A model is developed to test the hypothesis that kimberlites can form by low-degree melting of asthenospheric mantle followed by entrainment and assimilation of lithospheric mantle. The developed model uses inversion calculations based upon rare earth and compatible trace elements. For kimberlites (s.s.), an equation describing mass balance between a melt of unknown composition and a contaminant end-member of xenocrystic/assimilated material from the lithospheric mantle is inverted. This allows calculation of the mass fraction of xenocrystic minerals from the lithospheric mantle (olivine, orthopyroxene, clinopyroxene, garnet, ilmenite) entrained in the kimberlitic magma, as well as the source mineralogy and melt degree in the source region. The composition of the parental melt prior to interaction with the lithosphere is not assumed a priori but is calculated by the model. The CO2, H2O, K2O and P2O5 contents of the source are estimated assuming batch melting and the inversion models. The range and coupling of the model parameters are found using a non-linear most-squares inversion procedure, and the model space is visualised using a Self-Organising Map approach. Our earlier work supporting assimilation of xenocrystic opx is, however, not a precondition but provides a post-processing constraint , as well as the selection of a more likely set of solutions from the Self-Organising Map. The calculation is applied to a data set from the Majuagaa kimberlite dyke (southern West Greenland) including added whole rock analyses for CO2 and H2O. Major variations in whole rock compositions are related to flow differentiation of olivine macrocrysts. The textures of opx, cpx, gt and ilm megacrysts show evidence for reaction with the transporting melt and physical erosion in the kimberlitic mush. Using the bulk rocks in our inversion scheme results in a silico-carbonatite parental melt with major element concentrations consistent with experimental melts. The ol, opx, and cpx mass fractions in the source are not well-resolved by this calculation, but the proportion of gt in the source is comparatively well defined at 15–22 wt% and cpx is constrained to less than 14 wt%. The source assemblage required is 36–80 wt% ol, 2–49 wt% opx, 0–6 wt% cpx, and 15–19 wt% gt. This suggests a peridotitic rather than an eclogitic source. The inversion model gives an overall mass fraction of xenocrystic material in the Majuagaa kimberlite magma of 41–51 wt% The mass fractions of the xenocryst phases are as follows: 71–85 wt% ol, 0–13 wt% opx, 5 ± 1 wt% gt, and 10–14 wt% ilm. There is less than 3 wt% cpx in the xenocrystic and assimilated assemblage. These results agree with petrographic observations. Processing the model results using the Self-Organising Map clearly displays the extent and coupling within the statistically acceptable region of the model space and leads us to a preferred model of 49 wt% xenocrysts with a xenocryst assemblage of 71–76 wt% ol, 8–13 wt% opx, 4 wt% gt and 12 wt% ilm. A source with a REE pattern similar to that of primitive mantle is sufficient to form the parental melt and consistent with generation of the initial kimberlite melt in the convecting mantle. Calculated CO2 and H2O concentrations in the source of the Majuagaa kimberlite of 230–860 μg/g and 223–741 μg/g, respectively, are within the range of independent convecting mantle estimates. This is equivalent to <0.17 wt% magnesite and the H2O budget of the mantle source can be accommodated via storage in nominally anhydrous silicate phases. When applied to Majuagaa kimberlite, the inversions are consistent with a conceptually simple model of kimberlite formation: (1) low degree melting in carbonated asthenospheric peridotite, (2) melt extraction and concentration, and (3) entrainment and reaction with lithospheric mantle material. Lung cancer is the leading cause of cancer-related death in all Nordic countries which, though similar in demographics and healthcare systems, have noticeable differences in lung cancer survival. Historically, Denmark and Finland have had higher lung cancer incidences and lower survival than Norway and Sweden. All four countries have national cancer registries. Data in these registries are often compared, but their full potential as a source of learning across the Nordic countries is impeded by differences between the registries. In this paper, we describe and compare the Nordic registries on lung cancer–specific data and discuss how a more harmonized registration practice could increase their usefulness as a source for mutual learning and quality improvements. We describe and compare the characteristics of data on lung cancer cases from registries in Denmark, Finland, Norway and Sweden. Moreover, we compare the results from the latest annual reports and specify how data may be acquired from the registries for research. Denmark has a separate clinical lung cancer registry with more detailed data than the other Nordic countries. Finland and Norway report lung cancer survival as relative survival, whereas Denmark and Sweden report overall survival. The Danish Lung Cancer Registry and the Swedish Cancer Registry do not receive data from the Cause of Death registries in contrast to the Finnish Cancer Registry and the Cancer Registry of Norway. The lung cancer registries in Denmark, Finland, Norway and Sweden have high level of completeness. However, several important differences between the registries may bias comparative analyses. Helminthic therapy of immune-mediated diseases has gained attention in recent years, but we know little of how helminths modulate human immunity. In this study, we investigated how self-infection with Trichuris (T.) trichiura in an adult man without intestinal disease affected mucosal and systemic immunity. Colonic mucosal biopsies were obtained at baseline, during T. trichiura infection, and after its clearance following mebendazole treatment. Unexpectedly, the volunteer experienced a Campylobacter colitis following T. trichiura clearance, and this served as a positive infectious control. Trichuris trichiura colonization induced equally increased expressions of T-helper (h)1-, Th2-, Th17- and Treg-associated cytokines and transcription factors, measured by quantitative polymerase chain reaction. We observed several indicators of modulation of systemic immunity during the T. trichiura infection. Plasma eosinophils and anti- Trichuris antibodies rose markedly during the inoculation phase, and a shift towards a Th2-dominated T cell response at the expense of the Th1-response was observed in circulating T cells. During a severe outbreak of diarrhoea and vomiting in a pig herd in Central Eastern Europe, faecal samples were tested positive for porcine epidemic diarrhoea virus ( PEDV) and negative for transmissible gastroenteritis virus ( TGEV) using a commercial RT- qPCR assay that can detect both of these coronaviruses. However, further analyses, using other TGEV- and PEDV-specific RT- qPCR assays, provided results inconsistent with infection by either of these viruses. Sequencing of an amplicon (ca. 1.6 kb), generated by an RT- PCR specific for the PEDV S-gene, indicated a very close similarity (ca. 99% identity) to recently described chimeric viruses termed swine enteric coronaviruses (SeCoVs). These viruses (with an RNA genome of ca. 28 kb) were first identified in Italy in samples from 2009 but have not been detected there since 2012. A closely related virus was detected in archived samples in Germany from 2012, but has not been detected subsequently. Building on the initial sequence data, further amplicons were generated and over 9 kb of sequence corresponding to the 3′-terminus of the new SeCoV genome was determined. Sequence comparisons showed that the three known SeCoVs are ≥98% identical across this region and contain the S-gene and 3a sequences from PEDV within a backbone of TGEV, but the viruses are clearly distinct from each other. It is demonstrated, for the first time, that pigs from within the SeCoV-infected herd seroconverted against PEDV but tested negative in a TGEV-specific ELISA that detects antibodies against the S protein. These results indicate that SeCoV is continuing to circulate in Europe and suggest it can cause a disease that is very similar to PED. Specific detection of the chimeric SeCoVs either requires development of a new diagnostic RT- qPCR assay or the combined use of assays targeting the PEDV S-gene and another part of the TGEV genome. Radon-222 gas arises from the radioactive decay of radium-226 and has a half-life of 3.8 days. This gas percolates up through soil into buildings, and if it is not evacuated, there can be much higher exposure levels indoors than outdoors, which is where human exposure occurs. Radon exposure is classified as a human carcinogen, and new Danish homes must be constructed to ensure indoor radon levels below 100 Bq/m3. Our purpose was to assess how well 200 newly constructed single detached homes perform according to building regulations pertaining to radon and identify the association between indoor radon in these homes and municipality, home age, floor area, floor level, basement, and outer wall and roof construction. Median (5-95 percentile) indoor radon levels were 36.8 (9.0-118) Bq/m3, but indoor radon exceeded 100 Bq/m3 in 14 of these new homes. The investigated variables explained nine percent of the variation in indoor radon levels, and although associations were positive, none of these were statistically significant. In this study, radon levels were generally low, but we found that 14 (7%) of the 200 new homes had indoor radon levels over 100 Bq/m3. Background: Extremity injury with ischaemia is the most common pattern of vascular trauma and is a challenge for surgeons who must make decisions about the timing and mechanism of limb reperfusion. In modern military conflicts, effective use of limb tourniquets and rapid transport of the injured have increased the number of casualties who reach a medical service with potentially survivable vascular trauma. This report provides a review of extremity ischaemia and reperfusion following vascular trauma. Methods: A review was undertaken of extremity vascular injury with ischaemia, including a focus on adjuncts aimed at reducing reperfusion injury and improving neuromuscular recovery and limb salvage. Results: Findings from basic and clinical research support the need to restore perfusion to an ischaemic limb as soon as possible in order to achieve optimal neuromuscular recovery. Large-animal studies demonstrate that haemorrhagic shock worsens the impact of ischaemia on the neuromuscular structures of the limb and reduces the ischaemic threshold to as little as 1 h. Surgical adjuncts such as vascular shunts, fasciotomy, regional limb cooling and ischaemic conditioning may reduce the severity of ischaemic injury. Medical therapies have also been described including hypertonic saline, statins and ethyl pyruvate, which reduce the inflammatory response following limb reperfusion. Conclusion: Contemporary translational research refutes a casual approach to extremity vascular injury with ischaemia, instead emphasizing expedited reperfusion. Surgical and medical adjuncts exist to expedite reperfusion and mitigate reperfusion injury. Following the first ever case of bluetongue in Denmark during late 2007, further outbreaks were observed in Denmark during 2008, despite vaccination against bluetongue virus (BTV) serotype 8 (BTV-8) in the southern part of the country. In total, 15 separate outbreaks of infection were identified, mostly as a result of clinical suspicions but also because of surveillance of bulk milk samples. These outbreaks led to extensions of the original vaccination zone planned for 2008. Blood samples from clinical suspects were analysed using ELISA and real-time RT-PCR assays for the presence of anti-BTV antibodies and viral RNA, respectively. A newly infected calf from the primary outbreak in 2008 was studied for a period of three months, during which time it seroconverted to BTV, but the presence of viral RNA in its blood was maintained throughout this time. Each outbreak was caused by BTV-8, as determined by a serotype-specific real-time RT-PCR assay. Furthermore, the nucleotide sequence of a portion of segment 2 of the viral RNA (encoding the outer capsid protein VP2) from the samples analysed was identical to the BTV-8 segment 2 that circulated in the Netherlands during 2006. Coherent Raman detected electron spin resonance spectroscopy is a technique that bridges the established fields of magnetic resonance and magneto-optics. By exploiting the orientational selectivity of the microwave resonance condition it becomes possible to measure the relative orientations of the magnetic and optical anisotropies of paramagnetic chromophores, and thereby to test models of their electronic structure. This paper reports the application of this method to the CuA centre from Paracoccus pantotrophus nitrous oxide reductase, an unusual mixed valence copper, Cu(I)/Cu(II), dimer centre also found in some heme-copper terminal oxidases. Data from the principal visible bands (at 476, 514 and 750 nm) shows that their magnetic circular dichroism is almost entirely aligned with the g-value z-axis. This is consistent with previous models of the electronic structure in which the optical transitions are polarized within the copper-thiolate plane of the centre, and the g-value z-axis is orientated normal to this plane. Aim: To investigate the potential of quorum sensing inhibitors (QSI) as food preservative agents in a food product, where bacterial spoilage is controlled by quorum sensing (QS). Methods and Results: The effects of well-known QSI were tested on spoilage phenotypes and on QS-regulated genes of a bean sprout spoiling bacterial isolate ( Pectobacterium A2JM) in laboratory substrates and in a bean sprout model system. The acylated homoserine lactones (AHL) analogues PenS-AHL and HepS-AHL decreased the specific protease activity of Pectobacterium A2JM in broth but did not reduce the expression of a QS-regulated secretion protein, and were without effect on soft rot of bean sprouts. The QSI ProS-AHL, furanone C-30, patulin, penicillic acid and 4-nitropyridine- N-oxide did not have any effect on protease activity, on gene expression or bean sprout appearance at nongrowth inhibitory concentrations. Extracts from garlic and bean sprouts induced the QS system of Pectobacterium in bean sprouts and a broth system, respectively. Conclusions: Among the several well-known QSI compounds, only PenS-AHL and HepS-AHL, inhibited QS-regulated protease activity of Pectobacterium A2JM in broth cultures, but had no effect on bean sprout spoilage. Significance and Impact of the Study: The QSI compounds must be selected in the specific system in which they are to function and they cannot easily be transferred from one QS system to another. A real-time PCR assay based on primer-probe energy transfer (PriProET) was developed to detect swine vesicular disease virus (SVDV). Specificity tests of SVDV and heterologous virus showed specific amplification of SVDV strains only. The amplification plot for the closely related Coxsackievirus B5 remained negative. The sensitivity of assay was five copies of viral genome equivalents. A key point of the assay is tolerance toward mutations in the probe region. Melting curve analysis directly after PCR, with determination of probe melting point, confirmed specific hybridisation of the SVDV strains. Eight of twenty SVDV strains tested, revealed shifted melting points that indicated mutations in the probe region. All predicted mutations were confirmed by nucleotide sequencing. With the PriProET system there is a chance to identify phylogenetically divergent strains of SVDV, which may appear negative in other probe-based real-time PCR assays. At the same time, any difference in melting points may provide an indication of divergence in the probe region. The high sensitivity, specificity, and tolerance toward mutations in the probe region of the SVDV PriProET assay may improve the early and rapid detection of a wide range of SVDV strains, allowing reduced turnaround time and the use of high-throughput, automated technology. Multiple myeloma (MM) is an incurable B-cell malignancy characterized by accumulation of malignant plasma cells in bone marrow (BM) and recurrent or persistent infections. Toll-like receptors (TLRs) are essential in the host defense against infections and today 10 human TLRs (TLR1-TLR10) and one TLR-homolog (RP105) have been characterized. B cells express several TLRs (mainly TLR1, 6, 7, 9, 10 and RP105) and TLR-initiated responses in B cells include proliferation, anti-apoptosis effect and plasma cell (PC) differentiation. The present study was designed to analyze the role of TLRs in MM. We show that frequent expressions of TLRs were detected in cell lines from MM patients (minimum six TLRs in each). In comparison, only few TLRs (mainly TLR1 and or RP105) were found expressed in PCs from BM of healthy donors. In addition, TLR-specific ligands induce increased proliferation and survival of the MM cell lines, partially due to an autocrine interleukin-6 production. Importantly, we demonstrate that also PC from MM patients proliferates in response to TLR-specific ligands. Summary. Foot-and-mouth disease virus (FMDV) spreads extremely fast and the need for rapid and robust diagnostic virus detection systems was obvious during the recent European epidemic. Using a novel real-time RT-PCR system based on primer-probe energy transfer (PriProET) we present here an assay targeting the 3D gene of FMDV. The assay was validated for the efficacy to detect all known FMDV serotypes. The test method was linear over a range of at least 7 orders of magnitude and the detection limit was below the equivalent of 10 genomic copies. Analysing recent African probang samples the method was able to detect FMDV in materials from both cattle and buffalo. When compared to traditional virus cultivation the virus detection sensitivity was similar but the RT-PCR method can provide a laboratory result much faster than virus cultivation. The real-time PCR method confirms the identity of the amplicon by melting point analysis for added specificity and at the same time allows the detection of mutations in the probe region. As such, the described new method is suitable for the robust real-time detection of index cases caused by any serotype of FMDV. [ABSTRACT FROM AUTHOR]

The aim of this study was to examine proprioception with and without muscle tension and knee function in two groups of chronic (> 1 year post-injury) patients with deficient anterior cruciate ligaments with distinctly different functioning levels. The well (WF) and poor functioning (PF) group was defined as those with a Tegner/Lysholm score equal to or greater than 6/95 (WF, n = 7), and equal to or less than 4/83 (PF, n = 7), respectively. Clinical examination included a Lachman, and a Pivot shift test, as well as a KT2000 arthrometer assessment. Single and triple hop tests assessed one-legged performance. Proprioception was measured as threshold of passive movement detection, and as the ability to reproduce flexion angles with (20% and 50% MVC) and without muscle tension. Mean Tegner/Lysholm scores were: WF: 7 (range 6-8)/98 (range 95-100), PF: 2 (range 1-4)/71 (range 62-80). There were significant differences between the Pivot shift tests of the groups (p = 0.01). The laxity (KT2000) assessment yielded a significant (3 mm, p < 0.05) side-to-side difference in both groups. One-leg hop tests yielded no side-to-side differences in any of the groups or between the groups. There were no significant differences between the groups in any of the proprioceptive tests, except in one of the angle reproduction tests with muscle tension (20%, p < 0.05). WF had larger mean errors than PF. There was not found any side-to-side difference in any of the proprioceptive tests, except in one of the angle reproduction tests with muscle tension (WF, 20%, p < 0.05). In conclusion, these data suggest that subjects with long standing anterior cruciate ligament deficiency in the present study did not have a knee joint proprioceptive deficit as measured by some commonly accepted methods. We investigated the influence of electrical stimulation of the posterior cruciate ligament (PCL) on the motoneuron pool of the thigh and calf muscle during gait. The study group comprised eight young men without any history of injury to the knee joints. Multistranded teflon-insulated stainless steel wires were inserted into the PCL guided by sonography and in four subjects also into the fat pad of the knee. The PCL was electrically stimulated during gait on a treadmill at heel strike and 100 ms after heel strike. Electromyographic signals were recorded with bipolar surface electrodes placed over the vastus medialis, rectus femoris, vastus lateralis, biceps femoris caput longum, and semitendinosus muscles. The stimuli consisted of four pulses delivered at 200 Hz; the stimulus amplitude was two to three times the sensory threshold. The electrical stimulation of the PCL inhibited the ongoing muscle activity in both the quadriceps and the hamstrings. The latency of the inhibition ranged between 78 and 148 ms in the quadriceps, between 88 and 110 ms in the hamstrings and between 189 and 258 ms in m. gastrocnemius. Stimulation of the fat pad of the knee did not influence the thigh and calf muscle motoneuron pool as evidenced by electromyography. The response elicited from the stimulation of the PCL was not limited to a specific muscle group but depended on ongoing muscle contraction, which suggests that the mechanoreceptors in the PCL are involved in the control of all muscles acting on the knee joint during gait. We studied the effect of porcine CGRP (pCGRP) in concentrations from 10–10 to 10–8 mol L–1 on the motility and on the release of substance P, neurokinin A, somatostatin and gastrin in the antrum using the isolated perfused porcine antrum as experimental model. In addition, we studied the localization of CGRP by immunohistochemistry in the porcine antrum. CGRP-immunoreactive nerve fibres were found mainly in the submucous layer and in the external muscle coat, where they were seen in all layers, and in the ganglia of the myenteric nervous plexus. The frequency of contraction was significantly and dose-dependently increased from a basal level of 11.8 ± 0.5 contractions per 5 min to 24.4 ± 3.6 contractions per 5 min at pCGRP 10–8 mol L–1. At this dose, the release of substance P and neurokinin A was significantly increased to 470 ± 149% and 217 ± 26%, respectively, compared to basal release. The effect of pCGRP was unaffected by the addition of the nonpeptide antagonists for the NK-1 (CP-99994) and NK-2 receptors (SR48968), both at 10–6 mol L–1, whereas atropine (10–6 mol L–1) completely abolished the motor effect of pCGRP. The release of somatostatin was significantly increased by 154 ± 15% in response to CGRP at 10–8 mol L–1. The release of gastrin was unaffected by pCGRP. In conclusion, pCGRP increases contractile activity in the porcine antrum, an effect that involves cholinergic mechanisms but is independent of the release of substance P and neurokinin A. in addition, pCGRP increases the release of somatostatin but has no effect on gastrin release in the isolated perfused porcine antrum. We have performed atomic-scale simulations of cross-slip processes of screw dislocations in copper, simulating jog-free dislocations as well as different types of jogged screw dislocations. Minimum-energy paths and corresponding transition state energies are obtained using the nudged-elastic-band path technique. We find low barriers and effective masses for the conservative motion along the dislocations of elementary jogs on both ordinary {111}<110> and nonoctahedral {110}<110> slip systems. The jogs are found to be constricted and therefore effectively act as pre-existing constrictions; the cross-slip activation energy is thereby dramatically reduced, yielding values in agreement with experiment. Large scale atomistic simulations are used to investigate the properties of screw dislocation dipoles in copper. Spontaneous annihilation is observed for dipole heights less than 1 nm. Equilibrated dipoles of heights larger than 1 nm adopt a skew configuration due to the elastic anisotropy of Cu. The equilibrium splitting width of the screw dislocations decreases with decreasing dipole height, as expected from elasticity theory. The energy barriers, and corresponding transition states for annihilation of stable dipoles are determined for straight and for flexible dislocations for dipole heights up to 5.2 nm. In both cases the annihilation is initiated by cross-slip of one of the dislocations. For straight dislocations the activation energy shows a linear dependence on the inverse dipole height, and for flexible dislocations the dependence is roughly linear for the dipoles investigated. We studied the performance and proprioception of the knee joint in a group of non-reconstructed anterior cruclare ligament (ACL)-deficient (n=20) patients and compared them with a group of ACL-reconstructed patients (n= 18) and a group of healthy controls (n=20). Each patient was scored according to Lysholm and Tegner and was then asked to subjectively evaluate the performance of the injured knee and the degree of retropatellar discomfort. The knee joint laxity was measured. The performance was assessed based on the performance in a triple jump test and a one-leg one-step leap test. The proprioception in the knee was measured as the threshold when passive movement was detected and as the ability to reproduce a flexion angle from a start position of 60 degrees of flexion or from full extension of the knee. All tests were performed on both legs. The scoring systems and the subjective evaluation showed significant differences between the reconstructed and the non-reconstructed patients. No significant difference in knee joint laxity was found between the two groups. In the triple jump test and the one-step leap test, both groups performed significantly worse on the leg with the injured knee joint than on the non-injured leg, The proprioceptive tests showed decreased ability to recognize and reproduce a prior angle from a start position of 60°. The threshold to detection of passive movement with the injured knee was significantly increased in both groups of patients. No difference was found between the dominant and non-dominant knee in the control group. When reproduction of the same angles started from full extension, the groups did not differ. Objective: To assess whether maternal BMI before pregnancy and weight gain during pregnancy predicted the risk of islet autoimmunity in genetically susceptible children.Research Design and Methods: Of 46,939 newborns screened for the high-risk HLA genotype DR4-DQ8/DR3-DQ2, 1,003 were positive and 885 were followed with serial blood samples tested for autoantibodies to insulin, GAD, and insulinoma-associated protein 2 (IA2). The end point was defined as repeated positivity for two or three autoantibodies or the onset of type 1 diabetes (islet autoimmunity).Results: Thirty-six children developed islet autoimmunity, of whom 10 developed type 1 diabetes. Both maternal BMI > or =30 kg/m(2) before pregnancy and maternal weight gain > or =15 kg predicted the increased risk of islet autoimmunity (hazard ratio [HR] 2.5, P = 0.023, and HR 2.5, P = 0.015, respectively), independent of maternal diabetes.Conclusions: Maternal weight may predict risk of islet autoimmunity in offspring with a high genetic susceptibility for type 1 diabetes. Changes in cheese physical properties during aging are related to proteolysis by coagulant and culture enzymes. Storage temperature also affects rate of aging. Cultures are important for flavor development, but less is understood about their role in textural and melting properties. Our objective was to make cheddar cheese using different cultures, to age it at 5 and 13°C, and measure physical properties over 6 months. Cheese was manufactured using Lactococcus lactis starter culture either alone or combined with one or both of lac- Lc. lactis or Lactobacillus helveticus adjunct cultures. Three replicates of cheese were made using 1500 lb milk. Cheese composition was 35.5±1.0% moisture, 52.5±2.5% FDB, 1.65±0.05% salt, and pH 5.2±0.1. All cheeses were initially stored at 5°C, then half were moved to 13°C after 21 d. Texture profile analysis (TPA) was performed using 25% and 60% compression and melting measured using a Meltmeter at 65°C. After 2 months, differences in cheese properties were observed based on cultures and storage temperature. Cultures used had more effect on hardness and cohesiveness, while storage temperature had a large impact on adhesiveness. Using 25% or 60% compression produced different TPA values, especially when the cheese fractured during compression. At 25% compression, after 2 months, the cheeses made using Lb. helveticus had the highest hardness (av. 2365 g versus 1853 g), while the cheese stored at 13°C had more adhesiveness (av.18.3 versus 11.8). At 60% compression, the control cheese stored at 5°C had the highest hardness, as it was the only cheese that did not fracture during compression. Cheeses stored at 13°C melted faster (1.39 versus 1.29 mm/s) and further (82.5% versus 75.5% height reduction) than those stored at 5°C. Adding either adjunct cultures or raising the storage temperature can accelerate the aging of cheese thus changing its textural and melting properties. We investigated the temporal relationship of parental morbidity and mortality with offspring’s suicide attempt and whether any such association was modified by offspring’s age at attempt. We designed a matched case-control study through register linkage. Cases comprised all individuals in Sweden born 1973–1983 with inpatient care due to suicide attempt (15–31 years old, N = 15 193). Regression models were applied. Particularly for women, most parental markers showed the strongest effect sizes if exposure was short-term (within 2 years after exposure) and related to the mother. Regarding exposure to parental inpatient care due to psychiatric diagnoses, short-term as opposed to long-term (exceeding 2 years after exposure) effects were highest during adolescence and decreased significantly with age for female and male offspring, respectively. The results suggest that the high short-term risks of suicide attempt in case of exposure to parental psychopathology and suicidal behavior are of direct clinical importance. [ABSTRACT FROM AUTHOR]

Gas transport through sediments to the seabed and seepage occurs via advection through pores, faults, and fractures, and as solubility driven gas diffusion. The pore pressure gradient is a key factor in these processes. Yet, in situ measurements for quantitative studies of fluid dynamics and sediment deformation in deep ocean environments remain scarce. In this study, we integrate piezometer data, geotechnical tests, and sediment core analyses to study the pressure regime that controls gas transport along the Vestnesa Ridge in the eastern Fram Strait. The data show a progressive westward decrease in induced pore pressure (i.e., from c. 180 to c. 50 kPa) upon piezometer penetration and undrained shear strength of the sediments, interpreted as a decrease in sediment stiffness. In addition, the data suggest that the upper c. 6 m of sediments may be mechanically damaged due to variations in gas diffusion rates and exsolution. Background pore pressures are mostly at hydrostatic conditions, but localized excess pore pressures (i.e., up to 10 kPa) exist and point toward external controls. When analyzed in conjunction with observations from geophysical data and sediment core analyses, the pore pressure data suggest a spatial change from an advection dominated to a diffusion dominated fluid flow system, influenced by the behavior of sedimentary faults. Understanding gas transport mechanisms and their effect on fine‐grained sediments of deep ocean settings is critical for constraining gas hydrate inventories, seepage phenomena and sub‐seabed sediment deformations and instabilities. Plain Language Summary: Seafloor methane seepage occurs persistently in response to pressure changes. The scarcity of sub‐seabed pressure data limits our understanding of the mechanisms of methane release. Here we document observations from 4 days monitoring of pressure and temperature within the upper 10 m of sediment at a seepage site in the Fram Strait. The survey extends for 60 km between the continental shelf off west‐Svalbard and the mid‐ocean ridge. The data show that the geothermal gradient increases and that sub‐seabed sediments become softer and more susceptible to deformation as they approach the mid‐ocean ridge. When analyzed in conjunction with cross‐disciplinary data it seems plausible that the changes in sediment properties are associated with an increase in the amount of dissolved methane transported to the seafloor. We suggest that changes in the geological setting have resulted in the locking of fractures that would otherwise allow the release of trapped methane to the ocean. Trapped gas in the sediment pores is pumped by tides and causes damages beneath the seafloor. This study is an important step in understanding the mechanisms involved in the transport of methane into the ocean, with implications for climate research, assessment of geological hazards and alternative energy resources. Computed tomography (CT) examinations are increasingly used worldwide and incidental findings are growing likewise. Lung cancer stage at diagnosis is pivotal to survival. The earliest stage of lung cancer, stage IA is in most cases asymptomatic. Potentially, increased use of clinical CTs could induce a stage shift toward earlier lung cancer diagnosis. Data on the number of CT thorax in Denmark and the stage distribution of Danish lung cancer patients 2013–2020 were acquired from, respectively, the Danish Health Data Authority and the Danish Lung Cancer Registry. Clinical auditing of stage IA lung cancer patients was performed in the period 2019–2021 in a Danish region to assess the reasons for referral. Auditing of stage IV lung cancer patients was done to see whether a CT thorax was performed in a two-year period before diagnosis. All regions showed an increase in CTs per 1000 inhabitants. However, the number of CTs performed in 2013 differed by more than 50% among regions, and the increase per year also differed, from an increase of 1.9 to 3.4 more examinations per year. A significant correlation between CTs and fraction of stage IA lung cancers was seen in four out of the five regions. The audit of stage IA lung cancer cases revealed that 86.8% were incidental findings. Audit of stage IV lung cancer found that 4.3% had a nodule/infiltrate on a previous CT within a 2-year period prior to the diagnosis of lung cancer that was the probable origin of stage IV lung cancer. The study found that the vast majority of early-stage lung cancers were incidental findings. It highlights that follow-up algorithms of incidental findings should be used in accordance with guidelines and it should be unequivocally how the CT follow-up of pulmonary infiltrates is managed. MacLean (2012) Neuropathology and Applied Neurobiology 38, 162-174 S100β as a novel and accessible indicator for the presence of monocyte-driven encephalitis in AIDS Aims: The pathogenesis of human/simian immunodeficiency virus encephalitis (HIVE/SIVE) remains incompletely understood, but is associated with alterations in the blood-brain barrier. At present, it is not possible to easily determine if an individual has HIVE/SIVE before post mortem examination. Methods: We have examined serum levels of the astroglial protein S100β in SIV-infected macaques and show that it can be used to determine which animals have SIVE. We also checked for correlations with inflammatory markers such as CCL2/MCP-1, IL-6 and C-reactive protein. Results: We found that increased S100β protein in serum correlated with decreased expression of the tight junction protein zonula occludens-1 on brain microvessels. Furthermore, the decrease in zonula occludens-1 expression was spatially related to SIVE lesions and perivascular deposition of plasma fibrinogen. There was no correlation between encephalitis and plasma levels of IL-6, MCP-1/CCL2 or C-reactive protein. Conclusions: Together, these data indicate that SIVE lesions are associated with vascular leakage that can be determined by S100β protein in the periphery. Objective: To test whether the frequency of human enterovirus RNA in fecal samples collected monthly from early infancy was associated with development of multiple islet autoantibodies in children with the highest risk HLA genotype.Research Design and Methods: Individuals carrying the HLA DRB1*0401-DQA1*03-DQB1*0302/DRB1*03-DQA1*05-DQB1*02 genotype were identified at birth and followed with monthly stool samples from age 3 to 35 months. Blood samples taken at age 3, 6, 9, and 12 months and then annually were tested for autoantibodies to insulin, GAD 65 and IA-2. Among 911 children, 27 developed positivity for two or more islet autoantibodies in two or more consecutive samples (case subjects). Two control subjects per case subject were matched by follow-up time, date of birth, and county of residence. Stool samples were analyzed for enterovirus with a semiquantitative real-time RT-PCR.Results: The frequency of human enterovirus RNA in stool samples from case subjects before seroconversion (43 of 339, 12.7%) did not differ from the frequency in control subjects (94 of 692, 13.6%) (P = 0.97). Results remained essentially unchanged after adjustment for potential confounders, restriction to various time windows before seroconversion, or infections in the 1st year of life or after inclusion of samples collected after seroconversion. There was no difference in the average quantity of enterovirus RNA or in the frequency of repeatedly positive samples. The estimated relative risk for islet autoimmunity per enterovirus RNA-positive sample during follow-up (nested case-control analysis) was 1.12 (95% CI 0.66-1.91).Conclusions: There was no support for the hypothesis that fecal shedding of enteroviral RNA is a major predictor of advanced islet autoimmunity. STUDY DESIGN: A prospective randomized clinical study. OBJECTIVE: To compare 2 surgical methods in the treatment of cervical radiculopathy caused by hard or soft disc herniation; namely, simple discectomy versus discectomy with an additional interbody fusion with a Ray titanium cage. SUMMARY OF BACKGROUND DATA: Although an interbody fusion after anterior decompressive surgery for hard or soft disc herniation is widely accepted, there is no scientific evidence that convincingly demonstrates that insertion of graft material for interbody fusion is necessary after discectomy and decompression of the nervous elements have been performed. To date, no randomized studies have compared simple discectomy with discectomy followed by an interbody fusion with a titanium cage. METHODS: Eighty-six patients with symptoms of nerve root compression at 1 level were randomly allocated to either discectomy followed by fusion with a Ray titanium cage (40 patients) or to discectomy alone (46 patients). Clinical and radiologic follow-up was performed 3, 12, and 24 months after surgery. RESULTS: There was no statistically significant difference between the 2 groups concerning self-reported satisfaction or severity of pain in the neck and arm. Two years after the operation, 86.1% of the patients treated with cage stated a good outcome versus 76.7% in the discectomy group (P = 0.44). The rate of fusion was 83.3% in the cage group versus 81.0% in the discectomy group (P = 0.30). Furthermore, after 2 years, also the rates of new adjacent disc degeneration or spondylosis were the same in both groups. CONCLUSION: This study showed no statistically significant difference between simple discectomy and discectomy followed by interbody fusion with a titanium cage in the surgical treatment of cervical radiculopathy caused by disc herniation. In this study, short-term growth and feeding behaviour were compared among juvenile European flounder Platichthys flesus reared in enclosures in either their native habitat (bare sand or vegetation) or transferred to the opposite habitat. Growth was poorest in the vegetated habitat regardless of origin of the fish. The effect of the habitat shift differed between years. In 2000, the relatively small fish used grew fastest in their native habitat. In contrast, in 2001 when larger fish were used, growth was similar between native and introduced fish in the vegetated habitat, and introduced fish grew faster than native fish in the bare sand habitat. Diet composition and feeding intensity within a habitat were also similar among native and introduced fish in 2001, suggesting that the habitat switch had a minor influence on foraging efficiency. The different results obtained from the experiments in 2000 and 2001 suggest that fish size may determine the extent to which short-term habitat shifts influence feeding and growth in juvenile flounder, and, importantly, that the negative effects of habitat fragmentation are more severe for small compared to larger juvenile flounder. Many paleoceanographic reconstructions of the glacial North Atlantic include estimates of iceberg discharge, which are based on the variable abundance of ice-rafted detritus (IRD) in deep-sea sediments. IRD abundance is most often determined by the mechanical separation and painstaking counting of terrigenous particles larger than a specified threshold grain size, typically 150 μm. Here we present a new proxy for IRD based on X-ray diffraction (XRD) analysis of bulk sediments. This approach complements results obtained from standard techniques while offering several distinct advantages. In addition to the rapid production of objective data, XRD measurements on bulk sediments are sensitive to a broader and more characteristic grain size range than counts of individual coarse lithic fragments. The technique is demonstrated in a study of 12 sediment cores from the North Atlantic. Bulk quartz content and the quartz-to-plagioclase ratio exhibit peak-to-peak correspondence to manual counting results, which verifies the identification of large IRD influxes. The XRD data also reveal variations between the manually identified peaks, suggesting increased sensitivity to low-level, distal, or sea-ice sources of IRD. A saw-tooth pattern emerges in many IRD events, which supports a link between ice rafting and atmospheric temperature changes over Greenland, and providing further evidence of the influence of climate on iceberg discharges. Larval dispersal and spatial distribution of reproducing females of Pandalus borealis in the Barents Sea were investigated. The approach was based on particle tracking using flow fields and a biological model incorporating ontogenetic migration and temperature dependent development of the zoea stages. By the use of individual-based particle-tracking models it is possible to assess the life history of individuals, as opposed to an Eulerian model. The years simulated in this study were 1996–1998. New data on larval hatching, development and behaviour from laboratory experiments and field studies are presented. Annual shrimp surveys conducted by the Norwegian Institute of Fisheries and Aquaculture provided abundance estimates of spawning and one year old shrimp. The biological model simulated the larval development of shrimp from hatching to zoea V, and their vertical migratory behaviour. The results indicated large inter-annual variability with respect to dispersion. This is most likely explained by the position of the Polar Front and the inflow of Atlantic water into the Barents Sea. The spatial distribution of modelled settled larvae was in good agreement with the abundance of 1 year old juveniles obtained from trawl surveys. The distance travelled by a shrimp larvae varied between 0 and 330 km and the annual mean distance varied between 74 and 122 km. Female shrimp in the entire Barents Sea contributed to recruitment to the stock, which is considered to be one population. Therefore, the female component of the shrimp stock has to be the target of management throughout the Barents Sea. [Copyright &y& Elsevier]

Experiments on use of an agar-gel method for recovery of migrating Ascaris suum larvae from the liver and lungs of pigs were conducted to obtain fast standardized methods. Subsamples of blended tissues of pig liver and lungs were mixed with agar to a final concentration of 1% agar and the larvae allowed to migrate out of the agar-gel into 0.9% NaCl at 38°C. The results showed that within 3 h more than 88% of the recoverable larvae migrated out of the liver agargel and more than 83% of the obtained larvae migrated out of the lung agar-gel. The larvae were subsequently available in a very clean suspension which reduced the sample counting time. Blending the liver for 60 sec in a commercial blender showed significantly higher larvae recovery than blending for 30 sec. Addition of gentamycin to reduce bacterial growth during incubation, glucose to increase larval motility during migration or ice to increase sedimentation of migrated larvae did not influence larvae recovery significantly. Summary: Since the introduction of African swine fever virus (ASFV) into the Baltic states and Poland in 2014, the disease has continued to spread within these regions. In 2017, the virus spread further west and the first cases of disease were reported in the Czech Republic and Romania, in wild boar and domestic pigs, respectively. To control further spread, knowledge of different modes of transmission, including indirect transmission via a contaminated environment, is crucial. Up until now, such an indirect mode of transmission has not been demonstrated. In this study, transmission via an environment contaminated with excretions from ASFV‐infected pigs was investigated. Following euthanasia of pigs that were infected with an isolate of ASFV from Poland (POL/2015/Podlaskie/Lindholm), healthy pigs were introduced into the pens, in which the ASFV‐infected pigs had been housed. Introduction was performed at 1, 3, 5 or 7 days, following euthanasia of the infected pig groups. Pigs, that were introduced into the contaminated environment after 1 day, developed clinical disease within 1 week, and both ASFV DNA and infectious virus were isolated from their blood. However, pigs introduced into the contaminated pens after 3, 5 or 7 days did not develop any signs of ASFV infection and no viral DNA was detected in blood samples obtained from these pigs within the following 3 weeks. Thus, it was shown that exposure of pigs to an environment contaminated with ASFV can result in infection. However, the time window for transmissibility of ASFV seems very limited, and, within our experimental system, there appears to be a rapid decrease in the infectivity of ASFV in the environment. Outbreaks of porcine epidemic diarrhoea ( PED) were reported across Europe during the 1980s and 1990s, but only sporadic outbreaks occurred in recent years. PED virus ( PEDV) spread for the first time into the USA in 2013 and has caused severe economic losses. Retrospectively, it was found that two different strains of PEDV have been introduced into the United States, both are closely related to strains circulating in China where a new wave of the disease occurred from 2010 onwards. Since autumn 2014, new outbreaks of PED have occurred in Europe. In this study, weaned piglets were inoculated with an early European isolate (Br1/87) or faecal/intestinal suspensions derived from pigs infected with a recent European strain of PEDV (from Germany) or a US strain of PEDV. No evidence for infection resulted from inoculation of pigs with the German sample that contained high levels of PEDV RNA; there were no clinical signs, excretion of viral RNA or anti- PEDV antibody production. In contrast, all the pigs in the other two groups showed evidence of infection. Mild clinical signs of disease, mainly diarrhoea, occurred in piglets inoculated with the Br1/87 and US PEDV strains. PEDV RNA was detected throughout the intestine in euthanized animals at 4 days post-inoculation. In addition, within these animals, low levels of viral RNA were detected in lungs and livers with higher levels in spleens. Seroconversion against PEDV occurred in all surviving infected animals within 10 days. PEDV RNA excretion occurred for at least 2 weeks. The US PEDV RNA was detected at low levels in serum samples on multiple days. It is apparent that current diagnostic systems can detect infection by the different virus strains. Background Lower extremity vascular trauma ( LEVT) is a major cause of amputation. A clear understanding of prognostic factors for amputation is important to inform surgical decision-making, patient counselling and risk stratification. The aim was to develop an understanding of prognostic factors for amputation following surgical repair of LEVT. Methods A systematic review was conducted to identify potential prognostic factors. Bayesian meta-analysis was used to calculate an absolute (pooled proportion) and relative (pooled odds ratio, OR) measure of the amputation risk for each factor. Results Forty-five studies, totalling 3187 discrete LEVT repairs, were included. The overall amputation rate was 10·0 (95 per cent credible interval 7·4 to 13·1) per cent. Significant prognostic factors for secondary amputation included: associated major soft tissue injury (26 versus 8 per cent for no soft tissue injury; OR 5·80), compartment syndrome (28 versus 6 per cent; OR 5·11), multiple arterial injuries (18 versus 9 per cent; OR 4·85), duration of ischaemia exceeding 6 h (24 versus 5 per cent; OR 4·40), associated fracture (14 versus 2 per cent; OR 4·30), mechanism of injury (blast 19 per cent, blunt 16 per cent, penetrating 5 per cent), anatomical site of injury (iliac 18 per cent, popliteal 14 per cent, tibial 10 per cent, femoral 4 per cent), age over 55 years (16 versus 9 per cent; OR 3·03) and sex (men 7 per cent versus women 8 per cent; OR 0·64). Shock and nerve or venous injuries were not significant prognostic factors for secondary amputation. Conclusion A significant proportion of patients who undergo lower extremity vascular trauma repair will require secondary amputation. The aim of the study was to assess whether blood samples collected onto FTA® cards could be used in combination with real-time PCR for the detection of African swine fever virus ( ASFV) DNA in samples from resource-poor settings under the assumption that asymptomatically (sub-clinically) infected pigs may be present. Blood samples were collected from clinically healthy pigs from Mbeya Region, Tanzania. The blood samples were stored on FTA® cards and analysed by real-time PCR assays in duplicate; three pigs had high levels of viral DNA (Ct values of 27-29), and three pigs had a low level of viral DNA (Ct 36-45). Four pigs were positive in one of the duplicate samples only, but clear products of the expected size were obtained when the reactions were analysed by gel electrophoresis. For comparison, blood samples from pigs experimentally infected with either a pathogenic ( OURT T88/1) or a non-pathogenic ( OURT T88/3) isolate of ASFV were collected, stored on FTA® cards and analysed in the same way. The blood from pigs infected with the OURT T88/1 isolate showed high levels of viral DNA (Ct 22-33), whereas infection with non-pathogenic OURT T88/3 isolate resulted in only low levels of viral DNA (Ct 39) in samples collected at 10-14 days after inoculation. Detection of Schmallenberg virus RNA, using real-time RT- PCR, in biting midges ( Culicoides spp.) caught at 48 locations in 2011 and four well-separated farms during 2012 in Denmark, revealed a remarkably rapid spread of virus-infected midges across the country. During 2012, some 213 pools of obsoletus group midges (10 specimens per pool) were examined, and of these, 35 of the 174 parous pools were Schmallenberg virus RNA positive and 11 of them were positive in the heads. Culicoides species-specific PCRs identified both C. obsoletus and C. dewulfi as vectors of Schmallenberg virus. [ABSTRACT FROM AUTHOR]

Objectives: To examine whether dynamic contrast-enhanced CT (DCE-CT) could be used to characterise and safely distinguish between malignant and benign lung tumours in patients with suspected lung cancer. Methods: Using a quantitative approach to DCE-CT, two separate sets of regions of interest (ROIs) in tissues were placed in each tumour: large ROIs over the entire tumour and small ROIs over the maximally perfused parts of the tumour. Using mathematical modelling techniques and dedicated perfusion software, this yielded a plethora of results. Results: First, because of their non-normal distribution, DCE-CT measurements must be analysed using log scale data transformation. Second, there were highly significant differences between large ROI and small ROI measurements (p<0.001). Thus, the ROI method used in a given study should always be specified in advance. Third, neither quantitative parameters (blood flow and blood volume) nor semi-quantitative parameters (peak enhancement) could be used to distinguish between malignant and benign tumours. This was irrespective of the method of quantification used for large ROIs (0.13

Rabies is the oldest known zoonotic disease and was also the first recognized bat associated infection in humans. To date, four different lyssavirus species are the causative agents of rabies in European bats: the European Bat Lyssaviruses type 1 and 2 (EBLV-1, EBLV-2), the recently discovered putative new lyssavirus species Bokeloh Bat Lyssavirus (BBLV) and the West Caucasian Bat Virus (WCBV). Unlike in the new world, bat rabies cases in Europe are comparatively less frequent, possibly as a result of varying intensity of surveillance. Thus, the objective was to provide an assessment of the bat rabies surveillance data in Europe, taking both reported data to the WHO Rabies Bulletin Europe and published results into account. In Europe, 959 bat rabies cases were reported to the RBE in the time period 1977-2010 with the vast majority characterized as EBLV-1, frequently isolated in the Netherlands, North Germany, Denmark, Poland and also in parts of France and Spain. Most EBLV-2 isolates originated from the United Kingdom (UK) and the Netherlands, and EBLV-2 was also detected in Germany, Finland and Switzerland. Thus far, only one isolate of BBLV was found in Germany. Published passive bat rabies surveillance comprised testing of 28 of the 52 different European bat species for rabies. EBLV-1 was isolated exclusively from Serotine bats ( Eptesicus serotinus and Eptesicus isabellinus), while EBLV-2 was detected in 14 Daubenton′s bats ( Myotis daubentonii) and 5 Pond bats ( Myotis dasycneme). A virus from a single Natterer's bat ( Myotis nattereri) was characterized as BBLV. During active surveillance, only oral swabs from 2 Daubenton′s bats (EBLV-2) and from several Eptesicus bats (EBLV-1) yielded virus positive RNA. Innate lymphoid cells (ILCs) are an emerging subset of lymphocytes involved in surveillance against virally infected cells. Here, we show CD3−CD8high lymphocytes in macaque blood include major subsets of ILCs including natural killer (NK) cells expressing CD16, NKp46, and NKG2A, but also populations of ILCs in mucosal tissues having different properties. One ILC subset secreted interleukin (IL)-17 (ILC17), but these were restricted to mucosal tissues. Some mucosal ILC17 cells expressed classical NK-cell markers, but little NKG2A or NKG2D. Some ILC17 cells secreted IL-22 and tumor necrosis factor-α, but few produced interferon (IFN)-γ or contained granzyme B. IL-17 production by ILCs was induced by IL-6, transforming growth factor-β, and IL-23. Further, simian immunodeficiency virus (SIV) infection resulted in a significant loss of ILC17 cells, especially in the jejunum, which persisted throughout SIV infection. These findings indicate that ILC17 cells may be involved in innate mucosal immune responses, and their loss may contribute to loss of intestinal mucosal integrity and disease progression in human immunodeficiency virus (HIV)/SIV infection. Objectives: A contrast-enhanced multidetector CT (MDCT) scan is the first choice examination when evaluating patients with suspected lung cancer. However, while the clinical focus is on CT, research focus is on molecular biological methods whereby radiolabelled pharmaceuticals are injected into participants and target malignant lung tumours. We examined whether a contrast-enhanced MDCT scan supplied with an additional non-contrast enhanced high-resolution CT scan, or a newer but more expensive 99mTc depreotide single photon emission CT (SPECT) scan, was the better firstchoice examination for the work-up of pulmonary lesions. Furthermore, we examined whether a 99mTc depreotide SPECT scan was an appropriate second-choice examination for patients with indeterminate lesions. Methods: 140 participants were included in the analysis. CT images were given a malignancy potential rating of 1, 2 or 3 with higher rating being indicative of disease. 99mTc depreotide SPECT images were graded either positive or negative. Histopathology and CT follow-up were used as reference standard. Sensitivity, specificity and diagnostic accuracy were calculated. Results: Overall sensitivity, specificity and diagnostic accuracy of CT were 97%, 30% and 84%, respectively. Overall sensitivity, specificity and diagnostic accuracy of 99mTc depreotide SPECT were 94%, 58% and 76%, respectively. For indeterminate lesions sensitivity, specificity and diagnostic accuracy of 99mTc depreotide SPECT were 71%, 68% and 69%, respectively. Conclusion: Both CT and 99mTc depreotide SPECT made valuable contributions to the evaluation of pulmonary lesions. 99mTc depreotide SPECT results were not superior to CT results and did not contribute further to the diagnostic work-up. Human leucocyte antigen (HLA) polymorphisms among immigrants from Pakistan have not been well investigated. Immigration to Norway started in the late 1960s for working purposes. From 1975, immigration was mainly for marriages and family reunion. When recruiting couples for a birth cohort study, we ended up with 65.5% of the 374 parents genotyped being closely related. This was also reflected in that 21% of newborns were homozygotes for their DRB1-DQA1-DQB1 genotype. For being able to study HLA class II genes frequencies among unrelated individuals, we had to exclude 195 of the parents from data analysis. High-resolution typing for the DRB1 locus, low/intermediate for the DQA1 locus and resolution genotyping for the DQB1 locus were performed in all the 179 parents and their newborns from the Punjab province of Pakistan. We identified 25 DRB1, nine DQA1 and 14 DRB1 alleles in the 179 unrelated parents included in our analysis. The most frequent alleles were DRB1*03:01:01 (15.9%) and DRB1*07:01:01 (15.9%), DQA1*01:03 (22.1%) and DQB1*02:01:01 (26.0%). Forty-one haplotypes were identified, including DRB1*13:02:01-DQA1*01:02-DQB1*06:03:01, not earlier reported. Supported by the few earlier reports on Pakistani groups living in Pakistan, it appears that alleles found among those living in Norway are of Indo-European or mixed ethnic origin. This study provides the first comprehensive report of HLA class II alleles and haplotypes in Norwegian Pakistani immigrants. When the unrelated parents were compared with all parents genotyped, there were, however, no significant differences in allele frequencies, confirming that consanguineous marriages are usual in Pakistan. A flume study was conducted using a soil, yard waste compost, and an erosion control compost to investigate the response to concentrated flow and determine if the shear stress model could be used to describe the response. Yard waste compost (YWC) and the bare Cecil soil (CS) control responded to concentrated laboratory runoff similarly in rill formation, and the shear stress model was useful in estimating sediment yields from both of these land covers. Results indicated that critical shear stress values for yard waste compost and Cecil soil were not significantly different. However, the erodibility of the soil was higher, resulting in increased solids loss, on the Cecil soil. Micro-dams, the main mechanism for limiting soil erosion for the compost, often occurred in the rills of the erosion control compost (ECC), causing the concentrated flow to infiltrate and fan out through the compost. Intestinal CD4+ T cells are rapidly and profoundly depleted in human immunodeficiency virus (HIV)-infected patients and simian immunodeficiency virus (SIV)-infected macaques. However, monitoring intestinal cells in humans is difficult, and identifying surrogate markers in the blood, which correlate with loss or restoration of intestinal CD4+ T cells could be helpful in monitoring the success of therapeutic strategies and vaccine candidates. Recent studies indicate HIV utilizes the intestinal homing molecule α4β7 for attachment and signaling of CD4+ T cells, suggesting this molecule may have a central role in HIV pathogenesis. Here, we compared β7HIGH integrin expression on CD4+ T cells in blood with loss of CD4+ T cells in the intestine of macaques throughout SIV infection. The loss of β7HIGH CD4+ T cells in blood closely paralleled the loss of intestinal CD4+ T cells, and proved to be a more reliable marker of intestinal CD4+ T-cell loss than monitoring CCR5+ memory CD4+ T cells. These data are consistent with a recent hypothesis that α4β7 has a role in the selective depletion of intestinal CD4+ T cells, and indicate that monitoring β7HIGH expression on CD4+ T cells in the blood may be a useful surrogate for estimating intestinal CD4+ T cell loss and restoration in HIV-infected patients. Nine viral diseases included in the World Organization for Animal Health list of notifiable diseases (former list A) were chosen for their contagiousness and high capacity of spreading to improve their diagnosis using new and emerging technologies. All the selected diseases – foot-and-mouth disease, swine vesicular disease, vesicular stomatitis, classical swine fever, African swine fever, bluetongue, African horse sickness, Newcastle disease and highly pathogenic avian influenza – are considered as transboundary diseases, which detection causes the prohibition of livestock exportation, and, thus, it leads to high economical losses. The applied diagnostic techniques can fall into two categories: (i) nucleic-acid detection, including padlock probes, real-time PCR with TaqMan, minor groove binding probes and fluorescence energy transfer reaction probes, isothermal amplification like the Cleavase/Invader assay or the loop-mediated amplification technology and the development of rapid kits for ‘mobile’ PCR and (ii) antigen-antibody detection systems like simplified and more sensitive ELISA tests. Besides, internal controls have been improved for nucleic acid-detecting methods by using an RNA plant virus – Cowpea Mosaic Virus – to ensure the stability of the RNA used as a positive control in diagnostic real-time RT-PCR assays. The development of these diagnosis techniques has required the joint efforts of a European consortium in which nine diagnostic laboratories and an SME who have collaborated since 2004 within the European Union-funded Lab-on-site project. This study investigates the transmission of bovine viral diarrhoea virus (BVDV) 1f from a persistently infected (PI) lesser Malayan mousedeer to two bovine calves. Different contact routes to two calves were analysed: 1) aerosol contact between two adjacent pens without physical contact; 2) indirect contact by use of common utensils; 3) direct nose-to-nose contact for 30 seconds. One of the calves was infected either by aerosol or indirect contact. The virus sequence in 247 nucleotides in the 5′-UTR was 100% identical in mousedeer and calf. To elucidate the distribution of BVDV within the affected mousedeer family the captive population in a Zoo was analysed. The maternal line of PI animals was maintained, whereas a PI male was able to reproduce and have a non-PI calf. As a consequence of this, six female PI mousedeer were killed; subsequent autopsies did not reveal any lesions. Sequencing mousedeer BVD virus in the E2 region (420 nucleotides) through 4 generations showed only 7 mutations, which were maintained from mother to offspring. [ABSTRACT FROM AUTHOR]

Background Pre-transplant clinical evaluation of autografting is an important step in predicting post-transplant support, complications and safety. Today, unfavorable outcomes such as early death or graft failure are rare, making them unsuitable for quality assessment of supportive autografting. However, end-points constructed from frequently occurring clinical events may estimate clinically relevant prognostic models. Methods The present retrospective analysis was based on two consecutive clinical trials in the Nordic area, including up to 640 newly diagnosed multiple myeloma patients. Results In the model, the efficacy (time on antibiotics and use of transfusions) was influenced by pre-transplant variables, including sex, nationality, serum creatinine, hemoglobin, disease stage at diagnosis, response following induction therapy, length of priming and average graft CD34 + cell number per day of harvest. The toxicity end-point (time to blood cell recovery) was influenced by nationality, marrow plasma cell percentage, serum creatinine, M-component isotype, response to induction therapy, length of priming and graft CD34 + cell number. The safety (early disease recurrence or death) was influenced by serum creatinine, hemoglobin, treatment response and CD34 + cell number. Discussion In conclusion, the model illustrates that intervention strategies in quality assessment of autografting may benefit from probability estimates of graded clinical end-points.
