Biogenic amines (BAs), that is, spermine, spermidine, putrescine, histamine, tyramine, β-phenylethylamine, cadaverine, and serotonin, have been determined in several samples of tea leaves, tea infusions, and tea drinks by LC-UV method after derivatization with dansyl chloride. Different extraction solvents have been tested and TCA 5% showed better analytical performances in terms of linearity, recovery percentages, LOD, LOQ, and repeatability than HCl 0.1 M and HClO4 0.1 M and was finally exploited for the quantitative determination of BAs in all samples. In tea leaves total BAs concentration ranged from 2.23 μg g−1 to 11.24 μg g−1 and PUT (1.05–2.25 μg g−1) and SPD (1.01–1.95 μg g−1) were always present, while SER (nd–1.56 μg g−1), HIS (nd–2.44 μg g−1), and SPM (nd–1.64 μg g−1) were detected more rarely. CAD and PHE were determined in few samples at much lower concentrations while none of the samples contained TYR. Tea infusions showed the same trend with total BAs concentrations never exceeding 80.7 μg L−1. Black teas showed higher amounts of BAs than green teas and organic and decaffeinated samples always contained much lower BAs levels than their conventional counterparts. [ABSTRACT FROM AUTHOR]

Reverse-mode operation films can be achieved with polymer dispersed liquid crystals by many methods. One method consists in the use of either rough or polyimide treated supports in order to induce homeotropic alignment of the initial mixture that will separate in liquid crystal and polymer rich phases. The major disadvantage is the need for mesogenic monomers to keep a good alignment of the liquid crystal after the polymerization process. In this paper we present results concerning the use of an anisometric, non-mesogenic, tailor-made monomer for the preparation of excellent reverse-mode polymer dispersed liquid crystal shutters cast on rough surfaces. The combined use of rough surfaces and non-mesogenic monomers allows us to exceed earlier limits in physical and chemical properties, cost, and device dimensions by the use of a wider class of monomer molecules. Inclusion of lipids or polymers with a transition temperature closer to physiological body temperature (40–42 °C) is a strategy used in tumor therapy for more than 30 years, because it allows induction of drug release from delivery systems by mild hyperthermia. Unfortunately, most of these thermo-sensitive carriers are removed from circulation before completion of their function. Thus, novel multi-functional niosomes possessing spontaneous stealth and thermo-sensitive properties were developed from L64 Pluronic ® and L64ox as its derivative, in presence or absence of cholesterol. The use of L64 both as amphiphilic constituent and thermo-sensitive molecule, gave the possibility to bypass the use of additional excipients and increased the system biocompatibility. Niosomes diameter ranged from 400 to 750 nm and were long term stable. Calcein and 5-FU possess great affinity to niosomal matrices rich in PEO groups. Negative Z-potential values were attributed to the negative charges onto the niosomes surface and generally change according to the temperature. The in vitro drugs release studies were performed at 25 °C, 37 °C and 42 °C, that are representative of certain conditions (storage, physiological condition and mild hyperthermia, respectively). Results showed that L64-based niosomes possess spontaneous thermo-sensitive properties: drugs releases were found to be more pronounced at 42 °C. These early results are a promising first step for the development of multi-functional devices that combine several advantages such as stealth properties and temperature controllability at the desired location and time, for a more specific and efficient pharmacological therapy. Cromolyn sodium, or disodium cromoglycate (CS), is a surface active drug : a pharmacologically active compound with an amphiphilic nature. At certain conditions it is able to self-associate in several kind of supramolecular aggregates. Since CS could play the role of both carrier and drug, bypassing the use of additional excipients and increasing the system biocompatibility, the effects of cromolyn self-aggregates on diffusion and percutaneous permeation across rabbit ear skin were investigated. Niosomes (vesicular systems, 0.5 wt% of CS), monomeric and isotropic solutions (0.5 and 5 wt% of CS), nematic (15 wt% of CS) and hexagonal phases (30 wt% of CS) were selected as supramolecular systems and tested as transdermal delivery systems. Results demonstrated that CS was able to form vesicular structures of about 500 nm of diameter and this formulation gave the higher percutaneous permeation profile (systemic action), while isotropic solution and liquid crystals mesophases acted as slower release reservoir of drug on the skin surface (local action), as confirmed by diffusion coefficients. Diffusion rates through a synthetic membrane were dependent both on CS concentration present into the formulations and on its structural organization: maximum diffusion was noticed with isotropic solution, a lower amount of diffused cromolyn sodium was achieved by hexagonal phase. Consequently, CS appears as a versatile surfadrug as, depending on the disease degree, it is possible to modulate its permeation profile by choosing the most appropriate formulation. Biogenic amines (BAs) are detrimental to health and the consumption of food containing high concentrations of these compounds may cause toxic reactions. In particular, cases of tyramine intoxication have occurred subsequent to the consumption of cheese and the term “cheese reaction” has been coined to refer to it. The main pre-requisites for the presence of BAs in foods include: availability of free amino acids, the presence of microorganisms producing BA enzymes (mainly from raw materials and/or added starter cultures), conditions allowing their growth (particularly temperature, pH), as well as conditions affecting the enzyme production and activity (particularly low pH). The main technological aspects affecting aminogenesis in cheese are the presence of micro-organisms, their proteolytic and decarboxylase activities, ripening time, ripening temperature, pH and NaCl concentration. Moreover secondary parameters such as post-ripening processes and packaging can affect the accumulation of BAs as well. High temperature, high pH, low salt content, prolonged ageing and air packaging seem to accelerate the amino acid accumulation while the effect of grating seem to be mostly related with microbiological contamination, both aspects supporting the BAs increase. Abstract: In this study, we have designed novel chromanyl derivatives that share with α-tocopherol a chromanyl head but differ in the lateral chain in: (i) length and saturation (FEBL-45, 50, 70), (ii) position of double bonds in Z or E (FEBL-50 and 53 and their respective 6-chromanyl methyl derivatives FEBL-161 and 162), or (iii) presence of additional antioxidant molecules, such as the catechol compound hydroxytyrosol (FEBL-80) or dopamine (FEBL-82, 95). The efficiency of these compounds in preventing free-radical-induced oxidative stress was investigated in isolated membranes as well as intact cells. The results of this study clearly show that all compounds synthesized were active in: (i) inhibiting AAPH- or tert-BOOH-induced lipid peroxidation in microsomes and (ii) preventing H2O2-induced ROS production, cell damage, and heat-shock protein expression in immortalized RAT-1 fibroblasts. Such effects were dose- and time-dependent. Independent of the kind of pro-oxidant used, differences in the antioxidant potency of these compounds were found in relation to the chemical structure with respect to the natural α-tocopherol: (1) The concomitant presence of a chromanyl head and an additional aromatic ring markedly increased the antioxidant potency of the molecule. In particular, FEBL-82 and FEBL-95, resulting from the molecular combination of trolox and dopamine, were much more potent than α-tocopherol, α-tocotrienol, and the other synthetic compounds. Moreover, they were also more potent than trolox and dopamine, used alone or in combination, suggesting synergistic cooperative interactions in the molecule between chromanyl and catechol moieties. (2) The length of the side chain affected the antioxidant properties of the molecule: FEBL-70, which displays a bulky squalene side chain, was less effective than the natural α-tocotrienol and the synthetic FEBL-45 and FEBL-50. (3) The presence of polyunsaturated double bonds in the side chain in the Z configuration (FEBL-53, FEBL-162) increased the antioxidant potency of the molecule with respect to the E configuration (FEBL-50, FEBL-161). Electro-responsive hybrid hydrogels were synthesized by free radical polymerization using Gelatin-coated multi-walled carbon nanotubes as electro-conductive component, acrylamide and polyethylene glycol dimethacrylate as plasticizing and crosslinking monomer, respectively. Dynamic light scattering, Raman spectroscopy, scanning electron microscopy, resistivity measurement, cell viability assay, and evaluation of swelling degree upon application of an external voltage at 0, 12, 24, 36, and 48 V were performed as characterization tools. Composite materials were found to be highly versatile in modulating the drug delivery of neutral drugs (e.g. Curcumin) as a function of both nanotube content and voltage magnitude, with drug partition between carrier and releasing media being dependent on the balance between electrostatic attractive and repulsive forces and hydrogel swelling degree. Finally, suitable mathematical modelling were employed for the kinetic characterization of the release mechanism. Redox-responsive nanoparticles were synthesized by desolvation of bovine serum albumin followed by disulfide-bond crosslinking with N , N ʹ-Bis (acryloyl) cystamine. Dynamic light scattering and transmission electron microscopy studies revealed spherical nanoparticles (mean diameter: 83 nm, polydispersity index: 0.3) that were glutathione-responsive. Confocal microscopy revealed rapid, efficient internalization of the nanoparticles by Daoy medulloblastoma cells and healthy controls (HaCaT keratinocytes). Cisplatin-loaded nanoparticles with drug:carrier ratios of 5%, 10%, and 20% were tested in both cell lines. The formulation with the highest drug:carrier ratio reduced Daoy and HaCaT cell viability with IC 50 values of 6.19 and 11.17 μg mL −1 , respectively. The differential cytotoxicity reflects the cancer cells’ higher glutathione content, which triggers more extensive disruption of the disulfide bond-mediated intra-particle cross-links, decreasing particle stability and increasing their cisplatin release. These findings support continuing efforts to improve the safety and efficacy of antineoplastic drug therapy for pediatric brain tumors using selective nanoparticle-based drug delivery systems. The study evaluated the fatty acids and the biogenic amines (BAs) in ‘Nduja of Spilinga stored in different packaging materials (i.e. natural casing under vacuum, glass jar, aluminum tube and OVTENE®) during 15 months of shelf-life. Raw materials and pepper mixture were analysed as well. BAs concentrations increased with time, tyramine (TYR), putrescine (PUT) and cadaverine (CAD) were the most abundant. BAs in natural casing were always higher than those found in glass jar, aluminum tube and OVTENE®. Total fatty acids were characterized by higher level of unsaturated fatty acid that decreased with time (glass jar > natural casing under vacuum > aluminum tube > OVTENE®). The reduction of PUFA is the consequence of the increase of peroxides and carbonyls reacting with amino acids to form BAs. Hybrid hydrogel films able to modulate the release of anionic species, ketoprofen (Ket) and bovine serum albumin (BSA), as a function of their molecular size are prepared by UV-induced polymerization of methacrylated gelatin in the presence of oxidized multiwalled carbon nanotubes (MWNT_COOH). The dual-stimuli responsive composites can modulate the release in response to the variation of temperature and the application of an external voltage. Drug release profiles, analyzed by mathematical models, show that different temperatures (25 and 45 °C) and applied voltages (0 and 36 V) have a different effect on the release of the two therapeutics. Ket release is enhanced at 45 °C (77% vs 22% at 25 °C) with the maximum value recorded when 36 V is applied (94%). The loading of chemotherapics into smart nanocarriers that simultaneously possess more than one useful property for specifically targeting a tumor site improves their therapeutic effectiveness, reducing their side effects. Hence, we proposed a combined approach for the treatment of human breast cancer (BC) consisting of the co-encapsulation of doxorubicin and curcumin or doxorubicin and quercetin into multifunctional niosomes, which results in prolonged blood circulation and an ability to spontaneously accumulate at the tumor site (passive target) and to recognize and bind the tumor cells through dual ligand-receptor interactions (active target). The drug-loaded vesicles showed high stability and good capability of loading doxorubicin and antioxidants alone or in combination. Their diameter was around 400 nm. The drugs released from the vesicles were found to be controlled and sustained for over 24 h, with a strong dependence on the co-presence of the loaded molecules. Transferrin and/or folic acid were conjugated on the external surface of the niosomes as ligands, considerably improving the cellular uptake into MCF-7 and MDA-MB-231 malignant cells when compared with the uptake of nonconjugated samples. In vitro evaluation of anticancer activity demonstrated the strong potential of niosomes loaded with a doxorubicin/curcumin combination as useful devices in breast tumor treatment. Cotton gauze was inserted into a hydrogel network composed of acrylamide, sodium methacrylate, and polyethylene glycol dimethacrylate to fabricate an electroresponsive delivery system for wound dressing. The composite was characterized by swelling measurements, showing that shrinking or swelling depend on the applied voltage. The release profile of incorporated diclofenac sodium salt shows the possibility to modulate the kinetics by changes in the amplitude and duration of applied electric pulses. The present article reports the preparation, characterization and performance evaluation of solid lipid nanoparticles (SLNs) based on polyoxyethylene-40 stearate (PEG-40 stearate) for the administration of antifungal agents such as ketoconazole and clotrimazole. These nanoparticles could be useful in the treatment of vaginal infections sustained byCandida albicans. In particular, PEG-40 stearate was made to react with acryloyl chloride in order to introduce an easily polymerizable moiety for the creation of a second shell and to ensure a slow drug release. In addition, the differences on the release profiles between PEG-40 stearate-based nanoparticles, PEG-40 stearate acrylate based and polymerized ones, were analyzed under conditions, simulating the typical environment ofCandida albicansinfection. Then, the antifungal activity of nanoparticles was also evaluated in terms of minimal inhibitory concentration. Moreover, the nanoparticles were submitted toin vitrostudies for evaluating the drug permeability at the site of action. Objectives This study concerns the preparation and characterization of microspheres based on a mixture of triterpene saponins, from Physospermum verticillatum (Waldst & Kit), as a carrier for the specific release of gemcitabine. Methods Triterpene saponins were derivatized with acrylic acid. The obtained polymerizable product was characterized by Fourier transform infrared to confirm the ester linkage. Then, spherical microparticles were prepared by suspension radical copolymerization and impregnated with gemcitabine. Key findings Microspheres exhibited a mean diameter of 2.7 μ. The swelling studies showed that particles swell most at pH 6.2, typical of the tumour pathology, than at pH 7.4, miming physiological conditions. The microspheres were loaded with gemcitabine ( LE 72.2%). Their release profile showed an initial dot of around 24% and a further release for 24 h. Conclusions This carrier could be potentially release the drug in the lung, as a function of different pHs between tumour cells and healthy, reducing the systemic drug toxicity, allowing the reduction of the doses number, increasing the drug half-life and eliminating the problems related to the fast clearance of gemcitabine administration. In recent years, antioxidants have gained great importance because of their potential use in food, pharmaceutical, and cosmetic industries. This interest is rooted in the cumulative evidence connecting active oxygen and free radicals with numerous human degenerative disorders, such as cardiovascular diseases, cancer, aging, and atherosclerosis. Polyphenols are the major class of antioxidant able to reduce the oxidative damages of lipids, proteins, enzymes, carbohydrates, and DNA in living cells and tissues. Among the realm of polyphenol compounds, polyphenol conjugates have been proposed as innovative materials which, by combining the advantageous properties of both the components, can increase the efficiency of antioxidants and their range of application in nutritional and biomedical fields. This work is an overview of the different class of polyphenol conjugates, which will be analyzed in terms of nutritional and biological properties, showing how these bio-conjugates will positively affect the human health. Hydrogels sensitive to electric current are usually made of polyelectrolytes and undergo erosion, swelling, de-swelling or bending in the presence of an applied electric field. The electrical conductivity of many polymeric materials used for the fabrication of biomedical devices is not high enough to achieve an effective modulation of the functional properties, and thus, the incorporation of conducting materials (e.g., carbon nanotubes and nanographene oxide) was proposed as a valuable approach to overcome this limitation. By coupling the biological and chemical features of both natural and synthetic polymers with the favourable properties of carbon nanostructures (e.g., cellular uptake, electromagnetic and magnetic behaviour), it is possible to produce highly versatile and effective nanocomposite materials. Photostability studies were performed on topical formulations containing diclofenac (DC). Niosomal gels were designed as photostabilization systems and ascorbic acid was also added to the new topical formulations because of its antioxidant property. Photodegradation tests were applied on commercial formulations containing DC and novel prepared gels, according to the ICH rules. The experiments were monitored by spectrophotometry and the data processed by multivariate curve resolution analysis to estimate the spectra and concentration profiles of evolved components. Characterization of niosomes was evaluated by size and distribution measurement, morphological analysis and encapsulation efficiency. Permeation experiments were performed across rabbit ear skin up to 24 h. Photodegradation rate of DC was found very fast in commercial formulation, with a residual content of 90% after only 4.38 min under a radiant exposure of 450 W/m 2 . Photostability resulted increased significantly when the drug was entrapped in niosomal systems. The best results were obtained by reaching a 10% degradation after 50.00 min of light exposure after incorporation of DC in niosomes in presence of 5% ascorbic acid. Moreover, niosomal gel also influenced the permeation capability of DC by enhancing the transdermal delivery of the drug. Ferrogels (or magnetic hydrogels) are cross-linked polymer networks containing magnetic nanoparticles: they are mechanically soft and highly elastic and at the same time they exhibit a strong magnetic response. Our work focuses on an combinatorial strategy to improve the efficacy of 5-Fluorouracil (5-FU) assisted chemotherapy, by developing novel multifunctional pH-sensitive ferrogels . We designed gels based on N , N ′-dimethylacrylamide monomers polymerized in presence of methacrylic acid or 2-aminoethyl methacrylate hydrochloride, containing ferro-nanoparticles. The influence of polymeric matrix composition and exposition to magnetic field (MF) on swelling behavior and drugs release were investigated at pH 7.4 and 5. In particular, the magnetic field was obtained by using permanent magnetic bar (0.25 T) or electromagnet (0.5 and 1.2 T), with the aim to analyze quantitatively the magnetic effects. A strong influence of the magnetic field on ferrogels properties have been observed. Swelling analysis indicated a dependence on both pH and network composition, reaching a maximum at pH 7.4, for formulations containing methacrylic acid, while the application of MF appeared to decrease the swelling percentages. Release profiles of 5-FU showed effective modulation in release by application of MF: drug release is always higher in the presence of a magnetic field and generally increases with its intensity. The combining effect of pH sensitive properties and application of MF improved the performance of the systems. Cocoa contains many compounds such as biogenic amines (BAs), known to influence consumer health. Spermidine, spermidine, putrescine, histamine, tyramine, β-phenylethylamine, cadaverine and serotonine have been found in several cocoa-based products using HPLC with UV detection after derivatisation with dansyl-chloride. Once optimised in terms of linearity, percentage recovery, LOD, LOQ and repeatability, this method was applied to real samples. Total concentrations of BAs ranged from 5.7 to 79.0 µg g−1with wide variations depending on the type of sample. BAs present in all samples were in decreasing order: histamine (1.9–38.1 µg g−1) and tyramine (1.7–31.7 µg g−1), while putrescine (0.9–32.7 µg g−1), spermidine (1.0–9.7 µg g−1) and spermidine (0.6–9.3 µg g−1) were present in most of the samples. Cadaverine, serotonine and β-phenylethylamine were present in a few samples at much lower concentrations. Organic samples always contained much lower levels of BAs than their conventional counterparts and, generally speaking, the highest amounts of BAs were found in the most processed products. With the aim to modulate the flavonoid antioxidant properties, catechin and quercetin were covalently inserted into hydrogel networks by free radical grafting procedure. The effective conjugation of both flavonoids into the polymer chains was checked by FT-IR analyses. Water uptake measurements and calorimetric analyses proved the pH- and thermo-responsiveness. The effect of temperature on antioxidant properties were evaluated as scavenging ability and redox performance, while the determination of the ability to preserve folic acid from UV degradation proved the pH responsiveness. The aim of this study was to monitor the fatty acids and biogenic amines (BAs) profiles in mullet ( Mugil Cephalus ) and tuna ( Thunnus thynnus ) roe (bottarga) during storage (0–180 days) at 4 °C. Fatty acids were analyzed by gas chromatography (GC) as methyl ester derivatives while BAs were investigated using reverse-phase liquid chromatography (RP-LC) as dansyl derivatives. The content of total fatty acids decreased with time in mullet bottarga (BM) while in tuna bottarga (BT) the PUFA value showed a slight decrease up to 30 days followed by an intense reduction after 90 and 180 days. An increasing content of BAs was observed with time in all investigated samples without reaching hazardous concentrations. In general, the BAs concentrations were in decreasing order: tyramine, putrescine, cadaverine and histamine. In BM, phenylethylamine was always present, representing a possible discriminating compound as it was absent in tuna bottarga at any time of storage. A Pearson correlation matrix showed that lipid and protein degradation products were strongly and significantly correlated over the considered storage time implying that lipid oxidation processes are in some way linked to the chemical production of BAs. Monodispersed polymersomes based on a hydrophobically-modified protein with dimensional range between 154 and 254 nm were prepared employing a water addition/solvent evaporation method. Gelatin hydrolyzate and PEG 40 -stearate, acting as hydrophilic and hydrophobic blocks, respectively, underwent radical coupling and three materials with different hydrophilic/lipophilic ratio were obtained by varying the amount of PEG 40 -stearate in the reaction feed. Critical aggregation concentrations of each amphiphilic polymer were found ranged from 3.9 to 10.2 μg mL −1 , with lower values recorded for conjugate with lower hydrophilic/lipophilic ratio. Anticancer drug methotrexate was loaded into the vesicular systems with efficiency strictly dependent on the gelatin hydrolyzate content and the in vitro releasing profile assessed and analyzed by suitable mathematical models. To prove the applicability of the proposed delivery vehicle, the biocompatibility properties were assessed in normal human lung fibroblasts cells MRC-5, while the anticancer efficiency was tested on H1299 lung cancer cells, proving that the drug encapsulation into the polymersomes dramatically reduced the cytotoxicity on healthy cells while preserving its efficiency in killing cancer cells. Eight biogenic amines (spermine, spermidine, putrescine, histamine, tyramine, phenylethylamine, cadaverine and serotonin) were determined by LC-UV after derivatization with dansyl-chloride in both ground coffee and coffee beverages obtained by different methods. In ground coffee, the most relevant amine was PUT, followed by SPD, HIS, TYR, CAD, SPM, PHE, and SER, with the total BAs content decreasing as the roasting degree increased. In coffee brews, the order was PUT, SPM, TYR, CAD, SPD, PHE, HIS, and SER, but at a very low level in comparison with the amount of BAs determined in roasted ground coffee. Hybrid materials with enhanced antibacterial activity were prepared by incorporation of carbon nanotubes within gelatin-fluoroquinolones bioconjugates. Thermo-responsive hydrogel films, synthesized by UV-initiated radical polymerization, are proposed as delivery devices for non-steroidal anti-inflammatory drugs (Diclofenac sodium and Naproxen). N-isopropylacrylamide and N,N′-ethylenebisacrylamide were chosen as thermo-sensitive monomer and crosslinker, respectively. Infrared spectroscopy was used to assess the incorporation of monomers into the network, and the network density of hydrogel films was found to strictly depend on both feed composition and film thickness. Calorimetric analyses showed negative thermo-responsive behaviour with shrinking/swelling transition values in the range 32.8–36.1 °C. Equilibrium swelling studies around the LCST allowed the correlation between the structural changes and the temperature variations. The mesh size, indeed, rapidly changed from a collapsed to a swollen state, with beneficial effects in applications such as size-selective permeation or controlled drug delivery, while the crosslinking degree, the film thickness, and the loading method deeply influenced the drug release profiles at 25 and 40 °C. The aim of this study was to improve the transdermal permeation of sulfadiazine sodium, employing synergistic combination of surfactants (in the form of niosomes) and additives with different number of hydroxylic groups, (following referred to as “alcohol”), as component of the bilayer. In particular the effect of different concentration of each alcohol (ethanol, propylene glycol or glycerol, from 5%, to 40% v/v) on niosomes size and distribution, drug entrapment efficiencies and ex vivo drug percutaneous permeation were evaluated, identifying formulations giving the best performances. The findings revealed that the presence of alcohol critically affect the physico-chemical properties of niosomes, with regards to dimensions, drug encapsulation and permeation. Vesicular size increased with the amount of alcohol and at the same alcohol concentration, follow the sequence ethanol > propylene glycol > glycerol. Loaded niosomes were larger than empty ones. Low E % values were found for ethanol, even less in propylene glycol and glycerol based samples, confirming that the chemical structure of the alcohol and its physico-chemical properties, affected the sulfadiazine entrapment efficiency. The comparative evaluation of percutaneous permeation profiles showed that the cumulative amount of permeated drug increases with alcohol concentration up to 20% v/v. Higher concentration (40% v/v) resulted in a strong decrease of the potential skin permeation. Best performances were obtained with glycerol. In all cases ex vivo sulfadiazine percutaneous permeations are controlled and improved respect to the corresponding free drug solutions and traditional niosomes used as controls. A new approach for the synthesis of hydrogel films with thermo-sensitive enzymatic activity is reported. Pepsin (PEP) was covalently immobilized on thermo-responsive hydrogels by radical polymerization in the presence of N-isopropylacrylamide and poly-(ethylene glycol) dimethacrylate 750, acting as functional monomer and crosslinking agent, respectively. Hydrogels showing lower critical solution temperatures between 32.9 and 36.1 °C were synthesized by UV-irradiation of reaction batches differing in the PEP/monomers ratio. The derivatization degree of the hydrogels was expressed as mg of PEP per gram of matrix and found to be in the range of 6 to 11% as assessed by Lowry method. Scanning electron microscopy analysis and water affinity evaluation allowed to highlight the porous morphology and thermo-responsivity of hydrogels as a function of temperature. Using bovine serum albumin as a substrate, kinetics parameters were determined by Lineweaver–Burk plots and the catalyst efficiency evaluated. The use of biologics, polymers, silicon materials, carbon materials, and metals has been proposed for the preparation of innovative drug delivery devices. One of the most promising materials in this field are the carbon-nanotubes composites and hybrid materials coupling the advantages of polymers (biocompatibility and biodegradability) with those of carbon nanotubes (cellular uptake, stability, electromagnatic, and magnetic behavior). Objectives In this work, a new polymeric microparticle system based on gelatin covalently bound to isoniazid (ISN) and containing rifampicin (RFP) was prepared by spray-drying technique. Microparticle aptitude to nebulisation and their capability of interacting with A549, alveolar basal epithelial cells, were evaluated in vitro. Methods Microparticles were obtained by spray drying, and their morphology, size, zeta potential, thermotropic behaviour and nebulisation ability were evaluated. Key findings Microparticles were positively charged with a mean size of 4.88 ± 0.3 μm. Microspheres were able to incorporate both RFP and ISN: encapsulation efficiency was 51 ± 6% and 22 ± 1%, respectively. X-ray diffraction study showed a new extensive and flattened diffraction peak providing evidence that the drugs were dispersed into the microparticles. Differential scanning calorimetry analysis confirmed effective interactions between gelatin and drug molecules by the presence of new transition peaks. Fifty-nine per cent of used microparticles were aerosolised. In-vitro toxicity studies on A549 alveolar basal epithelial cells showed that microparticles decreased cytotoxicity in comparison with the RFP solution. Laser scanning confocal microscopy observation confirmed that fluorescent probes delivered by microparticles are efficiently internalised in A549 cells. Abstract: This study has the goal to evaluate the stability of B complex vitamins, loaded into catechin-based crosslinked hydrogels, in aqueous medium at accelerated conditions of oxidative stress. The formulations were prepared by coupling polyacrylic acid with catechin in the presence of dicycloexylcarbodiimide. Folic acid (FA) and Thiamine (TH) were chosen within the B complex vitamins because of the well-known oxidative degradation characteristics. The protective role of the hydrogels against the oxidation damage was monitored inducing oxidative stress, by means of UV irradiation and t-butyl hydroperoxide treatment for FA and TH, respectively. The characterization of the hydrogels was obtained by morphological, calorimetric, FT-IR analyses, antioxidant assays and evaluation of the swelling behavior in aqueous media at different pH. The experimental results confirmed the suitability of the hydrogels as pH-responsive devices, furthermore the presence of oxidative condition proved the protective effect of the hydrogels. Catechin-based polymers gave a pronounced improvement in the stability of vitamins compared to hydrogels prepared by the coupling reaction of polyacrylic acid in the presence of 1,6-hexandiol. Abstract: A three-functional nanocomposite was prepared by radical polymerization of methacrylic acid around carbon nanotubes in the presence of Quercetin as biologically active molecule and proposed as new anticancer therapeutic. The so-obtained hybrid material was characterized by FT-IR, Raman, SEM, TEM analyses, while the functionalization degree of 2.33mg of Quercetin per g of composite was assessed by Folin–Ciocalteu test. Antioxidant test (DPPH and ABTS) showed that the covalent coupling did not interfere with the antioxidant properties of the flavonoid, while the anticancer activity was greatly enhanced with a recorded IC50 value much lower than free Quercetin. Abstract: Novel spherical hybrid hydrogels composed of gelatin and multi-walled carbon nanotubes were synthesized by emulsion polymerization in the presence of sodium methacrylate and N,N′-ethylenebisacrylamide, and proposed as drug delivery microspheres for the electro-responsive release of Diclofenac sodium salt. Different amounts of nanotubes (up to 35% by weight) were covalently inserted into the polymeric network in order to determine the percentage conferring the highest electric sensitivity to the composite microspheres. Characterization of hydrogels was performed by electrical, morphological and calorimetric analyses, Raman measurements, biocompatibility assays and evaluation of the swelling behaviors in water (pH 1.0 and pH 7.4) with and without the application of an external electric field. Native gelatin, N, N′-ethylenebisacrylamide, and sodium methacrylate were inserted into a spherical crosslinked structure by a solvent-free emulsion polymerization method, in which sunflower seed oil containing different amounts of lecithin was selected as continuous phase. Nanogels were characterized by morphological analysis, particle size distribution, and determination of swelling degree. Different dimensional distributions (100-500 nm) and water affinities were obtained by varying the amount of surfactant in the polymerization feed. Nanogels were non-toxic on human bone marrow mesenchymal stromal cells and enzymatically stable in the gastric tract, with weight losses ranging from 58 to 20 % in pancreatin solution. Release profiles of diclofenac sodium salt from the nanogels were evaluated at different pH and found to depend on crosslinking degree and drug-polymer interactions; while in pancreatin solution, a complete release of the drug was observed. Abstract: A new hybrid material made of gelatin, catechin and carbon nanotubes was prepared by the non-covalent incorporation of carbon nanotubes (CNTs) into a gelatin–catechin covalent conjugate. The composite materials was tested by means of determination of the dispersion stability in water and the functionalization degree was assessed by the Folin–Ciocalteu method, finding a 0.9mg of CT/g of protein conjugate. Subsequently, the complete retention of the antioxidant properties of the flavonoid after incorporation into the composite was proved by DPPH and ABTS assays and IC50 values of 5.74mgmL−1 and 0.39mgmL−1 were recorded. The presence of CNT into the materials did not interfere with the scavenging activities of the catechin. Finally, the anticancer activity on HeLa cancer cells was evaluated and a considerable increase in the therapeutic activity of the flavonoid was recorded moving from the free to the conjugated form in the presence of CNT, while in absence of CNT a reduction of the efficiency was observed. Multi-walled carbon nanotubes were chemically functionalized with methacrylic acid and methacrylated bovine serum albumin by free radical grafting reaction to obtain novel nanocomposites. The nanotubes were synthesized by aerosol-assisted chemical vapor deposition, and then the monomers were directly grafted by the action of hydrogen peroxide/ascorbic acid redox pair which allows operating in water-compatible and eco-friendly environment without the generation of any toxic reaction by-product. A multi-technique approach was used to evaluate the effectiveness of the grafting process employing Fourier transform infrared, Raman, X-ray photoelectron spectroscopy, scanning electron microscopy, transmission electron microscopy, and thermogravimetric analyses. Abstract: The paper presents the application of liquid chromatography coupled with evaporative light scattering detector (LC-ELSD) for the determination of biogenic amines in different cheese samples, as their presence and relative amounts give useful information about freshness, level of ripening and quality of storage. Forty samples from different types of milk – hard-ripened, ripened and unripened – were considered. Results showed that the amine contents varied in relation to the manufacturing process, the highest concentration being in hard-ripened cheeses followed by ripened and then unripened. In hard-ripened cheeses amines were β-phenylethylamine (PHE) (69.8–136.6mgkg−1), tyramine (TYR) (19.7–147.1mgkg−1), spermidine (SPD) (nd–73.1mgkg−1), cadaverine (CAD) (nd–64.7mgkg−1), histamine (HIS) (17.6–48.2mgkg−1), spermine (SPM) (nd–47.4mgkg−1), putrescine (PUT) (nd–44.1mgkg−1) and agmatine (AGM) (nd–4.2mgkg−1); while in ripened cheese TYR (nd–116.7mgkg−1), PUT (nd–82.9mgkg−1), HIS (nd–57.7mgkg−1), PHE (nd–51.1mgkg−1), SPD (nd–31.5mgkg−1), CAD (nd–30.7mgkg−1), SPM (nd–26.9mgkg−1) and AGM (nd–4.8mgkg−1). The purpose of this work was the synthesis, characterization and efficacy evaluation of new biocompatible antioxidant polymers linking trans-ferulic acid or a-lipoic acid. In particular, ferulic or lipoic acid were introduced in the preformed polymeric backbone. The new antioxidant biopolymers were characterized by Fourier transform infrared spectroscopy and gel permeation chromatography. The degree of functionalization (moles of antioxidant per gram of polymer) was determined by the Gaur-Gupta method for free amino group determination and by the Folin method for the phenolic groups. Their ability to inhibit lipid peroxidation were estimated in rat liver microsomal membranes induced in vitro by tert-BOOH (tert-butyl hydroperoxide), as a source of free radicals. The DPPH (1,1-diphenyl-2-picrylhydrazyl) radical-scavenging effect was also evaluated. The obtained systems, with different solubility, showed strong antioxidant and antiradical activities, suggesting potential use as packaging materials for foods, cosmetics, pharmaceuticals and personal care products. Moreover, the cytotoxicity of the synthesized polymers was also evaluated on Caco-2 cell cultures in order to verify their biocompatibility when exposed to an absorptive epithelial cell line. Purpose: A polysaccharide-flavonoid conjugate was developend and proposed for the treatment of pancreatic ductal adenocarcinoma (PDAC). Methods: The conjugate was synthesized by free radical grafting reaction between catechin and dextran. The chemical characterization of the conjugate was obtained by UV-Vis, 1H-NMR, FT-IR and GPC analyses, while the functionalization degree was determined by the Folin-Ciocalteu assay. The biological activity of the catechin-dextran conjugate was tested on two different cell lines derived from human pancreatic cancer (MIA PaCa-2 and PL45 cells), and the toxicity towards human pancreatic nestin-expressing cells evaluated. Results: Both the cancer cell lines are killed when exposed to the conjugate, and undergo apoptosis after the incubation with catechin-dextran which resulted more effective in killing pancreatic tumor cells compared to the catechin alone. Moreover, our experimental data indicate that the conjugate was less cytotoxic to human pancreatic nestin-expressing cells which are considered a good model of non-neoplastic pancreatic cells. Abstract: An innovative biocompatible approach for the selective recovery of gallic acid (GA) from actual site olive mill wastewaters (OMWs) by means of Molecularly Imprinted Polymers (MIPs) was developed. Bulky MIPs with a high selectivity for GA were synthesized by a non-covalent approach using methacrylic acid as the functional monomer and ethylene glycol dimethacrylate as the crosslinker. Imprinting efficiency was evaluated in different water/ethanol solutions by determining α and ε parameter, by using pyrogallic acid as the template analogue: the highest GA recognition, both in terms of percentage and of imprinting effect (α =32.7; ε =4.2), was observed in pure ethanol. The efficiency of the developed extraction protocol was evaluated with an actual site OMW: a selective extraction of 80% of pure GA was achieved with high accuracy and precision, as shown by RSD% (4.3%) and recovery values (between 85% and 97%). Abstract: The main target of this study was the preparation, characterization and in vitro activities evaluation of microspheres curcumin-based able to incorporate azathioprine, useful for psoriasis treatment. Curcumin was derivatized with acryloyl chloride in order to introduce polymerizable groups in its structure. The obtained polymerizable drug was characterized by Fourier transform infrared (FT-IR), to confirm ester linkages, and by 1H-NMR, to establish the functionalization degree. The ability of curcumin derivative to inhibit cell proliferation, in human breast cancer cells, gave an IC50 value for viability of 20μM. Acrylated curcumin also showed a strong antioxidant activity against lipid peroxidation induced in rat liver microsomal membranes. Then, spherical microparticles, based on curcumin derivative, were prepared by suspension radical copolymerization. The obtained microparticles were characterized by FT-IR spectroscopy, dimensional and morphological analysis. Particle size investigation revealed a mean diameter of around 5.8μm. Microspheres showed a very good swelling behavior in simulating intestinal fluids. Their azathioprine loading efficiency, determined by spectophotometric analysis was equal to 80%. Objectives A novel and simple method to synthesize antitubercular-protein conjugate by solid phase synthesis was developed employing a carboxypolystyrene resin. The aim was to covalently bind a drug with antitubercular activity, isoniazid, to a biomacromolecule, gelatin, widely used in the pharmaceutical, cosmetic and food industry. Methods Calorimetric and 1H NMR analyses were performed to verify the bond formation between the antitubercular drug and gelatin. After absorption isoniazid delivers toxic metabolites and so an oxidation test with tert-butyl hydroperoxide was performed to assess the amount of toxic metabolites released from the prodrug (gelatin linked to isoniazid), compared with isoniazid itself. Key findings Spectrophotometric analysis revealed that the protein derivative was an excellent isoniazid prodrug since there was a 40% reduction in release of toxic metabolites (isonicotinic acid) by the prodrug. The results clearly showed that antitubercular moieties, covalently linked to a natural polymer, allowed the introduction of peculiar features for specific pharmaceutical applications into the macromolecule. In addition, antitubercular activity of the new polymer was determined by Middlebrook 7H11 medium against Mycobacterium tuberculosis complex. Conclusions The new isoniazid-gelatin conjugate showed significant antitubercular activity and for this reason should be useful as an efficacious tool in the treatment of tuberculosis. Phospholipids (PLs), the most important class of polar lipids in foods, are ubiquitous compounds because they are the major components of animal and plant cell membranes. Their technological and biological properties therefore make them important in human nutrition. The efficient separation and accurate quantification of PLs can be achieved with high-performance liquid chromatography–evaporative light scattering detection (HPLC-ELSD) because ELSD is a quasi-universal detector for liquid, countercurrent, and supercritical fluid chromatography and can detect any analyte less volatile than the mobile phase. In this article, the principles of ELSD operation (i.e., nebulization of the chromatographic effluent, evaporation of the mobile phase, and measurement of the scattered light) as well as the application of LC-ELSD to determine the amount of PLs in different food matrices are reviewed. Food matrices containing PLs include milk and dairy products, meat, fish, eggs, cereals, and oils. Both the technological aspects and analytical parameters affecting the concentration and quantitative determination of PLs in food matrices are evaluated. In particular, different extraction, purification, and chromatographic conditions were extensively investigated; moreover, wherever possible, the results obtained are reported and compared with other detection methods. Abstract: The aim of this work is to design, prepare and characterize l-lysine based prodrugs capable of targeting 6-mercaptopurine to the colon, an anti-tumor and immunosuppressant drug, and 5-aminosalicylic acid (5-ASA), drug of choice for inflammatory bowel disease (IBD). More specifically, Nɛ-feruloyl-S-(6-purinyl)-l-lysine and Nɛ-acryloyl-S-(6-purinyl)-l-lysine were synthesized and then characterized by FT-IR, 1H-NMR and GC/MS spectroscopies. The ability of feruloyl derivative in inhibiting lipid peroxidation in rat liver microsomal membranes, induced in vitro by tert-butyl hydroperoxide as source of free radicals, was evaluated. Moreover, Nɛ-acryloyl-S-(6-purinyl)-l-lysine, polymerizable prodrug, was used to microspheres realization for 5-ASA release. These lasts, obtained by emulsion inverse technique, were characterized by light scattering and scanning electron microscopy (SEM) analysis. The microspheres equilibrium swelling degree was evaluated and showed good swelling behaviour in simulating colonic fluids. Results confirm the possibility that the application range of l-lysine prodrug can be extended to the treatment of intestinal diseases whose conventional therapy envisages medications with serious side effects that, thanks to this new strategy, can be minimized in an optimal way. Abstract: The central motivation for this study was to evaluate if the increased hydrophilic drug permeation across the skin, which is always observed in presence of vesicular systems, is dependent on the structural organization of niosomes, that are used to transport the active molecules, or if it is only dependent on the surfactant dual nature. To answer this question, non-ionic surfactants belonging to the class of Pluronic and sucrose esters were used both as components of niosomal systems or in the form of sub-micellar solutions. The obtained niosomes were characterized by their entrapment efficiency, size and morphology. The enhancing effect of niosomes on the ex vivo percutaneous penetration of a model drug was investigated using a Franz-type diffusion chamber and compared to that obtained by using sub-micellar solution of surfactant or achieving pretreatment of the skin with surfactants’ sub-micellar solution or empty niosomes. The results suggest that the surfactants used in this study could be considered as percutaneous permeation enhancers only when they are in the form of drug-loaded vesicular systems: no percutaneous promotion was achieved by using sub-micellar solution containing free Sulfadiazine sodium salt or performing pretreatment with empty niosomes or sub-micellar solutions of the surfactant. Abstract: This paper presents a new LC method with evaporative light scattering detection (ELSD), for the separation and determination of the biogenic amines (histamine, spermidine, spermine, tyramine, putrescine and β-phenylethylamine) which are commonly present in cheese, as their presence and relative amounts give useful information about freshness, level of maturing, quality of storage and cheese authentication. The LC–ELSD method is validated by comparison of the results with those obtained through LC–UV determination, based on a pre-column dansyl chloride derivatisation step. The obtained data demonstrate that both methods can be interchangeably used for biogenic amines determination in cheese. The new LC–ELSD method shows good precision and permits to achieve, for standard solutions, limit of detection (LOD) values ranging from 1.4 to 3.6mgL−1 and limit of quantitation (LOQ) values ranging from 3.6 to 9.3mgL−1. The whole methodology, comprehensive of the homogenization–extraction process and LC–ELSD analysis, has been applied in the analysis of a typical Calabria (Southern Italy) POD cheese, known as Caciocavallo Silano. The most aboundant amine found was histamine, followed, in decreasing order, by tyramine, spermine, putrescine, β-phenylethylamine and spermidine, for a total amount of 127mgkg−1. Abstract: A new Molecularly Imprinted Solid Phase Extraction (MISPE) protocol was developed for the selective extraction and purification of glycyrrhizic acid (GL) from liquorice roots. Non-covalent MIP were synthesized using methacrylic acid (MAA), 2-(dimethylamino)ethyl methacrilate (DMAEM) or 2-hydroxyethylmetacrylate (HEMA) as functional monomer and ethylene glycol dimethacrylate (EGDMA) as crosslinking agent. After the evaluation of the selectivity of the GL imprinted polymers, the performance of these materials as Solid Phase Extraction (SPE) sorbents was investigated. MIP having HEMA as functional monomer were found to be able to selectively extract almost 80% of GL content in liquorice roots. The proposed MISPE-HPLC procedure has good precision, thus it can be successfully used for the purification of GL from natural sources. [ABSTRACT FROM AUTHOR]
