To address the problem of high false positive/negative rate and low accuracy in protein complex detection, we propose the Dual-population Firefly Algorithm Based on Gender Differences (DFAGD) based on the unique core-attachment structure of protein complexes and the biological properties of fireflies. This method divides the detection of protein complexes into two phases. Firstly, a global search of the male population is used to detect the core proteins, and then a local search of the female population is used to detect the attachment proteins, which improves the accuracy of detection. In the male population strategy, the population diversity is redefined, and when the diversity falls below the threshold, a spring model is introduced to bring the population into the repulsion phase so that it does not fall into a local optimum. The female population selects elite and excellent individuals from the detection results of the male population to perform guided neighborhood searches, which can effectively improve detection accuracy. Finally, the effectiveness of the protein complex detection method is tested by comparing it to eight classical detection methods using four datasets of Saccharomyces cerevisiae proteins. Clinical observations show that the current spinal fusion with internal fixation has a nonfusion rate of 5%–35%; however, methods to promote spinal fusion are limited. This study aimed to investigate the role of SDF-1-induced directional chemotaxis of BMSCs in bone marrow chimera spinal intervertebral fusion mouse model. BMSCs were isolated from bone marrow and identified by detecting CD44/CD34 positive cells. BMSCs (GFP-BMSCs) were labeled with GFP for tracking in vivo. Mice were inoculated with GFP-BMSCs to construct bone marrow chimera spinal intervertebral fusion model, which were divided into BM-SIF model, BMSIF+SDF-1, BM-SIF+SDF-1-Anta group. The callus area of intervertebral fusion site was detected by radiology. HE staining was used to detect trabeculae formation. Expressions of osteogenic molecules and fibroblast markers were detected by RT-PCR and Western blotting. GFP-BMSCs showed obvious osteogenic and adipogenic differentiation ability, according to oil-red O and alizarin-red staining. Bone marrow chimera spinal intervertebral fusion mouse model was successfully established, with efficient localization of GFP-BMSCs at intervertebral fusion site. SDF-1 significantly promoted bone trabeculae formation in callus at intervertebral fusion site. SDF-1 significantly increased osteogenic molecules transcription/expression in callus at intervertebral bone graft fusion site of mice; however, SDF-1-Anta (AMD3100) significantly decreased osteogenic molecules transcrition/expression, compared to those of mice from the BM-SIF model group (p < 0.05). SDF-1 markedly induced and SDF-1-Anta significantly decreased fibroblast proliferations in the callus at the intervertebral fusion site of mice, compared to those of mice from the BM-SIF model group (p < 0.05). SDF-1 enhanced expression of Wnt10b and β-catenin in callus at intervertebral fusion site of mice compared to mice of the BM-SIF model group (p < 0.05). In conclusion, SDF-1 induced directional chemotaxis of BMSCs to the intervertebral fusion site and promoted osteogenic differentiation in bone marrow chimera spinal intervertebral fusion mice by regulating Wnt/β-catenin pathway and modulating the proliferation of BMSCs. Aiming at the problem of a single influence measure method in the microblog network, a comprehensive measurement method integrating microblog content topology potential and attributes is proposed. In order to better reflect the dissemination of information in the real network, in order to improve the convergence accuracy of particle swarm optimization, a follow-bee strategy is introduced, and an elite cloning strategy based on Logistic is proposed. At the same time, the simulated annealing algorithm is introduced to make the particle swarm algorithm better jump out of the local optimum. Finally, the improved algorithm is applied to the problem of maximizing the influence of Weibo network. The traditional stress evaluation method cannot describe the chaotic characteristics of speech signals well or cannot fully approximate the complex nonlinear relationship between features, so that the exact location of stress cannot be accurately determined. In order to improve the recognition of spoken English stressed syllables, this study established a word stress recognition model. The model can accurately recognize the stressed syllables in the words and calculate the fundamental frequency change trajectory of the language tones using the fundamental frequency scaling function. According to this trajectory and using the time domain fundamental frequency synchronization superposition algorithm to modify the fundamental frequency parameters in the DIVA model motion instruction, the spoken language learner can master the rhythm in the spoken language. The present study aimed to determine whether maslinic acid effectively inhibits the proliferation of MKN28 cells, and to investigate the mechanisms underlying its antitumor functions. MKN28 cell viability was evaluated using a Cell Counting Kit-8, cell proliferation was analyzed by a colony formation assay and flow cytometry was used to investigate the rate of apoptosis. Western blot analysis was performed in order to determine the differential expression levels of Janus kinase (JAK), signal transducer and activator of transcription 3 (STAT3) and apoptosis associated proteins B-cell lymphoma 2 (Bcl-2), Bcl-2 associated X protein (Bax) and Bcl-2 associated agonist of cell death (Bad). Interleukin-6 (IL-6) concentration was evaluated using ELISA. IL-6 and anti-IL-6 antibodies were used to investigate the role of IL-6 in MKN28 cells treated with maslinic acid proliferation, and the STAT3 phosphorylation rates. The results demonstrated that maslinic acid treatment significantly reduced cell proliferation, induced apoptosis and was accompanied by a significant decrease in Bcl-2, Bax and Bad expression levels. Maslinic acid treatment also resulted in the downregulation of phosphorylated-STAT3 and JAK2, and significantly inhibited the protein expression of IL-6. Maslinic acid is able to inhibit MKN28 cell proliferation and the phosphorylation of STAT3 by downregulating the expression of IL-6. These results suggest that maslinic acid suppresses the growth of MKN28 cells by inducing apoptosis via its inhibition of the IL-6/JAK/STAT3 signaling cascade. [ABSTRACT FROM AUTHOR]

Novel targeted therapies need to be developed for gastric cancer, the third most common cancer type and the second most common cause of cancer-related mortality in China. Previous studies indicate that angiopoietin (Ang)-2 serves a role in the proliferation, migration, invasion and adhesion of malignant cells. The present study identified, using functional studies, that exogenous expression of miR-218 increased migration of NCI-87 and HGC-27 gastric cancer cells, which coincided with a reduction in the expression of Ang-2. In addition, intratumoral delivery of miR-218 inhibited proliferation and angiogenesis of gastric cancer cells in vivo, with a corresponding decreased in Ang-2 expression. These results indicate that miR-218 serves an important role in gastric cancer tumorigenesis through regulating the expression of Ang-2. Chloride intracellular channel 1 (CLIC1) has been demonstrated to be overexpressed in gastric cancer, and elevated CLIC1 expression levels are markedly associated with the processes of tumor cell migration and invasion. However, the regulatory mechanism and signaling pathway underlying these processes have remained to be elucidated. The present study examined the impact of N-acetyl cysteine (NAC), indanyloxyacetic acid (IAA)-94 and SB203580, inhibitors of reactive oxygen species (ROS), as well as CLIC1 and p38 mitogen-activated protein kinase (MAPK) on the migration and invasion of SGC-7901 gastric cancer cells in a hypoxia-reoxygenation (H-R) microenvironment. The results demonstrated that intracellular ROS and CLIC1 levels were increased under H-R conditions, and that functional inhibition of CLIC1 significantly decreased the H-R-elevated ROS generation and p-p38 MAPK levels in SGC-7901 cells, as well as inhibited the migration and invasion of SGC-7901 cells. In addition, the expression levels of MMP-2 and MMP-9 were inhibited by NAC, IAA-94 and SB203580. Diachasmimorpha longicaudata is the most widely used endoparasitoid in biocontrol programmes against various species of tephritids and is continuously mass-reared under laboratory conditions at a constant temperature; however, little is known about how varying the temperature might affect the effectiveness of the mass rearing of immature D. longicaudata. This study aimed to determine the optimum temperature for the development of larvae D. longicaudata the larvae of its host, Bactrocera dorsalis. Third-instar B. dorsalis were exposed for 4 h to fi ve pairs of parasitoids, and then they were kept at one of six temperatures ranging from 19°C to 34°C. The activities of three major antioxidant enzymes (CAT, POX, SOD) were individually measured 4, 24, 48 and 72 h after exposure to each of the six temperatures. The results were as follows: for CAT the highest CAT activities were recorded at 25°C (intervals 5-24 h and 25-48 h), at 28°C (interval 0-4 h) and at 31°C (interval 49-72 h), and for POX and SOD identically at 25°C (intervals 0-4 h, 5-24 h) and at 31°C (intervals 25-48 h, 49-72 h), respectively. Subsequently, the effects keeping the parasitized larvae at these different temperatures on the biological attributes of D. longicaudata reared under laboratory conditions were investigated.
