A seriesof novel hexacyclic tetracycline analogues (“hexacyclines”)was designed, synthesized, and evaluated for antibacterial activityagainst a wide range of clinically important bacteria isolates, includingmultidrug-resistant, Gram-negative pathogens. Valuable structure–activityrelationships were identified, and several hexacyclines displayedpotent, broad spectrum antibacterial activity, including promisinganti-Pseudomonas aeruginosaactivityin vitro and in vivo.
