Creativity, driving environmentally sustainable fashion design through innovation, has become increasingly more important. From an educational perspective, enhancing students' creative design thinking processes, presents several challenges, especially in blended learning. This article addresses this concern and reports on the implementation of a blended learning teaching framework, applied to support students' positive emotions and creativity during an environmentally sustainable fashion design project that required innovative design solutions. The qualitative inquiry adopted a case study research design, involving undergraduate fashion design students at a tertiary institution in South Africa. Two key findings have emerged from the research. Firstly, the significant level of creativity required by the undertaken design project requires re-thinking of support for creative and innovative thinking. Secondly, a clearer understanding of the anxiousness caused when participants are faced with innovative design practices is needed to find suitable solutions. Cardiac fibrosis contributes to the development of heart failure, and is the response of cardiac fibroblasts (CFs) to pressure or volume overload. Limiting factors in CFs research are the poor availability of human cells and the tendency of CFs to transdifferentiate into myofibroblasts when cultured in vitro. The possibility to generate CFs from induced pluripotent stem cells (iPSC), providing a nearly unlimited cell source, opens new possibilities. However, the behaviour of iPSC-CFs under mechanical stimulation has not been studied yet. Our study aimed to assess the behaviour of iPSC-CFs under mechanical stretch and pro-fibrotic conditions. First, we confirm that iPSC-CFs are comparable to primary CFs at gene, protein and functional level. Furthermore, iPSC-derived CFs adopt a pro-fibrotic response to transforming growth factor beta (TGF-β). In addition, mechanical stretch inhibits TGF-β-induced fibroblast activation in iPSC-CFs. Thus, the responsiveness to cytokines and mechanical stimulation of iPSC-CFs demonstrates they possess key characteristics of primary CFs and may be useful for disease modelling. This paper is devoted to the study of active and stable nickel catalysts for methane dry reforming based on Pr-doped ceria–zirconia obtained via the solvothermal continuous method. Studies on the physicochemical and catalytic properties of the 5%Ni/Ce0.75Zr0.25−xPrxO2 series have showed that Pr introduction leads to an increase in the amount of highly reactive oxygen in the oxide lattice. Praseodymium-based catalysts showed significantly higher reactant conversions. In addition to the nature of support, the method of nickel introduction was also studied; Ni was added both using impregnation and the one-pot procedure with mixed oxide preparation. The method of Ni addition was shown to have significant effect on the morphology of its particles and Ni-support interaction, and, respectively, on catalytic activity and coking stability. The 5%Ni/Ce0.75Zr0.15Pr0.1O2 catalyst prepared by one-pot method showed stable operation in the MDR reaction for 30 h at CO2 and CH4 conversions of ~40% and an H2 yield of ~18% (T = 700 °C, τ = 10 ms). A series of 5%Ni/Ce1-xTixO2 catalysts was prepared with nickel impregnation of mixed Ce–Ti oxides obtained via synthesis in supercritical isopropanol. All oxides have a cubic fluorite phase structure. Ti is incorporated into the fluorite structure. Small amounts of impurities of TiO2 or mixed Ce–Ti oxides appear with Ti introduction. Supported Ni is presented as the NiO or NiTiO3 perovskite phase. Ti introduction increases total samples reducibility and results in stronger interaction of supported Ni with the oxide support. The fraction of rapidly replaced oxygen and the average tracer diffusion coefficient also increase. The number of metallic nickel sites decreased with increasing Ti content. All catalysts except Ni-CeTi0.45 demonstrate close activity in tests of dry reforming of methane. The lower activity of Ni-CeTi0.45 can be connected to Ni decoration with species of the oxide support. The incorporation of Ti prevents detachment of Ni particles from the surface and their sintering during dry reforming of methane. [ABSTRACT FROM AUTHOR]

We have been observing large fluctuations and price increases in electricity markets in recent years. The COVID-19 pandemic, rising energy costs, political instability and increasing demand for electricity have been the factors intensifying the problems. This causes uncertainty related to maintaining energy security. Energy security is an element of the national security system. In this context, the question arises whether Polish energy companies are able to adapt to the growing demand for electricity while meeting the growing environmental requirements. Moreover, it remains to be seen how the current energy crisis will affect the financial condition of energy companies in Poland and whether companies from the energy sector will benefit from this crisis. Another issue is the impact of the current crisis on the sense of energy security of consumers. There are many factors affecting energy security. This study focuses on economic and financial factors. The article aims to assess the energy security of consumers from the perspective of the stability of energy prices and the financial condition of companies from the energy industry in Poland in the context of the global energy crisis. Brucellosis is a critical zoonotic disease impacting humans and animals globally, causing symptoms like fever and arthritis in humans and reproductive issues in animals. The disease stems from the Brucella genus, adept at evading the immune system and proliferating within host cells. This study explores how Brucella abortus manipulates host cellular mechanisms to sustain infection, focusing on the interaction with murine macrophages over 24 h. Initial host defenses involve innate immune responses, while Brucella 's survival strategies include evading lysosomal degradation and modulating host cell functions through various pathways. The research identified significant transcriptional changes in macrophages post-infection, highlighting pathways such as cytokine storm, pyroptosis signaling, Toll-like receptor pathways, and LXRs/RXRs signaling. The findings shed light on Brucella's complex mechanisms to undermine host defenses and underscore the need for further investigation into therapeutic targets to combat brucellosis. Two series of Ni/Ce(Ti/Nb)ZrO2 catalysts were prepared using citrate route and original solvothermal continuous flow synthesis in supercritical isopropanol and studied in dry reforming of methane (DRM). TEM, XPS and FTIRS of adsorbed CO confirm influence of support composition and preparation method on the catalysts' morphology and surface features. The oxygen mobility was studied by isotope heteroexchange with C18O2. After testing in DRM, carbon deposits after catalysts' testing in DRM were investigated by temperature-programmed oxidation with thermo-gravimetric analysis. The lowest amounts of carbon deposits were obtained for unmodified Ni-CeZr and Ni-CeNbZr compositions. Ti addition lead to an increased amount of carbon, which was removed at higher temperatures. The use of supercritical supports also resulted in the formation of a higher amount of coke. Catalysts prepared by the supercritical synthesis were tested in DRM for 25 h. Experiments were undertaken to identify the nature of a previously identified inhibitor of PAF-induced platelet aggregation (PA) in human saliva. Human saliva fractionated by preparative thin layer chromatography (TLC) yielded a fraction that co-migrated with fatty acids (FAs) and inhibited PAF-induced aggregation of platelets. Synthetic FAs tested for their capacities to inhibit 0.1 nM PAF-induced PA showed that only the cis-unsaturated compounds were inhibitory with activities of some of the polyunsaturated FAs (PUFA) reaching almost 100% at 20 μM. Eicosapentanoic acid (EPA) and 8,11,14-eicosatrienoic acid also deaggregated the PAF-induced aggregates. With the exception of oleic acid (OLA), cis-monounsaturated FAs, and elaidic acid, the trans isomer of OLA, were poor inhibitors. In a direct comparison with other platelet agonists, ADP, thrombin, and ionophore A23187, the active saliva fraction and selected individual FAs inhibited, to greater or lesser extent, PA induced by each of the agonists. EPA, OLA, linoleic acid (LNA), and the active saliva fraction were potent inhibitors of ADP-induced PA, EPA completely inhibited thrombin-induced PA and the saliva fraction showed only weak – moderate inhibitory activity to both thrombin- and ionophore A23187-induced PA. Other reports of endogenous PAF inhibitors in mammalian tissues are compared to the present results. In this work complex metal-oxide catalysts with the general formula 5 wt% Ni/Ce 0·75 Zr 0.25-x (Nb,Ti) x O 2-δ were synthesized by the solvothermal method using supercritical alcohols followed by nickel deposition. The catalysts were characterized and studied in ethanol dry reforming reaction (EDR) in the temperature range 600–750 °C. XRD and TEM showed that the synthesis method provides incorporation of doping cations into the ceria-zirconia fluorite structure, leading to mixed oxides formation. The effect of doping cations on structural and surface properties of 5 wt% Ni/Ce 0·75 Zr 0.25-x (Nb,Ti) x O 2-δ and activity in the EDR reaction was investigated. Oxygen deficiency δ increases with the introduction of titanium and niobium cations, which contributes to the bifunctional reforming mechanism implementation with rapid oxidation of coke precursors and activation of СО 2 at surface oxygen vacancies. TPO-O 2 analysis after reaction showed no carbon formation above 700 °C, and a few carbon deposits (not exceeding 4%) even after significant catalyst deactivation at 600 °C. The Ideation Tool (IT) emerged from a doctoral project, as a pedagogical strategy to assist students navigate and frame wicked design problems and design process activities. Subsequently, the IT was renamed as the Generative Tool (GT). Guided by action research, two teaching and learning iteration cycles with fashion design students at a South African higher education institution aimed to evaluate and refine the tool. The first iteration (Phase 1) focusses on two fourth-year fashion design students. For the second iteration (Phase 2), a refined GT was used by third-year fashion design students as a guide for the design process and a tool for reflection. This research explores the two independent student groups' experiences regarding their application of the tool, and propose strategies for refinement. Motivation Biological studies of dynamic processes in living cells often require accurate particle tracking as a first step toward quantitative analysis. Although many particle tracking methods have been developed for this purpose, they are typically based on prior assumptions about the particle dynamics, and/or they involve careful tuning of various algorithm parameters by the user for each application. This may make existing methods difficult to apply by non-expert users and to a broader range of tracking problems. Recent advances in deep-learning techniques hold great promise in eliminating these disadvantages, as they can learn how to optimally track particles from example data. Results Here, we present a deep-learning-based method for the data association stage of particle tracking. The proposed method uses convolutional neural networks and long short-term memory networks to extract relevant dynamics features and predict the motion of a particle and the cost of linking detected particles from one time point to the next. Comprehensive evaluations on datasets from the particle tracking challenge demonstrate the competitiveness of the proposed deep-learning method compared to the state of the art. Additional tests on real-time-lapse fluorescence microscopy images of various types of intracellular particles show the method performs comparably with human experts. X-ray computed tomography is a nondestructive technique often used in geosciences to obtain a three-dimensional representation of the pore space geometry that will be used for running fluid dynamic simulations. However, rocks often display a very large distribution of pore sizes and consequently pores of size smaller than the X-ray tomography resolution. Not taking such pores into account leads to incorrect calculations of petrophysical properties of rock, therefore the problem of characterizing sub-resolution porosity is identified as a critical issue for geosciences applications. In this paper we propose a new algorithm that allows localization and quantification of sub-resolution porosity. The proposed algorithm can operate in two modes: one of them could be considered as a multi-phase segmentation and could be applied for a variety of X-ray tomography image segmentation tasks; whereas second modification gives better porosity estimation. The algorithm is applied to two images. Good quality image is processed with automatic choice of parameters and without preprocessing. Image of lower quality is processed with preprocessing and manual choice of parameters. In both cases, the computed porosity fits reasonably well with the laboratory measured porosity. Despite the numerous papers on the statistical analyses o fpH, there is no explicit opinion on the use of arithmetic mean as a measure of the central tendency for pH and H+ activity. The problem arises because the transformation of the arithmetic mean for one does not give the arithmetic mean for the other. The paper presents 1) the theoretical considerations on the distribution of pH and H+ activity and relation between them, properties of these distributions, the choice of distributions which should be consistent with the distribution of pH and the distribution of H+ activity and measures of central tendency for features of such distributions and 2) examples of calculations of measures of central tendency for pH and H+ activity based on the literature data on soil and lake water pH. These data analyses included distributions of pH and H+ activities, properties of distribution, descriptive statistics for pH and for the H+ activity and comparison of arithmetic mean with the geometric mean. From the results, it could be concluded that a uniform approach to the choice of measure for the central tendency of pH and H+ activity requires the determination of the type of measure (mean) for one of them and then consistent transformation of this measure. The choice of measure of the central tendency for the variable should be preceded by determination of its distribution. Normal probability distribution of pH and thus lognormal distribution of H+ activity indicate that the arithmetic mean, and its corresponding geometric mean should be used as proper measures of the central tendency for pH and for H+ activity. In minimal invasive image guided catheterization procedures, physicians require information of the catheter position with respect to the patient’s vasculature. However, in fluoroscopic images, visualization of the vasculature requires toxic contrast agent. Static vasculature roadmapping, which can reduce the usage of iodine contrast, is hampered by the breathing motion in abdominal catheterization. In this paper, we propose a method to track the catheter tip inside the patient’s 3D vessel tree using intra-operative single-plane 2D X-ray image sequences and a peri-operative 3D rotational angiography (3DRA). The method is based on a hidden Markov model (HMM) where states of the model are the possible positions of the catheter tip inside the 3D vessel tree. The transitions from state to state model the probabilities for the catheter tip to move from one position to another. The HMM is updated following the observation scores, based on the registration between the 2D catheter centerline extracted from the 2D X-ray image, and the 2D projection of 3D vessel tree centerline extracted from the 3DRA. The method is extensively evaluated on simulated and clinical datasets acquired during liver abdominal catheterization. The evaluations show a median 3D tip tracking error of 2.3 mm with optimal settings in simulated data. The registered vessels close to the tip have a median distance error of 4.7 mm with angiographic data and optimal settings. Such accuracy is sufficient to help the physicians with an up-to-date roadmapping. The method tracks in real-time the catheter tip and enables roadmapping during catheterization procedures. We were particularly interested in the trend for students who received more training and had the shortest retention interval to do worst in the retention test. The competence level achieved was the same for all groups, but we would have expected the differences in retention interval and the amount of training to have had the opposite effect. The article presents the classification and application areas of adhesives while repairing weapon systems, research on adhesive materials' durability, research on mechanical and strength properties of adhesive composites, research on durability of adhesive composites and methodology of their selection for expedient (temporary) repair in the field. The conducted research indicates certain constraints concerning the application of the examined adhesive materials. Consequently, any repairs provided with the use of adhesives should be in most cases only of temporary nature so that certain damage can be fixed quickly but only for a time of the particular task. The TPM domain constitutes a family of recently characterized protein domains that are present in most living organisms. Although some progress has been made in understanding the cellular role of TPM-containing proteins, the relationship between structure and function is not clear yet. We have recently solved the solution and crystal structure of one TPM domain ( BA42) from the Antarctic bacterium Bizionia argentinensis. In this work, we demonstrate that BA42 has phosphoric-monoester hydrolase activity. The activity of BA42 is strictly dependent on the binding of divalent metals and retains nearly 70% of the maximum at 4 °C, a typical characteristic of cold-adapted enzymes. From HSQC, 15N relaxation measurements, and molecular dynamics studies, we determine that the flexibility of the crossing loops was associated to the protein activity. Thermal unfolding experiments showed that the local increment in flexibility of Mg2+-bound BA42, when compared with Ca2+-bound BA42, is associated to a decrease in global protein stability. Finally, through mutagenesis experiments, we unambiguously demonstrate that the region comprising the metal-binding site participates in the catalytic mechanism. The results shown here contribute to the understanding of the relationship between structure and function of this new family of TPM domains providing important cues on the regulatory role of Mg2+ and Ca2+ and the molecular mechanism underlying enzyme activity at low temperatures. The structure is based on a trinuclear {Fe3(μ-O)4} core, with an angular arrangement of the FeIII ions that can be explained by the geometrical restrictions of two bulky ligands, each coordinating to all of the metal cations. The travel and tourism industry has evolved over the past six decades into one of the world’s most significant economic sectors. Travel and tourism are critical to the American economy. This growing industry offers significant potential for job creation across all regions of the country. A considerable competitive advantage of the USA as a tourism destination lies in the variety of experiences the country offers, ranging from large cities and small-town to the many unique historic places and ethnic heritage sites, national parks, camping, hiking, environmental or ecological sites, and well-known theme park. The United States leads the world in revenues from international travel and tourism and ranks second in the number of international visitors. The United States welcomed nearly 75 million international visitors and received 177,2 billion dollars International Tourism Receipts in 2014. But the global travel market has become increasingly competitive. Other countries actively market themselves, and new destinations are aggressively competing for market share. According to World Bank data, the United States received 6,6% of global tourism arrivals in 2014 and accounted for nearly 14,2% of global spending on travel and tourism, down from 7,4% and 17%, respectively, in 2000. Because of that the U.S. market share of spending by international travelers has decreased and the government needs to work hard to support sectors of the economy such as the tourism industry. Sediments of two small dam reservoirs in Poland, Zalew Zemborzycki (ZZ) and Brody Iłżeckie (BI), were studied. Sediments from both reservoirs were sampled at 17 sites in the transects perpendicular to the shoreline, at the river inflow and the frontal dam and analysed using the BCR procedure for speciation of zinc (Zn), lead (Pb) and cadmium (Cd). The risk assessment code (RAC) and the individual contamination factor (ICF) were determined. In BI, the sediments were removed from the considerable part of the reservoir, creating an opportunity to study the effect of dredging on the speciation of trace metals. Trace metals partitioning was differentiated according to the transect/site and in the case of BI sediments also on the transect location in the dredged or undredged part of the reservoir. Considering ZZ sediments, the order of fractions for Zn, Pb and Cd according to a decreasing overall mean percentage contribution to total metal content was the same: F4 (residual) >F3 (oxidisable) >F2 (reducible) >F1 (acid soluble). In sediments of ZZ at most sites, the RAC for Zn, Pb and Cd revealed low or medium risk and ICF low or moderate contamination. For BI sediments, the order of fractions for Pb was similar while for Zn and Cd quite the opposite compared to the sediments of ZZ and it was: Pb–F4>F3>F1>F2, Zn–F1>F3>F2>F4, Cd–F1>F2>F3>F4. For BI sediments, RAC values for Zn, on average, indicated high and very high ecological risk; for Pb low and moderate risk and for Cd – high risk in the initial part and dredged part and according to the average value in the reservoir, while a medium risk in undredged part sediments. The ICF index showed high contamination with Cd for all BI sediments. The sequential analysis showed that Pb is poorly mobile as in sediments of both reservoirs residual fractions accounted, on average, for about 60% of the total content. Biological studies of intracellular dynamic processes commonly require motion analysis of large numbers of particles in live-cell time-lapse fluorescence microscopy imaging data. Many particle tracking methods have been developed in the past years as a first step toward fully automating this task and enabling high-throughput data processing. Two crucial aspects of any particle tracking method are the detection of relevant particles in the image frames and their linking or association from frame to frame to reconstruct the trajectories. The performance of detection techniques as well as specific combinations of detection and linking techniques for particle tracking have been extensively evaluated in recent studies. Comprehensive evaluations of linking techniques per se, on the other hand, are lacking in the literature. Here we present the results of a quantitative comparison of data association techniques for solving the linking problem in biological particle tracking applications. Nine multiframe and two more traditional two-frame techniques are evaluated as a function of the level of missing and spurious detections in various scenarios. The results indicate that linking techniques are generally more negatively affected by missing detections than by spurious detections. If misdetections can be avoided, there appears to be no need to use sophisticated multiframe linking techniques. However, in the practically likely case of imperfect detections, the latter are a safer choice. Cell migration is crucial in development, tissue repair and immunity and frequently aberrant in pathological processes including tumor metastasis. Focal adhesions (FAs) are integrin-based adhesion complexes that form the link between the cytoskeleton and the extracellular matrix and are thought to orchestrate cell migration. Understanding the regulation of FAs by (oncogenic) signaling pathways may identify strategies to target pathological cell migration. Here we describe the development of a robust FA tracker that enables the automatic, multi-parametric analysis of FA dynamics, morphology and composition from time-lapse image series generated by total internal reflection fluorescence (TIRF) microscopy. In control prostate carcinoma cells, this software recapitulates previous findings that relate morphological characteristics of FAs to their lifetime and their cellular location. We then investigated how FAs are altered when cell migration is induced by the metastasis-promoting hormone HGF and subsequently inhibited by activation of the small GTPase Rap1. We performed a detailed analysis of individual FA parameters, which identified FA size, sliding and intensity as primary targets of Rap1. HGF did not have strong effects on any of the FA parameters within the first hours of its addition. Subsequent Bayesian network inference (BNI), using all measured parameters as input, revealed little correlation between changes in cell migration and FA characteristics in this prostate carcinoma cell line. Instead BNI indicated a concerted coordination of cell size and FA parameters. Thus our results did not reveal a direct relation between the regulation of cell migration and the regulation of FA dynamics. [ABSTRACT FROM AUTHOR]

The results of experimental realization of the technology of diffusion doping of X-cut congruent lithium niobate crystals by copper and the results of studying their photorefractive and photovoltaic properties are presented. The latter was done by using the Bragg diffraction of the reading beam with a wavelength of 655 nm on the dynamic transmission volume-phase holograms recorded by laser beams having a wavelength of 532 nm. It is shown that by their photogalvanic properties the created samples, which are 1.8 mm thick, are similar to the single crystals grown by the conventional method from melt containing copper monooxide as a supplement. The development of pharmacological therapies for mitochondrial diseases is hampered by the lack of tissue‐level and circulating biomarkers reflecting effects of compounds on endothelial and mitochondrial function. This phase 0 study aimed to identify biomarkers differentiating between patients with mitochondrial disease and healthy volunteers (HVs). In this cross‐sectional case–control study, eight participants with mitochondrial disease and eight HVs matched on age, sex, and body mass index underwent study assessments consisting of blood collection for evaluation of plasma and serum biomarkers, mitochondrial function in peripheral blood mononuclear cells (PBMCs), and an array of imaging methods for assessment of (micro)circulation. Plasma biomarkers GDF‐15, IL‐6, NT‐proBNP, and cTNI were significantly elevated in patients compared to HVs, as were several clinical chemistry and hematology markers. No differences between groups were found for mitochondrial membrane potential, mitochondrial reactive oxygen production, oxygen consumption rate, or extracellular acidification rate in PBMCs. Imaging revealed significantly higher nicotinamide‐adenine‐dinucleotide‐hydrogen (NADH) content in skin as well as reduced passive leg movement‐induced hyperemia in patients. This study confirmed results of earlier studies regarding plasma biomarkers in mitochondrial disease and identified several imaging techniques that could detect functional differences at the tissue level between participants with mitochondrial disease and HVs. However, assays of mitochondrial function in PBMCs did not show differences between participants with mitochondrial disease and HVs, possibly reflecting compensatory mechanisms and heterogeneity in mutational load. Particle tracking is of key importance for quantitative analysis of intracellular dynamic processes from time-lapse microscopy image data. Because manually detecting and following large numbers of individual particles is not feasible, automated computational methods have been developed for these tasks by many groups. Aiming to perform an objective comparison of methods, we gathered the community and organized an open competition in which participating teams applied their own methods independently to a commonly defined data set including diverse scenarios. Performance was assessed using commonly defined measures. Although no single method performed best across all scenarios, the results revealed clear differences between the various approaches, leading to notable practical conclusions for users and developers. Bottom sediments of two small shallow dam reservoirs, the Zalew Zemborzycki (ZZ) on the Bystrzyca River near Lublin and Brody Iłżeckie (BI) on the Kamienna River, were investigated. The sediment samples from both water reservoirs were collected at 17 sampling sites in the transects perpendicular to the shoreline, at the river inflow and at the frontal dam. The contents of total nitrogen (TN) and total phosphorus (TP) were measured and the research results analyzed statistically. A relationship between TP concentration in sediments and TN, Fe, and Al levels was determined. The maps of spatial distribution of TN and TP were drawn. Sediments of the BI showed greater differentiation of TN and TP contents than sediments in the ZZ, which resulted from a sediment dredging operation performed in most of its area. The dredged part of the BI reservoir had many times lower levels of both TN and TP as compared to sediments in the undredged part. In the ZZ reservoir sediments, longitudinal zonation of TN spatial distribution was observed: the lowest content in the sediments in left-bank part of the reservoir, medium along the middle part, the highest in the right-bank area. In this water reservoir, the zone of the highest TP accumulation occurred in sediments of the upper part and at the frontal dam. It is likely to be caused by the high external loads of P from the Bystrzyca River and by its sedimentation process along with silt and clay particles, respectively. The TN and TP distribution in sediments in the BI was similar and displayed transverse zonation (parallel to the dam). Our research established a strong linear relationship between TP content and Fe and Al concentrations in BI sediments. Abstract: The evaluation of genome integrity in populations occupationally exposed to combine industrial factors is of medical importance. In the present study, the DNA-damage response was estimated by means of the alkaline comet assay in a sizeable cohort of volunteers recruited among workers in the automotive industry. For this purpose, freshly collected lymphocytes were treated with hydrogen peroxide (100μM, 1min, 4°C) in vitro, and the levels of basal and H2O2-induced DNA damage, and the kinetics and efficiency of DNA repair were measured during a 180-min interval after exposure. The parameters studied in the total cohort of workers were in a range of values prescribed for healthy adult residents of Belarus. Based on the 95th percentiles, individuals possessing enhanced cellular sensitivity to DNA damage were present in different groups, but the frequency was significantly higher among elderly persons and among individuals with chronic inflammatory diseases. The results indicate that the inter-individual variations in DNA-damage response should be taken into account to estimate adequately the environmental genotoxic effects and to identify individuals with an enhanced DNA-damage response due to the influence of some external factors or intrinsic properties of the organism. Abstract: Two innovative applications of Nd:YAG laser micromachining techniques are demonstrated in this publication. Research projects to determine the fission product transport mechanisms in TRISO coated particles necessitate heat treatment studies as well as the manufacturing of a unique sealed system for experimentation at very high temperatures. This article describes firstly the design and creation of an alumina jig designed to contain 500μm diameter ZrO2 spheres intended for annealing experiments at temperatures up to 1600°C. Functional requirements of this jig are the precision positioning of spheres for laser ablation, welding and post weld heat treatment in order to ensure process repeatability and accurate indexing of individual spheres. The design challenges and the performance of the holding device are reported. Secondly the manufacture of a sealing system using laser micromachining is reported. ZrO2 micro plugs isolate the openings of micro-machined cavities to produce a gas-tight seal fit for application in a high temperature environment. The technique is described along with a discussion of the problems experienced during the sealing process. Typical problems experienced were seating dimensions and the relative small size (∼200μm) of these plugs that posed handling challenges. Manufacturing processes for both the tapered seating cavity and the plug are demonstrated. In conclusion, this article demonstrates the application of Nd-YAG micromachining in an innovative way to solve practical research problems. [Copyright &y& Elsevier]

Context: The study of genome integrity in some genetic disorders has diagnostic and prognostic importance because of the evident relationship between genome instability and both DNA repair deficiencies and cancer predisposition. Objective: The objective was to compare the chromosomal and DNA damage responses in lymphocytes from patients with Nijmegen breakage syndrome (NBS), Fanconi anemia (FA) and Williams-Beuren syndrome (WBS) to find additional biomarkers of genome instability. Methods: The cytogenetic approaches were combined with the alkaline Comet assay to estimate genome integrity in cultured or freshly isolated and H2O2-treated lymphocytes. Results: Basal frequencies of chromosome aberrations were significantly increased in NBS/FA probands and NBS heterozygous carriers. The NBS diagnosis was confirmed by detecting site-specific rearrangements, while the mitomycin C (MMC)-stress test was highly positive in a FA patient. Among patients with suspected WBS, 12 individuals had a 7q11.23 microdeletion. In the Comet assay, genome instability was revealed in all three disorders, impaired capacity to repair oxidative damage being observed in NBS and WBS in contrast to FA and controls. Conclusion: The results indicate that the estimates of DNA damage response may be proposed as efficient biomarkers for detecting and characterizing genome instability in the genetic disorders under study. Background: Self-assembly is a common theme in proteins of unrelated sequences or functions. The human papillomavirus E7 oncoprotein is an extended dimer with an intrinsically disordered domain, that can form large spherical oligomers. These are the major species in the cytosol of HPV transformed and cancerous cells. E7 binds to a large number of targets, some of which lead to cell transformation. Thus, the assembly process not only is of biological relevance, but represents a model system to investigate a widely distributed mechanism. Methodology/Principal Findings: Using various techniques, we monitored changes in secondary, tertiary and quaternary structure in a time course manner. By applying a robust kinetic model developed by Zlotnik, we determined the slow formation of a monomeric ''Z-nucleus'' after zinc removal, followed by an elongation phase consisting of sequential secondorder events whereby one monomer is added at a time. This elongation process takes place at a strikingly slow overall average rate of one monomer added every 28 seconds at 20 μM protein concentration, strongly suggesting either a rearrangement of the growing complex after binding of each monomer or the existence of a ''conformation editing'' mechanism through which the monomer binds and releases until the appropriate conformation is adopted. The oligomerization determinant lies within its small 5 kDa C-terminal globular domain and, remarkably, the E7 N-terminal intrinsically disordered domain stabilizes the oligomer, preventing an insoluble amyloid route. Conclusion: We described a controlled ordered mechanism with features in common with soluble amyloid precursors, chaperones, and other spherical oligomers, thus sharing determining factors for symmetry, size and shape. In addition, such a controlled and discrete polymerization reaction provides a valuable tool for nanotechnological applications. Finally, its increased immunogenicity related to its supramolecular structure is the basis for the development of a promising therapeutic vaccine candidate for treating HPV cancerous lesions. Abstract: Tagged magnetic resonance imaging (tMRI) is a well-known noninvasive method for studying regional heart dynamics. It offers great potential for quantitative analysis of a variety of kine(ma)tic parameters, but its clinical use has so far been limited, in part due to the lack of robustness and accuracy of existing tag tracking algorithms in dealing with low (and intrinsically time-varying) image quality. In this paper, we evaluate the performance of four frequently used concepts found in the literature (optical flow, harmonic phase (HARP) magnetic resonance imaging, active contour fitting, and non-rigid image registration) for cardiac motion analysis in 2D tMRI image sequences, using both synthetic image data (with ground truth) and real data from preclinical (small animal) and clinical (human) studies. In addition we propose a new probabilistic method for tag tracking that serves as a complementary step to existing methods. The new method is based on a Bayesian estimation framework, implemented by means of reversible jump Markov chain Monte Carlo (MCMC) methods, and combines information about the heart dynamics, the imaging process, and tag appearance. The experimental results demonstrate that the new method improves the performance of even the best of the four previous methods. Abstract: Williams–Beuren syndrome (WBS) is the chromosomal disorder arising from a hemizygous microdeletion at 7q11.23. The present study was focused on a comparative investigation of genomic integrity in WBS patients by use of cytogenetic methods and the alkaline comet assay. Lymphocytes of whole peripheral blood were cultured and metaphases were examined for frequency and spectrum of chromosome aberrations. A WBS-related microdeletion was detected by means of the FISH (fluorescence in situ hybridization) technique. The blood samples from patients who were carriers of this microdeletion, were tested in the comet assay. For this purpose, freshly collected lymphocytes were exposed to hydrogen peroxide (100μM, 1min, 4°C). The frequencies of endogenous and exogenous DNA damage, and the kinetics and efficiency of DNA repair were measured during three subsequent hours of incubation. Comparison of the two data sets in this group of patients demonstrated a slightly elevated average frequency of chromosome aberrations, significantly increased levels of endogenous and H2O2-induced DNA damage, and somewhat impaired DNA repair. The relationship between an abnormal DNA-damage response and the 7q11.23 hemizygous microdeletion was confirmed experimentally when comparing the comet assay data in FISH-positive and FISH-negative lymphocytes from WBS-suspected patients. Briefly, our results indicate the impact of chromosomal instability within this region on susceptibility towards DNA damage, which may contribute to pathogenesis of this disease. Abstract: Xerogels obtained by drying the gels formed by glucofuranose derivatives with organic solvents were studied. The xerogels were characterized using the SEM, XRD, DSC and OPM techniques. The morphology of a xerogel observed by SEM may change from ‘amorphous, fibrillar’ to ‘crystal-like’, which may be caused by time or temperature. The results suggest that a similar transition may take place when the xerogel is being prepared. Thus SEM pictures of xerogels should be treated with great caution as they may not reflect the gelator network morphology in the bulk gel. [Copyright &y& Elsevier]

Abstract: Before the memory trace is stored long term, it must undergo a phase of consolidation during which it remains susceptible to modifications. It has previously been proposed that during consolidation, memories are kept from being stored long term, and can therefore be modified with additional information resulting from ongoing behavior. The Chasmagnathus associative memory model is used here to test whether it is possible during consolidation to modify the long-term expression of a memory generated by a weak training procedure. In this memory model, long-term memory expression is achieved after strong training protocols, a 15-spaced trial procedure. After a weak training protocol (WTP, six spaced trials), crabs do not show memory retention when tested in the long term. Nevertheless, the WTP builds a long-term memory that it is indeed consolidated, but remains unexpressed. Here we show that memory can be modified by experience during this short period after learning: memory expression can be enhanced by a Single Trial Session, on the condition that this session takes place contingent upon the consolidation period. We also found that during this time, the memory built by the WTP is behaviorally expressed, in contrast with what occurs at long term. Our results support the idea that during consolidation memories can be evaluated in the background of concurrent experiences. In particular, we propose that during the consolidation period it is possible for crabs to assess which experiences, among those stored long term, will be expressed long term. [ABSTRACT FROM AUTHOR]

Abstract: Deoxycytidine kinase (dCK) is a key enzyme in the salvage of deoxynucleosides and in the activation of several anticancer and antiviral nucleoside analogues. We recently showed that dCK was activated in vivo by phosphorylation of Ser-74. However, the protein kinase responsible was not identified. Ser-74 is located downstream a Glu-rich region, presenting similarity with the consensus phosphorylation motif of casein kinase 1 (CKI), and particularly of CKI δ. We showed that recombinant CKI δ phosphorylated several residues of bacterially overexpressed dCK: Ser-74, but also Ser-11, Ser-15, and Thr-72. Phosphorylation of dCK by CKI δ correlated with increased activity reaching at least 4-fold. Site-directed mutagenesis demonstrated that only Ser-74 phosphorylation was involved in dCK activation by CKI δ, strengthening the key role of this residue in the control of dCK activity. However, neither CKI δ inhibitors nor CKI δ siRNA-mediated knock-down modified Ser-74 phosphorylation or dCK activity in cultured cells. Moreover, these approaches did not prevent dCK activation induced by treatments enhancing Ser-74 phosphorylation. Taken together, the data preclude a role of CKI δ in the regulation of dCK activity in vivo. Nevertheless, phosphorylation of dCK by CKI δ could be a useful tool for elucidating the influence of Ser-74 phosphorylation on the structure–activity relationships in the enzyme. Studying intracellular dynamics is of fundamental importance for understanding healthy life at the molecular level and for developing drugs to target disease processes. One of the key technologies to enable this research is the automated tracking and motion analysis of these objects in microscopy image sequences. To make better use of the spatiotemporal information than common frame-by-frame tracking methods, two alternative approaches have recently been proposed, based upon either Bayesian estimation or space-time segmentation. In this paper, we propose to combine the power of both approaches, and develop a new probabilistic method to segment the traces of the moving objects in kymograph representations of the image data. It is based on variable-rate particle filtering and uses multiscale trend analysis of the extracted traces to estimate the relevant kinematic parameters. Experiments on realistic synthetically generated images as well as on real biological image data demonstrate the improved potential of the new method for the analysis of microtubule dynamics in vitro. Summary: In Chinese hamster ovary cells, microtubules originate at the microtubule organizing center (MTOC) and grow persistently toward the cell edge, where they undergo catastrophe []. In axons, microtubule dynamics must be regulated differently because microtubules grow parallel to the plasma membrane and there is no MTOC. GFP-tagged microtubule plus end tracking proteins (+TIPs) mark the ends of growing neuronal microtubules []. Their fluorescent “comet-like” pattern reflects turnover of +TIP binding sites []. Using GFP-tagged +TIPs and fluorescence-based segmentation and tracking tools, we show that axonal microtubules grow with a constant average velocity and that they undergo catastrophes at random positions, yet in a programmed fashion. Using protein depletion approaches, we find that the +TIPs CLIP-115 and CLIP-170 affect average microtubule growth rate and growth distance in neurons but not the duration of a microtubule growth event. In N1E-115 neuroblastoma cells, we find that EB1, the core +TIP [], regulates microtubule growth rate, growth distance, and duration, consistent with in vitro data []. Combined, our data suggest that CLIPs influence the axonal microtubule/tubulin ratio, whereas EB1 stimulates microtubule growth and structural transitions at microtubule ends, thereby regulating microtubule catastrophes and the turnover of +TIP binding sites. Abstract: Reconsolidation studies have led to the hypothesis that memory, when labile, would be modified in order to incorporate new information. This view has reinstated original propositions suggesting that short-term memory provides the organism with an opportunity to evaluate and rearrange information before storing it, since it is concurrent with the labile state of consolidation. The Chasmagnathus associative memory model is used here to test whether during consolidation it is possible to change some attribute of recently acquired memories. In addition, it is tested whether these changes in behavioral memory features can be explained as modifications on the consolidating memory trace or as a consequence of a new memory trace. We show that short-term memory is, unlike long-term memory, not context specific. During this short period after learning, behavioral memory can be updated in order to incorporate new contextual information. We found that, during this period, the cycloheximide retrograde amnesic effect can be reverted by a single trial in a new context. Finally, by means of memory sensitivity to cycloheximide during consolidation and reconsolidation, we show that the learning of a new context (CS) during this short-term memory period builds up a new memory trace that sustains the behavioral memory update. Deoxycytidine kinase (dCK) is a key enzyme in the salvage of deoxyribonucleosides and in the activation of several anticancer and antiviral nucleoside analogues. We have recently shown that dCK is a phosphoprotein. Four in vivo phosphorylation sites were identified: Thr-3, Ser-11, Ser-15, and Ser-74. Site-directed mutagenesis demonstrated that phosphorylation of Ser-74, the major phosphorylated residue, strongly influences dCK activity in eucaryotic cells. Here, we show that phosphorylation of the three other sites, located in the N-terminal extremity of the protein, does not significantly modify dCK activity, but phosphorylation of Thr-3 could promote dCK stability. Quantitative analysis of biological image data generally involves the detection of many subresolution spots. Especially in live cell imaging, for which fluorescence microscopy is often used, the signal-to-noise ratio (SNR) can be extremely low, making automated spot detection a very challenging task. In the past, many methods have been proposed to perform this task, but a thorough quantitative evaluation and comparison of these methods is lacking in the literature. In this paper, we evaluate the performance of the most frequently used detection methods for this purpose. These include seven unsupervised and two supervised methods. We perform experiments on synthetic images of three different types, for which the ground truth was available, as well as on real image data sets acquired for two different biological studies, for which we obtained expert manual annotations to compare with. The results from both types of experiments suggest that for very low SNRs (≈ 2), the supervised (machine learning) methods perform best overall. Of the unsupervised methods, the detectors based on the so-called -dome transform from mathematical morphology or the multiscale variance-stabilizing transform perform comparably, and have the advantage that they do not require a cumbersome learning stage. Transcription of the human papillomavirus E7 oncoprotein is negatively controlled by the viral E2 protein, and loss of this repression leads to irreversible transformation and carcinogenesis. Here we show that interaction of the HPV16 E7 protein with the DNA binding domain of the E2 protein (E2C) leads to ionic strength-dependent hetero-oligomerization even at the lowest concentrations measurable. Titration experiments followed by light scattering and native gel electrophoresis show insoluble oligomeric complexes with a =2000 nm diameter and intermediate soluble complexes 40 and 115 nm in diameter, respectively, formed in excess of E2C. A discrete oligomeric soluble complex formed in excess of E7 displays a diameter of 12 nm. The N-terminal domain of E7 interacts with E2C with a KD of 0.1 μM, where the stretch of residues 25-40 of E7, encompassing both a PEST motif and phosphorylation sites, is sufficient for the interaction. Displacement of the soluble E7-E2C complex by an E2 site DNA duplex and site-directed mutagenesis indicate that the protein-protein interface involves the DNA binding helix of E2. The formation of complexes of different sizes and properties in excess of either of the viral proteins reveals a finely tuned mechanism that could regulate the intracellular levels of both proteins as infection and transformation progress. Sequestering E2 into E7-E2 oligomers provides a possible additional route to uncontrolled E7 expression, in addition and prior to the disruption of the E2 gene during viral integration into the host genome. Background: Various studies have shown an association between low birth weight and the prevalence of elevated blood pressure later in life. However, a relationship between birth weight and blood pressure in the neonatal period has not been investigated yet. Objective: To study the relationship between birth weight and blood pressure during the first week of life. Methods: Systolic blood pressure (SBP), diastolic blood pressure (DBP) and mean arterial blood pressure (MBP) were measured every 8 h during days 1, 2, 3, 4 and 7 in 44 small for gestational age (SGA) infants and compared with those of infants born with a birth weight appropriate for gestational age (AGA). The infants were matched for gestational age and gender. Results: Meanbirth weight (range) in the SGA group was 778 (460–1,400) g versus 1,524 (960–2,730) g in the AGA group, and mean (range) gestational age of both groups was 30.5 (27.3–35.5) weeks. In the first week of life, SGA and AGA infants had similar SBP, DBP and MBP. Also, after adjustment for confounders (maternal medication, smoking), SBP, DBP and MBP rose equally in SGA and AGA infants during the first week of life. AGA infants showed a positive correlation between birth weight and blood pressure, but SGA infants showed an inverse correlation. Conclusions: Although differences in blood pressure were found between SGA and AGA infants in the first week of life, the inverse relationship between birth weight and blood pressure in SGA infants warrants follow-up studies to investigate a possible link with late cardiovascular disease. Pulmonary arterial hypertension (PAH) patients eventually die of right heart failure (RHF). Currently, there is no suitable pre-clinical model to study PAH. Therefore, we aim to develop a right heart dysfunction (RHD) model using the 3-dimensional engineered heart tissue (EHT) approach and cardiomyocytes derived from patient-induced pluripotent stem cells (iPSCs) to unravel the mechanisms that determine the fate of a pressure-overloaded right ventricle. iPSCs from PAH and healthy control subjects were differentiated into cardiomyocytes (iPSC-CMs), incorporated into the EHT, and maintained for 28 days. In comparison with control iPSC-CMs, PAH-derived iPSC-CMs exhibited decreased beating frequency and increased contraction and relaxation times. iPSC-CM alignment within the EHT was observed. PAH-derived EHTs exhibited higher force, and contraction and relaxation times compared with control EHTs. Increased afterload was induced using 2× stiffer posts from day 0. Due to high variability, there were no functional differences between normal and stiffer EHTs, and no differences in the hypertrophic gene expression. In conclusion, under baseline spontaneous conditions, PAH-derived iPSC-CMs and EHTs show prolonged contraction compared with controls, as observed clinically in PAH patients. Abstract: Time-lapse fluorescence microscopy imaging has rapidly evolved in the past decade and has opened new avenues for studying intracellular processes in vivo. Such studies generate vast amounts of noisy image data that cannot be analyzed efficiently and reliably by means of manual processing. Many popular tracking techniques exist but often fail to yield satisfactory results in the case of high object densities, high noise levels, and complex motion patterns. Probabilistic tracking algorithms, based on Bayesian estimation, have recently been shown to offer several improvements over classical approaches, by better integration of spatial and temporal information, and the possibility to more effectively incorporate prior knowledge about object dynamics and image formation. In this paper, we extend our previous work in this area and propose an improved, fully automated particle filtering algorithm for the tracking of many subresolution objects in fluorescence microscopy image sequences. It involves a new track management procedure and allows the use of multiple dynamics models. The accuracy and reliability of the algorithm are further improved by applying marginalization concepts. Experiments on synthetic as well as real image data from three different biological applications clearly demonstrate the superiority of the algorithm compared to previous particle filtering solutions. Objective: We describe the clinical course of an infant who presented with severe fetal anemia and fetal hydrops following congenital parvovirus B19 infection before 16 gestational weeks. The fetus was treated by cordocentesis and intrauterine transfusion at 18 weeks. Results: The infant demonstrated mild unilateral ventriculomegaly on antenatal magnetic resonance imaging, and polymicrogyria and heterotopia on postnatal magnetic resonance imaging. Despite the extensive literature on the effect on soil properties of afforestation of former arable land, we still lack full understanding of whether the changes proceed in the same direction and at the same rate, and of how long is required to achieve a state of soil equilibrium typical of a natural forest ecosystem. Therefore, as part of a study comparing post-arable sandy soils (Dystric Arenosols) afforested with Scots pine (Pinus silvestris L.) with arable soils and soils of continuous coniferous forests, the range and direction of changes in pH, organic carbon (Corg), total nitrogen (Ntot), ammonium (N-NH4) and nitrates (N-NO3) in soil solution, total (Ptot) and available (Pav) phosphorus were determined. The studies were carried out in south-east Poland (51°30′-51°37′N, 22°20′-22°35′E). Ten paired sites of afforested soils (five with 14- to 17-year-old stands and five with 32- to 36-year-old stands) with adjacent cultivated fields, and five sites of continuous forest with present stands of ca. 130–150 years old were selected. Soil samples were taken from the whole thickness of master horizons and, in the case of the A horizon of the afforested soils, from three layers: 0–5 (A0–5), 5–10 (A5–10) and 10–20 cm (A10–20). The cultivated soils in the Ap horizon showed higher pH (by ca. 1.0 unit), lower Corg and C:N, similar Ntot, lower N-NH4, higher N-NO3, higher Ptot and Pav contents compared with the Ah horizon of continuous forest soils. The results indicated decreased soil pH in the former plough layer of the afforested soils, with the greatest decrease observed in the 0–5 cm layer. In these soils, the Corg content was considerably higher in the A0–5 layer, but lower in the two deeper layers and in the whole A horizon (0–20 cm) compared with the Ap horizon of the arable soils. The results indicate that the Corg content, after an initial phase of decline, again achieved a level characteristic of arable soils. The Ntot content in all layers of the A horizon of the afforested soils was lower than in the Ap horizon of the arable soils, and showed a reduction with stand age, especially in deeper layers. The C:N ratios in the mineral topsoil increased with stand age. N-NH4 content increased and N-NO3 decreased after afforestation. The Ptot and Pav contents in all layers and in the whole A horizon of the afforested soils, on stands of both ages, was lower than in the Ap of the cultivated soils. From the results, it could be concluded that, after more than 30 years of tree growth, the soils of the A horizon were still more similar to arable than to continuous forest soils with respect to Corg, Ptot and Pav. With respect to pH, N-NH4 and N-NO3, especially in the 0–5 cm layer, they were more similar to continuous forest soils than to cultivated soils, but with respect to Ntot and C:N ratio they were somewhere in between. Despite the extensive literature on the effect of afforestation of former arable land on soil properties, we still do not fully understand whether the changes proceed in the same direction and at the same rate or how long it takes to achieve a state of soil equilibrium typical of a natural forest ecosystem. Therefore, as part of a comparative study of post-arable sandy soils (Distric Arenosols) afforested with Scots pine (Pinus silvestris L. ) with respect to arable soils and soils of continuous coniferous forests, a range and direction of the changes in some of their physical and sorptive properties were determined. The studies were carried out in SE Poland, 51°30′–51°37′N, 22°20′–22°35′E. Ten paired sites of the afforested soils (five with 14- to 17-year-old stands and five with 32- to 36-year-old stands) with adjacent cultivated fields and five sites of continuous forests with present stands of ca. 150 years were selected. For the physical properties, undisturbed soil cores were sampled from the upper part of each horizon while in the case of A horizon of the afforested soils, from two layers: 0–5 cm and 10–15 cm. For the remaining analyses, soil was taken from the whole thickness of the master horizons and in the case of A horizon of the afforested soils, from three layers: 0–5, 5–10 and 10–20 cm. The following properties were analysed: texture, bulk density (BD), total porosity (TP), water content at potential of −0.098, −9.81 and −49.03 kPa, hydrolytic acidity (Ha), base exchangeable cations: Ca2+, Mg2+, K+, Na+, total exchangeable bases (TEB), cation exchange capacity (CEC) and base saturation (BS). Afforestation caused a decrease in BD, an increase in TP and had no affect on water properties when compared with the cultivated soils. The changes referred to the A horizon, particularly to its 0–5 cm layer, and were related to the stand age. The CEC gradually rose in the former plough layer, beginning from the uppermost part, but during the first two decades its increase in the 0–5 cm layer was offset by a decline in the deeper layers. No substantial increase in CEC, in the whole A horizon, was recorded until three to four decades of afforestation. Afforestation also invoked an increase in Ha, a drop in TEB, particularly Ca2+, Mg2+ and K+, and reduction in BS. No differences between soils for all the studied properties for B and C horizons were observed. It was noted that more than 30 years after afforestation, the TEB and BS as well as Ca2+, Mg2+ and K+ content differed substantially, but in most cases not significantly, from their values in the cultivated soils and reached a level more similar to the soils of continuous coniferous forests. With respect to the water properties, Ha and CEC of the afforested soils still resembled arable soils, whereas regarding the TP and BD, they were somewhere in the middle. Abstract: 2-Chloroadenosine (2-CAdo) is an adenosine deaminase-resistant analogue of adenosine, widely used as an adenosine receptor agonist. This compound has been shown to induce apoptosis in several cell types either via activation of adenosine receptors or via intracellular metabolism. However, the molecular mechanisms of 2-CAdo-induced apoptosis are unclear. Here, we analyzed the effects of 2-CAdo in the leukemia cell line EHEB. 2-CAdo was found to induce apoptosis in EHEB cells, as shown by caspase-3 activation, DNA fragmentation, poly(ADP-ribose) polymerase (PARP) cleavage and phosphatidylserine exposure. Cytotoxicity of 2-CAdo was completely suppressed by 5-iodotubercidin, an adenosine kinase inhibitor, indicating that apoptosis induced by 2-CAdo was the result of its intracellular metabolism. Accordingly, we found that 2-CAdo was efficiently converted into 2-chloroATP. In parallel, a decrease of intracellular ATP concentration as well as a general inhibition of macromolecular synthesis, involving DNA, RNA and protein synthesis, was observed. Moreover, 2-CAdo induced cytochrome c release into the cytosol, indicating activation of the intrinsic pathway of apoptosis. This was found associated with a decline in Mcl-1 protein level and p53-independent. Inhibition of AMP deaminase by coformycin markedly prevented ATP depletion, and also significantly reduced 2-CAdo cytotoxicity and caspase-3 activation. In conclusion, our data show that intracellular metabolism of 2-CAdo can lead to activation of the intrinsic pathway of apoptosis and that ATP depletion, in addition to the accumulation of the triphosphate analogue, contributes to 2-CAdo-induced apoptosis. Abstract: Deoxycytidine kinase (dCK) activates several antileukaemic nucleoside analogues. We have recently reported that the activity of dCK, overexpressed in HEK 293T cells, correlates with its phosphorylation level on Ser-74. Here, we show that dCK from B-cell chronic lymphocytic leukaemia (B-CLL) lymphocytes can be detected by an anti-phospho-Ser-74 antibody and that interindividual variability in dCK activity is related to its phosphorylation level on Ser-74. Moreover, pharmacological intervention modified Ser-74 phosphorylation, in close parallel with changes in dCK activity. These results suggest that activation of dCK via phosphorylation of Ser-74 might constitute a new therapeutic strategy to enhance activation and efficacy of nucleoside analogues.
