Zaluzanin C (ZC), a sesquiterpene lactone isolated from Laurus nobilis L., has been reported to have anti-inflammatory and antioxidant effects. However, the mechanistic role of ZC in its protective effects in Kupffer cells and hepatocytes has not been elucidated. The purpose of this study was to elucidate the efficacy and mechanism of action of ZC in Kupffer cells and hepatocytes. ZC inhibited LPS-induced mitochondrial ROS (mtROS) production and subsequent mtROS-mediated NF-κB activity in Kupffer cells (KCs). ZC reduced mRNA levels of pro-inflammatory cytokines (Il1b and Tnfa) and chemokines (Ccl2, Ccl3, Ccl4, Cxcl2 and Cxcl9). Tumor necrosis factor (TNF)-α-induced hepatocyte mtROS production was inhibited by ZC. ZC was effective in alleviating mtROS-mediated mitochondrial dysfunction. ZC enhanced mitophagy and increased mRNA levels of fatty acid oxidation genes (Pparα, Cpt1, Acadm and Hadha) and mitochondrial biosynthetic factors (Pgc1α, Tfam, Nrf1 and Nrf2) in hepatocytes. ZC has proven its anti-lipid effect by improving lipid accumulation in hepatocytes by enhancing mitochondrial function to facilitate lipid metabolism. Purpose: To compare the radiographic migration profiles of primary cementless total hip arthroplasty (THA) between patients with rheumatoid arthritis (RA) and those with osteoarthritis (OA).Methods: A total of 197 patients (215 hips) who underwent cementless THA for RA or OA between January 2001 and January 2013 and followed up for a minimum of 5.5 years were included. Ninety-four RA patients (109 hips) were compared with 103 OA patients (106 hips). Radiological evaluation was performed for acetabular cup loosening, and cup migration was measured using Einzel-Bild-Röntgen-Analyse (EBRA) software. Multiple variables were assessed to identify influencing factors for cup migration.Results: Early cup migration was observed in 13 hips (11.9%) in the RA group and four hips (3.8%) in the OA group, showing a significant difference (p = 0.041). Acetabular cup loosening occurred in three cups (2.8%) in the RA group and in one cup (0.9%) in the OA group, showing no significant difference (p = 0.321). Total cup migration was higher in the RA group (2.62 mm) than in the OA group (1.44 mm, p = 0.005). Total cup migration was significantly higher in patients aged < 50 years than in those aged > 50 years (p = 0.005). Rheumatoid factor (RF) and anti-cyclic citrullinated peptide (anti-CCP) antibody influenced total cup migration. Patients with seropositive RA showed significantly higher total cup migration and early cup migration incidence than those with seronegative RA (p = 0.005, p = 0.038, respectively).Conclusions: Acetabular cups in primary cementless THAs of RA patients were less stable in terms of cup migration compared with that of OA patients. This review reports on the most updated technological aspects of Li–air battery cathode materials. It provides the reader with recent developments, alongside critical views. The requirements for air‐cathodes, as well as the classification and characterization of carbon‐based and carbon‐free air cathodes, are listed. The effects of two major substituent groups of materials, namely carbon and advanced materials (metals, metal‐oxides, metal‐carbides, and metal‐nitrides) aimed at replacing carbon, are discussed in terms of their chemical and electrochemical stability. The report covers aspects of surface chemistry and structure influence on the electrolyte and discharge products stability. The review also reports on the efforts to suppress side reactions and deterioration of the polymeric binders (if a composite electrode is being considered). This is recognized as a means to enhance Li–air battery performance. The report concludes with an outlook and perspective, providing the readers with some insight on other factors and their impact on the long road toward a viable air‐cathode suitable for Li–air battery operations. Semiconducting metal oxides (SMOs) based gas sensors are inherently hampered by requirement of high working temperature because of their wide bandgaps. Consequently, there are remain challenges to utilize SMOs based gas sensors with regard to continuous gas monitoring platform with low power consumption. To address the inherent limitation, nitridation (i.e., N doping) of WO 3 NFs to WO x N y NFs was herein introduced for realization of low-temperature NO 2 sensing. Importantly, nitridation was conducted at intervals of 50 °C from 400 to 600 °C to adjust the degree of phase transition from semiconducting WO 3 to conductive WON phases. To further optimize the conductivity and improve the sensing characteristics, i.e., reduction of optimal operating temperature and enhancement of sensitivity, WO x N y NFs were functionalized with platinum (Pt) catalytic nanoparticles (NPs) by physical mixing. Pt NPs functionalized WO x N y NFs nitrided at 550 °C exhibited improved normalized resistance change and superior selectivity against C 2 H 5 OH, C 7 H 8 , CH 4 , CO, NH 3 , and NO at 50 °C. Structural and electronic modification of titanium oxide (TiO 2 ) nanomaterials induced by the co-introduction of fully disordered glass phase and oxygen vacancies can lead to remarkable advances in the electrode performance in emerging energy storage systems. We report on the effective co-creation of fully amorphous nanofibers (NFs) composed of black TiO 2-x and conductive carbons throughout the NF structure, and evaluate the materials as potential anodes in sodium-ion batteries. The black TiO 2-x nanofiber is successfully fabricated by electrospinning a precursor solution followed by a two-step sequential thermal treatment in an air and reducing atmosphere. The NF electrode could deliver approximately two-fold higher 2nd discharge capacity and an excellent kinetic performance even under high rates compared to that delivered by anatase-structured white TiO 2 NFs used as reference, because of (i) an inherent free volume in the glass phase corresponding to the enlarged Na + sites, (ii) increased electrical conductivity (low bandgap) resulting from the presence of Ti 3+ , (iii) introduction of conductive carbon agents around the TiO 2-x domain, and (iv) one-dimensional NF feature allowing numerous Na + reaction sites at the electrochemical interface. Importance: Corneal refractive surgery may impact meibomian gland and tear film in post-refractive surgery patients. Background: To compare ocular surface parameters between post-refractive surgery patients and normal controls. Design: Cross-sectional single centre study. Participants: A total of 120 eyes of 120 subjects were divided into three groups: (i) 60 controls and 60 patients underwent corneal refractive surgery at least 12 months ago; (ii) 30 post-laser in-situ keratomileusis (LASIK) patients and (iii) 30 post-laser epithelial keratomileusis (LASEK)/photo-refractive keratectomy (PRK) patients. Methods: Tear meniscus height, non-invasive keratographic tear film break-up time and meibography were measured using the Keratograph® 5M. Fluorescein break-up time, ocular surface staining, examination of lid margins and meibomian glands, Schirmer's test and Ocular Surface Disease Index questionnaire were performed. Main Outcome Measures: Ordinary logistic regression was performed to evaluate the impact of clinical variables including refractive surgery on the meiboscores. Results: In post-LASIK patients, ocular surface parameters including Ocular Surface Disease Index scores, fluorescein break-up time and staining scores, except Schirmer's scores, were significantly worse than those in controls (P < 0.050). Ocular surface staining scores in post-LASEK/PRK patients was higher than that in the controls (P = 0.001). In post-refractive surgery patients, grade of meibomian gland parameters and meiboscores were worse than those of controls (all P < 0.050). Histories of refractive surgery were associated with high meiboscore (β = 1.100, P = 0.043 for LASIK and β = 1.039, P = 0.042 for LASEK/PRK). Conclusions and relevance: Corneal refractive surgery may adversely affect the ocular surface, and a reduction of functional meibomian glands can contribute to chronic tear film dysfunction after corneal refractive surgery. Hydrous ruthenium oxide (RuO 2) has been considered a potential electrode for supercapacitors. However, RuO 2 is not easy to be hydrous, which impedes advancement in RuO 2 -based supercapacitors. Here, strontium cation (Sr2+) – with a lower oxidation state than Ru4+ – doping effects on improving the capability of RuO 2 to be hydrous are reported. The Sr2+ doping in the RuO 2 was simply possible using electrospinning and subsequent calcination, affecting hydrous properties and promoting miniaturization of nanograins in one-dimensional (1D) nanofibers. The compositions of hydrous Sr2+-doped RuO 2 (H-Sr x Ru 1−x O 2−x) nanofibers were optimized considering correlations between synthesis temperature and degree of hydration. It was found that the H-Sr 0.2 Ru 0.8 O 1.8 nanofibers synthesized at 400 °C were hydrous compared with the pristine RuO 2 and H-Sr 0.1 Ru 0.9 O 1.9 nanofibers. The H-Sr 0.2 Ru 0.8 O 1.8 nanofibers (400) could possess the smallest nanocrystallites, increasing electrochemically active surface area and satisfying specific capacitance of 612.6 A/g at a scan rate of 50 mV s–1. • A Sr2+-doped RuO 2 nanofibers were successfully fabricated by electrospinning and calcination. • Sr2+ doping enabled RuO 2 nanofibers to be hydrous and to possess miniaturized nanograins. • Sr2+ doping in RuO 2 was optimized for annealing temperature and composition. • The highly hydrous Sr 0.2 Ru 1.8 O 1.8 nanofibers boost the capacitance performance. Ultra-stable pseudocapacitive electrodes for lithium-ion batteries (LIBs) are increasing in demand as highly sustainable energy storage system with excellent charge transport is important. The establishment of facile, controllable, and scalable synthesis of pseudocapacitive electrode materials is an attractive solution to realize such objectives. Here, we have successfully fabricated mesoporous orthorhombic Nb 2 O 5 nanofibers (m-T-Nb 2 O 5 NFs) by simple single-spinneret electrospinning followed by calcination at 600 °C. As-formed m-T-Nb 2 O 5 NFs exhibit high surface area (23.7 m 2 g −1 ) and a number of mesopores in the vacant sites where organic polymer was once decomposed. Such rationally designed m-T-Nb 2 O 5 -NFs allow facile Li ion and electron transport, with pseudocapacitive behavior. Arising from the high surface area coupled with mesopores in-between the Nb 2 O 5 nanograins, it exhibits ultra-long cycle retention (a capacity of ∼160 mAh g −1 at 500 mA g −1 after 2000 cycles and ∼88 mAh g −1 at 3000 mA g −1 after 5000 cycles) and higher rate capability (∼70 mAh g −1 at 5000 mA g −1 ). Such cycle retention characteristics of m-T-Nb 2 O 5 -NFs are at least 100-fold slower capacity decay compared with previously reported one-dimensional (1D) Nb 2 O 5 nanostructures and even superior or comparable to recently reported Nb 2 O 5 -graphene composite materials. Background To investigate the possible mechanisms by which cataract surgery aggravates meibomian gland dysfunction (MGD), we evaluated the changes in tear cytokines and ocular surface parameters after cataract surgery according to the preoperative MGD grade. Design Prospective, observational case series. Participants A total of 50 eyes from 50 patients who underwent cataract surgery were included. Methods Patients were classified into two groups: Group I had no or minimal MGD, and group II had grades 2-4 MGD. Ocular surface parameters were measured, including tear film break-up time, Schirmer I test, ocular surface staining and Ocular Surface Disease Index, and tear cytokine levels were measured. Main Outcome Measures The main outcomes were changes in ocular surface parameters and inflammatory tear cytokine concentrations. Results In group II, preoperative MGD grade, ocular surface staining, tear film break-up time and Ocular Surface Disease Index were worse, and mean interleukin (IL)-2, IL-6 and TNF-α levels were higher than those of group I. MGD and ocular surface parameters were worsened to a greater degree after surgery in group II than in group I ( P < 0.050). In group II, IL-6 and TNF-α levels significantly increased at postoperative 1 month, and there were significant correlations between changes in ocular surface parameters and tear cytokines (IL-2, IL-6 and TNF-α; P < 0.050). Conclusions The extent to which the MGD grade was aggravated following cataract surgery differed based on preoperative MGD grade. Purpose: The purpose of this study was to investigate efficient methods to evaluate the pigment layer location of tinted soft contact lenses and to assess various lens products on the market using those methods.Methods: Two types of tinted soft contact lenses with known pigment location embedded or back surface were manufactured and examined. Light microscopy (LM), focused ion beam milling and scanning electron microscopy (FIB-SEM), and Fourier-domain optical coherence tomography (FD-OCT) were used to examine the pigment layer. Lens surface roughness was also measured using atomic force microscopy. In the second part of the experiment, pigment location and surface roughness of a clear lens (Lens A) and eight commercially-available tinted soft contact lenses (Lens B-I) were evaluated using FIB-SEM and FD-OCT.Results: Pigment location could be reliably determined with FIB-SEM and FD-OCT. With LM, 40% of the lens sections were broken or deformed during slide preparation. The pigment particles in Lens B were buried below the front surface and there were no significant differences of roughness between the front and back surfaces. However, all tinted lenses with surface pigment had significant difference of roughness between front and back surfaces at the pigmented area.Conclusion: The FIB-SEM and FD-OCT could reliably locate the pigment layer of tinted soft contact lenses. We investigated the tear cytokine profiles in patients who underwent stem cell transplantation (SCT) and attempted to evaluate whether tear cytokines are associated with the presence of systemic chronic graft-versus-host disease (GVHD), regardless of ocular GVHD status. We also tested tear cytokines as biomarkers for chronic ocular GVHD severity. Forty-four patients who underwent SCT were enrolled and their diagnosis of chronic GVHD was confirmed. Ocular surface parameters and tear cytokine profiles were evaluated and the correlations between concentrations of cytokines and ocular surface parameters or several chronic ocular GVHD severity scales were evaluated. Tear interleukin (IL)-2, IL-10, IL-17α, interferon (IFN)-γ, IL-6, and tumor necrosis factor (TNF)-α were elevated in patients with chronic systemic GVHD compared with patients without chronic systemic GVHD. Receiver-operating characteristic curve analysis revealed that area under the curve (AUC) values for tear IL-10 (AUC = .795), IL-17α (AUC = .821), IL-6 (AUC = .912), and TNF-α (AUC = .910) were significantly correlated with the presence of chronic GVHD (all P < .001). Tear IL-10, IL-6, and TNF-α showed a stronger correlation with ocular surface parameters than other cytokines and these cytokines also correlated with several chronic ocular GVHD severity scales (all P < .05). Acoustic channels are characterized by long multipath spreads that cause intersymbol interference. The way in which this fact influences the design of the receiver structure is considered in this paper. To satisfy performance and throughput requirements, we propose a consecutive iterative BCJR equalization scheme. To achieve a low error performance, we resort to the powerful BCJR equalization algorithms to iteratively update probabilistic information between inner decoder and outer decoder. Also, to achieve a high throughput, we divide a long packet into a group of small consecutive packets, estimate channel information of the current packets, and use it to decode the next packets. The lack of bifunctional features of perovskite oxide toward the oxygen evolution reaction (OER) and oxygen reduction reaction (ORR) significantly limits its use as a cathode catalyst for rechargeable Zn–air batteries (ZABs). Thus far, numerous techniques to disperse additional catalysts on a perovskite host have been suggested to overcome this problem; however, cost-effectiveness and catalyst lifespan remain unsatisfactory. Herein, we present cobalt-based-nanoparticle-decorated Sr 0.95 Nb 0.1 Co 0.7 Fe 0.2 O 3–δ (S0.95NCF) by a simple ex-solution method and use it as a novel air-electrode catalyst. We successfully implemented the socketed nanoparticles with various compositions at different reduction temperatures, achieving notably enhanced activity towards OER and ORR. More importantly, the newly designed catalyst exhibits record-high charge/discharge durability over 500 h (or 1500 consequent cycles) when used as the ZABs cathode. Our findings provide essential guidelines for designing heterostructured electrocatalysts for future energy devices, in which multifunctionality is desirable. 1. Corydaline, an isoquinoline alkaloid, is one of the major active constituents in a new prokinetic botanical agent, DA-9701. It has been recommended that preclinical pharmacokinetic studies of natural medicines include both genders. Therefore, in this study, the pharmacokinetics of corydaline in male and female rats was evaluated following intravenous and oral administration of pure corydaline or DA-9701. 2. After intravenous administration of corydaline, the area under the plasma concentration-time curve (AUC) was significantly greater (by 46.4%) in female rats compared to male rats due to a 29.3% reduction in non-renal clearance in female rats. The gender difference in corydaline hepatic metabolic clearance was supported by a significantly slower metabolism of corydaline in hepatic microsomes of female rats mediated via male-specific (CYP2C11 and CYP3A2) or male-dominant (CYP3A1) CYP isozymes. 3. Following oral administration of pure corydaline or DA-9701, the AUC and Cmax values of corydaline in female rats were significantly greater (by 793% and 466% increase for corydaline administration or by 501% and 143% increase for DA-9701 administration) than in male rats. Greater F values of corydaline in female rats could be due to smaller hepatic first-pass extraction as a result of slower hepatic metabolism of corydaline. 4. However, we observed a comparable disappearance of corydaline in male and female human liver microsomes, consistent with little gender difference in CYP2C9 and CYP3A activities in humans compared to that in rats. 1. Few studies describing the pharmacokinetic properties of chlorogenic acid (CA) and corydaline (CRD) which are marker compounds of a new prokinetic botanical agent, DA-9701, have been reported. The aim of the present study is to evaluate the pharmacokinetic properties CA and CRD following intravenous and oral administration of pure CA (1-8 mg/kg) or CRD (1.1-4.5 mg/kg) and their equivalent dose of DA-9701 to rats. 2. Dose-proportional AUC and dose-independent clearance (10.3-12.1 ml/min/kg) of CA were observed following its administration. Oral administration of CA as DA-9701 did not influence the oral pharmacokinetic parameters of CA. Incomplete absorption of CA, its decomposition in the gastrointestinal tract, and/or pre-systemic metabolism resulted in extremely low oral bioavailability ( F) of CA (0.478-0.899%). 3. CRD showed greater dose-normalized AUC in the higher dose group than that in lower dose group(s) after its administration due to saturation of its metabolism via decreased non-renal clearance (by 51.3%) and first-pass extraction. As a result, the F of CRD following 4.5 mg/kg oral CRD (21.1%) was considerably greater than those of the lower dose groups (9.10 and 13.8%). However, oral administration of CRD as DA-9701 showed linear pharmacokinetics as a result of increased AUC and F in lower-dose groups (by 182% and 78.5%, respectively) compared to those of pure CRD. The greater oral AUC of CRD for DA-9701 than for pure CRD could be due to decreased hepatic and/or GI first-pass extraction of CRD by other components in DA-9701. [ABSTRACT FROM AUTHOR]

1. As promising anti-macular degeneration and/or anti-tumour agents, a better understanding of the pharmacokinetics of macrolactin A (MA) and 7- O-succinyl macrolactin A (SMA) is essential. Thus, we evaluated the pharmacokinetics of MA and SMA after intravenous, oral, or intraperitoneal administration of each drug to mice. 2. Both hepatic and extra-hepatic extractions of MA were expected based on the rapid total body clearance (CL) of MA. MA also showed a large steady-state volume of distribution ( Vss) in mice. A relatively slower CL (by 54.1%) and smaller Vss (by 85.8%) were observed for SMA than for MA. In accordance with the larger Vss values of MA than of SMA, the mouse tissues studied had good affinity to MA but less affinity to SMA. 3. Both MA and SMA had an extremely low oral extent of absolute bioavailability ( F). This could have been a result of the instability of MA and SMA in the gastrointestinal tract, supported by their unstable property in acidic buffer. Gastrointestinal and/or hepatic first-pass extraction of MA and SMA may be other reasons. 4. The pharmacokinetic profiles of both MA and SMA were much improved (greater AUC and F values) following intraperitoneal administration than following oral administration due to avoidance of acidic degradation and/or gastrointestinal first-pass extraction. Background: Acromioclavicular joint fixation using a hook plate is effective for the treatment of acute acromioclavicular joint dislocation. However, several studies have reported some complications including loss of reduction after surgery for acromioclavicular joint dislocation. This study aimed to identify the risk factors associated with the loss of reduction after acromioclavicular joint dislocation surgery using a hook plate. Methods: This was a retrospective study that assessed 118 patients with acromioclavicular joint dislocation, who were diagnosed between March 2013 and January 2019 and underwent surgical treatment using the hook plate (reduction loss group: n = 38; maintenance group: n = 80). The mean follow-up period was 29.9 months (range, 24–40 months). We assessed the range of motion, the American Shoulder and Elbow Surgeons score (ASES), visual analog scale score for pain, and a subjective shoulder value. Radiological assessment of coracoid clavicular distance was performed. The risk factors of reduction loss were analyzed using multivariable logistic regression analysis. Results: Age (p = 0.049), sex (female, p = 0.03, odds ratio OR = 4.81), Rockwood type V (p = 0.049, OR = 2.20), and time from injury to surgery > 7 days (p = 0.018, OR = 2.59) were statistically significant factors in the reduction loss group. There were no significant differences in the clinical outcomes for range of motion, ASES, subjective shoulder value, and visual analog scale scores between the two groups. In the radiological results, preoperative coracoid clavicular distance (p = 0.039) and ratio (p = 0.001), and over-reduction (p = 0.023, OR = 0.40) were significantly different between the two groups. The multivariate logistic regression analysis identified the female sex (p = 0.037, OR = 5.88), a time from injury to surgery > 7 days (p = 0.019, OR = 3.36), and the preoperative coracoid clavicular displacement ratio of the injured shoulder (p < 0.001, OR = 1.03) as risk factors associated with reduction loss following surgery using a hook plate for acromioclavicular dislocation. Conclusion: A delayed timing of surgery > 7 days, preoperative coracoid clavicular displacement ratio of the injured shoulder, and female sex were identified as risk factors for loss of reduction after surgery using a hook plate for acromioclavicular joint dislocation. A carotid-ophthalmic aneurysm often causes ocular symptoms and signs not only because of subarachnoid haemorrhage due to ruptured aneurysm but also because of the anatomical location, a proximity to the optic apparatus and ophthalmic artery. In addition, the permanent vision loss may be developed by direct injury to optic nerve and ophthalmic artery after surgical treatment. Recently, endovascular treatments such as coil embolisation have been frequently used for this aneurysm with a decreased complication. However, coil embolisation may rarely cause acute and delayed visual deterioration due to thromboembolism and perianeurysmal inflammation. Cellular senescence involves a reduction in adult stem cell self-renewal, and epigenetic regulation of gene expression is one of the main underlying mechanisms. Here, we observed that the cellular senescence of human umbilical cord blood-derived multipotent stem cells (hUCB-MSCs) caused by inhibition of histone deacetylase (HDAC) activity leads to down-regulation of high mobility group A2 (HMGA2) and, on the contrary, to up-regulation of p16, p21 and p27. We found that let-7a1, let-7d, let-7f1, miR-23a, miR-26a and miR-30a were increased during replicative and HDAC inhibitor-mediated senescence of hUCB-MSCs by microRNA microarray and real-time quantitative PCR. Furthermore, the configurations of chromatins beading on these miRNAs were prone to transcriptional activation during HDAC inhibitor-mediated senescence. We confirmed that miR-23a, miR-26a and miR-30a inhibit HMGA2 to accelerate the progress of senescence. Abstract: Flavonoids, a group of polyphenolic compounds abundant in plants, are known to prevent bone loss in ovariectomized (OVX) animal models. Inhibition of osteoclast differentiation and bone resorption is considered as an effective therapeutic approach in the treatment of postmenopausal bone loss. Luteolin, a plant flavonoid, has potent anti-inflammatory properties both in vivo and vitro. In this study, we found that luteolin markedly decreased the differentiation of both bone marrow mononuclear cells and Raw264.7 cells into osteoclasts. Luteolin also inhibited the bone resorptive activity of differentiated osteoclasts. We further investigated the effects of luteolin on ovariectomy-induced bone loss using micro-computed tomography, biomechanical tests and serum markers assay for bone remodeling. Oral administration of luteolin (5 and 20 mg/kg per day) to OVX mice caused significant increase in bone mineral density and bone mineral content of trabecular and cortical bones in the femur as compared to those of OVX controls, and prevented decreases of bone strength indexes induced by OVX surgery. Serum biochemical markers assays revealed that luteolin prevents OVX-induced increases in bone turnover. These data strongly suggest that luteolin has the potential for prevention of bone loss in postmenopausal osteoporosis by reducing both osteoclast differentiation and function. Abstract: The histone deacetylase (HDAC) inhibitors, trichostatin A (TSA) and sodium butyrate (NaBu) are considered as potent therapeutic agents for cancer treatment presenting therapeutic benefits with less risk of side effects. The microbial metabolite, TSA is a potent reversible and highly specific inhibitor of mammalian histone deacetylases. NaBu causes hyperacetylation of core histones with effects similar to TSA but it is not a specific inhibitor of HDACs. The gap junction is a channel in the plasma membrane of most cell types which allows direct communication (gap junctional intercellular communication; GJIC) of small molecules and ions. Modulation of GJIC is a known cellular event associated with tumor promotion. The effects of NaBu and TSA on the H2O2- and 12-O-tetradecanoylphorbol-13-acetate (TPA)-induced GJIC inhibition of WB cells and the mechanisms involved in the process were assessed. TSA and NaBu exerted differential preventive effects on the H2O2 and TPA-induced inhibition of GJIC as well as hyperphosphorylation of connexin43 (Cx43) in WB-F344 rat liver epithelial cells (WB cells). NaBu prevented the TPA-induced GJIC inhibition via ERK1/2 inactivation whilst TSA restored the H2O2-induced GJIC inhibition and Cx43 hyperphosphorylation by preventing p38 MAP kinase. The inhibition of tyrosine phosphorylation and down-regulation of src protein observed may also contribute to Connexin 43 dephosphorylation and GJIC restoration by TSA and NaBu partly through depletion of src protein pool. Abstract: Histone deacetylase inhibitors such as TSA, SAHA, and NaBu etc. are prospective cancer therapeutics of growing interest. Here, we demonstrated that oncogenic ras-transformed rat liver epithelial (WB-ras) cells were specifically undergone apoptosis by 48h treatment of NaBu. During this, inhibition of ras proteins, especially farnesylated form of ras, and down-regulation of ERK1/2 were observed, which suggest ras/raf/MEK/ERK down-regulation, while p38 MAP kinase was maintained up-regulated. In addition, up-regulation of pro-apoptotic proteins such as p53 and p21CIP1/WAF1, and down-regulation of cell cycle regulator/anti-apoptotic proteins such as cdk2, -4 and phosphorylated Akt were observed concurrently with an increase in apoptotic cell portion. A phosphatase inhibitor, sodium orthovanadate (SOV), efficiently blocked apoptosis and restored responsible proteins for each phenomenon including ERK1/2 while SB203580, a specific p38 MAP kinase inhibitor, showed minor effect on them. Thus, ras/ERK signaling pathway can be considered in chemotherapeutic strategies of NaBu regardless of its inhibitory action on histone deacetylase. [Copyright &y& Elsevier]

To achieve highly sensitive chemiresistive gas sensors, facile and effective synthesis for catalytic decoration is essential on an electrically conductive scaffold containing a large surface area and high porosity. Herein, we report a new synthetic strategy for the functionalization of one-dimensional carbon nanofibers (CNFs) using dual-catalytic nanoparticles (NPs) of both oxygen-incorporated MoS 2 (MoS x O y) and Pt. The functionalized CNFs were synthesized through an electrospinning process followed by heat treatment in a reducing ambient to ensure the decoration of ultra-small MoS x O y and Pt NPs along the CNFs (Pt-MoS x O y @CNFs). As a result, Pt-MoS x O y @CNFs exhibited an improved sensing response of 39.6% toward 5 ppm of NO 2 as compared to the responses of pristine CNFs (7.3%) and MoS x O y @CNFs (13.7%) at room temperature. At the optimized operating temperature of 150 °C, the Pt-MoS x O y @CNFs exhibited a 5.8-fold higher response than that of the pristine CNFs and also presented fully reversible and selective sensing properties. The enhanced NO 2 sensing properties were primarily attributed to the chemical sensitization of Pt NPs toward NO 2 and a p-n heterojunction between the CNFs and MoS x O y NPs. Carbon nanosheets (CNSs) have been used in various applications, ranging from energy storage to drug delivery. CNSs are conventionally synthesized by chemical vapor deposition, but their high cost and non-scalability prompted the development of alternative synthesis routes. More scalable methods such as the sol-gel process still suffer from its complicated synthetic steps with delicate conditions. Here, we demonstrate for the first time a unique top-down synthesis of CNSs from aronia fruit powder, which is simple, scalable, and cost-effective. Aronia-derived CNS can be fabricated at various heating temperatures (500, 700, and 900 °C) and under different atmospheres (N 2 /H 2 and NH 3), which can easily tune the overall physicochemical properties of the CNS. Background: Mesenchymal stem cells (MSCs) play important roles in tissue homeostasis by providing a supportive microenvironmental niche for the hematopoietic system. Cigarette smoking induces systemic abnormalities, including an impeded recovery process after hematopoietic stem cell transplantation. However, the role of cigarette smoking-mediated alterations in MSC niche function have not been investigated. Methods: In the present study, we investigated whether exposure to cigarette smoking extract (CSE) disrupts the hematopoietic niche function of MSCs, and pathways impacted. To investigate the effects on bone marrow (BM)-derived MSCs and support of hematopoietic stem and progenitor cells (HSPCs), mice were repeatedly infused with the CSE named 3R4F, and hematopoietic stem and progenitor cells (HSPCs) supporting function was determined. The impact of 3R4F on MSCs at cellular level were screened by bulk-RNA sequencing and subsequently validated through qRT-PCR. Specific inhibitors were treated to verify the ROS or NLRP3-specific effects, and the cells were then transplanted into the animal model or subjected to coculture with HSPCs. Results: Both direct ex vivo and systemic in vivo MSC exposure to 3R4F resulted in impaired engraftment in a humanized mouse model. Furthermore, transcriptomic profile analysis showed significantly upregulated signaling pathways related to reactive oxygen species (ROS), inflammation, and aging in 3R4F-treated MSCs. Notably, ingenuity pathway analysis revealed the activation of NLRP3 inflammasome signaling pathway in 3R4F-treated MSCs, and pretreatment with the NLRP3 inhibitor MCC950 rescued the HSPC-supporting ability of 3R4F-treated MSCs. Conclusion: In conclusion, these findings indicate that exposure to CSE reduces HSPCs supportive function of MSCs by inducing robust ROS production and subsequent NLRP3 activation. Although tetragonal phase (t-phase) in the multiple crystallographic structures of HfO 2 outperforms monoclinic phase (m-phase) most common phase that can be formed at low temperature in physicochemical properties such as dielectric constant, bandgap, high resistance to corrosion and hardness, it is extremely hard to stabilize t-phase below 1670 °C. Herein, we synthesized oxygen-deficient tetragonal hafnium oxide nanofibers (t-HfO 2–x NFs) using the electrospinning technique, which included the polymer-blended precursors and a magnesium-thermic reaction at a low processing temperature (<600 °C). The small grain size (<10 nm) of the t-HfO 2–x NFs and oxygen vacancies (V oxygen) synergistically stabilized t-phase in the nanofibrous structure. Oxygen reduction reaction (ORR) and oxygen evolution reaction (OER) performances of the t-HfO 2–x NFs in alkaline and acid media were evaluated as a potential application, which were higher than those of the m-HfO 2 NFs. Neuropathic pain (NP) is a debilitating condition stemming from damage to the somatosensory system frequently caused by nerve injuries or lesions. While existing treatments are widely employed, they often lead to side effects and lack specificity. This study aimed to alleviate NP by developing an innovative sustained-release thermosensitive hydrogel system. The system incorporates hyaluronic acid (HA)/Pluronic F127 injectable hydrogel and bupivacaine (Bup, B) in combination with poly(lactic-co-glycolic acid; P LGA)/modified magnesium hydroxide (M H)/luteolin (L ut; PML) microspheres (PML@B/Gel). The PML@B/Gel was designed for localized and prolonged co-delivery of Bup and Lut as an anesthetic and anti-inflammatory agent, respectively. Our studies demonstrated that PML@B/Gel had exceptional biocompatibility, anti-inflammatory, and antioxidant properties. In addition, it exhibited efficient pain relief in in vitro cellular assays. Moreover, this functional hydrogel showed substantial sustained drug release while diminishing microglial activation. Consequently, it effectively mitigated mechanical allodynia and thermal hyperalgesia in in vivo rat models of chronic constriction injury (CCI). Patients receiving immunosuppressive drugs to prevent organ transplant rejection exhibit a greatly increased risk of developing cutaneous squamous cell carcinoma (SCC). However, not all immunosuppressive drugs confer the same risk. Randomised, controlled trials demonstrate that switching renal transplant recipients receiving calcineurin inhibitor-based therapies to mammalian target of rapamycin (mTOR) inhibitors results in a reduced incidence of de novo SSC formation, and can even result in the regression of pre-existing premalignant lesions. However, the contribution played by residual immune function in this setting is unclear. We examined the hypotheses that mTOR inhibitors promote the enhanced differentiation and function of CD8+ memory T cells in the skin. Here, we demonstrate that the long-term oral administration of rapamycin to achieve clinically-relevant whole blood drug target thresholds, creates a “low rapamycin dose” environment in the skin. While both rapamycin and the calcineurin inhibitor tacrolimus elongated the survival of OVA-expressing skin grafts, and inhibited short-term antigen-specific CD8+ T cell responses, rapamycin but not tacrolimus permitted the statistically significant infiltration of CD8+ effector memory T cells into UV-induced SCC lesions. Furthermore, rapamycin uniquely enhanced the number and function of CD8+ effector and central memory T cells in a model of long-term contact hypersensitivity provided that rapamycin was present during the antigen sensitization phase. Thus, our findings suggest that patients switched to mTOR inhibitor regimens likely experience enhanced CD8+ memory T cell function to new antigen-challenges in their skin, which could contribute to their lower risk of de novo SSC formation and regression of pre-existing premalignant lesions. Cobalt oxide that has high energy density, is the next-generation candidate as the anode material for LIBs. However, the practical use of Co3O4 as anode material has been hindered by limitations, especially, low electrical conductivity and pulverization from large volume change upon cycling. These features lead to hindrance to its electrochemical properties for lithium-ion batteries. To improve electrochemical properties, we synthesized one-dimensional (1-D) Co3O4 nanofibers (NFs) overed with reduced graphene oxide (rGO) sheets by electrostatic self-assembly (Co3O4 NFs@rGO). The flexible graphene oxide sheets not only prevent volume changes of active materials upon cycling as a clamping layer but also provide efficient electrical pathways by three-dimensional (3-D) network architecture. When applied as an anode for LIBs, the Co3O4 NFs@rGO exhibits superior electrochemical performance: (i) high reversible capacity (615 mAh g−1 and 92% capacity retention after 400 cycles at 4.0 A g−1) and (ii) excellent rate capability. Lithium‐ion batteries (LIBs) and beyond‐LIB systems exhibit properties that are determined by electrochemical reactions occurring in their four essential components—the cathode, anode, electrolyte, and separator. Advanced analytical methods such as differential electrochemical mass spectrometry (DEMS) can assist in understanding the electrochemical behavior, which can help in advancing battery technologies. Recent studies have shown that the DEMS‐enabled real‐time gas analysis of electrochemical reactions can provide valuable information on aspects such as gaseous reactants or (side) products, which cannot be obtained appropriately through other characterization techniques. This review aims to provide a comprehensive overview of the latest developments and advancements in the use of DEMS as a rapid, operando gas‐monitoring method for advanced rechargeable battery systems. Moreover, the significance of DEMS in current and future battery development is also discussed and insights are provided into the various battery chemistries that can benefit from DEMS applications. We investigated the clinical efficacy of pinhole soft contact lenses for presbyopia correction. Twenty participants with presbyopia wore pinhole soft contact lenses in the non-dominant eye for 2 weeks. Manifest refraction, Goldmann binocular visual field tests, contrast sensitivity tests, and biomicroscopic examinations were performed along with evaluations of questionnaire responses and the binocular corrected distance visual acuity (CDVA), distance-corrected near visual acuity (DCNVA), distance-corrected intermediate visual acuity (DCIVA), and depth of focus, both before and after 2 weeks of lens wear. DCNVA at 33 and 40 cm and DCIVA at 50 and 70 cm showed significant improvements after pinhole lens wear (P-value: <0.001, <0.001, <0.001, and 0.046, respectively), with no changes in the binocular visual field and binocular CDVA. Contrast sensitivities under photopic and mesopic conditions decreased at some frequencies; however, visual function questionnaire scores significantly improved (all P-values <0.001). Covalent organic frameworks (COFs), featuring ordered nanopores with numerous accessible redox sites, have drawn much attention as promising electrode materials for rechargeable batteries. Thus far, however, COF‐based battery electrodes have exhibited limited capacity and unsatisfactory cycling stability due to the unwanted side reactions over their large surface area. Herein, a fluorine‐rich covalent organic framework (F‐COF) as an electrode material with improved stability and performance for potassium‐ion batteries is developed. The fluorinated COF not only stabilizes intercalation kinetics of K+ ions but also reinforces its electron affinity and conductivity, improving the reversibility of bond transitions during discharge–charge cycles. As a result, F‐COF affords a high specific capacity (95 mAh g−1 at fast rates up to 5 C) and excellent cycling stability (5000 cycles with ≈99.7% capacity retention), outperforming the pristine COF‐based electrodes devoid of F atoms. Notably, the experimental capacity of F‐COF approaches its theoretical value, confirming that a large proportion of electroactive sites are being actively utilized. Altogether, this work addresses the significant role of F atoms in improving the K+‐ion storage capability of COFs and provides the rational design principles for the continued development of stable and high‐performance organic electrode materials for energy storage devices. The ongoing COVID-19 pandemic highlights the urgent need for effective antiviral agents and vaccines. Drug repositioning, which involves modifying existing drugs, offers a promising approach for expediting the development of novel therapeutics. In this study, we developed a new drug, MDB-MDB-601a-NM, by modifying the existing drug nafamostat (NM) with the incorporation of glycyrrhizic acid (GA). We assessed the pharmacokinetic profiles of MDB-601a-NM and nafamostat in Sprague-Dawley rats, revealing rapid clearance of nafamostat and sustained drug concentration of MDB-601a-NM after subcutaneous administration. Single-dose toxicity studies showed potential toxicity and persistent swelling at the injection site with high-dose administration of MDB-601a-NM. Furthermore, we evaluated the efficacy of MDB-601a-NM in protecting against SARS-CoV-2 infection using the K18 hACE-2 transgenic mouse model. Mice treated with 60 mg/kg and 100 mg/kg of MDB-601a-NM exhibited improved protectivity in terms of weight loss and survival rates compared to the nafamostat-treated group. Histopathological analysis revealed dose-dependent improvements in histopathological changes and enhanced inhibitory efficacy in MDB-601a-NM-treated groups. Notably, no viral replication was detected in the brain tissue when mice were treated with 60 mg/kg and 100 mg/kg of MDB-601a-NM. Our developed MDB-601a-NM, a modified Nafamostat with glycyrrhizic acid, shows improved protectivity against SARS-CoV-2 infection. Abstract Lithium-ion batteries (LIBs) with higher energy density are necessary to meet the increasing demands of energy storage system (ESS) in near future. Tin (IV) oxide, SnO 2 , is one of highly promising anode candidates due to its high theoretical capacity (782 mAh g−1), abundance, environmental friendliness, and safety with organic electrolytes. However, a rapid capacity fading and poor rate capabilities arising from the large volume expansion and subsequent agglomeration of Sn nanoparticles have been major issues of SnO 2. Here, we have synthesized one-dimensional (1D) SnO 2 -amorphous titanium (IV) oxide NTs (SnO 2 -a-TiO 2 NTs), which allow both facile ionic and electron transport as well as easy penetration of electrolytes. The resultant SnO 2 -a-TiO 2 NTs not only alleviate volume expansion by maintaining their structural integrity but also possess minimal charge transfer resistance even after a number of cycles. SnO 2 -a-TiO 2 NTs exhibit both excellent cycle retention characteristics (1050.2 mAh g−1 after 250 cycles) and outstanding rate capability (522.3 mAh g−1 at a current density of 5000 mA g−1), which is attributed to the introduction of amorphous TiO 2 that not only acts as buffer agent for volume changes of SnO 2 but also allows fast surface-controlled diffusion process due to its pseudocapacitive charge storage mechanisms. Abstract: Edges of 2D transition metal dichalcogenides (TMDs) are well known as highly reactive sites, thus researchers have attempted to maximize the edge site density of 2D TMDs. In this work, metal‐organic framework (MOF) templates are introduced to synthesize few‐layered WS2 nanoplates (a lateral dimension of ≈10 nm) confined in Co, N‐doped hollow carbon nanocages (WS2_Co‐N‐HCNCs), for highly sensitive NO2 gas sensors. WS2 precursors are assembled in the surface cavity of Co‐based zeolite imidazole framework (ZIF‐67) and subsequent pyrolysis produced WS2_Co‐N‐HCNCs. During the pyrolysis, the carbonized ZIF‐67 are doped by Co and N elements, and the growth of WS2 is effectively suppressed, creating few‐layered WS2 nanoplates functionalized Co‐N‐HCNCs. The WS2_Co‐N‐HCNCs exhibit outstanding NO2 sensing characteristics at room temperature, in terms of response (48.2% to 5 ppm), selectivity, response and recovery speed, and detection limit (100 ppb). These results are attributed to the enhanced adsorption and desorption kinetics of NO2 on abundant WS2 edges, confined in the gas permeable HCNCs. This work opens up an efficient way for the facile synthesis of edge abundant few‐layered TMDs combined with porous carbon matrix via MOF templating route, for applications relying on highly active sites. Facile synthesis of rationally designed structures is critical to realize a high performance electrode for lithium-ion batteries (LIBs). Among different candidates, tin(IV) oxide (SnO 2 ) is one of the most actively researched electrode materials due to its high theoretical capacity (1493 mAh g −1 ), abundance, inexpensive costs, and environmental friendliness. However, severe capacity decay from the volume expansion and low conductivity of SnO 2 have hampered its use as a feasible electrode for LIBs. Rationally designed SnO 2 -based nanostructures with conductive materials can be an ideal solution to resolve such limitations. In this work, we have successfully fabricated porous SnO 2 -CuO composite nanotubes (SnO 2 -CuO p-NTs) by electrospinning and subsequent calcination step. The porous nanotubular structure is expected to mitigate the volume expansion of SnO 2 , while the as-formed Cu from CuO upon lithiation allows faster electron transport by improving the low conductivity of SnO 2 . With a synergistic effect of both Sn and Cu-based oxides, SnO 2 -CuO p-NTs deliver stable cycling performance (91.3% of capacity retention, ∼538 mAh g −1 ) even after 350 cycles at a current density of 500 mA g −1 , along with enhanced rate capabilities compared with SnO 2 . Anatase TiO2 has been suggested as a potential sodium anode material, but the low electrical conductivity of TiO2 often limits the rate capability, resulting in poor electrochemical properties. To address this limitation, we propose graphene-wrapped anatase TiO2 nanofibers (rGO@TiO2 NFs) through an effective wrapping of reduced graphene oxide (rGO) sheets on electrospun TiO2 NFs. To provide strong electrostatic interaction between the graphene oxide (GO) sheets and the TiO2 NFs, poly(allylamine hydrochloride) (PAH) was used to induce a positively charged TiO2 surface by the immobilization of the -NH3+ group and to promote bonding with the negatively charged carboxylic acid (-COO−) and hydroxyl (-O−) groups on the GO. A sodium anode electrode using rGO@TiO2 NFs exhibited a significantly improved initial capacity of 217 mAh g−1, high capacity retention (85% after 200 cycles at 0.2C), and a high average Coulombic efficiency (99.7% from the second cycle to the 200th cycle), even at a 5C rate, compared to those of pristine TiO2 NFs. The lithium metal anode is notoriously unstable and reactive to electrolytes, forming brittle solid‐electrolyte interphase (SEI) layers with uneven distribution, exacerbating Li dendrites that ultimately limit the battery life. Here, this work passivates Li metal with electrolyte‐swellable polymer nanolayers deposited monolithically by the initiated chemical vapor deposition (iCVD) to reinforce the native SEI layers and stabilize their interface. The 100 nm iCVD poly(dimethylaminomethyl styrene) (pDMAMS) layer is swelled by 264% under a carbonate electrolyte, establishing the electrolyte‐filled soft scaffold for Li‐ion transport. Notably, the solvogel accommodates homogeneous Li2O‐free and Li2CO3‐rich native SEI layers, providing a record‐high Li‐ion transference number of 0.95 and ionic conductivity of 6.54 mS cm−1. The developed pDMAMS‐Li anodes extend the cycle life by 550% in Li–Li symmetric cells and 600% in LiNi0.6Mn0.2Co0.2O2 full cells compared to pristine Li metal. The mechanistic details of the swollen‐soft‐scaffold strategy are elucidated by depth profile analysis of the pDMAMS homopolymer compared to pDMAMS/electrolyte‐phobic copolymers, providing new insights to manage the interface of liquid‐state electrolyte and solid‐state Li metal by using a novel class of bifunctional solvogel. Osteoporotic bone regeneration is a challenging process which involves the occurrence of sophisticated interactions. Although various polymeric scaffolds have been proposed for bone repair, research on osteoporotic bone regeneration remains practically limited. In particular, achieving satisfactory bone regeneration when using osteoporotic drugs is challenging including bisphosphonates. Here, a novel nitric oxide‐releasing bioinspired scaffold with bioactive agents for the exquisite regeneration of osteoporotic bone is proposed. The bone‐like biomimetic poly(lactic‐co‐glycolic acid) scaffold is first prepared in combination with organic/inorganic ECM and magnesium hydroxide as the base implant material. Nanoparticles containing bioactive agents of zinc oxide (ZO), alendronate, and BMP2 are incorporated to the biomimetic scaffold to impart multifunctionality such as anti‐inflammation, angiogenesis, anti‐osteoclastogenesis, and bone regeneration. Especially, nitric oxide (NO) generated from ZO stimulates the activity of cGMP and protein kinase G; in addition, ZO downregulates the RANKL/osteoprotegerin ratio by suppressing the Wnt/β‐catenin signaling pathway. The new bone is formed much better in the osteoporotic rat model than in the normal model through the regulation of bone homeostasis via the scaffold. Silicon-coated molybdenum sulfide nanosheets were prepared in a thin-film shape (Si–MoS 2 TFs) through metal organic chemical vapor deposition (MOCVD) and radio frequency (RF) sputtering. By using MOCVD, the MoS 2 thin films composed of nanosheets were vertically grown on stainless steel substrates. To address the problem of polysulfide dissolution and shuttling effect of MoS 2 during discharge and charge reactions in thin-film Li-ion batteries, the entire surfaces of MoS 2 TFs were covered with silicon layers using RF sputtering. Compared with pristine MoS 2 TFs without protection, the Si–MoS 2 TFs showed excellent reversibility with a high capacity of ∼2000 mAh g−1 for 500 cycles in thin-film Li-ion batteries. Both materials – MoS 2 and Si – were synergistically taken into account to enhance the ability to store Li ions in the host structures and complement each other during repeated conversion and alloying reactions. Nanostructuring has significantly contributed to alleviating the huge volume expansion problem of the Ge anodes. However, the practical use of nanostructured Ge anodes has been hindered due to several problems including a low tap density, poor scalability, and severe side reactions. Therefore, micrometer-sized Ge is desirable for practical use of Ge-based anode materials. Here, micronized Ge3N4 with a high tap density of 1.1 mg cm−2 has been successfully developed via a scalable wet oxidation and a subsequent nitridation process of commercially available micrometer-sized Ge as the starting material. The micronized Ge3N4 shows much-suppressed volume expansion compared to micrometer-sized Ge. After the carbon coating process, a thin carbon layer (≈3 nm) is uniformly coated on the micronized Ge3N4, which significantly improves electrical conductivity. As a result, micronized Ge3N4@C shows high reversible capacity of 924 mAh g−1 (2.1 mAh cm−2) with high mass loading of 3.5 mg cm−2 and retains 91% of initial capacity after 300 cycles at a rate of 0.5 C. Additionally, the effectiveness of Ge3N4@C as practical anodes is comprehensively demonstrated for the full cell, showing stable cycle retention and especially excellent rate capability, retaining 47% of its initial capacity at 0.2 C for 12 min discharge/charge condition. Due to the unique properties which are considerably different from macro‐scale or bulk materials, 1D) nanostructures have received great interests. In particular, they have greatly contributed to building next‐generation batteries by providing beneficial features such as short ion/electron pathways, structural versatility (i.e., formation of 3D current collectors, free‐standing electrodes, and separators), and excellent stress relaxation. Owing to these definite advantages, 1D nanostructure is often followed by discovery of new electrode materials. This review provides a systematic overview of the state‐of‐the‐art research progresses on 1D nanostructures, which are extensively used for rechargeable batteries. Specifically, a brief introduction of some important 1D nanostructuring methods is started and then, in situ structural characterizations using 1D nanostructures are summarized, which allow a great step forward in atomic‐scale monitoring of reaction kinetics and observing dynamic structural evolution of electrode materials. 1D nanstructuring in solid‐state electrolytes and stabilization of metal anodes are also highlighted which are not only substantially important in current research trends for future batteries but also rarely discussed in previous reviews. Hederacoside C (HDC) is one of the active ingredients in Hedera helix leaf extract (Ivy Ex.) and AG NPP709, a new botanical drug to treat acute respiratory infection and chronic inflammatory bronchitis. However, information regarding its pharmacokinetic properties remains limited. 2. Here, we report the pharmacokinetics of HDC in rats after intravenous administration of HDC (3, 12.5, and 25 mg/kg) and after oral administration of HDC, Ivy Ex., and AG NPP709 (equivalent to 12.5, 25, and 50 mg/kg HDC). 3. Linear pharmacokinetics of HDC were identified upon its intravenous administration at doses of 3-25 mg/kg. Intravenous administration of HDC results in relatively slow clearance (1.46-2.08 mL/min/kg) and a small volume of distribution at steady state (138-222 mL/kg), while oral administration results in a low absolute oral bioavailability ( F) of 0.118-0.250%. The extremely low F of HDC may be due to poor absorption of HDC from the gastrointestinal (GI) tract and/or its decomposition therein. 4. Abstract: Osteoclasts are multinucleated cells that have a unique role in bone degradation. Modulation of osteoclast formation and/or its activity is an important approach for the treatment of bone-destructive diseases such as osteoporosis and rheumatoid arthritis. In this study, Gymnasterkoreayne F (GK-F), a natural compound isolated from Gymnaster koraiensis, was found to inhibit osteoclast differentiation from primary bone marrow-derived macrophages (BMMs) in a dose-dependent manner. The inhibition occurred through the suppression of nuclear factor of activated T cells c1 (NFATc1) expression, which then led to the decreased levels of osteoclastogenic markers, including Cathepsin K and tartrate-resistant acid phosphatase (TRAP). In addition, GK-F abolished pre-osteoclast fusion induced by the receptor activator of nuclear factor kappa B ligand (RANKL), lipopolysaccharide (LPS) and TNF-α. Reflecting its inhibitory effects on cell–cell fusion, GK-F attenuated the gene expression of an essential molecule of osteoclast fusion, the dendritic cell-specific transmembrane protein (DC-STAMP). Furthermore, GK-F inhibited the bone resorptive activity of differentiated osteoclasts through its ability to block RANKL-induced actin ring formation. The effect was associated with a significant decrease in the induction of β3 integrin expression, which is an essential regulator of osteoclast cytoskeletal function. It has widely been reported that basic fibroblast growth factor (bFGF) promotes proliferation of human stem cells and contributes to the maintenance of their self-renewal capability through repeated replications. In contrast to embryonic stem cells (ESCs), the effects of growth factors on adult stem cells are poorly understood. In human umbilical cord blood-derived multipotent stem cells (hUCB-MSCs), bFGF is associated with an increased number of proliferating cells. Furthermore, expression levels of ESC markers were increased after treatment with bFGF. bFGF also increased the expression of FGFR, which in turn increased expression of insulin-like growth factor (IGFs). Since IGFs exert autocrine and paracrine effects on stem cells, bFGF-mediated release of IGFs from hUCB-MSCs might enhance FGFR1 and IGF1R expression in neighboring cells. These receptors could subsequently regulate the effects of bFGF and IGFs in adult stem cells. Abstract: Stem cell niches provide the micro-environment for the development of stem cells. Under our culturing regimen, a kind of osteoclast-centralized structure supports the proliferation of MSCs, derived from human cord blood, once they reside on osteoclasts. MSCs in this structure expressed Oct4 which is a marker of embryonic stem cells. Floating daughter cells of MSCs colony showed abilities to differentiate into osteocyte, adipocyte, and neuronal progenitor cells. Compared with the easy senescence of MSCs without this niche-like structure in vitro, these results suggested that osteoclasts might play an important role the development and maintenance of Umbilical cord blood (UCB)-derived MSCs and might provide a means to expand UCB-MSCs in vitro, more easily, through a stem cell niche-like structure. The room transfer function is computed by cross-correlation method There are some methods to measure the RFT. For example there are the image methods, ray tracing method to measure actually the RFT. In this we estimated RTF using white Gaussian noise that is broadband signal. It is important to extract feature vectors. Feature extraction is the process of computing a numerical representation. We used various features to detect intruder. The features based on the Short Time Fourier Transform (STFT) and the zero-crossing rates were used Several simulations and actually tests show that the proposed method provides good performance for intruder detection.
