How larvae of the many phyla of marine invertebrates find places appropriate for settlement, metamorphosis, growth, and reproduction is an enduring question in marine science. Biofilm-induced metamorphosis has been observed in marine invertebrate larvae from nearly every major marine phylum. Despite the widespread nature of this phenomenon, the mechanism of induction remains poorly understood. The serpulid polychaete Hydroides elegans is a well established model for investigating bacteria-induced larval development. A broad range of biofilm bacterial species elicit larval metamorphosis in H. elegans via at least two mechanisms, including outer membrane vesicles (OMVs) and complexes of phage-tail bacteriocins. We investigated the interaction between larvae of H. elegans and the inductive bacterium Cellulophaga lytica, which produces an abundance of OMVs but not phage-tail bacteriocins. We asked whether the OMVs of C. lytica induce larval settlement due to cell membrane components or through delivery of specific cargo. Employing a biochemical structure–function approach with a strong ecological focus, the cells and OMVs produced by C. lytica were interrogated to determine the class of the inductive compounds. Here, we report that larvae of H. elegans are induced to metamorphose by lipopolysaccharide produced by C. lytica. The widespread prevalence of lipopolysaccharide and its associated taxonomic and structural variability suggest it may be a broadly employed cue for bacterially induced larval settlement of marine invertebrates. In the biogeographical and taxonomical literature before the 1980s there was a wide perception that widespread, often referred to as 'cosmopolitan', species were very common among polychaetes. Here we discuss the origins of this perception, how it became challenged, and our current understanding of marine annelid distributions today. We comment on the presence of widely distributed species in the deep sea and on artificially extended ranges of invasive species that have been dispersed by anthropogenic means. We also suggest the measures needed to revolve the status of species with reported cosmopolitan distributions and stress the value of museum collections and vouchers to be associated with DNA sequences in resolving species distributions. The serpulid tubeworm, Hydroides elegans , is an ecologically and economically important species whose biology has been fairly well studied, especially in the context of larval development and settlement on man-made objects (biofouling). Nevertheless, ontogenetic changes associated with calcareous tube composition and structures have not yet been studied. Here, the ultrastructure and composition of the calcareous tubes built by H. elegans was examined in the three early calcifying juvenile stages and in the adult using XRD, FTIR, ICP-OES, SEM and Raman spectroscopy. Ontogenetic shifts in carbonate mineralogy were observed, for example, juvenile tubes contained more amorphous calcium carbonate and were predominantly aragonitic whereas adult tubes were bimineralic with considerably more calcite. The mineral composition gradually shifted during the tube development as shown by a decrease in Sr/Ca and an increase of Mg/Ca ratios with the tubeworm’s age. The inner tube layer contained calcite, whereas the outer layer contained aragonite. Similarly, the tube complexity in terms of ultrastructure was associated with development. The sequential appearance of unoriented ultrastructures followed by oriented ultrastructures may reflect the evolutionary history of serpulid tube biominerals. As aragonitic structures are more susceptible to dissolution under ocean acidification (OA) conditions but are more difficult to be removed by anti-fouling treatments, the early developmental stages of the tubeworms may be vulnerable to OA but act as the important target for biofouling control. Earlier studies have shown that biofilms can mediate the larval settlement of the polychaete Hydroides elegans and that changes in the bacterial community structure and density of biofilms often alter the larval settlement response. However, the chemical cues that mediate this response remain unknown. In this study, both successional changes in the bacterial community structure and the chemical profiles of subtidal biofilms are described and related to the larval settlement response. Multispecies biofilms were developed on polystyrene Petri dishes and granite rock in the subtidal zone over a period of 20 days. The effects of the substratum and age on the bacterial community structure and chemical profiles of the biofilms were evaluated with two molecular methods (microarray (PhyloChip) and denaturing gradient gel electrophoresis) and with gas chromatography–mass spectrometry, respectively. Both age and substratum altered the bacterial community structures and chemical profiles of the biofilms. Age had a greater effect in shaping the bacterial community structure than did the substratum. In contrast, the type of substratum more strongly affected the chemical profile. Extracts of biofilms of different ages, which developed on different substrata, were tested for the settlement of H. elegans larvae. The extracts induced larval settlement in a biofilm-age-dependent manner, and extracts originating from different substrata of the same age showed no differences in larval settlement. A toxicity test was developed to examine the effects of heavy metal contaminants on the early life stages of the marine polychaete. We have studied the effects of metals on fertilization and early development of marine polychaete Hydroides elegans. These heavy metals have often been found in polluted ground and water near industrial discharges, and have therefore been detected from time to time in the food chain. They have been reported to alter various reproduction functions in various animals including marine populations. The toxic effect of mercury, cadmium, lead, nickel and zinc on sperm viability, fertilization, embryogenesis and larvae of H. elegans was examined. We observed that the rate of fertilization decreased when the sperm was incubated with heavy metals. Treatment of eggs with each metal did not prevent fertilization, but delayed or blocked the first mitotic divisions, and altered early embryonic development. All these effects were observed at relatively high concentrations. However, bio-accumulation in sediments and aquatic organisms have been reported. Polychaete eggs may then be in contact with very high concentrations of these heavy metals in areas where these metals are not handled or stocked properly, and then develop into abnormal embryos. In addition to bivalves and sea-urchins, polychaete embryos can provide biological criteria for seawater quality standards taking into account the sensitivity of the invertebrates and their contribution in detection of harmful chemicals with no marked effect on the species. Our results indicate that the early development of H. elegans is highly sensitive to heavy metals and this polychaete can be routinely employed as a test organism for ecotoxicity bioassays in tropical and subtropical regions. Abstract: This study investigated the potential role of a fungal metabolite isolated from a sponge surface-associated fungus in a biofouling control. Succinic acid with both antibacterial and anti-larval settlement activity was isolated from the fungus associated with the sponge Acanthella cavernosa, using bioassay-guided isolation and purification procedure. Succinic acid was then embedded in hydrogel discs that were subsequently exposed to natural seawater for 24, 48 and 72 h to allow the development of biofilms on the disc surfaces. Both epifluorescence microscopy and DNA fingerprinting analysis were used for the analysis of cell density and species diversity of bacterial communities in the biofilms, respectively. The results indicated that succinic acid substantially altered the cell density and species diversity of the bacterial communities in the biofilms on hydrogel surfaces in comparison to the control hydrogel discs (without succinic acid). Laboratory larval-settlement bioassay with larvae of the fouling tubeworm Hydroides elegans showed that fewer larvae settled on the surface of hydrogel discs covered with the biofilms that had been altered by succinic acid than on the surface of the control discs. Field experiments using succinic acid-coated panels further confirmed the antifouling activity of the compound. It has been suggested that bacteria associated with soft-bodied organisms are suggested to produce bioactive compounds against the attachment of invertebrate larvae and bacteria onto the surface of these organisms. Our recent study has demonstrated that epibiotic bacteria from the surface of the soft coral Dendronephthya sp. (Coelenterata: Octocoralia, Alcyonacea) inhibit the growth of bacteria commonly found in marine natural biofilms. In the present study, the effect of 11 epibiotic bacteria isolated from the surface of Dendronephthya sp. on larval settlement of the tubeworms Hydroides elegans was examined using laboratory bioassay. Among 11 bacterial isolates, 2 strains (18%) inhibited the larval settlement of H. elegans (Haswell), 4 strains (36%) were “inductive” to larvae and the remaining 5 strains (46%) were “non-inductive”. There was no correlation between the antifouling activities of bacterial isolates and their phylogenetic origin, i.e. closely related bacterial strains showed different effects on larval settlement of H. elegans. When all “inductive”, “non-inductive” and “inhibitive” bacterial isolates were mixed in a 1:1:1 ratio, the effect of the resultant multispecies film on larval settlement became “inhibitive”. Waterborne compounds of Vibrio sp. and an unidentified α-Proteobacterium, which suppressed the settlement of H. elegans and Bugula neritina (L.) larvae, were further investigated using size fractionation and bioassay-guided enzymatic analysis. It was found that antilarval settlement compounds from these bacteria were heat-stable polysaccharides with a molecular weight >100 kDa. The results indicate that the bacteria associated with the soft coral Dendronephthya sp. may contribute to the antifouling mechanisms of the soft-bodied organisms by producing compounds that are against bacterial growth and settlement of macrofoulers on the surface of their host. Larvae of the biofouling tubeworm Hydroides elegans (Polychaeta) must contact a bacterial biofilm to induce settlement. To further understand the relationship between particular bacteria and the worm, this study analyzed associated microbiomes at multiple life-history stages with high-throughput 16S rRNA amplicon sequencing. Worms were collected from Pearl Harbor Hawai'i (21°21′27.2″N 157°57′34.8″W), and microbial communities were analyzed from adults, their tubes, spawned eggs, 6-day old nectochaete larvae, and filtered seawater from the larval cultures that included cells of the microalga Isochrysis galbana fed to the larvae. Microbiomes of females sampled at two different times were significantly different: Cellvibrionales dominated the microbiome of adult females in August 2016, whereas Oceanospirillales were dominant in females collected in April 2017. Additionally, in 2017, the microbiomes across all life stages were significantly different from each other; only one OTU in the genus Endozoicomonas was shared between females and their nectochaetes at a relative abundance > 1%. Because of the apparent absence of a consistent microbiome between stages, female worms, their eggs and both trochophore and nectochaete larvae were stained to reveal the presence of bacteria and analyzed with scanning confocal microscopy. The eggs were almost devoid of bacteria, and bacteria seen on larvae were almost all confined to the feeding apparatus. The evidence of inconsistent microbiomes across life stages and lack of bacteria on eggs demonstrates that H. elegans lacks a vertically transmitted microbiome, and bacteria found at each stage came from a varying environment.
