Microbiota and luminal components may affect epithelial integrity and thus participate in the pathophysiology of colon cancer (CC) and inflammatory bowel disease (IBD). Therefore, we aimed to determine the effects of fecal luminal factors derived from patients with CC and ulcerative colitis (UC) on the colonic epithelium using a standardized colon-derived two-dimensional epithelial monolayer. The complex primary human stem cell-derived intestinal epithelium model, termed RepliGut® Planar, was expanded and passaged in a two-dimensional culture which underwent stimulation for 48 h with fecal supernatants (FS) from CC patients (n = 6), UC patients with active disease (n = 6), and healthy subjects (HS) (n = 6). mRNA sequencing of monolayers was performed and cytokine secretion in the basolateral cell culture compartment was measured. The addition of fecal supernatants did not impair the integrity of the colon-derived epithelial monolayer. However, monolayers stimulated with fecal supernatants from CC patients and UC patients presented distinct gene expression patterns. Comparing UC vs. CC, 29 genes were downregulated and 33 genes were upregulated, for CC vs. HS, 17 genes were downregulated and five genes were upregulated, and for UC vs. HS, three genes were downregulated and one gene was upregulated. The addition of FS increased secretion of IL8 with no difference between the study groups. Fecal luminal factors from CC patients and UC patients induce distinct colonic epithelial gene expression patterns, potentially reflecting the disease pathophysiology. The culture of colonic epithelial monolayers with fecal supernatants derived from patients may facilitate the exploration of IBD- and CC-related intestinal microenvironmental and barrier interactions. Background We compared kidney and cardiorenal protection in patients without type 2 diabetes across urine albumin–creatinine ratio (UACR) levels after initiation on dapagliflozin for the treatment of chronic kidney disease (CKD). Methods OPTIMISE-CKD is an observational study describing dapagliflozin treatment for CKD. Adult patients with CKD without type 2 diabetes were included in the primary analysis. Baseline UACR was grouped as normal/mildly elevated (0–29 mg/g), low (30–200 mg/g) and high (>200 mg/g). Outcomes were estimated glomerular filtration rate (eGFR) trajectories/slopes, cardiorenal complications and all-cause mortality. Results In total, 1480 patients had low (n  = 796) and high (n  = 684) UACR. The two groups were similar at baseline, aged 75 and 74 years, and 42% and 39% female, respectively. After dapagliflozin initiation, an acute eGFR dip of 3 mL/min/1.73 m2 was observed, followed by a flat development in both groups. The eGFR slope [95% confidence interval (CI)] for patients with low UACR was 0.79 mL/min/1.73 m2 per year (–0.59, 2.56), and similar to patients with high UACR [0.40 mL/min/1.73 m2 per year (–0.46, 1.38)]. Risks of cardiorenal complications and all-cause mortality were similar, with adjusted hazard ratios of 0.89 (95% CI 0.66, 1.19) and 1.10 (95% CI 0.63, 1.92), respectively. Analogous results were found in those with normal/mildly elevated UACR. Conclusions Dapagliflozin in patients without type 2 diabetes for the treatment of CKD demonstrated similar kidney protection, cardiorenal and all-cause mortality risk across UACR levels. Summary Hydrogen–Deuterium Exchange Mass Spectrometry (HDX-MS) is a powerful protein characterization technique that provides insights into protein dynamics and flexibility at the peptide level. However, analyzing HDX-MS data presents a significant challenge due to the wealth of information it generates. Each experiment produces data for hundreds of peptides, often measured in triplicate across multiple time points. Comparisons between different protein states create distinct datasets containing thousands of peptides that require matching, rigorous statistical evaluation, and visualization. Our open-source R package, HDXBoxeR, is a comprehensive tool designed to facilitate statistical analysis and comparison of multiple sets among samples and time points for different protein states, along with data visualization. Clindamycin is a lincosamide antibiotic that has been used as a topical, oral, or injectable formulation for over five decades. It exhibits a narrow spectrum of microbiologic activity, primarily against gram-positive and anaerobic bacteria. In dermatology, clindamycin has been used primarily as a topical agent, usually for the treatment of acne vulgaris. Despite questions surrounding antibiotic resistance and/or its relative contribution to antibiotic treatment efficacy, a large body of data support the therapeutic value of topical clindamycin for acne vulgaris. As a systemic agent, clindamycin is used orally to treat a variety of cutaneous bacterial infections, and sometimes for acne vulgaris, with oral treatment for the latter less common in more recent years. The modes of action of clindamycin are supported by data showing both its anti-inflammatory and antibiotic mechanisms, which are discussed here along with pharmacokinetic profiles and structure-activity relationships. The diverse applications of clindamycin for multiple disease states, its efficacy, and safety considerations are also reviewed here, including for both topical and systemic formulations. Background Hyperkalaemia is a barrier to achieving optimal, guideline-directed treatment with renin–angiotensin–aldosterone system inhibitors (RAASis) in patients with chronic kidney disease (CKD) and/or heart failure (HF). This study describes the association between hyperkalaemia-related RAASi treatment reduction and the number of hospitalized days in patients with CKD and/or HF in Sweden and Japan. Methods Using data from health registers and hospital medical records, patients with CKD and/or HF currently receiving RAASis who experienced an index hyperkalaemia episode were identified and categorized as having maintained or reduced RAASi treatment post-index; propensity score matching (1:1) was applied to balance the groups in terms of baseline characteristics. Changes in the number of all-cause, CKD- and HF-related hospitalized days per patient-year during 6 months pre- versus post-index and the number of days alive and out of hospital (DAOH) during 6 months post-index were described. Results Overall, 20 824 and 7789 patients were included from Sweden and Japan, respectively, 42% and 38% of whom reduced their RAASi treatment after the index hyperkalaemia episode. During the 6 months post-index, all-cause hospitalization increased by 18.2 days [95% confidence interval (CI) 17.0–19.2] per person-year in Sweden and 17.9 days (95% CI 17.4–18.5) per person-year in Japan among patients with reduced RAASi treatment compared with increases of 9.4 days (95% CI 8.6–10.4) and 8.5 days (95% CI 8.0–9.0) per person-year, respectively, among patients with maintained RAASi treatment. The mean DAOH was 121.5 [standard deviation (SD) 75.0] in Sweden and 141.7 (SD 54.5) in Japan among patients with reduced RAASi treatment compared with 154.0 (SD 51.3) and 157.5 (SD 31.6), respectively, among patients with maintained RAASi treatment. Conclusion Patients whose RAASi treatment was reduced after a hyperkalaemia episode had more hospitalized days and fewer DAOH compared with patients whose RAASi treatment was maintained. Purpose: Sinonasal lymphoma (SL) is a rare lymphatic neoplasm of the nasal cavities, paranasal sinuses and nasopharynx. Whereas some risk factors for SL subtypes have been identified, their aetiology is unknown. Along with other predisposing factors, the viral association of lymphomas, such as Epstein-Barr virus (EBV) and Burkitt and Hodgkin lymphomas, is well-established. Modern molecular biology techniques have enabled the discovery of novel human viruses, exemplified by the protoparvovirus cutavirus (CuV), associated with cutaneous T-cell lymphoma. These findings, and the anatomical location of the sinonasal tract with its rich microbiome and infectious agents, justify in-depth studies among SL. Methods: We analysed the presence of 20 viruses of Orthoherpesviridae, Parvoviridae, and Polyomaviridae by qPCR in 24 SL tumours. We performed RNAscope in situ hybridisation (RISH) to localize the viruses. Parvovirus-specific IgG was analysed by enzyme immunoassay and targeted next-generation sequencing (NGS) was applied to detect CuV in plasma. Results: We detected viral DNA in 15/24 (63%) tumours; nine of EBV, six of human herpesvirus (HHV) -7, four each of HHV-6B and parvovirus B19, two of cytomegalovirus, and one each of CuV and Merkel-cell polyomavirus. We found tumours with up to four viruses per tumour, and localized CuV and EBV DNAs by RISH. Two of the ten plasma samples exhibited CuV IgG, and one plasma sample demonstrated CuV viremia by NGS. The present work describes the synthesis two novel derivatives of 4‐nitrobenzaldehyde through a single pot microwave assisted green synthetic method and the characterization of the synthesized drugs using UV, IR,1HNMR, Mass spectra and CHN analysis. The structure optimization of the synthesized anticancer agents was carried out using Gaussian 9. Also carbon quantum dots (CQDs) derived from amino acid precursors were synthesized (Cystine and threonine) by microwave assisted method, followed by the integration of CQDS together with 4‐nitrobenzaldehyde and its novel derivatives. The drugs were subjected to targeted drug delivery applications in cancer cell lines (PC3) The MTT assay results revealed that the CQDs conjugated with 4‐nitrobenzaldehye (D1) and one of the derivatives (D3) exhibited greater cytotoxic effects on PC3 cells compared to L929 cells. The outcomes demonstrated that this novel derivative could kill cancer cells as effectively as 4‐nitrobenzaldehyde conjugated with CQDs. Also, the potential cytotoxicity of drugs D1 and D3 were further confirmed by the apoptosis assay using flow cytometry, which also explains that apoptosis is the mechanism underlying cell death in PC3 cancer cells. The autofluorescence test were also carried out in PC3 cells lines and the drugs D1 and D3 showed significant autofluorescence in PE−A (29.9 % and 34.7 %), in Alexa Fluor‐488 (32.6 % and61.1 %) and in Bv510‐A (16.6 % and 23.2 %) respectively. Thus, the synergistic effect of CQDs together with 4‐nitrobenzaldehyde and its derivative drug D3 are indeed a promising medication towards the localized drug delivery applications in prostate cancer cells as well as the autofluorescence results will explore novel arenas in bioimaging applications. We investigated whether motivational and personality factors were related to PEAA in the domains of physical health, mental health, and social engagement.We used a nationally representative sample of German adults (SOEP-IS) covering the entire adult life span (N = 2,007, age range 16–94 years) to test our pre-registered hypotheses.Multiple regression analyses indicated that motivation (i.e. life goals and health-related worries) was consistently associated with PEAA in the matching domains and mediated the effects of openness to experience on PEAA. No other personality trait was associated with PEAA.Our findings suggest that individuals preferentially notice the expectations for active aging whose content relates to their personal concerns and goals. Proton Exchange Membrane Fuel Cells (PEMFCs) represent a promising green solution for energy production, traditionally relying on platinum-group-metal (PGM) electrocatalysts. However, the increasing cost and limited global availability of PGMs have motivated extensive research into alternative catalyst materials. PGM-free oxygen reduction reaction (ORR) catalysts typically consist of first-row transition metal ions (Fe, Co) embedded in a nitrogen-doped carbon framework. Key factors affecting their efficacy include intrinsic activity and catalyst degradation. Thus, alternative materials with improved characteristics and the elucidation of reaction and degradation mechanisms have been the main concerns and most frequently explored research paths. High intrinsic activity and active site density can ensure efficient reaction rates, while durability towards corrosion, carbon oxidation, demetallation, and deactivation affects cell longevity. However, when moving to the actual application in PEMFCs, electrode engineering, which involves designing the catalyst layer, and other critical operational factors affecting fuel cell performance play a critical role. Electrode fabrication parameters such as ink formulation and deposition techniques are thoroughly discussed herein, explicating their impact on the electrode microstructure and formed electrochemical interface and subsequent performance. Adjusting catalyst loading, ionomer content, and porosity are part of the optimization. More specifically, porosity and hydrophobicity determine reactant transport and water removal. High catalyst loadings can enhance performance but result in thicker layers that hinder mass transport and water management. Moreover, the interaction between ionomer and catalyst affects proton conductivity and catalyst utilization. Strategies to improve the three-phase boundary through the proper ionomer amount and distribution influence catalyst utilization and water management. It is critical to find the right balance, which is influenced by the catalyst–ionomer ratio and affinity, the catalyst properties, and the layer fabrication. Overall, understanding how composition and fabrication parameters impact electrode properties and behaviour such as proton conductivity, mass transport, water management, and electrode–electrolyte interfaces is essential to maximize electrochemical performance. This review highlights the necessity for integrated approaches to unlock the full potential of PGM-free materials in PEMFC technology. Objective: Effective communication between cancer patients and providers is critical for addressing psychological distress, reducing uncertainty, and promoting patient well‐being. This is particularly relevant during medical appointments that may elicit uncertainty, such as surgical consultations for newly diagnosed women with breast cancer. This study aimed to evaluate how pre‐appointment anxiety and illness uncertainty affect patient–provider communication in breast cancer surgical consultations and subsequent post‐appointment well‐being. Breast cancer patient anxiety has been studied as an outcome of provider communication, though less is known about the extent to which preexisting anxiety or uncertainty act as antecedents to effective patient–provider communication. Methods: This study analyzed videorecorded breast cancer surgical consultations (N = 51) and corresponding patient surveys to understand how pre‐appointment anxiety influences pre‐appointment patient uncertainty, patient–provider communication during the appointment, and subsequent post‐appointment uncertainty. Results: The proposed model achieved good fit to the data such that more pre‐appointment anxiety was associated with more pre‐appointment uncertainty, more pre‐appointment anxiety was associated with more empathic opportunities per minute, and more empathic opportunities were associated with less post‐appointment uncertainty. Conclusions: Results indicate breast cancer patients with anxiety pre‐appointment are at‐risk for more illness uncertainty and are more likely to explicitly provide empathic opportunities. This supports the need for added attention to empathic opportunities to not only address patients emotionally but to also assess whether a patient may be at higher risk of having preexisting anxiety. Proteus syndrome (PS) is an exceptionally uncommon genetic disorder that has been documented in only approximately 250 cases in the literature spanning the past four decades. It is characterized by a disproportionate, asymmetric overgrowth of all types of tissues, provoked by a somatic activating mutation in serine/threonine protein kinase 1. We report a case of PS in a two-year-old female patient with the following clinical features: unilateral overgrowth of connective tissue in the right buttock and right foot, where multiple surgeries were performed to achieve a desirable aesthetic outcome and ensure psychological comfort of the young patient. The insights provided by this case underscore the pivotal role of obtaining pleasing aesthetic outcomes in the surgical management of untreatable genetic disorders, with the aim of nurturing psychological contentment in affected children. Di-tert-butyldiphosphatetrahedrane (tBuCP)2 (A) is a reactive tetrahedral molecule which may serve as a source of new phosphaorganic molecules and ligands. However, the redox chemistry of this compound has not yet been investigated. Here, we show that the reduction of A with alkali metals (AM = Li, Na, K, Rb and Cs) affords 1,2,3-triphospholides [AM(crown ether)][1,2,3-P3C2tBu2] (1–5, [AM(crown ether)] = [Li([12]crown-4)2]+, [Na([15]crown-5)2]+, [K([18]crown-6)]+, [Rb([18]crown-6)]+, and Cs+) with 1,3-diphospholides [AM(crown ether)][1,3-P2C3tBu3] (6–10) formed as by-products. The potassium salt 3 was isolated on a preparative scale, allowing for reactivity studies. Transmetalation with iron(II) and ruthenium(II) chlorides yielded the sandwich complexes [Cp*M(η5-1,2,3-P3C2tBu2)] (11, M = Fe; 12, M = Ru, Cp* = C5Me5) featuring η5-coordinated triphospholide ligands. Treatment of 3 with [Cp2Fe][BAr4F] or [H(Et2O)2BAr4F] (BAr4F = B{C6H3(CF3)2}4) afforded the polyphosphorus compound tBu4C4P6 (13), which presumably results from the dimerisation of a 1,2,3-triphospholyl radical intermediate (1,2,3-P3C2tBu2)˙ (3˙). The control of transcription is crucial for homeostasis in mammals. A previous selective sweep analysis of horse racing performance revealed a 19.6 kb candidate regulatory region 50 kb downstream of the Endothelin3 (EDN3) gene. Here, the region was narrowed to a 5.5 kb span of 14 SNVs, with elite and sub-elite haplotypes analyzed for association to racing performance, blood pressure and plasma levels of EDN3 in Coldblooded trotters and Standardbreds. Comparative analysis of human HiCap data identified the span as an enhancer cluster active in endothelial cells, interacting with genes relevant to blood pressure regulation. Coldblooded trotters with the sub-elite haplotype had significantly higher blood pressure compared to horses with the elite performing haplotype during exercise. Alleles within the elite haplotype were part of the standing variation in pre-domestication horses, and have risen in frequency during the era of breed development and selection. These results advance our understanding of the molecular genetics of athletic performance and vascular traits in both horses and humans. Author summary: A previous study discovered that a genomic region close to the Endothelin3 gene was associated with harness racing performance. Here, careful phenotypic documentation of athletic performance and blood pressure measurements in horses, followed by state-of-the-art genomics, allowed us to identify a 5.5 kb regulatory region located approximately 50 kb 3' of the EDN3 gene. A comparative analysis of the region using human HiCap data supported a regulatory role as, in endothelial cells, interaction was observed between the region and multiple genes relevant to blood pressure regulation and athletic performance. Long range cis-regulatory modules are critical for cooperatively controlling multiple genes located within transcriptionally active domains. We measured blood pressure in Coldblooded trotters during exercise and demonstrated that horses with two copies of the elite-performing haplotype had lower blood pressure during exercise and better racing performance results, compared to horses with two copies of the sub-elite performing haplotype. In addition, horses with the elite-performing haplotype also had higher levels of Endothelin3 in plasma. The results reported here are important for understanding the biological mechanisms behind blood pressure regulation in relation to racing performance in both horses and humans. In the present study, we examined the effect of extreme temperatures on different life stages of Ephestia kuehniella Zeller (Lepidoptera: Pyralidae). The individuals were exposed to different temperatures ranging between 5 and −18 °C for cold treatment, and between 30 and 55 °C for heat treatment, and different exposure intervals, ranging from minutes to days. Complete control was achieved at −10 and −15 °C for cold treatment and 50 and 55 °C for heat treatment at all exposure intervals. Considering the efficacy of extreme temperatures for the control of E. kuehniella, our study provides specific temperature exposure modules that can be effective for the control of this species. Cancer patients often attend medical interactions with at least one companion. The degree to which companions participate varies, ranging from passive observer to active advocate. However, the structure of the medical interaction often promotes dyadic rather than triadic communication, creating ambiguity about to the degree to which companions can and should participate. Participants (N = 34, 16 dyads) included gynecologic cancer patients who were undergoing chemotherapy treatment (n = 18) and their companions (n = 16); all participants were separately interviewed. Interviews included discussion of dyadic communication patterns within medical interactions. The normative rhetorical theory (Goldsmith, 2019) was applied as a guiding framework. Patients discussed the dilemma they experience when companions are expected but absent. Patients and companions provided positive reports of companion communication when behavior aligned with expectations. Alternatively, patients and companions experience dilemmas when companions participate more than or differently from how patients and/or companions had expected. The first representatives of new fused tricyclic heterosystems, namely 3,3a‐dihydro‐1H‐imidazo[4′,5′:4,5]imidazo[2,1‐b]thiazoles and 3,3a‐dihydro‐1H‐imidazo[4′,5′:4,5]imidazo[2,1‐b][1,3]selenazoles have been synthesized through the regioselective reaction of diethyl acetylenedicarboxylate with semithioglycolurils, thioglycolurils or selenoglycolurils. Study of antifungal properties of the obtained compounds against Venturia inaequalis (V. i.), Rhizoctonia solani (R. s.), Fusarium oxysporum, Fusarium moniliforme, Bipolaris sorokiniana (B. s.) and Sclerotinia sclerotiorum phytopathogenic fungi has been performed. Tested compounds possessed good inhibition activity against R. s., B. s. with 45–94 % by mycelium growth inhibition test method, which were more active than the well–known azole fungicide triadimefon as positive control. The best active thiazole derivative was established to be effective against R. s. (94 %), B.s. (65 %) and V. i. (60 %). Objective: Support people of cancer patients are at significant risk for psychological distress. Additionally, cancer patients' well‐being is reciprocally associated with support peoples' psychological well‐being. Informed by Uncertainty in Illness Theory, this study tests whether support person psychological well‐being is influenced by provider communication and uncertainty reduction. Methods: We tested a multiple mediation model to investigate how empathic communication facilitates psychological adjustment in support people of cancer patients and how this process is mediated by support peoples' illness uncertainty and caregiver burden. Support people of cancer patients (N = 121; including spouses, adult children, etc.) completed an online questionnaire about their perceptions of oncologists' empathy, uncertainty about the cancer patients' illness, perceived caregiving burden, and their psychological adjustment to diagnoses. Results: Path analysis revealed that (1) more perceived oncologist empathy was associated with less illness uncertainty, (2) more illness uncertainty was associated with worse psychological adjustment and more perceived caregiver burden, and (3) more burden was associated with worse adjustment (χ2(2) = 1.19, p = 0.55; RMSEA < 0.01; CFI = 1.00; SRMR = 0.02). Conclusions: Given the reciprocal nature of well‐being between cancer patients and their support people, it is critical to understand and bolster support people's psychological well‐being. Results demonstrated how empathic provider communication can support psychological well‐being for support people of cancer patients. In the present work, the operating results from an innovative, renewable, energy-based space-heating and domestic hot water (DHW) system are shown. The system used solar thermal energy as its primary source and was assisted by a shallow geothermal application in order to accommodate the space-heating and DHW needs of a domestic building in Austria. The system incorporated phase-change materials (PCMs) in specially designed containers to function as heat-storage modules and provide an energy storage capability for both the space-heating and DHW subsystems. This system was designed, implemented, and tested under real operating conditions in a building for a period of one year. The operating and energy results for the system are demonstrated in this work. The system was compared with a conventional one, and a reduction in the primary energy consumption equal to 84.3% was achieved. The maintenance and operating costs of the system were reduced by 79.7% compared to the conventional system, thus significantly contributing to the NZEB target of the building. The newly proposed system, although presenting an increased operating complexity, utilizes an innovative self-learning control system that manages all of its operations. The combination of a solar thermal energy source with thermal energy storage increases the use of renewable energy by extending the capacity of the system beyond the solar hours and using excess solar energy for space-heating needs. The thermal energy storage unit also increases the energy and economic efficiency of the geothermal heat pump by operating it during the hours of a reduced electricity tariff and using the stored energy during hours of a high electricity demand. The cost for the installation of such a system is higher than a conventional one, but due to the significantly decreased operating costs, the pay-back period was calculated to be 8.7 years. Climate change mitigation requires countries to report their annual greenhouse gas (GHG) emissions and sinks, including those from land use, land use change, and forestry (LULUCF). In Finland, the LULUCF sector plays a crucial role in achieving net-zero GHG emissions, as the sector is expected to be a net sink. However, accurate estimates of LULUCF-related GHG emissions, such as methane (CH 4 ), remain challenging. We estimated LULUCF-related CH 4 emissions in Finland in 2013–2020 by combining national land cover and remote-sensed surface wetness data with CH 4 emissions estimated by an inversion model. According to our inversion model, most of Finland's CH 4 emissions were attributed to natural sources such as open pristine peatlands. However, our research indicated that forests with thin tree cover surrounding open peatlands may also be a significant source of CH 4 . Unlike open pristine peatlands and pristine peatlands with thin tree cover, surrounding transient forests are included in the Finnish GHG inventory if they meet the criteria used for forest land. The current Finnish national GHG inventory may therefore underestimate CH 4 emissions from forested organic soils surrounding open peatlands, although more precise methods and data are needed to verify this. Given the potential impact on net GHG emissions, CH 4 emissions from transitional forests on organic soils should be further investigated. Furthermore, the results demonstrate the potential of combining atmospheric inversion modelling of GHGs with diverse data sources and highlight the need for methods to more easily combine atmospheric inversions with national GHG inventories. Clustering time activity curves of PET images have been used to separate clinically relevant areas of the brain or tumours. However, PET image segmentation in multiorgan level is much less studied due to the available total-body data being limited to animal studies. Now, the new PET scanners providing the opportunity to acquire total-body PET scans also from humans are becoming more common, which opens plenty of new clinically interesting opportunities. Therefore, organ-level segmentation of PET images has important applications, yet it lacks sufficient research. In this proof of concept study, we evaluate if the previously used segmentation approaches are suitable for segmenting dynamic human total-body PET images in organ level. Our focus is on general-purpose unsupervised methods that are independent of external data and can be used for all tracers, organisms, and health conditions. Additional anatomical image modalities, such as CT or MRI, are not used, but the segmentation is done purely based on the dynamic PET images. The tested methods are commonly used building blocks of the more sophisticated methods rather than final methods as such, and our goal is to evaluate if these basic tools are suited for the arising human total-body PET image segmentation. First, we excluded methods that were computationally too demanding for the large datasets from human total-body PET scanners. These criteria filtered out most of the commonly used approaches, leaving only two clustering methods, k -means and Gaussian mixture model (GMM), for further analyses. We combined k -means with two different preprocessing approaches, namely, principal component analysis (PCA) and independent component analysis (ICA). Then, we selected a suitable number of clusters using 10 images. Finally, we tested how well the usable approaches segment the remaining PET images in organ level, highlight the best approaches together with their limitations, and discuss how further research could tackle the observed shortcomings. In this study, we utilised 40 total-body [18F] fluorodeoxyglucose PET images of rats to mimic the coming large human PET images and a few actual human total-body images to ensure that our conclusions from the rat data generalise to the human data. Our results show that ICA combined with k -means has weaker performance than the other two computationally usable approaches and that certain organs are easier to segment than others. While GMM performed sufficiently, it was by far the slowest one among the tested approaches, making k -means combined with PCA the most promising candidate for further development. However, even with the best methods, the mean Jaccard index was slightly below 0.5 for the easiest tested organ and below 0.2 for the most challenging organ. Type II diabetes is a chronic health condition and its successful management requires effective patient-provider communication. Responding to a call to model pathways between provider communication and patient health outcomes, this study tested four models of type II diabetic patient adherence with four mediators. Given the complex nature of type II diabetic care, patient adherence was conceptualized as wellness, screening, medication, and treatment adherence. Mediators included patient understanding, agreement, trust, and motivation. A sample of U.S. patients with type II diabetes patients who were both under the care of a medical provider and taking medication for their type II diabetes completed online surveys (n = 793). Findings indicated that the relationships between patient-centered communication and adherence outcomes were mediated by proximal outcomes. The results contribute to the understanding of patient-centered communication, adherence behaviors, and proximal outcomes of patient understanding, agreement, trust, and motivation. Findings indicate that relationships between patient-centered communication and wellness adherence is mediated by patient motivation, patient-centered communication and screening adherence is mediated by patient agreement, trust, and motivation, and patient-centered communication and treatment adherence is mediated by patient agreement, trust, and motivation. Communicating about hardships with close others can be challenging, leading to avoidance of hardship‐related topics. Although typically considered relationally damaging, topic avoidance could serve as a beneficial or neutral strategy when paired with relationally affirming communication. The current research investigates if the resilience communication processes outlined in the communication theory of resilience mitigate the negative relational effects of topic avoidance. Hypotheses are tested in two different contexts: sibling communication during parental caregiving (N = 207) and spousal communication in the first months of the COVID‐19 pandemic (N = 598). The processes of crafting normalcy, communication networks, and productive action were beneficial across contexts. When participants reported higher engagement in these processes, topic avoidance was not significantly related to relationship satisfaction. At lower engagement levels, topic avoidance was negatively related to relationship satisfaction. Nuance between contexts existed. The goal of this study was to develop an imaging approach to assess the integrity of the thymic epithelium in preclinical mouse models. The thymus is the most sensitive organ to cytoreductive treatments, and it rapidly involutes after chemotherapy insult in pediatric cancer patients. By using automated fluorescent whole slide scanning and complementary 3D SEM array tomography, we were able to visualize the biological structure of the thymic epithelial cell (TEC) network, analyze branching of this network, and obtain a more complete three-dimensional morphological structure of the cortical thymic epithelial cell (cTEC) compartment. The establishment of this imaging method will allow researchers to move forward in comparing the integrity of the cTEC in different conditions, such as different treatment conditions, for example, chemotherapy and irradiation, as well as different ages. Exposure to expectations for active aging may be modulated by age and individual resources (socioeconomic status, social integration, and health) via multiple pathways. Using a cross-sectional, nationally representative sample of adults aged 17 to 94 (N = 2,007), we investigated the relations between age, individual resources, and perceived expectations for active aging (PEAA) in three domains (physical health, mental health, and social engagement). Across domains, young adults and individuals aged 70+ reported slightly lower PEAA than emerging adults did; no other age differences emerged. Multiple regression showed that a higher subjective socioeconomic status, better perceived general health, and partnership (in older adults) predicted higher PEAA (almost) across domains, whereas church attendance, employment status, and occupational prestige yielded domain- and age-specific effects, which were not always positive. Previous research suggests Deaf signers may have different short-term and working memory processes compared with hearing nonsigners due to prolonged auditory deprivation. The direction and magnitude of these reported differences, however, are variable and dependent on memory modality (e.g., visual, verbal), stimulus type, and research design. These discrepancies have made consensus difficult to reach which, in turn, slows progress in areas such as education, medical decision-making, and cognitive sciences. The present systematic review and meta-analysis included 35 studies (N = 1,701 participants) that examined verbal (n = 15), visuospatial (n = 10), or both verbal and visuospatial (n = 10) serial-memory tasks comparing nonimplanted, Deaf signers to hearing nonsigners across the life span. Multivariate meta-analyses indicated a significant, negative effect of deafness on verbal short-term memory (forward recall), g = −1.33, SE = 0.17, p <.001, 95% CI [−1.68, −0.98], and working memory (backward recall), g = −0.66, SE = 0.11, p <.001, 95% CI [−0.89, −0.45], but no significant effect of deafness on visuospatial short-term memory, g = −0.055, SE = 0.17, p = 0.75, 95% CI [−0.39, 0.28]. Visuospatial working memory was not analyzed due to limited power. Population estimates for verbal and visuospatial short-term memory were moderated by age wherein studies with adults demonstrated a stronger hearing advantage than studies with children/adolescents. Quality estimates indicated most studies were of fair quality, with only 38% of studies involving Deaf authors. Findings are discussed in the context of both Deaf equity and models of serial memory. Intra-operative fluorescent imaging of endometriosis could help to optimize surgical treatment. Potential biomarkers to use as target for endometriosis-binding fluorescent probes were identified using a new five-phase transcriptomics-based approach to broaden the search for biomarkers. Using publicly available datasets, a differentially expressed gene (DEG) analysis was performed for endometriosis versus surgically relevant surrounding tissue (peritoneum, bladder, sigmoid, rectum, transverse colon, small intestine, vagina, and fallopian tubes) for which data was available. The remaining relevant surrounding tissues were analyzed for low expression levels. DEGs with a predicted membranous or extracellular location and with low expression levels in surrounding tissue were identified as candidate targets. Modified Target Selection Criteria were used to rank candidate targets based on the highest potential for use in fluorescent imaging. 29 potential biomarkers were ranked, resulting in Folate receptor 1 as the most potential biomarker. This is a first step towards finding a fluorescent tracer for intra-operative visualization of endometriosis. Additionally, this approach, using transcriptomics analysis to identifying candidate targets for a specific type of tissue for use in fluorescence-guided surgery could be translated to other surgical fields. Declining thymic functions associated either with old age (i.e., age-related thymic involution), or with acute involution as a result of stress, infectious disease, or cytoreductive therapies (e.g., chemotherapy/radiotherapy), have been associated with cancer development. A key mechanism underlying such increased cancer risk is the thymus-dependent debilitation of adaptive immunity, which is responsible for orchestrating immunoediting mechanisms and tumor immune surveillance. In the past few years, a blooming set of evidence has intriguingly linked obesity with cancer development and progression. The majority of such studies has focused on obesity-driven chronic inflammation, steroid/sex hormone and adipokine production, and hyperinsulinemia, as principal factors affecting the tumor microenvironment and driving the development of primary malignancy. However, experimental observations about the negative impact of obesity on T cell development and maturation have existed for more than half a century. Here, we critically discuss the molecular and cellular mechanisms of obesity-driven thymic involution as a previously underrepresented intermediary pathology leading to cancer development and progression. This knowledge could be especially relevant in the context of childhood obesity, because impaired thymic function in young individuals leads to immune system abnormalities, and predisposes to various pediatric cancers. A thorough understanding behind the molecular and cellular circuitries governing obesity-induced thymic involution could therefore help towards the rationalized development of targeted thymic regeneration strategies for obese individuals at high risk of cancer development. Young women in India report that avoidant talk within families hinders their ability to communicate about menstruation, further promoting stigma and limiting knowledge. Participants included 287 college-attending women from one of two colleges in different locations in northern India. Participants completed surveys that were guided by the disclosure decision-making model (DD-MM) and asked about menstruation-related topic avoidance with mothers. Results find that the stigma of being dramatic is a consistent predictor across the two locations but that predictors otherwise differ. Theoretical and practical implications are discussed. People's names and the places, animals, and items named by people are deeply personal and reflective of their culture and identity, yet in qualitative research, the standard is to use pseudonyms. This practice is thought to protect research participants, but when are "real" names most respectful and appropriate? How might researchers include research participants in these considerations? The purpose of this methodological article is to increase transparency and collaboration around the participant naming process, bringing much-needed attention to issues of power in the naming process in research. The authors review the literature and detail their reflexive engagement with pseudonyms; they advance issues for consideration and provide recommendations in the areas of power in participant naming, culturally responsive research, and pseudonym use with trans people and incarcerated people. Aim: The aim of the study was to estimate the annual health care cost by number of comorbid mental and somatic disorders in persons with a mental disorder. Methods: All persons living in Denmark between 2004 and 2017 with a hospital diagnosis of a mental disorder were identified. We investigated the cost of different health care services: psychiatric hospitals, somatic hospitals, primary health care (e.g. general practitioners, psychologists and so on) and subsidised prescriptions. Within those with at least one mental disorder, we examined the costs for people with (a) counts of different types of mental disorders (e.g. exactly 1, exactly 2 and so on up to 8 or more) and (b) counts of different types of somatic disorders (e.g. no somatic disorders, exactly 1, exactly 2 and so on up to 15 or more). The estimates are reported in average cost per case and nationwide annual cost in Euro 2017. Results: In total, 447,209 persons (238,659 females and 208,550 males) were diagnosed with at least one mental disorder in the study period. The average annual health care cost per case and nationwide cost was 4471 Euros and 786 million Euro, respectively, for persons with exactly one mental disorder, and 33,273 Euro and 3.6 million Euro for persons with eight or more mental disorders. The annual health care cost was 4613 Euro per case and 386 million Euro for persons without any somatic disorders, while the cost per case was 16,344 Euro and 0.7 million Euro in nationwide cost for persons with 15 or more disorders. The amount and proportion of the different health care costs varied by type of comorbidity and count of disorders. Conclusions: The annual health care cost per case was higher with increasing number of comorbid mental and somatic disorders, while the nationwide annual health care cost was lower with increasing number of comorbid disorders for persons with a mental disorder in Denmark. Simple Summary: Nitrogen treatment is a promising method for controlling stored product insects. It is based on the alternation of the oxygen level in the atmosphere (lower than 3%), which can be lethal to insects. In the present study, we tested nitrogen treatment (>99%) on a commercial scale to control major stored product insects such as Sitophilus granarius (L.), Sitophilus oryzae (L.), Rhyzopertha dominica (F.), Prostephanus truncatus (Horn), Tribolium confusum Jacquelin du Val, and Oryzaephilus surinamensis (L.). The current work indicates that nitrogen treatment is effective for all life stages and populations tested. In this study, we evaluated the insecticidal effect of nitrogen against Sitophilus granarius (L.), Sitophilus oryzae (L.), Rhyzopertha dominica (F.), Prostephanus truncatus (Horn), Tribolium confusum Jacquelin du Val, and Oryzaephilus surinamensis (L.). Four trials were conducted in chambers containing flour in bags or sacks with >99% nitrogen level. Adults of all the above species, as well as immature life stages (eggs, larvae, and pupae) of T. confusum were used in the trials. Our results showed that nitrogen caused high mortality for all species and life stages tested. Some survival was recorded for R. dominica and T. confusum pupae. Low progeny production was recorded for S. granarius, S. oryzae, and R. dominica. The study of rare events in molecular and atomic systems such as conformal changes and cluster rearrangements has been one of the most important research themes in chemical physics. Key challenges are associated with long waiting times rendering molecular simulations inefficient, high dimensionality impeding the use of PDE-based approaches, and the complexity or breadth of transition processes limiting the predictive power of asymptotic methods. Diffusion maps are promising algorithms to avoid or mitigate all these issues. We adapt the diffusion map with Mahalanobis kernel proposed by Singer and Coifman (2008) for the SDE describing molecular dynamics in collective variables in which the diffusion matrix is position-dependent and, unlike the case considered by Singer and Coifman, is not associated with a diffeomorphism. We offer an elementary proof showing that one can approximate the generator for this SDE discretized to a point cloud via the Mahalanobis diffusion map. We use it to calculate the committor functions in collective variables for two benchmark systems: alanine dipeptide, and Lennard-Jones-7 in 2D. For validating our committor results, we compare our committor functions to the finite-difference solution or by conducting a "committor analysis" as used by molecular dynamics practitioners. We contrast the outputs of the Mahalanobis diffusion map with those of the standard diffusion map with isotropic kernel and show that the former gives significantly more accurate estimates for the committors than the latter. Because of the close proximity of the cochlea, vestibular apparatus, and shared neurovascular structures, the static postural control of athletes who are deaf or hard of hearing (D/HoH) may be different from that of athletes who are hearing. Limited research is available to quantify differences between these athletes. To determine the effect of hearing status and stance condition on the static postural control of athletes. Cross-sectional study. Athletic training facilities. Fifty-five collegiate varsity athletes who were D/HoH (age = 20.62 ± 1.80 years, height = 1.73 ± 0.08 m, mass = 80.34 ± 18.92 kg) and 100 university club athletes who were hearing (age = 20.11 ± 1.59 years, height = 1.76 ± 0.09 m, mass = 77.66 ± 14.37 kg). Participants completed the Modified Clinical Test of Sensory Interaction and Balance on a triaxial force plate. Anteroposterior and mediolateral (ML) center-of-pressure (CoP) velocity, anteroposterior and ML CoP amplitude root mean square, and 95% ellipse sway area were calculated. Athletes who were D/HoH had a larger CoP velocity, larger ML root mean square, and larger sway area than those who were hearing (P values <.01). A significant main effect of stance condition was observed for all postural control variables (P values <.01). During the Modified Clinical Test of Sensory Interaction and Balance, athletes who were D/HoH demonstrated a larger sway area compared with athletes who were hearing. Etiology of vestibular schwannoma (VS) is unknown. Viruses can infect and reside in neural tissues for decades, and new viruses with unknown tumorigenic potential have been discovered. The presence of herpesvirus, polyomavirus, parvovirus, and anellovirus DNA was analyzed by quantitative PCR in 46 formalin-fixed paraffin-embedded VS samples. Five samples were analyzed by targeted next-generation sequencing. Viral DNA was detected altogether in 24/46 (52%) tumor samples, mostly representing anelloviruses (46%). Our findings show frequent persistence of anelloviruses, considered normal virome, in VS. None of the other viruses showed an extensive presence, thereby suggesting insignificant role in VS. The United States, through research ethics review, is complicit in ethically recolonizing countries into U.S. dominant understandings. This review and consent are seen as an all-encompassing ethical understanding and a discrete, one-time experience. Drawing on personal experiences and in-depth interviews with international and domestic subculture research participants and researchers, the authors argue that the United States ethics review must move toward a nuanced understanding of human ethics review that exhibits cultural responsiveness within culturally complex research, with a focus on cultural humility. The increasing demand for food, the lack of natural resources and arable land, and the recent restrictions on energy consumption require an immediate solution in terms of agricultural activities. This paper's objective was to review hydroponics (a new soilless cultivation technology) and compare it with conventional agriculture (soil cultivation) regarding its environmental impact and water and energy consumption. The soil loss, the crop/soil contamination, and the greenhouse gas emissions were the criteria for the environmental comparison of conventional agriculture and hydroponics. As for resource consumption, the water consumption rates (L/kg), energy consumption rates (kWh), and energy required (kW) were the criteria for comparing conventional agriculture with hydroponics. Tomato and cannabis cultivation were used as case studies in this review. The review results showed that the advantages of hydroponics over conventional cultivation include zero-soil cultivation, land-use efficiency, planting environment cleanliness, fertilizer and resource saving, water consumption reduction, and conservation. The disadvantages of hydroponics versus conventional cultivation were found to include the high investment costs, technical know-how requirements, and higher amount of demanded energy. Simple Summary: Side biases observed in behavior are thought to reflect underlying asymmetric brain function or hemispheric specialization. These asymmetries occur at the individual and population level, although population-level laterality normally is only evident in social species. In a previous study, we found both individual- and population-level laterality in a solitary mole rat species. Here, we assessed laterality in a eusocial mole rat species, the Damaraland mole rat, Fukomys damarensis, using turning biases. All individuals combined demonstrated left-turning biases, which was also significant at the population level. Wild-caught animals were more strongly lateralized, but lacked the population-level left-turning bias that was observed in captive mole rats. This emphasizes the importance of context and animal handling when measuring and interpreting behavioral asymmetries. Lateralization is the functional control of certain behaviors in the brain being processed by either the left or right hemisphere. Behavioral asymmetries can occur at an individual and population level, although population-level lateralization is less common amongst solitary species, whereas social species can benefit more from aligning and coordinating their activities. We assessed laterality (individual and population) through turning biases in the eusocial Damaraland mole rat, Fukomys damarensis. We considered factors such as breeding status (queen or subordinate), environment (wild-caught or captive), sex (male or female), colony and body mass. All individuals together demonstrated significant left-turning biases, which was also significant at the population level. Wild-caught animals were more strongly lateralized, had a wider spread over a laterality index and lacked the population-level left-turning bias as compared to captive mole rats. Subordinate animals were more lateralized than queens, demonstrating social status differences in turning biases for social mole rats. Voluntary participation is thought to promote the well-being of engaged individuals, especially in old age, but prior evidence on this link is mixed. In the present studies, we used the cross-sectional data from Round 6 (2012) of the European Social Survey (ESS) to investigate the variation in the associations between voluntary participation and eudaimonic (e.g., sense of direction) and social (e.g., perceived social support) well-being across types of participation (nonpolitical volunteering vs. political participation), age groups, and European countries. Study 1 addressed individual-level associations and age differences therein (preregistered at https://osf.io/2p9sz and https://osf.io/6twqe). Two-level multiple regression with an extensive set of control variables showed that at the within-country level, the associations between voluntary participation and well-being indicators were small on average. Nonpolitical volunteering had significantly more positive effects than did political participation, whereas few significant age differences emerged. Study 2 focused on the country-level variables that might explain the substantial cross-national variation in the main effects of voluntary participation (preregistered at https://osf.io/mq3dx). Only GDP per capita was a significant moderator at the country level: The associations of nonpolitical volunteering with eudaimonic well-being were more positive in the European countries with lower GDP. Other country-level variables (Gini coefficient, social welfare spending, and democracy indices) yielded no consistent moderation effects. Study 3 considered potential country-level explanations for the substantial cross-national variation in whether younger or older adults appeared to benefit more (preregistered at https://osf.io/7ks45). None of the country-level variables considered (effective retirement age in men, life expectancy at 65, average age of members of the national parliament and cabinet, and youth unemployment rate) could account for this variation. Single photon sources based on semiconductor quantum dots are one of the most prospective elements for optical quantum computing and cryptography. Such systems are often based on Bragg resonators, which provide several ways to control the emission of quantum dots. However, the fabrication of periodic structures with many thin layers is difficult. On the other hand, the coupling of single-photon sources with resonant nanoclusters made of high-index dielectric materials is known as a promising way for emission control. Our experiments and calculations show that the excitation of magnetic Mie-type resonance by linearly polarized light in a GaAs nanopillar oligomer with embedded InAs quantum dots leads to quantum emitters absorption efficiency enhancement. Moreover, the nanoresonator at the wavelength of magnetic dipole resonance also acts as a nanoantenna for a generated signal, allowing control over its radiation spatial profile. We experimentally demonstrated an order of magnitude emission enhancement and numerically reached forty times gain in comparison with unstructured film. These findings highlight the potential of quantum dots coupling with Mie-resonant oligomers collective modes for nanoscale single-photon sources development. [ABSTRACT FROM AUTHOR]

Endoplasmic reticulum (ER) stress and inflammation are hallmarks of myocardial impairment. Here, we investigated the role of the stress response protein regulated in development and DNA damage 1 (REDD1) as a molecular link between ER stress and inflammation in cardiomyocytes. In mice fed a high-fat high-sucrose (HFHS, 42% kcal fat, 34% sucrose by weight) diet for 12 wk, REDD1 expression in the heart was increased in coordination with markers of ER stress and inflammation. In human AC16 cardiomyocytes exposed to either hyperglycemic conditions or the saturated fatty acid palmitate, REDD1 expression was increased coincident with ER stress and upregulated expression of the proinflammatory cytokines IL-1Β, IL-6, and TNFɑ. In cardiomyocytes exposed to hyperglycemic/hyperlipidemic conditions, pharmacological inhibition of the ER kinase protein kinase RNA-like endoplasmic reticulum kinase (PERK) or knockdown of the transcription factor ATF4 prevented the increase in REDD1 expression. REDD1 deletion reduced proinflammatory cytokine expression in both cardiomyocytes exposed to hyperglycemic/hyperlipidemic conditions and in the hearts of obese mice. In the present study, we examined the insecticidal effect of Carifend® (BASF AG, Ludwigshafen, Germany; 163.2 mg m−2), an alpha-cypermethrin-coated polyester net, as well as Carifend-like nets containing different rates of alpha-cypermethrin, specifically 10, 30, 80, and 325 mg m−2, against two major stored tobacco insect pests, i.e., Lasioderma serricorne (Fabricius) (Coleoptera: Anobiidae) and Ephestia elutella (Hübner) (Lepidoptera: Pyralidae). The nets were glued at the bottom of plastic Petri dishes, and adults of L. serricorne and E. elutella were exposed to the nets for different exposure times (e.g., 1, 5, 15, 30, 60, 90, and 120 min, as well as 12 and 24 h). After each exposure interval, insect mortality was recorded, whereas after the final evaluation, alive individuals were transferred to untreated dishes, and delayed mortality was recorded after an additional period of 1, 3, 5, and 7 d. Based on our results, efficient control of both insect species was achieved with all nets, even the one containing a lower concentration of alpha-cypermethrin (10 mg m−2). High mortality rates were recorded for all treatments, indicating that even short exposure of adults of both tested species to the alpha-cypermethrin-coated nets tested can lead to mortality. For instance, at 30 mg m−2, mortality of E. elutella reached 40% 1 d after exposure for 30 min. Moreover, at the highest concentration (325 mg m−2), mortality reached 70% and 99% 7 d post exposure for 24 h of L. serricorne and E. elutella, respectively. Based on these data, Carifend®, as well as the rest of the Carifend-like nets tested, can be used as an alternative insecticide method for the control of two major stored tobacco insect species, i.e., L. serricorne and E. elutella. To measure the potential clinical impact of neoadjuvant treatment response, change in thyroid neck surgical morbidity score was assessed using CT neck with contrast (1.25 mm cuts) at baseline and preoperatively at the completion of neoadjuvant therapy. The first patient was treated with the study drug LOXO-292 (provided by Loxo Oncology) after an investigational drug application approval by the FDA, which has been previously published.[3] The remaining three patients were treated outside of a clinical trial. Single-task assessments may not identify lingering effects following a concussion that may be detected under dual-task (DT) paradigms. The purpose of this study was to determine the effects of a novel DT paradigm and concussion history on gait and cognitive performance. Hockey and rugby club college athletes (n = 26) completed a box drill and the color and word Stroop test under single task and DT. Distance ambulated around the box, response rate, and accuracy were recorded to calculate dual-task cost. Mean comparisons and linear mixed-effects regression models were performed. Compared to athletes with no concussion history, those with a history had a greater motor than cognitive dual-task cost and were 3.15% less accurate in Stroop responses (p < .01). Athletes walked 0.72-m shorter distance under DT compared to single task (p = .04). A multidirectional, low-tech DT assessment may highlight long-term motor and cognitive deficits among athletes with a concussion history, which will provide valuable information to prepare and track performance within an athletic season. We found two functional populations of hemocytes in the hemolymph of the pulmonate snail Planorbarius corneus: hyalinocytes and granulocytes. The hyalinocytes were much more numerous than granulocytes. Each population was subdivided into subpopulations based on morphological and functional characteristics. The hyalinocyte population was represented by two subpopulations differing in size, granularity, and ability to form pseudopodia. In snails infected with the trematodes Plagiorchis sp., the cellular composition of the hemolymph changes, with hemocytes being mostly represented by granulocytes. This phenomenon is associated with the fact that granulocytes form a hemocytic paletot on the surface of sporocysts of Plagiorchis sp. It is suggested that pulmonate molluscs have one lineage of hemolymph cells. Hematopoietic stem cells can divide, ensuring multiplication of prohemocytes. Mutations in the Ca V 1.2 L-type calcium channel can cause a profound form of long-QT syndrome known as long-QT type 8 (LQT8), which results in cardiac arrhythmias that are often fatal in early childhood. A growing number of such pathogenic mutations in Ca V 1.2 have been identified, increasing the need for targeted therapies. As many of these mutations reduce channel inactivation; resulting in excess Ca2+ entry during the action potential, calcium channel blockers (CCBs) would seem to represent a promising treatment option. Yet CCBs have been unsuccessful in the treatment of LQT8. Here, we demonstrate that this lack of efficacy likely stems from the impact of the mutations on Ca V 1.2 channel inactivation. As CCBs are known to preferentially bind to the inactivated state of the channel, mutation-dependent deficits in inactivation result in a decrease in use-dependent block of the mutant channel. Further, application of the CCB verapamil to induced pluripotent stem cell (iPSC) derived cardiomyocytes from an LQT8 patient demonstrates that this loss of use-dependent block translates to a lack of efficacy in correcting the LQT phenotype. As a growing number of channelopathic mutations demonstrate effects on channel inactivation, reliance on state-dependent blockers may leave a growing population of patients without a viable treatment option. This biophysical understanding of the interplay between inactivation deficits and state-dependent block may provide a new avenue to guide the development of improved therapies. The Salmon gill poxvirus (SGPV) has emerged in recent years as the cause of an acute respiratory disease that can lead to high mortality in farmed Atlantic salmon presmolts, known as Salmon gill poxvirus disease. SGPV was first identified in Norway in the 1990s, and its large DNA genome, consisting of over 206 predicted protein-coding genes, was characterized in 2015. This review summarizes current knowledge relating to disease manifestation and its effects on the host immune system and describes dissemination of the virus. It also demonstrates how newly established molecular tools can help us to understand SGPV and its pathogenesis. Antibody-dependent enhancement (ADE) has been associated with severe disease outcomes in several viral infections, including respiratory infections. In vitro and in vivo studies showed that antibody-response to SARS-CoV and MERS-CoV could exacerbate infection via ADE. Recently in SARS CoV-2, the in vitro studies and structural analysis shows a risk of disease severity via ADE. This phenomenon is partially attributed to non-neutralizing antibodies or antibodies at sub-neutralizing levels. These antibodies result in antigen-antibody complexes' deposition and propagation of a chronic inflammatory process that destroys affected tissues. Further, antigen-antibody complexes may enhance the internalization of the virus into cells through the Fc gamma receptor (FcγR) and lead to further virus replication. Thus, ADE occur via two mechanisms; 1. Antibody mediated replication and 2. Enhanced immune activation. Antibody-mediated effector functions are mainly driven by complement activation, and the first complement in the cascade is complement 1q (C1q) which binds to the virus-antibody complex. Reports say that deficiency in circulating plasma levels of C1q, an independent predictor of mortality in high-risk patients, including diabetes, is associated with severe viral infections. Complement mediated ADE is reported in several viral infections such as dengue, West Nile virus, measles, RSV, Human immunodeficiency virus (HIV), and Ebola virus. This review discusses ADE in viral infections and the in vitro evidence of ADE in coronaviruses. In the present study we examined the effect of the presence of wheat bugs (Eurygaster spp. and Aelia spp.) on wheat on the mortality, progeny production and infestation patterns of three stored product insects, Trogoderma granarium Everts, Sitophilus oryzae (L.) and Rhyzopertha dominica (F.). The bioassays were carried out under laboratory conditions (25 ± 1 °C, 60 ± 5% relative humidity (r.h.) and continuous darkness), and 20 adults of each stored product insect species were placed in plastic vials (3 cm in diameter, 8 cm in height) with wheat that contained different numbers of wheat bugs (1, 2, 3 or 5 adults). In general, stored product insect mortality was not affected by the presence of wheat bugs, in most of the combinations examined. In most of the cases, mortality of T. granarium was higher on wheat which contained Eurygaster that wheat containing Aelia, while progeny production of S. oryzae was also affected by the presence of wheat bugs. Frass production for all combinations was similar, but greater frass production occurred by all combinations of R. dominica with Aelia. Overall, the presence of wheat bugs could affect the survival and the progeny production of both S. oryzae and R. dominica. In general, our results showed that beetle longevity and fecundity can be affected by the presence of wheat bugs that are likely to be present in stored grains after harvest. [ABSTRACT FROM AUTHOR]

Drawing on the theory of planned behavior (TPB) and the volunteer process model (VPM), we compared proximal (the TPB variables) and distal (general social support and sense of community) antecedents and quality of experience between civic (CP) and political (PP) participation. Outcome variables were future intentions. We used data from a mail survey of 3,231 adults with or without prior CP or PP experience. Both inexperienced and experienced individuals perceived PP more negatively than CP. Quality of experience was also rated substantially more negatively for PP than for CP. Distal antecedents had similar effects for CP and PP, but they were mostly significant for CP only. In line with the VPM, quality of experience was the strongest predictor of future intentions in experienced individuals. Perceived behavioral control was the strongest mediator. Findings corroborate the distinction between CP and PP and suggest that PP is much less attractive on average. [ABSTRACT FROM AUTHOR]
