-ethylenedioxythiophene)-poly(styrenesulfonate) (PEDOT:PSS) solution to produce conductive PEDOT:PSS coated membranes. The coated and uncoated membranes were investigated using scanning electron microscopy (SEM), field emission scanning electron microscopy (FESEM), energy dispersive X-ray (EDX) and water uptake measurement. PEDOT:PSS coated membranes were then loaded with drug (tetracycline hydrochloride) and their antibacterial activity was investigated. Beadless PLA/PHBV membranes were successfully fabricated. The fabricated PLA/PHBV membranes were rendered conductive by dipping them into poly(3,4-ethylenedioxythiophene)-poly(styrenesulfonate) (PEDOT:PSS) solution. Well-characterized and scalable downstream processes for the purification of biologics are extremely demanding for delivering quality therapeutics to patients at a reasonable price. Erythropoietin (EPO) is a blockbuster biologic with diverse clinical applications, but its application is limited to financially well-off societies due to its high price. The high price of EPO is associated with the technical difficulties related to the purification challenge to obtain qualified products with a cost-effective defined process. Though there are reports for the purification of EPO there is no report of a well-characterized downstream process with critical process parameters (CPPs) that can deliver EPO consistently satisfying the quality target product profile (QTPP), which is a critical regulatory requirement. To advance the field, we applied the quality by design (QbD) principle and design of experiment (DoE) protocol to establish an effective process, which is scalable up to 100× batch size satisfying QTPP. We have successfully transformed the process from static mode to dynamic mode and validated it. Insignificant variation (p > 0.05) within and between 1×, 10×, and 100× batches showed that the process is reproducible and seamlessly scalable. The biochemical analysis along with the biofunctionality data ensures that the products from different scale batches were indifferent and comparable to a reference product. Our study thereby established a robust and scalable downstream process of EPO biosimilar satisfying QTPP. The technological scheme presented here can speed up the production of not only EPO but also many other life-saving biologics and make them available to the mass population at a reduced cost. A field experiment was conducted at Bangladesh Agricultural University farm, Mymensingh, Bangladesh, during the Boro season, 2017 with six different K fertilizer rates: 0, 10, 20, 30, 40 and 50 kg ha-1 to determine the optimum rate of potassium (K) fertilization for the improved yield of a specific rice variety in the irrigated ecosystem under floodplain area. Compared with no K fertilizer, adding K increased the rice grain and straw yields significantly, with all other yield contributing components, except 1000-grain weight. The highest yield of grain (7.07 ton ha-1) and straw (8.48 ton ha-1) were recorded in recommended fertilizer dose (RFD) of NPS + 50kg K treatment, which were statistically identical with RFD of NPS + 40 kg K. Rice grain and straw yields due to the different treatments increased by 18.65% to 53.74% and 18.67% to 53.78%, respectively over control. K content and uptake through grain and straw were significantly influenced by applying different levels of K. These results specified that the use of 40 kg K ha-1 had better performance on the grain and straw yields. Therefore, we conclude that the application of 40 kg K ha-1 along with the RFD of NPS for BRRI dhan29 cultivation is the best option for higher yield in Old Brahmaputra Floodplain soil. In the current study, electrospinning technique was used to fabricate composite membranes by blending of a synthetic polymer, polylactic acid (PLA) and a natural polymer, poly(3-hydroxybutyrate-co-3-hydroxyvalerate), PHBV. Conductive membranes were prepared by dipping PLA/PHBV electrospun membranes into poly(3,4-ethylenedioxythiophene)–poly(styrenesulfonate) (PEDOT:PSS) solution, which is a biocompatible polymer. The coated and uncoated membranes were evaluated using several techniques. The electrical conductivity of the coated membranes was measured using a digital multimeter. In vitro cell cytotoxicity and cell viability were measured by culturing human skin fibroblast (HSF) cells onto the membranes using MTT assays. It was observed that electrospinning of 20% (w/v) PLA/PHBV with a weight ratio of 50:50 produced the most uniform fibers with no beads. It was observed that the wettability and surface roughness of the PEDOT:PSS coated PLA/PHBV membranes were greatly increased than uncoated membrane. The results of cell viability using MTT assay, cell attachment and cell proliferation showed that the conductive PEDOT:PSS coated PLA/PHBV membrane were more favorable for tissue engineering application than their uncoated counterparts. Zebrafish connexin 36.7 (cx36.7/ecx) has been identified as a key molecule in the early stages of heart development in this species. A defect in cx36.7 causes severe heart malformation due to the downregulation of nkx2.5 expression, a result which resembles congenital heart disease in humans. It has been shown that cx36.7 is expressed specifically in early developing heart cardiomyocytes. However, the regulatory mechanism for the cardiac-restricted expression of cx36.7 remains to be elucidated. In this study we isolated the 5′-flanking promoter region of the cx36.7 gene and characterized its promoter activity in zebrafish embryos. Deletion analysis showed that a 316-bp upstream region is essential for cardiac-restricted expression. This region contains four GATA elements, the proximal two of which are responsible for promoter activation in the embryonic heart and serve as binding sites for gata4. When gata4, gata5 and gata6 were simultaneously knocked down, the promoter activity was significantly decreased. Moreover, the deletion of the region between − 316 and − 133 bp led to EGFP expression in the embryonic trunk muscle. The distal two GATA and A/T-rich elements in this region act as repressors of promoter activity in skeletal muscle. These results suggest that cx36.7 expression is directed by cardiac promoter activation via the two proximal GATA elements as well as by skeletal muscle-specific promoter repression via the two distal GATA elements. Investigation of the serum level of copper, zinc, magnesium and manganese in type-2 diabetes mellitus and their possible association with lipid profile was carried out. The comparative study included 100 type-2 diabetic patients in Gr-II and 100 non-diabetic as control in (Gr-I). Results indicated that there is a significant lower level (p < .001) of serum Zn, Cu, Mg and Mn in diabetic patients compared with the control group, showing p value < .001. In type-2 DM patients (Gr-II) there were significant correlations between serum Zn and TAG (r = 0.209) and between Zn and HDL-C level (r = .199) showing p value <.05. Non significant relationships were found in between Zn and lipid profile (TAG, Cholesterol, HDL-C and LDL-C) of control (Gr-I) group. Significant correlation was found between serum magnesium and TAG of control (Gr-I) where p < .01 and non significant correlations were found in serum Mg and total cholesterol, HDL-C, LDL-C of both type-2 diabetic (Gr-II) and control (Gr-I). Significant correlations were also found in between serum of Cu and Mn and TAG of control (Gr-I) where p < .05 and non significant correlations were found in other component of lipid profile of both cases (Gr-II) and control (Gr-I). Abstract: Calcitonin (CT), a 32-amino acid peptide, was initially isolated from fish. Fish CT has higher affinity to mammalian CT receptor (CTR), and has activity on calcium homeostasis. Therefore, fish CT has been used as a drug for the treatment of human bone diseases. However, the physiological roles of CT in fish as well as the characteristics of the fish CTR have not been clarified. Here, we cloned and characterized CTR from mefugu (Takifugu obscurus). Full-length cDNA sequencing revealed that mfCTR (mf, mefugu) consists of N-terminal four tandem putative hormone-binding domains (HBDs). Database mining showed that the multiple HBD-containing CTR is a common feature for some other fishes. Detailed pharmacological studies revealed that mfCTR generated cAMP in response to (1) fish CT, (2) calcitonin gene-related peptide (CGRP) in combinations with receptor activity-modifying proteins (mfRAMPs) 1 and 4, and (3) amylin in combinations with mfRAMPs 1−5. Unlike mammalian CTR, mfCTR showed dual affinity sites. Corresponding EC50 values of those are in close proximity of the in vivo concentration of CT in fish. Analyses of the deletion mutants of mfCTR demonstrated that only the nearmost HBD to the first transmembrane region is functional to the ligands. Although, fish CT has higher affinity to the human CTR, human CT did not bind to the mfCTR. This is the first report that demonstrates the structure and property of fish receptor for CT, CGRP, and amylin. Fish CTR is the first example that has multiple HBD-like sequences. Lipid nanoparticle (LNP) technology has become extremely demanding for delivering RNA-products and other drugs. However, there is no platform to manufacture pharmaceutical-grade LNPs with desired particle size from a wide range in continuous mode. We have developed a unique platform to obtain any specific size-range of LNPs from 60 to 180 nm satisfying pharmaceutical regulatory requirements for polydispersity index, sterility, dose uniformity and bio-functionality. We applied design of experiment (DoE) methodology and identified the critical process parameters to establish the process for global application. Cross-point validation within the response map of DoE confirmed that the platform is robust to produce specific size (± 10 nm) of LNPs within the design-range. The technology is successfully transformed to production scale and validated. Products from R&D, pilot and production batches for a candidate SARS-CoV-2 mRNA-vaccine generated equivalent biological responses. This study investigated groundwater pollution and potential human health risks from arsenic, iron, and manganese in the rural area of Jashore, Bangladesh. Study results show that the mean value of groundwater pH is 7.25+0.31, with a mean conductivity of 633.94+ 327.41 µs/cm, while about 73, 97, and 91% of groundwater samples exceeded the Bangladesh drinking water standard limits for As, Fe, and Mn, respectively. Groundwater pollution evaluation indices, including the heavy metal pollution index, the heavy metal evaluation index, the degree of contamination, and the Nemerow pollution index, show that approximately 97, 82, 100, and 100% of samples are in the high degree of pollution category, respectively. Spatial distribution exhibited that the study area is highly exposed to As (73%), Fe (82%), and Mn (46%). In the case of non-carcinogenic health risk via oral exposure, about 94% of samples suggest a high category of risk for infants, and 97% of samples are found to be at high risk for children and adults. The carcinogenic risk of arsenic via an oral exposure pathway suggests that approximately 97% of the samples are found to be at high risk for infants, and all of the samples are at high risk for both adults and children. Non-thermal atmospheric plasma jet (NTAPPJ) is believed to be effective in treating monolayer cells cultured in-vitro due to the oxidative stresses induced. However, it is controversial whether studying the impact of plasma to cells in monolayer can be representative to a model close to the in-vivo tissues. This study revealed and compared the effects of NTAPPJ treatment directly to both monolayer of HeLa cells and microspheroids in the culture media, at different exposure time from 15 to 60 seconds at a fixed power of 80 W. Optical emission spectroscopy (OES) revealed that the plasma applied to the cells is rich in hydroxyl, nitrogen, oxygen and argon species. The experimental results revealed that the treatment time from 15 seconds onwards was detrimental to the monolayer of cells, but not to the spheroids compared with the negative controls. After long NTAPPJ exposure, the HeLa cells in the spheroids were found viable, regardless of the plasma exposure time applied. We aimed to assess how suicidality has been depicted in Bangla movies and dramas. We conducted a search on YouTube by using search terms to identify movies and dramas with suicidal scripts. The search was performed between February and May 2022 resulting in 71 items consisting of 35 Bangla movies and 36 Bangla dramas. We scrutinized the contents of movies and dramas against our pre-designed instrument and we assessed their quality against World Health Organization guidelines. Among the 71 suicidal behaviors, 46.5% were suicides, 72% of the suicidal behavior was noted in young adults, 63.9% were unmarried, and 69% attempts were found in prominent characters. Hanging was found as the most prominent method (25.4%) and premarital and extramarital affairs and sexual harassment were the most prominent risk factors (60.6%). • Generation of HeLa spheroids in Ca-alginate-PEG microbeads using the flicking technique. • An analytical model of the flicking-microencapsulation process was developed. • The Ca-alginate-PEG enhanced the biophysical properties of microbeads. • The microbead of Ca-alginate-PEG prolonged the 3D cell culture and reduced the cell leaking problem. In a conventional three-dimensional (3D) cell culture system based on microencapsulation, the calcium alginate microcapsules tend to rupture within 7 days of culture, causing unwanted cell leakage. The microencapsulation based on flicking model of calcium-alginate-polyethylene glycol (Ca-alg-PEG) was proposed to rectify this problem. An analytical model to simulate the flicking process based on the deflection of a needle cantilever was also successfully developed and used to predict the size of the microbeads. The size of the microbeads were ranged from 300 to 500 μm and it is controlled by varying the liquid flow rate from 4.8 to 366 μL/min and flicking speed from 70 to 120 rpm. Under a flicking force of 0.58 N, uniform sized and spherical shaped Ca-alginate-PEG microbeads were produced at a liquid flow rate of 40 μL/min and a flicking rate of 100 rpm. The microbeads were characterized by Field Emission Scanning Microscopy, Atomic Force Microscopy, Raman Spectroscopy and Nanoindentation, and the results indicated improved bio-physical properties of the ca-alginate microbeads after added with PEG. The cell viability test demonstrated that Ca-alginate-PEG microbeads were able to support the growth of viable HeLa cells into spheroids. Resilient calcium-alginate-polyethylene glycol (Ca-alg-PEG) microbeads were found to be able to last up to 15 days before rupturing and greatly reduced the cell leakage problem. [ABSTRACT FROM AUTHOR]
