In the present study, the phytochemical composition, antioxidant and anti-hemolytic activities of root and aerial part Rubia tinctorum hydromethanolic extracts were investigated. Phytochemical screening reveals the presence of phenol, alkaloids, coumarin, flavonoids, and tannins in both extracts. LC-ESI-MS/MS analysis showed the presence of 15 and 17 components in the root and the aerial part extracts, respectively. The quantitative analysis indicates the richness of root extracts in phenolic compounds (118.38 mg GAE/g) and flavonoids (45.29±0.04 mg GAE/g), the aerial part extract has the highest levels in tannins (134.1±0.1 mg GAE/g). Besides, aerial part extracts revealed the highest antioxidant activity for DPPH (83.23 %) and FRAP (1.51±0.22), while the root extract exhibited the highest potential for TAC and antihemolytic activity (61.09 %). 2,3'-Biindole (2) was synthesized via bromination of indole (1) with molecular bromine and underwent simultaneous dimerization. Antiproliferative and cytotoxic activity of 2 was investigated in vitro on C6 (rat brain tumor), HeLa (human cervix carcinoma), and HT29 (human colon carcinoma) cells lines by using BrdU cell proliferation ELISA and lactate dehydrogenase (LDH) assays. In contrast to 5-fluorouracil (5-FU), 2,3'-biindole (2) significantly inhibited proliferation of HeLa and HT29 cell lines. According to LDH assay, the cytotoxicity of compound 2 was low on HT29 cell lines and high on HeLa and C6 cell lines. Moreover, 2 did not cause any DNA laddering on the DNA of tested cells; therefore, it is suggested that the mechanism of action of this compound may not involve apoptosis. In addition, 2 inhibited relaxation of supercoiled plasmid DNA by topoisomerase activity. The main objective of this study is to highlight the dietary risk of the prevalence of blaESBL producing multidrug-resistant Enterobacteriaceae in raw milk, besides evaluating the effect of Artemesia herba-alba and Thymus algeriensis essential oils against blaESBL producing Enterobacteriaceae isolates from raw milk. Isolation and molecular identification of ESBL producing Enterobacteriaceae and blaTEM, blaSHV, blaCTX-M genes encoding resistance were investigated by PCR and sequencing. The chemical composition of oils was effectuated by GC/MS. Antibacterial activity was tested by disc diffusion and microdilution methods. Six ceftazidime-resistant Enterobacteriaceae were identified; four of them were confirmed as ESBL producers and were found to be multi-drug resistant. blaSHV and blaTEM were found to be having the most dominant gene encoding resistance. The results of GC/MS analysis bestowed 31 and 35 compounds for A. herba-alba and T. algeriensis respectively. Klebsiella pneumonia SB6 (17.04 mm for A. herba-alba and 16.22 mm for T. algeriensis) revealed the highest zones of inhibition. MIC values were 1.56-12.5 mg/mL for both essential oils, while the MBC values were 6.25-25 mg/mL (A. herba-alba) and 12.5-25 mg/mL (T. algeriensis). [ABSTRACT FROM AUTHOR]

Objective(s): The current study was conducted to assess the protective mechanisms of n-BuOH fraction from the aerial part of Genista cephontala (BEGC) on APAP-induced liver injury compared to necrostatine-1 (Nec-1). Materials and Methods: A model of APAP-induced hepatotoxicity was created in male rats by injecting a single dose; 1000 mg/kg APAP, the protective effect was performed with (200 mg/kg; 10 days) BEGC compared to Nec-1, (1.8 mg/kg). Results: BEGC or NeC-1 pretreatment significantly abolished impaired effects in APAP-rats, by decreasing the generation of TBARS and ROS in mitochondrial and cytosolic fractions and maintaining liver function activities. A marked response was observed in the levels of both GSH and GSH-system enzymes in liver homogenates and mitochondrial fractions to BEGC. BEGC/Nec-1 successfully regulated the inflammatory mediators (IL-β, TNF-α, HMGB1, and acHMGB1) and MPO levels. During APAP treatment, no caspase-3 or -8 activity was detected, and the level of fk18; M30 was higher than the levels of cck18; M65. Moreover, RIPK3 and MLKL levels were increased in the APAP group. These results suggested that necroptosis predominates during the APAP liver injury model. Interestingly, these necroptotic factors were significantly down-regulated by BEGC treatment. Both biochemical and histopathological findings were consistent with each other. Conclusion: From all these findings, the hepatoprotective effect of BEGC could be due to the abundance of polyphenols identified by LC-MS/MS analysis, as well as the synergistic interactions of all contents.
