In order to study the relationship between rock mechanical properties and resistivity under deep underground environmental conditions, a rock resistivity test system was developed which can realize simultaneous control of high and low temperature, pressure, seepage and chemical environment, and the corresponding specimen sealing method was explored. The system mainly includes triaxial system, chemical permeation system, temperature control system and test control system. The reliability of the system was verified by tests and preliminary experiments. Background and Objectives: The popularity of takeaway has caused health problems. To analyse the basic nutrients and composition of popular takeaway meals in Chengdu, China.Methods and Study Design: We randomly collected 105 takeaway meals from takeaway platforms. The quality of ingredients such as grains, vegetables, and meat were assessed and weighed. The samples were then homogenised, and the nutrients were detected following the AOAC Official Methods of Analysis.Results: Compared with Chinese and US dietary reference intakes, the average energy, protein, salt, fat, vitamin, and available carbohydrate contents exceeded dietary recommendations for one takeaway meal. By contrast, the whole grain, vegetable, fruit, dairy product, egg, mineral, and dietary fibre contents were insufficient. Food compositions and basic nutrients differed among takeaway meals prepared with various cooking methods and meats. Fried rice had the lowest nutritional value. The fried dish set meal had high energy density. The nutrient content of poultry takeaway meals was more balanced compared with other meals assessed, and salt and fat were excessive in mixed meat meals. In addition, meatless takeaway meals tended to have high fat content because of excess vegetable oil added for better taste.Conclusions: Takeaway meals should have lower contents of energy, fat, carbohydrate, and salt and higher contents of whole grains, vegetables, fruits, dairy products, and eggs. Attention should be paid to the high energy density of the fried dish set meal to prevent resultant health problems such as obesity. The transcription factor forkhead box P3 (FOXP3) is a biomarker for regulatory T cells and can also be expressed in cancer cells, but its function in cancer appears to be divergent. The role of hepatocyte-expressed FOXP3 in hepatocellular carcinoma (HCC) is unknown. Here, we collected tumor samples and clinical information from 115 HCC patients and used five human cancer cell lines. We examinedFOXP3mRNAsequences for mutations, used a luciferase assay to assess promoter activities of FOXP3's target genes, and employed mouse tumor models to confirm in vitro results. We detected mutations in the FKH domain of FOXP3 mRNAs in 33% of the HCC tumor tissues, but in none of the adjacent nontumor tissues. None of the mutations occurred at high frequency, indicating that they occurred randomly. Notably, the mutations were not detected in the corresponding regions of FOXP3 genomic DNA, and many of them resulted in amino acid substitutions in the FKH region, altering FOXP3's subcellular localization. FOXP3 delocalization from the nucleus to the cytoplasm caused loss of transcriptional regulation of its target genes, inactivated its tumor-inhibitory capability, and changed cellular responses to histone deacetylase (HDAC) inhibitors. More complexFKHmutations appeared to be associated with worse prognosis in HCC patients. We conclude that mutations in the FKH domain of FOXP3 mRNA frequently occur in HCC and that these mutations are caused by errors in transcription and are not derived from genomicDNAmutations. The study analyzed the influence of random defects on plateau stresses of honeycomb materials with varied relative densities and established a computational model of honeycomb materials considering random defects. The results show that the plateau stress decreases evidently as the random defects increase, which is closely related to the relative density of honeycomb materials. It also set up a functional relationship between relative plateau stresses and random defects as well as that between relative plateau stresses and relative densities. Taken topological structure, random defects and strain rate effect into consideration, and it proposed a dynamic constitutive model of honeycomb materials under low-middle impact loading. And the proposed constitutive model possesses a better applicability to match the stress-strain relationship of honeycomb materials in existing impact experiments. Background: The role of estrogen receptor alpha (ERa), estrogen receptor beta (ERb) and ERa36 signaling in hepatocellular carcinoma (HCC) is not fully addressed. Methods: In this study, three cohorts were included: (i) primary HCC patients (N = 76, cohort P), (ii) colorectal liver metastasis (mCRC) (N = 32, cohort S), and (iii) HCC from The Cancer Genome Atlas (TCGA) (N = 121). The levels of ERa36 and wtER36 were measured and their correlation with clinicopathologic features was determined. Results: WtERa was downregulated and that ERa36 was upregulated in tumor tissues in both cohort P and TCGA data set. ERa36 was downregulated in tumor tissues in cohort S. In cohort P, wtERa was differentially expressed in gender (P < 0.000), age (P = 0.004), tumor number (P = 0.043), tumor size (P = 0.002), intrahepatic recurrence (P = 0.054). ERa36 was unequally expressed in different non-tumor liver status (P = 0.040). WtERa was negatively associated with overall survival (OS) and disease free survival (DFS) in cohort P. Compared with non-tumor tissues, the expression of ERa36 was increased in primary HCC but decreased in secondary HCC, showing opposite expression patterns of ERa36 between primary HCC and secondary HCC. Conclusions: Primary HCC is associated with the decreased WtERa but increased ERa36. The expression pattern of ERa36 is different between primary HCC and secondary HCC, as the former with the increased ERa36 but the latter with the decreased ERa36. Modulated synthesis of MIL-101(Cr) in high yield and with good reproducibility using formic acid as a modulator is reported. Higher molar ratio of formic acid/CrCl3 was found to form better shape-defined MIL-101(Cr) crystals with higher surface area, larger pore volume and better hydrogen uptake performance. The highly crystalline MIL-101(Cr), composed of crystals in the size range of 100-150 nm with multifaceted surface, could be obtained in an optimized molar regime of CrCl3·6H2O/H2BDC/100HCOOH/550H2O at 210 °C for 8 h. The MIL-101(Cr) obtained from the modulated synthesis also showed high thermal and moisture stabilities as well as enhanced hydrogen storage capacity, making this material particularly promising for practical hydrogen storage applications. A type of novel material with a high hydrogen storage capacity was prepared by supporting PdNi18 alloy nanoparticles, which were synthesized by using a new colloid method, on the surface of pretreated multiwal led carbon nanotubes (MWCNTs). The average PdNi18 alloy particle sizes calculated from XRD patterns were ca. 3 nm, and the high dispersion of these particles on MWCNTs was confirmed by TEM image. Hydrogen storage performance of the composite was investigated under moderate pressure (0.1-1.5 MPa) at room temperature, and a maximum storage capacity of ca. 2.3 wt% was achieved under 1.5 MPa at room temperature, which was much higher than that reported previously under the same conditions. Aneuploidy is a key process in tumorigenesis. Dysfunction of the mitotic spindle checkpoint proteins has been implicated as a cause of aneuploidy in cells. We have previously reported that FAT10, a member of the ubiquitin-like modifier family of proteins, is overexpressed in several gastrointestinal and gynecological cancers. Here we show that FAT10 interacts with MAD2, a spindle checkpoint protein, during mitosis. Notably, we show that localization of MAD2 at the kinetochore during the prometaphase stage of the cell cycle was greatly reduced in FAT10-overexpressing cells. Furthermore, compared with parental HCT116 cells, fewer mitotic cells were observed after double thymidine-synchronized FAT10-overexpressing cells were released into nocodazole for more than 4 h. Nonetheless, when these double thymidine-treated cells were released into media, a similar number of G1 parental and FAT10-overexpressing HCT116 cells was observed throughout the 10-h time course. Additionally, more nocodazole-treated FAT10-overexpressing cells escape mitotic controls and are multinucleate compared with parental cells. Significantly, we observed a higher degree of variability in chromosome number in cells overexpressing FAT10. Hence, our data suggest that high levels of FAT10 protein in cells lead to increased mitotic nondisjunction and chromosome instability, and this effect is mediated by an abbreviated mitotic phase and the reduction in the kinetochore localization of MAD2 during the prometaphase stage of the cell cycle. Abstract: A combined quaternary ammonium salt was used as a collector for the flotation of quartz. It shows better selectivity and collectability than dodecyl amine chloride and cetyl trimethylammonium bromide for the flotation of quartz from magnetite and specularite, but the quebracho should be used in the flotation separation of quartz from specularite. By the measurement of zeta potentials, contact angles, and adsorption of the combined quaternary ammonium salt, the results show that the combined quaternary ammonium salt is preferred to adsorb on the surface of quartz, changes its zeta potentials and contact angles, and increases its hydrophobicity. The results of FTIR show that the combined quaternary ammonium salt adsorbs on the quartz surface in physical adsorption with no new products. [Copyright &y& Elsevier]

Various reports have implicated the virally encoded HBx protein as a cofactor in hepatocarcinogenesis. However, direct evidence of the role of HBx as a promoter of oncogenesis in response to an initiating factor such as DNA damage remains inadequate. Here, we report the effects of HBx in HepG2 cells exposed to UV light-induced DNA damage. HBx expression was found not to affect the morphology, viability, and cell cycle/apoptotic profiles or DNA repair machinery of untreated cells. Nonetheless, upon UV treatment, HBx protein levels increased concomitant3y with p53 levels. Both HBx and p53 proteins were found to interact and colocalize primarily in the nucleus. The binding of HBx to p53 modulated (but did not inhibit) the transcriptional activation function of p53. Notably, HBx-expressing cells exhibited increased sensitivity to UV damage, resulting in greater G2/M arrest and apoptosis of these cells. Additionally, these cells displayed a reduced DNA repair capacity in response to UV damage. In conclusion, this work suggests that DNA damage may be an initiating factor in hepatocarcinogenesis and that HBx may act as the promoting factor by inhibiting DNA repair. In hepatitis B virus-infected hepatocytes, a chronic infection may present the opportunity for such a DNA-damaging event to occur, and accumulated errors caused by the inhibition of DNA repair by HBx may result in oncogenesis. Cyr61 is a secreted, cysteine-rich, heparin-binding protein that mediates diverse functions including extracellular matrix formation, differentiation, cell proliferation, adhesion, migration, survival, as well as angiogenesis and tumorigenesis. In this study, we found that Cyr61 gene expression is significantly downregulated in the tumors of hepatocellular carcinoma (HCC) patients. To elucidate its mechanism of gene regulation, we examined the promoter of Cyr61 which contains two long stretches of repeats, each comprising d(CA) dinucleotide repeats downstream of HNF3β- and ATF-binding sites. We hypothesized that the d(CA) repeats may play an important role in regulating Cyr61 promoter activity and performed promoter reporter assays to examine this. We found that a greater number of d(CA) repeats resulted in significantly lower promoter activity of the Cyr61 gene in the KB3-1 and HepG2 cell lines, but not in the MCF-7 cell line. In addition, the d(CA) repeats, but not other random sequences, were found to be important for Cyr61 promoter activity. We further demonstrate that the ATF- and HNF3β-binding sites upstream the d(CA) repeats positively and negatively modulate Cyr61 promoter activity, respectively. Strength of loess that experienced cyclic freeze and thaw is of great significance for evaluating stability of slopes and foundations in loess regions. This paper takes the frequently encountered loess in the Northwestern China as the study object and carried out three kinds of laboratory tests including freeze-thaw test, direct shear test and SEM test to investigate the strength behaviors of loess after cyclic freeze and thaw, and the correlation with meso-level changes in soil structure. Results show that for loess specimens at four dry densities, the cohesion decreases with freeze-thaw cycles until a residual value is reached and thus an exponential equation is proposed. Besides, little change in the angle of internal friction was observed as freeze-thaw proceeds. This may depend on the varying of soil structure, based on which a clue can be found from the surface morphology and mesoscopic scanning of loess specimens. Clearly we observed significant changes in surface morphology of loess and it tends to aggravate at higher water contents or more cycles of freeze and thaw. Moreover, freeze-thaw cycling leads to obvious changes in the meso-structure of loess including lowering the particle aggregates and increasing both the proportion of fine particles and porosity area ratio. The typical US diet has >30% calories from fat; yet, typical laboratory diets contain 17% calories from fat. This disparity could confound the clinical relevance of findings in cerebral ischemia models. We compared outcomes after neonatal brain injury in offspring of rat dams fed standard low-fat chow (17% fat calories) or a higher fat diet (34% fat calories) from day 7 of pregnancy. On postnatal day 7, hypoxic-ischemic injury was induced by right carotid ligation, followed by 60, 75 or 90 min 8% oxygen exposure. Sensorimotor function, brain damage, and serum and brain fatty acid content were compared 1 to 4 weeks later. All lesioned animals developed left forepaw placing deficits; scores were worse in the high-fat groups (p < 0.0001, ANOVA). Similarly, reductions in left forepaw grip strength were more pronounced in the high-fat groups. Severity of right hemisphere damage increased with hypoxia-ischemia duration but did not differ between diet groups. Serum and brain docosahexaenoic acid fatty acid fractions were lower in high-fat progeny (p < 0.05, ANOVA). We speculate that the high-fat diet disrupted docosahexaenoic acid-dependent recovery mechanisms. Like other cancers, aberrant gene regulation features significantly in hepatocellular carcinoma (HCC). MicroRNAs (miRNAs) were recently found to regulate gene expression at the post-transcriptional/translational levels. The expression profiles of 157 miRNAs were examined in 19 HCC patients, and 19 up-regulated and 3 down-regulated miRNAs were found to be associated with HCC. Putative gene targets of these 22 miRNAs were predicted in silico and were significantly enriched in 34 biological pathways, most of which are frequently dysregulated during carcinogenesis. Further characterization of microRNA-224 (miR-224), the most significantly up-regulated miRNA in HCC patients, revealed that miR-224 increases apoptotic cell death as well as proliferation and targets apoptosis inhibitor-5 (API-5) to inhibit API-S transcript expression. Significantly, miR-224 expression was found to be inversely correlated with API-5 expression in HCC patients (p < 0.05). Hence, our findings define a true in vivo target of miR-224 and reaffirm the important role of miRNAs in the dysregulation of cellular processes that may ultimately lead to tumorigenesis.
