Currently, important methodological approaches are available to make systematic reviews more robust and transparent. One of these approaches is the Grading of Recommendation, Assessment, Development, and Evaluation (GRADE) system.A perspective for rehabilitation was conducted aiming to (1) shed light on the meaning and relevance of the GRADE system in systematic reviews with meta-analysis of rehabilitation science, and (2) propose suggestions for helping clinicians improve the interpretation of the GRADE findings.The meaning and relevance of GRADE in rehabilitation science was discussed. Suggestions were proposed to help clinicians in the interpretation of the GRADE findings. We discussed the use of meta-analyses, meta-regressions, subgroups meta-analyses, and sensitivity analyses to increase the objectivity of the domains of GRADE. Finally, a future agenda was provided.The use of GRADE is essential for improving the synthesis of evidence that clinicians may often use in rehabilitation practice. However, GRADE is only one hallmark when the findings of systematic reviews are interpreted. The issues of sample size, futile research, pre-registration, switching outcomes, or narrative bias should be also considered.Grading of Recommendation, Assessment, Development, and Evaluation (GRADE) is helpful in systematic reviews for understanding the certainty of evidence is analyzed.GRADE could detect limitation in the certainty of evidence helping clinicians to choose the best therapeutic option for their patients.Some domains of GRADE (indirectness and inconsistency) are particularly useful for clinicians who aim to translate the conclusions of a synthesis of evidence into clinical practice. The article is based on a series of interviews (61) and a multi-sited ethnography conducted during 2019–2021 which traced archival records of 20 feminist organizations in Israel: local women's peace organizations (FPAs) and Rape Crisis Centres (RCCs). We describe the study and the complex methodological concerns and meta-questions relating to the study of feminist community archives in relation to content (types of testimonials or records), method of organization (archival practices like cataloging or digitization) and activists' perspectives concerning future preservation and access. In order to overcome these challenges, we suggest six methodological principles which may apply to the study of civil society organizations that were established between the 1970s–1990s: the importance of identifying researchers' positionality vis a-vis the archive; the politics of knowledge and intersectional identities; avoiding judgment of informal archival practices; identifying who sets the rules; silence and self-silencing; and recognition of invisible labor. The use of routinely collected electronic healthcare records (EHR) for outcome assessment in clinical trials has been described as a 'disruptive' new technique more than a decade ago. Despite this potential, significant methodological issues and regulatory barriers have hampered the progress in this area. This article discusses the key considerations that trialists should take into account when incorporating EHR into their trials. These include considerations of the clinical relevance of the outcome, data timeliness and quality, ethical and regulatory issues, and some practical considerations for clinical trials units. In addition, this article describes the benefits of using EHR which include cost, reduced trial burden for participants and staff, follow up efficiencies, and improved health economic evaluation procedures. We also describe the major regulatory and start up costs of using EHR in clinical trials. This article focuses on the UK specific EHR landscape in clinical trials and would help researchers and trials units considering the use of this method of outcome data collection in their next trial. Telocytes (TCs) are CD34-positive interstitial cells that have long cytoplasmic projections, called telopodes; they have been identified in several organs and in various species. These cells establish a complex communication network between different stromal and epithelial cell types, and there is growing evidence that they play a key role in physiology and pathology. In many tissues, TC network impairment has been implicated in the onset and progression of pathological conditions, which makes the study of TCs of great interest for the development of novel therapies. In this review, we summarise the main methods involved in the characterisation of these cells as well as their inherent difficulties and then discuss the functional assays that are used to uncover the role of TCs in normal and pathological conditions, from the most traditional to the most recent. Furthermore, we provide future perspectives in the study of TCs, especially regarding the establishment of more precise markers, commercial lineages and means for drug delivery and genetic editing that directly target TCs. Multiconjugate adaptive optics (MCAO) stands as an essential technology for the development of future large-aperture solar telescopes. Its primary objective is to empower telescopes to achieve nearly diffraction-limited performance while substantially extending the correction field of view (FoV). Conventional solar MCAO relies on the combination of adaptive optics and high-altitude correction (AO + HAC) modules for multiconjugate correction. However, this architectural approach excels in correction performance primarily at the central position, with performance deteriorating as one moves farther from the centre. Consequently, it results in poor consistency of FoV correction performance. To address these limitations, a new architectural approach was introduced, which combines ground layer AO with HAC (GLAO + HAC). Preliminary results have shown that, compared to AO + HAC, this approach significantly enhances FoV correction uniformity. Building upon these initial findings, this paper undertakes a more extensive research of the GLAO + HAC system. Its objective is to compare various solar MCAO system architectures, including AO + HAC, GLAO + HAC, and general MCAO, to finally propose optimization tailored to GLAO + HAC. Through this analysis, the paper conducts the performance comparison between GLAO + HAC and general MCAO. It underscores that, under equivalent configuration parameters, the differences between these two systems are marginal. However, due to the advantage of the independent control of dual correction modules in GLAO + HAC, it can introduce an optimization strategy by increasing the number of subapertures at the cost of reducing the GLAO guide star sensing FoV. Finally, the results of this strategy demonstrate an obvious enhancement in performance and FoV correction consistency within the GLAO + HAC system. [ABSTRACT FROM AUTHOR]

This paper presents an agenda for the future of gender research in small‐scale fisheries (SSF). Building on expert insight from scholars who gathered during the 4th World Small‐Scale Fisheries Congress Africa (4WSFC) with a synthesis of existing literature, we identify six topics that warrant future investigation in SSF, along with methodological considerations for addressing them. Research priorities include identifying pathways towards (1) equitable participation in governance and decision‐making, (2) valuing all actors' contributions to aquatic food systems, (3) increasing access to financial services, (4) inclusive infrastructural development, (5) livelihood diversification and (6) reducing occupational health hazards. Several important methodological considerations include (i) using multiple methodologies, (ii) applying participatory methods, (iii) collecting gender‐disaggregated data, (iv) integrating gender into a food systems approach in fisheries, (v) engaging an intersectional approach and (vi) operationalising equity. Decision-makers rely extensively on expert groups and commissions for policy advice. Many see in this a growing technocracy or expertization of decision-making. Yet, we know little about the actual influence of these expert bodies in policy-making. When does expert group advice shape policy-making rather than being brushed aside? The article goes beyond existing research by measuring and explaining variation in expert group influence. Using the number of citations to expert group reports in government white papers and later expert group reports as measures of influence, the article analyzes citations to 1545 Norwegian advisory commission reports published 1972–2017. It concentrates on three dimensions of the organization of expert groups as potential determinants of influence: member composition, appointing ministry and resources. The results suggest that expert groups have greater influence when they include more politicians and have a bigger secretariat. Connected speech is often used to assess many aspects of an individual's language abilities after stroke. However, it is unknown the degree to which elicitation methods differ in generating structural and syntactic aspects of connected speech, two critical components of successful communication. Quantifying the degree to which elicitation methods differ in eliciting structurally, syntactically, and lexically complex connected speech at the earliest stage of stroke before reorganization and rehabilitation of function independent of clinical diagnosis of aphasia has not been examined to date. Addressing this gap has implications for early clinical intervention as well as empirical studies of connected speech production. We compared two common elicitation methods, picture description and storytelling on lexical, structural, and syntactic measures of connected speech in speakers during the acute stage of left hemisphere stroke. We measured connected speech using an automated quantitative production analysis approach (Fromm et al., 2021) in 71 native-English speaking participants (27 female; 59 ± 13 years) within an average 3.9 days from left hemisphere stroke onset. We tested the degree of agreement and consistency between elicitation methods for lexical, structural, and syntactic measures of connected speech, as well as the degree of concordance in classifying deficits across individuals. Storytelling elicited significantly more words and more structurally complex, lexically diverse, and syntactically accurate speech in comparison to picture description. Elicitation methods differed in measuring outcomes across participants for the lexical and syntactic, but not structural complexity aspects of connected speech where storytelling classified more participants with impairments in comparison to picture description. These differences suggest storytelling provides assessment of connected speech abilities more reflective of real-world abilities where its use is particularly critical for examining individual differences and providing diagnoses of acute stroke language deficits. As a result, using storytelling as a connected speech elicitation method more effectively captures a patient's language capabilities after stroke, consequently informing clinical diagnosis and treatment. Many people with aphasia and people without brain injury talk to themselves in their heads, i.e., have "inner speech." Inner speech may be more preserved compared with spoken speech for some people with aphasia and may serve a variety of functions (e.g., emotion regulation), which motivates us to provide a high-fidelity characterization of it. Researchers have used multiple methods to measure this internal phenomenon in the past, which we combine here for the first time in a single study. We compare performance between individuals with and without aphasia on inner speech tasks that assess inner speech "in-the-moment" to general subjective impressions of inner speech to tease apart the relationship of aphasia severity to inner speech. Twenty people with mild-moderate aphasia and twenty neurotypical controls completed several inner speech tasks, including objective silent rhyme judgements (picture, written, and auditory), subjective reports of inner speech during naming, and subjective rating scales about inner speech experience more generally. In-the-moment inner speech during silent rhyming tasks was associated with aphasia severity only for picture and written rhyming but not auditory rhyming. In-the-moment inner speech reports during silent naming were not associated with aphasia severity, nor were the subjective ratings about general inner speech experience. Individuals with and without aphasia demonstrated a variety of subjective general inner speech experiences, demonstrating heterogeneity of this phenomenon more broadly. Methods of measuring inner speech complement each other and speak to different facets of the inner speech phenomenon, and clinicians and researchers must carefully choose the method(s) that will provide the information about inner speech that they desire. Yellowing viruses are an increasing threat to sugar beet cultivation, due to limitations on insecticide usage and climate change. Virus detection, monitoring and resistance breeding are key to secure high sugar beet yields in the future. For this research, a one‐step multiplex reverse transcription (mRT)‐PCR method was designed to detect simultaneously beet mild yellowing virus, beet chlorosis virus, beet yellows virus (BYV), beet mosaic virus and turnip yellows virus. The addition of Luminex xTAG array technology was used as a follow‐up method to increase assay specificity. The one‐step mRT‐PCR was evaluated on 22 field samples with single and mixed virus infections. The xTAG assay works as expected both in a simplex and multiplex setting, except that BYV detection needs optimization in the multiplex setting. In the future, the Luminex xTAG assay would be an excellent method for the detection of beet yellowing viruses due to its high specificity and the potential to increase the number of targets. Frequency-domain near-infrared spectroscopy (FD-NIRS) has been used for non-invasive assessment of cortical oxygenation since the late 1990s. However, there is limited research demonstrating clinical validity and general reproducibility. To address this limitation, recording duration for adequate validity and within- and between-day reproducibility of prefrontal cortical oxygenation was evaluated. To assess validity, a reverse analysis of 10-min-long measurements (n = 52) at different recording durations (1–10-min) was quantified via coefficients of variation and Bland–Altman plots. To assess within- and between-day within-subject reproducibility, participants (n = 15) completed 2-min measurements twice a day (morning/afternoon) for five consecutive days. While 1-min recordings demonstrated sufficient validity for the assessment of oxygen saturation (StO2) and total hemoglobin concentration (THb), recordings ≥4 min revealed greater clinical utility for oxy- (HbO) and deoxyhemoglobin (HHb) concentration. Females had lower StO2, THb, HbO, and HHb values than males, but variability was approximately equal between sexes. Intraclass correlation coefficients ranged from 0.50–0.96. The minimal detectable change for StO2 was 1.15% (95% CI: 0.336–1.96%) and 3.12 µM for THb (95% CI: 0.915–5.33 µM) for females and 2.75% (95%CI: 0.807–4.70%) for StO2 and 5.51 µM (95%CI: 1.62–9.42 µM) for THb in males. Overall, FD-NIRS demonstrated good levels of between-day reliability. These findings support the application of FD-NIRS in field-based settings and indicate a recording duration of 1 min allows for valid measures; however, data recordings of ≥4 min are recommended when feasible. Background: To assess the frequency of patient‐reported outcomes (PROs) and non‐PROs in randomized controlled trials (RCTs) addressing head/neck cancers. Methods: We included RCTs about interventions to treat head/neck cancers. PubMed was searched on September 16, 2022 and included studies published during three periods (2000–2002, 2010–2012, and 2020–2022). Data on types of outcomes and instruments to measure them were extracted and organized into PROs and non‐PROs, and temporal trends for reporting outcomes were determined. Results: There was a reduction in the frequency of non‐PROs (40% to 22%) and an increase in PROs (5% to 19%) over 20 years. The frequency of reporting both non‐PROs and PROs seemed to be stable over the same period (55% to 58%). A great variety of instruments to measure PROs and non‐PROs was identified. Conclusions: There has been a growth in the types of PROs in more recent years, and they were more frequently reported in RCTs. However, head/neck cancer trials with a combination of PROs and non‐PROs were the most prevalent. [ABSTRACT FROM AUTHOR]

There are currently no methodological frameworks available to conduct semi-structured interviews in the education and social sciences fields with children of primary school age, between 6–12, having no history of trauma or disabilities. To fill this gap, we conducted an integrative review of the literature to uncover current methodological considerations using the PRISMA procedure to search the ERIC, SAGE, ProQuest CBCA, SciELO and Redined databases. 19 methodological articles across multiple disciplines were retrieved and analyzed through inductive content analysis. We found that using traditional semi-structured interviews with children brings inherent challenges and that adaptation of current methods is not sufficient. A preliminary set of interviewing guidelines emphasizing a flexible and laissez-faire approach as well as a list of alternatives to the semi-structured interview for elementary-aged children are proposed, laying the foundations for a much-needed overhaul of current interviewing methods. AbstractThis article examines the potential of the ‘biography of things’ to enhance mobility studies in the context of the climate emergency of the Anthropocene. It suggests that mobility research should thoroughly consider non-human entities to address this unprecedented crisis. To achieve this goal, the article draws on anthropological literature about the biography of things and new materialist discourses about non-human agency. The text begins by exploring vitalism as a philosophical foundation for investigating the agency of things and their relationship to the biography of things in mobility studies. To address this issue, the ‘mobility biography of things’ methodology is proposed as a valuable approach. This methodology acknowledges the agency and ‘personhood’ of non-human entities, revealing the temporal and spatial entanglement of things by studying their mobility throughout their lifespan. There is little evidence however, of the training either achieving its required learning outcomes, or delivering improvement in outcomes for patients. Whist organisations may be hitting their compliance target for mandatory training, is the purpose missing the point? This systematic review aims to synthesize and evaluate the efficacy of statutory and mandatory training.PubMed, EMBASE, CNAHL, ERIC and Cochrane Central registers were searched on 23rd May 2023. All research designs were included and reported training had to specify an organisational mandate within a healthcare setting. Data was coded using a modified Kirkpatrick (KP) rating system. Critical appraisal was undertaken using the Modified Medical Education Research Study Quality Instrument, Critical Appraisal Skills Programme Qualitative Studies checklist and Mixed Methods Assessment Tool.Twenty-five studies were included, featuring 9132 participants and 1348 patient cases audited. Studies described evaluation of mandatory training according to Kirkpatrick’s outcomes levels 1–4b, with the majority (68%) undertaken in the UK and within acute settings. Training duration varied from 5 min to 3 days. There is a lack of consensus regarding mandatory training rationale, core topics, duration, and optimum refresher training period. Currently, mandatory training does not consistently translate to widescale improvements in safe practice or improved patient outcomes.Due to the lack of international consensus regarding the need for mandated training, most papers originated from countries with centrally administered national health care systems. The rationale for mandating training programmes remains undefined. The assumption that mandatory training is delivering safe practice outcomes is not supported by studies included in this review. The objective of this study was to reveal the efficiency of Open-Ended Emotive Projection Tests (OEEPTs) and Open-Ended Questions (OEQs) in understanding consumers' reactions to products processed with innovative food processing technologies (IPTs) and the influence of such communication on consumer attitudes, perception of appropriateness and willingness to perform different behaviors. A representative sample of 1206 Norwegian participants were organized in six experimental groups and responded to OEEPTs, an Emotive Projection Test (EPT), OEQs and survey questions. OEEPTs and EPTs showed no significant differences in consumers' responses between experimental conditions. OEQs indicated that the information on food processed as usual was preferred, while responses to food processed using IPT were not influenced by information about its benefits. This result was confirmed by willingness to try, buy and eat the same products. The main managerial implication is that communicating about benefits should be prioritized over information about IPTs. Virtual reality (VR) is a technology that is seeing increasing use in medical education as a means to complement or prepare students for clinical practice in a safe space. Whilst effective for learning, it can be difficult to use effectively and requires significant planning to avoid the technological tail wagging the educational dog. We have run educational sessions using the technology to teach anatomy and clinical reasoning that have been well received by students at Queen Mary, University of London. The RNA world hypothesis suggests that early cellular ancestors relied solely on RNA molecules for both genetic information storage and cellular functions. RNA, composed of four nucleosides—adenosine, guanosine, cytidine, and uridine—forms the basis of this theory. These nucleosides consist of purine nucleobases, adenine and guanine, and pyrimidine nucleobases, cytosine and uracil, bonded to ribose sugar. Notably, carbonaceous chondrite meteorites have revealed the presence of these bases and sugar, hinting at the potential existence of nucleosides in space. This study aims to present the infrared spectra of four RNA nucleosides commonly found in terrestrial biochemistry, facilitating their detection in space, especially in astrobiological and astrochemical contexts. Laboratory measurements involved obtaining mid- and far-IR spectra at three temperatures (−180 °C, room temperature, and +180 °C), followed by calculating molar extinction coefficients (ε) and integrated molar absorptivities (ψ) for corresponding bands. These spectral data, along with ε and ψ values, serve to provide quantitative insights into the presence and relative abundance of nucleosides in space and aid in their detection. Integration and coordination among monitoring programs are needed to better assess the impacts of climate change on Arctic marine ecosystems. The use of common indicators and methods could facilitate this comprehensive understanding. We completed a systematic review of published marine research in the western Canadian Arctic from 1962 to 2021 to identify the commonalities in methods and indicators present in assessments of coastal and offshore ecosystems. Most abundant in our sample were indicators addressing the environmental context, followed by indicators concerning trophic webs and biological organisms, and finally indicators associated with anthropogenic stressors and threats. Ship-based studies located far offshore were by far the most common and focused on indicators that characterized the physical environment and lower trophic levels. Commonalities in data collection methods suggest possibilities for standardization among programs for some parameters. Differences and data gaps highlighted areas for future coordination and the potential to integrate among indicators, especially as some indicators may span coastal to offshore ecosystems whereas individual monitoring programs may not. The results of this review could be used to identify and gather data into broad-spanning datasets. Overall, this systematic review highlights opportunities to link indicators and methods among coastal to offshore programs and will therefore facilitate connectivity and coordination of ecological research and monitoring in the western Canadian Arctic. Much of psychological science relies on collaboration—from generating new theories and study ideas, to collecting and analyzing data, to writing and sharing results with the broader community. Learning how to collaborate with others is an important skill, yet this process is not often explicitly discussed in academia. Here, five researchers from diverse backgrounds share their experiences and advice on starting and sustaining collaborations. In doing so, they reflect on aspects of both successful (and failed) collaborations with students, colleagues within and outside of psychology, and members of industry and organizational partners beyond academia. Recommendations and challenges of productive collaborations are discussed, along with examples of how collaborative teams can contribute to psychological science, address real‐world issues, and make the process of conducting research more enjoyable and rewarding. Cannabis use disorder (CUD) is increasing in the United States, yet, specific neural mechanisms of CUD are not well understood. Disordered substance use is characterized by heightened drug cue incentive salience, which can be measured using the late positive potential (LPP), an event‐related potential (ERP) evoked by motivationally significant stimuli. The drug cue LPP is typically quantified by averaging the slow wave's scalp‐recorded amplitude across its entire time course, which may obscure distinct underlying factors with differential predictive validity; however, no study to date has examined this possibility. In a sample of 105 cannabis users, temporo‐spatial Principal Component Analysis was used to decompose cannabis cue modulation of the LPP into its underlying factors. Acute stress was also inducted to allow for identification of specific cannabis LPP factors sensitive to stress. Factor associations with CUD severity were also explored. Eight factors showed significantly increased amplitudes to cannabis images relative to neutral images. These factors spanned early (~372 ms), middle (~824 ms), and late (>1000 ms) windows across frontal, central, and parietal‐occipital sites. CUD phenotype individual differences were primarily associated with frontal, middle/late latency factor amplitudes. Acute stress effects were limited to one early central and one late frontal factor. Taken together, results suggest that the cannabis LPP can be decomposed into distinct, temporal‐spatial factors with differential responsivity to acute stress and CUD phenotype variability. Future individual difference studies examining drug cue modulation of the LPP should consider (1) frontalcentral poolings in addition to conventional central‐parietal sites, and (2) later LPP time windows. The current study is the first to utilize temporo‐spatial principal component analysis (PCA) to decompose the cannabis cue modulation of the late positive potential (LPP) before and after an acute laboratory stressor. Objectives: Ancient human dental calculus is a unique, nonrenewable biological resource encapsulating key information about the diets, lifestyles, and health conditions of past individuals and populations. With compounding calls its destructive analysis, it is imperative to refine the ways in which the scientific community documents, samples, and analyzes dental calculus so as to maximize its utility to the public and scientific community. Materials and Methods: Our research team conducted an IRB‐approved survey of dental calculus researchers with diverse academic backgrounds, research foci, and analytical specializations. Results: This survey reveals variation in how metadata is collected and utilized across different subdisciplines and highlights how these differences have profound implications for dental calculus research. Moreover, the survey suggests the need for more communication between those who excavate, curate, and analyze biomolecular data from dental calculus. Discussion: Challenges in cross‐disciplinary communication limit researchers' ability to effectively utilize samples in rigorous and reproducible ways. Specifically, the lack of standardized skeletal and dental metadata recording and contamination avoidance procedures hinder downstream anthropological applications, as well as the pursuit of broader paleodemographic and paleoepidemiological inquiries that rely on more complete information about the individuals sampled. To provide a path forward toward more ethical and standardized dental calculus sampling and documentation approaches, we review the current methods by which skeletal and dental metadata are recorded. We also describe trends in sampling and contamination‐control approaches. Finally, we use that information to suggest new guidelines for ancient dental calculus documentation and sampling strategies that will improve research practices in the future. The parietal P300 wave of event‐related potentials (ERPs) has been associated with various psychological operations in numerous laboratory tasks. This study aims to decompose the P3 wave of ERPs into subcomponents and link them with behavioral parameters, such as the strength of stimulus–response (S‐R) links and GO/NOGO responses. EEGs (31 channels), referenced to linked ears, were recorded from 172 healthy adults (107 women) who participated in two cued GO/NOGO tasks, where the strength of S‐R links was manipulated through instructions. P300 waves were observed in active conditions in response to cues, GO/NOGO stimuli, and in passive conditions when no manual response was required. Utilizing a combination of current source density transformation and blind source separation methods, we decomposed the P300 wave into two distinct components, purportedly originating from different parts of the parietal lobules. The amplitude of the parietal midline component (with current sources around Pz) closely mirrored the strength of the S‐R link across proactive, reactive, and passive conditions. AbstractIn this guide we provide instructions and recommendations about creating and running escape rooms for healthcare education. In recent years there has been a growing interest in adopting escape rooms as an educational tool to be included in healthcare curricula, and we attempt to explain why and how these tools are fit for the particularities of this type of education. We first describe the steps that a design team will have to follow to create an educational escape room from scratch, from core characteristics like target audience and learning goals to actual puzzle design and testing. We then continue by providing recommendations to operators and lecturers about how to run such escape room as part of an overall teaching session that also includes a lecture, briefing, debriefing and evaluation. Recent research observed 92% accuracy for age‐at‐death estimations by U.S. forensic anthropologists. The present study compares this case report level accuracy to method level accuracy for the most commonly used methods in U.S. casework, drawing from the Forensic Anthropology Database for Assessing Methods Accuracy (FADAMA). Method application rate (i.e., how often a method is used in casework) was analyzed for n = 641 cases and identified 15 methods with an application rate >45 cases, and the present study focused further analyses on these 15 methods. Of the 15, only four yielded accuracies greater than or equal to the 92% documented for case‐report level accuracy. The other 11 methods produced accuracy rates ranging from 54% to 91%, with six of these below 70% This disconnect between highly accurate age estimations at the case report level compared to the poor performance at method level suggests that practitioner interpretation and synthesis of the methods' outcomes is a critical step for increasing the accuracy rates of the age estimations as reported on the final case report. This inference was further supported by the study's results which indicated that practitioner interpretations of frequently used method combinations improve accuracy and age range width of age estimation. The study also performed a Fisher's Exact test to assess whether case report‐level accuracy differed with the number of aging methods used in a case, and found no significant differences. Cerebrospinal fluid (CSF) matrix biomarkers have become increasingly valuable surrogate markers of neuropsychiatric diseases in research and clinical practice. In contrast, CSF cells have been rarely investigated due to their relative scarcity and fragility, and lack of common collection and cryopreservation protocols, with limited exceptions for neurooncology and primary immune-based diseases like multiple sclerosis. the advent of a microfluidics-based multi-omics approach to studying individual cells has allowed for the study of cellular phenotyping, intracellular dynamics, and intercellular relationships that provide multidimensionality unable to be obtained through acellular fluid-phase analyses. challenges to cell-based research include site-to-site differences in handling, storage, and thawing methods, which can lead to inaccuracy and inter-assay variability. In the present study, we performed single-cell RNA sequencing (10x Genomics) on fresh or previously cryopreserved human CSF samples from three alternative cryopreservation methods: Fetal Bovine Serum with Dimethyl sulfoxide (FBS/DMSO), FBS/DMSO after a DNase step (a step often included in epigenetic studies), and cryopreservation using commercially available Recovery© media. In comparing relative differences between fresh and cryopreserved samples, we found little effect of the cryopreservation method on being able to resolve donor-linked cell type proportions, markers of cellular stress, and overall gene expression at the single-cell level, whereas donor-specific differences were readily discernable. We further demonstrate the compatibility of fresh and cryopreserved CSF immune cell sequencing using biologically relevant sexually dimorphic gene expression differences by donor. Our findings support the utility and interchangeability of FBS/DMSO and Recovery cryopreservation with fresh sample analysis, providing a methodological grounding that will enable researchers to further expand our understanding of the CSF immune cell contributions to neurological and psychiatric disease. AbstractHuman users are increasingly recognized as a vector of cybersecurity attack. One problem that contributes to this condition is the growing complexity of digital tools. Such complexity can make it difficult for users to understand how tools work and how their actions will impact security. This work sought to answer the research question: Can mental modeling analyses (from human factors engineering and human-automation interaction) be developed to effectively discover cybersecurity risks? To answer this, we extend mental models with cybersecurity-specific concepts. The resulting models are then incorporated into model checking analyses (an automated approach to formal verification) to discover if and when mismatches between human mental models and systems can cause security failures. We evaluated our approach by successfully applying it to a case study regarding the security configuration of a popular cloud data storage service. The exaggerators (Ghulāt) were the first group of interpretive currents in Shiite history to introduce heresy into Islamic teachings by incorporating their exaggerated ideas into Shi'i interpretive and hadith texts. They were divided into two groups of atheists and nonatheists, each of whom entered Quranic interpretation with their own presuppositions and interpreted the verses of the Quran according to their own methods. Therefore, it is very important to know their bases and methods of interpretation in order to recognize them better. In this article, an attempt is made to extract the exaggerations in Shi'a Quranic commentaries. Using the descriptive-analytical-critical method, their foundations and methods are examined and criticized and validated with the help of hadith criticism tools. Based on the hadiths of the great exaggerators, it is found that most of their hadiths in the Shi'ah commentaries have fundamental problems in terms of sanad and hadith text. The Food Finding Test (FFT) olfactory paradigm without overnight food deprivation examined olfaction in aged (16-months-old) animals. Ethograms of three goal-directed behaviors towards hidden food (sniffing, finding and eating) elicited in male and female 3xTg-AD mice for Alzheimer's disease (AD) and their age-matched C57BL/6 wild-type counterparts with normal aging were meticulously analyzed with the support of video recordings. The new FFT protocol elicited longer ethograms than previously reported with the standard deprivation protocol. However, it was sensitive when identifying genotype- and sex-dependent olfactory signatures for the temporal patterns of slow sniffing, finding, and eating in AD and males, but it had a striking consistency in females. The impact of forced social isolation was studied and it was found to exert sex-dependent modifications of the ethogram, mostly in males. Still, in both sexes, a functional derangement was detected since the internal correlations among the behaviors decreased or were lost under isolated conditions. In conclusion, the new paradigm without overnight deprivation was sensitive to sex (males), genotype (AD), and social context (isolation-dependent changes) in its ethogram and functional correlation. Understanding the numerous roles that colouration serves in the natural world has remained a central focus in many evolutionary and ecological studies. However, to accurately characterise and then compare colours or patterns among individuals or species has been historically challenging. In recent years, there have been a myriad of new resources developed that allow researchers to characterise biological colours and patterns, specifically from digital imagery. However, each resource has its own strengths and weaknesses, answers a specific question and requires a detailed understanding of how it functions to be used properly. These nuances can make navigating this emerging field rather difficult. Herein, we evaluate several new techniques for analysing biological colouration, with a specific focus on digital images. First, we introduce fundamental background knowledge about light and perception to be considered when designing and implementing a study of colouration. We then show how numerous modifications can be made to images to ensure consistent formatting prior to analysis. After, we describe many of the new image analysis approaches and their respective functions, highlighting the type of research questions that they can address. We demonstrate how these various techniques can be brought together to examine novel research questions and test specific hypotheses. Finally, we outline potential future directions in colour pattern studies. Background: Clinical educators in geriatrics are often tasked with presenting a literature update at annual conferences and scientific meetings, which is a highly regarded continuing medical education (CME) activity. Preparation of an annual literature update cannot rely on bibliometric analysis due to time lag and poor correlation between bibliometrics and expert opinion on clinical relevance. The methodology of how top research articles of the year are selected and presented is not often reported. Methods: We conducted a scoping review for published reports of a curated selection of recent articles critically appraised for high impact to clinical practice in general geriatrics, published from 2010 to 2022. Results: Six annual literature updates were included for study. Three updates detailed their article sources, ranging from a survey of clinicians, consulting seven individual journals, searching up to four bibliographic databases, scanning social media outlets, and reviewing previous literature updates. One update reported a detailed method of article selection and consensus development. Critical appraisal of articles followed a structured reporting of clinical context, methods, results, and a statement of clinical implication or bottom line. Three of the six updates' results were disseminated in an annual conference update and did not evaluate learning outcomes of the audience. We mapped the results on a four‐step framework of article search, selection, critical appraisal, and dissemination of knowledge. Conclusions: Educators in geriatrics consult numerous article sources spanning multiple journals, databases, social media, and peer suggestions to create an annual literature update. The methodology of article search and selection is inconsistently described. Aims: To discuss the construction and use of vignettes as a novel approach in spiritual care research and education. Design: Methods paper. Methods: In this methods paper, the authors introduce the use of vignettes in spiritual care research and provide insight into the construction of vignettes. The vignette presented was part of a study of neurosurgical nurses' attitudes and responses to the spiritual needs of neuro-oncology patients. The development process, consisting of four steps, is explained in this paper. Results: Using a vignette to explore nurses' attitudes towards spiritual care is an innovative way to understand what behaviours nurses consider appropriate in situations where the patient is seeking meaning and connection. Transparent description of the development process is crucial to ensure reproducibility. Conclusion: The use of theoretically constructed and validated vignettes in spiritual care research is new. Vignettes used in surveys have the potential to elicit nurses' responses to patients' search for meaning and connectedness. Implications: In order to investigate nurses' attitudes and behaviours towards patients' spiritual needs, carefully constructed and validated vignettes are valuable research tools. Impact: Vignettes have proven to be a valuable research tool in the social and health sciences. So far, their use as a survey instrument in spiritual care research has not been investigated. Screening for adolescent sexual activity is a vital aspect of comprehensive pediatric care. Adolescents engage in risky sexual behaviors. Thus, a complete and accurate sexual health history can assist in the prevention and treatment of disease, prevention of unwanted pregnancy, treatment of existing diseases, and optimal planning of future healthcare for adolescents. Current evidence shows that provider-focused strategies improve the delivery of preventive services, including sexual health screenings. In this initiative, we assessed and examined pre- and post-screening rates for sexual activity among adolescents by advanced practice providers. This multi-site initiative was implemented in four school-based health centers and a school-linked center that included 2,102 unique patients ages 9 to 24 years. Our biphasic intervention included education for advanced practice providers and electronic health record modifications. Pre- and post-data collection was conducted to determine changes in the rate of screening for sexual activity during a primary care adolescent health visit over a 3-year period. Data were collected via retrospective medical chart review and analyzed in three time periods for comparison from 2018 to 2021. Screening rates for sexual activity increased significantly after the intervention (all p <.001) with the likelihood more than double that of the year before it was implemented. The intervention was deemed to be a feasible and cost-effective strategy to improve the provider's willingness and ability to provide more adolescent sexual health screenings. [ABSTRACT FROM AUTHOR]

Less than 1% of studies on racialized health inequities have empirically examined their root cause: structural racism. Moreover, there has been a disconnect between the conceptualization and measurement of structural racism. This study advances the field by (1) distilling central tenets of theories of structural racism to inform measurement approaches, (2) conceptualizing U.S. states as racializing institutional actors shaping health, (3) developing a novel latent measure of structural racism in states, (4) using multilevel models to quantify the association between structural racism and five individual-level health outcomes among respondents from the Health and Retirement Study (N = 9,020) and the Behavioral Risk Factor Surveillance System (N = 308,029), and (5) making our measure of structural racism publicly available to catalyze research. Results show that structural racism is consistently associated with worse health for Black people but not White people. Background: Intervention fidelity is a critical element of randomized controlled trials, yet reporting of intervention fidelity among attention control arms is limited. Lack of fidelity to attention control procedures can affect study outcomes by either overestimating or underestimating the efficacy of the intervention under examination. Objectives: This brief report describes the approach researchers took to promote fidelity to the attention control arm of a pediatric palliative care randomized controlled trial funded by the National Institutes of Health. Methods: The Informational Meetings for Planning and Coordinating Treatment trial aims to determine the efficacy of a communication intervention that uses care team dyads (i.e., physicians partnered with nurses or advanced practice providers) to engage parents of children with cancer who have a poor prognosis in structured conversations about prognostic information, goals of care, and care planning. The intervention is compared with an attention control arm, which provides parents with structured conversations on common pediatric cancer education topics, such as talking to their child about their cancer, clinical trials, cancer treatment, side effects, and so forth. National Institutes of Health guidelines for assessing and implementing strategies to promote intervention fidelity were used to design (a) the attention control arm of a randomized controlled trial, (b) related attention control arm training, and (c) quality assurance monitoring. Results: Attention control study procedures were designed to mirror that of the intervention arm (i.e., same number, frequency, and time spent in study visits). Cluster randomization was used to allocate care team dyads to one arm of the randomized controlled trial. Care team dyads assigned to the attention control arm participated in online training sessions to learn attention control procedures, the different roles of research team members, and quality assurance methods. Fidelity to attention control procedures is assessed by both the interveners themselves and a quality assurance team. Discussion: Study design, training, and delivery are all critical to attention control fidelity. Baseline training often needs to be supplemented with booster training when time gaps occur between study start-up and implementation. As education preparation programs align their instruction to the contemporary technologies used in schools, they may begin to design and deliver virtual reality instruction in the teaching methods classroom. In this study, we examined the experiences of 24 pre-service teachers as they used virtual reality for the first time through the lens of the Technology Acceptance Model, and we explored their perspectives on virtual reality in the elementary classroom. Qualitative data were obtained through two surveys. The findings indicated that pre-service teachers experienced discomfort with virtual reality and perceived a variety of barriers toward integrating it into their elementary classrooms. Based on the study's findings, practical implications are shared for teaching methods instructors as they design and integrate virtual reality instruction into the teaching methods classroom. This study aimed to assess whether recently proposed alternatives to the quality-adjusted life-year (QALY), intended to address concerns about discrimination, are suitable for informing resource allocation decisions. We consider 2 alternatives to the QALY: the health years in total (HYT), recently proposed by Basu et al, and the equal value of life-years gained (evLYG), currently used by the Institute for Clinical and Economic Review. For completeness we also consider unweighted life-years (LYs). Using a hypothetical example comparing 3 mutually exclusive treatment options, we consider how calculations are performed under each approach and whether the resulting rankings are logically consistent. We also explore some further challenges that arise from the unique properties of the HYT approach. The HYT and evLYG approaches can result in logical inconsistencies that do not arise under the QALY or LY approaches. HYT can violate the independence of irrelevant alternatives axiom, whereas the evLYG can produce an unstable ranking of treatment options. HYT have additional issues, including an implausible assumption that the utilities associated with health-related quality of life and LYs are "separable," and a consideration of "counterfactual" health-related quality of life for patients who are dead. The HYT and evLYG approaches can result in logically inconsistent decisions. We recommend that decision makers avoid these approaches and that the logical consistency of any approaches proposed in future be thoroughly explored before considering their use in practice. • A common criticism of the quality-adjusted life-year (QALY) is that life extensions generate fewer QALYs for those with poorer health-related quality of life. The equal value of life-years gained (evLYG) approach addresses this by assigning equal value to life extensions of similar length; however, this does not incentivize development of technologies that also improve health-related quality of life during life extension. The recently proposed health years in total (HYT) approach attempts to overcome both these issues. • We assess whether these recently proposed alternatives to the QALY are suitable for informing healthcare resource allocation decisions. Purpose: Polyaniline (PANI) has garnered attention for its potential applications in anticorrosion fields because of its unique properties. Satisfactory outcomes have been achieved when using PANI as a functional filler in organic coatings. More recently, research has extensively explored PANI-based organic coatings with self-healing properties. The purpose of this paper is to provide a summary of the active agents, methods and mechanisms involved in the self-healing of organic coatings. Design/methodology/approach: This study uses specific doped acids and metal corrosion inhibitors as active and self-healing agents to modify PANI using the methods of oxidation polymerization, template synthesis, nanosheet carrier and nanocontainer loading methods. The anticorrosion performance of the coatings is evaluated using EIS, LEIS and salt spray tests. Findings: Specific doped acids and metal corrosion inhibitors are used as active agents to modify PANI and confer self-healing properties to the coatings. The coatings' active protection mechanism encompasses PANI's own passivation ability, the adsorption of active agents and the creation of insoluble compounds or complexes. Originality/value: This paper summarizes the active agents used to modify PANI, the procedures used for modification and the self-healing mechanism of the composite coatings. It also proposes future directions for developing PANI organic coatings with self-healing capabilities. The summaries and proposals presented may facilitate large-scale production of the PANI organic coatings, which exhibit outstanding anticorrosion competence and self-healing properties. We argue that walking as a method provides an integrative approach to advance epistemic justice in sustainability research. The theory and practice of walking as a method has grown quickly within the social sciences and arts but remains underrepresented in sustainability research, where walking is typically an object of study (e.g., urban walkability). We argue that walking should be valued as an important mode of knowledge production that simultaneously widens sustainability knowledge, integrates diverse knowledge systems, and supports transdisciplinary sustainability solutions. In this perspective article, we consider the following questions: (1) Why is walking important to sustainability knowledge? (2) How can walk-based methods advance epistemic justice in sustainability knowledge? (3) What outcomes might we expect from cultivating walking as a method for sustainability knowledge? Current methods used to investigate emotional inference and conflict resolution knowledge are limited in their suitability for use with children with language disorders due to a reliance on language processing. This is problematic, as nearly 8% of the population are estimated to have developmental language disorder (DLD). In this paper, we present 'Zoti's Social Toolkit', a set of animated scenarios that can be used to assess emotion inferencing and conflict resolution knowledge. All animated scenarios contain interpersonal situations centred around a gender‐neutral alien named Zoti. Four studies investigated the face and construct validity of the stimuli. Iron deficiency is a common health issue in active and athlete populations. Accordingly, research into iron status, regulation, absorption, and iron deficiency treatment strategies is increasing at a rapid rate. However, despite the increase in the quantity of research, various methodological issues need to be addressed as we progress our knowledge in this area. The purpose of this review is to highlight specific considerations for conducting iron-related research in active and athlete populations. First, we discuss the methodological importance of assessment and interpretation of iron status, with reference to blood collection protocols, participant screening procedures, and biomarker selection. Next, we consider numerous variables that should be accounted for in the design of iron-related research studies, such as the iron regulatory hormone hepcidin and its interaction with exercise, in addition to an examination of female physiology and its impact on iron metabolism. Subsequently, we explore dietary iron and nutrient interactions that impact iron regulation and absorption, with recommendations made for optimal methodological control. Consideration is then given to key features of long-term study designs, such as the monitoring of training load, oral iron supplementation, dietary analysis, and general lifestyle factors. Finally, we conclude our recommendations with an exploration of stable iron isotope tracers as a methodology to measure iron absorption. Ultimately, it is our intention that this review can be used as a guide to improve study design, biomarker analysis, and reporting of findings, to maximize the quality of future research outputs in iron-related research focused on active and athlete populations. In this article, I make the case for a methodological turn in the field based on the growing interest in a methodology-oriented RE. That is, the idea that we should make academic methods and methodology not just a part of, but part and parcel of the core of teaching about religion and worldviews in public education. This interest is found in research articles, new RE curricula and debates about them, and concrete approaches and teaching materials. The article has three sections. In the first I bring these different contributions from England, Norway and Switzerland together, highlighting key terms and conceptualisations. In the second, I present eight arguments in support of a methodology-oriented RE. In the third, I discuss three questions raised by a methodological turn in the field: (1) How do we translate academic methods into the classroom? (2) How should we think about the relationship between traditional knowledge acquisition and exploration with different methods? (3) What methods should we prioritise, and why? Skateboarding is an informal activity with a relatively low cost of entry, and a range of potential practice grounds. Without formal gatekeepers, it is potentially inclusive on social, economic, and cultural levels. Participation has increased overall in recent years, including by girls and young women, who are increasingly visible in skateboarding organisations, international competition, and media. However, skateboarding spaces remain dominated by white, middle-class, male participants. Why, then, is the increased diversity of participation not diversifying the wider culture of the sport? Furthermore, skateboarding research has not been methodologically innovative, limiting its potential to see what is happening. This paper charts the development of a mapping tool as part of a wider study of young woman skateboarders, designed to better understand how different skateboarders (and others) use, move, and interact within skateboarding spaces. Drawing on behavioural mapping frameworks, we show how our mapping tool developed into a comprehensive, transferable system through which complex, fast moving, leisure settings can be studied. We conclude that being in a space does not always confer full access to participate in it. Drawing on PhD fieldwork on the practice and experience of ‘welcome’ in Berlin in the wake of the so‐called ‘refugee crisis’ of 2015–2016, this article engages with the ethics and practicalities of note‐taking. It does so by focusing on the production of particular spaces of welcome and hostility in and around one neighbourhood in particular, Neukölln, an important place of arrival for newcomers both historically and in the present day. At present, river estuaries are constantly being explored; therefore, it is necessary to receive up-to-date information on their actual environmental condition, hydrological regime, and geographical features. Therefore, it is important to develop and improve the methods and technical instruments for monitoring river estuaries. The article presents a developed methodology for integrated research of small-river estuaries using new methods and techniques. The article discusses the experience in using modern domestic devices for solving practical problems of integrated water-environmental monitoring with a case study of expeditionary work in estuaries of the Chernaya River (Crimean Peninsula, Sevastopol). The possibilities of the following technical instruments used are described: The GAP-AK-12R CTD (MHI RAS), the Condor biophysical complex (Aquastandard), and the Garmin echo sounder, which make it possible to obtain field data on turbidity, the concentration of suspended particulate matter, speed and direction of currents, the average size of suspended particles, and salinity and temperature of the aquatic environment. The results of applying a new method for determining the flow velocity and water consumption based on video image processing are presented. Application of the developed methodology makes it possible to obtain the information necessary to analyze the hydrological regime and ecological state of small-river estuaries. While the use of video recording as a method of data collection has helped researchers to resolve the challenge of capturing classroom interactions between teachers and students, it can be challenging for the researchers and teachers to make sense of the rich data collected. This paper describes an approach of analysing and visualising a language lesson with lesson microgenre and network graphs to provide an overview map of the lesson enactment. Studying the language lesson from a lesson microgenre perspective can provide both the co-text and context of the lesson when specific segments of the lesson are identified for interpretation and reflection by the teacher. The lesson map is visualised using network graphs to show the progression, connections, and patterns of the lesson microgenres. The paper describes the application of the approach in a study of the English lessons conducted by two teachers in a Singapore primary school and discuss the implications of the approach on teacher training. • There is a shift from a perspective biased towards visual methods to a broader understanding of the multisensory phenomenon known as "sensescape". Yet, the interplay among senses remains underexplored. • There is a lack of a universally agreed-upon definition for certain affects which are often characterized by their longer durations, ambiguity, and irreducibility. • Some functional studies explore attitudes to enhance cycling rates, while identity research emphasizes local context. Identities of cyclists in our sample face marginalization, stigma, disadvantage, and invisibility. • Retrospective surveys and interviews are dominant, but growing interest in mobile methods enhances real-time and context-specific data collection. • The positive values of cycling experiences have been largely ignored in our sample of studies, suggesting that the dominant narrative is still guided by the assumption that mobility is a disutility. When evaluating cycling subjective experiences (CSE), mobility researchers have questioned the depictions of cycling as an efficient, fast, and solitary mobility mode. By reframing cycling in terms of its emotional impact on the cyclist, research to date has explored dimensions such as fun, relaxation, and sociability of cycling experiences. Yet, these insights have not been integrated into a holistic understanding of CSE. Addressing this gap, this paper asks: what is the CSE exactly and how do we measure it? We selected and analysed in-depth 50 articles with the aim of unpacking the innate characteristics of CSE. The paper makes a contribution to the research on CSE by presenting a novel framework that clarifies the relationships within existing literature, and identifies measurement methods aligned with this framework. The three interrelated core aspects of CSE are 1) sensory interpretation, 2) affective states, and 3) cognitive construction. Additionally, the three identified methods are 1) retrospective, 2) interceptive, and 3) mobile methods. Notably, retrospective surveys and interviews emerged as the dominant methods employed in the field. However, there has been a growing interest in mobile methods, which enable the collection of real-time and context-specific data, thereby enhancing the generalizability of research findings. Through our analysis we have found that the positive values of cycling experiences have been largely ignored in our sample of studies, revealing a bias of researchers to focus on mobility as a disutility. Based on our findings, we urge planners and scholars to rethink their implicit efforts to mitigate the negative effects of cycling experiences and look for opportunities to optimize for positive cycling experiences. AbstractObjectiveMethodsResultsConclusionAlthough non-suicidal self-injury (i.e., NSSI) has been suggested as a robust risk factor of suicide, NSSI related information that is most related to suicide risk remains unclear. Commonly studied NSSI characteristics are its frequency and the number of methods endorsed. However, it may not be merely how frequent or how many different methods that matters, but “why,” which alludes to the importance of NSSI functions (or why individuals engage in NSSI). Thus, this study examined how the interactions between NSSI characteristics and functions are associated with suicide risk.Undergraduate students (n = 820) with a lifetime history of NSSI, filled out self-report measures on NSSI and suicide risk. A hierarchical regression analysis was conducted to examine the moderation effects between four 2-way interactions (i.e., method X intrapersonal; method X interpersonal; frequency X intrapersonal; frequency X interpersonal) on suicide risk.Main effects of all four independent variables were statistically significant. In terms of interactions, the intrapersonal function moderated both the effects of NSSI frequency and methods on suicide risk, whereas the interpersonal function moderated the effects of NSSI frequency on suicide risk.Our results highlight that some NSSI related information than others are more indicative of suicide risk. In particular, the combination of NSSI functions, along with its frequency and number of methods, holds promise when assessing for current and lifetime suicidal thoughts and behaviors.
