In shallow tropical waters, ultraviolet radiation (UVR) occurs at high intensity simultaneously with high water temperatures, and both stressors are predicted to increase in the future and to have a major impact on reef coral survival. The poor knowledge of the interactive effects of those two factors, however, prevents a good estimation of the coral resistance to climate change pressure. The results obtained in this study on two genetically distant scleractinian coral species, Pocillopora damicornis and Turbinaria reniformis, highlight an interaction of temperature and UVR on the corals' physiology as well as a species-specific response. Overall, increase in UVR level exacerbated the negative impact of thermal stress. Whereas stressors did not reduce the autotrophic capacity (symbiont density, net photosynthetic rates) of T. reniformis, they significantly lowered it in P. damicornis, which initially contained twice more symbionts in its tissue. Only grazing rates (heterotrophy) were significantly decreased in T. reniformis under thermal stress. For both corals, calcification was slowed by the combination of thermal and UVR stress. Calcification was impaired likely due to a decrease in autotrophic energy supply in P. damicornis, and in heterotrophic energy supply in T. reniformis. Photosynthetic organisms have developed various photoprotective mechanisms to cope with exposure to high light intensities. In photosynthetic dinoflagellates that live in symbiosis with cnidarians, the nature and relative amplitude of these regulatory mechanisms are a matter of debate. In our study, the amplitude of photosynthetic alternative electron flows ( AEF) to oxygen (chlororespiration, Mehler reaction), the mitochondrial respiration and the Photosystem I ( PSI) cyclic electron flow were investigated in strains belonging to three clades (A1, B1 and F1) of Symbiodinium., Cultured S ymbiodinium strains were maintained under identical environmental conditions, and measurements of oxygen evolution, fluorescence emission and absorption changes at specific wavelengths were used to evaluate PSI and PSII electron transfer rates ( ETR)., A light- and O2-dependent ETR was observed in all strains. This electron transfer chain involves PSII and PSI and is insensitive to inhibitors of mitochondrial activity and carbon fixation., We demonstrate that in all strains, the Mehler reaction responsible for photoreduction of oxygen by the PSI under high light, is the main AEF at the onset and at the steady state of photosynthesis. This sustained photosynthetic AEF under high light intensities acts as a photoprotective mechanism and leads to an increase of the ATP/ NADPH ratio. [ABSTRACT FROM AUTHOR]

Corals exposed to environmental stresses need to engage appropriate physiological strategies to survive. Here, we examined tissue modifications following algal dysfunction. Aiptasia pallida was exposed during 1 week to 3-(3,4-dichlorophenyl)-1,1-dimethylurea (DCMU), the herbicide called diuron. DCMU treatment produced a drastic loss in photosynthetic efficiency and a subsequent diminution of algae density over the following days. Cell proliferation evaluated by measuring the number of cells labeled with a thymidine analogue (EdU) revealed a significant increase in EdU+ cells in the epidermis after 1 week of DCMU incubation and in the gastrodermis at 4 weeks. TUNEL histology showed that the extent of cell death was, however, similar in the epidermis of control and treated specimens. In addition, we noticed a significant effect of DCMU treatment on the density of epidermal mucocytes after 1, 2 and 4 weeks. These results show that inhibition of Symbiodinium photosynthesis in the absence of any known direct effect of DCMU on host cells can induce an increase in epidermal host cell proliferation in both the epidermis and the gastrodermis. While new host gastrodermal cells are likely to promote tissue regeneration in order to recruit new algae, the new host epidermal cells may contribute to tissue adaptation following a decrease in energy income. Some of these new epidermal cells, such as mucocytes, may contribute to an eventual increase in the host heterotrophic ability until restoration of algal autotrophic contribution. Recovery of coral after bleaching episodes is a critical period for the health of the reef ecosystem. While events such as symbiont (genus Symbiodinium) shifting/shuffling or tissue apoptosis have been demonstrated to occur following bleaching, little is known concerning tissue recovery or cell proliferation. Here, we studied the sea anemone Aiptasia pallida exposed to a transient elevation of water temperature combined with high illumination (33°C and 1900 µmolphotons.m−2.s−1 for 30h). Following such treatment bleached anemones showed a significant reduction of their Symbiodinium density. Cell proliferation in the ectodermis and gastrodermis was determined by assessing the densities of cells labeled with a thymidine analogue (EdU). Cell proliferation significantly increased during the first day following stress in both tissue types. This increased cell proliferation returned to pre-stress values after one week. Although cell proliferation was higher in the ectodermis in absence of stress, it was relatively more pronounced in the gastrodermis of stressed anemones. In addition, the ratio of ectodermal mucocytes significantly increased three weeks after induced stress. These results suggest that thermal/photic stress coupled with the loss of the symbionts is able to enhance cell proliferation in both gastrodermis and ectodermis of cnidarians. While new cells formed in the gastrodermis are likely to host new Symbiodinium, the fate of new cells in the ectodermis was only partially revealed. Some new ectodermal cells may, in part, contribute to the increased number of mucocytes which could eventually help strengthen the heterotrophic state until restoration of the symbiosis. Abstract: The symbiosis between cnidarians and Symbiodinium algae (dinoflagellates) is the keystone responsible for the formation of the huge and important structures that are coral reefs. Today many environmental and/or anthropogenic threats compromise this tight relationship and lead to more frequent events of drastic loss of Symbiodinium pigments and eventually of algae themselves from cnidarians, better known as cnidarian bleaching. While the mechanisms underlying the collapse of the algae–coral symbiosis are progressively getting unraveled, the understanding of the mechanisms involved in the de novo infection of bleached cnidarians by Symbiodinium remains elusive. In this review, we describe the various steps needed to establish a stable symbiotic relationship between Symbiodinium and cnidarians. We review the mechanisms implicated in host–symbiont recognition and in symbiosome formation and persistence, with a special emphasis on the role played by lectins and Rab proteins. A better understanding of these molecular mechanisms may contribute to the development of strategies to promote post-bleaching recovery of corals. [Copyright &y& Elsevier]

The mutualistic relationship existing between scleractinian corals and their photosynthetic endosymbionts involves a complex integration of the metabolic pathways within the holobiont. Respiration and photosynthesis are the most important of these processes and although they have been extensively studied, our understanding of their interactions and regulatory mechanisms is still limited. In this work we performed chlorophyll-a fluorescence, oxygen exchange and time-resolved absorption spectroscopy measurements on small and thin fragments (0.3 cm2) of the coral Stylophora pistillata. We showed that the capacity of mitochondrial alternative oxidase accounted for ca. 25% of total coral respiration, and that the high-light dependent oxygen uptake, commonly present in isolated Symbiodiniaceae, was negligible. The ratio between photosystem I (PSI) and photosystem II (PSII) active centers as well as their respective electron transport rates, indicated that PSI cyclic electron flow occurred in high light in S. pistillata and in some branching and lamellar coral species freshly collected in the field. Altogether, these results show the potential of applying advanced biophysical and spectroscopic methods on small coral fragments to understand the complex mechanisms of coral photosynthesis and respiration and their responses to environmental changes. The sea anemone Entacmaea medusivora (Actiniaria, Anthozoa) commonly feeds on the golden jellyfish Mastigias papua (Rhizostomeae, Scyphozoa) which harbours an endosymbiotic dinoflagellate of the genus Cladocopium (Symbiodiniaceae). In this study, we monitored the photosynthetic activity of the endosymbiotic microalgae while their host jellyfish were ingested and digested by starved medusivorous anemones. By analyzing the photosynthetic yield of photosystem II, we observed that Cladocopium cells remain photosynthetically competent during the whole digestion process, thus confirming the exceptional resistance of Symbiodiniaceae to digestive enzymes. In the gastric cavity of E. medusivora , Cladocopium cells release oxygen, which could broadly stimulate the gastric microbiotic flora of the sea anemone. Ultimately, E. medusivora is not able to retain Cladocopium cells more than few days and physiologically-unaltered cells are therefore expelled in faecal pellets. The potential contribution of E. medusivora to maintain a reservoir of Cladocopium symbionts and its role in the life cycle of M. papua is discussed. [ABSTRACT FROM AUTHOR]
