Summary Early stress detection of crops requires a thorough understanding of the signals showing the very first symptoms of the alterations in the photosynthetic light reactions. Detection of the activation of the regulated heat dissipation mechanism is crucial to complement passively induced fluorescence to resolve ambuiguities in energy partitioning. Using leaf spectroscopy, we evaluated the capability of pigment spectral unmixing to calculate the fluorescence quantum efficiency (FQE) and simultaneously retrieve fast absorption changes in a drought and nitrogen deficiency experiment with tomato. In addition, active fluorescence measurements and pigment analyses of xanthophylls, carotenes and chlorophylls were conducted. We observed notable responses in noninvasive proximal sensing‐retrieved FQE values under stress, but as expected, these alone were not enough to identify the constraints in photosynthetic efficiency. Reflectance‐based detection of the 535‐nm peak absorption change was able to complement FQE and indicate the activation of regulated heat dissipation for both stress treatments under growing light conditions. However, further complexity in the light harvesting energy regulation needs to be accounted for when considering additional light stress. Our results underscore the potential of complementary in vivo quantitative spectroscopy‐based products in the early and nondestructive stress diagnosis of plants, marking the path for further applications. ABSTRACT Nitrogen is a crucial macroelement essential for plant growth and development. In Arabidopsis Thaliana, classical phytohormones such as auxin and cytokinin orchestrate local and systemic signalling networks coordinate plant growth and development in response to nitrogen deficiency. Nowadays, emerging signalling pathways involving small peptides like CLAVATA3/EMBRYO SURROUNDINGR REGION (CLE) and C‐TERMINALLY ENCODED PEPTIDE (CEP) and their corresponding kinase receptors, also regulate Arabidopsis' adaptation to nitrogen scarcity. Unlike Arabidopsis, which adapts to nitrogen deficiency by changing root development, legumes have the unique ability to form nitrogen‐fixing root nodules through symbiotic interactions with soil rhizobia. During the symbiotic nodulation in Medicago, CLE and CEP peptides and their receptors consist of an autoregulatory network governing the number of nodules in accordance with the soil nitrogen level. Additionally, other plant peptides, such as phytosulfokine (PSK) and root meristem growth factors (RGF), have been identified as new regulators of leguminous root nodule development under nitrogen‐limited condition. However, the precise mechanism by which these peptides coordinate nitrogen deficiency response and the development of nitrogen‐fixing organs remains to be fully elucidated. This review summarises the adaptive strategies of dicotyledons to nitrogen deficiency, with a particular focus on the regulation of Medicago nitrogen‐fixing nodule development by the peptides. Root growth, nutrient uptake, and nutrient use efficiency of a plant is influenced by the nutrient availability in its growing environment. The aim of this study was to investigate the root morphology, nutrient uptake and use efficiency of Grevillea robusta grown in nitrogen (N)- and phosphorus (P)-limited conditions. A hydroponic experiment was conducted for three months in a glasshouse with four nutrient treatments; (i) N-deficient, (ii) P-deficient, (iii) N/P-deficient (co-limitation of N and P), and (iv) control (sufficient amounts of N, P, and other nutrients in the growth medium). The composition and the concentration of each nutrient in the control solution were custom-made based on the growth requirement of G. robusta. N and P concentration in nutrient-deficient media were maintained at 2 ppm. The impact of N- or P-deficiency and their co-limitation on cluster root formation, organ-specific dry matter and nutrient accumulation, N and P uptake and their use efficiencies were measured. P-deficient conditions produced the highest number of cluster roots (24 ± 4) which was comparatively lower than those of N- and N/P-deficient conditions indicating that P-deficiency is the primary factor influencing the formation of cluster roots in G. robusta. The use efficiency of N and P in biomass formation was significantly (p < 0.05) enhanced when their uptake was reduced in response to N- and P-limitation. The efficient internal use of P by G. robusta under N- and N/P-deficiency is indicated by the significant increase in P-remobilization efficiency under these nutrient-deficient conditions. Proper crop management requires rapid detection methods for abiotic and biotic stresses to ensure plant health and yield. Hemp (Cannabis sativa L.) is an emerging economically and environmentally sustainable crop capable of yielding high biomass. Nitrogen deficiency significantly reduces hemp plant growth, affecting photosynthetic capacity and ultimately decreasing yield. When symptoms of nitrogen deficiency are visible to humans, there is often already lost yield. A real-time, non-destructive detection method, such as Raman spectroscopy, is therefore critical to identify nitrogen deficiency in living hemp plant tissue for fast, precise crop remediation. A two-part experiment was conducted to investigate portable Raman spectroscopy as a viable hemp nitrogen deficiency detection method and to compare the technique's predictive ability against a handheld SPAD (chlorophyll index) meter. Raman spectra and SPAD readings were used to train separate nitrogen deficiency discrimination models. Raman scans displayed characteristic spectral markers indicative of nitrogen deficiency corresponding to vibrational modes of carotenoids, essential pigments for photosynthesis. The Raman-based model consistently predicted nitrogen deficiency in hemp prior to the onset of visible stress symptoms across both experiments, while SPAD only differentiated nitrogen deficiency in the second experiment when the stress was more pronounced. Our findings add to the repertoire of plant stresses that hand-held Raman spectroscopy can detect by demonstrating the ability to provide assessments of nitrogen deficiency. Abiotic stress factors can be detected using visible and near-infrared spectral signatures. Previous work demonstrated the potential of this technology in crop monitoring, although a large majority used vegetation indices, which did not consider the complete spectral information. This work explored the capabilities of spectral information for abiotic stress detection using supervised machine learning techniques such as support vector machine (SVM), random forest (RF), and neural network (NN). This study used avocados grown under various water treatments, maize submitted to nitrogen deficiency, and common beans under phosphorous restriction. The spectral characterization of the crops subjected to abiotic stress was studied on the visible to near-infrared (450 to 900 nm) spectrum, identifying discriminative bands and spectral ranges. Then, the advantages of using an integrated approach based on machine learning to detect abiotic stress in crops were demonstrated. Instead of relying on vegetation indices, the proposed approach used several spectral features obtained by analyzing the discriminative signature shape, applying a spectral subset band selection algorithm based on similarity, and using the minimum redundancy maximum relevance (MRMR), F-test and chi-square test ranks for feature selection. The results showed that supervised classifiers applied to the spectral features outperform the accuracies obtained from vegetation indices. The best common bean results were obtained using SVM with accuracies up to 91%; for maize and avocado, NN obtained 90% and 82%, respectively. Decreasing nitrogen (N) supply affected the normal growth of Oryza sativa (O. sativa) seedlings, reducing CO2 assimilation, stomatal conductance (gs), the contents of chlorophylls (Chl) and the ratio of Chl a/Chl b, but increasing the intercellular CO2 concentration. Polyphasic chlorophyll a fluorescence transient and relative fluorescence parameters (JIP test) results indicated that N deficiency increased Fo, but decreased the maximum quantum yield of primary photochemistry (Fv/Fm) and the maximum of the IPphase, implying that N-limiting condition impaired the whole photo electron transport chain from the donor side of photosystem II (PSII) to the end acceptor side of PSI in O. sativa. N deficiency enhanced the activities of the antioxidant enzymes, such as ascorbate peroxidase (APX), guaiacol peroxidase (GuPX), dehydro–ascorbate reductase (DHAR), superoxide dismutase (SOD), glutathione peroxidase (GlPX), glutathione reductase (GR), glutathione S-transferase (GST) and O-acetylserine (thiol) lyase (OASTL), and the contents of antioxidant compounds including reduced glutathione (GSH), total glutathione (GSH+GSSG) and non-protein thiol compounds in O. sativa leaves. In contrast, the enhanced activities of catalase (CAT), DHAR, GR, GST and OASTL, the enhanced ASC–GSH cycle and content of sulfur-containing compounds might provide protective roles against oxidative stress in O. sativa roots under N-limiting conditions. Quantitative real-time PCR (qRT-PCR) analysis indicated that 70% of the enzymes have a consistence between the gene expression pattern and the dynamic of enzyme activity in O. sativa leaves under different N supplies, whereas only 60% of the enzymes have a consistence in O. sativa roots. Our results suggested that the antioxidant system and sulfur metabolism take part in the response of N limiting condition in O. sativa, and this response was different between leaves and roots. Future work should focus on the responsive mechanisms underlying the metabolism of sulfur-containing compounds in O. sativa under nutrient deficient especially N-limiting conditions. The development of a nonconventional nitrogen fertilizer, which can be fixed in agricultural fields using decentralized renewable energy sources, presents a feasible solution for sustainable on‐site nitrogen fixation and fertilization. This study focuses on plasma‐generated dinitrogen pentoxide (N2O5) as a prospective mediator for the on‐site nitrogen fertilization, allowing nitrogen fertilization directly into culture media. Basal dinitrogen pentoxide fertilization demonstrated almost 100% dinitrogen pentoxide dissolution efficiency as nitrate in a culture medium and nitrogen fertilization effect on plant growth without explicit symptoms of damage. Top‐dressing of dinitrogen pentoxide was also an efficient method for transferring nitrogen into the soil as nitrate, which improved plant growth and suppressed nitrogen deficiency symptoms, while overdose caused adverse effects. Background: Konjac corms are known for their alkaloid content, which possesses pharmacological properties. In the primary cultivation areas of konjac, nitrogen deficiency is a common problem that significantly influences alkaloid synthesis. The impact of nitrogen deficiency on the alkaloids in konjac corms remains unclear, further complicated by the transition from mother to daughter corms during their growth cycle. Results: This study examined 21 alkaloids, including eight indole alkaloids, five isoquinoline alkaloids, and eight other types of alkaloids, along with the associated gene expressions throughout the development of Amorphophallus muelleri Blume under varying nitrogen levels. Nitrogen deficiency significantly reduced corm diameter and fresh weight and delayed the transformation process. Under low nitrogen conditions, the content of indole alkaloids and the expression of genes involved in their biosynthesis, such as tryptophan synthase (TRP) and tryptophan decarboxylase (TDC), exhibited a substantial increase in daughter corms, with fold changes of 61.99 and 19.31, respectively. Conversely, in the mother corm, TDC expression was markedly reduced, showing only 0.04 times the expression level observed under 10 N treatment. The patterns of isoquinoline alkaloid accumulation in corms subjected to nitrogen deficiency were notably distinct from those observed for indole alkaloids. The accumulation of isoquinoline alkaloids was significantly higher in mother corms, with expression levels of aspartate aminotransferase (GOT), chorismate mutase (CM), tyrosine aminotransferase (TAT), and pyruvate decarboxylase (PD) being 4.30, 2.89, 921.18, and 191.40 times greater, respectively. Conversely, in daughter corms, the expression levels of GOT and CM in the 0 N treatment were markedly lower (0.01 and 0.83, respectively) compared to the 10 N treatment. Conclusions: The study suggests that under nitrogen deficiency, daughter corms preferentially convert chorismate into tryptophan to synthesize indole alkaloids, while mother corms convert it into tyrosine, boosting the production of isoquinoline alkaloids. This research provides valuable insights into the mechanisms of alkaloid biosynthesis in A. muelleri and can aid in developing nitrogen fertilization strategies and in the extraction and utilization of alkaloids. This article discusses the relationship between root cortical parenchyma wall width (CPW) and plant functioning under drought conditions. The study found that thicker CPW in maize roots promotes growth and yield under suboptimal water and nitrogen supply. The authors used a functional-structural root anatomy model to predict the effects of increased CPW on root respiration and conducted field experiments to confirm their findings. Optimizing plant nitrogen (N) usage and inhibiting N leaching loss in the soil–crop system is crucial to maintaining crop yield and reducing environmental pollution. This study aimed at identifying quantitative trait loci (QTLs) and differentially expressed genes (DEGs) between two N treatments in order to list candidate genes related to nitrogen-related contrasting traits in tomato varieties. We characterized a genetic diversity core-collection (CC) and a multi-parental advanced generation intercross (MAGIC) tomato population grown in a greenhouse under two nitrogen levels and assessed several N-related traits and mapped QTLs. Transcriptome response under the two N conditions was also investigated through RNA sequencing of fruit and leaves in four parents of the MAGIC population. Significant differences in response to N input reduction were observed at the phenotypic level for biomass and N-related traits. Twenty-seven QTLs were detected for three target traits (leaf N content, leaf nitrogen balance index, and petiole NO3− content), 10 and six in the low and high N condition, respectively, while 19 QTLs were identified for plasticity traits. At the transcriptome level, 4752 and 2405 DEGs were detected between the two N conditions in leaves and fruits, respectively, among which 3628 (50.6%) in leaves and 1717 (71.4%) in fruit were genotype specific. When considering all the genotypes, 1677 DEGs were shared between organs or tissues. Finally, we integrated DEG and QTL analyses to identify the most promising candidate genes. The use of plant biostimulants (PBs), integrated with good agronomic practices, offers a sustainable solution.The study explores the potential use of exhausted cell culture media from plant cell cultures (Nicotiana sp. and Gardenia sp.) as PBs, assessing their role in enhancing plant physiological status and NUE. It also compares their effectiveness against commercial PBs.Two experiments (Experiments 1 and 2) were conducted using cherry tomato plants. In the first one, the two media were applied at different concentrations as a foliar spray to define the optimal dose. This outcome was used to set up Experiment 2, in which both media were compared to commercial PBs under optimal and reduced N supply. Measurements included plant growth, chlorophyll (Chl) content, and NUE indices. Moreover, Experiment 2 examined gene expression related to N assimilation, transport, and response to oxidative stress.Both experiments demonstrated a significant increase in leaf area (≈11%) and aboveground biomass (≈13%) using exhausted media. Furthermore, Experiment 2 demonstrated enhancements in Chl content (≈10%) and plant N accumulation (≈20%). These findings indicate that exhausted media has a comparable efficacy to commercial PBs.The study underscores the practical viability of exhausted cell culture media as effective PBs for tomato growth in both N conditions. Abiotic stresses such as nitrogen deficiency, drought, and salinity significantly impact coconut production, yet the molecular mechanisms underlying coconut's response to these stresses are poorly understood. MYB proteins, a large and diverse family of transcription factors (TF), play crucial roles in plant responses to various abiotic stresses, but their genome-wide characterization and functional roles in coconut have not been comprehensively explored. This study identified 214 CnMYB genes (39 1R–MYB, 171 R2R3–MYB, 2 3R–MYB, and 2 4R–MYB) in the coconut genome. Phylogenetic analysis revealed that these genes are unevenly distributed across the 16 chromosomes, with conserved consensus sequences, motifs, and gene structures within the same subgroups. Synteny analysis indicated that segmental duplication primarily drove CnMYB evolution in coconut, with low nonsynonymous/synonymous ratios suggesting strong purifying selection. The gene ontology (GO) annotation of protein sequences provided insights into the biological functions of the CnMYB gene family. CnMYB47/70/83/119/186 and CnMYB2/45/85/158/195 were identified as homologous genes linked to nitrogen deficiency, drought, and salinity stress through BLAST, highlighting the key role of CnMYB genes in abiotic stress tolerance. Quantitative analysis of PCR showed 10 CnMYB genes in leaves and petioles and found that the expression of CnMYB45/47/70/83/85/119/186 was higher in 3-month-old than one-year-old coconut, whereas CnMYB2/158/195 was higher in one-year-old coconut. Moreover, the expression of CnMYB70, CnMYB2, and CnMYB2/158 was high under nitrogen deficiency, drought, and salinity stress, respectively. The predicted secondary and tertiary structures of three key CnMYB proteins involved in abiotic stress revealed distinct inter-proteomic features. The predicted interaction between CnMYB2/158 and Hsp70 supports its role in coconut's drought and salinity stress responses. These results expand our understanding of the relationships between the evolution and function of MYB genes, and provide valuable insights into the MYB gene family's role in abiotic stress in coconut. Compensatory effects are common biological phenomena in nature. In this study, we investigated the changes in root nitrogen uptake, root morphological and physiological responses, and changes in the rhizosphere soil microbial communities of indica and japonica rice during a nitrogen deficiency‐sensitive period and an effective compensation period with double the nitrogen supply. We conducted a bucket experiment using Suxiu 867 (a japonica rice variety) and Yangxian You 918 (an indica rice variety). Treatments included CK (constant distribution of nitrogen fertilizer at each growth stage, represented by CK867 and CK918) and NDC (nitrogen deficiency in the tillering stage, double nitrogen application in the ear differentiation stage to compensate, represented by NDC867 and NDC918) variations. Both varieties presented the highest δ15N and 15N abundances and Ndff (refers to the proportion of nitrogen in a plant's body that comes directly from the fertilizer applied.) in rice under the NDC treatment. Metagenomic sequencing of rhizospheric soil showed that the dominant bacterial groups at the phylum level among each treatment were Actinobacteria, Proteobacteria, Chloroflexi, Acidobacteria, Gemmatimonadetes, and Nitrospirae. The rhizosphere of indica rice was more enriched with the microbial communities involved in nitrogen metabolism, which contributed to higher nitrogen utilization efficiency. A correlation‐based network was constructed and provides insights into the formation of nitrogen deficiency compensation effects and contributes to the enhancement of nitrogen uptake and utilization efficiency in rice production. Understanding plant responses to individual stresses does not mean that we understand real-world situations, where stresses usually combine and interact. These interactions arise at different levels, from stress exposure to the molecular networks of the stress response. Here, we built an in-depth multiomic description of plant responses to mild water (W) and nitrogen (N) limitations, either individually or combined, among 5 genetically different Arabidopsis (Arabidopsis thaliana) accessions. We highlight the different dynamics in stress response through integrative traits such as rosette growth and the physiological status of the plants. We also used transcriptomic and metabolomic profiling during a stage when the plant response was stabilized to determine the wide diversity in stress-induced changes among accessions, highlighting the limited reality of a "universal" stress response. The main effect of the W × N interaction was an attenuation of the N-deficiency syndrome when combined with mild drought, but to a variable extent depending on the accession. Other traits subject to W × N interactions are often accession specific. Multiomic analyses identified a subset of transcript–metabolite clusters that are critical to stress responses but essentially variable according to the genotype factor. Including intraspecific diversity in our descriptions of plant stress response places our findings in perspective. Proper nitrogen management in crops is crucial to ensure optimal growth and yield maximization. While hyperspectral imagery is often used for nitrogen status estimation in crops, it is not feasible for real-time applications due to the complexity and high cost associated with it. Much of the research utilizing RGB data for detecting nitrogen stress in plants relies on datasets obtained under laboratory settings, which limits its usability in practical applications. This study focuses on identifying nitrogen deficiency in maize crops using RGB imaging data from a publicly available dataset obtained under field conditions. We have proposed a custom-built vision transformer model for the classification of maize into three stress classes. Additionally, we have analyzed the performance of convolutional neural network models, including ResNet50, EfficientNetB0, InceptionV3, and DenseNet121, for nitrogen stress estimation. Our approach involves transfer learning with fine-tuning, adding layers tailored to our specific application. Our detailed analysis shows that while vision transformer models generalize well, they converge prematurely with a higher loss value, indicating the need for further optimization. In contrast, the fine-tuned CNN models classify the crop into stressed, non-stressed, and semi-stressed classes with higher accuracy, achieving a maximum accuracy of 97% with EfficientNetB0 as the base model. In traditional farming, fertilizers are often used without precision, resulting in unnecessary expenses and potential damage to the environment. This study introduces a new method for accurately identifying macronutrient deficiencies in Rhodena lettuce crops. We have developed a four-stage process. First, we gathered two sets of data for lettuce seedlings: one is composed of color images and the other of point clouds. In the second stage, we employed the interactive closest point (ICP) method to align the point clouds and extract 3D morphology features for detecting nitrogen deficiencies using machine learning techniques. Next, we trained and compared multiple detection models to identify potassium deficiencies. Finally, we compared the outcomes with traditional lab tests and expert analysis. Our results show that the decision tree classifier achieved 90.87% accuracy in detecting nitrogen deficiencies, while YOLOv9c attained an mAP of 0.79 for identifying potassium deficiencies. As the global climate continues to change, plants will increasingly experience abiotic stress(es). Stomata on leaf surfaces are the gatekeepers to plant interiors, regulating gaseous exchanges that are crucial for both photosynthesis and outward water release. To optimise future crop productivity, accurate modelling of how stomata govern plant–environment interactions will be crucial. Here, we synergise optical and thermal imaging data to improve modelled transpiration estimates during water and/or nutrient stress (where leaf N is reduced). By utilising hyperspectral data and partial least squares regression analysis of six plant traits and fluxes in wheat (Triticum aestivum), we develop a new spectral vegetation index; the Combined Nitrogen and Drought Index (CNDI), which can be used to detect both water stress and/or nitrogen deficiency. Upon full stomatal closure during drought, CNDI shows a strong relationship with leaf water content (r2 = 0.70), with confounding changes in leaf biochemistry. By incorporating CNDI transformed with a sigmoid function into thermal‐based transpiration modelling, we have increased the accuracy of modelling water fluxes during abiotic stress. These findings demonstrate the potential of using combined optical and thermal remote sensing‐based modelling approaches to dynamically model water fluxes to improve both agricultural water usage and yields. Process analytical technologies (PATs) have transformed the beverage production management by providing real-time monitoring and control of critical process parameters through non-destructive measurements, such as those obtained with infrared (IR) spectroscopy and enabling process readjustment if necessary. New requirements in the analysis of beverages call for new methods, so in this article, we propose a method based on the construction of multivariate statistical process control (MSPC) charts from a new dissimilarity index (the evolving window dissimilarity index, EWDI) to monitor fermentation processes. The EWDI was applied to monitor wine alcoholic fermentation, the biochemical transformation of sugars into ethanol. Small-scale fermentations were carried out and analyzed using a portable mid-infrared spectrometer. In some of them, process deviations due to nitrogen deficiency or temperature changes were intentionally promoted to evaluate the performance of the EWDI. The MSPC charts build by using the fermentations carried out under normal operating conditions allowed identifying deviations of the fermentation in its early stages. Furthermore, the shape of the EWDI curve over time provides insights about the specific type of deviation occurring. These results show the potential of this new approach to improve the monitoring and control of key process stages in biochemical processes in the food industry, which allows maximizing quality and minimizing losses. Microalgae have a great potential to change their metabolism pathway under stress condition, and nitrogen deficiency is an effective technique for maximizing lipids synthesis. This study focus on the growth performance, and biochemical composition of Tetradesmus obliquus (Turpin) M.J. Wynne cultured in different nitrogen concentrations (BG 11-Blue-Green Medium, 50% N+, 50% N-, 75% N+, 75% N-). Specific growth rate and protein content of T. obliquus decreased with nitrogen deficiency, but its lipid content was increased. Maximum specific growth rate (0.20-0.21) and protein content (47.82%, 47.62% DW-dried weight) values were obtained in 75% N+, 50% N+ medium. The highest lipid percentage was obtained in 50% N- medium. The biochemical composition of T. obliquus changes based on nitrogen concentration. Polyunsaturated fatty acids (PUFAs) are more abundant (49.69%) in this microalga under deficient nitrogen. To obtain more lipid and PUFAs in T. obliquus, N deficiency is an effective method, but it is not applicable to protein content. T. obliquus can be used as an alternative nutritional supplement with its high PUFAs content. This study documented the contribution of precise positioning involving a global navigation satellite system (GNSS) and a real-time kinematic (RTK) system in unmanned aerial vehicle (UAV) photogrammetry, particularly for establishing the coordinate data of ground control points (GCPs). Without augmentation, GNSS positioning solutions are inaccurate and pose a high degree of uncertainty if such data are used in UAV data processing for mapping. The evaluation included a comparative assessment of sample coordinates involving RTK and an ordinary GPS device and the application of precise GCP data for UAV photogrammetry in field crop research, monitoring nitrogen deficiency stress in maize. This study confirmed the superior performance of the RTK system in providing positional data, with 4 cm bias as compared to 311 cm with the non-augmented GNSS technique, making it suitable for use in agronomic research involving row crops. Precise GCP data in this study allow the UAV-based Normalized Difference Red-Edge Index (NDRE) data to effectively characterize maize crop responses to N nutrition during the growing season, with detailed analyses revealing the causal relationship in that a compromised optimum canopy chlorophyll content under limiting nitrogen environment was the reason for reduced canopy cover under an N-deficiency environment. Nitrogen (N) is essential for sugar beet (Beta vulgaris L.), a highly N-demanding sugar crop. This study investigated the morphological, subcellular, and microRNA-regulated responses of sugar beet roots to low N (LN) stress (0.5 mmol/L N) to better understand the N perception, uptake, and utilization in this species. The results showed that LN led to decreased dry weight of roots, N accumulation, and N dry matter production efficiency, along with damage to cell walls and membranes and a reduction in organelle numbers (particularly mitochondria). Meanwhile, there was an increase in root length (7.2%) and branch numbers (29.2%) and a decrease in root surface area (6.14%) and root volume (6.23%) in sugar beet after 7 d of LN exposure compared to the control (5 mmol/L N). Transcriptomics analysis was confirmed by qRT-PCR for 6 randomly selected microRNAs, and we identified 22 differentially expressed microRNAs (DEMs) in beet root under LN treatment. They were primarily enriched in functions related to binding (1125), ion binding (641), intracellular (437) and intracellular parts (428), and organelles (350) and associated with starch and sucrose metabolism, tyrosine metabolism, pyrimidine metabolism, amino sugar and nucleotide sugar metabolism, and isoquinoline alkaloid biosynthesis, as indicated by the GO and KEGG analyses. Among them, the upregulated miR156a, with conserved sequences, was identified as a key DEM that potentially targets and regulates squamosa promoter-binding-like proteins (SPLs, 104889216 and 104897537) through the microRNA-mRNA network. Overexpression of miR156a (MIR) promoted root growth in transgenic Arabidopsis, increasing the length, surface area, and volume. In contrast, silencing miR156a (STTM) had the opposite effect. Notably, the fresh root weight decreased by 45.6% in STTM lines, while it increased by 27.4% in MIR lines, compared to the wild type (WT). It can be inferred that microRNAs, especially miR156, play crucial roles in sugar beet root's development and acclimation to LN conditions. Trehalose-6-phosphate phosphatase (TPP), a key enzyme for trehalose biosynthesis in plants, plays a pivotal role in the growth and development of higher plants, as well as their adaptations to various abiotic stresses. Employing bioinformatics techniques, 45 TPP genes distributed across 17 chromosomes were identified with conserved Trehalose-PPase domains in the peanut genome, aiming to screen those involved in salt tolerance. Collinearity analysis showed that 22 TPP genes from peanut formed homologous gene pairs with 9 TPP genes from Arabidopsis and 31 TPP genes from soybean, respectively. Analysis of cis-acting elements in the promoters revealed the presence of multiple hormone- and abiotic stress-responsive elements in the promoter regions of AhTPPs. Expression pattern analysis showed that members of the TPP gene family in peanut responded significantly to various abiotic stresses, including low temperature, drought, and nitrogen deficiency, and exhibited certain tissue specificity. Salt stress significantly upregulated AhTPPs, with a higher number of responsive genes observed at the seedling stage compared to the podding stage. The intuitive physiological effect was reflected in the significantly higher accumulation of trehalose content in the leaves of plants under salt stress compared to the control. Background: Yield gap is distance between potential yield and attainable yield (yield gap= potential yield-attainable yield) in order to increase the crop yield and the goal of sustainable agriculture. Methods: This study was conducted in Bam, Narmashir and Fahraj counties in 219 separate farms (79 farms in Bam, 72 farms in Narmashir and 68 farms in Fahraj) during the years 2019 and 2020 to investigate the yield gap of wheat and determine the limiting factors and their contributions to yield reduction, using the Comparative Performance Analysis (CPA) method. All information related to soil factors, management factors, climatic factors and agronomic traits (53 variables) were measured and recorded. The relationship between wheat yield and all variables was examined using a stepwise multiple regression method. A production model was determined for soil and management factors. Result: The results showed a yield gap between actual yield and potential yield in the regions of Bam, Narmashir and Fahraj, which were 1770.8, 1817.61 and 1605 kg per hectare, respectively. Since the actual yield in these regions was 4248, 4228 and 3161 kg per hectare, the yield gap was observed to be 33.6%, 30% and 29.4%, respectively. The effective factors in reducing the yield in the Bam region included water shortage (19.48%), nitrogen deficiency (16.09%), delayed planting (15.40%), soil available phosphorus deficiency (10.14%), weed density (10.8%), soil organic carbon (13.59%) and inappropriate plant density (14.47%). In the Narmashir region, water shortage (26.68%), nitrogen deficiency (18.86%), soil organic carbon (11.78%), inappropriate plant density (12.43%), wheat aphid (19.58%) and weed density (10.64%) caused yield reduction. To investigate the contents of carbon, nitrogen, and phosphorus in tea plantation soils and their ecological stoichiometric characteristics, as well as their response to environmental factors in high-latitude regions of China, soil samples from 0 to 20 cm depth were collected from tea plantations at different altitudes and cultivation years in the main tea-producing areas of Shaanxi Province. These samples were used to determine the soil organic carbon (SOC), total nitrogen (TN), and total phosphorus (TP) contents and to calculate their stoichiometric ratios. The findings revealed the following: the average soil SOC and TN content in tea gardens were 13.15 and 1.30 g·kg−1, respectively, exceeding the national soil average. These values met the Class I tea garden fertility standards. However, the average soil TP content, at 0.45 g·kg−1, fell below the national soil average, meeting the Class II tea garden fertility standards. In tea gardens, the average ratios of carbon to nitrogen (C:N), carbon to phosphorus (C:P), and nitrogen to phosphorus (N:P) in the soil were 10.42, 30.98, and 3.32, respectively. These ratios were all lower than the national soil average, indicating relatively high phosphorus availability but nitrogen deficiency in tea garden soils. As altitude increased, there was a decline in soil SOC content, C N, and C P ratios, followed by a subsequent increase. No significant changes were seen in TN, TP, and N P ratio in the soil, but there was an increase in SOC content, TN content, and C P ratio during cultivation. The N-to-P ratio initially increased before decreasing, while the C-to-N ratio decreased before increasing. Soil TP content did not change significantly. Eucalyptus grandis is an important planted hardwood tree worldwide with fast growth and good wood performance. The nitrate transporter (NRT) gene family is a major core involved in nitrogen (N) absorption and utilization in plants, but the comprehensive characterization of NRT genes in E. grandis remains mostly elusive. In this study, a total of 75 EgNRT genes were identified from the genome of E. grandis that were distributed unevenly across ten chromosomes, except Chr9. A phylogenetic analysis showed that the EgNRT proteins could be divided into three classes, namely NRT1, NRT2 and NRT3, which contained 69, 4 and 2 members, respectively. The cis-regulatory elements in the promoter regions of EgNRT genes were mainly involved in phytohormone and stress response. The transcriptome analysis indicated that the differentially expressed genes of leaf and root in E. grandis under different N supply conditions were mainly involved in the metabolic process and plant hormone signal transduction. In addition, the transcriptome-based and RT-qPCR analysis revealed that the expression of 13 EgNRT genes, especially EgNRT1.3, EgNRT1.38, EgNRT1.39 and EgNRT1.52, was significantly upregulated in the root under low-N-supply treatment, suggesting that those genes might play a critical role in root response to nitrate deficiency. Summary: Elucidating the mechanisms regulating nitrogen (N) deficiency responses in plants is of great agricultural importance. Previous studies revealed that decreased expression of NITRATE‐INDUCIBLE GARP‐TYPE TRANSCRIPTIONAL REPRESSOR1 (NIGT1) transcriptional repressor genes upon N deficiency is involved in N deficiency‐inducible gene expression in Arabidopsis thaliana. However, our knowledge of the mechanisms controlling N deficiency‐induced changes in gene expression is still limited.Through the identification of Dof1.7 as a direct target of NIGT1 repressors and a novel N deficiency response‐related transcriptional activator gene, we here show that NIGT1 and Dof1.7 transcription factors (TFs) differentially regulate N deficiency‐inducible expression of three high‐affinity nitrate transporter genes, NRT2.1, NRT2.4, and NRT2.5, which are responsible for most of the soil nitrate uptake activity of Arabidopsis plants under N‐deficient conditions.Unlike NIGT1 repressors, which directly suppress NRT2.1, NRT2.4, and NRT2.5 under N‐sufficient conditions, Dof1.7 directly activated only NRT2.5 but indirectly and moderately activated NRT2.1 and NRT2.4 under N‐deficient conditions, probably by indirectly decreasing NIGT1 expression. Thus, Dof1.7 converted passive transcriptional activation into active and potent transcriptional activation, further differentially enhancing the expression of NRT2 genes.These findings clarify the mechanism underlying different expression patterns of NRT2 genes upon N deficiency, suggesting that time‐dependent multilayered transcriptional regulation generates complicated expression patterns of N deficiency‐inducible genes. Enhancing nitrogen use efficiency (NUE) in crops, particularly under limited nitrogen availability, is pivotal for advancing sustainable agricultural practices. This study investigates the NUE and associated physiological responses in foxtail millets (Setaria spp.) subjected to varying nitrogen regimes. We selected two genotypes of S. viridis and four of S. italica, representing three distinct domestication stages. The study employed four nitrogen treatments: 0 mg/kg (N0), 25 mg/kg (N25), 50 mg/kg (N50), and 100 mg/kg (N100), ranging from nitrogen deficiency to sufficiency. Our findings highlight marked variances in phenotypic adaptations to nitrogen availability among these domestication stages. Notably, wild genotypes (Q4, Q23) performed well in lower nitrogen conditions, evidenced by substantial increases in shoot and root biomass. Conversely, cultivated genotypes (Yugu18, C28) flourished predominantly under higher nitrogen conditions, displaying significant growth in shoot and root biomass, alongside improvements in root length, surface area, and volume. A critical observation was that under nitrogen-limited conditions, wild genotypes exhibited superior NUE compared to their cultivated counterparts. Structural equation modeling (SEM) elucidated a positive correlation between NUE and root architecture in wild genotypes, contrasted by a negative association with plant nitrogen content. Our findings indicate that modern elite varieties do not perform as well as the wild species in adapting to poor soil conditions. Two genes of nitrate transporters SaNRT2.1 and SaNRT2.5, putative orthologs of high-affinity nitrate transporter genes AtNRT2.1 and AtNRT2.5 from Arabidopsis thaliana, were cloned from the euhalophyte Suaeda altissima. Phylogenetic bioinformatic analysis demonstrated that the proteins SaNRT2.1 and SaNRT2.5 exhibited higher levels of homology to the corresponding proteins from the plants of family Amaranthaceae; the similarity of amino acid sequences between proteins SaNRT2.1 and SaNRT2.5 was lower (54%). Both SaNRT2.1 and SaNRT2.5 are integral membrane proteins forming 12 transmembrane helices as predicted by topological modeling. An attempt to demonstrate nitrate transporting activity of SaNRT2.1 or SaNRT2.5 by heterologous expression of the genes in the yeast Hansenula (Ogataea) polymorpha mutant strain Δynt1 lacking the only yeast nitrate transporter was not successful. The expression patterns of SaNRT2.1 and SaNRT2.5 were studied in S. altissima plants that were grown in hydroponics under either low (0.5 mM) or high (15 mM) nitrate and salinity from 0 to 750 mM NaCl. The growth of the plants was strongly inhibited by low nitrogen supply while stimulated by NaCl; it peaked at 250 mM NaCl for high nitrate and at 500 mM NaCl for low nitrate. Under low nitrate supply, nitrate contents in S. altissima roots, leaves and stems were reduced but increased in leaves and stems as salinity in the medium increased. Potassium contents remained stable under salinity treatment from 250 to 750 mM NaCl. Quantitative real-time PCR demonstrated that without salinity, SaNRT2.1 was expressed in all organs, its expression was not influenced by nitrate supply, while SaNRT2.5 was expressed exclusively in roots—its expression rose about 10-fold under low nitrate. Salinity increased expression of both SaNRT2.1 and SaNRT2.5 under low nitrate. SaNRT2.1 peaked in roots at 500 mM NaCl with 15-fold increase; SaNRT2.5 peaked in roots at 500 mM NaCl with 150-fold increase. It is suggested that SaNRT2.5 ensures effective nitrate uptake by roots and functions as an essential high-affinity nitrate transporter to support growth of adult S. altissima plants under nitrogen deficiency. Nitrogen is one of the most essential macronutrients for plant growth and its availability in soil is vital for agricultural sustainability and productivity. However, excessive nitrogen application could reduce the nitrogen use efficiency and produce environmental pollution. Here, we systematically determined the response in lipidome and metabolome in rapeseed during nitrogen starvation. Plant growth was severely retarded during nitrogen deficiency, while the levels of most amino acids were significantly decreased. The level of monogalactosyldiacyglycerol (MGDG) in leaves and roots was significantly decreased, while the level of digalactosyldiacylglycerol (DGDG) was significantly decreased in roots, resulting in a significant reduction of the MGDG/DGDG ratio during nitrogen starvation. Meanwhile, the levels of sulfoquinovosyl diacylglycerol, phosphatidylglycerol and glucuronosyl diacylglycerol were reduced to varying extents. Moreover, the levels of metabolites in the tricarboxylic acid cycle, Calvin cycle and energy metabolism were changed during nitrogen deficiency. These findings show that nitrogen deprivation alters the membrane lipid metabolism and carbon metabolism, and our study provides valuable information to further understand the response of rapeseed to nitrogen deficiency at the metabolism level. Nitrogen is an essential element for maize growth, but excessive application can lead to various environmental and ecological issues, including water pollution, air pollution, greenhouse gas emissions, and biodiversity loss. Hence, developing maize hybrids resilient to low-N conditions is vital for sustainable agriculture, particularly in nitrogen-deficient soils. Combining ability and genetic relationships among parental lines is crucial for breeding superior hybrids under diverse nitrogen levels. This study aimed to assess the genetic diversity of maize inbred lines using simple sequence repeat (SSR) markers and evaluate their combining ability to identify superior hybrids under low-N and recommended conditions. Local and exotic inbred lines were genotyped using SSR markers, revealing substantial genetic variation with high gene diversity (He = 0.60), moderate polymorphism information content (PIC = 0.54), and an average of 3.64 alleles per locus. Twenty-one F1 hybrids were generated through a diallel mating design using these diverse lines. These hybrids and a high yielding commercial check (SC-131) were field-tested under low-N and recommended N conditions. Significant variations (p < 0.01) were observed among nitrogen levels, hybrids, and their interaction for all recorded traits. Additive genetic variances predominated over non-additive genetic variances for grain yield and most traits. Inbred IL3 emerged as an effective combiner for developing early maturing genotypes with lower ear placement. Additionally, inbreds IL1, IL2, and IL3 showed promise as superior combiners for enhancing grain yield and related traits under both low-N and recommended conditions. Notably, hybrids IL1×IL4, IL2×IL5, IL2×IL6, and IL5×IL7 exhibited specific combining abilities for increasing grain yield and associated traits under low-N stress conditions. Furthermore, strong positive associations were identified between grain yield and specific traits like plant height, ear length, number of rows per ear, and number of kernels per row. Due to their straightforward measurability, these relationships underscore the potential of using these traits as proxies for indirect selection in early breeding generations, particularly under low-N stress. SUMMARY: The conservation of GOLVEN (GLV)/ROOT MERISTEM GROWTH FACTOR (RGF) peptide encoding genes across plant genomes capable of forming roots or root‐like structures underscores their potential significance in the terrestrial adaptation of plants. This study investigates the function and role of GOLVEN peptide‐coding genes in Medicago truncatula. Five out of fifteen GLV/RGF genes were notably upregulated during nodule organogenesis and were differentially responsive to nitrogen deficiency and auxin treatment. Specifically, the expression of MtGLV9 and MtGLV10 at nodule initiation sites was contingent upon the NODULE INCEPTION transcription factor. Overexpression of these five nodule‐induced GLV genes in hairy roots of M. truncatula and application of their synthetic peptide analogues led to a decrease in nodule count by 25–50%. Uniquely, the GOLVEN10 peptide altered the positioning of the first formed lateral root and nodule on the primary root axis, an observation we term 'noduletaxis'; this decreased the length of the lateral organ formation zone on roots. Histological section of roots treated with synthetic GOLVEN10 peptide revealed an increased cell number within the root cortical cell layers without a corresponding increase in cell length, leading to an elongation of the root likely introducing a spatiotemporal delay in organ formation. At the transcription level, the GOLVEN10 peptide suppressed expression of microtubule‐related genes and exerted its effects by changing expression of a large subset of Auxin responsive genes. These findings advance our understanding of the molecular mechanisms by which GOLVEN peptides modulate root morphology, nodule ontogeny, and interactions with key transcriptional pathways. Nitrogen (N) is an essential element for plant growth, and its deficiency influences plants at several physiological and gene expression levels. Barley (Hordeum vulgare) is one of the most important food grains from the Poaceae family and one of the most important staple food crops. However, the seed yield is limited by a number of stresses, the most important of which is the insufficient use of N. Thus, there is a need to develop N-use effective cultivars. In this study, comparative physiological and molecular analyses were performed using leaf and root tissues from 10 locally grown barley cultivars. The expression levels of nitrate transporters, HvNRT2 genes, were analyzed in the leaf and root tissues of N-deficient (ND) treatments of barley cultivars after 7 and 14 days following ND treatment as compared to the normal condition. Based on the correlation between the traits, root length (RL) had a positive and highly significant correlation with fresh leaf weight (FLW) and ascorbate peroxidase (APX) concentration in roots, indicating a direct root and leaf relationship with the plant development under ND. From the physiological aspects, ND enhanced carotenoids, chlorophylls a/b (Chla/b), total chlorophyll (TCH), leaf antioxidant enzymes such as ascorbate peroxidase (APX), peroxidase (POD), and catalase (CAT), and root antioxidant enzymes (APX and POD) in the Sahra cultivar. The expression levels of HvNRT2.1, HvNRT2.2, and HvNRT2.4 genes were up-regulated under ND conditions. For the morphological traits, ND maintained root dry weight among the cultivars, except for Sahra. Among the studied cultivars, Sahra responded well to ND stress, making it a suitable candidate for barely improvement programs. Nitrogen fertilizer levels significantly affect crop growth and development, necessitating precision management. Most studies focus on nitrogen nutrient estimation using vegetation indices and textural features, overlooking the diagnostic potential of color features. Hence, we investigated cotton nitrogen nutrition status using unmanned aerial vehicle (UAV) image features and the nitrogen nutrient index (NNI). Random frog algorithm - and random forest-screened image feature sets significantly correlated with the NNI, which were substituted into four machine learning algorithms for NNI estimation modeling. The composite scores (F) of optimal image feature sets were calculated using the coefficient of variation method for comprehensive cotton nitrogen nutrient diagnosis. Validation of the model for determining the critical nitrogen concentration in cotton yielded a coefficient of determination R2 = 0.89, root mean square error RMSE = 0.50 g (100 g)-1, and mean absolute error MAE = 0.44, demonstrating improved performance. Additionally, our novel NNI estimation model constructed based on the optimal image feature sets exhibited R2 c = 0.97, RMSEc = 0.02, MAEc = 0.02, R2 v = 0.85, RMSEv = 0.05, and MAEv = 0.04. Polynomial fitting of the composite index with NNI indicated that the model was reliable and yielded the following diagnostic criterion: 0.48 < F2 < 0.67 indicated nitrogen overapplication, whereas F2 < 0.48 or F2 > 0.67 indicated nitrogen deficiency. This study demonstrates the superior effectiveness of using UAV RGB image feature sets for NNI estimation and the quick, accurate diagnosis of cotton nitrogen levels, which will help guide nitrogen fertilizer application. Aims: Nitrogen deficiency in eco-engineered technosol from iron (Fe) ore tailings limits the productivity of colonising soil microbes and pioneer plants, which are critical to further development of the technosol. Symbiotic biological N2 fixation may be a strategy to supply N in the moderately alkaline early technosols since native legumes such as Acacia auriculiformis are tolerant of saline and alkaline soil conditions as those in the technosol. It is hypothesized that tolerant native legume A. auriculiformis could form functional nodules to fix N2 when grown in early eco-engineered technosols. Methods: A. auriculiformis growth and root nodulation in the early tailing technosols were investigated using a glasshouse experiment, and plant N2 fixation was evaluated using the 15 N natural abundance isotope method. Key factors influencing root nodulation and N2 fixation have also been evaluated, including water supply and phosphorous nutrition. Results: The results indicated that A. auriculiformis grew well in the tailing technosols and naturally formed nodules with rhizobia. The nodules were functional in N2 fixation, leading to improved plant N nutrition. The nodulation and N2 fixation were severely limited by water deficiency stress. Improved phosphorous supply favoured nodulation and N2 fixation by A. auriculiformis plants under water deficiency stress. Conclusions: These findings suggested that A. auriculiformis could grow in early tailings technosols and fixed N2, and proper water and phosphorous fertilizer management could improve Acacia plant's performance and N2 fixation functions. SUMMARY: The yield of maize (Zea mays L.) crops depends on their ability to intercept sunlight throughout the growing cycle, transform this energy into biomass and allocate it to the kernels. Abiotic stresses affect these eco‐physiological determinants, reducing crop grain yield below the potential of each environment. Here we analyse the impact of combined abiotic stresses, such as water restriction and nitrogen deficiency or water restriction and elevated temperatures. Crop yield depends on the product of kernel yield per plant and the number of plants per unit soil area, but increasing plant population density imposes a crowding stress that reduces yield per plant, even within the range that maximises crop yield per unit soil area. Therefore, we also analyse the impact of abiotic stresses under different plant densities. We show that the magnitude of the detrimental effects of two combined stresses on field‐grown plants can be lower, similar or higher than the sum of the individual stresses. These patterns depend on the timing and intensity of each one of the combined stresses and on the effects of one of the stresses on the status of the resource whose limitation causes the other. The analysis of the eco‐physiological determinants of crop yield is useful to guide and prioritise the rapidly progressing studies aimed at understanding the molecular mechanisms underlying plant responses to combined stresses. Oil palm (Elaeis guineensis) is a widely cultivated crop known for its high oil yield. It is cultivated extensively across tropical regions, notably in Southeast Asia, Africa, and Latin America. It plays a vital role in global vegetable oil supply, meeting approximately 35% of the world's demand. However, the expansion of oil palm plantations often involves the utilization of degraded soils where nutrient availability, particularly nitrogen, is limited, posing challenges to plant growth and productivity. Roots are crucial in responding to nitrogen deficiency by adjusting their growth and distribution; however, research on root system distribution patterns in oil palm still needs to be completed. This study analyzes the root system architecture using RhizoVision Explorer, a 2D root image processing software while assessing its relationship with nitrogen availability across two commercial cultivars: Deli × La Mé of African oil palm (Elaeis guineensis) and the interspecific hybrid O×G Coari × La Mé. Our findings reveal significant associations between eight root traits in oil palm seedlings and treatments with and without nitrogen availability. Notably, total root morphology (length, surface area, and volume), rotation angle, solidity, and hole characteristics decreased under nitrogen deprivation, whereas surface angle frequency increased. We highlight the variability of these traits across cultivars, suggesting genetic dependence and potential utility in breeding programs. Moreover, interactions observed in primary root morphology and hole size indicate greater differences between control and nitrogen-treated groups in C × LM than in D × LM cultivars. On the other hand, cultivar differences, regardless of nitrogen availability, influenced lateral root morphology, while nitrogen availability, irrespective of cultivar, affected inclined angle frequency. Significant differences were observed in growth and development parameters such as root and shoot biomass, root-to-shoot ratio, and leaf emission numbers between nitrogen-optimal and nitrogen-starved conditions. SUMMARY: Nitrogen (N) is an essential nutrient for crop growth and development, significantly influencing both yield and quality. Melatonin (MT), a known enhancer of abiotic stress tolerance, has been extensively studied. However, its relationship with nutrient stress, particularly N deficiency, and the underlying regulatory mechanisms of MT on N absorption remain unclear. In this study, exogenous MT treatment was found to improve the tolerance of apple plants to N deficiency. Apple plants overexpressing the MT biosynthetic gene N‐acetylserotonin methyltransferase 9 (MdASMT9) were used to further investigate the effects of endogenous MT on low‐N stress. Overexpression of MdASMT9 improved the light harvesting and heat transfer capability of apple plants, thereby mitigating the detrimental effects of N deficiency on the photosynthetic system. Proteomic and physiological data analyses indicated that MdASMT9 overexpression enhanced the trichloroacetic acid cycle and positively modulated amino acid metabolism to counteract N‐deficiency stress. Additionally, both exogenous and endogenous MT promoted the transcription of MdHY5, which in turn bound to the MdNRT2.1 and MdNRT2.4 promoters and activated their expression. Notably, MT‐mediated promotion of MdNRT2.1 and MdNRT2.4 expression through regulating MdHY5, ultimately enhancing N absorption. Taken together, these findings shed light on the association between MdASMT9‐mediated MT biosynthesis and N absorption in apple plants under N‐deficiency conditions. Background: Nitrogen (N) metabolism-related key genes and conserved amino acid sites in key enzymes play a crucial role in improving N use efficiency (NUE) under N stress. However, it is not clearly known about the molecular mechanism of N deficiency-induced improvement of NUE in the N-sensitive rhizomatous medicinal plant Panax notoginseng (Burk.) F. H. Chen. To explore the potential regulatory mechanism, the transcriptome and proteome were analyzed and the three-dimensional (3D) information and molecular docking models of key genes were compared in the roots of P. notoginseng grown under N regimes. Results: Total N uptake and the proportion of N distribution to roots were significantly reduced, but the NUE, N use efficiency in biomass production (NUEb), the recovery of N fertilizer (RNF) and the proportion of N distribution to shoot were increased in the N0-treated (without N addition) plants. The expression of N uptake- and transport-related genes NPF1.2, NRT2.4, NPF8.1, NPF4.6, AVP, proteins AMT and NRT2 were obviously up-regulated in the N0-grown plants. Meanwhile, the expression of CIPK23, PLC2, NLP6, TCP20, and BT1 related to the nitrate signal-sensing and transduction were up-regulated under the N0 condition. Glutamine synthetase (GS) activity was decreased in the N-deficient plants, while the activity of glutamate dehydrogenase (GDH) increased. The expression of genes GS1-1 and GDH1, and proteins GDH1 and GDH2 were up-regulated in the N0-grown plants, there was a significantly positive correlation between the expression of protein GDH1 and of gene GDH1. Glu192, Glu199 and Glu400 in PnGS1 and PnGDH1were the key amino acid residues that affect the NUE and lead to the differences in GDH enzyme activity. The 3D structure, docking model, and residues of Solanum tuberosum and P. notoginseng was similar. Conclusions: N deficiency might promote the expression of key genes for N uptake (genes NPF8.1, NPF4.6, AMT, AVP and NRT2), transport (NPF1.2 and NRT2.4), assimilation (proteins GS1 and GDH1), signaling and transduction (genes CIPK23, PLC2, NLP6, TCP20, and BT1) to enhance NUE in the rhizomatous species. N deficiency might induce Glu192, Glu199 and Glu400 to improve the biological activity of GS1 and GDH, this has been hypothesized to be the main reason for the enhanced ability of N assimilation in N-deficient rhizomatous species. Exploration of genetic resources constitutes an eminent device for the selection of interesting tolerant forage species for nitrogen deficiency. In this regard, the performance of two legume forage species (Sulla carnosa (Desf.) B.H.Choi & H.Ohashi, wild halophytic species; Trifolium alexandrinum L., commercially cultivated species) to nitrogen deficiency was compared on the basis of the total dry weight, relative growth rate, nitrogen accumulation, nitrogen use efficiency, photosynthetic activity and chlorophyll a fluorescence. Plants were supplied with mixed nitrogen form (NO3NH4) at a concentration of 5.0 mM (NS–nitrogen sufficient) or 0.5 mM (ND–nitrogen deficiency) and cultivated under hydroponic controlled conditions. At S. carnosa, nitrogen deficiency reduced growth rate (68%), nitrogen use efficiency (16%), net photosynthetic assimilation rate (36%), maximum photochemical efficiency of PSII (7%), PSII operating efficiency (19%), photochemical quenching coefficient (8%) and non-cyclic electron transport rate (19%), while it had no deleterious effects on growth, photosynthesis and PSII photochemistry at T. alexandrinum. The data in the present research clearly indicated that S. carnosa was more sensitive to nitrogen deficiency than T. alexandrinum, which was mainly related to decline of NUE and to the damage of the photosynthetic apparatus and consequently to the decline of the photosynthetic activity. Despite its sensitivity, S. carnosa had the ability to use nitrogen more efficiently (27%) and to produce a biomass comparable to that of the cultivated legume species T. alexandrinum. Experiments to evaluate the performance of 10 bread wheat genotypes with nitrogen deficiency and diagnose the optimal genotype for the appropriate environment commenced in the winter seasons, 2021-2022 and 2022-2023, in three locations of Iraq, i.e., Baghdad, Wasit, and Diwaniyah. All the experiments had a randomized complete block design (RCBD) with split-plot arrangement. Nitrogen levels (0, 50, and 100 kg N ha-1) occupied the prime plots, with 10 wheat genotypes (T2, T3, S1, S2, S148, IPA99, Adana99, Rasheed, Buhooth-10, and Buhooth-22) allocated to the subplots with three replications in each location. The interaction of wheat genotypes, nitrogen levels, and locations revealed that genotype S2 with 50 kg N ha-1 at Baghdad showed the highest mean nitrogen use efficiency at 105.00 and 96.00 for 2021-2022 and 2022- 2023, respectively. However, these values showed nonsignificant differences from the obtained values of wheat genotypes S148 and the IPA-99 during the second season, in the same location and fertilizer level, i.e., 92.87 and 90.00, respectively. Also, the genotype S2 gave the highest grain yield (6.53 t ha-1) with N2 fertilizer level in the Baghdad site, not differing significantly from two other wheat genotypes, S148 and IPA99 (6.13 and 5.85 t ha-1, respectively) in the second season. The presented study authenticated that some wheat genotypes provided good yield under nitrogen stress (50 kg N ha-1), as there were no significant increases in grain yield when doubling the nitrogen level (100 kg N ha-1). The TvV-SM2 virus, isolated fromthe coastal waters of the Black Sea, causes lysis of its host, the algae Tetraselmis viridis (Chlorophyta). Under optimal conditions for nutrients, an increase in the initial abundance of algae cells by four times caused a 3-fold reduction in the latent period of viral infection. During the period of the most rapid cell lysis of T. viridis, nitrogen deficiency leads to a decrease in the average daily rate of death of cells affected by the virus by 3.2 times relative to the replete conditions, while in the case of phosphorus deficiency, this process slows down by up to 2.4 times. Under deplete conditions, the rate of cell death was only 34% lower than under replete conditions. The effect of copper ions (100 µg L-1) on the viral suspension for 6 h led to the complete suppression of its activity. In the presence of the host of this virus, its activity is only partially suppressed. As a result, cell lysis under the influence of a viral infection occurred in two stages. The first stage was noted only during the first 6 h of the experiment. The second main stage took place within 78-170 h. Background: Lack of nutrients, in particular nitrogen and phosphorus, has been known in the field to sense glutamate production via 2-oxoglutarate and subsequently accelerate carbon storage, including glycogen and polyhydroxybutyrate (PHB), in cyanobacteria, but a few studies have focused on arginine catabolism. In this study, we first time demonstrated that gene manipulation on proC and adc1, related to proline and polyamine syntheses in arginine catabolism, had a significant impact on enhanced PHB production during late growth phase and nutrient-modified conditions. We constructed Synechocystis sp. PCC 6803 with an overexpressing proC gene, encoding Δ1pyrroline-5-carboxylate reductase in proline production, and adc1 disruption resulted in lower polyamine synthesis. Results: Three engineered Synechocystis sp. PCC 6803 strains, including a ProC-overexpressing strain (OXP), adc1 mutant, and an OXP strain lacking the adc1 gene (OXP/Δadc1), certainly increased the PHB accumulation under nitrogen and phosphorus deficiency. The possible advantages of single proC overexpression include improved PHB and glycogen storage in late phase of growth and long-term stress situations. However, on day 7 of treatment, the synergistic impact created by OXP/Δadc1 increased PHB synthesis by approximately 48.9% of dry cell weight, resulting in a shorter response to nutrient stress than the OXP strain. Notably, changes in proline and glutamate contents in engineered strains, in particular OXP and OXP/Δadc1, not only partially balanced the intracellular C/N metabolism but also helped cells acclimate under nitrogen (N) and phosphorus (P) stress with higher chlorophyll a content in comparison with wild-type control. Conclusions: In Synechocystis sp. PCC 6803, overexpression of proC resulted in a striking signal to PHB and glycogen accumulation after prolonged nutrient deprivation. Understanding plant-microbe interactions requires examination of root exudation under nutrient stress using standardized and reproducible experimental systems. We grew Brachypodium distachyon hydroponically in fabricated ecosystem devices (EcoFAB 2.0) under three inorganic nitrogen forms (nitrate, ammonium, and ammonium nitrate), followed by nitrogen starvation. Analyses of exudates with liquid chromatography-tandem mass spectrometry, biomass, medium pH, and nitrogen uptake showed EcoFAB 2.0's low intratreatment data variability. Furthermore, the three inorganic nitrogen forms caused differential exudation, generalized by abundant amino acids-peptides and alkaloids. Comparatively, nitrogen deficiency decreased nitrogen-containing compounds but increased shikimates-phenylpropanoids. Subsequent bioassays with two shikimates-phenylpropanoids (shikimic and p-coumaric acids) on soil bacteria or Brachypodium seedlings revealed their distinct capacity to regulate both bacterial and plant growth. Our results suggest that (i) Brachypodium alters exudation in response to nitrogen status, which can affect rhizobacterial growth, and (ii) EcoFAB 2.0 is a valuable standardized plant research tool. Soil contamination by cadmium (Cd) has presented a major challenge in China. The objective of the field experiments in this study was to examine the influence of nitrogen fertilizer application at the full heading and milky stages on minimizing the absorption of Cd in rice. This was achieved by affecting the distribution of Cd in root plaques and subcellular compartments of the root and flag leaf. The hydroponic culture experiments aimed to examine the effect of nitrogen and Cd interaction or deficiency on Cd accumulation in rice during the late growth stage. The findings revealed that adequate nitrogen supply during the early growth stage, coupled with nitrogen application during the full heading and milky stages, led to a notable increase in Fe concentration in the root plaques during the milk and mature stages. Furthermore, it elevated the Cd proportion in the soluble fraction of the flag leaves at the milky stage. Conversely, nitrogen deficiency during the early growth stage resulted in a significant increase in Fe concentration in the root plaques, along with a decrease in Cd concentration. Additionally, the proportion of Cd in the flag leaf cell walls increased significantly, while the proportion in the soluble fraction decreased notably. Irrespective of nitrogen supply during the early growth stage, applying nitrogen at the full heading stage significantly reduced Cd transport from shoots to brown rice, leading to a considerable reduction in the Cd concentration in brown rice. Under hydroponic culture conditions, combined Cd exposure with nitrogen supply significantly increased the Cd concentration in brown rice. Nitrogen supply had no impact on the Cd concentration in brown rice in the absence of Cd. The study showed that applying nitrogen fertilizer at the full heading stage effectively decreased the brown rice Cd concentration. This was achieved by elevating the concentration of Fe in the root plaque, thereby influencing the adsorption of Cd by the roots. Additionally, nitrogen application at the full heading stage can influence the distribution of Cd in flag leaf cells during the filling stage. Ensuring ample nitrogen supply in the early stage of rice growth is crucial, and nitrogen application at the full heading stage can effectively reduce the Cd concentration in brown rice. Leaf nitrogen content (LNC) is an important index to measure the nitrogen deficiency in cotton. The rapid and accurate monitoring of LNC is of great significance for understanding the growth status of cotton and guiding precise fertilization in the field. At present, the hyperspectral technology monitoring of LNC is very mature, but it is interfered with by external factors such as shadow and soil, and data acquisition is still dependent on manpower. Therefore, on the basis of clarifying the correlation and quantitative relationship between physiological parameters and cotton LNC, the 400-2500 nm spectral curve was simulated based on PROSPECT-5 model. Combined with the measured spectra, the sensitive bands of leaf nitrogen content were screened, and four machine learning algorithms based on the reflectance of the sensitive bands were compared to construct a model for the estimation of LNC in cotton and determine the optimal model. The results show the following: (1) The parameter with the best correlation with nitrogen content was Cab, and the linear relationship was y=0.3942x+12.521, R2=0.81, RMSE=12.87 g/kg. (2) The shuffled frog leaping algorithm (SFLA) and the successive projections algorithm (SPA) were used to screen the relevant bands sensitive to LNC. SFLA selected nine characteristic bands, mainly distributed between 700 and 750 nm. SPA screened seven characteristic bands, mainly distributed between 670 and 760 nm. The characteristic bands of both screening methods were distributed near the red edge. (3) Based on the sensitive bands, the four machine learning algorithms were compared. Among them, the band modeling of SFLA screening under the random forest (RF) algorithm was the best (modeling set R2=0.973, RMSE=1.001 g/kg, rRMSE=3.41%, validation set R2=0.803, RMSE=3.191 g/kg, rRMSE=10.85%). In summary, this study proposes an optimal estimation model of cotton leaf nitrogen content based on the radiative transfer model, which provides a theoretical basis for the dynamic, accurate, and non-destructive monitoring of cotton leaf nitrogen content. Background and aims: Legumes can obtain nitrogen from mineral nitrogen and symbiotic nitrogen; however, the regulatory mechanisms by which legume plants respond to various nitrogen sources are still unclear. Methods: Here, the flavonoid metabolism profiles of Robinia pseudoacacia (black locust) in response to different nitrogen sources during the early growth stages were performed using the hydroponics method. Results: The flavonoids were accumulated specifically responding to different nitrogen sources (symbiotic nitrogen and NO3−) in the roots and root exudates of R. pseudoacacia and showed growth-stage-dependent changes in the plant growth process. More flavonoids were synthesized and secreted under nitrogen deficiency. The differential flavonoids among different treatments were significantly enriched in the flavone and flavonol biosynthesis pathway and isoflavonoid biosynthesis pathway. Liquiritigenin, naringenin, apigenin and kaempferol were not only the metabolic nodes of these two metabolic pathways but also the key nodes in the differential flavonoid network. Therefore, these four flavonoids may be the key flavonoids of R. pseudoacacia responding to different nitrogen sources. Conclusion: Our findings provide an important insight into the adaptability of legumes to different nitrogen environments and are of great significance for understanding the molecular mechanisms underlying nitrogen utilization in legumes and will contribute to the development of sustainable agricultural practices and crop production.
