Atavisms have attracted people's attention for a long time. First, atavisms excited their imagination and created fertile ground for myths and superstitions. With the development of science, atavisms became the subject of investigation, which soon provided evidence to support evolutionary theory. However, at the molecular level, the formation of atavisms remained insufficiently understood. Recent progress in comparative genomics and molecular developmental biology has helped in understanding the processes underlying the formation of one of the human atavisms: the vestigial tail. Neurulation is traditionally defined as the process of closure of the neural tube. New data have shown that the major driving forces of neurulation continue to operate with the closure of the neural tube, at least until the central canal of the neural tube has formed. Owing to this, the paper proposes to distinguish two periods of neurulation. According to these notions, early neurulation corresponds to the period of closure of the neural tube, and late neurulation corresponds to the period of formation of the central canal. The book "Objects of Developmental Biology" provides detailed description of the developmental stages of loach, which is widely used as a model for research in developmental biology, biochemistry, and cryobiology.

The database “Objects for Storage and Disposal of Radioactive Wastes from Nuclear Fuel Cycle Enterprises” has been developed. Criteria were chosen for assessment of the hazard of storage and disposal (SD) objects for staff and population, and parameters based on these criteria were suggested: local hazard parameter and geospheric hazard parameter. To calculate the latter parameter, it is recommended to use simple models of radionuclide egress into the geosphere. The database can be used as an effective tool for ranging of SD objects of nuclear fuel cycle (NFC) enterprises in the order of decreasing hazard parameters. The use of a procedure for ranging of SD objects, tested with one of NFC enterprises as example, makes it possible to reveal the most hazardous SD objects at NFC enterprises and to decide on priority in decommissioning. Myogenesis is a fundamental process governing the formation of muscle in multicellular organisms. Recent studies in zebrafish Danio rerio have described the molecular events occurring during embryonic morphogenesis and have thus greatly clarified this process, helping to distinguish between the events that give rise to fast v. slow muscle. Coupled with the well-known Hedgehog signalling cascade and a wide variety of cellular processes during early development, the continual research on D. rerio slow muscle precursors has provided novel insights into their cellular behaviours in this organism. Similarly, analyses on fast muscle precursors have provided knowledge of the behaviour of a sub-set of epitheloid cells residing in the anterior domain of somites. Additionally, the findings by various groups on the roles of several molecules in somitic myogenesis have been clarified in the past year. RhoA small GTPase, as a key regulator for actin cytoskeletal rearrangement, plays pivotal roles during morphogenesis, cytokinesis, phagocytosis and cell migration, but little is known about its signaling mechanism that controls cell survival in vivo. Using zebrafish as a model, we show that non-overlapping antisense morpholinos that block either translation or splicing of rhoA lead to extensive apoptosis during embryogenesis, resulting in overall reduction of body size and body length. These defects are associated with reduced activation of growth-promoting Erk and decreased expression of anti-apoptotic bcl-2. Moreover, ectopic expression of rhoA, Mek or BCL-2 mRNA rescues such phenotypes. Consistently, combined suppression of RhoA and Mek/Erk or Bcl-2 pathways by sub-optimal dose of rhoA morpholino and pharmacological inhibitors for either Mek (U0126) or Bcl-2 (HA 14-1) can induce developmental abnormalities and enhanced apoptosis, similar to those caused by effective RhoA knockdown. Furthermore, U0126 abrogates the rescue by RhoA and MEK but not BCL-2. In contrast, HA 14-1 effectively abolishes all functional rescues by RhoA, MEK or BCL-2, supporting that RhoA prevents apoptosis by activation of Mek/Erk pathway and requiring Bcl-2. The GFP gene controlled by a mini-promoter of the keratin-8 gene was cloned into a modified nonautonomous transposon Tol2 from the medaka genome. Such transposon cannot itself transpose, but external transposase allows it to integrate into the germ cell genome, where it remains after transposase is depleted. This gives rise to a transgenic zebrafish line stably maintained for many generations. This approach was used to obtain several tens of transgenic zebrafish lines, each of which demonstrates stably inherited pattern of GFP expression. This pattern depends on the enhancer activity in the vicinity of the transposon integration site. These lines can be used for the analysis of DNA sequences of enhancers, in vivo studies of complex cell morphogenesis, description of organs whose development has not been described in zebrafish, and analysis of activity of genes controlled by the identified promoters during development. Current data on the transgenic lines were integrated into the dedicated ZETRAP database. At the second stage of this project, the transposon was mobilized by a short-term exposure of two transgenic lines to transposase and left the initial chromosomal location. This allowed us to isolate about 20 transgenic lines with new expression patterns. This approach can be successfully used to study the zebrafish biology and anatomy, activity of the regulatory part of the genome, and development of new promising techniques in developmental biology and, in the foreseeable future, gene therapy. The article recounts the introduction of embryologist Boris Balinsky, author of the book "An Introduction to Embryology," to embryology. Balinsky, an ex-professor of embryology of the Kiev University, got interested in the science of embryology upon reading Sergei Aksakov's story about collecting butterflies. In 1992, he carried his first research project following the experimental scheme set by embryologist Dmitrii Filatov. During and after the writing of the book, Balinsky was amidst social political changes in Russia. The book was finished and published outside Russia, hence Balinksy and his work, were never highly regarded in his country.

The surface instability of laminated composites of regular structure is analyzed in a three-dimensional linearized formulation. Hearing loss is a common sensory impairment with a heterogeneous genetic etiology. Genetic variants in the TBC1D24 gene have recently emerged as an important cause of the non-syndromic autosomal dominant hearing loss (ADHL). However, the molecular mechanism behind the TBC1D24 -associated ADHL is unknown. Using a zebrafish model, we investigated involvement of TBC1D24 in hearing and the functional effects of the associated ADHL-causing genetic variants. We show that the morpholino-mediated knock-down of Tbc1d24 resulted in defective ear kinocilia structure and reduced locomotor activity of the embryos. The observed phenotypes were rescued by a wild-type TBC1D24 mRNA but not by a mutant mRNA carrying the ADHL-causing variant c.553G>A (p.Asp185Asn), supporting its pathogenic potential. CRISPR-Cas9-mediated knock-out of tbc1d24 led to mechanosensory deficiency of lateral line neuromasts. Overexpression of TBC1D24 mRNA resulted in developmental abnormalities associated with ciliary dysfunction and mesendodermal mispatterning. We observed that the ADHL-causing TBC1D24 variants: c.553G>A (p.Asp185Asn); c.1460A>T (p.His487Leu), c.1461C>G (p.His487Gln) or a novel variant c.905T>G (p.Leu302Arg) alleviated the effect of overexpression, indicating that these variants disrupt the TBC1D24 function. Furthermore, the zebrafish phenotypes correspond to the severity of ADHL. Specific changes in ear structures upon TBC1D24 overexpression further highlighted its tissue-specific role in ciliary function and inner ear development. Our findings provide functional evidence for the pathogenic potential of the ADHL-causing TBC1D24 variants and lead to new insights into the function of TBC1D24 in cilia morphogenesis. Congenital hearing loss can be caused by genetic abnormalities leading to inner ear malformations (IEM). SLC26A4 (pendrin) is an anion exchanger expressed in the inner ear, thyroid and β-intercalated cells in the kidney. Mutations in SLC26A4 cause a common form of IEMS: enlarged vestibular aqueduct (EVA), often accompanied by incomplete partition type 2 (EVA/IP2). However, only 25% of patients have confirmed mutations in SLC26A4. It has been reported that a group of polymorphisms upstream of the SLC26A4 gene, the CEVA haplotype, is frequently found in patients with monoallelic SLC26A4 mutations. Since the publication of the zebrafish genome in 2001, the zebrafish has become an increasingly popular animal model for studying human diseases. The transparency of the larvae, the large number of offspring and the accessibility of various methods of genetic, chemical and physical manipulation make it a very useful model for studying inner ear development. During early development, the inner ear undergoes dynamic changes from an otic placode and otic vesicle to a labyrinth of semicircular canals. Since data on the expression of genes associated with hearing are incomplete, we present here a detailed account of expression during early development. Secondly, we plan to create a zebrafish model for human SLC26A4 gene expression by introducing human upstream regulatory elements into the zebrafish genome. [ABSTRACT FROM AUTHOR]

TBC1D24 genetic variants are causally involved in the development of both autosomal recessive hearing loss and epilepsy syndromes, and autosomal dominant hearing loss (ADHL). So far, our group published four novel ADHL-causative TBC1D24 probably pathogenic variants by performing high-throughput genetic testing in families with ADHL, and more variants are yet to be revealed. In the light of current discoveries, variants in TBC1D24 emerge as a more significant cause of ADHL. The molecular mechanism behind the TBC1D24-associated ADHL is unknown. Using a zebrafish model, we investigated involvement of TBC1D24 in hearing and the functional effects of the associated ADHL-causing genetic variants. Different methodological approaches were used in the study, including (i) expression studies by whole mount in situ hybridization (WISH), qPCR on different developmental stages and cryosections, (ii) assessment of the zebrafish ear and neuromast hair cell morphology by high-resolution imaging and (iii) behavioral studies in a developed tbc1d24-deficient zebrafish models (by knock-down or knock-out of tbc1d24) and in overexpression and rescue tbc1d24 models. Mixed lineage leukemia 5 (MLL5) has been implicated in multiple aspects of cell physiology, such as hematopoiesis, cell cycle control and chromatin regulatory network. In this study, we present evidence that MLL5 is involved in the camptothecin (CPT)-induced p53 activation. CPT promoted the degradation of MLL5 protein in a time- and dose-dependent manner in actively replicating cells. The downregulation of MLL5 led to phosphorylation of p53 at Ser392, which was abrogated by exogenous overexpression of MLL5. In MLL5-knockdown cells, p53 protein was stabilized and bound to DNA with higher affinity, leading to activation of downstream genes. Co-immunoprecipitation showed that MLL5 preferentially interacted with the tetramerized form of p53, and knockdown of MLL5 promoted chromatin accumulation of p53 tetramers, suggesting that the association of MLL5 with p53 may prevent the p53 tetramers from binding to the chromatin target sites. The role of MLL5 in CPT-induced p53 activation was conserved in developing zebrafish, where CPT downregulated zebrafish Mll5 protein, and the microinjection of zebrafish mll5 mRNA substantially blocked the CPT-induced apoptosis. The results of experimental investigation of detection properties of the planar microwave diodes of various configuration on DC magnetic field are presented in this paper. The detection of microwave radiation was measured at 51 GHz, 72 GHz and 144 GHz frequencies. The magnetic field was applied in plane and perpendicularly to the plane of the diodes. The experiment was performed at room temperature. Dependence of the detected voltage of the diodes on the magnetic field had asymmetric character with respect to the polarity of the magnetic field. This fact allowed us to suspect the magnetic rectification influencing the detected voltage. Therefore, average value of the detected voltage with respect to the polarity of the applied magnetic field gives its dependence on the applied magnetic field. [ABSTRACT FROM AUTHOR]
