Beneficial utilization of yuzu (Citrus junos) peel was investigated to improve the commercial value of cultured yellowtail fish. The antioxidant effects of grated yuzu peel from pomace (yuzu paste) on prevention of dark muscle discoloration during storage in sliced yellowtail fish were tested. Four diets were prepared, containing 0, 10, 50, and 100 g yuzu paste in a 1,450-g diet (wet basis). Fish were fed one of the four diets for 10 weeks. Growth performance was not significantly different among the dietary groups, but fish fed a diet containing 100-g yuzu paste showed the least average body weight and a significant increase in fecal amount and fat levels in the feces, which suggest low digestibility and low absorption. Dark muscle discoloration was significantly reduced in all yuzu dietary groups compared to the control group during storage at 4°C. The optimal supplementation of yuzu paste in the fish diet is very effective at maintaining fish flesh freshness as a finishing diet without causing additional phosphorus or nitrogen pollution of the sea and inhibiting fish growth. Neuropeptide Y (NPY) and cholecystokinin (CCK) play important roles in regulating appetite in vertebrates, including mammals and fish. Understanding the appetite mechanism is important in aquaculture to improve production performance. The yellowtail Seriola quinqueradiata is one of the most cultured fish in Japan, but little is known about its appetite hormones. In the present study, complementary DNA encoding for NPY was cloned in yellowtail and consists of 604 bp, in which deduced amino acid sequences show high identity to those of other teleosts. In tissue distribution, the npy and cck mRNAs were detected in all examined tissues (whole brain, telencephalon, optic tectum, hypothalamus, cerebellum, pituitary, retina, stomach, pyloric caeca, anterior intestine, liver, and kidney). In the fasting experiment, only npy mRNA expression in the hypothalamus responded to fasting, showing a significantly high value compared with that in control fish. The expression of cck mRNA in the examined tissues did not change with fasting. The npy mRNA expression in the hypothalamus might be involved in feeding regulation in yellowtail. Abstract: In vertebrates, the peptide cholecystokinin (CCK) is one of the most important neuroregulatory digestive hormones. CCK acts via CCK receptors that are classified into two subtypes, CCK-1 receptor (CCK-1R; formally CCK-A) and CCK-2 receptor (formally CCK-B). In particular, the CCK-1R is involved in digestion and is regulated by CCK. However, very little information is known about CCK-1R in fish. Therefore, we performed molecular cloning of CCK-1R cDNA from the digestive tract of yellowtail Seriola quinqueradiata. Phylogenetic tree analysis showed a high sequence identity between the cloned yellowtail CCK receptor cDNA and CCK-1R, which belongs to the CCK-1R cluster. Furthermore, the expression of yellowtail CCK receptor mRNA was observed in gallbladder, pyloric caeca, and intestines, similarly to CCK-1R mRNA expression in mammals, suggesting that the cloned cDNA is of CCK-1R from yellowtail. In in vivo experiments, the CCK-1R mRNA levels increased in the gallbladder and pyloric caeca after feeding, whereas in vitro, mRNA levels of CCK-1R and digestive enzymes in cultured pyloric caeca increased by the addition of CCK. Abstract: The polymerization of N-isopropylacrylamide (NIPAAm) with dilithium tetra-tert-butylzincate (TBZL) has been investigated in toluene at low temperatures in the presence of alkyl and fluorinated alcohols. Of the alcohols examined, 1,1,1,3,3,3-hexafluoro-2-propanol induced isotactic specificity and accelerated the polymerization process, affording the corresponding poly(NIPAAm)s with meso (m) diad contents of 82% in relatively high yields. It is worthy of note that the introduction of a fluorinated alcohol, which is typically used as an inhibitor in conventional anionic polymerization processes, enabled control over the stereospecificity and rate of the anionic polymerization of NIPAAm when TBZL was used as an initiator. Yttrium trifluoromethanesulfonate also induced isotactic specificity in the NIPAAm polymerization process in methanol and gave poly(NIPAAm) with an m diad content of 88% in high yield. Cholecystokinin (CCK) is the key regulator hormone that stimulates the secretion of digestive pancreatic enzymes in vertebrates. In fish, little is known about the mechanism of induction of CCK in the digestive tract by feed ingredients. To investigate the response of CCK and digestive enzymes to fish feed ingredients in yellowtail Seriola quinqueradiata, we performed a number of experiments in which we measured the mRNA levels of CCK, trypsin, and lipase after oral administration of a single bolus of the ingredients. We administered fish meal and fish oil in experiment 1; high and low concentrations of fish meal in experiment 2; and five different dietary protein sources (fish meal, soybean meal, soy protein concentrate, corn gluten meal, and glutamic acid fermentation by-products) in experiment 3. In experiments 1 and 3, only fish meal significantly increased the mRNA levels of CCK and digestive enzyme. In experiment 2, a high concentration of fish meal [20 % (w/v)] significantly increased CCK and trypsin mRNA levels, but a low concentration of fish meal [1 % (w/v)] did not. These results suggest that high concentrations of fish meal (the protein source in fish feed) has the most potent effect on stimulation of CCK synthesis and secretion of digestive enzymes in yellowtail. Radical polymerization of N- n-propyl-α-fluoroacrylamide (NNPFAAm) was investigated in several solvents at low temperatures in the presence or absence of Lewis bases, Lewis acids, alkyl alcohols, silyl alcohols, or fluorinated alcohols. Different effects of solvents and additives on stereospecificity were observed in the radical polymerizations of NNPFAAm and its hydrocarbon analogs such as N-isopropylacrylamide (NIPAAm) and N- n-propylacrylamide (NNPAAm); for instance, syndiotactic (and heterotactic) specificities were induced in radical polymerization of NNPFAAm in polar solvents (and in toluene in the presence of alkyl and silyl alcohols), whereas isotactic (and syndiotactic) specificities were induced in radical polymerizations of the hydrocarbon analogs under the corresponding conditions. In contrast, heterotactic specificity induced by fluorinated alcohols was further enhanced in radical polymerization of NNPFAAm. The effects of stereoregularity on the phase-transition behaviors of aqueous solutions of poly(NNPFAAm) were also investigated. Different tendencies in stereoregularity were observed in aqueous solutions of poly(NNPFAAm)s from those in solutions of the hydrocarbon analogs such as poly(NIPAAm) and poly (NNPAAm). The polymerization behavior of NNPFAAm and the phase-transition behavior of aqueous poly(NNPFAAm) are discussed based on possible fluorine-fluorine repulsion between the monomer and propagating chain-end, and neighboring monomeric units. © 2012 Wiley Periodicals, Inc. The effects of alcohol extract of soybean meal on growth performance, digestibility, and pancreatic enzyme and bile acid status of yellowtail Seriola quinqueradiata were studied. Fish were fed one of the following for 8 weeks: a fish meal (FM) control diet (FMD), a defatted soybean meal diet (SBMD), an alcohol-extracted SBM diet (ExSBMD), an ExSBMD with alcohol-extract diet (ExSBM + ExtD), and an FMD with alcohol-extract diet (FM + ExtD). Growth performance was significantly inferior in fish fed with SBM, ExSBM + ExtD, and FM + ExtD compared to FMD- or ExSBMD-fed fish. Total bile acid levels, and trypsin and lipase activities in the anterior intestine were significantly lower in fish fed with SBM, ExSBM + ExtD, and FM + ExtD than in fish fed with ExSBMD and FMD, despite similar values in gallbladder or pyloric caeca. Cholecystokinin mRNA levels of the former diet-fed fish were significantly lower than those of FMD- or ExSBMD-fed fish. Lipid and protein digestibility of fish fed with SBMD, ExSBM + ExtD, and FM + ExtD was significantly reduced in comparison with that of fish fed with FMD or ExSBMD. These findings indicate that the alcohol extract of soybean meal inhibited the secretion of bile acids and pancreatic enzymes by a decrease of cholecystokinin stimulation.
