Abstract It is a challenge to ascertain the quality of polysaccharides due to their complex chemical structure; therefore, multi-fingerprint profiling was used to investigate the quality of Astragalus polysaccharides (APS) harvested from Inner Mongolia (NM) and Gansu (GS) with the help of chemometric analysis. Additionally, FT-IR and 1H NMR were applied to characterize the chemical structure of the harvested APS. The spectral fingerprinting results indicated that APS had reduced similarity when they were from different origins. Further, PCA showed that NM and GS could be distinguished and that the main differences from the loading plots were in the absorption intensity of carbonyls and H1 signals of Galp and β-glucose. Moreover, UPLC/Q-TOF-MS fingerprints were established based on the monosaccharide composition of the APS. The concentration of monosaccharides and results of cluster analysis indicated that GlcA might be an indicator that can be used to distinguish NM and GS. Overall, this multiple fingerprint method was stable, comprehensive and valid for monitoring APS quality. A novel electrochemiluminescence resonance energy transfer (ECL-RET) system from Eu 3 + -doped CdS nanocrystals (CdS:Eu NCs) to Ru(bpy) 3 2 + -doped silica nanoparticle (RuSi) was designed for ultrasensitive detection of prostate-specific antigen (PSA). The platform consists of a CdS:Eu NCs film on glassy carbon electrode (GCE) as ECL emitter and a sandwich-type immunocomplex formed by the primary antibody, PSA and RuSi-tagged second antibody. This system showed a maximum of 4.83-fold enhancement of ECL intensity at 620 nm due to the efficient ECL-RET and high Ru(bpy) 3 2 + quantum yields. By the signal amplification of RuSi and the specific antibody–antigen interactions, this ECL-RET system could sensitively respond down to 1.0 fg mL − 1 PSA. [ABSTRACT FROM AUTHOR]

Many malignant tumors express high levels of the chemokine receptor CXCR4, and the interaction between CXCR4 and its ligand, SDF-1, promotes migration, invasion, and metastasis of breast cancer cells. Therefore, blocking the interaction between CXCR4 and SDF-1 could alter the tumor's metastatic phenotype and control the development and progression of cancers. We used a cellular phenotypic knockout strategy and developed a novel recombinant gene, AdSDF-1α/54/KDEL, which contains an adenovirus vector, a mutant form of SDF-1 that lacks a C-terminal α-helix, and a KDEL tetrapeptide sequence that promotes retention at the endoplasmic reticulum (ER). We hypothesized that SDF-1α/54/KDEL could efficiently block metastasis of breast cancer cells with less inflammatory side effects than SDF-1α/KDEL. Using the MCF-7 cell line, which expresses a stable, high level of CXCR4, we found that SDF-1α/54/KDEL efficiently becomes localized at the ER of tumor cells, where it specifically binds to newly synthesized CXCR4 and prevents it from reaching the cell surface. Chemotaxis and invasion assays revealed that the cells treated with SDF-1α/54/KDEL failed to migrate toward SDF-1. We also found that SDF-1α/54/KDEL impaired lung metastasis of metastatic breast cancer by decreasing CXCR4 on the cell surface. The novel recombinant gene, SDF-1α/54/KDEL, played an instrumental role in blocking SDF-1/CXCR4-mediated cell migration, and we found that this gene-based strategy for targeting the SDF-1/CXCR4 axis offers a very effective alternative method for preventing metastasis of breast cancer and other cancers expressing high levels of CXCR4. Transconductance (gm) is one important figure of merit for benchmarking organic electrochemical transistor (OECT) biosensors, which nevertheless experiences undesired fluctuation instead of keep constant during the target detection of varied concentrations. Light‐matter interplay is recently shown as a potential way to regulate the OECT characteristics, e.g. maximum gm at zero gate bias is achieved. In this work, the challenge of unstable gm is addressed by using a unique metal‐organic framework (MOF)‐derived hollow tube indium sulfide (m‐In2S3) as the photogate. Interestingly, the light irradiation on m‐In2S3 generate a straight transfer curve, leading to a stable gm that is desirable for biological application. Exemplifying by an aptasening, such a device is then tested for protein quantification with a stable sensitivity during the detection. This study explores the mechanism of enhanced electrochemiluminescence (ECL) due to the coupling effect in gold nanodimers (Au NDs) with precisely controlled interparticle distances via electrochemiluminescence microscopy (ECLM). Our research revealed that the enhancement in ECL was predominantly attributed to increased charge density and elevated electric fields resulting from overlapping electrochemical double layers. These findings offer new insights into the fundamental processes that govern nanostructure-mediated electrocatalysis, opening up exciting possibilities for future applications. Designing dynamic assemblies in living cells is crucial for creating organelle‐like structures, yet precisely controlling their morphological transitions in response to specific signals is a significant challenge. In this study, a DNA framework is combined with hybridization chain reaction (HCR) to achieve specific assembly of hyperbranched aggregates in cancer cells. HCR, distinguished for its signal amplification and linear extension capabilities, enables the morphological transition of precursors to be specifically triggered by trace amounts of endogenous microRNA‐21 (miR‐21). The spatial constraints of the framework and the diversity of hairpin orientations significantly accelerate the assembly kinetics of hyperbranched networks, and the resulting micrometer‐scale aggregates possess enhanced intracellular retention capabilities. Introducing Ce6 molecules as a proof of concept, the regulatory function of aggregates can be activated under light irradiation and remains effective over a long period. The probe we constructed demonstrates good stability and biocompatibility, offers easy functionalization, and works inside cells long‐term, making it an ideal candidate material for the construction of organelle‐like structures. Purpose: MicroRNAs play important roles in cancer development, progression, and metastasis. The aim of this study was to determine whether altered microRNA-155 expression is associated with hepatocellular carcinoma (HCC) recurrence and prognosis following orthotopic liver transplantation (OLT). Methods: Tissue specimens from 100 HCC patients following OLT were recruited. MicroRNA-155 expression levels were detected using quantitative reverse transcription-polymerase chain reaction (qRT-PCR). Kaplan-Meier and Cox proportional regression analyses were utilized to determine the association of microRNA-155 expression with survival of patients. MicroRNA-155 expression levels of two HCC cell lines (HepG2 and SMMC-7721) and normal liver tissue were quantified using qRT-PCR. The potential function of miR-155 on invasiveness was evaluated in the above HCC cell lines. Results: We found that microRNA-155 expression levels were high in tumor tissues in patients with post-OLT HCC recurrence ( n = 45) compared with those in patients with non-recurrence ( n = 55) ( P = 0.001) and correlated with micro-vascular invasion of HCC tissue samples ( P = 0.001). Patients with higher miR-155 expression had significantly poorer recurrence-free survival (RFS, log rank P < 0.001) and overall survival (OS, log rank P < 0.001). Multivariate analysis revealed that high miR-155 expression was an independent predictor of poor prognosis (HR 2.748, P = 0.001 for RFS; HR 5.752, P < 0.001 for OS). In addition, the invasiveness of HCC cells was significantly increased by higher microRNA-155 expression. Conclusions: MicroRNA-155 is a candidate oncogenic microRNA and plays an important role in promoting HCC cells invasion. Although the potential value of phenotypic/functional knockout technology with intrabody/kine in prevention and cure of some serious diseases, such as AIDS and cancer, is being regarded, there are still several technical difficulties. One of the the most critical problems is how to directly deliver the intrabody/kine proteins into endoplasm reticulum (ER). In this study, a novel recombinant protein, TAT–GFP–KDEL, was designed and constructed. In this recombinant protein, HIV-derived TAT (47–57) and an ER retention four-peptide sequence KDEL were fused at the N-terminal and C-terminal of GFP respectively. The results showed that TAT–GFP–KDEL had been successfully expressed in bacteria BL21 and its purity reached to 95%. Moreover, we observed that this recombinant protein was able to efficiently transduce into MOLT-4 cells and accurately locate at ER. Herein, we report the rapid mass spectrometric identification of the short-lived intermediates generated under AC electrolysis via combining bipolar electrochemistry with nanoelectrospray ionization in a hybrid ultramicroelectrode/ion emitter. The key reactive intermediates involved in the C–O/O–H cross-metathesis between 4-alkoxy anilines and alcohols were successfully captured and identified for the first time, providing direct evidence for the previously proposed mechanism. A platinum electrode modified with lanthanum hexacyanoferrate (LaHCF) was prepared by electrochemical scanning and characterized by Fourier transform infrared spectroscopy, X-ray powder diffraction (XRD), scanning electron microscopy (SEM) and voltammetry. The results show that the deposit on the platinum substrate is composed of lanthanum potassium hexacyanoferrate (II) tetrahydrate. The electrochemical behavior of the LaHCF modified electrode was investigated preliminarily by voltammetry, and the experiments showed that hydrogen ion has a considerable effect on the electrochemical behavior of the modified electrode. This study explores plasmon-induced electrochemical reactions on single nanoparticles using electrogenerated chemiluminescence microscopy (ECLM). Under laser irradiation, real-time screening showed lower plasmon-induced reaction efficiency for bimetallic Au@Pt nanoparticles compared to monometallic Au nanoparticles. ECLM offers a high-throughput imaging and precise quantitative approach for analyzing photo-electrochemical conversion at single nanoparticle level, valuable for both theoretical exploration and optimization of plasmonic nanocatalysts. For the first time, we report novel aggregation-induced electrochemiluminescence (AIECL) of organic dots in aqueous media, with near-infrared II (NIR-II) luminescence peaked at 906 nm. Furthermore, a hybrid mechanism of ECL generation is revealed by various experiments in conjunction with theoretical calculations. The transport behavior of ions in the nanopores has an important impact on the performance of the electrochemical devices. Although the classical Transmission‐Line (TL) model has long been used to describe ion transport in pores, the boundary conditions for the applicability of the TL model remain controversial. Here, we investigated the transport kinetics of different ions, within nanochannels of different lengths, by using transient single‐particle imaging with temporal resolution up to microseconds. We found that the ion transport kinetics within short nanochannels may deviate significantly from the TL model. The reason is that the ion transport under nanoconfinement is composed of multi basic stages, and the kinetics differ much under different stage domination. With the shortening of nanochannels, the electrical double layer (EDL) formation would become the "rate‐determining step" and dominate the apparent ion kinetics. Our results imply that using the TL model directly and treating the in‐pore mobility as an unchanged parameter to estimate the ion transport kinetics in short nanopores/nanochannels may lead to orders of magnitude bias. Alzheimer's disease (AD), a progressive neurological disorder, presents challenges in early detection due to the limitations and costs of neuropsychological tests and neuroimaging. This study introduces a visual electrochemiluminescence (ECL) sensing device employing red and green signal lights on dual-channel closed bipolar electrodes (BPEs) for simultaneous Aβ oligomers and Tau proteins detection. To mitigate cross-chemical reactions, we physically separated target detection and ECL production within different chambers connected by BPEs. Utilizing efficient gold clusters (Au NCs) and Ru(bpy) 3 2+ as green and red emitters, respectively, along with methylene blue (MB) as the recognition probe on the cathode sensing platform, we achieved a detection limit of 2.6 pM for Aβ oligomers and 0.3 pM for Tau proteins by measuring visual ECL intensity at two anodic poles of BPEs. This approach minimizes cross-chemical reactions, ensures robust ECL signal sensors, and allows integration of multiple sensors on a single chip. Construction of white‐light emitters is of great importance in practical applications but is challenging for metal nanoclusters. Herein, arginine (Arg)‐mediated copper nanoclusters (Cu NCs) assemblies with white‐light emission (WLE) are reported for the first time. The multidentate ligand, Arg, not only effectively tailors the high stability and locally‐excited emission (LES) of aqueous phase Cu NCs, but also bridges 1‐naphthalenol methylcarbamate (NMC) to generate a blue emitter via through‐space interactions (TSIs), resulting in a stable mixed light. Density functional theory calculation and mass spectrum further confirm that Arg can allow for the formation of strong hydrogen‐bonds with the ligand 2,4‐diaminotoluene of Cu NCs, and formation of TSIs‐based complexes with NMC. More importantly, pure white‐light emission (WLE) with Commission Internationale de L'Eclairagecoordination (0.30, 0.31) is observed by regulating the ratio between Arg and the interfacial ligand of NMC. The findings not only provide a new perspective for the construction of WLE emitters via interface modification chemistry but also open a window to gain insight into the mechanism of interface engineering. Engineered nanopipette technology has gained momentum in single‐cell studies; however, solutions to epigenetic questions remain unachieved. This study addresses the challenge by exploring N6‐methyladenine (m6A)‐bearing deoxyribozyme (DNAzyme) confined within a nanopipette for profiling a representative m6A‐modifying enzyme, fat mass and obesity‐associated protein (FTO). Electroosmotic intracellular extraction of FTO could remove the m6A and cause DNAzyme cleavage, leading to the altered ionic current signal. Because the cleavage can release a DNA sequence, we simultaneously program it as an antisense strand against FTO‐mRNA, intracellular injection of which has been shown to induce early stage apoptosis. Classic tris(bipyridine)ruthenium(II) complex (Ru-bpy) with high electrochemiluminescence (ECL) efficiency still suffers from a serious aggregation-caused quenching (ACQ) problem, which greatly weakens its ECL efficiency to restrict further applications in solid-state ECL imaging and light-emitting devices. Herein, the crystallization-induced enhanced ECL (CIE-ECL) of tris(bipyridine) ruthenium(II) derivatives (Ru-TPE) are reported by decorating Ru-bpy with inherent aggregation-induced emission active tetraphenylethene (TPE), which effectively eliminates the detrimental π-π stacking interactions and thus helps Ru-bpy surmount notorious ACQ effect in the aqueous phase. The Ru-TPE shows negligible ECL emission in solution but produces a strong ECL emission upon crystallization. Surprisingly, the ECL efficiency of Ru-TPE crystals is 20 and 3 times higher than that of its solution and Ru-bpy crystals, respectively. Experimental and structural analysis reveals that such a CIE-ECL effect originates from the restricted intramolecular rotation and ordered molecular packing. Moreover, the high-resolution ECL imaging of a single Ru-TPE crystal is successfully demonstrated. Cuproptosis is a new form of programmed cell death and exhibits enormous potential in cancer treatment. However, reducing the undesirable Cu ion release in normal tissue and maximizing the copper‐induced therapeutic effect in cancer sites are two main challenges. In this study, we constructed a photothermally triggered nanoplatform (Au@MSN‐Cu/PEG/DSF) to realize on‐demand delivery for synergistic therapy. The released disulfiram (DSF) chelated with Cu2+ in situ to generate highly cytotoxic bis(diethyldithiocarbamate)copper (CuET), causing cell apoptosis, and the formed Cu+ species promoted toxic mitochondrial protein aggregation, leading to cell cuproptosis. Synergistic with photothermal therapy, Au@MSN‐Cu/PEG/DSF could effectively kill tumor cells and inhibit tumor growth (inhibition rate up to 80.1 %). Enzymatic biosensors have been valuable bioanalytical devices for analysis of diverse targets in disease diagnosis, biological and biomedical research, etc. Photoelectrochemical (PEC) bioanalysis is a recently emerged method that promptly becoming a subject of new research interests due to its attractive potential for future bioanalysis with high sensitivity and specificity. PEC enzymatic biosensors integrate the inherent sensitivities of PEC bioanalysis and the selectivity of enzymes and thus share their both advantages. Currently, PEC enzymatic biosensors have become a hot topic of significant research and the recent impetus has grown rapidly as demonstrated by increased research papers. Given the pace of advances in this area, this review will make a thorough discussion and survey on the fundamentals, sensing strategies, applications and the state of the art in PEC enzymatic biosensors, followed by future prospects based on our own opinions. Rational utilization of the rich light‐bio‐matter interplay taking place in single‐cell analysis represents a new technological direction in the field. The light‐fueled operation is expected to achieve advanced photoelectrochemical (PEC) single‐cell analysis with unknown possibilities. Here, a PEC nanoreactor capable of single‐cell sampling and near zero‐background Faradaic detection of intracellular microRNA (miR) is devised by the construction of a small reaction chamber accommodating the target‐triggered hybridization chain reaction for binding the metallointercalator of [Ru(bpy)2(dppz)]2+ as the signal reporter. Light stimulation of the dsDNA/metallointercalator adduct will induce the generation of photocurrents, underpinning a zero‐biased and near zero‐background PEC method toward Faradaic detection of non‐electrogenic miR at the single‐cell level. Saccharides play critical roles in many forms of cellular activities. Saccharide structures are however complicated and similar, setting a technical hurdle for direct identification. Nanopores, which are emerging single molecule tools sensitive to minor structural differences between analytes, can be engineered to identity saccharides. A hetero‐octameric Mycobacterium smegmatis porin A nanopore containing a phenylboronic acid was prepared, and was able to clearly identify nine monosaccharide types, including D‐fructose, D‐galactose, D‐mannose, D‐glucose, L‐sorbose, D‐ribose, D‐xylose, L‐rhamnose and N‐acetyl‐D‐galactosamine. Minor structural differences between saccharide epimers can also be distinguished. To assist automatic event classification, a machine learning algorithm was developed, with which a general accuracy score of 0.96 was achieved. Acidic catecholamine metabolites, which could serve as diagnostic markers for many diseases, demonstrate an importance of accurate sensing. However, they share a highly similar chemical structure, which is a challenge in the design of sensing strategies. A nanopore may be engineered to sense these metabolites in a single molecule manner. To achieve this, a recently developed programmable nano‐reactor for stochastic sensing (PNRSS) technique adapted with a phenylboronic acid (PBA) adaptor was applied. Three acidic catecholamine metabolites, including 3,4‐dihydroxyphenylacetic acid (DOPAC), 3,4‐dihydroxymandelic acid (DHMA) and 3‐methoxy‐4‐hydroxymandetic acid (VMA) were investigated by PNRSS. Specifically, DHMA, which contains an α‐hydroxycarboxylate moiety and an adjacent cis‐hydroxyl groups on its benzene ring, reports two binding modes simultaneously resolvable by PNRSS. Assisted with the high resolution of PNRSS, direct regulation of these two binding modes by pH can also be observed. Photoelectrochemical (PEC) bioanalysis represents a unique and dynamically developing methodology that offers an elegant route for sensitive biomolecular detection. Aptamers are synthetic nucleic acid molecules whose binding characteristics can rival those of protein antibodies. Originated from the fusion of PEC bioanalysis and aptamers, PEC aptasensing has rapidly becoming a subject of new research interests in recent years. Using illustrative examples, this review provides the introductory concept, bioanalysis development, signaling strategies, and the state of the art in this field. Depending on the situation, metal ions may either play beneficial roles or be harmful to human health and ecosystems. Sensitive and accurate detection of metal ions is thus a critical issue in the field of analytical sciences and great efforts have been devoted to the development of various metal ion sensors. Photoelectrochemical (PEC) detection is an emerging technique for the bio/chemical detection of metal ions, and features a fast response, low cost and high sensitivity. Using representative examples, this review will first introduce the fundamentals and summarize recent progress in the PEC detection of metal ions. By assembling nanotweezers with ATP-splitting aptamers on gold nanorods (AuT123L), we constructed a near-infrared-activated ATP sensing device that could time-controllably image ATP levels in living cells. By replacing the aptamers on the nanotweezers, the nanoplatform can be applied to other important biomolecules, opening up more possibilities for the study of time controllable nanodevices. Since the concept of surface plasmon amplification by stimulated emission of radiation (SPASER) was proposed in 2003, great progresses have been made on two kinds of plasmonic nanolasers, namely SPASER device and SPASER nanoparticle. In comparison, SPASER nanoparticle, with ultra‐narrow emission line, small size, and good biocompatibility, is a kind of promising luminescent nanoprobe and has a bright application prospect in biomedical imaging and sensing. Hence, it is gradually becoming an attractive research hotspot in the world. However, as the research on SPASER nanoparticle is still in its infancy, there are still lots of problems to be solved before it is widely applied. In this paper, the latest research advances of SPASER nanoparticles and their biological applications are reviewed. A novel electrochemiluminescence (ECL) Janus emitter realizes dual‐mode biosensing for multiple disease‐related biomarkers from cells. Each Janus emitter consists of ruthenium (II) complex and luminol polymer doped hemispheres that are spatially separated but function simultaneously, generating intense dual‐ECL emissions. This design effectively eliminates optical and electrochemical interferences between different ECL mono‐luminophores in comparison with the homogenous nanostructures, thereby improving ECL efficiency and detection sensitivity. Accordingly, the ECL Janus emitters provide a proof of concept for the construction of the ratiometric ECL sensing of H2O2. Furthermore, this Janus emitter enables simultaneous identification and quantification of the intracellular cancer‐related dual microRNAs. Photonic (bio)electronic circuits and devices have been an enduring goal in modern photonics and electronics. Organic photoelectrochemical transistor (OPECT) interfacing light with biological world creates a substantial opportunity to this end due to the immense light−bio−matter interplay. Through modulating the invisible ionic circuit of organic electrochemical transistor (OECT) with a bipolar electrode (BPE) accommodating different semiconductor combinations, the unique photonic electronics toward logic and sensor applications are realized. Six combinations of BPEs are produced by the representative n‐type CdS/TiO2 and p‐type BiOI/NiO semiconductors, and the corresponding photonic characteristics and operation rationale are systematically investigated and explained. Designation of opto‐logic circuits and feasibility as a sensory platform are subsequently demonstrated. This mini review provides a brief introduction of this arena and summaries the key steps about the development of PEC cytosensors with representative examples, followed by future prospects based on our own opinions. A large collection of unique molecular barcodes is useful in the simultaneous sensing or screening of molecular analytes. Though the sequence of DNA has been widely applied to encode for molecular barcodes, decoding of these barcodes is normally assisted by sequencing. We here demonstrate a barcode system based solely on self‐assembly of synthetic nucleic acids and direct nanopore decoding. Each molecular barcode is composed of "n" distinct information nodes in a non‐binary manner and can be sequentially scanned and decoded by a Mycobacterium smegmatis porin A (MspA) nanopore. Nanopore events containing step‐shaped features were consistently reported. 14 unique information nodes were developed which in principle could encode for 14n unique molecular barcodes in a barcode containing "n" information nodes. These barcode probes were adapted to detect different antibody proteins or cancer‐related microRNAs, suggesting their immediate application in a wide variety of sensing applications. G-quadruplex secondary structures are naturally found in genome sequences and play essential roles in regulating a wide variety of important biological processes. Although stabilizing effects of monovalent cations (e.g., K+ and Na+) has been recognized during the past decades, a general and reliable analytical method for accurate characterization of specific interactions of K+/Na+ with G-quartets is still not well established. In the present study, we demonstrate a practical strategy that combined the use of a nanoscale ion emitter, a low-flow drying gas and a volatile salt (trimethylammonium acetate) to almost totally suppress the nonspecific cationic adduction to G-quadruplexes during the ionization process. Our combined strategy takes full advantage of the ultrasmall initial charged droplets when employing a nanoscale ion emitter, the maximum uneven fission of charged droplets under the gentle desolvation conditions, and the effective shielding of the negatively charged phosphate groups by trimethylammonium ions, to eventually producing ions of G-quadruplexes free of non-specific K+/Na+ adduction. For the first time, the accurate binding states as well as the quantitative binding constants between K+/Na+ and G-quadruplexes can be directly obtained even in the presence of tens of millimolar non-volatile salts, which has long been a notorious challenge in mass spectrometry. Accurate binding state between Na + and 24TTG directly obtained by electrospray ionization mass spectrometry using a combined strategy for suppressing nonspecific cationic adduction. The evolution of photoelectrochemical (PEC) bioanalysis has resulted in substantial progress in its analytical performance and biodetection applications. The aim of this review is to provide a panoramic snapshot of the state of the art in this dynamically developing field, with special emphasis on PEC DNA analysis, immunoassay, enzymatic biosensing and cell-related detection. The future prospects in this area are also evaluated and discussed. The luminescence generated by Ru(bpy)32+ and co‐reactant tripropylamine (TPA) pair acts as both optical readout for ECL imaging, and light source for the excitation of photosensitizer to produce reactive oxygen species (ROS) in photodynamic therapy (PDT) system. The ECL‐driven PDT (ECL‐PDT) relies on the effective energy transfer from ECL emission to photosensitizer chlorin e6 (Ce6), which sensitizes the surrounding O2 into ROS. The dynamic process of gradual morphological changes, the variation of cell‐matrix adhesions, as well as the increase of cell membrane permeability in the process of ECL‐PDT were monitored under ECL microscopy (ECLM) with good spatiotemporal resolution. Combining real‐time imaging with ECL‐PDT, this new strategy provides not only new insights into dynamic cellular processes, but also promising potential of ECL in clinical applications. Single‐nanoparticle electrochemistry offers electrochemical behaviors of individual entities beyond the ensemble system. An electric double layer (EDL) exists on any charged particle–liquid interface because of counter‐ion accumulation, while direct measuring of the interfacial ion migration remains a challenge. Herein, a plasmonic‐based transient microscopic method, with a temporal resolution of 1–2 μs, was demonstrated to directly track the ion migration dynamics on single charged nanoparticles. We found that the dynamics of EDL formation might deviate significantly from the prediction made by using the classical resistance–capacitance (RC) model under nanoscale and transient conditions. Under ultrafast charging, due to the limit migration rate of ions in the solution, the actual time scale of the EDL formation could be up to 5 times slower than the predicted value from the RC model. We then proposed a new theoretical model to describe the transient dynamics of EDL formation. Comprehensive Summary: SPASER nanoparticle (NP), with small size, ultranarrow spectral‐line and good biocompatibility, is a potential biomedical nanoprobe. However, owning to the striking light absorption capacity of Au‐resonator, huge energy could accumulate under strong pumping‐laser, which would lead to poor photo‐stability and unexpected photo‐damage to SPASER NP, and is harmful to its future practical application but hasn't been systematically studied in experiment. In this paper, our experimental and theoretical researches revealed that the photo‐stability of SPASER NP is largely determined by its structure parameters: smaller Au‐core diameter, thicker silica‐shell will contribute to better photo‐stability. And the possibility and extent of photo‐damage of SPASER NP depend on not only its intrinsic stability but also the external pumping‐laser. Notably, the strong pumping condition for two‐level SPASER system could lead to heating‐melting‐evaporation or even phase‐explosion of SPASER NP, resulting in obvious photo‐damage, while, the weak pumping condition for three‐level SPASER system is quite friendly. This study provides reliable bases for the structural‐properties optimization of SPASER NPs, which is of great significance for its future application as a novel luminescence probe.
