Pesticides can help reduce yield losses caused by pests, pathogens, and weeds, but their overuse causes serious environmental pollution. They are persistent in the environment and are biomagnified through the food chain, becoming a serious health hazard for humankind. Bioremediation, where microbes are used to degrade pesticides in situ, is a useful technology. This review summarizes data on the fungi involved in the biodegradation of chemical pesticides and their application in soil and water bioremediation. Experiments in skinned fibres at low temperature (10-12°C) have shown that [Pi] increase depresses isometric force and alters the kinetics of actomyosin interaction. However, the effects of Pi decrease with temperature and this raises the question of the role of Pi under physiological conditions. The present experiments were performed to investigate this point. Intact fibre bundles isolated from the flexor digitorum brevis of C57BL/6 mice were stimulated with a series of tetanic contractions at 1.5 s intervals at 33°C. As show previously the most significant change induced by a bout of contractile activity similar to the initial 10 tetani of the series was an increase of [Pi] without significant Ca2+ or pH changes. Measurements of force, stiffness and responses to fast stretches and releases were therefore made on the 10th tetanus of the series and compared with control. We found that (i) tetanic force at the 10th tetanus was ∼20% smaller than control without a significant decrease of crossbridge stiffness; and (ii) the force recovery following quick stretches and releases was faster than in control. These results indicate that at physiological temperature the increase of [Pi] occurring during early fatigue reduces tetanic force mainly by depressing the individual crossbridge force and accelerating crossbridge kinetics. Discovering brain activity patterns associated with specific stimuli, behaviors, or mental disorders is a primary objective of many neuroimaging studies. In particular, functional magnetic resonance imaging analyses are often concerned with finding brain areas "most relevant" to a mental state of interest. In this paper, we consider the predictive accuracy of a subset of voxels (smallest units of a threedimensional image) identified by multivariate sparse regression as a better relevance measure than the commonly used mass-univariate correlations between the task and individual voxels. Sparse regression detects synergistic multivoxel interactions missed by univariate techniques. Moreover, when invoked iteratively, it allows us to discover novel patterns of information distribution through the brain. Indeed, unlike univariate techniques that typically imply a sharp transition from task-relevant to task-irrelevant brain areas, our approach reveals that for some tasks, task-relevant information is distributed rather "holographically" through the entire brain, yielding multiple relevant subsets of voxels, with gradually declining predictive accuracy, and without any clear separation between taskrelevant and task-irrelevant ones. Interestingly, as our preliminary results show, such phenomena occur in certain "complex" experiences such as pain perception, but not in relatively "simple" tasks such as finger tapping. We describe recent preliminary studies demonstrating that computational analysis of spoken and written language can provide for highly accurate diagnostics over a wide variety of psychiatric and neurologic conditions, including psychosis, drug abuse, Parkinson's, and Alzheimer's. These results are based on the mathematical formalization of psychiatric qualitative knowledge related to the characterization of the conditions (e.g., "derailment" in psychosis) and drug effects (e.g., increased intimacy/affection with the use of the recreational drug ecstasy), as well as novel linguistic feature extraction approaches. Moreover, we show novel results using publicly available text sources suggesting that 1) it is possible to define an embedding space that allows modeling of the vast language dimension into a smaller space to map and compare different conditions, and 2) computational studies of a public personality (Ronald Reagan) can likely yield novel insights into normal aging and neurodegenerative disorders. Finally, we discuss the implications for mental health (and possibly, computer science) of a systematic application of this methodology, extended to include similar readily available behavioral data such as voice and video. The Old World screwworm (OWS) fly, Chrysomya bezziana, is an obligate parasite of livestock, and the myiasis caused by its larval infestations is economically important in Indonesia. The current spatial distribution of such a pest depends on two main factors: the current environmental conditions in which it can survive; and, its ability to occupy those environments by dispersal, which can be inferred from phylogeography and population genetics. These indicate that all OWS flies in Indonesia have mitochondrial cytochrome b (cyt b) haplotypes of the Asian lineage, and the regional separation of its four sub-lineages is the result of infrequent long-distance dispersal. We report the first investigation to associate regional cyt b sub-lineages of the OWS fly with environmental variables. Principal Components Analysis was used to demonstrate that these sub-lineages are associated with very similar macro-climates throughout Indonesia. Then, a species distribution model for the OWS fly in Indonesia was obtained by using the Maxent program. This indicated that elevation captured information not given by other environmental variables, and cattle density provided the most useful information by itself. Tsetse flies, like many insects, use pheromones for inter- and intra-specific communication. Several of their pheromones are cuticular hydrocarbons (CHCs) that are perceived by contact at close range. We hypothesized that for a successful implementation of the Sterile Insect Technique (SIT), along with proper identification of target area and target species, the target tsetse populations and the sterile flies must chemically communicate with each other. To study the population structuring of Glossina pallidipes in Ethiopia, CHCs were extracted and analyzed from three tsetse belts. As a comparative approach, wing morphometric analysis was performed. The analysis of the relative abundance of CHCs revealed that populations of G. pallidipes from the Rift Valley tsetse belt showed a distinct clustering compared to populations from the other two belts. The spatial pattern of CHC differences was complemented by the wing morphometric analysis. Our data suggest that CHCs of known biological and ecological role, when combined with wing morphometric data, will provide an alternative means for the study of population structuring of Glossina populations. Abstract: Endemic animal diseases such as tsetse-transmitted trypanosomosis are a constant drain on the financial resources of African livestock keepers and on the productivity of their livestock. Knowing where the potential benefits of removing animal trypanosomosis are distributed geographically would provide crucial evidence for prioritising and targeting cost-effective interventions as well as a powerful tool for advocacy. To this end, a study was conducted on six tsetse-infested countries in Eastern Africa: Ethiopia, Kenya, Somalia, South Sudan, Sudan and Uganda. First, a map of cattle production systems was generated, with particular attention to the presence of draught and dairy animals. Second, herd models for each production system were developed for two scenarios: with or without trypanosomosis. The herd models were based on publications and reports on cattle productivity (fertility, mortality, yields, sales), from which the income from, and growth of cattle populations were estimated over a twenty-year period. Third, a step-wise spatial expansion model was used to estimate how cattle populations might migrate to new areas when maximum stocking rates are exceeded. Last, differences in income between the two scenarios were mapped, thus providing a measure of the maximum benefits that could be obtained from intervening against tsetse and trypanosomosis. For this information to be readily mappable, benefits were calculated per bovine and converted to US$ per square kilometre. Results indicate that the potential benefits from dealing with trypanosomosis in Eastern Africa are both very high and geographically highly variable. The estimated total maximum benefit to livestock keepers for the whole of the study area amounts to nearly US$ 2.5 billion, discounted at 10% over twenty years – an average of approximately US$ 3300 per square kilometre of tsetse-infested area – but with great regional variation from less than US$ 500 per square kilometre to well over US$ 10,000. The greatest potential benefits accrue to Ethiopia, because of its very high livestock densities and the importance of animal traction, but also to parts of Kenya and Uganda. Abstract: Fluorescence lidar is a non-invasive, remote sensing technique that makes it possible to extend the application of the laser-induced fluorescence technique to the outdoor environment, where uncontrolled external conditions must be met. Although initially developed for the investigation of marine environment and vegetation, in the past decade this technique has been successfully applied to the field of the cultural heritage. Among other applications, the detection and characterisation of photoautotrophic biodeteriogens has become very promising: the method is based on the detection of chlorophyll a fluorescence, while fluorescent accessory pigments can be exploited for a rough classification of the biodeteriogens present on the surface. Early experiments on monuments date back to the mid 1990s, when fluorescence lidar point measurements were conducted on the Cathedral and Baptistery of Parma, Italy. Subsequently, the technique has taken further advantage of the introduction of imaging capabilities in lidar instrumentation and this has led to the acquisition of hyperspectral fluorescence maps over extended areas of several monuments from distances as great as 80 m. Here, we present the main achievements obtained in the outdoor cultural heritage so far and the latest developments in the technique. [Copyright &y& Elsevier]

This study aims to provide trypanosomiasis-affected countries with standardized datasets and methodologies for mapping the habitat of the tsetse fly ( Glossina spp., the disease vector) by customizing and integrating state-of-the-art land cover maps on different spatial scales. Using a combination of inductive and deductive approaches, land cover and fly distribution maps are analysed in a geographic information system (GIS) to estimate the suitability of different land cover units for the three groups (subgenera) of Glossina. All land cover datasets used for and produced by the study comply with the Land Cover Classification System (LCCS). At the continental scale, a strong correlation between land cover and tsetse habitat is found for both the fusca and palpalis groups, whereas a weaker correlation found for the morsitans group may be indicative of less restrictive ecological requirements. At the regional and national levels, thematic aggregation of the multi-purpose Africover datasets yielded high-resolution, standardized land cover maps tailored for tsetse habitat for eight East African countries. The national maps provide remarkable spatial resolution, thematic detail and geographical coverage. They may be applied in subsequent phases of tsetse and trypanosomiasis control projects, including the planning of entomological surveys, actual tsetse control operations and planning for land use in reclaimed areas. The methodology and datasets discussed in the paper may have applications beyond the tsetse and trypanosomiasis issue and may be used with reference to other arthropod vectors, vector-borne and parasitic diseases. Extensive fluorescence multispectral imaging of the cathedral and baptistery of Parma, Italy, is reported and discussed. In particular, the first fluorescence imaging data from protection-treated stony materials were recorded. Fluorescence spectra were taken with a mobile lidar system scanning the monument surfaces with a frequency-tripled Nd:YAG laser beam from a distance of about 80 m. For each pixel of the area investigated, a high-spectral-resolution spectrum in the full visible range was acquired. The principal-component analysis technique was used to obtain thematic maps that outlined areas subject to protective treatment and biological growth, and other features, such as different types of stones and decoration pigments. This study builds upon earlier work mapping the potential benefits from bovine trypanosomosis control and analysing the costs of different approaches. Updated costs were derived for five intervention techniques: trypanocides, targets, insecticide-treated cattle, aerial spraying and the release of sterile males. Two strategies were considered: continuous control and elimination. For mapping the costs, cattle densities, environmental constraints, and the presence of savannah or riverine tsetse species were taken into account. These were combined with maps of potential benefits to produce maps of benefit-cost ratios. The results illustrate a diverse picture, and they clearly indicate that no single technique or strategy is universally profitable. For control using trypanocide prophylaxis, returns are modest, even without accounting for the risk of drug resistance but, in areas of low cattle densities, this is the only approach that yields a positive return. Where cattle densities are sufficient to support it, the use of insecticide-treated cattle stands out as the most consistently profitable technique, widely achieving benefit-cost ratios above 5. In parts of the high-potential areas such as the mixed farming, high-oxen-use zones of western Ethiopia, the fertile crescent north of Lake Victoria and the dairy production areas in western and central Kenya, all tsetse control strategies achieve benefit-cost ratios from 2 to over 15, and for elimination strategies, ratios from 5 to over 20. By contrast, in some areas, notably where cattle densities are below 20 per km 2 , the costs of interventions against tsetse match or even outweigh the benefits, especially for control scenarios using aerial spraying or the deployment of targets where both savannah and riverine flies are present. If the burden of human African trypanosomosis were factored in, the benefit-cost ratios of some of the low-return areas would be considerably increased. Comparatively, elimination strategies give rise to higher benefit-cost ratios than do those for continuous control. However, the costs calculated for elimination assume problem-free, large scale operations, and they rest on the outputs of entomological models that are difficult to validate in the field. Experience indicates that the conditions underlying successful and sustained elimination campaigns are seldom met. By choosing the most appropriate thresholds for benefit-cost ratios, decision-makers and planners can use the maps to define strategies, assist in prioritising areas for intervention, and help choose among intervention techniques and approaches. Abstract: Baseline surveys were conducted in the Upper West Region of Ghana to assess the distribution and densities of tsetse species, as well as the prevalence of bovine trypanosomosis. The entomological survey was designed to cover the suitable tsetse habitats along the three main rivers in the study area (i.e. Black Volta, Kulpawn and Sissili). Results indicated the presence of Glossina tachinoides in all three river basins, whilst Glossina palpalis gambiensis was only found close to the southern limit of the study area. A random sampling of 1800 cattle of the West African Short Horn, Sanga and Zebu breeds from 36 randomly selected grid cells covering the study area showed substantial differences between parasitological and serological prevalences. The average parasitological prevalence was estimated at 2.5% (95% CI: 1.06–5.77) with the majority of the infections due to Trypanosoma vivax. Most of the infected cattle were found close to the major river systems. The serological prevalence, measured using Enzyme Linked Immunosorbent Assay (ELISA), test was 19% (95% CI: 14.03–25.35). Background: Trypanocidal drugs have been used to control African animal trypanosomosis for several decades. In Ethiopia, these drugs are available from both authorized (legal) and unauthorized (illegal) sources but documentation on utilization practices and quality of circulating products is scanty. This study looked at the practices of trypanocidal drug utilization by farmers and the integrity of active ingredient in trypanocides sold in Gurage zone, south western Ethiopia. The surveys were based on a structured questionnaire and drug quality determination of commonly used brands originating from European and Asian companies and sold at both authorized and unauthorized markets. One hundred farmers were interviewed and 50 drug samples were collected in 2013 (Diminazene aceturate = 33 and Isometamidium chloride = 17; 25 from authorized and 25 from unauthorized sources). Samples were tested at the OIE-certified Veterinary Drug Control Laboratory (LACOMEV) in Dakar, Senegal, by using galenic standards and high performance liquid chromatography. Results: Trypanosomosis was found to be a major threat according to all interviewed livestock keepers in the study area. Diminazene aceturate and isometamidium chloride were preferred by 79% and 21% of the respondents respectively, and 85% of them indicated that an animal receives more than six treatments per year. About 60% of these treatments were reported to be administered by untrained farmers. Trypanocidal drug sources included both unauthorized outlets (56%) and authorized government and private sources (44%). A wide availability and usage of substandard quality drugs was revealed. Twenty eight percent of trypanocidal drugs tested failed to comply with quality requirements. There was no significant difference in the frequency of non-compliance between diminazene-based and isometamidium chloride products (P = 0.87) irrespective of the marketing channel (official and unofficial). However, higher rates of non-compliant trypanocides were detected for drugs originating from Asia than from Europe (P = 0.029). Conclusion: The findings revealed the presence of risk factors for the development of drug resistance, i.e. wide distribution of poor quality drugs as well as substandard administration practices. Therefore, it is strongly recommended to enforce regulatory measures for quality control of veterinary drugs, to expand and strengthen veterinary services and to undertake trypanocidal drug efficacy studies of wider coverage. African Animal Trypanosomosis (AAT) is a major disease of cattle in Togo and its control is essentially based on chemotherapy. However, because of excessive use of trypanocides during the past decades, chemo-resistance in the parasites has developed. In order to assess the current situation of AAT and resistance to trypanocidal drugs in Northern Togo, a study was conducted on cattle from December 2012 to August 2013 in the regions of Kara and Savanes. An initial cross-sectional survey was carried out in 40 villages using the Haematocrit Centrifugation Technique (HCT). Out of these, 5 villages with a trypanosome prevalence of >10% were selected for a block treatment study (BT) with diminazene diaceturate (DA: 3.5 mg/kg for a 14-day follow-up) and isometamidium chloride (ISM: 0.5 mg/kg for a 28-day follow-up). Positive blood samples collected during the parasitological surveys and an equivalent number of negatives were further analyzed by PCR-RFLP for trypanosome species confirmation and molecular diagnosis of resistance to DA in Trypanosoma congolense . The results from 1883 bovine blood samples confirmed a high overall trypanosome prevalence of 10.8% in Northern Togo. PCR-RFLP revealed that T. congolense is the dominant pathogenic trypanosome species (50.5%) followed by T. vivax (27.3%), and T. brucei (16.2%). The BT showed varying levels of treatment failures ranging from 0 to 30% and from 0 to 50% for DA and for ISM respectively, suggesting the existence of resistant trypanosome populations in the study area. Our results show that AAT still represents a major obstacle to the development of cattle husbandry in Northern Togo. In areas of high AAT risk, a community-based integrated strategy combining vector control, rational use of trypanocidal drugs and improving the general condition of the animals is recommended to decision makers. Computational psychiatry is a relatively new field that integrates computational models, biological knowledge, and clinical data to provide much needed predictive power to our understanding of brain disorders. The enterprise is far from straightforward, and difficulties include the definition of psychiatric illnesses, variability from subjective elements in measurement, lack of large systematic databases, and mastering subtle technical aspects behind different measurements. In this perspective article, we make the case for studying Huntington's disease as a model system of neurodegeneration that is largely exempt from many of these limitations. The combination of its well-defined genetic basis and the availability of large neuroimaging databases makes it an outstanding candidate for marrying computational and clinical approaches. We highlight particularly promising directions and sketch a roadmap for a research program, with a focus on producing clinical insights based on translating hypotheses concerning brain disorders into features to be used in predictive models. BackgroundLanguage use is of increasing interest in the study of mental illness. Analytical approaches range from phenomenological and qualitative to formal computational quantitative methods. Practically, the approach may have utility in predicting clinical outcomes. We harnessed a real-world sample (blog entries) from groups with psychosis, strong beliefs, odd beliefs, illness, mental illness and/or social isolation to validate and extend laboratory findings about lexical differences between psychosis and control subjects.MethodWe describe the results of two experiments using Linguistic Inquiry and Word Count software to assess word category frequencies. In experiment 1, we compared word use in psychosis and control subjects in the laboratory (23 per group), and related results to subject symptoms. In experiment 2, we examined lexical patterns in blog entries written by people with psychosis and eight comparison groups. In addition to between-group comparisons, we used factor analysis followed by clustering to discern the contributions of strong belief, odd belief and illness identity to lexical patterns.ResultsConsistent with others’ work, we found that first-person pronouns, biological process words and negative emotion words were more frequent in psychosis language. We tested lexical differences between bloggers with psychosis and multiple relevant comparison groups. Clustering analysis revealed that word use frequencies did not group individuals with strong or odd beliefs, but instead grouped individuals with any illness (mental or physical).ConclusionsPairing of laboratory and real-world samples reveals that lexical markers previously identified as specific language changes in depression and psychosis are probably markers of illness in general. Abstract: Decision-making and financial planning for tsetse control is complex, with a particularly wide range of choices to be made on location, timing, strategy and methods. This paper presents full cost estimates for eliminating or continuously controlling tsetse in a hypothetical area of 10,000km2 located in south-eastern Uganda. Four tsetse control techniques were analysed: (i) artificial baits (insecticide-treated traps/targets), (ii) insecticide-treated cattle (ITC), (iii) aerial spraying using the sequential aerosol technique (SAT) and (iv) the addition of the sterile insect technique (SIT) to the insecticide-based methods (i–iii). For the creation of fly-free zones and using a 10% discount rate, the field costs per km2 came to US$283 for traps (4 traps per km2), US$30 for ITC (5 treated cattle per km2 using restricted application), US$380 for SAT and US$758 for adding SIT. The inclusion of entomological and other preliminary studies plus administrative overheads adds substantially to the overall cost, so that the total costs become US$482 for traps, US$220 for ITC, US$552 for SAT and US$993 – 1365 if SIT is added following suppression using another method. These basic costs would apply to trouble-free operations dealing with isolated tsetse populations. Estimates were also made for non-isolated populations, allowing for a barrier covering 10% of the intervention area, maintained for 3 years. Where traps were used as a barrier, the total cost of elimination increased by between 29% and 57% and for ITC barriers the increase was between 12% and 30%. In the case of continuous tsetse control operations, costs were estimated over a 20-year period and discounted at 10%. Total costs per km2 came to US$368 for ITC, US$2114 for traps, all deployed continuously, and US$2442 for SAT applied at 3-year intervals. The lower costs compared favourably with the regular treatment of cattle with prophylactic trypanocides (US$3862 per km2 assuming four doses per annum at 45 cattle per km2). Throughout the study, sensitivity analyses were conducted to explore the impact on cost estimates of different densities of ITC and traps, costs of baseline studies and discount rates. The present analysis highlights the cost differentials between the different intervention techniques, whilst attesting to the significant progress made over the years in reducing field costs. Results indicate that continuous control activities can be cost-effective in reducing tsetse populations, especially where the creation of fly-free zones is challenging and reinvasion pressure high. M3 reflection intensity (IM3) from tetanized, intact skeletal muscle fiber bundles was measured during sinusoidal length oscillations at 2.8 kHz, a frequency at which the myosin motor's power stroke is greatly reduced. IM3 signals were approximately sinusoidal, but showed a “double peak” distortion previously observed only at lower oscillation frequencies. A tilting lever arm model simulated this distortion, where IM3 was calculated from the molecular structure of myosin subfragment 1 (S1). Simulations showed an isometric lever arm disposition close to normal to the filament axis at isometric tension, similar to that found using lower oscillation frequencies, where the power stroke contributes more toward total S1 movement. Inclusion of a second detached S1 in each actin-bound myosin dimer increased simulated IM3 signal amplitude and improved agreement with the experimental data. The best agreement was obtained when detached heads have a fixed orientation, insensitive to length changes, and similar to that of attached heads at tetanus plateau. This configuration also accounts for the variations in relative intensity of the two main peaks of the M3 reflection substructure after a length change. This evidence of an IM3 signal distortion when power stroke tilting is suppressed, provided that a large enough amplitude of length oscillation is used, is consistent with the tilting lever arm model of the power stroke. At the end of the force transient elicited by a fast stretch applied to an activated frog muscle fiber, the force settles to a steady level exceeding the isometric level preceding the stretch. We showed previously that this excess of tension, referred to as "static tension," is due to the elongation of some elastic sarcomere structure, outside the cross bridges. The stiffness of this structure, "static stiffness," increased upon stimulation following a time course well distinct from tension and roughly similar to intracellular Ca2+ concentration. In the experiments reported here, we investigated the possible role of Ca2+ in static stiffness by comparing static stiffness measurements in the presence of Ca2+ release inhibitors (D600, Dantrolene, ²H2O) and cross-bridge formation inhibitors [2,3-butanedione monoxime (BDM), hypertonicity]. Both series of agents inhibited tension; however, only D600, Dantrolene, and ²H2O decreased at the same time static stiffness, whereas BDM and hypertonicity left static stiffness unaltered. These results indicate that Ca2+, in addition to promoting cross-bridge formation, increases the stiffness of an (unidentified) elastic structure of the sarcomere. We designed and implemented a new mitoxantrone-based high-dose chemotherapy regimen to minimize pulmonary injury (seen in carmustine-based regimens) in patients with breast cancer. One hundred and ninety-one breast cancer patients (99 stage II/IIIA; 27 stage IIIB; 65 stage IV responsive to conventional-dose chemotherapy) were treated with high-dose chemotherapy (CTM) delivered over 4 days (cyclophosphamide (6 g/m2), thiotepa (600 mg/m2), and mitoxantrone (24–60 mg/m2)) followed by autologous hematopoietic stem cell rescue. Stage II/III patients received chest wall radiation and tamoxifen (if hormone-receptor positive) after CTM. The 5-year event-free survival (EFS) for stage II/IIIA patients with 10 or more involved axillary lymph nodes (n = 80) was 62 ± 12%. Hormone receptor-positive patients with 10 or more nodes did significantly better than negative patients. The EFS for stage IIIB patients at 5 years was 44 ± 19%; for stage IV patients at 5 years was 17 ± 10%. Stage IV patients achieving complete response in viscera and/or soft tissue prior to CTM did significantly better than those achieving a partial response. There were six (3%) treatment-related deaths including two due to diffuse alveolar hemorrhage. There were no episodes of delayed interstitial pneumonitis. There were six severe cardiac events in 91 patients (6.6%) but none after instituting mitoxantrone dose-adjustment in the final 100 patients. We conclude that CTM is associated with a low treatment-related mortality and little pulmonary toxicity. Abstract. The total fluxes of sulphur dioxide from the Italian volcanoes Etna, Stromboli, and Vulcano were studied using optical remote sensing techniques in three shipbornefield experiments (1992, 1994, and 1997). The main purpose of the experiments was to compare active (laser) techniques with passive monitoring. Differential absorption lidar (DIAL) measurements were implemented by placing the Swedish mobile lidar system on board the Italian research vessel Urania,sailing under the volcanic plumes. Simultaneously, the passive differential optical absorption spectroscopy (DOAS) technique was used for assessing the total overhead gas burden. Finally, correlation spectroscopy (COSPEC) was also implemented in one of the campaigns. Differences in integrated gas column assessment are expected and observed, mostly connected to complex scattering conditions influencing the passive measurements. Since such measurements are much employed in routine volcanic monitoring it is of great interest to model and provide corrections to the raw data obtained. Lidar measurements proved to be quite useful for this purpose. By combining the integrated gas concentration overthe plume cross section with wind velocity data, SO[sub 2] fluxes ofthe order of 1000, 100, and 10 tonnes/day were measured for Mt.
