As the prevalence and complexity of goose astrovirus genotype 1 and 2 (GAstV 1 and GAstV 2) infections continue to rise in China's goose farming industry, a rapid, reliable and cost-effective diagnostic tool is urgently needed. Duplex SYBR Green I real-time RT-PCR (qRT-PCR) method was developed, which is capable of simultaneous detection of GAstV 1 and GAstV 2. We designed two primer pairs, targeting the viruses conserved genomic regions, facilitating the distinction of GAstV 1 and GAstV 2 through unique melting curve peaks. This method exhibited strong linear relationships in standard curves (R2 = 0.99), and detection limits were 3.18 × 102 and 3.16 × 102 copies/µL for GAstV 1 and GAstV 2, respectively. With intra- and inter-assay variation below 2%, the assay exhibits excellent repeatability. This duplex qRT-PCR, displaying consistent detection rates comparable to a TaqMan-based method, offers a promising, rapid, and cost-effective diagnostic strategy for waterfowl farming. The impact of additional activators (sodium sulfate, sodium nitrite, and calcium nitrite) and curing temperature (20 and 35 °C) on chloride binding and ingress resistance of ground granulated blast furnace slag activated by Ca(OH) 2 was studied using X-ray diffraction, scanning electron microscopy, thermogravimetric and thermodynamic modeling. The outcomes of the study indicated that the difference in the chloride binding capacity at low chloride concentrations was primarily affected by the types of the AFm phase. The solid solution formed between Friedel's salt and various types of AFm phases resulted in a decrease in the chloride binding capacity. At a curing temperature of 35 °C, the chloride binding capacity reduced due to the leaching of calcium ions and carbonation induced by the coarse pore structure. It was found that the chloride ingress resistance was significantly related to the leaching of calcium and carbonation, and the denser calcite layer formed at the exposure layer had a positive impact on the chloride ingress resistance. • Curing at 35 °C led to a lower chloride binding capacity than 20 °C. • Na 2 SO 4 addition led to lower chloride binding capacity. • The samples with added nitrites showed good chloride binding behaviors. • Leaching and carbonation had a positive effect on the chloride resistance. PCV2e shows high sequence similarity to other PCV2 genotype, causing greater difficulties in the molecular biological identification of pathogens. In this study, we developed a TaqMan-based RT-qPCR for efficient detection of PCV2e in clinical samples. We designed specific primers and a probe for the PCV2e ORF2 gene and then identified target genes using the optimized PCR reaction system and reaction conditions. The results indicated that the cycle threshold (Ct) and the standard DNA template had a linear relationship between 103-108 copies/μL, and the determination coefficient was 0.998. The assay has a detection limit of 100 copies per reaction and a standard deviation of cycle thresholds below 1.00. The PCR method we established can specifically identify PCV2e, did not cross-react with other genotypes of PCV, as well as PRV and PPV. In genome-wide association studies (GWASs) to detect the disease-associated genetic variants, two-stage design has received much attention because of its cost effectiveness and high efficiency. Under the framework of a two-stage design, it has been shown that joint analysis is more powerful than replication-based analysis. Several robust tests have been proposed for joint analysis to handle the problem of unknown genetic mode of inheritance. However, existing joint analysis of combining test statistics from both stages might suffer from a loss of efficiency if the combined test statistics are not sufficient or the weight of the statistic for each stage is not appropriate. In this article, we propose a new strategy for joint analysis by combining the raw data rather than the test statistics across stages and construct a robust MAX3-based test for two-staged GWASs, which can make full use of the information of the data from both stages. Our numerical results show that the proposed procedure is more powerful and computationally much faster than the existing joint analysis procedures. Three new species of the predaceous midges of genus Alluaudomyia Kieffer, 1913: A. flavinotum Wu & Li, sp. nov. of the maculipennis group, and A. reflexuralis Wu & Li, sp. nov. and A. limu Wu & Li, sp. nov. of the parva group, are described from the National Park of Hainan Tropical Rainforest, Hainan Island, China. Illustrations and COI barcodes (a fragment of the mitochondrial cytochrome c oxidase subunit 1) of the three new species are also provided. Associations of male and female specimens of two species (A. reflexuralis Wu & Li, sp. nov. and A. limu Wu & Li, sp. nov.) are supported by DNA barcodes. Ginkgetin, a bioflavonoid derived from Ginkgo biloba leaves has been shown to promote tumor cell apoptosis and suppress cell growth in a variety of tumors. This study has examined the role and mechanism of ginkgetin on the tumorigenesis of endometrial carcinoma. Results of the studies show that ginkgetin treatment led to a dose-dependent decrease in cell viability, enhanced expression of cleaved caspase-3 protein, and reduced expression of Bcl-2 protein. In addition, cell migration and invasion of HEC1B and Ishikawa cells were repressed by ginkgetin treatment. Levels of phosphoprotein kinase B, Janus kinase 1, and signal transducers and activators of transcription 3 were decreased in HEC1B and Ishikawa cells by ginkgetin treatment. The digital measurement and processing is an important direction in the measurement and control field. The quantization error widely existing in the digital processing is always the decisive factor that restricts the development and applications of the digital technology. In this paper, we find that the stability of the digital quantization system is obviously better than the quantization resolution. The application of a border effect in the digital quantization can greatly improve the accuracy of digital processing. Its effective precision has nothing to do with the number of quantization bits, which is only related to the stability of the quantization system. The high precision measurement results obtained in the low level quantization system with high sampling rate have an important application value for the progress in the digital measurement and processing field.
