Background: Little is known on the effects of delirium onset and duration on outcome in critically ill patients with cancer. Objectives: To determine the impact of delirium onset and duration on intensive care unit (ICU) and hospital mortality and length of stay (LOS) in patients with cancer. Methods: Of the 915 ICU patients admitted in 2018, 371 were included for analysis after excluding for terminal disease, <24-h ICU stay, lack of active cancer and delirium. Delirium was defined as early if onset was within 2 days of ICU admission, late if onset was on day 3 or later, short if duration was 2 days or less, and long if duration was 3 days or longer. Patients were placed into 4 combination groups: early-short, early-long, late-short, and late-long delirium. Multivariate analysis controlling for sex, age, metastatic disease, and predelirium hospital LOS was performed to determine ICU and hospital mortality and LOS. Exploratory analysis of long-term survival was also performed. Restricted cubic splines were performed to confirm the use of 2 days to distinguish between early versus late onset and short versus long duration. Results: A total of 32.9% (n = 122) patients had early-short, 39.1% (n = 145) early-long, 16.2% (n = 60) late-short, and 11.9% (n = 44) late-long delirium. Late-long delirium was independently associated with increased ICU (OR 4.45, CI 1.92-10.30; P <.001) and hospital (OR 2.91, CI 1.37-6.19; P =.005) mortality and longer ICU (OR 1.97, CI 1.58-2.47; P <.001) LOS compared to early-short delirium. Early delirium had better overall survival at 18 months than late delirium. Long-term survival further improved when delirium duration was 2 days or less. Prediction heatmaps confirm the use of a 2-day cutoff. Conclusion: Late delirium, especially with long duration, significantly worsens outcome in ICU patients with cancer and should be considered a harbinger of poor overall condition. Cryptostegia grandiflora and C. madagascariensis (Apocynaceae) are the only two species of this Madagascan plant genus. Both have been transported around the world as ornamentals due to their attractive flowers and based on a perceived potential as sources of rubber - hence, the common name rubber vine - because of their copious latex, which also contains toxic cardiac glycosides. As a result of their vigorous growth and ability to climb over and smother vegetation, both species have become invasive, posing an actual or potential threat to native ecosystems in many tropical and sub-tropical countries, as well as to human and animal health. Classical biological control (CBC), or the introduction of co-evolved natural enemies to control an invasive alien species in its exotic range, has successfully been used to tackle C. grandiflora in northern Queensland, Australia. This strategy is currently being evaluated for its suitability to manage C. madagascariensis in north-eastern Brazil using the same Madagascan rust fungus, Maravalia cryptostegiae, released as a CBC agent in Australia. For CBC to be successful, it is critical to understand the taxonomy of the invader as well as the origin(s) of its weedy biotype(s) in order to select the best-matched co-evolved natural enemies. Based on an exhaustive search in published and unpublished sources, we summarise the taxonomy and uses of these rubber vines, follow their historical movements and track their earliest records and current weed status in more than 80 countries and territories around the world. Food preservation is an essential application for polymers, particularly in packaging. Complex multilayer films, such as those used for modified atmosphere packaging (MAP), extend the shelf life of sensitive foods. These mostly contain various polymers to achieve the necessary combination of mechanic, optic, and barrier properties that limit their recyclability. As the European Union's Circular Economy Action Plan calls for sustainable products and business models, including waste prevention policies and recycling quotas, with plastic packaging being a high priority, solutions towards more sustainable multilayer packaging are urgently needed. This study evaluated and compared the recycling potential of functionally equivalent PET (polyethylene terephthalate) and PP (polypropylene) post-consumer MAP through structure analysis and recycling simulation. The structure analysis revealed that both types of MAP contained functional (stability) and barrier layers (oxygen and moisture). The recycling simulation showed that the PP-based packaging was recyclable 10 times, maintaining its mechanical properties and functionality. At the same time, the PET-based MAP resulted in a highly brittle material that was unsuitable for reprocessing into similar economic value products. The secondary material from the PP-based MAP was successfully manufactured into films, demonstrating the functional possibility of closed-loop recycling. The transition from a linear to a circular economy for MAP is currently still limited by safety concerns due to a lack of sufficient and efficient purification methods, but the proper design of multilayers for recyclability is a first step towards circularity. The mitochondrion is a maternally inherited organelle that produces approximately 90% of cellular energy. The mitochondrial genome (mtDNA) consists of 13 genes that are critical components of oxidative phosphorylation. Identifying and understanding variation within this genome could elucidate differences in metabolic and physiological traits, specifically growth efficiency in cattle. Our aim is to identify how variation in the mitochondrial genome influences efficiency in beef cattle. Low-pass sequencing data (average 0.5X coverage of the autosomes) were collected (Neogen) from 49 animals within the UNL herd (composite Simmental, Red Angus, and Gelbvieh). The data were trimmed and mapped (BWA-MEM) to the ARS-UCD1.2 cattle genome. After variant impacts were predicted (Ensembl VEP) haplotype networks were created to visualize variation among samples. Over 60X coverage was achieved across the mitochondrial genome, on average. Excluding the D-loop, 62 variants were identified in the 49 animals: 2 tRNA, 6 rRNA, and 54 coding single nucleotide polymorphisms (SNPs). These 62 variants were encompassed in 23 unique haplotypes. Of the coding variants, 13 were nonsynonymous and found in the following genes encoding components of oxidative phosphorylation: ND1 (n = 3), ND2 (n = 1), ND3 (n = 1), ND4 (n = 1), ND5 (n = 1), ND6 (n = 3), COX3 (n = 2), and CYTB (n = 1). Nine unique haplotypes were present considering only nonsynonymous variants. Nonsynonymous mutations were identified in complex I genes (ND1-6), in the complex IV gene COX3, and in CYTB, which is involved in multiple processes including the electron transport chain. These data, from a closed herd at UNL, support that there is a great deal of mtDNA variation present among beef cattle for which to analyze the impact of mitochondrial genotype on animal efficiency. Performance data demonstrate variation in average daily gain (ADG) and residual feed intake (RFI) in this herd, with a mean ADG of 1.95 kg/d (SD = 0.23 kg/d) and a mean RFI of -0.11 kg (SD = 0.57 kg). Data on individual feed intake, growth rates, and carcass performance values will continue to be collected on all animals sequenced. Based upon these data we hypothesize that variation in the mitochondrial genome will be associated with altered complex I and IV activity. For a more comprehensive understanding of this variation, the sample size will be expanded to over 1,500 animals in the next 2 yr. Overall, from our initial analysis of the mitochondrial genomes from 49 animals, we have identified 62 variants that we anticipate will explain variation in cellular and growth efficiency. Simple Summary: This study reveals that circulating Hsp70 levels could serve as a tumor biomarker for patients with NSCLC in advanced UICC stages. All patients in advanced tumor stages had significantly elevated Hsp70 levels in the circulation compared to a healthy control cohort and an early-stage tumor cohort, and Hsp70 levels progressively increased with higher UICC tumor stages. These findings demonstrate the potential of Hsp70 measurements to predict an advanced tumor stage in NSCLC patients. We have also demonstrated that the prevalence of CD3−/CD94+ NK cells and CD8+ cytotoxic T cells were greater in advanced tumor stages, whereas that of CD4+ T helper cells was decreased. We hypothesize that raised levels of circulating Hsp70 in higher tumor stages might support NK cell proliferation, but that a lowered prevalence of CD4+ T helper cells could temper the capacity of cytolytic CD8+ T cells and NK cells to control tumor growth. Non-small cell lung cancer (NSCLC) is the second most frequently diagnosed tumor worldwide. Despite the clinical progress which has been achieved by multimodal therapies, including radiochemotherapy, and immune checkpoint inhibitor blockade, the overall survival of patients with advanced-stage NSCLC remains poor, with less than 16 months. It is well established that many aggressive tumor entities, including NSCLC, overexpress the major stress-inducible heat shock protein 70 (Hsp70) in the cytosol, present it on the plasma membrane in a tumor-specific manner, and release Hsp70 into circulation. Although high Hsp70 levels are associated with tumor aggressiveness and therapy resistance, membrane-bound Hsp70 can serve as a tumor-specific antigen for Hsp70-primed natural killer (NK) cells, expressing the C-type lectin receptor CD94, which is part of the activator receptor complex CD94/NKG2C. Therefore, we investigated circulating Hsp70 levels and changes in the composition of peripheral blood lymphocyte subsets as potential biomarkers for the advanced Union for International Cancer Control (UICC) stages in NSCLC. As expected, circulating Hsp70 levels were significantly higher in NSCLC patients compared to the healthy controls, as well as in patients with advanced UICC stages compared to those in UICC stage I. Smoking status did not influence the circulating Hsp70 levels significantly. Concomitantly, the proportions of CD4+ T helper cells were lower compared to the healthy controls and stage I tumor patients, whereas that of CD8+ cytotoxic T cells was progressively higher. The prevalence of CD3−/CD56+, CD3−/NKp30, CD3−/NKp46+, and CD3−/NKG2D+ NK cells was higher in stage IV/IIIB of the disease than in stage IIIA but were not statistically different from that in healthy individuals. However, the proportion of NK cells expressing CD94 and the activation/exhaustion marker CD69 significantly increased in higher tumor stages compared with stage I and the healthy controls. We speculate that although elevated circulating Hsp70 levels might promote the prevalence of CD94+ NK cells in patients with advanced-stage NSCLC, the cytolytic activity of these NK cells also failed to control tumor growth due to insufficient support by pro-inflammatory cytokines from CD4+ T helper cells. This hypothesis is supported by a comparative multiplex cytokine analysis of the blood in lung cancer patients with a low proportion of CD4+ T cells, a high proportion of NK cells, and high Hsp70 levels versus patients with a high proportion of CD4+ T cells exhibiting lower IL-2, IL-4, IL-6, IFN-γ, granzyme B levels. Various bioactive food compounds may confer health and longevity benefits, possibly through altering or preserving the epigenome. While bioactive food compounds are widely being marketed for human consumption as 'improving health and longevity' by counteracting harmful effects of poor nutrition and lifestyle, claimed effects are often not adequately documented. Using the honey bee (Apis mellifera) as a model species, we here employed a multi-step screening approach to investigate seven compounds for effects on lifespan and DNA methylation using ELISA and whole genome bisulfite sequencing (WGBS). A positive longevity effect was detected for valproic acid, isovaleric acid, and cyanocobalamin. For curcumin, we found that lifespan shortening caused by ethanol intake, was restored when curcumin and ethanol were co-administered. Furthermore, we identified region specific DNA methylation changes as a result of ethanol intake. Ethanol specific changes in DNA methylation were fully or partially blocked in honey bees receiving ethanol and curcumin together. Ethanol-affected and curcumin-blocked differentially methylated regions covered genes involved in fertility, temperature regulation and tubulin transport. Our results demonstrate fundamental negative effects of low dose ethanol consumption on lifespan and associated DNA methylation changes and present a proof-of-principle on how longevity and DNA methylation changes can be negated by the bioactive food component curcumin. As a result of global warming, glaciers worldwide recede and lose mass. Glacier ablation is influenced by surface characteristics, in particular the existence of debris cover. The evolution of glaciers may also be influenced by the existence of proglacial lakes, which are worldwide increasing in number and size. We examine the recent changes of the terminus of Pasterze Glacier by discussing the thinning and dynamics of the glacier terminus along with the changes of the glacial-proglacial transition zone between September 2016 and September 2021. By considering land cover in the interpretation, we find that glacier surface characteristics show different rates of elevation changes and surface movement. We mainly use orthophotos and digital elevation models (DEMs) resulting from airborne surveys of 12 different acquisition dates, but also draw on ablation stake measurements, lake water level data, water temperature data, as well as thermal infrared (TIR) imagery. While debris-covered ice lowered typically −5.0 m a−1 ± 4.6 m a−1 (mean ± standard deviation), debris-free ice was affected by ablation in the range of −7.0 m a−1 ± 2.7 m a−1, thus demonstrating that melt rates in similar elevations are related to surface characteristics. The elevation change rates were, in most cases, in a similar range throughout the observed periods of time. The horizontal displacement close to glacier-marginal crevasses amounted to up to ∼8 m a−1, but was mostly in the range of ∼1–4 m a−1. Considering both horizontal displacement and elevation changes, we find that clean and dirty ice surfaces show larger thickness losses and are moving faster compared to debris-covered glacier surfaces. Diurnal and seasonal fluctuations of lake water levels in the range of decimeters and lake water temperatures mostly clearly exceeding 0 °C are indicative of the potential influence of the proglacial lake on the glacier terminus. Given the decelerating and decaying behavior of the Pasterze Glacier in recent decades, we interpret the change rates of surface lowering and horizontal displacement 2016–2021 to be influenced by other factors such as the recently formed proglacial lake. This is also supported by direct ablation measurements that mostly show lesser ice melt differences compared to DEM-based surface elevation changes, suggesting that other factors such as thermally-induced melting contributed to an increased elevation difference. • Change rates of glacier surface lowering and horizontal displacement varied only slightly throughout the period 2016-2021. • We found evidence that the changes of glacier elevation and displacement rates are related to surface characteristics. • We found a discrepancy between direct ice melt measurements compared to DEM-based glacier surface changes. • The thermal condition of a proglacial lake and a fluctuating water level indicate conditions that evoke increased ice melt. Introduction: Neoadjuvant imatinib (Neo-IM) therapy may facilitate R0 resection in primary gastrointestinal stromal tumors (GISTs) that are large or in difficult anatomic locations. While response to preoperative tyrosine kinase inhibitors is associated with better outcome in metastatic GIST, little is known about prognostic factors after Neo-IM in primary GIST. Study Design: Patients with primary GIST with or without synchronous metastases who underwent Neo-IM were retrospectively analyzed from a prospective maintained institutional database for Response Evaluation Criteria in Solid Tumors (RECIST), tumor viability, and mitotic rate. Overall survival (OS) was estimated by Kaplan-Meier and compared by log-rank test. Cox proportionate hazard models were used for univariate and multivariate analysis. Results: One hundred and fifty patients were treated for a median of 7.1 months (range 0.2–160). By RECIST, partial response, stable disease, and progressive disease were seen in 40%, 51%, and 9%, respectively. By pathologic analysis, ≤ 50% of the tumor was viable in 72%, and the mitotic rate was ≤ 5/50HPF in 74%. On multivariate analysis, RECIST response and tumor viability were not associated with OS, while post-treatment high mitotic rate (hazard ratio (HR) for death 5.3, CI 2.3–12.4), R2 margins (HR 6.0, CI 2.3–15.5), and adjuvant imatinib (HR 0.4, CI 0.2–0.9) were (p < 0.05). Five-year OS was 81 vs. 38% for low vs. high mitotic rate; 81, 59, and 39% for R0, R1, and R2 margins; and 75 vs 61% for adjuvant vs. no adjuvant imatinib therapy (p < 0.05). Conclusions: In primary GIST undergoing Neo-IM therapy, progression was uncommon, but substantial down-sizing occurred in the minority. High tumor mitotic rate and incomplete resection following Neo-IM were associated with poor outcome, while adjuvant imatinib was associated with prolonged survival. [ABSTRACT FROM AUTHOR]

Low central venous pressure (LCVP) anesthesia reduces blood loss during hepatic resection and historically has required a central venous catheter (CVC) for intra-operative monitoring. The aim of this study was to assess the effect of an evolution of practice to CVP monitoring without CVC on the perioperative outcomes after liver resection. A retrospective study of partial hepatectomy patients from 2007 to 2016 who were over 18 years of age was performed. Of 3903 patients having partial hepatectomy, 2445 (62%) met inclusion criteria, and 404 (16%) had a CVC. Overall morbidity (33% non-CVC vs 38% CVC P = 0.076), major morbidity (16% vs 20% P = 0.067), and infective complications (superficial wound infection) 3% vs 4% P = 0.429; deep wound infection (5% vs 6% P = 0.720) did not differ between the two groups. In multivariate analysis, superficial wound infection, deep wound infection, and major complications were not associated with the presence of a CVC. All-cause mortality at 90 days was associated with CVC presence (OR 3.45, CI 1.74–6.85, P = 0.001) and age (OR 1.05, CI 1.02–1.08, P < 0.001). μ‐Opioid receptors (μ‐ORs) play a critical role in the modulation of pain and mediate the effects of the most powerful analgesic drugs. Despite extensive efforts, it remains insufficiently understood how μ‐ORs produce specific effects in living cells. We developed new fluorescent ligands based on the μ‐OR antagonist E‐p‐nitrocinnamoylamino‐dihydrocodeinone (CACO), that display high affinity, long residence time and pronounced selectivity. Using these ligands, we achieved single‐molecule imaging of μ‐ORs on the surface of living cells at physiological expression levels. Our results reveal a high heterogeneity in the diffusion of μ‐ORs, with a relevant immobile fraction. Using a pair of fluorescent ligands of different color, we provide evidence that μ‐ORs interact with each other to form short‐lived homodimers on the plasma membrane. Background: DBS in the ventral intermediate nucleus (VIM) of the thalamus has been a revolutionary treatment for patients with essential tremor (ET) by reducing tremor. Unfortunately, some patients develop habituation to DBS and thus experience reduced efficacy and loss of tremor control. There are no standardized methods of addressing habituation to DBS. We propose alternating stimulation patterns as a way to reduce habituation. Methods: This was a randomized, placebo‐controlled trial for patients with VIM DBS for ET. Patients were randomized to either experimental treatment arm of alternating stimulation patterns on a weekly basis or standard care arm of continuous stimulation settings for 12 weeks. Primary outcome was change in the performance subscale of The Essential Tremor Rating Assessment Scale (TETRAS), which was performed at initial visit and 12‐week follow‐up. Secondary outcome included change in the activities of daily living subscale of TETRAS. Results: Twenty‐two patients were enrolled in the trial, and 16 were analyzed at follow‐up. Experimental treatment subjects displayed sustained tremor control compared to standard care, as measured by the change in TETRAS performance subscale (–0.6 vs. 6.7 point change, respectively) with a 7.3 difference between the arms (P = 0.006). Conclusion: Alternating stimulation patterns on a weekly basis for ET patients with VIM DBS reduced habituation in this pilot study. In April 2001, a rock fall occurred at the landslide Lärchberg- Galgenwald, Austria. The movement of this landslide has been under observation for decades, but the methods used so far have several disadvantages. This article aims to show that unmanned aerial systems (UAS) provide a reasonable alternative in terms for monitoring the landslide. At the landslide, we focused on the most active area to (i) record changes of the landslide's surface, (ii) assess the accuracy of the resulting data, (iii) assess whether the number of ground control points (GCPs) could be reduced in subsequent UAS surveys, and (iv) conclude whether UAS-based recording is favourable compared to common terrestrial methods. UAS imagery, acquired under challenging site conditions in November 2015 and May 2016, were processed to digital elevation models and orthophotos with a ground sampling distance of 0.04 m, which should display expected changes of up to ~0.17 m (relating to previously reported movements of up to 30 cm a-1) for the same period of time. Using DEM differencing, we calculated a vertical difference that was in the range of a few centimetres in most cases with only a few localized areas of much larger change. The latter are mainly related to single objects moving such as rock blocks or tree trunks. Apart from that, no general surface elevation lowering (a threshold of 0.27 m was used to distinguish significant from insignificant changes) could be detected. Based on the resulting orthophotos, we calculated vectors of horizontal displacement, which confirmed that the changes are related to single objects and revealed that in upcoming flight campaigns the number of GCPs can be generously reduced with an acceptable loss of positional accuracy. Summary: The FRAX algorithm assesses the patient’s probability of sustaining an osteoporotic fracture and can be calculated with or without densitometric data. This study seeks to determine whether in men, FRAX scores calculated without BMD, correctly identify patients with BMD-defined osteoporosis.Introduction: The diagnosis of osteoporosis is based on densitometric data, the presence of a fragility fracture or increased fracture risk. The FRAX algorithm estimates the patient’s 10-year probability of sustaining an osteoporotic fracture and can be calculated with or without BMD data. The purpose of this study is to determine whether in men, FRAX calculated without BMD, can correctly identify patients with BMD-defined osteoporosis.Methods: Retrospectively retrieved data from 726 consecutive Caucasian males, 50 to 70 years old referred to our Osteoporosis Center.Results: In the population studied, 11.8 and 25.3% had BMD-defined osteoporosis when female and male reference populations were used respectively. When the National Osteoporosis Foundation thresholds to initiate treatment are used, only 27% of patients with BMD-defined osteoporosis, but 4% with normal BMD reached/exceeded these thresholds. Lowering the threshold increased sensitivity, but decreased specificity.Conclusions: Our results suggest that FRAX without BMD is not sensitive/specific enough to be used to identify Caucasian men 50 to 70 years old with BMD-defined osteoporosis. Energy systems with a high share of renewable electricity generation face a challenge associated with surplus electricity generation. Flexibility options may help to integrate renewable electricity and reduce the need for electricity from fossil fuels. Load shifting is one of the flexibility options that lead to residual load smoothing in energy systems. In this paper, the effects of load shifting by dynamic electricity purchase on future residual load smoothing was assessed using wastewater treatment plants (WWTPs) in Germany as a case study. As dynamic electricity purchase is motivated economically, a consumer’s demand for supplying flexibility options needs a price signal. Therefore, electricity wholesale prices on an hourly basis were modeled for 2030 using a merit order simulation for two scenarios. Relevant data for WWTPs was obtained from primary data suppliers, which was then aggregated. The effects on residual load smoothing were assessed by optimizing WWTPs electricity purchase costs on a daily basis. Our results show that German WWTPs have a potential to integrate 120 MW el of surplus electric power. The developed methodology has a high potential to be applied to other potential load shifting actors when assessing the potential of residual load smoothing and renewable electricity integration. Pregnancy does not often occur in the setting of Parkinson's disease (PD) as the most common age of onset is beyond the childbearing years, yet management of these two conditions is crucial for the health of both mother and child. Here we review treatment data of PD during pregnancy, primarily from case reports and drug registries, and focus on available evidence regarding the pregnancy risks for patient and fetus. Historically, it was reported that many women had worsening of symptoms during pregnancy but this may be because anti-parkinsonian medications were not recommended or were under dosed. Levodopa has the best safety data for use in pregnancy and amantadine should be avoided in women who are pregnant or trying to become pregnant. The data for other pharmacological and surgical treatments is less clear. There is no evidence that women with PD have higher rates of birth or fetal complications. [ABSTRACT FROM AUTHOR]

Glacier-related applications of unmanned aircraft systems (UAS) in high mountain regions with steep topography are relatively rare. This study makes a contribution to the lack of UAS applications in studying alpine glaciers in the European Alps. We transferred an established workflow of UAS-based change detection procedures to Austria's largest glacier, the Pasterze Glacier. We focused on a selected part of the glacier tongue and its proglacial vicinity to obtain detailed knowledge of (i) the behavior of a lateral crevasse field, (ii) the evolution of glacier surface structures and velocity fields, (iii) glacier ablation behavior and the current glacier margin, and (iv) proglacial dead ice conditions and dead ice ablation. Based on two UAS flight campaigns, accomplished in 2016 (51 days apart), we produced digital elevation models (DEMs) and orthophotos with a ground sampling distance (GSD) of 0.15 m using Structure-from-Motion (SfM) photogrammetry. Electrical resistivity tomography (ERT) profiling was additionally conducted in the proglacial area. Results indicate distinct changes in the crevasse field with massive ice collapses, rapid glacier recession, surface lowering (mean of --0.9 m), and ice disintegration at the margins, calculated degree day factors on the order of --7 to --11 mm d--1.°C--1 for clean ice parts, and minimal changes of the debris-covered dead ice in the proglacial area. Flood events often endangered and damaged the small town of Oberwölz (Styria, Austria) and its nearby surroundings. In 2011 the last bigger flood occurred. After that, the main watercourse, the Schöttlbach creek, became a centre of a national funded research project. A wide variety of methods were applied together with intra- and extra-universitarian project partners to investigate that creek from a fluvial-geomorphological point of view. Apart from hydrological, meteorological, and geophysical methods, both, terrestrial and airborne remote-sensing techniques were applied. In particular, terrestrial laser scanning data as well as terrestrial and aerial photographs were frequently recorded since 2012. On the one hand the terrestrial monitoring focused on some representative erosion sites to quantify detailed erosion rates. On the other hand recording the sediment catchment output at the flood detention basin was intended. The aerial-based monitoring included three flight campaigns using mainly a fixed-wing unmanned aerial vehicle to cover approximately an area of about 170 ha that corresponds roughly to the major parts of the Schöttlbach creek. The main focus of the study lies on the adequate processing, georeferencing, and qualitative and quantitative comparison of the recorded elevation data. This was primarily done by using structure from motion photogrammetry and geodetic measurements. The resulting point clouds, digital elevation models, and orthophotos allow a visualization and quantification of morphodynamic processes. As a main outcome of this study, the combined usage of the mentioned methods can be seen. In this way, data recording from diverse perspectives is possible and allows a consideration of the varying terrain inclination. In addition, the significance of the resulting data for a geomorphological interpretation was assessed by describing the uncertainties. Testosterone has in recent years been proven essential for normal growth and maturation of small growing follicles. Concomitantly, low functional ovarian reserve (LFOR), characterized by a small growing follicle pool, has been associated with low testosterone levels, which can be of ovarian and/or adrenal origin. In this study we, therefore, investigated whether peripheral sex steroid precursors and testosterone levels potentially reflect on adrenal function. In a retrospective cohort study of 355 consecutive infertile women, who presented to an academically affiliated fertility center in New York City, we investigated in a series of statistical models whether low peripheral sex steroid precursors and testosterone are associated with peripheral cortisol (C) levels, reflecting adrenal function. To determine potential correlations, we investigated the dehydroepiandrosterone (DHEA), DHEA sulfate (DHEAS), androstenedione (AD), total testosterone (TT), free testosterone (FT); sex hormone binding globulin (SHBG), anti-Müllerian hormone (AMH), thyroid stimulating hormone (TSH) and C in a series of multivariate and logistic regression analyses, utilizing C either as a continuous variable or with cut off <5.0 μg/dL, and TT only as a continuous variable. Practically all models demonstrated significant predictability of peripheral sex hormone precursors for C levels, with DHEA demonstrating the strongest and most consistent predictability as an individual parameter and as part of the DHEAS/DHEA ratio. We conclude that in infertile women peripheral sex hormone precursors, especially DHEA, reflect C levels and, therefore, adrenal function. Objectives: Methicillin-resistant Staphylococcus aureus (MRSA), in particular clonal complex (CC) 398, is increasingly found in livestock. Recently, MRSA CC30 was identified in Danish pigs. We determined the susceptibility of porcine S. aureus isolates of CC398 and CC30 to disinfectants used in pig farming (benzalkonium chloride, hydrogen peroxide, formaldehyde, sodium hypochlorite, and caustic soda). Furthermore, efflux pump activity, antimicrobial resistance profiles, hemolysis properties, and the presence of toxic shock syndrome toxin-1 (TSST-1) and Panton-Valentine Leukocidin (PVL)-encoding virulence factors were investigated. Methods: Susceptibilities to biocides and antimicrobial agents of 79 porcine S. aureus isolates were determined by the microdilution method. Isolates comprised 21 methicillin-sensitive S. aureus (MSSA) and 40 MRSA isolates belonging to CC398 and 13 MSSA and 5 MRSA isolates belonging to CC30. The presence of quaternary ammonium compound (QAC) resistance efflux pumps was analyzed using an ethidium bromide accumulation assay. The presence of qac resistance genes in active efflux pump positive isolates was determined by whole-genome sequencing data. All isolates were screened for lukPV and tst genes with PCR, and hemolytic activities were determined using an agar plate assay. Results: S. aureus isolates did not show reduced susceptibility to the biocides tested. However, the QAC resistance gene, qacG, was detected in three MRSA CC30 isolates and the qacC in one MRSA CC30 isolate. CC30 isolates were generally more susceptible to non-beta-lactam antibiotics than CC398. Isolates generally had low hemolytic activity and none encoded PVL or TSST-1. Conclusion: The presence of qac genes in European porcine S. aureus isolates and in livestock-associated MRSA CC30 is for the first time described in this study. Objectives Large amounts of biocides are used to reduce and control bacterial growth in the healthcare sector, food production and agriculture. This work explores the effect of subinhibitory concentrations of four commonly used biocides (ethanol, hydrogen peroxide, chlorhexidine digluconate and sodium hypochlorite) on the conjugative transposition of the mobile genetic element Tn916. Methods Conjugation assays were carried out between Bacillus subtilis strains. The donor containing Tn916 was pre-exposed to subinhibitory concentrations of each biocide for a defined length of time, which was determined by an analysis of the transcriptional response of the promoter upstream of tet(M) using β-glucuronidase reporter assays. Results Ethanol significantly (P = 0.01) increased the transfer of Tn916 by 5-fold, whereas hydrogen peroxide, chlorhexidine digluconate and sodium hypochlorite did not significantly affect the transfer frequency. Conclusions These results suggest that exposure to subinhibitory concentrations of ethanol may induce the transfer of Tn916-like elements and any resistance genes they contain. Objectives The aim of the study was to investigate the pharmacokinetic properties of insulin aspart ( IAsp) in three different concentrations given as a continuous subcutaneous insulin infusion ( CSII). Methods A randomized cross-over study was performed in pigs, where IAsp U200, U100 or U20 was given for 8 h with the same total dose. Six pigs were included and blood was sampled during the CSII and 3 h after. Key findings The half-life (t1/2) was 24.3 (range 17.3-41.3), 28.8 (range 19.6-54.3) and 23.6 (range 17.4-36.8) min for U200, U100 and U20, respectively. The area under the curve per dose ( AUC/ D) was determined to be 51.2 ± 19.5, 52.3 ± 12.5 and 51.6 ± 6.7 p m × min/kg for U200, U100 and U20, respectively. The steady state plasma concentration ( Css) was 57.5 ± 27.1, 54.3 ± 10.3 and 55.1 ± 8.0 p m (mean ± SD) for U200, U100 and U20, respectively. Time to steady state ( Tss) was 110 ± 36, 98 ± 48 and 90 ± 27 min for U200, U100 and U20, respectively. Cirsium arvense (Asteraceae) is known as creeping thistle in its native range in the UK and Canada thistle in its invasive North American range. Recently, the fungus Phoma macrostoma was registered in Canada as a bioherbicide in turfgrass, where it causes severe chlorosis (White Tip disease) and death of C. arvense and other broadleaved weeds. It was hypothesised that the disease originated in the UK on its thistle host and, therefore, that fungal isolates from both countries should be biologically and genetically similar. Twenty-six strains in the genus Phoma- isolated during surveys in the UK - were compared morphologically with the type culture of P. macrostoma, tested for bioherbicidal activity using the inoculum mat bioassay and genetically screened with bioherbicide-specific primers. White tip disease was found to be restricted to the eastern and southern counties of England. Phoma macrostoma was isolated consistently from diseased bleached tissues. Bioherbicidal isolates of P. macrostoma occupy a unique clade, which is phylogenetically distinct but morphologically indistinguishable from the type culture. Most isolates from the UK had the same bioherbicidal activity and similar genetic make-up as strain SRC 94-44B, the active ingredient in the registered Canadian product. The origin of all bioherbicidal strains found to date has a clear presence in both Canada and the UK, with strong genetic similarities, supporting the view of a common ancestry. Thus, on the evidence presented, the 'white tip' clade of P. macrostoma evolved in southern England. Therefore, the bioherbicide based on strain SRC 94-44B should also be eligible for registration in the UK, based on the pest risk assessment data already available. [ABSTRACT FROM AUTHOR]

The accumulation of mutations causes cell lethality and can lead to carcinogenesis. An important class of mutations, which are associated with mutational hotspots in many organisms, are those that arise by nascent strand misalignment and template-switching at the site of short repetitive sequences in DNA. Mutagens that strongly and specifically affect this class, which is mechanistically distinct from other mutations that arise from polymerase errors or by DNA template damage, are unknown. Using Escherichia coli and assays for specific mutational events, this study defines such a mutagen, 3'-azidothymidine [zidovudine (AZT)], used widely in the treatment and prevention of HIV/AIDS. At sublethal doses, AZT has no significant effect on frame shifts and most base-substitution mutations. AT-to-CG transversions and deletions at microhomologies were enhanced modestly by AZT. AZT strongly stimulated the "template-switch" class of mutations that arise in imperfect inverted repeat sequences by DNA-strand misalignments during replication, presumably through its action as a chain terminator during DNA replication. Chain-terminating 2'-3'-didehydro 3'-deoxythymidine [stavudine (D4T)] and 2'-3'-dideoxyinosine [didanosine (ddI)] likewise stimulated template-switch mutagenesis. Animal models are important tools in the development of new drug candidates against the inflammatory bowel diseases (IBDs) Crohn's disease and ulcerative colitis. In order to increase the translational value of these models, it is important to increase knowledge relating to standard drugs. Using the SCID adoptive transfer colitis model, we have evaluated the effect of currently used IBD drugs and IBD drug candidates, that is, anti-TNF-α, TNFR-Fc, anti-IL-12p40, anti-IL-6, CTLA4-Ig, anti-α4β7 integrin, enrofloxacin/metronidazole, and cyclosporine.We found that anti-TNF-α, antibiotics, anti-IL-12p40, anti-α4β7 integrin, CTLA4- Ig, and anti-IL-6 effectively prevented onset of colitis, whereas TNFR-Fc and cyclosporine did not. In intervention studies, antibiotics, anti-IL-12p40, and CTLA4-Ig induced remission, whereas the other compounds did not. The data suggest that the adoptive transfer model and the inflammatory bowel diseases have some main inflammatory pathways in common. The finding that some well-established IBD therapeutics do not have any effect in the model highlights important differences between the experimental model and the human disease. Strand misalignments at DNA repeats during replication are implicated in mutational hotspots. To study these events, we have generated strains carrying mutations in the Escherichia coli chromosomal lacZ gene that revert via deletion of a short duplicated sequence or by template switching within imperfect inverted repeat (quasipalindrome, QP) sequences. Using these strains, we demonstrate that mutation of the distal repeat of a quasipalindrome, with respect to replication fork movement, is about 10-fold higher than the proximal repeat, consistent with more common template switching on the leading strand. The leading strand bias was lost in the absence of exonucleases I and VII, suggesting that it results from more efficient suppression of template switching by 39 exonucleases targeted to the lagging strand. The loss of 39 exonucleases has no effect on strand misalignment at direct repeats to produce deletion. To compare these events to other mutations, we have reengineered reporters (designed by Cupples and Miller 1989) that detect specific base substitutions or frameshifts in lacZ with the reverting lacZ locus on the chromosome rather than an F9 element. This set allows rapid screening of potential mutagens, environmental conditions, or genetic loci for effects on a broad set of mutational events. We found that hydroxyurea (HU), which depletes dNTP pools, slightly elevated templated mutations at inverted repeats but had no effect on deletions, simple frameshifts, or base substitutions. Mutations in nucleotide diphosphate kinase, ndk, signicantly elevated simple mutations but had little effect on the templated class. Zebularine, a cytosine analog, elevated all classes. Abstract: The taxonomic relationship between two microcyclic rust species, Puccinia melampodii and Puccinia xanthii, recorded from a number of Asteraceae hosts, was explored by comparing teliospore morphology, host specificity and sequence data. Teliospore morphology varied between and within individual rust accessions but, in general, teliospores of P. xanthii were larger than those of P. melampodii. Field observations and cross-inoculation greenhouse studies showed that P. melampodii accessions were highly specific to their original host species and unable to establish compatible interactions with related Asteraceae species. Though small amounts of genetic variation were detected between P. melampodii and P. xanthii accessions, the ITS and TEF phylogenetic analyses comprising several other rust species grouped these accessions within a well supported clade. Our data indicate that despite differences between P. xanthii and P. melampodii accessions these rusts cannot readily be separated at the species level, supporting earlier taxonomic conclusions and the recent proposal to group them into a morphospecies. Based on host-specificity data, the name Puccinia xanthii var. parthenii-hysterophorae is proposed to accommodate all Mesoamerican records of P. melampodii associated with the host Parthenium hysterophorus. Abstract: Enzyme-linked immunosorbent assay (ELISA) for measurement of antibodies to pertussis toxin (PT) is a widely used method for diagnosis of whooping cough and is also frequently used for seroprevalence studies and for assessment of antibodies after vaccinations against whooping cough. The recommended ELISA procedure for assessment of PT antibodies in human serum does, however, have a serious problem with false-positive results when heat-treated sera are analyzed. Historic sera might have been exposed to routine heat treatment, and diagnostic sera from warm geographic regions are at risk of unintentional exposure to heat. A modified version of the PT ELISA, incorporating a blocking step with 1% milk and addition of 0.1% milk to the dilution buffer, eliminates the false-positive phenomenon occurring with heat-treated sera. Results for serum antibody concentrations correlate well with the currently recommended method. This ELISA modification is straightforward and cheap, and it should be recommended at all analyses incorporating sera with unknown history of heat exposure. [Copyright &y& Elsevier]

Abstract: The aim of the current study was to evaluate the potential of the dynamic lipolysis model to simulate the absorption of a poorly soluble model drug compound, probucol, from three lipid-based formulations and to predict the in vitro–in vivo correlation (IVIVC) using neuro-fuzzy networks. An oil solution and two self-micro and nano-emulsifying drug delivery systems were tested in the lipolysis model. The release of probucol to the aqueous (micellar) phase was monitored during the progress of lipolysis. These release profiles compared with plasma profiles obtained in a previous bioavailability study conducted in mini-pigs at the same conditions. The release rate and extent of release from the oil formulation were found to be significantly lower than from SMEDDS and SNEDDS. The rank order of probucol released (SMEDDS∼SNEDDS>oil formulation) was similar to the rank order of bioavailability from the in vivo study. The employed neuro-fuzzy model (AFM-IVIVC) achieved significantly high prediction ability for different data formations (correlation greater than 0.91 and prediction error close to zero), without employing complex configurations. These preliminary results suggest that the dynamic lipolysis model combined with the AFM-IVIVC can be a useful tool in the prediction of the in vivo behavior of lipid-based formulations. Abstract: The usefulness of selected biorelevant dissolution media (BDM) to predict in vivo drug absorption was studied. Dissolution profiles of solid formulations of a poorly soluble model compound were compared in BDM simulating fasted and two levels of fed state. A non-physiologically relevant medium containing the cationic surfactant, cetrimide, was also investigated. All the media studied were capable of differentiating between the formulations employed, with formulation A consistently ranking high and formulations C and D ranking low. An in vivo dog study was carried out and an attempt was made to obtain a level A correlation between the plasma absorption curves and in vitro dissolution curves, using non-linear regression software. The in vitro–in vivo correlation (IVIVC) models developed indicated that fed state media (BDM 3) containing high levels of both bile salts (BS) and lipolysis products (LP) were best able to predict in vivo pharmacokinetic parameters (C max and AUC) with prediction errors lower than 10%. Overall, design and use of appropriate media for in vitro dissolution is extremely important. Abstract: The influence of droplet size on the absorption from lipid and surfactant based formulations was evaluated from two self-emulsifying formulations, a surfactant solution, and an oil solution. The self-emulsifying formulations was a self-emulsifying (SEDDS) and a self-nanoemulsifying (SNEDDS) formulation containing equal lipid and surfactant load, but exhibiting a large difference (approx 100 times) in the mean particle diameter of the resultant emulsion. The formulations were evaluated in a bioavailability study in fasted and fed Göttingen minipigs using probucol as model drug. In order to determine the bioavailability, an oil-in-water emulsion was included as i.v. formulation. The fasted group was fasted overnight and offered the first daily meal approx 4h after treatment. The fed group was offered the first daily meal (50% energy from fat) 30min prior to treatment. In the fasted group the SNEDDS exhibited a slightly faster absorption and higher bioavailability than the SEDDS, though non-significant. Furthermore, the bioavailability from the surfactant solution and the oil solution were slightly lower compared to the SNEDDS, indicating that both small particle size and digestibility are important in ensuring optimal bioavailability. Comparing the absorption in fasted and fed minipigs showed that probucol exhibited no significant food effect, when formulated in a lipid and surfactant based formulations. [Copyright &y& Elsevier]

Background Pure compounds and herbal medicines, had to be administered orally to vervet monkeys for testing. A reliable method was sought to avoid daily gavaging. Methods Substances were incorporated into the maize meal based diet of this facility for voluntary consumption. Food intake was monitored. Results In all but one case, consumption of the treated food was in excess of 90%. In all cases, the purpose of the study was achieved. Habituation was usually only required to the vehicle, and was accomplished by incremental increases to the desired concentration. Aversion was overcome by the addition of a small amount of honey. Plasma concentrations of a compound metabolite could be measured in one case after gavage and food consumption, and were similar for both methods. Conclusion The method enables reliable voluntary consumption of diverse substances. Since stressful gavaging is avoided, it is a valuable contribution to the 3Rs. [ABSTRACT FROM AUTHOR]

Abstract: The present work introduces a simple and robust in vitro method for enzymatic characterisation of surface properties of lipid dispersions in aqueous media. The initial lipolysis rate in biorelevant media, using pancreatic lipase and a self-microemulsifying formulation (SMEDDS) containing digestible lipids as substrate, was determined. The impact of incorporating two sparingly water soluble model drugs, probucol and halofantrine, into the SMEDDS was studied. It was found that both model drugs reduced the initial rate of lipolysis compared with the vehicle, probucol having a larger effect than halofantrine. The reduction of initial lipolysis rate indicates that probucol and halofantrine are bound in the water/emulsion interface limiting the substrate availability. [Copyright &y& Elsevier]

Abstract: Background: Peptides of the trefoil factor family (TFF1, TFF2 and TFF3) are cosecreted with mucus from mucus-producing cells in most organ systems and are believed to interact with mucus to form high-viscosity stable gel complexes. In the gastrointestinal tract, they sustain the mucosal barrier, and both injected and orally administered TFF peptide have protective and healing functions in the gastric mucosa. Aim: To investigate the possible treatment effect of luminally and parenterally administered TFF peptides in experimental colitis in rats. Methods: Colitis was induced by administration of 5% dextran sodium sulphate in the drinking water or by one intraperitoneal injection of mitomycin C, 3.75 mg/kg. TFF peptides were administered as subcutaneous injections or directly into the lumen via a catheter placed in the proximal colon. Treatments were saline, TFF2, TFF3 monomer or TFF3 dimer 5 mg/kg twice per day throughout the study [dextran sulphate sodium (DSS)] or from day 4 to 7 (mitomycin C). Colitis severity was scored in a stereomicroscope and histologically. Results: Luminal treatment with TFF3 in its dimeric form significantly improved the colitis score in both colitis models, whereas TFF2 had positive effect only in DSS-induced colitis. The TFF3 monomer was without any effects in both models. Treatment effect was most pronounced in the middle part of the colon, closest to the tip of the catheter. Injected TFF peptides, especially the TFF3 monomer, aggravated the colitis score in both colitis models. Conclusions: Intracolonic administration of TFF3 dimer and TFF2 improves experimentally induced colitis in rats. The TFF3 monomer has no effect. Background: Primates reared in captivity may display stereotypic behaviors. These behaviors are arguably reminiscent of human obsessive–compulsive or posttraumatic symptoms, which respond to selective serotonin reuptake inhibitors (SSRIs). Captive primates with marked stereotypic behaviors were entered into a randomized controlled study of the SSRI, fluoxetine. Methods: A sample of 10 vervet monkeys with behaviors such as marked saluting, somersaulting, weaving, or head tossing was selected. Subjects were randomized to receive fluoxetine 1 mg/kg for 6 weeks (n=5) or no treatment (n=5). A rater blind to the medication status of subjects noted the frequency of the stereotypic behaviors. Results: Repeated-measures analysis of variance (RM-ANOVA) demonstrated a significant Group×Time difference with significantly fewer stereotypic symptoms in the fluoxetine group by endpoint. At this time, three of the five fluoxetine-treated subjects (but none of the no-treatment subjects) were responders on the Clinical Global Impressions (CGI) change item (CGI ≤2). Conclusions: Stereotypic behaviors in captive vervets gradually and partially decrease in response to administration of an SSRI, paralleling research on human anxiety symptoms. Further research on animal stereotypies may be useful in providing appropriate veterinary care, and in exploring the underlying neurobiology of certain psychiatric disorders. The expression of endometrial β3 integrin and insulin-like growth factor binding protein-1 (IGFBP-1) was studied in cycling and pregnant vervet monkeys. There were clear changes of β3 integrin expression during the menstrual cycle, with the strongest immunostaining observed on day 26. Moderate to strong expression was observed during pregnancy. The expression of IGFBP-1 during the menstrual cycle was weak but upregulated during pregnancy with moderate to strong staining. The administration of a single dose of onapristone at 10 mg/kg on days 17, 21 and 22 of the menstrual cycle, followed by a biopsy on days 22, 22 and 26, respectively, and during pregnancy (34-44 days menstrual age) 24 h before the biopsy, disrupted and desynchronized the endometrium. However, no effect on β3 integrin expression could be observed and staining reflected the untreated patterns. The same applied to IGFBP-1 except that during pregnancy the expression of this protein was reduced or abolished. The results suggest that β3 integrin is associated with endometrial receptivity in vervet monkeys and that IGFBP-1 plays an important role during pregnancy in this species. The administration of onapristone appeared to only influence IGFBP-1 expression. To our knowledge, this is the first time that these endometrial proteins have been investigated in vervet monkeys. During a period of 6 years, uni- or bilateral cataracts occurred in 55 captive-bred vervet monkeys, representing 27.6% of all offspring produced in that time. Fifty-eight percent of all cases were detectable only by ophthalmoscopy. A total of 30% of cases were offspring of wild-caught parents, 40% from first-generation captive-bred parents, 4% from second-generation captive-bred parents, and 26% from parents of which one was wild caught. Cataracts that could be observed macroscopically appeared at about 6–8 mo of age. A sample of juvenile vervet monkeys with cataracts and their parents had no antibodies against rubella virus, herpes simplex virus, or toxoplasma gondi , and all except two wild-caught breeding females had no antibodies against cytomegalovirus. Plasma vitamin D3 (25 [OH] D), calcium, phosphorus, and glucose concentrations of six cataract cases were comparable to a control group of healthy individuals. The exclusion of these extraneous factors, as well as the fact that the majority of all cases were related, was considered evidence of hereditary etiology. Males and females that had either produced offspring with cataracts or were related to individuals with cataracts were replaced. Among the diverse facets of sperm maturation, changes in motility are conspicuous and hence studies of sperm kinematics might provide good indices for sperm maturation. Accordingly, the maturation of sperm motility in the epididymis and vas deferens of the vervet monkey, Cercopithecus aethiops, was assessed using a computer-aided sperm motility analysis system. The results revealed clear trends in the development of both sperm motility per se and in the movement characteristics of motile spermatozoa from different regions of the epididymis, the vas deferens and the ejaculate, reflecting maturational changes associated with the attainment of functional motility and fertility. Motion of spermatozoa from the caput epididymis was sluggish and irregular. As the spermatozoa moved through the corpus epididymis, motility increased sharply, and continued to improve through the cauda epididymis and vas deferens. Despite the high proportion of motile cells, full maturation of motion capabilities was not completed in spermatozoa from the corpus epididymis. Only once spermatozoa had reached the cauda epididymis and vas deferens did they attain their full vigour, and swam rapidly (greater VCL, VSL and VAP) with straightline trajectories (greater LIN, WOB and STR; lower ALH, MAD and CURV). After acquiring their maximal percentage motility and progressive velocity in the cauda epididymis and vas deferens, a slight decline in motility and vigour occurred in ejaculated spermatozoa, and was possibly associated with the ageing of stored spermatozoa. The results from this investigation have revealed clear trends in the maturation of the motility of vervet monkey spermatozoa during their transit through the epididymis and vas deferens and final emergence in the ejaculate, and have provided crucial baseline information on the reproductive physiology of this potentially valuable biomedical model to serve as a reference for future studies in reproductive toxicology.
