Simple Summary: Underwater mountains, or seamounts, are deep-sea habitats collectively forming an area as large as Europe. Yet, they are one of the least studied ecosystems on earth. Known for supporting rich marine life compared to surrounding deep-sea environments, we have no information on how seamounts truly compare to other iconic biodiversity hotspots like shallow coral reefs. To assess the effective ecological value of seamounts, we compared fish communities in coral reefs and seamounts up to 500 m deep using two techniques: environmental DNA to detect the presence of species by filtering fragments of DNA lost by organisms in seawater, and underwater cameras to directly measure fish abundance and size. We found that the deepest seamounts had almost 10 times fewer fish species than coral reefs. However, the shallowest seamounts had larger fish species, including sharks, than coral reefs. We conclude that while seamounts are important and unique ecosystems, they may not be as diverse for fish species as previously thought (diversity hotspots) but rather biomass oases and refuges for endangered species. This study therefore calls for protecting the shallowest seamounts, as they are critical areas for marine life. Seamounts are the least known ocean biome. Considered biodiversity hotspots, biomass oases, and refuges for megafauna, large gaps exist in their real diversity relative to other ecosystems like coral reefs. Using environmental DNA metabarcoding (eDNA) and baited video (BRUVS), we compared fish assemblages across five environments of different depths: coral reefs (15 m), shallow seamounts (50 m), continental slopes (150 m), intermediate seamounts (250 m), and deep seamounts (500 m). We modeled assemblages using 12 environmental variables and found depth to be the main driver of fish diversity and biomass, although other variables like human accessibility were important. Boosted Regression Trees (BRT) revealed a strong negative effect of depth on species richness, segregating coral reefs from deep-sea environments. Surprisingly, BRT showed a hump-shaped effect of depth on fish biomass, with significantly lower biomass on coral reefs than in shallowest deep-sea environments. Biomass of large predators like sharks was three times higher on shallow seamounts (50 m) than on coral reefs. The five studied environments showed quite distinct assemblages. However, species shared between coral reefs and deeper-sea environments were dominated by highly mobile large predators. Our results suggest that seamounts are no diversity hotspots for fish. However, we show that shallower seamounts form biomass oases and refuges for threatened megafauna, suggesting that priority should be given to their protection. [ABSTRACT FROM AUTHOR]

Motivation: Host to intricate networks of marine species, coral reefs are among the most biologically diverse ecosystems on Earth. Over the past few decades, major degradations of coral reefs have been observed worldwide, which is largely attributed to the effects of climate change and local stressors related to human activities. Now more than ever, characterizing how the environment shapes the dynamics of the reef ecosystem (e.g., shifts in species abundance, community changes, emergence of locally adapted populations) is key to uncovering the environmental drivers of reef degradation, and developing efficient conservation strategies in response. To achieve these objectives, it is pivotal that environmental data describing the processes driving such ecosystem dynamics, which occur across specific spatial and temporal scales, are easily accessible to coral reef researchers and conservation stakeholders alike. Main types of variable contained: Multiple environmental variables characterizing various facets of the reef environment, including water chemistry and physics (e.g., temperature, pH, chlorophyll concentration), local anthropogenic pressures (e.g., boat traffic, distance from agricultural or urban areas) and sea currents patterns. Anomalous heat waves are causing a major decline of hard corals around the world and threatening the persistence of coral reefs. There are, however, reefs that have been exposed to recurrent thermal stress over the years and whose corals appear to have been tolerant against heat. One of the mechanisms that could explain this phenomenon is local adaptation, but the underlying molecular mechanisms are poorly known. In this work, we applied a seascape genomics approach to study heat stress adaptation in three coral species of New Caledonia (southwestern Pacific) and to uncover the molecular actors potentially involved. We used remote sensing data to characterize the environmental trends across the reef system, and sampled corals living at the most contrasted sites. These samples underwent next generation sequencing to reveal single nucleotide polymorphisms (SNPs), frequencies of which were associated with heat stress gradients. As these SNPs might underpin an adaptive role, we characterized the functional roles of the genes located in their genomic region. In each of the studied species, we found heat stress‐associated SNPs located in proximity of genes involved in pathways well known to contribute to the cellular responses against heat, such as protein folding, oxidative stress homeostasis, inflammatory and apoptotic pathways, and DNA damage‐repair. In some cases, the same candidate molecular targets of heat stress adaptation recurred among species. Together, these results underline the relevance and the power of the seascape genomics approach for the discovery of adaptive traits that could allow corals to persist across wider thermal ranges. Marine ecosystems are both stressed and threatened by pesticides that are used on land. Nevertheless, research on the impact of pesticides on coral reefs and the underlying mechanisms is still in its infancy. The insecticide chlordecone is a persistent organic pollutant with carcinogenic effects in rats and mice. Chlordecone has been detected in diverse marine organisms in the Caribbean, but unexpectedly, also in French Polynesia. We combined transcriptomic and morphologic analyses of analyses the response of the coral Pocillopora damicornis to chlordecone stress. We compared chlordecone stress with thermal stress to determine a chlordecone-specific response. We found eight transcripts related to the P450-1A or P450-3A families that were specifically overexpressed in response to chlordecone. There was also sequential overexpression of transcripts involved in apoptosis and degradation of cellular matrix proteins. Finally, we report the first observation of chlordecone-induced P. damicornis polyp bail-out. Altogether, these results strongly suggest that apoptosis and expression of genes belonging to the cathepsin family are sequentially regulated processes leading to coenosarc dissociation and loss. Coral disease outbreaks have increased over the last three decades, but their causal agents remain mostly unclear (e.g., bacteria, viruses, fungi, protists). This study details a 14-month-long survey of coral colonies in which observations of the development of disease was observed in nearly half of the sampled colonies. A bimonthly qPCR method was used to quantitatively and qualitatively evaluate Symbiodinium assemblages of tagged colonies, and to detect the presence of Vibrio spp. Firstly, our data showed that predisposition to disease development in general, and, more specifically, infection by Vibrio spp. in Acropora cytherea depended on which clades of Symbiodinium were harbored. In both cases, harboring clade D rather than A was beneficial to the coral host. Secondly, the detection of Vibrio spp. in only colonies that developed disease strongly suggests opportunistic traits of the bacteria. Finally, even if sporadic cases of switching and probably shuffling were observed, this long-term survey does not suggest specific-clade recruitment in response to stressors. Altogether, our results demonstrate that the fitness of the coral holobiont depends on its initial consortium of Symbiodinium, which is distinct among colonies, rather than a temporary adaptation achieved through acquiring different Symbiodinium clades. As anomalous heat waves are causing the widespread decline of coral reefs worldwide, there is an urgent need to identify coral populations tolerant to thermal stress. Heat stress adaptive potential is the degree of tolerance expected from evolutionary processes and, for a given reef, depends on the arrival of propagules from reefs exposed to recurrent thermal stress. For this reason, assessing spatial patterns of thermal adaptation and reef connectivity is of paramount importance to inform conservation strategies. In this work, we applied a seascape genomics framework to characterize the spatial patterns of thermal adaptation and connectivity for coral reefs of New Caledonia (Southern Pacific). In this approach, remote sensing of seascape conditions was combined with genomic data from three coral species. For every reef of the region, we computed a probability of heat stress adaptation, and two indices forecasting inbound and outbound connectivity. We then compared our indicators to field survey data, and observed that decrease of coral cover after heat stress was lower at reefs predicted with high probability of adaptation and inbound connectivity. Accelerating rate of human impact and environmental change severely affects marine biodiversity and increases the urgency to implement the Convention on Biological Diversity (CBD) 30×30 plan for conserving 30% of sea areas by 2030. However, area‐based conservation targets are complex to identify in a 3‐dimensional (3D) ocean where deep‐sea features such as seamounts have been seldom studied mostly due to challenging methodologies to implement at great depths. Yet, the use of emerging technologies, such as environmental DNA combined with modern modeling frameworks, could help address the problem. We collected environmental DNA, echosounder acoustic, and video data at 15 seamounts and deep island slopes across the Coral Sea. We modeled 7 fish community metrics and the abundances of 45 individual species and molecular operational taxonomic units (MOTUs) in benthic and pelagic waters (down to 600‐m deep) with boosted regression trees and generalized joint attribute models to describe biodiversity on seamounts and deep slopes and identify 3D protection solutions for achieving the CBD area target in New Caledonia (1.4 million km2). We prioritized the identified conservation units in a 3D space, based on various biodiversity targets, to meet the goal of protecting at least 30% of the spatial domain, with a focus on areas with high biodiversity. The relationship between biodiversity protection targets and the spatial area protected by the solution was linear. The scenario protecting 30% of each biodiversity metric preserved almost 30% of the considered spatial domain and accounted for the 3D distribution of biodiversity. Our study paves the way for the use of combined data collection methodologies to improve biodiversity estimates in 3D structured marine environments for the selection of conservation areas and for the use of biodiversity targets to achieve area‐based international targets. Adaptation of reef-building corals to global warming depends upon standing heritable variation in tolerance traits upon which selection can act. Yet limited knowledge exists on heat-tolerance variation among conspecific individuals separated by metres to hundreds of kilometres. Here, we performed standardized acute heat-stress assays to quantify the thermal tolerance traits of 709 colonies of Acropora spathulata from 13 reefs spanning 1060 km (9.5° latitude) of the Great Barrier Reef. Thermal thresholds for photochemical efficiency and chlorophyll retention varied considerably among individual colonies both among reefs (approximately 6°C) and within reefs (approximately 3°C). Although tolerance rankings of colonies varied between traits, the most heat-tolerant corals (i.e. top 25% of each trait) were found at virtually all reefs, indicating widespread phenotypic variation. Reef-scale environmental predictors explained 12–62% of trait variation. Corals exposed to high thermal averages and recent thermal stress exhibited the greatest photochemical performance, probably reflecting local adaptation and stress pre-acclimatization, and the lowest chlorophyll retention suggesting stress pre-sensitization. Importantly, heat tolerance relative to local summer temperatures was the greatest on higher latitude reefs suggestive of higher adaptive potential. Coral recruitment is a vital process for the maintenance and recovery of coral reefs, particularly due to their decline from global change. While it is well established that larval settlement cues significantly influence coral recruitment, the investigation of recruitment success associated with the surrounding community mainly focuses on algae. To investigate other factors controlling this process, we examined the impact of benthic invertebrate assemblages on coral recruitment using artificial reefs. Three types of artificial reefs (mono‐, bi‐, and tri‐species) with different assemblages of three common species, Pocillopora acuta, Acropora cytherea, and Tridacna maxima, were studied over 8 months. This study revealed that benthic assemblages play a significant role in coral recruitment and survival. High biodiversity was found to enhance coral recruitment and inhibit potential negative cues from A. cytherea. Our findings underscore the importance of preserving high biodiversity using not only hard coral but a wide range of phyla, including bivalves, in coral restoration efforts. Background: Cosmetic plants and their uses have often been neglected in ethnobotanical surveys which focus mainly on plants with medicinal or food uses. Thus, this survey was carried out to specifically investigate cosmetics in a small community and to establish a cosmetopoeia, based on the model of pharmacopoeia for medicinal plants. The geographic spread of the survey covered the Marquesas Islands, one of the five archipelagos of French Polynesia (Pacific Ocean). This archipelago was also recently investigated for its pharmacopoeia. Methods: This survey is based on individual interviews of Marquesan informants on the islands of Tahiti (Society archipelago) and Nuku Hiva (Marquesas archipelago). The methodological approach was semi-directive with open-ended questions based on cosmetic criteria (application area, cosmetic use, plant). Before each interview, researchers and the informant signed a Prior Informed Consent (PIC). Quantitative analyses were performed using basic statistics and the indice of Fidelity Level (FL). Results: Twenty-eight informants from five of the six inhabited Marquesan islands were interviewed and yielded more than 500 cosmetic recipes. Marquesan cosmetopoeia included 79 plant taxa, of which 5% are Marquesan endemics, 23% are indigenous, 28% are Polynesian introductions and 44% are modern introductions. Among the introduced species, half were cultivated whereas the other half were weedy species. Most of the plants were abundant and only eight species were considered rare, of which four were Marquesan endemics. Main cosmetic plants were identified through informant citations and fidelity levels, and included Calophyllum inophyllum, Cananga odorata, Citrus aurantiifolia, Cocos nucifera, Curcuma longa, Gardenia taitensis, Mentha spp., Ocimum basilicum, Rauvolfia nukuhivensis and Santalum insulare var. marchionense. The most referred application areas were skin, hair and private parts whereas the main cosmetic uses were perfume, hydration, medicinal care and healing. Conclusions: Through this survey, Marquesan cosmetopoeia was investigated in detail and uncovered a majority of introduced and abundant plants, and a minority of endemic and rare plants which required proper management to avoid future shortage. The well known perfumed coconut oil or monoi appeared as the main Marquesan cosmetic preparation either for the skin and the hair. When fish larvae recruit back to a reef, chemical cues are often used to find suitable habitat or to find juvenile or adult conspecifics. We tested if the chemical information used by larvae was intentionally produced by juvenile and adult conspecifics already on the reef (communication process) or whether the cues used result from normal biochemical processes with no active involvement by conspecifics (“spying” behavior by larvae). Conspecific chemical cues attracted the majority of larvae (four out of the seven species tested); although while some species were equally attracted to cues from adults and juveniles ( Chromis viridis , Apogon novemfasciatus ), two exhibited greater sensitivity to adult cues ( Pomacentrus pavo , Dascyllus aruanus ). Our results indicate also that spying cues are those most commonly used by settling fishes ( C. viridis , P. pavo , A. novemfasciatus ). Elasmobranchs are critical to tourism industries worldwide. In French Polynesia, where ecotourism is the second most important industry, little is known about the spatiotemporal distribution of eagle rays. This study represents the first investigation into habitat use and behaviour of ocellated eagle rays (Aetobatus ocellatus) in French Polynesia focused on two sites with different levels of anthropogenic noise. Environmental variables and biological data were recorded over a one-year period to explore the distribution patterns of 113 eagle rays identified at these sites. Results revealed distinct patterns in habitat use between the two sites, with the eagle ray population structured according to ontogenetic stage. Young rays preferred the site with lower levels of noise pollution, where they foraged at the end of the day. Adult eagle rays gathered in the mornings at the louder site, where the only noticeable activity was group swimming. Overall, our results could help conservation initiatives to manage this important species in French Polynesia, and potentially other coral reef areas, especially in the context of rising human impacts on the environment. Calophyllum inophyllum L. (Calophyllaceae), locally called "tamanu" in French Polynesia, is an evergreen pantropical tree growing mostly along the seashores. Its barks, leaves, and fruits are still used in traditional medicine. The oil expressed from the nuts has been also traditionally used. Tamanu oil is topically applied on skins as well as mucous membrane lesions. This oil is especially recommended to heal all kinds of skin ailments. Bioassays and different assessments of Tamanu oil revealed numerous biological activities (antioxidant, anti-inflammatory, antibacterial, wound healing...), so bringing scientific evidence of beneficial effects of this oil on human skin healing. Such biological properties may explain the use of tamanu oil as an active cosmetic ingredient recorded as "Calophyllum inophyllum seed oil" by the INCI (International Nomenclature of Cosmetic Ingredients). Most of the bioactive properties of tamanu oil are attributed to oil composition including the presence of resinous compounds in tamanu oil beside common fatty acids, which constitutes a unique characteristic of this healing oil. Actually, resinous part of tamanu oil is known to contain bioactive secondary metabolites mostly constituted by neoflavonoids including pyranocoumarin derivatives. Symbioses with the dinoflagellate Symbiodinium are widespread among marine invertebrates and protists, especially in nutritionally demanding habitats, such as tropical coral reefs, where they play a major role in ecosystem survival. Moreover, apart from corals and sea anemones, many of the Symbiodinium species and clades involved in these partnerships remain to be characterized. This study provides new insights into nudibranch and sponge associations with Symbiodinium by sequencing regions of the Symbiodinium 28S rDNA and the host mitochondrial COI oxidase. Specimens were sampled between 2011 and 2013 from locations around the islands of Moorea and Tahiti, French Polynesia. Our results revealed that some of the sponges and nudibranchs harbored typical Symbiodinium from clade B or C while others harbored new, undescribed Symbiodinium -like dinoflagellates. A detailed analysis of the different life stages of the nudibranch Phestilla lugubris and of its specific coral prey, Porites rus , suggests a prey-predator horizontal transfer of the symbiont and its vertical inheritance from the parent to the eggs. In addition to the already reported nukuhivensiums 1 and 2 , 11 indole alkaloids were isolated from the bark of the plant Rauvolfia nukuhivensis , growing in the Marquesas archipelago. The known sandwicine ( 3 ), isosandwicine ( 4 ), spegatrine ( 8 ), lochneram ( 9 ), flavopereirine ( 13 ) have been found in this plant together with the norsandwicine ( 5 ), isonorsandwicine ( 6 ), N b -methylisosandwicine ( 7 ), 10-methoxypanarine ( 10 ), nortueiaoine ( 11 ), tueiaoine ( 12 ). The structure elucidation was performed on the basis of a deep exploration of the NMR and HRESIMS data as well as comparison with literature data for similar compounds. Norsandwicine, 10-methoxypanarine, tueiaoine, and more importantly nukuhivensiums, were shown to significantly induce a reduction of IKr amplitude (HERG current). Abstract: The study investigated visual recognition of conspecifics and predators by settlement-stage coral reef fish larvae in a set of three experiments using a dual-choice aquarium (Moorea Island). Experiments 1 and 2 were conducted under artificial light conditions. Experiment 3 was conducted under natural light during new and full moon nights. In experiment 1, five out of six species preferred conspecifics rather than heterospecifics (Acanthurus triostegus, Chromis viridis, Ostorhinchus angustatus, Stegastes fasciolatus, Valenciaenna strigata). In experiment 2, three out of six species were repulsed by predators (Mulloidichtys flavolineatus, O. angustatus, V. strigata). In experiment 3 (conducted on one species), A. triostegus was attracted to conspecifics during bright nights, but did not show such behavior during dark nights. Our study raises the question of trade-off for fish larvae to settle during the night with high light intensities to favor the visual recognition of conspecifics and predators, or during darker nights to reduce reef predation. Abstract: Microsatellite length polymorphism (MLP) typing is a PCR-based method used for genotyping of the diploid yeast Candida albicans. However, MLP is subject to homoplasia which can hamper the accuracy of the results. We combined fragment length analysis, high-resolution DNA melting (HRM) analysis, and SNaPshot minisequencing after a single amplification of the CDC3 locus to study 95 epidemiologically independent C. albicans isolates. HRM analysis for a given electrophoretic group led to a maximum of three different curves due to the presence of a SNP upstream of the tandem repeat which could be characterized using the SNaPshot assay. The combination of the three methods had a discriminatory index of 0.88 in complete congruence with previous MLP typing (Mantel test R =0.99, P <10− 4). Psoriasis is a chronic inflammatory skin disease that is mediated by complex crosstalk between immune cells and keratinocytes (KCs). Emerging studies have showed a specific psoriatic microRNAs signature, in which miR-21 is one of the most upregulated and dynamic miRNAs. In this study, we focused our investigations on the passenger miR-21-3p strand, which is poorly studied in skin and in psoriasis pathogenesis. Here, we showed the upregulation of miR-21-3p in an IMQ-induced psoriasiform mouse model. This upregulation was correlated with IL-22 expression and functionality, both in vitro and in vivo, and it occurred via STAT3 and NF-κB signaling. We identified a network of differentially expressed genes involved in abnormal proliferation control and immune regulatory genes implicated in the molecular pathogenesis of psoriasis in response to miR-21-3p overexpression in KCs. These results were confirmed by functional assays that validated the proliferative potential of miR-21-3p. All these findings highlight the importance of miR-21-3p, an underestimated miRNA, in psoriasis and provide novel molecular targets for therapeutic purposes. [ABSTRACT FROM AUTHOR]
