Aims: To establish which components of energy balance mediate the clinically significant weight loss demonstrated with use of cotadutide, a glucagon‐like peptide‐1 (GLP‐1)/glucagon receptor dual agonist, in early‐phase studies. Materials and Methods: We conducted a phase 2a, single‐centre, randomized, placebo‐controlled trial in overweight and obese adults with type 2 diabetes. Following a 16‐day single‐blind placebo run‐in, participants were randomized 2:1 to double‐blind 42‐day subcutaneous treatment with cotadutide (100–300 μg daily) or placebo. The primary outcome was percentage weight change. Secondary outcomes included change in energy intake (EI) and energy expenditure (EE). Results: A total of 12 participants (63%) in the cotadutide group and seven (78%) in the placebo group completed the study. The mean (90% confidence interval [CI]) weight change was −4.0% (−4.9%, −3.1%) and −1.4% (−2.7%, −0.1%) for the cotadutide and placebo groups, respectively (p = 0.011). EI was lower with cotadutide versus placebo (−41.3% [−66.7, −15.9]; p = 0.011). Difference in EE (per kJ/kg lean body mass) for cotadutide versus placebo was 1.0% (90% CI −8.4, 10.4; p = 0.784), assessed by doubly labelled water, and −6.5% (90% CI −9.3, −3.7; p < 0.001), assessed by indirect calorimetry. Aim: Evidence from mouse models suggests that brain serotonergic pathways control blood glucose. We hypothesized that sumatriptan (5HT1B‐receptor agonist) would alter glucose homeostasis in humans. Materials and Methods: We conducted a two‐visit random‐order double‐blinded placebo‐controlled cross‐over trial in 10 overweight adults that were otherwise healthy. Participants received sumatriptan (single dose, 100 mg) or placebo before undergoing a 60‐min intravenous glucose tolerance test, followed by a 120‐min hyperinsulinaemic euglycaemic clamp. Results: Glucose excursion was greater during intravenous glucose tolerance test with sumatriptan compared with placebo [iAUC0‐60 min 316 (268‐333) vs. 251 (197‐319) min/mmol/L p =.047]. This was probably explained by a combination of reduced circulating insulin levels [iAUC0‐10 min 1626 (1103‐2733) vs. 2336 (1702‐3269) min/pmol/L, p =.005], reduced insulin sensitivity [M/I‐value 2.11 (1.15, 4.05) vs. 3.03 (1.14, 4.90) mg/kg/min per pmol/L, p =.010] and glucose effectiveness [SG 0.17 (0.12, 0.21) vs. 0.22 (0.18, 0.65)/min, p =.027]. This work challenges the widely accepted model of sensory gating as a preattention inhibitory process by investigating whether attention directed at the second tone (S2) within a paired‐click paradigm could affect gating at the cortical level. We utilized magnetoencephalography, magnetic resonance imaging and spatio‐temporal source localization to compare the cortical dynamics underlying gating responses across two conditions (passive and attention) in 19 healthy subjects. Source localization results reaffirmed the existence of a fast processing pathway between the prefrontal cortex (PFC) and bilateral superior temporal gyri (STG) that underlies the auditory gating process. STG source dynamics comprised two gating sub‐components, Mb1 and Mb2, both of which showed significant gating suppression (>51%). The attention directed to the S2 tone changed the gating network topology by switching the prefrontal generator from a dorsolateral location, which was active in the passive condition (18/19), to a medial location, active in the attention condition (19/19). Enhanced responses to the attended stimulus caused a significant reduction in gating suppression in both STG gating components (>50%). Our results demonstrate that attention not only modulates sensory gating dynamics, but also exerts topological rerouting of information processing within the PFC. The present data, suggesting that the cortical levels of early sensory processing are subject to top‐down influences, change the current view of gating as a purely automatic bottom‐up process. Magnetoencephalography (MEG), a direct measure of neuronal activity, is an underexplored tool in the search for biomarkers of Alzheimer's disease (AD). In this study, we used MEG source estimates of auditory gating generators, nonlinear correlations with neuropsychological results, and multivariate analyses to examine the sensitivity and specificity of gating topology modulation to detect AD. Our results demonstrated the use of MEG localization of a medial prefrontal (mPFC) gating generator as a discrete (binary) detector of AD at the individual level and resulted in recategorizing the participant categories in: (1) controls with mPFC generator localized in response to both the standard and deviant tones; (2) a possible preclinical stage of AD participants (a lower functioning group of controls) in which mPFC activation was localized to the deviant tone only; and (3) symptomatic AD in which mPFC activation was not localized to either the deviant or standard tones. This approach showed a large effect size (0.9) and high accuracy, sensitivity, and specificity (100%) in identifying symptomatic AD patients within a limited research sample. The present results demonstrate high potential of mPFC activation as a noninvasive biomarker of AD pathology during putative preclinical and clinical stages. Travertine deposition is a landscape-forming process, usually building a series of calcareous barriers differentiating the river flow into a series of cascades and ponds. The process of carbonate precipitation is a complex relationship between biogenic and abiotic causative agents, involving adapted microbial assemblages but also requiring high levels of carbonate saturation, spontaneous degassing of carbon dioxide and slightly alkaline pH. We have analysed calcareous crusts and water chemistry from four sampling sites along the Hoyoux River and its Triffoy tributary (Belgium) in winter, spring, summer and autumn 2014. Different surface textures of travertine deposits correlated with particular microenvironments and were influenced by the local water flow. In all microenvironments, we have identified the cyanobacterium Phormidium incrustatum (Nägeli) Gomont as the organism primarily responsible for carbonate precipitation and travertine fabric by combining morphological analysis with molecular sequencing (16S rRNA gene and ITS, the Internal Transcribed Spacer fragments), targeting both field populations and cultures to exclude opportunistic microorganisms responding favourably to culture conditions. Several closely related cyanobacterial strains were cultured; however, only one proved identical with the sequences obtained from the field population by direct PCR. This strain was the dominant primary producer in the calcareous deposits under study and in similar streams in Europe. The dominance of one organism that had a demonstrated association with carbonate precipitation presented a valuable opportunity to study its function in construction, preservation and fossilisation potential of ambient temperature travertine deposits. A review of the article "Automated insulin dosing guidance to optimize insulin management in patients with type 2 diabetes: a multicentre, randomised controlled trial," by Richard M. An important personnel management challenge is to explore factors that stimulate or hinder the development of individual work ability and quality of life throughout a career. Design. A cross-sectional study. Methods. The study was performed during 2007-2008 in six randomly selected hospitals in Croatia. The self-administered questionnaires included the Work Ability Index (WAI) developed by the Finnish Institute of Occupational Health, the Quality of Life questionnaire (WHOQL-BREF) developed by the World Health Organization and additional socio-demographic questions. A total number of 1212 nurses completed the questionnaires, giving a response rate of 67.3%. Binary logistic regression was performed to assess how socio-demographic characteristics and work ability groups predict each of the WHOQL-BREF domains. Results. Having a satisfactory WAI score (WAI ≥ 37) was significantly the most important predictor for all quality of life domains, with the odds ratios (OR) being as follows: OR = 6.8 (95% CI: 4.8-9.6) for the physical domain, OR = 2.3 (95% CI: 1.7-3.1) for the psychological domain, OR = 1.7 (95% CI: 1.3-2.4) for the social relationship domain and OR = 1.7 (95% CI: 1.3-2.3) for the environmental domain. Conclusions. Satisfactory work ability was a major quality of life determinant in all WHOQL-BREF domains with the highest odds ratio for the physical domain. Maintaining clinical nurses' work ability is an important issue, because it is foundational for the quality of life of the workforce. Relevance to clinical practice. Our study provides quantified estimates of the extent to which a satisfactory WAI score predicts a better score in physical, psychosocial, social relationships and environmental domain of nurses' quality of life. Extracellular UV-screening pigments gloeocapsin and scytonemin present in the exopolysaccharide (EPS) envelopes of extremophilic cyanobacteria of freshwater and marine environments were studied by Raman spectroscopy and compared to their intracellular photosynthetic pigments. This Raman spectral analysis of the extracellular pigment gloeocapsin showed that it shared Raman spectral signatures with parietin, a radiation-protective pigment known in lichens. The UV-light spectra also show similarities. Gloeocapsin occurs in some cyanobacterial species, mostly with exclusion of scytonemin, indicating that these pigments have evolved in cyanobacteria as separate protective strategies. Both gloeocapsin and scytonemin are widely and species-specifically distributed in different cyanobacterial genera and families. The widespread occurrence of these pigments may suggest an early origin, while their detection by Raman spectroscopy makes them potential biosignatures for cyanobacteria in the fossil record and demonstrates the usefulness of nondestructive Raman spectroscopy analyses for the search for complex organics, including possible photosynthetic pigments, if preservable in early Earth and extraterrestrial samples. Abstract: The amplitude variability of the M50 component of neuromagnetic responses is commonly used to explore the brain's ability to modulate its response to incoming repetitive or novel auditory stimuli, a process conceptualized as a gating mechanism. The goal of this study was to identify the spatial and temporal characteristics of the cortical sources underlying the M50 network evoked by tones in a passive oddball paradigm. Twenty elderly subjects [10 patients diagnosed with mild cognitive impairment (MCI) or probable Alzheimer disease (AD) and 10 age-matched controls] were examined using magnetoencephalographic (MEG) recordings and the multi-dipole Calibrated Start Spatio-Temporal (CSST) source localization method. We identified three cortical regions underlying the M50 network: prefrontal cortex (PF) in addition to bilateral activation of the superior temporal gyrus (STG). The cortical dynamics of the PF source within the 30–100ms post-stimulus interval was characterized and was found to be comprised of two subcomponents, Mb1c and Mb2c. The PF source was localized for 10/10 healthy subjects, whereas 9/10 MCI/AD patients were lacking the PF source for both tone conditions. The selective activation of the PF source in healthy controls along with the inactivation of the PF region for MCI/AD patients, enabled us to examine the dynamics of this network of activity when it was functional and dysfunctional, respectively. We found significantly enhanced activity of the STG sources in response to both tone conditions for all subjects who lacked a PF source. The reported results provide novel insights into the topology and neurodynamics of the M50 auditory network, which suggest an inhibitory role of the PF source that normally suppresses activity of the STG sources. Background Detailed assessment of physical activity (PA) in older adults is required to comprehensively describe habitual PA-levels in this growing population segment. Current evidence of population PA-levels is predominantly based on self-report. Methods We examined PA and sedentary behaviour in a nationally representative sample of British people aged 60-64, using individually-calibrated combined heart-rate and movement sensing and a validated questionnaire (EPAQ2), and the socio-demographic and behavioural factors that may explain between-individual variation in PA. Results Between 2006-2010, 2224 participants completed EPAQ2 capturing the past year's activity in four domains (leisure, work, transportation and domestic life) and 1787 participants provided 2-5 days of combined-sensing data. According to objective estimates, median(IQR) physical activity energy expenditure (PAEE) was 33.5 (25.3-42.2) and 35.5 (26.6- 47.3) kJ/kg/day for women and men, respectively. Median (IQR) time spent in moderate-to-vigorous PA (MVPA; >3MET), light-intensity PA (1.5-3MET) and sedentary (<1.5MET) was 26.0 (12.3-48.1) min/day, 5.4 (4.2-6.7) h/day and 18.0 (16.6-19.4) h/day, respectively, in women; and 41.0 (18.8-73.0) min/day, 5.2 (4.0-6.5) h/day and 17.9 (16.3-19.4) h/day in men. PAEE and time spent in MVPA were lower and sedentary time was greater in obese individuals, those with poor health, and those with lower educational attainment (women only). Questionnaire-derived PAEE and MVPA tended to have similar patterns of variation across socio-demographic strata. In the whole sample, domestic PA had the greatest relative contribution to total questionnaire-derived PAEE (58%), whereas occupational PA was the main driver among employed participants (54%). Only 2.2% of participants achieved an average of >30 min MVPA per day combined with >60 min strength-training per week. Conclusions The use of both self-report and objective monitoring to assess PA in early old age provides important information on the domains of PA, PAEE and time spent at different intensity levels. Our findings suggest PA levels are generally low and observed patterns of variation indicate specific subgroups who might benefit from targeted interventions to increase PA. [ABSTRACT FROM AUTHOR]

A complex microboring trace of fungal affinity is described in shells as a new ichnotaxonSaccomorpha stereodiktyonisp. nov. and compared with the earlier established ichnotaxonSaccomorpha terminalisRadtke, 1991. The new trace is characterized by a three-dimensional network of tunnels composed of a bifurcate horizontal (parallel to substrate surface) network with an upright (perpendicular to surface) system of tunnels and by the formation of cylindrical to multilobate terminal sporangial swellings. The new trace shares withSaccomorpha terminalisRadtke, 1991 the terminal position of sporangial swellings but differs from this ichnotaxon by its complexity in spatial arrangement, segmented construction, and ramification of tunnels. The horizontal parts of the network in the new taxon adhere to the substrate surface and regularly produce thinner tunnels that explore the interior of the substrate, allowing the producer to participate in digestion of organic lamellae incorporated in the shell. Microborings similar to the new trace fossil have been observed in modern bivalve shells of the Atlantic Ocean, North Sea, Adriatic Sea and Red Sea at depths ranging from the intertidal down to 1,550 m. The fossil record of the trace reaches back to the Jurassic and the type material stems from a Lower Oligocene oyster shell. Objective: To examine the validity of the Recent Physical Activity Questionnaire (RPAQ) which assesses physical activity (PA) in 4 domains (leisure, work, commuting, home) during past month. Methods: 580 men and 1343 women from 10 European countries attended 2 visits at which PA energy expenditure (PAEE), time at moderate-to-vigorous PA (MVPA) and sedentary time were measured using individually-calibrated combined heart-rate and movement sensing. At the second visit, RPAQ was administered electronically. Validity was assessed using agreement analysis. Results: RPAQ significantly underestimated PAEE in women [median(IQR) 34.1 (22.1, 52.2) vs. 40.6 (32.4, 50.9) kJ/kg/day, 95%LoA: −44.4, 63.4 kJ/kg/day) and in men (43.7 (29.0, 69.0) vs. 45.5 (34.1, 57.6) kJ/kg/day, 95%LoA: −47.2, 101.3 kJ/kg/day]. Using individualised definition of 1MET, RPAQ significantly underestimated MVPA in women [median(IQR): 62.1 (29.4, 124.3) vs. 73.6 (47.8, 107.2) min/day, 95%LoA: −130.5, 305.3 min/day] and men [82.7 (38.8, 185.6) vs. 83.3 (55.1, 125.0) min/day, 95%LoA: −136.4, 400.1 min/day]. Correlations (95%CI) between subjective and objective estimates were statistically significant [PAEE: women, rho = 0.20 (0.15–0.26); men, rho = 0.37 (0.30–0.44); MVPA: women, rho = 0.18 (0.13–0.23); men, rho = 0.31 (0.24–0.39)]. When using non-individualised definition of 1MET (3.5 mlO2/kg/min), MVPA was substantially overestimated (∼30 min/day). Revisiting occupational intensity assumptions in questionnaire estimation algorithms with occupational group-level empirical distributions reduced median PAEE-bias in manual (25.1 kJ/kg/day vs. −9.0 kJ/kg/day, p<0.001) and heavy manual workers (64.1 vs. −4.6 kJ/kg/day, p<0.001) in an independent hold-out sample. Conclusion: Relative validity of RPAQ-derived PAEE and MVPA is comparable to previous studies but underestimation of PAEE is smaller. Objectives: To compare physical activity (PA) subcomponents from EPIC Physical Activity Questionnaire (EPAQ2) and combined heart rate and movement sensing in older adults. Methods: Participants aged 60–64y from the MRC National Survey of Health and Development in Great Britain completed EPAQ2, which assesses self-report PA in 4 domains (leisure time, occupation, transportation and domestic life) during the past year and wore a combined sensor for 5 consecutive days. Estimates of PA energy expenditure (PAEE), sedentary behaviour, light (LPA) and moderate-to-vigorous PA (MVPA) were obtained from EPAQ2 and combined sensing and compared. Complete data were available in 1689 participants (52% women). Results: EPAQ2 estimates of PAEE and MVPA were higher than objective estimates and sedentary time and LPA estimates were lower [bias (95% limits of agreement) in men and women were 32.3 (−61.5 to 122.6) and 29.0 (−39.2 to 94.6) kJ/kg/day for PAEE; −4.6 (−10.6 to 1.3) and −6.0 (−10.9 to −1.0) h/day for sedentary time; −171.8 (−454.5 to 110.8) and −60.4 (−367.5 to 246.6) min/day for LPA; 91.1 (−159.5 to 341.8) and 55.4 (−117.2 to 228.0) min/day for MVPA]. There were significant positive correlations between all self-reported and objectively assessed PA subcomponents (rho  = 0.12 to 0.36); the strongest were observed for MVPA (rho = 0.30 men; rho = 0.36 women) and PAEE (rho = 0.26 men; rho = 0.25 women). Conclusion: EPAQ2 produces higher estimates of PAEE and MVPA and lower estimates of sedentary and LPA than objective assessment. Objective:To investigate the relationship of body weight and its changes over time with physical activity (PA).Design:Population-based prospective cohort study (Norfolk cohort of the European Prospective Investigation into Cancer and Nutrition, EPIC-Norfolk, United Kingdom).Subjects:A total of 25 639 men and women aged 39-79 years at baseline. PA was self-reported. Weight and height were measured by standard clinical procedures at baseline and self-reported at 18-month and 10-year follow-ups (calibrated against clinical measures). Main outcome measure was PA at the 10-year follow-up.Results:Body weight and PA were inversely associated in cross-sectional analyses. In longitudinal analyses, an increase in weight was associated with higher risk of being inactive 10 years later, after adjusting for baseline activity, 18-month activity, sex, baseline age, prevalent diseases, socioeconomic status, education, smoking, total daily energy intake and alcohol intake. Compared with stable weight, a gain in weight of >2 kg per year in the short-, medium- and long-term was consistently and significantly associated with greater likelihood of physical inactivity after 10 years, with the most pronounced effect for long-term weight gain, OR=1.89 (95% CI: 1.30-2.70) in fully adjusted analysis. Weight gain of 0.5-2 kg per year over long-term was substantially associated with physical inactivity after full adjustment, OR=1.26 (95% CI: 1.11-1.41).Conclusion:Weight gain (during short-, medium- and long-term) is a significant determinant of future physical inactivity independent of baseline weight and activity. Compared with maintaining weight, moderate (0.5-2 kg per year) and large weight gain (>2 kg per year) significantly predict future inactivity; a potentially vicious cycle including further weight gain, obesity and complications associated with a sedentary lifestyle. We report on the molecular evidence that Dermacentor reticulatus ticks in Croatia are infected with Rickettsia helvetica (10%) or Rickettsia slovaca (2%) or co-infected with both species (1%). Kołodziej, B., Golubic, S., Bucur, I.I., Radtke, G. & Tribollet, A. 2011: Early Cretaceous record of microboring organisms in skeletons of growing corals. Lethaia, Vol. 45, pp. 34-45. A spectacularly preserved assemblage of microbial euendoliths, penetrating into skeletons of growing scleractinian corals, has been recognized in Early Aptian (Early Cretaceous) reef limestone of the Rarău Mountains (East Carpathians, NE Romania). Microboring euendolithic filaments were found in five coral colonies of the suborder Microsolenina. They remained in part well-preserved, often impregnated with iron oxides, which made them visible even in strongly recrystallized parts of coral skeletons. Filaments of a wide range of sizes (2-40 μm in diameter) were concentrated within medium parts of coral septa, oriented along the septa in the direction of the coral growth. The larger filaments were tubular, occurring in bundles and branched into finer, often tapering branches. Their behaviour and organization were quite similar to the modern euendolithic siphonalean chlorophyte Ostreobium. Filament diameters exceeded those reported for the modern species, but covered a similarly wide size range. Narrower frequently branching filaments, 4-8 μm in diameter, resemble distal branching patterns of modern Ostreobium quekettii. Some very thin filaments (ca. 1-2 μm) observed within skeleton or inside the large tubular filaments, sometimes associated with globular swellings, may represent euendolithic fungi. While the relationship between sensory stimulation and tasks and the size of the cortical activations is generally unknown, the visual modality offers a unique possibility of an experimental manipulation of stimulus size-related increases of the spatial extent of cortical activation even during the earliest activity in the retinotopically organized primary visual cortex. We used magnetoecephalography (MEG), visual stimuli of increasing size, and numerical simulations on realistic cortical surfaces to explore the effects of increasing spatial extent of the activated cortical sources on the neuromagnetic fields, location estimation biases, and source resolution. Source localization was performed assuming multiple dipoles in a sphere model using an efficient, automatically restarted multi-start simplex minimizer within the Calibrated Start Spatio-Temporal (CSST) algorithm. We found size-related effects on amplitude and latencies and differences in relative locations of the earliest occipital sources evoked by stimuli of increasing size presented at the same eccentricity. This finding was confirmed by single patch simulations. Additionally, simulations of multiple extended sources demonstrated size-related increase in limits in source resolution for bilaterally simulated sources, biases in location estimates for a given separation of sources, and limits in source resolution due to source multiplicity within a hemisphere. Abstract: Toxic cyanobacterial blooms have been a primary concern predominantly in the plankton of freshwater bodies. Recently, however, the toxicity of benthic cyanobacteria is increasingly attracting attention of the scientific community and environmental agencies. The occurrence of toxic strains in benthic cyanobacteria is intimately linked to our understanding of the diversity and ecological responses of these organisms under field conditions. To that effect, we are engaged in combined morphotypic and genotypic characterization (polyphasic) of benthic natural populations of cyanobacteria in tropical lagoons and coral reefs, with the objective to provide a reliable reference for further comparative work. The methods of identification based on phenotypic properties and those based on molecular tools for genotypic identification are correlated. The approach is based on identifying the occurrences of cyanobacterial benthic blooms, tested for purity and analyzed by application of molecular tools. The questions addressed include the distinction between marine and freshwater taxa, between populations in geographically separate regions as well as between their potential vs. expressed toxicity. Aim. This paper is a report of a study conducted to determine which occupational stressors are present in nurses' working environment; to describe and compare occupational stress between two educational groups of nurses; to estimate which stressors and to what extent predict nurses' work ability; and to determine if educational level predicts nurses' work ability. Background. Nurses' occupational stress adversely affects their health and nursing quality. Higher educational level has been shown to have positive effects on the preservation of good work ability. Method. A cross-sectional study was conducted in 2006--2007. Questionnaires were distributed to a convenience sample of 1392 (59%) nurses employed at four university hospitals in Croatia (n = 2364). The response rate was 78% (n = 1086). Data were collected using the Occupational Stress Assessment Questionnaire and Work Ability Index Questionnaire. Findings. We identified six major groups of occupational stressors: 'Organization of work and financial issues', 'public criticism', 'hazards at workplace', 'interpersonal conflicts at workplace', 'shift work' and 'professional and intellectual demands'. Nurses with secondary school qualifications perceived Hazards at workplace and Shift work as statistically significantly more stressful than nurses a with college degree. Predictors statistically significantly related with low work ability were: Organization of work and financial issues (odds ratio = 1.69, 95% confidence interval 1.22--2.36), lower educational level (odds ratio = 1.69, 95% confidence interval 1.22--2.36) and older age (odds ratio = 1.07, 95% confidence interval 1.05--1.09). Conclusion. Hospital managers should develop strategies to address and improve the quality of working conditions for nurses in Croatian hospitals. Stress at work is a specific type of stress arising from the work environment. The individual's estimate of the objective situation or event greatly affects the level of stress in the individual's response. It was the aim of the study to test the sample of hospital healthcare workers on the stressors at work which they find stressful and/or greatly stressful and to establish whether there are differences in the experience of stress between physicians and nurses/lab technicians. The study was based on a sample of 1,900 participants. Of the total number of participants, 1,477 healthcare workers completed the questionnaire and the response rate was 78%. Results indicated that hospital healthcare workers experience great intensity of stress from several stressors. Among the most common for both groups were the financial and organisational factors. Physicians, as opposed to nurses and technicians, experience some of these stressors with greater intensity. At the top of the scale for most physicians are the stressors related to financial aspects of their work, organisation and inadequate workspace, whereas nurses and technicians listed inadequate (insufficient) number of workers as a stressor alongside the inappropriate financial remuneration. Nurses and technicians, more frequently than physicians, experience as stressful the fear of specific work-related dangers and harms in healthcare, indicating a need to improve education and safety measures at work. On the other hand, physicians find stressful the exposure to unjust public criticism and, much more so, the psychological pressure from the patients poorly informed and labouring under unrealistic expectations, indicating a need to improve physician-patient communication. Background and Objectives We prospectively evaluated the infusion-related toxicity of autologous peripheral blood progenitor cells (PBPC) in 215 patients with haematologic malignancies or solid tumours. Materials and Methods PBPCs were collected by apheresis after mobilization with chemotherapy and/or granulocyte–colony-stimulating factor (G-CSF). The grafts were cryopreserved in 10% dimethyl sulfoxide (DMSO) and stored in liquid nitrogen. Patients were monitored for vital signs and symptoms of the toxicity during and after infusion. Results The adverse reactions were reported during 149 (56·9%) infusions. During 21·0% infusions occurred just one symptom classified as grade 1, while during 35·9% occurred multiple symptoms classified as grade 2. Logistic regression analysis showed that female gender, diagnosis of multiple myeloma and number of granulocytes infused per kg body weight were significant predictors of occurrence of adverse reactions during infusion. Conclusion Our results indicate that beside the infused DMSO dose, the composition of graft as well as patient's diagnosis are also very important factors for infusion-related toxicity. Cyanobacterial populations, the primary producers and builders of 'kopara' microbial mats were studied in four selected ponds along the rim of the Rangiroa Atoll, French Polynesia using a polyphasic approach. Seven isolates were maintained in uni-cyanobacterial cultures, characterized morphotypically and phylogenetically by evaluating sequences of the 16S rRNA gene of about 620 base pairs in length. Cyanobacteria in natural populations were analyzed microscopically, characterized morphotypically, and compared with cultured strains. Three of the isolates were identified in the field samples: Lyngbya aestuarii, Johannesbaptistia pellucida and Chroococcus submarinus were present in the mats only as minor components, whereas the species of Schizothrix that dominated the mat community could not be cultured. The sequence of Johannesbaptistia pellucida is published for the first time. The phylogenetic and taxonomic relations are discussed on the basis of a reconstructed phylogenetic tree in relation to morphotypic characters. Sequences of Kopara isolates plot separately from those cultured from the lagoon of the neighbouring atoll Tikehau, indicating a narrow niche differentiation of benthic cyanobacterial taxa. Six cyanobacterial isolates recovered from Polynesian microbial mats, called “kopara,” were cultured using laboratory-closed photobioreactors and were shown to produce exopolymers as released and capsular exopolysaccharides (EPS). These polymers have been chemically characterized using colorimetric and elemental assays, infrared spectrometry, and gas chromatography. Both capsular and released EPS consisted of 7 to 10 different monosaccharides with neutral sugars predominating. Interestingly, four isolates exhibited sulfate contents ranging from 6% to 19%. On the basis of preliminary data, cyanobacteria from this unusual ecosystem appear to be an important source of novel EPS of a great interest in terms of their biological activities. Fungi are an important constituent of microbial endolithic assemblages in marine ecosystems. As euendoliths, they penetrate limestone, mollusk shells and other carbonate substrates, where they can exploit mineralized organic matter, attack their hosts, or engage in symbiotic relationships. They leave specific boring traces, which can be identified in the fossil record and described as trace fossils. Their distribution is independent of light and extends from the intertidal ranges to abyssal oceanic depths. Important, but insufficiently studied, is the role of aggressive endolithic fungi in skeletons of corals where they are ubiquitous and globally distributed. In healthy growing reef corals, the relationship between the coral coelenterate, endolithic algae and fungi is in a state of equilibrium, but can turn detrimental to coral health when reefs are exposed to environmental stress. Bacterial contamination of blood products causes significant patient morbidity and mortality. Contaminated platelet transfusion is a frequent cause of bacteraemia and sepsis because of the storage conditions of platelets. A fatal case of Morganella morganii platelet transfusion associated with sepsis is described, along with procedures traced back to the isolation of M. morganii from a donor's stool. Molecular typing was performed, and the same M. morganii strain was found in blood and post-mortem organ cultures of platelet recipient and platelet bag and in the donor's stool. The route of contamination is unknown. The contamination could be due to either insufficient venipuncture site disinfection or the donor's transient bacteraemia. Natural populations of benthic cyanobacteria in the lagoon of Tikehau Atoll in French Polynesia were studied using a polyphasic approach that combined field observations, LM, culturing, and molecular sequencing. The approach assessed their phenotypic (morphotypic and ecological) and genotypic diversity. Partial sequences (approximately 450 bp long) of the PCR-amplified 16S rRNA gene were obtained from both natural and cultured populations using cyanobacteria-specific primers. The sequences obtained clustered into six separate phylogenetic clusters in relation to the complete set of 16S rRNA sequences available in public databases. Phylogenetic clustering correlated in many cases with some morphological characteristics. For example, Spirulina subsalsa could be identified to the morphospecies level using both molecular and microscopic approaches. Morphotypes identified as Symploca hydnoides (Kütz. ex Gomont) TK22, Phormidium sp. TK1, and P. laysanense (Lemmerman) TK14 formed a distinct cluster. The morphogenus Hydrocoleum (Blennothrix) clustered interestingly close to the morphologically similar, but planktonic, marine cyanobacterium Trichodesmium , suggesting a relationship. Other sequences belonging to morphotaxa with very narrow trichomes were found to be polyphyletic. Enrichment cultures, with inoculum obtained from field populations, were followed over a period of 18 months. Dominance in all cultures shifted over time in favor of a set of cyanobacterial strains with narrow trichomes, which were phenotypically and phylogenetically different from natural populations dominating the original samples. Sequences from strains enriched by cultivation clustered into two well-defined phylogenetic groups, possibly identifying new taxa. BackgroundMethodsResultsConclusionWhile numerous studies have explored treatment outcomes for the overall ACC patient cohort, data on the subpopulation of patients with recurrent disease are limited. Therefore, the aim of this study was to assess treatment outcomes in patients with recurrent ACC.In this retrospective study, we included 18 patients median age 49 years (42–62); 67% female) diagnosed with recurrent ENSAT stage I-III ACC who underwent either R0 (n = 16) or Rx (n = 2) surgical resection of the tumor.The median time from the initial surgery to ACC recurrence was 29 months (IQR 18–50). Seven patients (39%) manifested local recurrence, while 11 patients (61%) developed distant metastases. The median follow-up duration after tumor recurrence was 32 months (IQR 25–53). Regarding the treatment of ACC recurrence, 10 patients underwent a second surgery either as an alone procedure (n = 4), or in combination with mitotane (n = 4), mitotane and chemotherapy (n = 1), or mitotane combined with radiotherapy (n = 1). The remaining patients received treatment involving chemotherapy±mitotane (n = 4) and locoregional therapy ±chemotherapy (n = 3). One patient chose not to proceed with further management and follow-up. The median PFS was 17 (95% CI 8–26) months while the median OS was not reached. In the multivariate model, increased mortality was associated with advanced age (p = 0.04) and a shorter interval to ACC recurrence (p = 0.03).A significant proportion of patients with ACC recurrence experience disease progression or second recurrence, despite all treatment efforts. Purpose: The recent development and approval of new diagnostic imaging and therapy approaches in the field of theranostics have revolutionised nuclear medicine practice. To ensure the provision of these new imaging and therapy approaches in a safe and high-quality manner, training of nuclear medicine physicians and qualified specialists is paramount. This is required for trainees who are learning theranostics practice, and for ensuring minimum standards for knowledge and competency in existing practising specialists. Methods: To address the need for a training curriculum in theranostics that would be utilised at a global level, a Consultancy Meeting was held at the IAEA in May 2023, with participation by experts in radiopharmaceutical therapy and theranostics including representatives of major international organisations relevant to theranostics practice. Results: Through extensive discussions and review of existing curriculum and guidelines, a harmonised training program for theranostics was developed, which aims to ensure safe and high quality theranostics practice in all countries. Background: Cotadutide is a GLP1/glucagon receptor dual agonist under development for the treatment of nonalcoholic steatohepatitis and type 2 diabetes (T2D) with diabetic kidney disease. Early-phase clinical studies show that cotadutide achieves clinically significant weight loss. We aimed to establish mechanistically to what extent this effect is mediated via changes in energy intake (EI) or energy expenditure (EE). Methods: We conducted a phase 2a single-center randomized double-blind placebo-controlled trial in overweight/obese adults (BMI 27-40 kg/m2) with T2D (HbA1c 6.5-8%). Participants underwent a 16-day single-blind placebo run-in and were randomized to double-blind 42-day cotadutide or placebo. Cotadutide was given subcutaneously, up to 300 µg daily. Primary outcome: % weight change from day 17 to day 59. Secondary outcomes: % and absolute changes in EI (ad libitum lunch), and EE (indirect calorimetry). Results: A total of 19 (63%) cotadutide and 9 (78%) placebo participants completed the study. Least-squares (LS) mean weight change was -3.98% (90% CI -4.85%, -3.10%) and -1.40% (90% CI -2.66%, -0.13%) for cotadutide and placebo, respectively, p= 0.011 between groups. The cotadutide group had a striking 35% lower EI vs. placebo: LS mean -35.0% (90% CI -61.8%, -8.2%), p=0.037. Total EE (kJ/kg lean mass) after cotadutide treatment decreased vs. placebo: -6.0% vs. 0.3%, p<0.001. Significant reductions in absolute resting and activity EE were noted with cotadutide after 42 days, but there was no change in resting EE (8.5%; cotadutide vs. placebo 1.2%, p=0.177) after 16 days of therapy despite a reduction in body weight. No additional safety concerns were raised. Conclusions: Weight loss caused by cotadutide is predominantly due to reduced EI rather than increased EE. We analyzed children with urolithiasis with age- and gender-matched healthy children. Calcium (mmol/mmol creatinine) and the calcium/citrate ratio (mol/mmol) are the only variables that differentiate children before puberty from healthy children (ROC analysis confirmed only calcium/citrate as a significant variable with cut-off value > 0.84). Peri-pubertal children are distinguished from age- and gender-matched healthy children by the following variables: citrate (mmol/mol creatinine), calcium/citrate (mol/mmol), oxalate/glycosaminoglycans (mmol/g), oxalate/citrate ratios (mmol/mmol) and oxalate/(citrate × glycosaminoglycans) (mol oxalate × mol creatinine)/(mol citrate × g glycosaminoglycans). All variables were confirmed by ROC analysis with cut-off values ≤ 327.87, >1.02, >11.24, >0.12 and >0.03, respectively. These results indicate a different risk of urinary stones development before puberty vs. pubertal/postpubertal children and increasing importance (deficiency) of citrate and glycosaminoglycans in such children. A new species of endolithic cyanobacteria, Hyella racemus sp. nov. (order Pleuroeapsales), penetrates ooid grians in actively shoaling sands in shallow (0.5-5 m) waters of the Bahama carbonate platform and the arabian Gulf. Hyella racemus is characterized by prolific branching, resulting in dense grape-like cell clusters that penetrate the carbonate substrate. The clusters radiate inward and form deep, isodiametric pits. Resin casts of these pits are frameboid (raspberry-like) in shape. The borehole morphology conforms with the shape of the endolith body and can be identified in resin-cast preparations. Cell division is by cleavage with regular alternation of division planes. The sheath is tight, layered, and colorless or yellow-brown when under strong illumination. The new species is one of the most common endoliths in ooids of the Bahamas and the Arabian Gulf. A new species of endolithic cyanophyte, Hyella gigas Lukas and Golubic (Order: Pleurocapsales), differs from other species of Hyella by its larger cell and filament dimensions and its laminated, often pigmented sheath. It bores into mollusk shells and other marine carbonate sub-strates on the east coast of North America. The Bahama islands and the Northern Mediterranean Sea, where it is found from near mean low water down to 25 m depth. This comparatively shallow maximum depth is probably due to the limited capacity of Hyella gigas to adapt to changing light quality and intensity. This paper describes a new genus and species of endolithic microorganism, Cyanosaccus piriformis Lukas and Golubic (Cyanophyta, Pleurocapsales). It is characterized by 1) having one to four spherical to pear-shaped cells within a stalked, gelatinous sheath, 2) its mode of reproduction by release of a large number of baeocytes produced by the proximal daughter cell following transverse binary fission of the parent cell, and 3) its ability to bore into and dwell within carbonate substrates in the marine environment. Its habit of asymmetrical sheath production allies it with the common microboring genera Hyella and Solentia within the family Hyellaceae. Cyanosaccus is a common inhabitant of mollusk shells found in water depths ranging from +0.5 to -75 m MLW on the eastern Florida continental margin and in carbonate sands or rocks in the intertidal and subtidal zones of the Bermuda and Bahama Islands. Understanding optical and radiative properties of aerosols and clouds is critical to reduce uncertainties in climate models. For over 10 years, the Observatory of Atmospheric Physics of La Réunion (OPAR) has been operating three active lidar instruments (named Li1200, LiO3S and LiO3T) providing time-series of vertical profiles from 3 to 45 km of the aerosol extinction and backscatter coefficients at 355 and 532 nm, as well as the linear depolarization ratio at 532 nm. This work provides a full technical description of the three systems, details about the methods chosen for the signal preprocessing and processing, and an uncertainty analysis. About 1737 night-time averaged profiles were manually screened to provide cloud-free and artifact-free profiles. Data processing consisted in Klett inversion to retrieve aerosol optical products from preprocessed files. The measurement frequency was lower during the wet season and the holiday periods. There is a good correlation between the Li1200 and LiO3S in terms of stratospheric AOD at 355 nm (0.001–0.107; R = 0.92 ± 0.01), and with the LiO3T in terms of Angström exponent 355/532 (0.079–1.288; R = 0.90 ± 0.13). The lowest values of the averaged uncertainty of the aerosol backscatter coefficient for the three time-series are 64.4 ± 31.6 % for the LiO3S, 50.3 ± 29.0 % for the Li1200, and 69.1 ± 42.7 % for the LiO3T. These relative uncertainties are high for the three instruments because of the very low values of extinction and backscatter coefficients for background aerosols above Maïdo observatory. Uncertainty increases due to SNR decrease above 25 km for the LIO3S and Li1200, and 20 km for the LiO3T. The LR is responsible for an uncertainty increase below 18 km (10 km) for the LiO3S and Li1200 (LiO3T). The LiO3S is the most stable instrument at 355 nm due to less technical modifications and less misalignments. The Li1200 is a valuable addition to fill in the gaps in the LiO3S time-series at 355 nm or for specific case-studies about the middle and low troposphere. Diagnostic criteria for determination of inclination towards idiopathic calcium oxalate (CaOx) urolithiasis based on biochemical urine parameters are not sufficiently well defined in children. The aim of this study was to determine the risk of CaOx urolithiasis in children from concentrations of calcium, oxalate, citrate, and glycosaminoglycans in urine and their ratios, all standardized in respect to creatinine. We collected and analyzed 24-h urine samples of children with CaOx urolithiasis (n = 61) and compared with urine samples of matched control group of healthy children (n = 25). The study has showed that all stone formers have higher excretion of calcium (mmol/mmol creatinine), calcium/citrate (mol/mmol), and oxalate/(citrate × glycosaminoglycans) ratio (mol Ox × mol cr)/(mol Cit × g GAGs). ROC analysis of these variables gave criteria (>0.28, >1.07, and >0.08, respectively) for distinguishing stone formers from healthy children. Biochemical urine parameters and their ratios (calcium, calcium citrate, and oxalate/(citrate × glycosaminoglycans) enable one to discriminate idiopathic calcium oxalate stone formers from healthy children. Oxalate/(citrate × glycosaminoglycans) ratio per se can serve as an independent risk for stone formation.Conclusion: Using biochemical urine parameters and their ratios such as calcium, calcium/citrate, and oxalate/(citrate × glycosaminoglycans) enables one to determine diagnostic criteria towards idiopathic calcium oxalate urolithiasis in children. The participation of microorganisms in construction and destruction of sedimentary structures is widely recognized, and so is the importance of studying such geological processes in modern systems, where the conditions, participating forces, and the results can be observed and recorded. This information is important for understanding and interpreting corresponding processes if their effects were preserved as part of the fossil record. The present contribution refers to topics discussed during the 9th International Bioerosion Workshop in Rome on Oct. 23–27, 2017, dedicated to the evaluation of microbial traces as paleoecological and paleobathymetric indicators. The paper reviews the habitats, methods of collection, and preparation of samples, followed by observation of extracted microbial euendoliths. This approach is complemented by producing images of three-dimensional display of inhabited microborings in their original positions using resin-casting and double embedding of the microbially invaded substrates. This contribution stresses the value of recognizing the microboring organisms' identities as a key aspect of the interpretation of their traces. It discusses different and complementary ways of how to achieve such parallel assessments. It reports on the importance of photo-documentation and morphometric evaluation of microbial populations, while avoiding possible artefacts caused by the methods used. The study also briefly summarizes the distribution patterns of microboring organisms and their boring and etching traces along depth profiles. Cardiovascular disease (CVD) is the most common non-communicable disease occurring globally. Although previous literature has provided useful insights into the important role that diet plays in CVD prevention and treatment, understanding the causal role of diets is a difficult task considering inherent and introduced weaknesses of observational (e.g. not properly addressing confounders and mediators) and experimental research designs (e.g. not appropriate or well designed). In this narrative review, we organised current evidence linking diet, as well as conventional and emerging physiological risk factors, with CVD risk, incidence and mortality in a series of diagrams. The diagrams presented can aid causal inference studies as they provide a visual representation of the types of studies underlying the associations between potential risk markers/factors for CVD. This may facilitate the selection of variables to be considered and the creation of analytical models. Evidence depicted in the diagrams was systematically collected from studies included in the British Nutrition Task Force report on diet and CVD and database searches, including Medline and Embase. Although several markers and disorders linked to conventional and emerging risk factors for CVD were identified, the causal link between many remains unknown. There is a need to address the multifactorial nature of CVD and the complex interplay between conventional and emerging risk factors with natural and built environments, while bringing the life course into the spotlight. Due to their ability to induce immunological tolerance in the recipient, mesenchymal stromal cells (MSCs) have been utilized in the treatment of various hematological and immune- and inflammation-mediated diseases. The clinical application of MSCs implies prior in vitro expansion that usually includes the use of fetal bovine serum (FBS). The present study evaluated the effect of different platelet lysate (PL) media content on the biological properties of MSCs. MSCs were isolated from the bone marrow of 13 healthy individuals and subsequently expanded in three different culture conditions (10% PL, 5% PL, 10% FBS) during 4 passages. The cells cultured in different conditions had comparable immunophenotype, clonogenic potential, and differentiation capacity. However, MSC growth was significantly enhanced in the presence of PL. Cultures supplemented with 10% PL had a higher number of cumulative population doublings in all passages when compared to the 5% PL condition (p < 0.03). Such a difference was also observed when 10% PL and 10% FBS conditions were compared (p < 0.005). A statistically significant difference in population doubling time was determined only between the 10% PL and 10% FBS conditions (p < 0.005). Furthermore, MSCs cultured in 10% PL were able to cause a 66.9% reduction of mitogen-induced lymphocyte proliferation. Three chromosome aberrations were detected in PL conditions. Since two changes occurred in the same do nor, it is possible they were donor dependent rather than caused by the culture condition. These findings demonstrate that a 10% PL condition enables a higher yield of MSCs within a shorter time without altering MSC properties, and should be favored over the 5% PL condition.
