Under the influence of novel corona virus pneumonia epidemic prevention and control, higher requirements for behavior recognition in complex environment are put forward. The accuracy of traditional methods for sports training is not high, so a method is needed to improve the local action recognition to assist sports training. In the process of behavior recognition, if only the track is regarded as an independent individual, the information of its neighbor will be ignored. Therefore, we use KNN algorithm to get the nearest neighbor trajectory. In order to calculate the rich neighborhood information around the track, this paper calculates the complex relationship between the center track and the neighborhood track from four different angles, including absolute motion, relative motion, distance relationship and direction relationship. Then, from the four different perspectives of variance, discrete coefficient, skewness and kurtosis, this paper proposes a large interval nearest neighbor coding method. This method makes the four eigenvalues complement each other and improves the ability of describing complex and changeable behaviors. The experimental results show that the coding method proposed in this paper can be used for behavior recognition according to different transformation matrix. [ABSTRACT FROM AUTHOR]

Ten-eleven translocation (TET) methylcytosine dioxygenases are enzymes that catalyze the demethylation of 5-methylcytosine on DNA. Through global and site-specific demethylation, they regulate cell fate decisions during development and in embryonic stem cells by maintaining pluripotency or by regulating differentiation. In this Primer, we provide an updated overview of TET functions in development and stem cells. We discuss the catalytic and noncatalytic activities of TETs, and their roles as epigenetic regulators of both DNA and RNA hydroxymethylation, highlighting how TET proteins function in regulating gene expression at both the transcriptional and post-transcriptional levels. 4. Knockdown of TMEM16A suppressed MAPK and inhibited cell proliferation and migration in hepatocellular carcinoma.

TMEM16A plays an important role in cell proliferation in various cancers. However, less was known about the expression and role of TMEM16A in hepatocellular carcinoma. We screened the expression of TMEM16A in patients' hepatocellular carcinoma tissues, and also analyzed the biological function of hepatocellular carcinoma cells by knockdown of TMEM16A, as well as the expression of MAPK signaling proteins, including p38, p-p38, ERK1/2, p-ERK1/2, JNK, and p-JNK, and cell cycle regulatory protein cyclin D1 in TMEM16A siRNA-transfected SMMC-7721 cells by Western blot. Our results showed that TMEM16A was overexpressed in hepatocellular carcinoma tissues. Inhibition of TMEM16A suppressed the cell proliferation, migration, and invasion, and cell cycle progression but did not influence the cell apoptosis. TMEM16A siRNAsuppressed cancer cell proliferation and tumor growth were accompanied by a reduction of p38 and ERK1/2 activation and cyclin D1 induction, and were not influenced by other tested MAPK signaling proteins. In addition, inhibition of TMEM16A suppressed tumorigenicity in vivo. TMEM16A is overexpressed in hepatocellular carcinoma, and that inhibition of TMEM16A suppressed MAPK and growth of hepatocellular carcinoma. Type 2 diabetes mellitus (T2D), affecting >90% of the diabetic patients, is one of the major threats to human health. A comprehensive understanding of the mechanisms of T2D at molecular level is essential to facilitate the related translational research. Here, we introduce a comprehensive and up-to-date knowledgebase for T2D, i.e. T2D@ZJU. T2D@ZJU contains three levels of heterogeneous connections associated with T2D, which is retrieved from pathway databases, protein-protein interaction databases and literature, respectively. In current release, T2D@ZJU contains 1078 T2D related entities such as proteins, protein complexes, drugs and others together with their corresponding relationships, which include 3069 manually curated connections, 14 893 protein-protein interactions and 26 716 relationships identified by text-mining technology. Moreover, T2D@ZJU provides a user-friendly web interface for users to browse and search data. A Cytoscape Web-based interactive network browser is available to visualize the corresponding network relationships between T2D-related entities. Abstract: Salacia chinensis is a traditional South and Southeast Asian herb medicine and has been reported to have an antidiabetic function via α-glucosidases inhibitory activity. In this study, the effects of S. chinensis extract (SCE) on reproductive functions of F0 males and females and the effects on survival and growth of F1 offspring were examined using Sprague–Dawley rats. SCE was administered at dose levels of 0, 500, 1000 and 2000mg/kg/day orally to groups consisting of 25 males and 25 females. Males were dosed once a day in the morning from 8weeks before mating, throughout the mating period and until the day before necropsy and females were dosed once a day in the morning for 2weeks before mating and through the mating, gestation and lactation periods (until day 20 of lactation). In all SCE treatment groups, no toxic signs were noted on reproductive outcome such as estrous cycle of F0 females or any parameters for reproductive function or survival, growth, sensory reflex or function development of F1 pups. Therefore, we concluded that SCE has no effects on the reproductive outcome even at a remarkably high dosage level, 2000mg/kg/day, in Sprague–Dawley rats. [Copyright &y& Elsevier]

Of 322 stool specimens collected from children with diarrhea from October 2005 through September 2006 in Wuhan, China, group A rotavirus was identified in 101 (31.4%). The most prevalent group A rotavirus genotype was G3P[8] (62.6%), followed by G1P[8](17.6%), G1+G3P[8](8.8%), G3P[4](6.6%), G1P[4](2.2%), and G9P[8](2.2%). Coronary heart disease (CHD), the leading cause of global morbidity and mortality in adults, has been reported to be associated with hundreds of genes. A comprehensive understanding of the CHD-related genes and their corresponding interactions is essential to advance the translational research on CHD. Accordingly, we construct this knowledgebase, CHD@ZJU, which records CHD-related information (genes, pathways, drugs and references) collected from different resources and through text-mining method followed by manual confirmation. In current release, CHD@ZJU contains 660 CHD-related genes, 45 common pathways and 1405 drugs accompanied with >8000 supporting references. Almost half of the genes collected in CHD@ZJU were novel to other publicly available CHD databases. Additionally, CHD@ZJU incorporated the protein-protein interactions to investigate the cross-talk within the pathways from a multi-layer network view. These functions offered by CHD@ZJU would allow researchers to dissect the molecular mechanism of CHD in a systematic manner and therefore facilitate the research on CHD-related multi-target therapeutic discovery. 9. LINC00978 promotes hepatocellular carcinoma carcinogenesis partly via activating the MAPK/ERK pathway.

Objective: To study the role of long non-coding RNA (IncRNA) LINC00978 in hepatocellular carcinoma (HCC) carcinogenesis. Materials and methods: LINC00978 expression level was measured by reverse transcription quantitative real-time PCR (RT-qPCR) in HCC tissues and adjacent healthy liver tissues from 49 HCC patients. MTT assay, colony forming assay, and flow cytometry were performed to evaluate the effects of shRNA-mediated LINC00978 knockdown on HCC cell proliferation, cell cycle progression, and apoptosis in vitro. Xenograft tumor model was performed to determine the effects of LINC00978 knockdown on HCC tumor growth in vivo. Western blot was used to assess the activation of signaling molecules in the apoptosis and mitogen-activated protein kinase (MAPK)/extracellular signal-regulated kinase (ERK) pathway. Results: LINC00978 expression was significantly up-regulated in human HCC tissue relative to adjacent normal tissue, and LINC00978 high expression was correlated with poor HCC overall survival. LINC00978 was up-regulated in HCC cell lines. ShRNA-mediated LINC00978 knockdown significantly decreased HCC cell proliferation, and induced HCC cell cycle arrest and apoptosis in vitro. LINC00978 knockdown led to significant decrease in tumor xenograft size in vivo. Western blots revealed LINC00978 inhibition decreased ERK, p38, and c-Jun N-terminal kinase (JNK) phosphorylation in HCC cells. Conclusions: LINC00978 is highly expressed in human HCC tissue and correlates with poor HCC prognosis. LINC00978 promotes HCC cell proliferation, cell cycle progression, and survival, partially by activating the MAPK/ERK pathway. Our findings partially elucidated the roles of LINC00978 in HCC carcinogenesis, and identified a therapeutic target for HCC. [ABSTRACT FROM AUTHOR]

10. Distinct functions and prognostic values of RORs in gastric cancer.

- Abstract
Retinoic acid receptor-related orphan receptors (RORs) are frequently abnormally expressed in several human malignancies, including gastric cancer (GC). RORs are involved in the development and progression of GC through Wnt signaling pathway receptors and other common receptors. However, the prognostic roles of individual RORs in patients with GC remain elusive. We accessed the prognostic roles of three RORs (RORα, RORβ, and RORγ) through “The Kaplan–Meier plotter” (KM plotter) database in patients with GC. For all patients with GC who were followed for 20 years, the low mRNA expression of all three RORs showed a significant correlation with better outcomes. We further accessed the prognostic value of individual RORs in different clinical pathological features including Lauren classification, clinical stages, pathological grades, HER2 status, and different treatments methods. The RORs demonstrated critical prognostic roles in GC. Expressions of RORs were higher in GC tissues when compared with normal gastric tissues. Moreover, knockdown of RORs significantly inhibited cell proliferation and migration, suggesting an oncogenic role of RORs in human GC. These findings suggest potential roles of RORs as biomarkers for GC prognosis and as oncogenes in GC. [ABSTRACT FROM AUTHOR]

11. Study on Climate and Grassland Fire in HulunBuir, Inner Mongolia Autonomous Region, China.

Grassland fire is one of the most important disturbance factors of the natural ecosystem. Climate factors influence the occurrence and development of grassland fire. An analysis of the climate conditions of fire occurrence can form the basis for a study of the temporal and spatial variability of grassland fire. The purpose of this paper is to study the effects of monthly time scale climate factors on the occurrence of grassland fire in HulunBuir, located in the northeast of the Inner Mongolia Autonomous Region in China. Based on the logistic regression method, we used the moderate-resolution imaging spectroradiometer (MODIS) active fire data products named thermal anomalies/fire daily L3 Global 1km (MOD14A1 (Terra) and MYD14A1 (Aqua)) and associated climate data for HulunBuir from 2000 to 2010, and established the model of grassland fire climate index. The results showed that monthly maximum temperature, monthly sunshine hours and monthly average wind speed were all positively correlated with the fire climate index; monthly precipitation, monthly average temperature, monthly average relative humidity, monthly minimum relative humidity and the number of days with monthly precipitation greater than or equal to 5 mm were all negatively correlated with the fire climate index. We used the active fire data from 2011 to 2014 to validate the fire climate index during this time period, and the validation result was good (Pearson’s correlation coefficient was 0.578), which showed that the fire climate index model was suitable for analyzing the occurrence of grassland fire in HulunBuir. Analyses were conducted on the temporal and spatial distribution of the fire climate index from January to December in the years 2011–2014; it could be seen that from March to May and from September to October, the fire climate index was higher, and that the fire climate index of the other months is relatively low. The zones with higher fire climate index are mainly distributed in Xin Barag Youqi, Xin Barag Zuoqi, Zalantun Shi, Oroqen Zizhiqi, and Molidawa Zizhiqi; the zones with medium fire climate index are mainly distributed in Chen Barag Qi, Ewenkizu Zizhiqi, Manzhouli Shi, and Arun Qi; and the zones with lower fire climate index are mainly distributed in Genhe Shi, Ergun city, Yakeshi Shi, and Hailar Shi. The results of this study will contribute to the quantitative assessment and management of early warning and forecasting for mid-to long-term grassland fire risk in HulunBuir. [ABSTRACT FROM AUTHOR]

Grassland, as one of the most important ecosystems on Earth, experiences fires that affect the local ecology, economy and society. Notably, grassland fires occur frequently each year in northeastern China. Fire occurrence is a complex problem with multiple causes, such as natural factors, human activities and land use. This paper investigates the disruptive effects of grassland fire in the northeastern Inner Mongolia Autonomous Region of China. In this study, we relied on thermal anomaly detection from the Moderate Resolution Imaging Spectroradiometer (MODIS) sensor to identify fire occurrences, and land use data were acquired by Landsat Thematic Mapper/Enhanced Thematic Mapper (TM/ETM). We discussed the relationship between land use and the spatial distribution of grassland fires. The results showed that the impact of land use on grassland fires was significant. Spatially, approximately 80% of grassland fires were clustered within 10 km of cultivated land, and grassland fires generally occurred in areas of intense human activity. The correlation between the spatial distribution of grassland fires and the land use degree in 2000, 2005 and 2010 was high, with R2 values of 0.686, 0.716, 0.633, respectively (p < 0.01). These results highlight the importance of the relationship between land use and grassland fire occurrence in the northeastern Inner Mongolia Autonomous Region. 13. Identification and characterization of a novel stress-responsive outer membrane protein Lip40 from Actinobacillus pleuropneumoniae.

Background: Actinobacillus pleuropneumoniae, a Gram-negative bacterium, is the causative agent of porcine pleuropneumonia, a highly contagious and often fatal disease. Because current vaccines confer limited protection against A. pleuropneumoniae infection, the development of more effective vaccines is urgently required. The identification of immunogenic and protective antigens, such as an outer-membrane lipoprotein, will advance this purpose. Results: Sixty putative lipoproteins were predicted from the genomic sequence of A. pleuropneumoniae using multiple algorithms. Here, we focused on the characteristics of the putative lipoprotein Lip40 from A. pleuropneumoniae strain SLW01 (serovar 1). Lip40 shares sequence similarity with many bacterial lipoproteins, and the structural prediction of Lip40 suggests that it is similar to A. pleuropneumoniae TbpB. The N-terminus of Lip40 contains an interesting tandemly repeated sequence, Q(E/D/P)QPK. Real-time RT-PCR indicated that the expression of lip40 was significantly upregulated at 42 °C, at 16 °C, and under anaerobic conditions. Recombinant Lip40 (rLip40) produced in Escherichia coli BL21(DE3) was specifically recognized by porcine convalescent serum directed against A. pleuropneumoniae. Lip40 was confirmed to localize at the bacterial outer membrane, and its expression was significantly stimulated when A. pleuropneumoniae was cultured under various stress conditions. Lip40 also protected 75 % of mice from fatal virulent A. pleuropneumoniae infection. Conclusions: The immunogenic outer-membrane protein Lip40 is stress responsive, protects mice against infection, and might be a virulence determinant. Further investigation of Lip40 should expedite vaccine development and provide insight into the pathogenesis of A. pleuropneumoniae. [ABSTRACT FROM AUTHOR]

Chinese medicine has evolved from thousands of years of empirical applications and experiences of combating diseases. It has become widely recognized that the Chinese medicine acts through complex mechanisms featured as multicompound, multitarget and multipathway. However, there is still a lack of systematic experimental studies to elucidate the mechanisms of Chinese medicine. In this study, the differentially expressed genes (DEGs) were identified from myocardial infarction rat mo del treated with Xuesaitong Injection (XST), a Chinese medicine consisting of the total saponins from Panax notoginseng (Burk.) F. H. Chen (Chinese Sanqi). A network-based approach was developed to combine DEGs related to cardiovascular diseases (CVD) with lines of evidence from the literature miningto investigate the mechanism of action (MOA) of XST on antimyocardial infarction. A compound-target-pathway network of XST was constructed by connecting compounds to DEGs validated with literature lines of evidence and the pathways that are functionally enriched. Seventy potential targets of XST were identified in this study, of which 32 were experimentally validated either by our in vitro assays or by CVD-related literatures. 15. Influence of genetic background on albuminuria and kidney injury in Ins2+/C96Y (Akita) mice.

Previous studies have shown that Akita mice bearing the Ins2+/C96Y mutation have significant advantages as a type I diabetes platform for developing models of diabetic nephropathy (DN; Gurley SB, Clare SE, Snow KP, Hu A, Meyer TW, Coffman TM. Am J Physiol Renal Physiol 290: F214-F222, 2006). In view of the critical role for genetic factors in determining susceptibility to DN in humans, we investigated the role of genetic background on kidney injury in Akita mice. To generate a series of inbred Akita mouse lines, we back-crossed the Ins2C96Y mutation more than six generations onto the 129/SvEv and DBA/2 backgrounds and compared the extent of hyperglycemia and renal disease with the standard C57BL/6-Ins2+/C96Y line. Male mice from all three Akita strains developed marked and equivalent hyperglycemia. However, there were significant differences in the level of albuminuria among the lines with a hierarchy of DBA/2 > 129/SvEv > C57BL/6. Renal and glomerular hypertrophy was seen in all of the lines, but significant increases in mesangial matrix compared with baseline nondiabetic controls were observed only in the 129 and C57BL/6 backgrounds. In F1(DBA/2 x C57BL/6)-Ins2+/C96Y mice, the extent of albuminuria was similar to the parental DBA/2-Ins2+/C96Y line; they also developed marked hyperfiltration. These studies identify strong effects of genetic background to modify the renal phenotype associated with the Ins2C96Y mutation. Identification of these naturally occurring strain differences should prove useful for nephropathy modeling and may be exploited to allow identification of novel susceptibility alleles for albuminuria in diabetes. Virus-like particles (VLPs) represent a promising vaccine against severe acute respiratory syndrome coronavirus (SARS CoV). In this study, recombinant baculovirus vAcS and vAcME were constructed to express the S protein and the M and E proteins of SARS CoV, respectively. Electron microscope analysis demonstrated the formation of VLPs in vAcME and vAcS coinfected insect cells. Mice immunized four times with VLPs developed high antibody titres against SARS CoV. In addition, VLPs elicited cell-mediated immunity as demonstrated by enhanced interferon-γ and interleukin-4 production. VLPs also conferred protective immunity against the infection of Spike protein pseudotyped murine leukaemia virus. Our findings demonstrate that SARS CoV VLPs are immunogenic and can elicit strong SARS CoV-specific humoral and cellular immune responses in mice. This is the first study describing the immunogenicity of SARS CoV VLPs, providing valuable data for developing a protective vaccine against SARS CoV infection. [ABSTRACT FROM AUTHOR]

Adenovirus (Ad) type 5 DNA packaging is initiated in a polar fashion from the left end of the genome. The packaging process is dependent upon the cis-acting packaging domain located between nucleotides 194 and 380. Seven A/T-rich repeats have been identified within this domain that direct packaging. A1, A2, A5, and A6 are the most important repeats functionally and share a bipartite sequence motif. Several lines of evidence suggest that there is a limiting trans-acting factor(s) that plays a role in packaging. Two cellular activities that bind to minimal packaging domains in vitro have been previously identified. These binding activities are P complex, an uncharacterized protein(s), and chicken ovalbumin upstream promoter transcription factor (COUP-TF). In this work, we report that a third cellular protein, octamer-1 protein (Oct-1), binds to minimal packaging domains. In vitro binding analyses and in vivo packaging assays were used to examine the relevance of these DNA binding activities to Ad DNA packaging. The results of these experiments reveal that COUP-TF and Oct-1 binding does not play a functional role in Ad packaging, whereas P-complex binding directly correlates with packaging function. We demonstrate that P complex contains the cellular protein CCAAT displacement protein (CDP) and that full-length CDP is found in purified virus particles. In addition to cellular factors, previous evidence indicates that viral factors play a role in the initiation of viral DNA packaging. We propose that CDP, in conjunction with one or more viral proteins, binds to the packaging sequences of Ad to initiate the encapsidation process. [ABSTRACT FROM AUTHOR]
