Objective: To compare the characteristics and short-term outcomes in extremely preterm infants, who developed necrotizing enterocolitis (NEC) following a packed red blood cell transfusion (pRBC) within 48 h (TANEC), with those who developed NEC beyond 48 h (non-TANEC). Setting: A single-center retrospective cohort study in a Tertiary neonatal intensive care unit in the UK over a 5-year period Patients and methods: Extremely premature infants (23–27 weeks gestation) were selected. TANEC and non-TANEC incidence were calculated from the confirmed NEC group (defined as modified Bell's stage II and beyond). The characteristics and short-term outcomes of infants with TANEC in the first 8 weeks of life were compared to infants with non-TANEC. Results and interpretation: Incidence of confirmed NEC was 14% (28/207). On further subgroup analysis of the confirmed NEC cases, 46% (13/28) of infants were identified with TANEC and 54% (15/28) with non-TANEC. The incidence of TANEC did not correlate with the number of antecedent pRBC transfusions or the pre-transfusion median hemoglobin (Hb) levels. There were no significant differences in characteristics between the TANEC and non-TANEC groups. Infants within the TANEC group required more intensive neonatal care support, greater surgical intervention (p-value 0.043) with loss of gut integrity and an increase in number of TPN dependency days (p-value 0.014). Multiple endocrine neoplasia type 1 (MEN1), caused by mutations in the MEN1 gene encoding menin, is an autosomal dominant disorder characterised by the combined occurrence of parathyroid, pituitary and pancreatic neuroendocrine tumours (NETs). Development of these tumours is associated with wide variations in their severity, order and ages (from <5 to >80 years), requiring life-long scre ening. To improve tumour surveillance and quality of life, better circulating biomarkers, particularly for pancreatic NETs that are associated with higher mortality, are required. We, therefore, examined the expression of circulating miRNA in the serum of MEN1 patien ts. Initial profiling analysis followed by qRT-PCR validation studies identified miR-3156-5p to be significantly downregulated (-1.3 to 5.8-fold, P < 0.05-0.0005) in nine MEN1 patients, compared to matched unaffected relatives. MEN1 knock-down experiments in BON-1 human pancreatic NET cells resulted in reduced MEN1 (49%, P < 0.05), menin (54%, P < 0.05) and miR-3156-5p expression (20%, P < 0.005), compared to control-treated cells, suggesting that miR-3156-5p downregulation is a consequence of loss of MEN1 expression. In silico analysis identified mortality factor 4-like 2 (MOR4FL2) as a potential target of miR-3156-5p, and in vitro functional studies in BON-1 cells transfected with either miR-3156-5p mimic or inhibitors showed that the miR-3156-5p mimic significantly reduced MORF4L2 protein expression (46%, P < 0.005), while miR-3156-5p inhibitor significantly increased MORF4L2 expression (1.5-fold, P < 0.05), compared to control-treated cells, thereby confirming that miR-3156-5p regulates MORF4L2 expression. Thus, the inverse relationship between miR-3156-5p and MORF4L2 expression represents a potential serum biomarker that could facilitate the detection of NET occurrence in MEN1 patients. Natural regeneration can reduce costs compared with replanting. However, its use requires knowledge about how either active or passive management will affect the balance between quality and quantity in the timber supply. This study aimed to quantify the effects of respacing on volume recovery and wood properties. Two British forest experiments using Picea sitchensis with various respacing distances and an un-respaced control were assessed 21–22 years after the treatments were applied. Tree dimensions were measured and used to quantify slenderness, merchantable volume, and sawlog volume. Wood properties were assessed on a sub-sample using mechanical testing. Generalised linear mixed models were used to examine differences between treatments and sites. Respacing decreased slenderness and increased relative sawlog volume and branch size. Wider respacing reduced wood strength and the widest respacing reduced wood stiffness. Respacing did not affect wood density. However, at the relatively low productivity sites considered here, respacing to 2.1 m represented the best compromise for current markets. In summary, not respacing improved some wood properties but reduced tree stability and the proportion and volume of sawlogs, which will negatively affect forest value. [ABSTRACT FROM AUTHOR]

Pacific silver fir ( Abies amabilis Douglas ex J. Forbes) was first introduced to Britain in 1830 but has not been widely planted and occupies a minute part of the forest estate. The results of six experiments established in the uplands of Britain examining material from 30 collection sites in 14 seed zones clearly demonstrate that its potential has not been recognised. The trials were assessed after 28 years and show that Pacific silver fir has the potential to be as productive as other common species options. There was little variation in performance between the 14 seed zones, and future seed collections could be carried out within a wide geographical range, including mainland British Columbia and Vancouver Island and the Olympic Mountains and western Cascades of Washington. The silvicultural characteristics of the species mean that it could be used more widely to diversify forests in Britain both as a plantation species and in the wider use of continuous cover management. More work is justified to determine its susceptibility to Annosum root rot ( Heterobasidion annosum (Fr.) Bref), confirm its productivity on sites with rainfall below 800 mm·a-1 and (or) with a high peat content, and provide more detail on its wood properties. [ABSTRACT FROM AUTHOR]

Adiposity is associated with reduced fertility in men. The aetiology is multifactorial, with obese men at greater risk of suffering from impaired spermatogenesis, reduced circulating testosterone levels, erectile dysfunction and poor libido. The diagnosis and treatment of reduced fertility observed in obese men therefore requires insight into the underlying pathology, which has hormonal, mechanical and psychosocial aspects. This article summarises the current epidemiological, experimental and clinical trial evidence from the perspective of a practicing clinician. The following conclusions and recommendations can be drawn: Obesity is associated with low serum testosterone concentrations, but treatment with exogenous testosterone is likely to adversely impact on fertility. It is important to discuss this with men prior to initiation of testosterone therapy. Obesity adversely affects sperm concentration and may affect sperm quality. However, whether or not weight loss will correct these factors remain to be established. Oestrogen receptor modulators (and aromatase inhibitors) are unlicensed in the treatment for male hypogonadism and/or infertility. European silver fir ( Abies alba Mill.) was the first true fir to be introduced to Britain in 1603. Despite early enthusiasm for the species, it has not been widely planted and work on the effects of silver fir woolly aphid ( Adelges nordmannianae Eckstein ) in the mid-twentieth century concluded that large scale use of the species could not be justified. This was interpreted as a research challenge and resulted in the establishment of a series of provenance experiments being established between 1966 and 1970. Results from these experiments after 46 years clearly show that European silver fir is a productive species and could be deployed in British forests as a component to diversify species composition and in the wider use of continuous cover forestry. In general, the trials show that good provenances survive and grow on a wide range of sites and it is not necessary to select different provenances depending on site location or climate. Provenances from Calabria in Italy performed very well and should be the first choice of origin for future planting in Britain. However, provenances from a wide range of the species natural distribution performed well demonstrating that European silver fir is a very adaptable species. [ABSTRACT FROM AUTHOR]

Stands of Sitka spruce ( Picea sitchensis (Bong.) Carr.) originating from natural regeneration can be extremely dense with high mortality, poor growth form and low volume production. Although costly, respacing (pre-commercial thinning) can reduce resource competition for the remaining trees, altering the final crop produced. Three experiments were established to examine the effect of respacing 1-2 m tall trees to different target densities. The impact on stand growth and yield was measured 11-17 years after respacing, and the longer-term impact was modelled to year 50. Unrespaced control treatments self-thinned at a similar and constant rate at two sites. At the third, extremely high initial density resulted in a higher rate of self-thinning and lower density after 11 years. Wider respacing treatments resulted in larger mean tree diameter, but there was no significant effect of respacing on stand volume 11-17 years after respacing; greater diameter growth did not compensate for low tree number. Results indicate a volume penalty associated with delaying respacing until trees were 4 m tall, but this treatment was unreplicated. Modelled stand volume in year 50 was higher for the 2.6 m × 2.6 m respacing treatment than for the 1.8 m × 1.8 m, 2.1 m × 2.1 m or 3.3 m × 3.3 m treatments. However, open-grown conditions may reduce timber quality compared to closer spacing treatments. The results are presented using a stand density management diagram for Sitka spruce growing in Canada and support recent suggestions that British stands have a shallower size-density relationship than Canadian stands. Abstract: A new method of constructing light, flexible and more realistic replica leaves for continuous determination of leaf boundary layer conductance to heat transfer () was developed and tested in a mature oak (Quercus robur L.) and sycamore (Acer pseudoplatanus L.) tree canopy. The replicas were used to determine the difference between oak and sycamore leaf in exposed sites in the upper canopy, the relationship of with wind speed, the seasonal changes in in the canopy as leaf cover developed, and values for Ω, the decoupling coefficient. The replicas showed similar gradients in temperature at their margins to those in real leaves. When exposed, the of the larger sycamore leaves was 66% of that of oak leaves under the same conditions. Linear relationships were found with and wind speed across the measured range of 0.3–3.5ms−1, and flow in the replica boundary layers was laminar in all conditions. The leafless canopy produced a substantial sheltering effect, reducing by 12–28% in light winds. Sycamore replicas in the leafed canopy showed a 19–29% lower at a given external wind speed than when outside, but there was little difference between ‘sun’ and ‘shade’ position shoots, because of the density of the shoots, and the close branching pattern. In contrast, in oak at a given wind speed was 15–21% lower for ‘sun’ leaves than that for replicas outside the canopy, with a larger reduction (approximately 28%) in denser ‘shade’ sites. Although wind speed in the canopy was often low, leaves of both species were usually well coupled to the canopy airstream (Ω <0.3). Sun leaves were substantially less well coupled than shade leaves, despite the lower shelter effect, because of their higher stomatal conductance values. In the lightest winds (<0.5ms−1) and with high stomatal conductance, coupling may on many occasions be poor for sun leaves, particularly for the larger sycamore leaves. [Copyright &y& Elsevier]

Aspen (Populus tremula L.) is a keystone species and a model system for forest tree genomics. We present an updated resource comprising a chromosome‐scale assembly, population genetics and genomics data. Using the resource, we explore the genetic basis of natural variation in leaf size and shape, traits with complex genetic architecture. We generated the genome assembly using long‐read sequencing, optical and high‐density genetic maps. We conducted whole‐genome resequencing of the Umeå Aspen (UmAsp) collection. Using the assembly and re‐sequencing data from the UmAsp, Swedish Aspen (SwAsp) and Scottish Aspen (ScotAsp) collections we performed genome‐wide association analyses (GWAS) using Single Nucleotide Polymorphisms (SNPs) for 26 leaf physiognomy phenotypes. We conducted Assay of Transposase Accessible Chromatin sequencing (ATAC‐Seq), identified genomic regions of accessible chromatin, and subset SNPs to these regions, improving the GWAS detection rate. We identified candidate long non‐coding RNAs in leaf samples, quantified their expression in an updated co‐expression network, and used this to explore the functions of candidate genes identified from the GWAS. A GWAS found SNP associations for seven traits. The associated SNPs were in or near genes annotated with developmental functions, which represent candidates for further study. Of particular interest was a ~177‐kbp region harbouring associations with several leaf phenotypes in ScotAsp. We have incorporated the assembly, population genetics, genomics, and GWAS data into the PlantGenIE.org web resource, including updating existing genomics data to the new genome version, to enable easy exploration and visualisation. Objective: Primary hyperparathyroidism is a common endocrine disorder, with 80% of all cases usually caused by one single hyperfunctioning parathyroid adenoma. Conventional imaging modalities for the diagnostic work‐up of primary hyperparathyroidism (PHPT) include ultrasound of the neck, 99mTc‐sestamibi scintigraphy, and four‐dimensional computed tomography (4D‐CT). However, the role of other imaging modalities, such as 11C‐methionine PET/CT, in the care pathway for PHPT is currently unclear. Here, we report our experience of the diagnostic utility of 11C‐methionine PET/CT in a single‐center patient cohort (n = 45). Design: Retrospective single‐center cohort study. Patients and Measurements: The data of eligible patients that underwent 11C‐methionine PET/CT between 2014 and 2022 at Addenbrooke's Hospital (Cambridge, UK) were collected and analyzed. The clinical utility of imaging modalities was determined by comparing the imaging result with histopathological and biochemical outcomes following surgery. Results: In patients with persistent primary hyperparathyroidism following previous surgery, 11C‐methionine PET/CT identified a candidate lesion in 6 of 10 patients (60.0%), and histologically confirmed in 5 (50.0%). 11C‐methionine PET/CT also correctly identified a parathyroid adenoma in 9 out of 12 patients (75.0%) that failed to be localized on other imaging modalities. 11C‐methionine PET/CT had a sensitivity of 70.0% (95% CI 55.8 – 84.2%) for the detection of parathyroid adenomas. Conclusions: This study highlights a diagnostic role for 11C‐methionine PET/CT in patients that have undergone unsuccessful prior surgery or have equivocal or negative prior imaging results, aiding localization and a targeted surgical approach. Prenatal ethanol exposure increases collagen deposition and alters surfactant protein (SP) expression and immune status in lungs of near-term fetal sheep. Our objectives were to determine 1) whether these prenatal effects of repeated gestational ethanol exposure persist after birth and 2) whether surfactant phospholipid composition is altered following prenatal ethanol exposure. Pregnant ewes were chronically catheterized at 90 days of gestational age (DGA) and given a 1-h daily infusion of ethanol (0.75 g/kg, n = 9) or saline (n = 7) from 95 to 135 DGA; ethanol administration ceased after 135 DGA. Lambs were born naturally at full term (146 ± 0.5 DGA). Lung tissue was examined at 9 wk postnatal age for alterations in structure, SP expression, and inflammation; bronchoalveolar lavage fluid was examined for alterations in surfactant phospholipid composition. At 134 DGA, surfactant phospholipid concentration in amniotic fluid was significantly reduced (P < 0.05) by ethanol exposure, and the composition was altered. In postnatal lambs, there were no significant differences between treatment groups in birth weight, postnatal growth, blood gas parameters, and lung weight, volume, tissue fraction, mean linear intercept, collagen content, proinflammatory cytokine gene expression, and bronchoalveolar lavage fluid surfactant phospholipid composition. Although SP-A, SP-B, and SP-C mRNA levels were not significantly different between treatment groups, SP-D mRNA levels were significantly greater (P < 0.05) in ethanol-treated animals; as SP-D has immunomodulatory roles, innate immunity may be altered. The adverse effects of daily ethanol exposure during late gestation on the fetal lung do not persist to 2 mo after birth, indicating that the developing lung is capable of repair. Our aim was to characterize an ovine model of preterm birth that allows analysis of the developmental effects of preterm birth in the absence of postnatal confounding factors. Preterm birth was induced at 131 days of gestation in 82 lambs; controls (n = 31) were born at term (145 days). Overall survival of preterm lambs was 60%; males had significantly lower survival than females (44% vs 76%); 94% of term lambs survived. Although the birth weight of preterm lambs was ∼0.9 kg lower than in term lambs, the crown-to-rump and forelimb lengths were similar. At 9 weeks after term-equivalent age, there were no differences in body weight or dimensions between preterm and term lambs; when adjusted for body weight, the heart was 21% heavier in preterm than term lambs. We conclude that moderately preterm birth in sheep is characterized by a greater survival of female lambs than males and has significant effects on organ development. [ABSTRACT FROM AUTHOR]

Non‐native plant pests and pathogens are increasing exponentially, causing extirpation of foundation species. The impact of large‐scale declines in a single host on associated biodiversity is widely documented. However, the impact of multiple host loss on biodiversity and whether these impacts are multiplicative has not been assessed. Ecological theory suggests that systems with greater functional redundancy (alternative hosts) will be more resilient to the loss of sympatric hosts. We test this theory and show its importance in relation to pest/pathogen impact assessments.We assessed the potential impact on biodiversity of the loss of two widely occurring sympatric European tree species, Fraxinus excelsior and Quercus petraea/robur, both of which are currently threatened by a range of pests and pathogens.At the UK scale, the total number of associated species at risk of extirpation from plant diseases affecting these two sympatric hosts is greater than the sum of the associated species at risk from declines in either host alone. F. excelsior hosts 45 obligate species (species only found on that host) and Q. petraea/robur hosts 326. However, a decline in both these trees would impact 512 associated species, across multiple taxon groups, a 38% increase. Assessments at a local scale, 24 mixed F. excelsior–Q. petraea/robur woodlands revealed that these impacts may be even greater due to a lack of functional redundancy. Only 21% of sites were able to provide functional redundancy for F. excelsior and Q. petraea/robur associated species which can use other tree species. In most woodlands, the tree species required to provide functional redundancy were not present, although the site conditions were often suitable for them to grow.Synthesis. Understanding of functional redundancy should be applied to assessments of pests/pathogens impact on biodiversity. In risk assessments, higher impact scores should be given to pests/pathogens affecting hosts occurring with other host plant species already impacted by pests/pathogens. Current pest/pathogen risk assessment approaches that ignore the cumulative, cascading effects shown in this study may allow an insidious, mostly overlooked, driver of biodiversity loss to continue. Associational resistance theory predicts that insect herbivory decreases with increasing tree diversity in forest ecosystems. However, the generality of this effect and its underlying mechanisms are still debated, particularly since evidence has accumulated that climate may influence the direction and strength of the relationship between diversity and herbivory.We quantified insect leaf herbivory and leaf chemical defences (phenolic compounds) of silver birch Betula pendula in pure and mixed plots with different tree species composition across 12 tree diversity experiments in different climates. We investigated whether the effects of neighbouring tree species diversity on insect herbivory in birch, that is, associational effects, were dependent on the climatic context, and whether neighbour‐induced changes in birch chemical defences were involved in associational resistance to insect herbivory.We showed that herbivory on birch decreased with tree species richness (i.e. associational resistance) in colder environments but that this relationship faded as mean annual temperature increased.Birch leaf chemical defences increased with tree species richness but decreased with the phylogenetic distinctiveness of birch from its neighbours, particularly in warmer and more humid environments.Herbivory was negatively correlated with leaf chemical defences, particularly when birch was associated with closely related species. The interactive effect of tree diversity and climate on herbivory was partially mediated by changes in leaf chemical defences.Our findings confirm that tree species diversity can modify the leaf chemistry of a focal species, hence its quality for herbivores. They further stress that such neighbour‐induced changes are dependent on climate and that tree diversity effects on insect herbivory are partially mediated by these neighbour‐induced changes in chemical defences. High levels of ethanol (EtOH) consumption during pregnancy adversely affect fetal development; however, the effects of lower levels of exposure are less clear. Our objectives were to assess the effects of daily EtOH exposure (3.8 USA standard drinks) on fetal-maternal physiological variables and the fetal brain, particularly white matter. Pregnant ewes received daily intravenous infusions of EtOH (0.75 g/kg maternal body wt over 1 h, 8 fetuses) or saline (8 fetuses) from 95 to 133 days of gestational age (DGA; term ∼145 DGA). Maternal and fetal arterial blood was sampled at 131-133 DGA. At necropsy (134 DGA) fetal brains were collected for analysis. Maternal and fetal plasma EtOH concentrations reached similar maximal concentration (∼0.11 g/dl) and declined at the same rate. EtOH infusions produced mild reductions in fetal arterial oxygenation but there were no changes in maternal oxygenation, maternal and fetal PaCO2, or in fetal mean arterial pressure or heart rate. Following EtOH infusions, plasma lactate levels were elevated in ewes and fetuses, but arterial pH fell only in ewes. Fetal body and brain weights were similar between groups. In three of eight EtOH-exposed fetuses there were small subarachnoid hemorrhages in the cerebrum and cerebellum associated with focal cortical neuronal death and gliosis. Overall, there was no evidence of cystic lesions, inflammation, increased apoptosis, or white matter injury. We conclude that daily EtOH exposure during the third trimester-equivalent of ovine pregnancy has modest physiological effects on the fetus and no gross effects on fetal white matter development.
